1. We assessed the terrestrial export of organic carbon, which effectively supported aquatic bacterial production (BP), from small boreal catchments during spring flood. We analysed stream runoff from nine small catchments with different proportions of peat mires and coniferous forests by monitoring the dissolved organic carbon (DOC) flux in combination with conducting bacterial bioassays.
2. Multiple linear regression analysis showed that BP during 7-day-dark bioassays (BP7; μg C L−1day−1) was explained by both the quantity and quality (low-molecular weight fractions) of the DOC. BP7 can be used as a measure of export of terrestrial organic carbon that is highly bioavailable.
3. Total export of DOC during spring flood from the different catchments ranged from 20 to 27 kg ha−1 and was negatively correlated to forest cover (%). However, the export of BP7 carbon was positively correlated to forest cover and varied from about 0.1 kg ha−1 in mire-dominated streams to about 0.2 kg ha−1 in forest-dominated streams.
4. The high bioavailability of forest carbon suggests that forests are the main contributors of BP-supporting carbon in boreal streams although mires have higher area-specific export of DOC.  相似文献

SUMMARY. 1. Sephadex gel filtration of filtered water from small, Finnish forest lakes demonstrated abiotic movement of 33P from added PO4 to two higher molecular weight fractions. This movement was most pronounced in waters with high humic content which also had high iron content. The two fractions which took up 13P had nominal molecular weights of > 100,000 and 10,000-20,000.
2. An equilibrium existed between free PO4 and the two fractions. However, one fraction, at least, appeared to exist in two phases, with one phase in rapid equilibrium with free PO4 but the other in only slow equilibrium.
3. Additions of ferric iron up to 1 mg Fe l−1 to the filtered lake water stimulated movement from free PO4, provided high concentrations of humic materials were present. In the absence of humic materials even 0.1 mg Fe 1−1 would precipitate all added 33PO4.
4. The high molecular weight P was only partially reactive with standard molybdate reagents. Exposure of the high molecular weight P to sunlight caused a small release of PO4 under the experimental conditions employed.
5. Possible implications for biological phosphorus demand of such sequestration of free PO4 by humic materials in combination with iron are discussed.  相似文献

SUMMARY 1. The influences of temperature and dissolved nitrates and phosphates on microbial activity associated with suspended fine particulate organic matter (seston) were evaluated in four headwater streams in the southern Appalachian Mountains.
2. Temperature manipulations of ± 5°C always induced significant changes in [14C] glucose mineralization (ANOVA; P <0.05) and [3H]thymidine incorporation (ANOVA; P <0.05).
3. Nutrient amendments of 1.0 mg NO3 I−1 and 0.05 mg PO4I−1 induced no significant alterations in bacterial mineralization of [14C]glucose (ANOVA; P >0.05) or incorporation of [3H]thymidine (ANOVA; P >0.05) in short-term (i.e. 3 h) experiments.
4. Microorganisms attached to refractory particulate organic matter do not appear to be limited by nitrogen or phosphorus even in streams with ambient nutrient concentrations as low as 0.06 mg NO3 I−1 and <0.03 mg PO4 I−1.
5. Our results indicate that variations in water temperature resulting from diurnal and seasonal temperature fluctuations, forest clear-cutting, and catchment elevation and aspect can have marked effects upon microbial activity and production, while short-term alterations in nutrient regime appear to have no significant effect on microbial activity associated with seston.  相似文献

1.  The δ13C and δ15N signatures of zooplankton vary with dissolved organic carbon (DOC), but inconsistent and limited taxonomic resolution of previous studies have masked differences that may exist among orders, genera or species and are attributable to dietary and/or habitat differences. Here we investigate differences among the isotopic signatures of five zooplankton taxa ( Daphnia , Holopedium , large Calanoida, small Calanoida and Cyclopoida) in Precambrian shield lakes with a sixfold range of DOC concentration.
2.  δ13C signatures of Daphnia , small calanoids and large calanoids became more depleted with increasing lake DOC, whereas Holopedium and cyclopoid δ13C became enriched with increasing DOC concentration.
3.  The variability of δ13C and δ15N isotopic signatures among zooplankton groups was reduced in high-DOC, compared to low-DOC lakes, especially for δ13C. Differences in δ13C and POM-corrected δ15N accounted for up to 33.7% and 19.5% of the variance, respectively, among lakes of varying DOC concentration.
4.  The narrow range of signatures found in higher DOC lakes suggests that different taxa have similar food sources and/or habitats. In contrast, the wide range of signatures in low-DOC lakes suggests that different taxa are exploiting different food sources and/or habitats. Together with the variable trends in zooplankton isotopic signatures along our DOC gradient, these results suggest that food web dynamics within the zooplankton community of temperate lakes will change as climate and lake DOC concentrations change.  相似文献

