In an experimental cross between Meishan and Dutch Large White and Landrace lines, 619 F(2) animals and their parents were typed for molecular markers covering the entire porcine genome. Associations were studied between these markers and two fatness traits: intramuscular fat content and backfat thickness. Association analyses were performed using interval mapping by regression under two genetic models: (1) an outbred line-cross model where the founder lines were assumed to be fixed for different QTL alleles; and (2) a half-sib model where a unique allele substitution effect was fitted within each of the 19 half-sib families. Both approaches revealed for backfat thickness a highly significant QTL on chromosome 7 and suggestive evidence for a QTL at chromosome 2. Furthermore, suggestive QTL affecting backfat thickness were detected on chromosomes 1 and 6 under the line-cross model. For intramuscular fat content the line-cross approach showed suggestive evidence for QTL on chromosomes 2, 4, and 6, whereas the half-sib analysis showed suggestive linkage for chromosomes 4 and 7. The nature of the QTL effects and assumptions underlying both models could explain discrepancies between the findings under the two models. It is concluded that both approaches can complement each other in the analysis of data from outbred line crosses.  相似文献

A multivariate QTL detection was carried out on fatness and carcass composition traits on porcine chromosome 7 (SSC7). Single-trait QTLs have already been detected in the SLA region, and multivariate approaches have been used to exploit the correlations between the traits to obtain more information on their pattern: almost 500 measurements were recorded for backfat thickness (BFT1, BFT2), backfat weight (BFW) and leaf fat weight (LFW) but only about half that number for intramuscular fat content (IMF), affecting the detection. First, groups of traits were selected using a backward selection procedure: traits were selected based on their contribution to the linear combination of traits discriminating the putative QTL haplotypes. Three groups of traits could be distinguished based on successive discriminant analyses: external fat (BFT1, BFT2), internal fat (LFW, IMF) and BFW. At least four regions were distinguished, preferentially affecting one or the other group, with the SLA region always influencing all the traits. Meishan alleles decreased all trait values except IMF, confirming an opportunity for marker-assisted selection to improve meat quality with maintenance of carcass composition based on Meishan alleles.  相似文献

Drip loss, one of the most important meat quality traits, is characterized by low heritability. To date, the genetic factors affecting the drip loss trait have not been clearly elucidated. The objective of this study was to identify critical candidate genes affecting drip loss. First, we generated a Pietrain × Duroc × Landrace × Yorkshire commercial pig population and obtained phenotypic values for the drip loss trait. Furthermore, we constructed two RNA libraries from pooled samples of longissimus dorsi muscles with the highest (H group) and lowest (L group) drip loss and identified the differentially expressed genes (DEGs) between these extreme phenotypes using RNA‐seq technology. In total, 25 883 genes were detected in the H and L group libraries, and none was specifically expressed in only one library. Comparative analysis of gene expression levels found that 150 genes were differentially expressed, of which 127 were upregulated and 23 were downregulated in the H group relative to the L group. In addition, 68 drip loss quantitative trait loci (QTL) overlapping with 63 DEGs were identified, and these QTL were distributed mainly on chromosomes 1, 2, 5 and 6. Interestingly, the triadin (TRDN) gene, which is involved in muscle contraction and fat deposition, and the myostatin (MSTN) gene, which has a role in muscle growth, were localized to more than two drip loss QTL, suggesting that both are critical candidate genes responsible for drip loss.  相似文献

为鉴定对猪生产性状有重要影响的新分子标记,文章采用电子克隆结合PCR方法获得了猪肌动蛋白α2(Actin alpha 2, ACTA2)基因编码区序列和部分基因组序列,建立了第2内含子C1554T替换的PCR-HinfⅠ- RFLP基因分型方法,在所检测的7个不同猪群中除大白和梅大群体外,其他群体中均是C等位基因频率高于T等位基因的频率。标记与性状关联分析发现ACTA2基因型与肩部背膘厚、臀部背膘厚、肥肉率、瘦肉率、股二头肌pH和肌内脂肪显著或极显著相关,TT基因型与CC基因型相比具有更高的瘦肉率以及更低的肥肉率和背膘厚。通过Real-time RT-PCR分析发现ACTA2在大白和梅山两个品种猪骨骼肌中的表达量都随着日龄的增加而降低,在各个阶段,梅山猪中的表达量都比大白猪中的表达量高。  相似文献

