Detailed planktonic foraminiferal zonations have been established for the Neogene (Latest Oligocene through present) in six DSDP sites in the South Pacific ranging from equatorial to subantarctic latitudes (48°S). Two basic zonal schemes are readily recognized: tropical and temperate. The tropical zonation is best developed in DSDP Site 289 and the temperate zonation in Sites 206, 207A and 284. Tropical and temperate zonations can be linked by a warm subtropical scheme in Site 208, because this sequence includes a mixture of tropical and temperate elements. A site located close to the Subantarctic Convergence (Site 281) contains a zonation largely of temperate character, but the present of cooler elements and some differences in biostratigraphic ranges have required a slightly different biostratigraphic scheme.Although two broad schemes are recognized, none of the biostratigraphic sequences are identical between any of the sites. This reflects differences in biogeography, evolution and diachronous extinction at various latitudes during the entire Neogene. Diachronism in biostratigraphic ranges continue to create difficulties in correlation across such wide latitudes.Our detailed work has required the establishment of new biostratigraphic zonations in certain parts of the Neogene sequence and modifications in some other parts. Otherwise, previously established schemes are followed as closely as possible. In the temperate region, a new zonation has been established for the Early Miocene to early Middle Miocene. For the remainder of the Neogene the zonation of Kennett (1973) has been largely used. The tropical zonation of Blow (1969) is employed in the equatorial Site 289, but with further subdivisions for Zones N4 and N17. For areas intermediate between tropical and temperate latitudes (Site 208), a modified Early Miocene zonation is established based on changes in tropical and temperate elements.The zonal schemes are established on taxa that exhibit both diachronous and isochronous ranges across the latitudes. Zones that are at least partly diachronous include the Globigerinoides trilobus and Globorotalia miozea Zones of Early Miocene age; perhaps the Globorotalia mayeri Zone (its base) of the Middle Miocene; the Globorotalia conomiozea Zone of the Late Miocene; and the Globorotalia crassaformis Zone of the Early Pliocene.A large number of datum levels are recognized based on first evolutionary appearances or extinctions. The most widely applicable datums are as follows: latest Oligocene — Globigerinoides F.A.; Early Miocene — Globoquadrina dehiscens, F.A., Globorotalia kugleri L.A., Catapsydrax dissimilis L.A. and Praeorbulina glomerosa F.A.; Middle Miocene — Orbulina suturalis F.A., Globorotalia peripheroacuta F.A., Fohsella lineage L.A., Globorotalia mayeri L.A.; Late Miocene — “Neogloboquadrina” continuosa L.A., Globoquadrina dehiscens L.A., Globorotalia cibaoensis F.A.; Early Pliocene — Globorotalia puncticulata F.A., Globorotalia margaritae F.A.; Early Pleistocene — Globorotalia truncatulinoides F.A. A number of other datums are identified which assist with correlation over more restricted latitudinal ranges.The evolution of most Neogene planktonic foraminifera is now well established for a wide range of water masses. Evolutionary lineages are primarily centered in the temperate and tropical regions. Tropical lineages have recently been reviewed by Srinivasan and Kennett (1981) and are not discussed in detail here. However, Sphaeroidinellopsis seminulina is now considered to have evolved directly into S. paenedehiscens during the Late Miocene and S. subdehiscens Blow is considered to be junior synonym of S. seminulina.A new evolutionary lineage is recognized in the warm subtropics (Site 208) whereby Globigerina woodi woodi gave rise to Globigerinoides subquadratus via Globigerina brazieri. The discovery of this lineage clearly demonstrates that Globigerinoides is a polyphyletic “genus”. Another major phylogenetic lineage is recognized within the temperate globorotaliids of Early Miocene age as follows: “N.” continuosa → Globorotalia zealandica incognita → G. zealandica zelandica → G. praescitula → G. miozea. Although parts of this lineage have been recognized earlier, the entire phylogeny has previously been underscribed.A new Early to Middle Miocene lineage is recognized in the subantarctic to temperate areas which involve a transition from Globorotalia praescitula to G. challengeri n. sp. via intermediate forms.Two major Neogene globorotaliid lineages — the Menardella of the tropics and Middle Miocene to Recent forms of Globoconella of the temperate areas — are both considered to have evolved from Globorotalia praescitula beginning in the Early Miocene. This evolution initially was restricted to temperate areas but has since separated into distinctly tropical and temperate phylogenetic elements.  相似文献

