The in vitro effects of cadmium, copper, lead and zinc, and various cadmium compounds (chloride, sulphate and nitrate) on common carp (Cyprinus carpio) lymphocyte viability and phagocyte activity, were evaluated. The percentage of dead lymphocytes was determined after Trypan blue staining, and phagocyte activity was measured by using the nitroblue tetrazolium (NBT) reduction test. Lead was the most toxic to lymphocytes--the maximum mortality exceeded 30%, and was significantly higher at 1 microM of lead, compared to the control. The maximum mortality caused by cadmium was below 10%, but was significantly elevated with 5 microM or more of cadmium. Zinc induced lymphocyte mortality from 10 microM, whilst no effect was observed with copper. The incubation of full blood with the three cadmium compounds (at 5mg/l of cadmium) showed that cadmium nitrate and cadmium sulphate were more toxic (over 35% and 25% mortality, respectively) than cadmium chloride (about 15% mortality). This was confirmed by the results of tests on isolated cells--1mg/l of cadmium as nitrate and sulphate increased lymphocyte mortality compared to the control and cadmium chloride. Phagocytic activity was less sensitive to heavy metals than was lymphocyte viability. It was significantly reduced following exposure to 50 microM and 100 microM cadmium, and 100 microM zinc, but no effects were observed with either lead or copper.  相似文献

Human exposure to heavy metals is of increasing concern due to their well-documented toxicological and carcinogenic effects and rising environmental levels through industrial processes and pollution. It has been widely reported that such metals can be genotoxic by several modes of action including generation of reactive oxygen species and inhibition of DNA repair. However, although it has been observed that certain heavy metals can inhibit single strand break (SSB) rejoining, the effects of these metals on SSB end-processing enzymes has not previously been investigated. Accordingly, we have investigated the potential inhibition of polynucleotide kinase (PNK)-dependent single strand break repair by six metals: cadmium, cobalt, copper, nickel, lead and zinc. It was found that micromolar concentrations of cadmium and copper are able to inhibit the phosphatase and kinase activities of PNK in both human cell extracts and purified recombinant protein, while the other metals had no effect at the concentrations tested. The inhibition of PNK by environmentally and physiologically relevant concentrations of cadmium and copper suggests a novel means by which these toxic heavy metals may exert their carcinogenic and neurotoxic effects.  相似文献

To better understand the relationship between prenatal exposure to heavy metals and trace elements and the risk of adverse pregnancy outcomes, we investigated the status of heavy metals and trace elements level in a Chinese population by collecting umbilical cord blood. Umbilical cord blood heavy metals and trace elements concentrations were determined by inductively coupled plasma–mass spectrometry. No differences with statistical significance in the median arsenic (As), cadmium (Cd), cobalt (Co), chromium (Cr), copper (Cu), manganese (Mn), nickel (Ni), lead (Pb), strontium (Sr), thallium (Tl), vanadium (V), and zinc (Zn) concentrations were observed between the adverse pregnancy outcome group and the reference group. Titanium (Ti) and antimony (Sb) were found at higher levels with statistical significance in the cord blood samples with adverse pregnancy group when compared to the ones in the reference group. The association between Ti levels and the risk of adverse pregnancy outcomes remained significant after adjusting for potential confounding factors, including newborn weight. These results indicated that environmental exposure to Ti may increase the risk of adverse pregnancy outcomes in Chinese women without occupational exposure.  相似文献

为了进一步研究镉、锌、铅 3种重金属元素间的相互作用以及对植物吸收重金属能力的影响 ,在模拟单一重金属污染试验研究的基础上 ,采用正交回归设计方案 ,研究了 Cd、Zn、Pb复合污染情况下紫花苜蓿和披碱草两种敏感性植物对 3种重金属的吸收效应。结果表明 ,在单一污染条件下 ,镉元素对紫花苜蓿生长的影响大于锌、铅 ,铅元素对披碱草生长的影响大于锌、镉 ;紫花苜蓿对于镉的吸收累积显著高于披碱草 ,植物内镉元素浓度最高达到 1 0 88.5 mg/kg,而披碱草对于铅元素的吸收则高于紫花苜蓿 ,植物内铅元素浓度最高达到 1 3 4 5 .5 mg/kg。在复合污染条件下 ,两种植物对铅、锌和铅、镉的吸收在不同浓度范围内分别存在存在着协同效应和拮抗效应 ;同时两种植物对锌、镉元素在实验涉及浓度范围内都存在着拮抗效应。这对于深入研究复合污染条件下重金属的土壤环境化学行为 ,对植物的综合毒性以及不同植物对重金属的吸收累积效应等 ,具有一定的参考意义  相似文献

