The preformed root primordia in stems of Salk viminalis L. consist of undifferentiated cells. Forty-eight hours after activation of the primordia in cuttings a root cap meristem was initiated four to five cell tiers from the surface of the primordia. The cells distal to the meristem divided only in an anticlinal plane, while in the meristem they divided mostly periclinally but sometimes anticlinally. After 72 hours a columella was established and the amyloplasts began to sediment in response to gravity. Shortly after this stage the roots began to bend slightly downward, probably as a geo-tropic response. Six days after activation the root cap consisted of up to 15 tiers of cells. The ultrastucture of the cap cells just prior to emergence was studied in more detail. The plastids in the cells adjoining the root proper were typical proplastids. Distal to this cell tier starch accumulated in the plastids. In the fifth tier the amyloplasts were fully sedimented to the lowermost cell walls. The amount of ER increased with the distance from the initial cells and most of it was located at the distal periclinal cell wall. The nucleus and the vacuoles in the geo-sensitive cells occurred in the space above the sedimented amyloplasts. The cytoplasm was less electron opaque than in the initial cells and the mitochondria had more cristae. In the distal cells of the columella and the lateral root cap secretion of mucilage seemed to have started. Numerous large dictyosomes were associated with large vesicles containing a fibrillar or granular material. The plasmalemma lining the distal periclinal cell wall had separated from the wall. A fibrillar material was present between the plasmalemma and the wall and also in intercellular spaces outside the root cap.  相似文献

Formation and structure of adventitious roots in cuttings of two clones of S. caprea and a hybrid betwen S. caprea and 5. viminalis was studied with light microscopy (LM) and transmission electron microscopy (TEM). The hybrid contained large preformed root primordia, which in cuttings placed in water developed into roots in only three days. One of the S. caprea clones contained minute preformed root primordia, which developed into roots in about eight days. Treatment with indolebutyric acid (IBA) increased the percentage of rooted cuttings and the number of roots formed. Roots emerging from IBA-treated cuttings contained both mature protophloem and protoxylem, while in roots of untreated cuttings only some sieve elements of the protophloem were mature. In the other 5. caprea clone no preformed root primordia were detected, but after treatment with IBA roots appeared in about two weeks. The cambium in treated stems produced a large number of cells, most of which differentiated into xylem. Root primordia were initiated in the newly produced tissue external to the cambium. The roots contained both mature protophloem and protoxylem at emergence. A few roots emerged also from extensive callus tissues formed external to the basal end of the cuttings. Cell enlargement and cell divisions in various parts of the base of the cuttings caused disruption of the peripheral tissues, which made the cuttings susceptible to infection by microorganisms.  相似文献

Root primordia initiate in poplar stems in the secondary growing parts, that is in the parts where the elongation growth is terminated and the leaves are mature. Their initiation is connected with the occurrence of unusual biseriate, rarely multiseriate rays. A small cell group in the secondary phloem is initiated by cell division of the ray. It gradually enlarges by continuing cell division, by the addition of cells adjacent to the cell group and by cambial activity. Thus, a hemispherical root primordium is formed, for which a permanent occurrence of reserve lipids is characteristic. In stems several years old the intraprimordial mitotic activity is rhythmically renewed together with the cambium function renewal. Latent root primordia slightly enlarge with the passing years, whereas mainly the cells localized in their centre divide. Further organization and root histogenesis was not observed either in older root primordia. Adjacent to root primordia, cambial initials produce the secondary xylem elements increasingly. Xylem protuberances are thus formed under root primordia. Primordia initiation is most frequent within the first year of stem development, though they can also initiate in later years.  相似文献