Occasional spontaneous “action potentiais” are found in mature hyphae of the fungus Neurospora crassa. They can arise either from low-level sinusoidal oscillations of the membrane potential or from a linear slow depolarization which accelerates into a rapid upstroke at a voltage 5–20 mV depolarized from the normal resting potential (near − 180 mV). The “action potentiais” are long-lasting, 1–2 min and at the peak reach a membrane potential near −40 mV. A 2− to 8−fold increase of membrane conductance accompanies the main depolarization, but a slight decrease of membrane conductance occurs during the slow depolarization. Two plausible mechanisms for the phenomenon are (a) periodic increases of membrane permeability to inorganic ions, particularly H+ or Cl- and (b) periodic decreases in activity of the major electrogenic pump (H+) of the Neurospora membrane, coupled with a nonlinear (inverse sigmoid) current-voltage relationship.Identification of action potential-like disturbances in fungi means that such behavior has now been found in all major biologic taxa which have been probed with suitable electrodes. As yet there is no obvious function for the events in fungi.  相似文献

Faunal analyses of planktonic foraminifera and upper-water temperature reconstructions with the modern analog technique are studied and compared to the magnetic susceptibility and gamma ray logs of ODP Core 999A (western Caribbean) for the past 560 kyr in order to explore changes in paleoceanographic conditions in the western Caribbean Sea. Long-term trends in the percentage abundance of planktonic foraminifera in ODP Core 999A suggest two hydrographic scenarios: before and after 480 ka. High percentage abundances of Neogloboquadrina pachyderma and Globorotalia inflata, low abundances of Globorotalia menardii and Globorotalia truncatulinoides, low diversity, and sea-surface temperatures (SST) under 24 °C are typical characteristics occurring from 480 to 560 ka. These characteristics suggest a “shallow” well-oxygenated upper thermocline and the influx of nutrients by either seasonal upwelling plumes and/or eddy-mediated entrainment. The second scenario occurred after 480 ka, and it is characterized by high and fluctuating percentage abundances of Neogloboquadrina dutertrei, G. truncatulinoides, G. menardii, Globigerinita glutinata, Globigerinella siphonifera, and Globigerinoides ruber; a declining trend in diversity; and large SSTs. These characteristics suggest a steady change from conditions characterized by a “shallow” thermocline and chlorophyll maximum to conditions characterized by a “deep” thermocline (mainly during glacial stages) and by more oligotrophic conditions. The influence of the subtropical North Atlantic on the upper thermocline was apparently larger during glacial stages, thus favoring a deepening of the thermocline, an increase in sea-surface salinity, and a dramatic reduction of nutrients in the Guajira upwelling system. During interglacial stages, the influx of nutrients from the Magdalena River is stronger, thus resulting in a deep chlorophyll maximum and a fresher upper ocean. The eddy entrainment of nutrients is the probable mechanism responsible of transport from the Guajira upwelling and Magdalena River plumes into ODP 999A site.  相似文献