SUMMARY. 1. Single-station diel oxygen curves were used to monitor the oxygen metabolism of the Ogeechee River, a sixth-order blackwater river in the Coastal Plain of southeastern U.S.A., over a period of 4 years. Ecosystem production ( P) and respiration (R) were estimated, and P/R ratios calculated to determine the extent of autotrophy characteristic of this type of river. The potential error in oxygen metabolism caused by photo-oxidation of dissolved organic carbon (DOC) in the water was measured and found to be minor.
2. Rates of ecosystem primary production measured were intermediate compared to other rivers, ranging from 0.49 to 13.99g O2m−2 day −1.Primary production rates were highest during the summer when water levels were low. Regression analysis indicated that water depth and light absorption by DOM were significant predictors of primary production in this river. Incident light intensities were not significantly correlated with production rate.
3. Respiration rates were unusually high, varying between 3.70 and 11.5 g O2 m−2 day − 1. System respiration also varied seasonally, but less than primary production. Rates were slightly higher in spring and summer.
4. With one exception, P/R ratios were considerably lower than l throughout the study period, indicating that the Ogeechee River was highly heterotrophic. PIR ratios ranged from 0.09 to 1.3, and averaged 0.25.
5. A carbon budget calculated for this river showed floodplain inputs were 7 times autochthonous production. Organic carbon turnover length was 690 km, considerably longer than has been reported for lower-order rivers.  相似文献

SUMMARY. 1. Regeneration of ammonium and phosphate by macro-zooplankton (Cladocera. adult copepods. and copepodites) was measured in Lake Calado. an Amazon floodplain lake, Macrozooplanktonabundances ranged between 1×104 and 3×105 individuals m−2.
2. Phosphate regeneration ranged from 0.2 to 1.3 μ mol PO4 m−2 b−1at station 1. located 2 km from the Solimoes River, and from 1.6 to8.3 μ mol PO4 m−2 h −1 at station 3, located 7 km from the SolimoesRiver. Ammonium regeneration at stations 1 and 3 ranged from 1.7 to11.9 and from 13.4 to 77.2 μ mol NH4 m−2 h−1. respectively.
3. Zooplankton regenerated ammonium and phosphate at similarrates during rising and falling waier. Regeneration by macrozooplankton was low compared to other tropical lakes and compared to microbesand microzooplankton in Lake Calado.  相似文献

SUMMARY. 1. Published data are used to construct a tentative carbon budget for the ecosystem of the lower river Rhine and its sedimentation areas in The Netherlands.
2. It is estimated that 287 × 106kg Cy−1 of particulate organic material is transported by the river Rhine, and 100 × 106 kgCy−1 of this material is deposited in the delta.
3. Phytoplankton and nitrifying bacteria seem to contribute significantly to the carbon budget, by producing c. 78 × 106kgCy−l.
4. The mineralization in the water (estimated from routine determinations of biological oxygen demand and from in situ diurnal fluctuations of oxygen) and in the bottom (estimated from the denitrification rates in the delta, from the production rate of methane and from overall oxygen consumption) is shown to degrade c. 50% of the carbon input plus autochthonous production.
5. The carbon budget and oxygen regime in the lower Rhine suggest that after decades of severe organic pollution the river has more or less resumed the normal, slightly heterotrophic state of a large lowland river.  相似文献