Adventitious root formation in vitro in 1-mm stem slices cut from microshoots of apple cv. Jork 9 was studied using light and electron microscopy. When indole-3-butyric acid (IBA) had been added to the medium, starch grains accumulated during the first 24 h of culture in cells of the cambial region and in cells in the vicinity of vascular tissue and in the primary rays. This accumulation occurred only in the basal part of explants. After that, the nuclei in these cells were activated, and the density of the cytoplasm and the number of cell organelles increased, whereas starch was broken down. Cambium cells started to divide transversely and at 96 h, after several divisions, a continuous ring of isodiametric cytoplasmic cells had appeared around the xylem near the basal cutting surface. The cells in this ring were rich in cell structures, and did not contain large starch grains and a central vacuole. Root meristemoids regenerated from the portions of the ring that were localized in the primary rays. From the other cells in the ring, callus developed. The meristemoids did not grow into the direction of the epidermis as in shoots, but along the vascular bundles. After emergence from the cutting surface, the meristemoids were transformed into small, dome-like primordia. They developed a typical root apex with root cap, root ground meristem and tracheid connection with shoot vascular tissue. Effects of exogenous cytokinins on root formation in pea cuttings   总被引:4,自引:0,他引:4  
Benzylaminopurine (BAP) or zeatin continuously supplied through the rooting solution to cuttings of pea ( Pisum sativum L. cv. Weibull's Marma), inhibited root formation down to a concentration of 3.10−9 M . The inhibitory effect of BAP in the concentration range 10−8–10−7 M was readily reversible if the cuttings were transferred to solutions without cytokinin after treatment for 1–4 days. A slight increase in the number of roots formed was obtained after treatment with low cytokinin concentrations for 1–2 days. Evidence from microscopic studies of primordia formation indicates that BAP inhibits differentiation of primordia at an early stage in their development. Growth of already formed primordia, or root elongation, was considerably less sensitive to the inhibitory effect of BAP. The results indirectly support the hypothesis that endogenous cytokinins prevent root formation in stems of intact plants and may be of importance for the regulation of rooting in cuttings.  相似文献

Adventitious root formation (ARF) was studied in woody leaf bud cuttings of Ficus pumila L., creeping fig. Juvenile cuttings rooted easily, whereas only mature cuttings treated with indole-3-butyric acid (IBA) attained any rooting success. In the rooting process, both juvenile and mature material exhibited dedifferentiation of phloem ray parenchyma, root initial formation, primordia differentiation, and root elongation. The early stages of adventitious rooting were most critical since few primordia were observed in mature controls. The stages leading up to root primordia differentiation and elongation occurred more rapidly in IBA-treated juvenile vs. mature cuttings; however, time differences in both types between first observable roots and maximum rooting were comparable. Root primordia differentiated from basal callus of some cuttings, but neither these nor the few primordia in mature controls elongated into well-developed roots. Anatomical differences between the juvenile and mature material did not account for rooting disparity, nor did presence of perivascular fibers, sclereids, and laticifers retard rooting.  相似文献

In disbudded Azukia stem cuttings, auxin exerted a dual effecton root formation. The first phase of auxin action is identifiedwith the acceleration of cell division, especially longitudinaldivision. In cuttings treated with auxin during the first 24hr, longitudinally divided cells were observed in all 12 rootprimordia, while in water-treated cuttings, such cells wereobserved only in 8 root primordia. The second phase is the promotionof the reaction in which root primordia unable to develop furtherwithout auxin supply develop into roots. Irrespective of thetreatment during the first 24 hr, the auxin-treatment duringthe second 24 hr increased the number of roots protruding fromthe cuttings. Stomata of various sizes are produced on the primary root of Ceratonia siliqua L. Most are generated during embryogenesis, prior to seed desiccation. They can be detected on the dry embryo in a wide zone just above the root tip. Initially, large stomata are formed. These have the ability to induce divisions of their neighbouring cells, creating particular cell patterns around them. Later, small perigenous stomata are generated. As the root grows following seed germination, the stomatal zone overlaps with that of the root hairs. Although root stomata of C. siliqua undergo a structural differentiation that seems almost identical to that of the elliptical stomata formed on leaves, they are unable to move and remain permanently open. Polarizing microscopy of fully differentiated stomata and young stomata at the stage of stomatal pore formation revealed deposition of radial cellulose microfibril systems on their periclinal walls. However, these systems were less developed than those on leaf stomata, a feature that might be responsible for their inactivity. Besides, plastids of the root guard cells (GCs) do not differentiate into chloroplasts but function solely as amyloplasts. Root stomata have a short life span. During rapid and intense root growth, GCs cannot keep pace with the elongation of their neighbouring rhizodermal cells. They therefore split in their mid-region, transversely to the stoma axis. The two parts of the transversely torn stoma are dragged apart and a large opening is formed on the root surface, just above the substomatal cavity. The root stomata, together with these openings, may facilitate increased gaseous exchange during respiration and/or an increased transfer of some nutrients and water in the rapidly growing primary root.  相似文献