The South African members of Australopithecus form a single group, trending from earlier, more gracile or smaller forms, to later, more robust or larger forms, in accordance with the “Law of Cope”. This is supported by the evidence of the lower first deciduous molar: it is only slightly molarized in the earlier, smaller forms and astonishingly heavily molarized in the later, robust forms, such as that of Kromdraai. Hence, Robinson's view that two different genera are represented by the gracile and robust forms is not supported here. A. robustus is seen as a late offshoot of the Australopithecus stem. The resemblance of the Taung dm1 to that of Sinanthropus, except for the more differentiated talonid of the Taung specimen, suggests that the separation of the Australopithecinae and Homininae must have taken place at an earlier stage than that represented by the oldest South African Australopithecinae. The lower jaw of Meganthropus of Java combines certain characteristics of A. africanus with those of A. robustus: Meganthropus might provisionally be called “Australopithecoid”. The geographically intermediate India has yielded a hominid, Ramapithecus punjabicus, but the author does not consider “Kenyapithecus” to be a hominid. “K. wickeri” is a pongid species of its own and “K. africanus” a Proconsul. Ramapithecus sensu stricto is known only from the Indian Siwaliks and the author suggests that the transition from Ramapithecus to a still unknown Australopithecus took place in the same region prior to their migration into Africa and Southeast Asia.  相似文献

Members of the RNA-dependent RNA polymerase (RdRP) gene family have been shown to be essential for dsRNA-mediated gene silencing based on genetic screens in a variety of organisms, including Caenorhabditis elegans, Arabidopsis, Neurospora, and Dictyostelium. A hallmark of this process is the formation of small 21- to 25-bp dsRNAs, termed siRNAs for small interfering RNAs, which are derived from the dsRNA that initiates gene silencing. We have developed methods to demonstrate that these siRNAs produced in Drosophila embryo extract can be uniformly incorporated into dsRNA in a template-specific manner that is subsequently degraded by RNase III-related enzyme activity to create a second generation of siRNAs. SiRNA function in dsRNA synthesis and mRNA degradation depends upon the integrity of the 3-hydroxyl of the siRNA, consistent with the interpretation that siRNAs serve as primers for RdRP activity in the formation of dsRNA. This process of siRNA incorporation into dsRNA followed by degradation and the formation of new siRNAs has been termed “degradative PCR” and the proposed mechanism is consistent with the genetic and biochemical data derived from studies in C. elegans, Arabidopsis, Drosophila, and Dictyostelium. The methods used to study the function of both natural and synthetic siRNAs in RNA interference in Drosophila embryo extracts are detailed. The importance of the 3-hydroxyl group for siRNA function and its incorporation into dsRNA is emphasized and the results support a model that places RNA-dependent RNA polymerase as a key mediator in the RNA interference mechanism in Drosophila.  相似文献

Aspergillus flavus is a common filamentous fungus that produces aflatoxins and presents a major threat to agriculture and human health. Previous phylogenetic studies of A. flavus have shown that it consists of two subgroups, called groups I and II, and morphological studies indicated that it consists of two morphological groups based on sclerotium size, called “S” and “L.” The industrially important non-aflatoxin-producing fungus A. oryzae is nested within group I. Three different gene regions, including part of a gene involved in aflatoxin biosynthesis (omt12), were sequenced in 33 S and L strains of A. flavus collected from various regions around the world, along with three isolates of A. oryzae and two isolates of A. parasiticus that were used as outgroups. The production of B and G aflatoxins and cyclopiazonic acid was analyzed in the A. flavus isolates, and each isolate was identified as “S” or “L” based on sclerotium size. Phylogenetic analysis of all three genes confirmed the inference that group I and group II represent a deep divergence within A. flavus. Most group I strains produced B aflatoxins to some degree, and none produced G aflatoxins. Four of six group II strains produced both B and G aflatoxins. All group II isolates were of the “S” sclerotium phenotype, whereas group I strains consisted of both “S” and “L” isolates. Based on the omt12 gene region, phylogenetic structure in sclerotium phenotype and aflatoxin production was evident within group I. Some non-aflatoxin-producing isolates of group I had an omt12 allele that was identical to that found in isolates of A. oryzae.  相似文献