Even in the current era of growing technology, the concentration of heavy metals present in drinking water is still not within the recommended limits as set by the regulatory authorities in different countries of the world. Drinking water contaminated with heavy metals namely; arsenic, cadmium, nickel, mercury, chromium, zinc, and lead is becoming a major health concern for public and health care professionals. Occupational exposure to heavy metals is known to occur by the utilization of these metals in various industrial processes and/or contents including color pigments and alloys. However, the predominant source resulting in measurable human exposure to heavy metals is the consumption of contaminated drinking water and the resulting health issues may include cardiovascular disorders, neuronal damage, renal injuries, and risk of cancer and diabetes. The general mechanism involved in heavy metal‐induced toxicity is recognized to be the production of reactive oxygen species resulting oxidative damage and health related adverse effects. Thus utilization of heavy metal‐contaminated water is resulting in high morbidity and mortality rates all over the world. Thereby, feeling the need to raise the concerns about contribution of different heavy metals in various health related issues, this article has discussed the global contamination of drinking water with heavy metals to assess the health hazards associated with consumption of heavy metal‐contaminated water. A relationship between exposure limits and ultimate responses produced as well as the major organs affected have been reviewed. Acute and chronic poisoning symptoms and mechanisms responsible for such toxicities have also been discussed.  相似文献

Lead and zinc tolerant genotypes ofHolcus lanatus L. were grown in culture solution at different cadmium, lead and zinc concentrations, and combinations. In all treatments, an increased inhibition of root length with increasing concentrations of heavy metals was observed. Growth of genotype 1 was better than that of genotype 2 in all treatments, suggesting that genotype 1 is more tolerant. The better root growth of genotype 1, at different cadmium concentrations, than that of genotype 2, indicated the existence of a co-tolerance or greater tolerance of genotype 1 to cadmium. Heavy metal combinations resulted in increased lead or zinc uptake by plants, while cadmium was decreased. In a lead-zinc combination, decreased lead and increased zinc uptake were detected. The different interactive effects of heavy metals on root growth of genotype 1 (additive or synergistic) and genotype 2 (additive or antagonistic) may suggest their differential susceptibility to the above metals.  相似文献

Background

Interleukin-6 (IL-6) may have a protective role in acute liver disease but a detrimental effect in chronic liver disease. It is unknown whether IL-6 is associated with risk of liver-related mortality in humans.

Aims

To determine if IL-6 is associated with an increased risk of all-cause, cardiovascular disease (CVD), cancer, and liver-related mortality.

Methods

A prospective cohort study included 1843 participants who attended a research visit in 1984–87. Multiple covariates were ascertained including serum IL-6. Multivariable-adjusted Cox proportional hazards regression analyses were used to examine the association between serum IL-6 as a continuous (log transformed) variable with all-cause, CVD, cancer, and liver-related mortality. Patients with prevalent CVD, cancer and liver disease were excluded for cause-specific mortality.

Results

The mean (± standard deviation) age and body-mass-index (BMI) of participants was 68 (±10.6) years and 25 (±3.7) Kg/m2, respectively. During the 25,802 person-years of follow-up, the cumulative all-cause, CVD, cancer, and liver-related mortality were 53.1% (N = 978), 25.5%, 11.3%, and 1.3%, respectively. The median (±IQR) length of follow-up was 15.3±10.6 years. In multivariable analyses, adjusted for age, sex, alcohol, BMI, diabetes, hypertension, total cholesterol, HDL, and smoking, one-SD increment in log-transformed serum IL-6 was associated with increased risk of all-cause, CVD, cancer, and liver-related mortality, with hazard ratios of 1.48 (95% CI, 1.33–1.64), 1.38 (95% CI, 1.16–1.65), 1.35 (95% CI, 1.02–1.79), and 1.88 (95% CI, 0.97–3.67), respectively. CRP adjustment attenuated the effects but the association between IL-6 and all-cause and CVD mortality remained statistically significant, independent of CRP levels.