The present paper reports results of experiments to develop a system for studying adventitious root initiation in cuttings derived from seedlings. Hypocotyl cuttings of 2-week-old eastern white pine (Pinus strobus L.) seedlings were treated for 5 min with 0, 100, 200, 300, 400, 500 or 600 mg l?1 (0, 0.54, 1.07, 1.61, 2.15, 2.69 or 3.22 mM) 1-naphthaleneacetic acid (NAA) to determine the effect on root initiation. The number of root primordia per cutting was correlated with NAA concentration and the square of NAA concentration. Thus, the number increased from less than one per cutting in the 0 NAA treatment to approximately 40 per cutting at 300 mg l-1 NAA, above which no substantial further increase was observed. The larger number of root primordia formed in response to increasing concentrations of NAA was due to the formation of primordia over a larger proportion of the hypocotyls. Histological analysis of the timing of root primordium formation in hypocotyl cuttings revealed three discernible stages. Progression through these stages was relatively synchronous among NAA-treated hypocotyl cuttings and within a given cutting, but variation was observed in the portion of different cuttings undergoing root formation. Control-treated hypocotyl cuttings formed root primordia at lower frequencies and more slowly than NAA-treated cuttings, with fewer primordia per cutting. Epicotyl cuttings from 11-week-old seedlings also formed adventitious roots, but more slowly than hypocotyl cuttings. NAA treatment of epicotyl cuttings caused more rapid root initiation and also affected the origin of adventitious roots in comparison with nontreated cuttings. NAA-treated epicotyl cuttings formed roots in a manner analogous to that of the hypocotyl cuttings, directly from preformed vascular tissue, while control-treated epicotyl cuttings first formed a wound or callus tissue and subsequently differentiated root primordia within that tissue. This system of inducing adventitious roots in pine stem cuttings lends itself to studying the molecular and biochemical steps that occur during root initiation and development.  相似文献

Three-day-old light-grown sunflower seedlings were de-rootedand incubated in nutrient solutions that either contained orlacked boric acid (B). In the absence of B, in the majorityof the seedlings, no adventitious roots were formed. The micronutrientB caused the development of numerous roots in the lower partof the hypocotyl. The effect of B occurred without the supplyof any phytohormones. A dose-response curve of B-induced rootingyielded an optimum concentration of 0.1 mM boric acid. Histologicalstudies revealed that cell divisions occurred in the controlbut no root primordia developed. In cuttings that were incubatedin B (0.1 m M) root primordia were observed that rapidly developedinto well-differentiated adventitious roots. Sunflower cuttingsthat were planted with their cut end in vermiculite that wasmoistened with nutrient solutions without B degenerated afterseveral weeks. In the presence of B the cuttings formed numerousadventitious roots that entirely replaced the tap root systemof intact seedlings. The rooted cuttings developed into sturdyadult sunflower plants. Summary Root nodule initiation in Pisum sativum begins with cell divisions in the inner cortex at some distance from the advancing infection thread. After penetrating almost the entire cortex, the branches of the thread infiltrate the meristematic area previously initiated in the inner cortical cells. These cells are soon invaded by bacteria released from the infection thread and subsequently differentiate into non-dividing, bacteriod-containing cells. As the initial meristematic centre in the inner cortex is thus lost to bacteroid formation, new meristematic activity is initiated in neighbouring cortical cells. As development proceeds, more cortical layers contribute to the nodule, with the peripheral layer and apical meristem of the nodule not invaded by bacteria.Lateral root primordia are initiated in a region separate from that in which nodules are formed, with the lateral primordia being closer to the root apex. This is interpreted to indicate that the physiological basis for nodule initiation is distinct from that for initiation of lateral roots. The role of a single tetraploid cell in nodule initiation is refuted, as is the existence of incipient meristematic foci in the root. It is suggested that the tetraploid cells in nodule meristems arise from pre-existing endoreduplicated cells, or by the induction of endoreduplication in diploid cortical cells by Rhizobium.  相似文献

DNA synthesis and cell divisions in the quiescent center as well as initiation of lateral root primordia were investigated in the course of incubation of the roots of 3-day-old wheat (Triticum aestivum L.) seedlings on the medium with 0.1 mM NiSO4 for 72 h. It was found that the earliest effect of nickel on proliferation of the quiescent center cells was associated with an increase in the mitotic index 6 h after the beginning of its action. This effect was assumed to depend on an increase in mitosis time. Twelve hours after the beginning of the effect of nickel, mitotic index became somewhat lower, and in 18 h it sharply decreased. Some dividing cells were observed among the initial cells of certain tissues and near the quiescent center even in 72 h. The portion of DNA synthesizing cell sharply decreased in 12 h, and in 48 h such cells were lacking. The main mechanism governing the termination of cell proliferation in the quiescent center as well as in the meristem and calyptrogen of the cap is the inhibition of cell transition to DNA synthesis. The cells that had time to start DNA synthesis or already finished it and were in other phases of the cycle continued a slow progression through the cycle and completed it. Sister cells, produced as a result of divisions, left the mitotic cycle in the phase G1 and transited to dormancy. Nickel did not inhibit initiation and development of lateral root primordia. Resumption of DNA synthesis and cell divisions occurred not only in the pericycle and endodermis participating in the initiation of lateral root primordia but also in the cortex cells in the vicinity of developing primordia. In 18 h after the beginning of the experiment when the rate of the root growth considerably decreased, the region, where primordia were initiated, was located closer to the root tip. Subsequently, when elongation of the cells was inhibited, this region moved closer to the tip until structural disturbances occurred in the nuclei of the endodermal cells located near the root tip and elongated under the effect of nickel. The results concerning the effect of nickel and other heavy metals on root cell proliferation obtained by other researchers and the role of pericycle organization in the translocation and accumulation of nickel in the tissues are discussed.  相似文献