对脂肪分化相关蛋白(Adipocyte Differentiation-Related Protein, ADFP)基因的外显子进行SNPs 检测, 探讨其作为鸡脂肪性状候选基因的可能性。实验以四川省畜牧科学研究院和大恒家禽育种有限公司培育的优质肉鸡新品系为素材, 采用PCR-SSCP的方法进行SNPs 检测和基因型的分析。结果找到3个SNPs位点: 4 079位由A→T(位点A)、4 843位由C→T(位点B)和7 070位由A→G(位点C)。单位点基因型对屠宰性状的遗传效应分析表明, 位点A的基因型对腿肌率、腹脂重、腹脂率和肌内脂肪含量有显著性影响(P < 0.05), 位点B的基因型对活重和屠体重均有显著性影响(P < 0.05), 位点C的基因型对胸肌重和肌内脂肪含量有显著性影响(P < 0.05), 对胸肌率有极显著性影响(P < 0.01)。初步推断ADFP基因可能是影响鸡脂肪性状的主效基因或与主效基因连锁, 推测可以利用多态位点A和C对鸡腹脂重、腹脂率和肌内脂肪含量进行标记辅助选择。  相似文献

实验采用PCR-RFLP技术分析了不同品种猪MyoG基因3′端MspⅠ位点的多态性,应用单标记回归模型分析了不同基因型与相关性状间的关联性及不同等位基因的遗传效应。结果表明:N等位基因能极显著地增加胴体瘦肉率和眼肌面积,降低皮脂含量(P<0.01),改善胴体产肉量和提高胴体品质;同时,不同基因型对肉质性状的遗传影响作用较大,表现为N等位基因能极显著地降低猪肉品质,使pH值、肉色和肌内脂肪含量降低,并使肌肉的系水力变差(P<0.01)。N等位基因对增加胴体瘦肉率的加性效应值和显性效应值分别为3.929%和-0.602%;对增加眼肌面积的加性效应值和显性效应值分别为2.0985 cm2和-0.5775 cm2;对皮脂率的加性效应值为-3.0245%,显性效应值为-0.4045%。N等位基因对pH1的加性效应值和显性效应值分别为-0.167和0.034;对贮藏损失的加性效应值和显性效应值分别为0.558和-0.347;对肌内脂肪含量的加性效应值和显性效应值分别为-0.963和-0.217。但MyoG基因3′端MspⅠ位点的突变对FOM肉脂仪测定的胴体等级性状的影响不显著(P>0.05)。  相似文献

具有不同遗传特性的猪种具有不同的肉质性状。尤其是地方品种和引进品种间在肉质性状存在极大的差异。在已有的研究中H-FABP,MC4R,ADD1基因同肌内脂肪或背膘相关。利用梅山猪、苏太猪和杜×长×大猪为试验动物,研究上述3个基因的多态性分布和多态性同肌内脂肪和背膘的相关性。结果表明:3个基因的多态性分布在不同猪种间存在极显著的差异,这种差异可能是肌内脂肪(IMF)或背膘(BF)不同的主要原因之一。连锁分析表明:H-FABPADD1基因多态性同IMF有显著的相关,但是同BF没有显著的相关;MC4R基因的多态性同IMF和BF都有显著相关性。说明:H-FABPADD1基因多态性有可能应用到提高IMF,同时不影响BF的育种实践中。   相似文献

Drip loss is an essential evaluation indicator for pork quality. It is closely related to other meat quality indicators, including water-holding capacity, water loss rate and pH value at 45 min (pH1) and 24 h post-mortem (pH2), and is influenced by environmental and genetic factors and their interactions. We previously conducted differentially expressed gene analysis to identify candidate genes affecting drip loss using eight individuals with extremely high- and low-drip loss selected from 28 purebred Duroc pigs. Using 28 identical samples, in the present study, we performed weighted gene co-expression network analysis with drip loss and drip loss-related traits, including water-holding capacity, water loss rate, pH1 and pH2. A total of 25 modules were identified, and five of them correlated with at least two drip loss or drip loss-related traits. After functional enrichment analysis of genes in the five modules, three modules were found to be critical, as their genes were significantly involved in amino acid metabolism, immune response and apoptosis, which have potential relationships with drip loss. Furthermore, we identified five candidate genes affecting drip loss in one critical module, AASS, BCKDHB, ALDH6A1, MUT and MCCC1, as they overlapped with differentially expressed genes detected in our previous study, exhibited protein–protein interactions and had potential biological functions in affecting drip loss according to the literature. The outcomes of the present study enhance our understanding of the molecular mechanisms underlying drip loss and will aid in improving the pork quality.  相似文献