Conclusions

In community-dwelling older adults, serum IL-6 is associated with all-cause, CVD, cancer, and liver-related mortality.  相似文献

Phytochelatins (PCs) are known to play an essential role in the heavy metal detoxification of some higher plants and fungi by chelating heavy metals. However, three recent papers reported that no PCs could be detected in the hyperaccumulator Sedum alfredii Hance upon cadmium, lead or zinc treatment, respectively. In this paper, PC synthesis was assayed again in the mine population of S. alfredii with the help of reversed phase high-performance liquid chromatography (HPLC), HPLC-mass spectrometry, and HPLC-tandem mass spectrometry. Our data showed that PC formation could be induced in the leaf, stem and root tissues of S. alfredii upon exposure to 400 microM cadmium, and only in the stem and root when exposed to 700 microM lead. However, no PCs were found in any part of S. alfredii when it was subjected to exposure to 1600 microM zinc.  相似文献

Soil heavy metal contamination is a major environmental concern, and health risk associated with heavy metals is not fully explored. A combination of spatial analysis and Monte Carlo simulation was successfully used to identify the possible sources and health risk of cadmium (Cd), arsenic (As), mercury (Hg), lead (Pb), chromium (Cr), and copper (Cu) in soils collected from a rapidly developing region of China. It was found that mean concentrations of Cd (0.17 mg/kg ), As (8.74 mg/kg ), Hg (0.15 mg/kg ), Pb (27.28 mg/kg ), and Cu (33.32 mg/kg ) were greater than the soil background values. Accumulation and spatial variability of heavy metals were significantly affected by anthropogenic activities and soil properties. The risk assessment indicated that non-carcinogenic risk was not significant. However, 95% of the total cumulative carcinogenic risk of children was greater than 1E-05, implying high potential carcinogenic risk with As and Pb representing the major contributors. Ingestion of heavy metals in the soils was the main exposure pathway compared with the inhalation and the dermal exposure. Concentration of heavy metals in the soils, particulate emission factor, and dermal exposure ratio were the major parameters affecting health risk. This study highlights the importance of assessment of soil direct exposure health risk in studying heavy metal exposures.  相似文献

Abstract

Phytoremediation with vetiver was investigated in relation to heavy metal contaminated soil in Thailand. The work compared the performance of two species of vetiver named Songkhla 3 (Chrysopogon zizaniodes) and Prachuap Khiri Khan (Chrysopogon nemoralis) in absorbing lead, zinc, and cadmium in contaminated soils. Toxicity Characteristic Leaching Procedure (TCLP), and Allium tests were conducted to determine toxicity of treated soil. Ethylenediaminetetraacetic acid (EDTA) was also used to increase heavy metals concentration in solution in soil, which led to an increase in translocation and bioaccumulation factors. In general, results showed that concentration of heavy metals decreased in soil and increased in both the shoots and roots of vetivers during a 4-month treatment period. TCLP results indicated that the concentration of zinc and cadmium in contaminated soil was reduced over treatment time, and significantly increased after EDTA was applied. To confirm vetiver performance in phytoremediation, Allium testing showed that remained heavy metals in treated soils had no effect on nucleus aberration. Songkhla 3 and Prachuap Khiri Khan showed similar trends in their ability to remediate lead, zinc, and cadmium from contaminated soil. Both species could accumulate higher concentrations of heavy metals in their shoots and roots over time, and with EDTA application.  相似文献