In Azukia stem cuttings, root primordia always appeared in theinterfascicular regions between the endodermis and the interfascicularcambium. Transverse cell divisions were observed as the first eventsin the process of root formation. They began to occur 10 hrafter cuttings had been made and were restricted to the interfascicularregions about 1 mm above the basal cut end of the cutting. Ineach of interfascicular region, 10 to 20 cells divided. Transversedivisions were followed by longitudinal divisions, which beganto occur 18 hr after cuttings had been made. The extent of rooting in cuttings of Phaseolus vulgaris L., and Vigna radiata Wilcz. was affected by 4-chlororesorcinol, a polyphenol oxidase inhibitor. More root primordia and more roots were formed after 4-chlororesorcinol treatment both with and without 10-5M Indole butyric acid. Promotion of rooting was observed also in cuttings of Elaeagnus pungens, Gypsophilia elegans and Kalanchoe blossfeldiana. The enhancement in bean and mung bean was accompanied by a concomitant wider spatial distribution of the primordia and the resulting adventitious roots. The formation of primordia in the treated cuttings was delayed by 12–24 hours, compared to untreated cuttings. The treatment was effective only when given during the first hours after the preparation of the cutting of bean and mung bean, suggesting involvement in the initiation stage. Hypocotyl extracts of mung bean cuttings, pretreated with 4-chlororesorcinol, exhibited reduced polyphenol oxidase activity. Starch grain morphology in laticifer amyloplasts of Euphorbia pulcherrima Willd. (poinsettia) was examined for evidence of starch metabolism in vegetative and flowering plants. Laticifer starch grains in vegetative plants were rod shaped with lengths ranging from 3 to 60 μm. Average grain size was significantly larger in stems than leaves, and in older than younger tissues. Starch grain length frequency was unimodal and approximated a normal probability distribution in stems, but was skewed positively toward smaller grains in leaves. Frequency distributions were shifted toward larger grains in older tissues. Under short-day photoperiod (flowering) conditions, round starch grains formed in latex of stems, and the average length of rod-shaped grains decreased in latex of stems and leaves. Round grains did not occur in laticifers of leaves or bracts. Round starch grains often occurred in aggregates of two or more subunits. Changes in size and shape of latex starch grains indicate that amyloplasts in fully differentiated laticifers metabolize starch. Identification of metabolically active amyloplasts in differentiated laticifers suggests that the function of these organelles may involve starch mobilization under certain physiological conditions.  相似文献

Cuttings of Agathis australis (D. Don) Lindl passed through a well-defined series of morphological changes prior to root emergence. These phases were incorporated into a morphological index which can be used as a guide for the selection of cuttings at known developmental and anatomical stages. After a variable period (lag phase) during which no external change occurred there was an increase in stem diameter a few milimetres above the cut base. This swelling gradually increased in size and isolated bulges developed. Longitudinal splits then arose in the epidermis over the bulges, followed by root emergence through the splits. Root initiation occurred shortly after the sub-basal swelling commenced in cuttings that eventually rooted. Removal of the basal 8 mm of a rooted cutting (which included the roots) usually led to re-rooting of the cuttings. However, if the roots were merely trimmed off, the cutting never formed new roots and always died. The basal region apparently has the capacity to produce only one set of roots. Occasionally the stem diameter continued to increase and the swelling extended to include the basal region. Such cuttings never formed isolated wellings and never rooted.
In general the younger the plant from which the cutting was taken, the shorter the lag phase and the higher the final percentage rooting. Cuttings taken from older plants had a lower rooting percentage and a more variable lag phase, which was related to the time of year the cuttings were taken since root emergence always occurred in spring. Irrespective of the age of the original material there was a constant time period (3–4 weeks) from root initiation to root emergence.  相似文献