Lipogenic enzyme activities of porcine intra- and intermuscular adipose tissues were determined in growing lean (Large White) and fat (Meishan) pigs. The activities of acetyl-CoA-carboxylase (ACX), malic enzyme (ME) and glucose-6-phosphate dehydrogenase (G6PDH) were compared in both breeds and at both adipose sites. All three enzyme activities were much lower in the intramuscular adipose tissue than in the intermuscular site. Although the lipogenic activity of the intramuscular adipose site was low, it appeared, however, to possess adequate levels of enzymes for in situ lipid synthesis. The highest differences in lipogenic enzyme activities between Meishan and Large White pigs were found in intramuscular adipose tissue, and essentially concerned the activity of malic enzyme which was much higher in Meishan pigs. A close relationship between ME activity and lipid content of intramuscular adipose tissue was observed in both breeds. It was concluded that ME appeared to be a major factor affecting the incidence of higher intramuscular fat in the pig.  相似文献

The mRNA differential display technique was performed to investigate the differences of gene expression in the longissimus muscle tissues from Meishan and Large White pigs. One novel mRNA that was differentially expressed was identified through semi-quantitative RT-PCR and the cDNA complete sequence was then obtained using the rapid amplification of cDNA ends (RACE) method. The nucleotide sequence of the mRNA is not homologous to any of the known porcine genes. Sequence prediction analysis revealed that the this mRNA is not protein-coding mRNA. Polymorphism analyses revealed that there was a C-T mutation on the position of 669 bp and PCR -Dra I-RFLP analyses revealed that Chinese indigenous pig breeds and exotic pig breeds displayed obvious genotype and allele frequency differences at this locus. Association analyses revealed that this polymorphic locus was significantly associated with the drip loss rate, skin percentage, meat color value (m.Longissimus Dorsi, LD), loin eye width, loin eye area, water holding capacity, carcass length, caul fat weight, intramuscular fat (m.Longissimus Dorsi, LD), lean meat weight, lean meat percentage, backfat thickness at buttock (< 0.05).  相似文献

An autosomal scan of the swine genome with 119 polymorphic microsatellite (ms) markers and data from 116 F2 barrows of the University of Illinois Meishan x Yorkshire Swine Resource Families identified genomic regions with effects on variance in carcass composition and meat quality at nominal significance (p-value <0.05). Marker intervals on chromosomes 1, 6, 7, 8 and 12 (SSC1, SSC6, SSC7, SSC8, SSC12) with phenotypic effects on carcass length, 10th rib backfat thickness, average backfat thickness, leaf fat, loin eye area and intramuscular fat content confirm QTL effects identified previously based on genome wide significance (p-value <0.05). Several marker intervals included nominally significant (p-value <0.05) dominance effects on leaf fat, 10th rib backfat thickness, loin eye area, muscle pH and intramuscular fat content.  相似文献

钙蛋白酶抑制蛋白基因是影响猪肉质性状的候选基因之一。本研究以125头地方猪和117头外来猪为材料,研究CAST基因的多态性。结果在CAST基因上检测到一个多态性位点(A876G),并引起了氨基酸残基的改变Lys250 Arg。在地方猪种中仅检测到G (Arg)等位基因,而在外来猪种中A (Lys) 和 G (Arg)两个等位基因均检测到。基因型与肉质性状的关联性分析结果表明,CAST基因型与肌肉的嫩度,屠宰45 min后的pH值及滴水损失存在强相关,又由于地方猪种与外来猪种的肉质性状间存在显著差异。因此,在CAST基因上检测到的多态型位点Lys250Arg的基因型效应有待于进一步研究,并将其有效应用于商品猪生产中。  相似文献

Intramuscular fat content and water-holding capacity are important traits in livestock as they influence meat quality, nutritive value of the muscle, and animal health. As a model for livestock, two inbred lines of the Berlin Muscle Mouse population, which had been long-term selected for high muscle mass, were used to identify genomic regions affecting intramuscular fat content and water-holding capacity. The intramuscular fat content of the Musculus longissimus was on average 1.4 times higher in BMMI806 than in BMMI816 mice. This was accompanied by a 1.5 times lower water-holding capacity of the Musculus quadriceps in BMMI816 mice. Linkage analyses with 332 G3 animals of reciprocal crosses between these two lines revealed quantitative trait loci for intramuscular fat content on chromosome 7 and for water-holding capacity on chromosome 2. In part, the identified loci coincide with syntenic regions in pigs in which genetic effects for the same traits were found. Therefore, these muscle-weight-selected mouse lines and the produced intercross populations are valuable genetic resources to identify genes that could also contribute to meat quality in other species.  相似文献