Stigmaria asiatica Jongmans et Gothan was discovered in situ in a volcanic tuff layer between two coal seams in the lowermost Permian rocks of the Wuda coal district in Inner Mongolia, China. The species is characterized by its consistently small axial width of 20–30 mm and the combination of vertical and horizontal orientation of each axis. It is interpreted as being attached below the stigmarian axes of trees of the Sigillaria brardii-ichthyolepis group forming a deeper reaching “stigmariopsid” system. Such a unique two-tier rooting system gave this group advantages over other arboreous (tree-sized) lycopsids of the late Palaeozoic. Trees of the S. brardii-ichthyolepis group could reach deeper water resources and thus survive in wetlands that experienced dry seasons with a lowered water table. This complex underground rooting system appears to be one factor that enabled this group to survive the regional extirpation of arboreous lycopsids at the Moscovian–Kasimovian boundary in Euramerica and at the same time be a major peat-forming plant in Cathaysia.  相似文献

“Bryndza” is a traditional Slovak dairy product (type of soft cheese) made from sheep cheese which was ripened for 14 days. Because its manufacture, transporting and/or storing represent conditions which facilitate contamination, the effect of enterocin CCM4231 in “bryndza” was investigated with the aim to reduce the contaminant agents. “Bryndza” was divided into equal portions (50 g). The experimental sample (ES) as well as the control sample one (C1) were inoculated with Listeria innocua Li1 strain. The other control samples C2 and C3 were without Li1 strain. C3 control was selected as a reference control. ES and C2 portions were treated with purified enterocin CCM4231 in a concentration of 6400 AU/ml. Before the experimental inoculation, “bryndza” was checked for the presence of contaminant agents. The experiment lasted 1 week and the samples were stored in the refrigerator at 4 °C. Sampling was performed on day 1, on day 4 and on day 7. The control samples C2 and C3 were checked only on day 1 and then after 1 week. The following contaminant agents were detected in “bryndza” before its experimental inoculation with L. innocua Li1 strain: Escherichia coli in the amount 103 cfu/ml/g, Staphylococcus aureus (102 cfu/ml/g) and enterococci (104 cfu/ml/g). In the control sample C2, the number of E. coli was reduced to 102 cfu/ml/g. Enterococci and staphylococci were totally eliminated there. Concerning C3 control, natural decrease of bacteria was found and/or their unchanged counts. The value of pH (5) was stable during the whole experiment. In the experimental sample inoculated with Li1 strain, its counts were decreased immediately after enterocin CCM4231 addition approximately by one order of magnitude. This inhibitory effect was also detectable on day 4 by the difference of one order of magnitude between ES and C1. On day 7, 103 cfu/ml/g of Li1 strain were detected in both samples (ES, C1). The difference by one order of magnitude indicated, an inhibitory effect of enterocin CCM4231 in “bryndza”. However, bacteriocin activity was not determined by laboratory analyses.  相似文献

A ‘tigrina’ mutant stock of xHaynaldoticum sardoum Meletti et Onnis grown under field conditions produced high frequency spikes with abnormal stamens and pistils. The abnormalities occurred mostly in the stamens, which frequently changed to either pseudoovaries, or leaf-shaped structures or, in extreme cases, did not form at all. The developmental abnormalities of the stamens resulted in male sterility in the whole or in part of the spike. It is proposed that the “tigrina” status changes the physiology of the plant (e.g. photosynthetic activity) in such a way that it responds with floral abnormalities brought about by environmental conditions which permit normal flower development in control (non-mutant) plants.  相似文献

We have determined the sequence of an oligonucleotide from the large ribosomal subunit RNA of Staphylococcus aureus whose methylation renders the organism resistant to erythromycin and other antibiotics (the “MLS” phenotype). Analysis of RNase A digests of [3H]methyl-, 32P-labeled RNA yielded the sequence GG · m26A · AAGACp, where m26A is an N6-dimethylated adenosine residue that in sensitive cells is unmethylated. Comparison with homologous sequences recently reported for Saccharomyces cerevesiae mitochondria indicates that an A to G mutation in this latter system mimics dimethylation in St. aureus with regard to functional consequences.  相似文献