The effects of heavy metals, both toxic (Pb, Cd) and essential (Cu, Zn) on the ultrastructure and the induction of Heat Shock Protein 70 (HSP70) have been studied in the aquatic moss Leptodictyum riparium Hedw. In vitro cultured L. riparium was treated with different heavy metals, both toxic, as cadmium or lead; and essential microelements such as Copper or Zinc concentrations ranging from 10(-3) to 10(-6) M to investigate both ultrastructural damage and HSP induction. TEM observations showed that sub-lethal concentrations of heavy metals caused only slight changes, largely localized in the chloroplasts. Among all the heavy metals tested, cadmium caused the most severe modifications. Heavy metals caused the decrease of the soluble protein content and the enhancement of proteins reacting versus HSP70 antibodies, suggesting that molecular chaperons might be involved in the resistance to toxic effects of lead, cadmium, copper and zinc. Therefore, the induction of HSP70 in L. riparium would confer a higher resistance to pollutants under stressful conditions lethal for other mosses and higher plant species. These results suggest that the moss L. riparium can tolerate heavy metals stress without incurring severe cellular/subcellular damage. Therefore it can be used as a useful indicator of heavy metals accumulation.  相似文献

The distributions of cadmium, chromium, lead, and zinc in eelgrass were studied in samples collected from the field, and the loss/accumulation of the metals during decomposition of eelgrass leaves was studied in laboratory experiments.Concentrations of heavy metals in the below grounds parts were greater in the roots than in the different age groups of the rhizomes. In the rhizomes, the highest concentrations of lead were recorded in the oldest parts, whereas highest chromium and zinc concentrations were found in the youngest parts. The concentration of cadmium did not vary. In the above ground parts, the concentrations of cadmium, lead, and zinc increased with age of the leaves, and concentrations in the leaves were greater than in the stem fraction. The concentrations of chromium decreased with age of the leaves.In the laboratory study of decomposition of leaf material, the concentrations of chromium, lead and zinc increased significantly and a net absorption from the surrounding water was recorded. Cadmium concentrations were relatively constant and a loss of cadmium was proportional to the release of soluble organic compounds indicating an association of cadmium with the soluble phase.The investigation demonstrated the utility of compositional analyses and decomposition experiments in assessing the significance of eelgrass in the heavy metal cycling in coastal areas. Furthermore, significant differences in the fate of heavy metals associated with eelgrass detritus are discussed.  相似文献

Objective

Low-carbohydrate diets and their combination with high-protein diets have been gaining widespread popularity to control weight. In addition to weight loss, they may have favorable short-term effects on the risk factors of cardiovascular disease (CVD). Our objective was to elucidate their long-term effects on mortality and CVD incidence.

Data sources

MEDLINE, EMBASE, ISI Web of Science, Cochrane Library, and ClinicalTrials.gov for relevant articles published as of September 2012. Cohort studies of at least one year’s follow-up period were included.

Review methods

Identified articles were systematically reviewed and those with pertinent data were selected for meta-analysis. Pooled risk ratios (RRs) with 95% confidence intervals (CIs) for all-cause mortality, CVD mortality and CVD incidence were calculated using the random-effects model with inverse-variance weighting.

Results

We included 17 studies for a systematic review, followed by a meta-analysis using pertinent data. Of the 272,216 people in 4 cohort studies using the low-carbohydrate score, 15,981 (5.9%) cases of death from all-cause were reported. The risk of all-cause mortality among those with high low-carbohydrate score was significantly elevated: the pooled RR (95% CI) was 1.31 (1.07–1.59). A total of 3,214 (1.3%) cases of CVD death among 249,272 subjects in 3 cohort studies and 5,081 (2.3%) incident CVD cases among 220,691 people in different 4 cohort studies were reported. The risks of CVD mortality and incidence were not statistically increased: the pooled RRs (95% CIs) were 1.10 (0.98–1.24) and 0.98 (0.78–1.24), respectively. Analyses using low-carbohydrate/high-protein score yielded similar results.

