Effect of aurothioglucose on some allergic and non-allergic skin reactions in animals.

The antiphlogistic effect of aurothioglucose was examined in reversed passive Arthus reaction, delayed-type hypersensitivity and carrageenan oedema. A close association existed between the antiphlogistic effect of aurothioglucose and the pathogenetic role of polymorphonuclear leukocytes. In reversed passive Arthus reaction and in carrageenan oedema the drug decreased the inflammation considerably, whereas in delayed hypersensitivity, where the leukocytes play a less important part, it exerted only a mitigating effect.     

Effect of disodium cromoglycate on mast cell-mediated immediate-type allergic reactions

We investigated the effect of disodium cromoglycate (DSCG) on mast cell-mediated immediate-type hypersensitivity. DSCG inhibited systemic allergic reaction induced by compound 48/80 dose-dependently. Passive cutaneous anaphylaxis was inhibited by 71.6% by oral administration of DSCG (1 g/kg). When DSCG was pretreated at concentration rang from 0.01-1000 g/kg, the serum histamine levels were reduced in a dose dependent manner. DSCG also significantly inhibited histamine release from rat peritoneal mast cell (RPMC) by compound 48/80. We confirmed that DSCG inhibited compound 48/80-induced degranulation of RPMC by alcian blue/nuclear fast red staining. In addition, DSCG showed a significant inhibitory effect on anti-dinitrophenyl IgE-mediated tumor necrosis factor-alpha production. These results indicate that DSCG inhibits mast cell-mediated immediate-type allergic reaction.     

The course of skin radiation reactions after treatment with the preparation nucliderm-gel

The process of epithelialization and the antiseptic effect of nucliderm-gel applied locally have been studied on 63 oncologic patients subjected to radiotherapy. In all cases under study, nucliderm-gel promotes skin epithelialization and has a high antiseptic effect as compared with the drugs applied hitherto.     

Effect of organic solvents in the course of professional exposure on the E rosette test and skin reactions against distreptase and tuberculin

The study comprised 106 workers having professional contact with organic solvents containing benzene, toluene, and xylene during 1 to 122 months. The reduction of the total lymphocyte and the T lymphocyte count (T cells forming both "early" and "late" rosettes with sheep erythrocytes) corresponding to the prolongation of the exposure time to the solvents (the negative correlation) was observed. These alterations are not associated with disturbances of delayed skin hypersensitivity against tuberculin and distreptase. The E rosette test may serve as a practical test for monitoring the biological effects of exposure to organic solvents.     

Activation of plasma Hageman factor and kallikrein in ongoing allergic reactions in the skin

Ten atopic subjects, sensitive to intradermal injection of less than or equal to 10 protein nitrogen units of ragweed or grass pollen antigen, underwent paired antigen and buffer skin chamber incubation over the base of denuded skin blisters. The chamber fluids were sampled over a 6-hr period for histamine and activated Hageman factor and plasma kallikrein which were complexed to C1 inhibitor. In 9 of 10 subjects significantly (p less than 0.01) increased histamine levels (74 +/- 11 ng/ml vs 1.5 +/- 0.55 ng/ml) and kallikrein-C1 inhibitor complexes (2.15 +/- 0.78 ng/ml/hr vs 0.51 +/- 0.09 ng/ml/hr, p less than 0.25) were detected at antigen sites compared with buffer sites, respectively. Increased levels of activated Hageman factor (ng/ml/hr) were detected at antigen sites (1.35 +/- 0.60) compared with buffer sites (0.11 +/- 0.05), (p less than 0.01), in 8 of 10 subjects. Whereas peak levels of histamine were obtained after 1 hr of challenge, both Hageman factor and kallikrein activation, as assessed by complex formation, tended to peak later from the 2nd to the 5th hr. This represents the first demonstration that cutaneous IgE-mediated allergic responses are associated with local activation of the intrinsic plasma coagulation-kinin pathways.     

Redox processes in allergic reactions of the delayed type to microbial antigens]

The influence of common allergic response of delayed type to brucellosis antigen on the processes of glycolysis and dehydrogenase activity of Krebs' cycle in guinea pigs' blood and organs was studied. Along with inhibitory activity of four dehydrogenases investigated there was a depression of the glucolysis processes connected with reduction of the lactate, pyruvate content, and with depression of the LDH activity. An increase of the anaerobic fractions and lowering of aerobic fractions content and of the spectrum excess was seen in the isoenzyme LDH spectrum.     

Effect of ethacrynic acid on the course of cytotoxic glomerulonephritis]

Ethacrynic acid (10 mg/kg subcutaneously 15 days) significantly facilitated the experimental glomerulonephritis in rats. The efficacy of diuretic was confirmed by the improvement of the functional characteristics (the degree of proteinuria, the plasma contents of protein and urea, the creatinine excretion), and by the results of the histological investigation, especially at the second phase of the pathological process.     

Importance of some modern methods of diagnosing allergic reactions to antibiotics]

For diagnosis of drug allergy and antibiotic allergy in particular it is necessary to use several cell reactions in combination in vitro and to estimate them in complex with clinical data. To obtain reliable results it is recommended to observe the conditions of choosing the time of the test, antibiotic optimal concentrations and pH 6.8.