Mutant frequency “saturation” in Arabidopsis' “main” inflorescence after fast neutron or X-ray treatment of seeds was observed using cell populations with heterogeneous radiation-sensitivities. Mutant frequencies were determined in (1) morphologically normal and conspicuous plants, (2) main and lateral inflorescences, and (3) different M1-fertility classes. At moderate radiation doses (47 Gy fast neutrons and 233 Gy X-rays) significant differences in mutant frequencies between different M1-fertility classes were observed. This suggests that cell populations with heterogeneous sensitivities were studied. No significant differences were observed either between the mutant frequencies in main and lateral inflorescences or between the ones in inflorescences of morphologically normal and conspicuous plants. This suggests that these inflorescences were formed by cell populations with similar but heterogeneous sensitivities.  相似文献

The fossil evidence of the “robust” australopithecines is reviewed with an emphasis on the taxonomic divisions and evolutionary relationships among this group of hominids. The hypodigms of A. robustus, A. crassidens and A. boisei are described, and the significance of morphological variation within and between these species is assessed. Phylogenetic relationships among the “robust” australopithecines are examined using maximum parsimony analysis, and evolutionary scenarios are evaluated in the light of recent discoveries in East Africa.  相似文献

A water-soluble glucuronan “protuberic acid”, [α]d22 −83.6° and purified from Kobayashia Nipponica, and its physicochemical properties were investigated.The purified protuberic acid was homogeneous as shown by zone electrophoresis, gel filtration over Sepharose 4B, and ultracentrifugation. The sedimentation coefficient was 1.8 S and its intrinsic viscosity was 1.1 dl/g. By gel filtration the molecular weight was estimated to be about 170 000. The results of periodate oxidation, methylation analysis, and partial acid hydrolysis indicated that this acidic polysaccharide has a linear structure of mainly 1,4-linkages and containing an acid-labile linkage. Reduced protuberic acid, [α]d22 −44°, is also described.  相似文献

A review of species in the genus Praon Haliday, 1833 is presented. Twenty described species are keyed and illustrated with scanning electron micrographs and line drawings. The Praon species presented in this work have been identified from 67 aphid taxa occurring on 120 plant taxa. Furthermore, 87 original parasitoid–aphid–plant associations of the species mentioned in the key are presented. Phylogenetic relationships among Praon species are reconstructed using parsimony and cladistic distance methods. Praon abjectum is the sister taxon to the remaining Praon species. We recognized three species group: “Parapraon”, “dorsale-yomenae” and “rosaecola”. Monophyly is suggested for “Parapraon” species group and paraphyly for “dorsale-yomenae” group. Finally, by phylogenetic reconstruction, a close phylogenetic relationship between “Parapraon” and “dorsale-yomenae” species group was found.  相似文献

Using ion-selective microprobes, interactive signalling between barley and Blumeria graminis or Bipolaris sorokiniana has been investigated. The question was raised whether a biotrophically growing fungus manipulates the electrical driving forces (membrane potential, transmembrane pH), required for H+ cotransport of energy-rich compounds. Electrodes were positioned in the substomatal cavity of open stomata or on the leaf surface, and pH was measured continuously up to several days during fungal development. We demonstrate that surface and apoplastic fluids are electrically coupled and respond in a similar manner to stimuli. Apoplastic pH, monitored from the moment of inoculation with conidia, reveals several phases: 2–4 h after inoculation of the barley leaf with either fungus, the host displays rapid transient responses after its first contact with the fungal cell wall; apoplastic pH and pCa increases, cytoplasmic pH and pCa decreases. About 1 day after inoculation, the apoplastic pH increases by up to 2 pH units, which is thought to reflect a resistance response against the intruder. Whereas barley leaf cells possess a membrane potential of −152±5 mV, hyphae of B. graminis yield −251±8 mV, indicative of a substantial driving force advantage for the fungus. Although the resting membrane potential of barley remains constant during the first days after inoculation, leaves infected with B. sorokiniana get confronted with an energy problem, indicated by a retarded repolarization following a “light-off” stimulus. Five days after inoculation, apoplastic pH has increased to 5.97±0.47 (n=11) and does no longer respond to “light-off” when measured within lesions. In contrast, it stays at near normal values outside the lesions and responds to “light-off”.It is concluded that biotrophically growing fungi do not manipulate the cotransport driving forces since (i) any change in apoplastic pH would be experienced by both partners; (ii) the resting membrane potential is not changed. It is suggested that measured pH changes reflect defence responses of the host against the fungus rather than fungal action to increase compatibility.  相似文献