Conclusion

Low-carbohydrate diets were associated with a significantly higher risk of all-cause mortality and they were not significantly associated with a risk of CVD mortality and incidence. However, this analysis is based on limited observational studies and large-scale trials on the complex interactions between low-carbohydrate diets and long-term outcomes are needed.  相似文献

Changes in essential trace elements and heavy metals may affect the atherosclerotic state of patients on maintenance hemodialysis (HD). The aim of the study was to evaluate the relation between the serum levels of some trace elements and heavy metals (iron, zinc, manganese, copper, magnesium, cobalt, cadmium, lead, and copper/zinc ratio) and carotid artery intima-media thickness (CIMT) in HD patients. Fifty chronic HD patients without known atherosclerotic disease and 48 age- and sex-matched healthy individuals were included in the study. The serum levels of trace elements (iron, zinc, manganese, copper, and magnesium) and heavy metals (cobalt, cadmium, and lead) were measured by Atomic Adsorption Spectrophotometer (UNICAM-929). CIMT was assessed by carotid artery ultrasonography. The serum levels of iron, zinc, and manganese were lower; levels of copper, magnesium, cobalt, cadmium, lead, and copper/zinc ratio were higher in HD patients compared to controls. BackgroundIn general, smoking increases the risk of mortality. However, it is less clear how the relative risk varies by cause of death. The exact impact of changes in smoking habits throughout life on different mortality risks is less studied.MethodsWe studied the impact of baseline and lifetime smoking habits, and duration of smoking on the risk of all-cause mortality, mortality of cardiovascular diseases (CVD), chronic obstructive pulmonary disease (COPD), any cancer and of the four most common types of cancer (lung, colorectal, prostate, and breast cancer) in a cohort study (Vlagtwedde-Vlaardingen 1965–1990, with a follow-up on mortality status until 2009, n = 8,645). We used Cox regression models adjusted for age, BMI, sex, and place of residence. Since previous studies suggested a potential effect modification of sex, we additionally stratified by sex and tested for interactions. In addition, to determine which cause of death carried the highest risk we performed competing-risk analyses on mortality due to CVD, cancer, COPD and other causes.ResultsCurrent smoking (light, moderate, and heavy cigarette smoking) and lifetime persistent smoking were associated with an increased risk of all-cause, CVD, COPD, any cancer, and lung cancer mortality. Higher numbers of pack years at baseline were associated with an increased risk of all-cause, CVD, COPD, any cancer, lung, colorectal, and prostate cancer mortality. Males who were lifetime persistent pipe/cigar smokers had a higher risk of lung cancer [HR (95% CI) = 7.72 (1.72–34.75)] as well as all-cause and any cancer mortality. A longer duration of smoking was associated with a higher risk of COPD, any and lung cancer [HR (95% CI) = 1.06 (1.00–1.12), 1.03 (1.00–1.06) and 1.10 (1.03–1.17) respectively], but not with other mortality causes. The competing risk analyses showed that ex- and current smokers had a higher risk of cancer, CVD, and COPD mortality compared to all other mortality causes. In addition, heavy smokers had a higher risk for COPD mortality compared to cancer, and CVD mortality.ConclusionOur study indicates that lifetime numbers of cigarettes smoked and the duration of smoking have different impacts for different causes of mortality. Moreover, our findings emphasize the importance of smoking-related competing risks when studying the smoking-related cancer mortality in a general population and that smoking cessation immediately effectively reduces the risk of all-cause and any cancer mortality.  相似文献

Background

Current guidelines for the prevention of cardiovascular disease (CVD) recommend diabetes as a CVD risk equivalent. However, reports that have examined the risk of diabetes in comparison to pre-existing CVD are lacking among older women. We aimed to assess whether diabetes was associated with a similar risk of total and cause-specific mortality as a history of CVD in older women.

Methodology/Principal Findings

We studied 9218 women aged 68 years or older enrolled in a prospective cohort study (Study of Osteoporotic Fracture) during a mean follow-up period of 11.7 years and compared all-cause, cardiovascular and coronary heart disease mortality among 4 groups: non-diabetic women with and without existing CVD, diabetic women with and without existing CVD. Mean (SD) age of the participants was 75.2 (5.3) years, 3.5% reported diabetes and 6.8% reported existing CVD. During follow-up, 5117 women died with 36% from CVD. The multivariate adjusted risk of cardiovascular mortality was increased among both non-diabetic women with CVD (hazard ratio (HR) 2.32, 95% CI: 1.97–2.74, P<0.001) and diabetic women without CVD (HR 2.06, CI: 1.62–2.64, P<0.001) compared to non-diabetic women without existing CVD. All-cause, cardiovascular and coronary mortality of non-diabetic women with CVD were not significantly different from diabetic women without CVD.