大白×梅山杂交组合肉质性状的数量性状位点定位分析   总被引:4,自引:0,他引:4  
为寻找影响猪肉质数量性状基因位点的染色体区域 ,以 3头英系大白公猪和 7头梅山母猪建立F2 资源家系。随机选留 14 7头F2 代个体 (1998年 81头 ,2 0 0 0年 66头 ) ,经检测均获得肉质性状表型数据。对资源家系内的所有个体位于染色体 1、2、3、4、6和 7上的 48个微卫星位点进行扩增。利用线性模型最小二乘法分别对各年度及两年综合后的肉质性状进行数量性状位点 (QTL)区间定位 ,利用置换法确定显著性阈值。研究结果表明 :在 2 0 0 0年群体中 ,猪 4号染色体 (SSC4)上定位了肌内脂肪QTL ,达到染色体极显著水平 (P <0 0 1)和基因组显著水平 (P <0 0 5) ,解释表型变异为 5 2 4% ,梅山猪具有增加肌内脂肪QTL ;两年度群体综合后 ,在上述 4号染色体同一区间 ,肌内脂肪QTL接近染色体显著水平 ;股二头肌pH值和半棘肌pH值QTL分别定位在SSC1和 3上 ;在 1998年和 2 0 0 0年群体中分别发现 1个和 3个达染色体显著水平 (P <0 0 5)的系水力QTL ;在 1998年群体中 ,肌肉含水量QTL位于SSC6;两年综合群体中 ,SSC2、6和 7上定位了肌肉含水量QTL ,达到染色体显著水平 ,含水量QTL均有印迹效应 ,梅山和大白猪各有增效基因  相似文献

A resource population was established by crossing one modern broiler sire from a commercial broiler breeder male line with dams from two unrelated highly inbred lines; F1 birds were intercrossed to produce two F2 populations. A variety of phe notypic measurements related to growth, muscling, internal organs, and skeleton were recorded for the F2 populations and contemporary pure inbred and broiler birds. Based on the means and phenotypic distributions of the F2 populations com pared to their parental lines, the effective number of genes affecting each trait and heterosis were estimated and discussed relative to the known genetic selection history for each trait. The results suggest that a high number of genes with small epistatic effects are involved in determining the phenotype for traits that broilers were traditionally selected for, and a lower number of genes with major effects are involved in determining the phenotype for traits related to fitness. The estimated number of genes and the phenotypic distributions of the different traits suggest that a quantitative trait loci (QTL) search might be more effectively applied for traits with a low number of involved genes and a high phenotypic distribution among the F2 birds than for traits that show a lower phenotypic distribution and a high number of genes.  相似文献

The contractile protein troponin I (TnI), a constituent protein of the troponin complex located on the thin filaments of striated muscle, is involved in inhibition of calcium-induced myosin AT Pase activity (and thus contraction). TnI-slow (slow-twitch skeletal muscle isoform, named TNNI1) and TnI-fast (fast-twitch skeletal muscle isoform, named TNNI2) are muscle-fiber-type-specific proteins, and expression of their genes may affect the composition of muscle fiber, thereby influencing the meat quality traits. Thus, the TnI genes are potential candidate genes for traits related to meat quality in animals. Association of 2 SNPs (EU743939:g.5174T>C in intron 4, and EU743939:g.8350C>A in intron 7) of theTNNI1 gene and a SNP (EU696779:g.1167C>T in intron 3) of theTNNI2 gene with 11 meat quality traits were studied on 334 Large White × Meishan F2 pigs. In theTNNI1 gene, g.5174T>C and g.8350C>A were found to be significantly associated with intramuscular fat content and meat color value of biceps femoris. The g.5174T>C also showed significant effects on meat color value and marbling score of longissimus dorsi, as well as pH of longissimus dorsi and semispinalis capitis. The g.1167C>T polymorphism in theTNNI2 gene affected significantly the pH of longissimus dorsi, meat color value of longissimus dorsi and semispinalis capitis, marbling score of longissimus dorsi, and intramuscular fat.  相似文献

Elucidation of the pig microRNAome is essential for interpreting functional elements of the genome and understanding the genetic architecture of complex traits. Here, we extracted small RNAs from skeletal muscle and adipose tissue, and we compared their expression levels between one Western breed (Yorkshire) and seven indigenous Chinese breeds. We detected the expression of 172 known porcine microRNAs (miRNAs) and 181 novel miRNAs. Differential expression analysis found 92 and 12 differentially expressed miRNAs in adipose and muscle tissue respectively. We found that different Chinese breeds shared common directional miRNA expression changes compared to Yorkshire pigs. Some miRNAs differentially expressed across multiple Chinese breeds, including ssc‐miR‐129‐5p, ssc‐miR‐30 and ssc‐miR‐150, are involved in adipose tissue function. Functional enrichment analysis revealed that the target genes of the differentially expressed miRNAs are associated mainly with signaling pathways rather than metabolic and biosynthetic processes. The miRNA–target gene and miRNA–phenotypic traits networks identified many hub miRNAs that regulate a large number of target genes or phenotypic traits. Specifically, we found that intramuscular fat content is regulated by the greatest number of miRNAs in muscle tissue. This study provides valuable new candidate miRNAs that will aid in the improvement of meat quality and production.  相似文献