The outer-most layer (“exo-layer”) of the wall was isolated from cell walls of Epidermophyton floccosum. The pure cell walls, obtained by disruption in a Ribi cell fractionator, sonication and centrifugation, were digested with snail enzyme for 12 h. Thereafter, the exo-layer preparation was obtained as the fraction resistant to the snail enzyme. Electron microscopy showed that the exo-layer is a thin, stranded network structure 10–20 nm thick. Chemical analysis of the exo-layer showed that the main components are protein (63 %), mannose (10 %) and glucosamine (17 %). Sodium dodecyl sulfate polyacrylamide gel electrophoresis has revealed that the main band is a glycoprotein containing mannose.  相似文献

Francisella tularensis is a highly infectious Gram-negative bacterium that is the causative agent of tularemia. Very little is known about the molecular mechanisms responsible for F. tularensis virulence, in part due to the paucity of genetic tools available for the study of F. tularensis. We have developed a gene knockout system for F. tularensis that utilizes retargeted mobile group II introns, or “targetrons”. These targetrons disrupt both single and duplicated target genes at high efficiency in three different F. tularensis subspecies. Here we describe in detail the targetron-based method for insertional mutagenesis of F. tularensis genes, which should facilitate a better understanding of F. tularensis pathogenesis. Group II introns can be adapted to inactivate genes in bacteria for which few genetic tools exist, thus providing a powerful tool to study the genetic basis of bacterial pathogenesis.  相似文献

Shifts between adaptive peaks, caused by sampling drift, are involved in both speciation and adaptation via Wright's “shifting balance.” We use techniques from statistical mechanics to calculate the rate of such transitions for a population in a single panmictic deme and for a population which is continuously distributed over one- and two-dimensional regions. This calculation applies in the limit where transitions are rare. Our results indicate that stochastic divergence is feasible despite free gene flow, provided that neighbourhood size is low enough. In two dimensions, the rate of transition depends primarily on neighbourhood size N and only weakly on selection pressure (≈sk exp(− cN)), where k is a number determined by the local population structure, in contrast with the exponential dependence on selection pressure in one dimension (≈exp(− cNs)) or in a single deme (≈exp(− cNs)). Our calculations agree with simulations of a single deme and a one-dimensional population.  相似文献

engrailed has been postulated to be the “selector gene” involved in the establishment of the anterior-posterior compartment border in several imaginal discs and in at least the first two abdominal segments of Drosophila melanogaster. Our study of the effects of different mutant engrailed genotypes on genital disc development provided the following major results: All three terminal primordia (female and male genitalia, and analia) were affected. Different heteroallelic combinations showed different expressivities, and the three terminal primordia were differently affected by the same mutant genotype. The engrailed genotypes deleted specific elements of the adult terminalia without causing associated pattern duplications. The reduced morphology of the male engrailed genital disc was analogous to the pattern deletions observed in the adult terminalia. That the engrailed phenotype is stable was demonstrated by culturing in vivo intact and fragmented engrailed genital discs. Cell death was found in a significant number of mature male en2/en3 genital discs. The results are discussed in terms of the segmental organization of the genital disc and in terms of the “selector gene” function postulated for the engrailed locus. The interpretation that each terminal primordium has an anterior and a posterior compartment is presented and it is assumed that in the genital disc engrailed transforms posterior cells into anterior cells that do not develop, thereby causing the deficiency pattern of the engrailed phenotype.  相似文献