Conclusions/Significance

Older diabetic women without CVD have a similar risk of cardiovascular mortality compared to non-diabetic women with pre-existing CVD. The equivalence of diabetes and CVD seems to extend to older women, supporting current guidelines for cardiovascular prevention.  相似文献

The effects of 45 days exposure to mercury, cadmium and lead on tissue GSH levels were studied in Oreochromis aureus (Steindachner). Liver, brain, gill filaments, intestine and caudal muscle were assayed after exposure to these heavy metals singly, or in combination. Significant increases in intestinal GSH concentrations consistently occurred after exposure to mixtures of heavy metals. Exposure to cadmium or lead did not change hepatic GSH levels, while exposure to two different concentrations of mercury caused significant increases in hepatic GSH.  相似文献

Microbially mediated leaching and solubilization of zinc ore concentrate by native aquatic microbial communities incubated under aerobic and oxygen‐limited conditions were examined in static microcosms consisting of stream sediment and water. Sterile controls provided information on abiotic sulfide oxidation and leaching of zinc, lead, cadmium, and copper. The flux of these heavy metals from the sediments to the water column was greatest under biotic oxygen‐limited conditions. When calculated as the percentage of total metal available in zinc concentrate‐amended microcosms, the order of metal solubilization under oxygen‐limited conditions was lead, copper, zinc, and cadmium. Under biotic aerobic conditions, the order of solubilization was zinc, lead, cadmium, and copper. This study indicates that aquatic heterotrophs are capable of leaching and solubilizing metallic sulfides under conditions of neutral to slightly acidic pH and are effective in releasing heavy metals to the water column under oxygen‐limited conditions.  相似文献

我国铜铅锌有色冶炼固废种类多、存量大,富含多种环境毒害元素如镉铬汞砷等,而目前固废末端处置方式单一,对其所含的大量铜铅锌银镍等有价金属组分也未能有效分离,固废污染防控与高效资源化利用问题突出,已成为制约产业绿色可持续发展的关键因素。以长江经济带典型铜铅锌综合冶炼基地为例,设置直接处理(情景1)、内部循环(情景2)和协同利用(情景3)三种锌冶炼固废处置情景模式,结合物质投入产出法、层次分析法与熵值法,构建资源能耗、环境风险、物质循环与经济效益的多维生态效率指标核算方法体系,对三种情景模式开展生态效率综合评估分析。结果表明,情景3因加强了锌冶炼固废在铜、铅冶炼系统间的协同转化,其生态效率综合评价指数最高,为0.2246,较情景1和情景2分别高出313.6%和25.5%;从资源能耗角度来看,情景2单位产品能耗最高,达0.3086;而从物质循环和经济效益角度来看,情景3固废综合利用率最高,达97.6%,铜铅锌三种有价金属回收率达97.2%,利润总额达48131.3万元;但在环境风险方面,尽管情景3固废中重金属污染排放减少1470.4 t/a,但废气中铅砷镉汞等重金属污染排放却比情景1增加了3倍...  相似文献

利用生态系统研究了白银有色金属冶炼矿区周围环境中重金属的分布及生物有效性。结果表明 ,工厂在冶炼过程中已造成 Pb、Cd、Cu、Zn对周围环境不同程度的污染 ,其含量与距工厂的距离呈负相关 ;重金属在各种生物体内均有不同程度的吸收和累积 ,其吸收累积量随重金属和生物种类的不同而有差异 ;土壤的污染 ,使农作物和牧草中 Pb、Cd含量超过动物的最大耐受量和中毒的临界值 ;动物研究发现 ,肾脏、骨骼和肝脏是机体内重金属蓄积的主要器官。因此 ,放牧动物可作为环境重金属污染状况的标识 ,对评价重金属环境污染对当地人群的危害也有重要意义  相似文献