In Arabidopsis thaliana, lateral roots are formed from root pericycle cells adjacent to the xylem poles. Lateral root development is regulated antagonistically by the plant hormones auxin and cytokinin. While a great deal is known about how auxin promotes lateral root development, the mechanism of cytokinin repression is still unclear. Elevating cytokinin levels was observed to disrupt lateral root initiation and the regular pattern of divisions that characterizes lateral root development in Arabidopsis. To identify the stage of lateral root development that is sensitive to cytokinins, we targeted the expression of the Agrobacterium tumefaciens cytokinin biosynthesis enzyme isopentenyltransferase to either xylem-pole pericycle cells or young lateral root primordia using GAL4-GFP enhancer trap lines. Transactivation experiments revealed that xylem-pole pericycle cells are sensitive to cytokinins, whereas young lateral root primordia are not. This effect is physiologically significant because transactivation of the Arabidopsis cytokinin degrading enzyme cytokinin oxidase 1 in lateral root founder cells results in increased lateral root formation. We observed that cytokinins perturb the expression of PIN genes in lateral root founder cells and prevent the formation of an auxin gradient that is required to pattern lateral root primordia.  相似文献

MI was found to decrease in LP with increase in cell number, and reached minimal values just before the emergence of LP to form lateral roots. These changes in MI have been correlated with the accumulation of cells in G1, 24 hours before a lateral root is formed. The durations of C and the various phases of the mitotic cycle were also investigated in LP, and compared with those in small primordia. As lateral root primordia increase in cell number, the durations of C, S and G2 become longer, while G1 becomes shorter. Also MI and GF decrease while the proportion of quiescent cells increases. Thus, there is a gradual decrease in the rate of cell proliferation as primordia increase in size. The changes which take place in these parameters during lateral root primordium development have been compared with the events which occur in seed proliferating tissues at the onset of dormancy.  相似文献

BACKGROUND AND AIMS: The aim of this study was to determine the role of nitrogen- and storage-affected carbohydrate availability in rooting of pelargonium cuttings, focusing on the environmental conditions of stock plant cultivation at low latitudes, transport of cuttings, and rooting under the low light that prevails during the winter rooting period in Central European greenhouses. METHODS: Carbohydrate partitioning in high-light-adapted cuttings of the cultivar 'Isabell' was studied in relation to survival and adventitious root formation under low light. Effects of a graduated supply of mineral nitrogen to stock plants and of cutting storage were examined. KEY RESULTS: Nitrogen deficiency raised starch levels in excised cuttings, whereas the concentrations of glucose and total sugars in leaves and the basal stem were positively correlated with internal total nitrogen (Nt). Storage reduced starch to trace levels in all leaves, but sugar levels were only reduced in tissues of non-nitrogen deficient cuttings. Sugars accumulated in the leaf lamina of stored cuttings during the rooting period, whereas carbohydrates were simultaneously exhausted in all other cutting parts including the petioles, thereby promoting leaf senescence. The positive correlation between initial Nt and root number disappeared after storage. Irrespectively of storage, higher pre-rooting leaf glucose promoted subsequent sugar accumulation in the basal stem and final root number. The positive relationships between initial sugar levels in the stems with cutting survival and in leaves with root formation under low light were confirmed in a sample survey with 21 cultivars provided from different sources at low latitudes. CONCLUSIONS: The results indicate that adventitious rooting of pelargonium cuttings can be limited by the initial amount of nitrogen reserves. However, this relationship reveals only small plasticity and is superimposed by a predominant effect of carbohydrate availability that depends on the initial leaf sugar levels, when high-light adaptation and low current light conditions impair net carbon assimilation.  相似文献

Seasonal changes in amyloplast, starch, total soluble sugars, non-reducing sugars and reducing sugars of perennial root nodules of beach pea (Lathyrus maritimus) were studied. Ultrastructural changes in nodule cells of beach pea indicated an accumulation of large amounts of amyloplasts with multiple starch grains in summer months. As the winter season sets in, degradation of amyloplasts and starch grains was detected. The membranes of amyloplasts faded out in winter and the structure of the amyloplasts was lost. The degradation of starch grains showed some electron-dense fiber-like material and amorphous structures. Quantitative data revealed an increase in starch reserves during the summer and depletion during the winter. Total soluble sugar and non-reducing sugar concentrations peaked in the middle of the winter, whereas reducing sugar concentrations showed an increase in the fall. These results indicate that elevated levels of various sugars most likely help to maintain high osmolarity of cells so that the dormant nodules do not freeze during the prolonged periods of cold in the winter.  相似文献