Abstract: Substrate utilization of microbial cells extracted from soil with a 0.85% aqueous sodium chloride solution, was determined to estimate effects on soil microorganisms at the community level with microtiter plates (Biolog GN®) containing 95 different sources of organic carbon. A consistent pattern of utilized substrates was obtained after 24 h of microtiter plate incubation at 28°C. The absorbance values (OD590) obtained from a microtiter plate reader after background correction were transformed by using the average absorbance values of oxidized substrates as a threshold to distinguish between well utilized and poorly or non-utilized substrates and thereby reduce variances between replicates. Doubling times of the extracted soil microorganisms in the microtiter plates were tested with 12 substrates and ranged from 1.96 h to 3.23 h, depending on the carbon source. The carbon source utilization assay was used to assess the effects of soil inoculation with Corynebacterium glutamicum with and without a genetically engineered plasmid (pUN1; 6.3 kb), which encoded for the synthesis of the mammalian protease inhibiting peptide, aprotinin. Additionally, aprotinin itself was added at two concentrations to soil samples. An identical decrease in the number of carbon sources utilized, especially carbohydrates, occurred upon soil inoculation with both C. glutamicum strains after inoculation with 106 cells g−1 soil. This effect was only detectable during the first three weeks of incubation, as long as cell numbers of C. glutamicum (pUN1) were above 105 cfu g−1. Soil amendment with aprotinin resulted in utilization of additional substrates, most of them carbohydrates. With 0.1 mg aprotinin g−1 soil this stimulation lasted 2 days and with 10 mg g−1 it lasted for 7 days.  相似文献

1. Increasing carbon dioxide concentration (E: 680 μl CO2 litre–1 vs ambient, A: 355 μl CO2 litre–1) around late-successional Alpine sedge communities of the Swiss Central Alps (2450 m) for four growing seasons (1992–1995) had no detectable effect on symbiotic N2 fixation in Trifolium alpinum —the sole N2-fixing plant species in these communities (74 ± 30 mg N m–2 year–1, A and E plots pooled).
2. This result is based on data collected in the fourth growing season showing that elevated CO2 had no effect on Trifolium above-ground biomass (4·4 ± 1·7 g m–2, A and E plots pooled, n = 24) or N content per unit land area (124 ± 51 mg N m–2, A and E pooled), or on the percentage of N Trifolium derived from the atmosphere through symbiotic N2 fixation (%Ndfa: 61·0 ± 4·1 across A and E plots) estimated using the 15N dilution method.
3. Thus, it appears that N inputs to this ecosystem via symbiotic N2 fixation will not be dramatically affected in the foreseeable future even as atmospheric CO2 continues to rise.  相似文献

1. Littoral biota in boreal lakes are known to assimilate epilithon. Being able to predict the stable isotopic composition of these alga will help to identify those systems in which δ13C and δ15N analysis can be used in foodweb investigations of allochthony and biomagnification.
2. In a survey of 15 boreal lakes, the concentration of dissolved organic carbon (DOC) explained 76% of the variation in epilithon δ13C, and 86% of the variation in epilithon δ15N.
3. Because both δ13C and δ15N values were depressed and similar to terrestrial values in humic (high DOC) lakes, it will be more difficult to successfully employ stable isotopic techniques for estimating allochthony in such systems. Lower δ15N values in humic lakes also indicate that trophic positions estimated by stable isotopes are not directly comparable to those of similar biota inhabiting clearwater lakes, unless autochthonous baseline corrections are made.  相似文献

SUMMARY. 1. The effect of ionic strength on the configuration of aquatic humic substances was studied by gel filtration and dialysis of water from small. Finnish forest lakes of varying colour.
2. Sephadex gel filtration of water from the most humic lake gave similar elution profiles of UV absorbance and dissolved organic carbon (DOC). Gel filtration of unconcentrated samples from all three lakes under natural conditions of ionic strength (I = 1.7 × 10−4) and pH (5.5–6.0) gave similar fractionation of humic substances, despite their widely differing colour (30–350 mg Pt l−1) and DOC (5–25mg C l−1).
3. Increasing the ionic strength by two orders of magnitude caused considerable retardation on the Sephadex columns of the humic substances, suggesting a decrease in their molecular size and/or shape.
4. Dialysis experiments strongly indicated that ionic strength-induced changes in the configuration of the aquatic humic substances are indeed real. Hence it is probable that the elution behaviour of aquatic humic substances on Sephadex gels has previously been wrongly attributed to ionic strength-dependent interactions between the gel and the humic substances.  相似文献

Aims:  To evaluate the effectiveness of organic acids and supercritical carbon dioxide (SC-CO2) treatments as well as their combined effect for the reduction of nonpathogenic Escherichia coli and three pathogenic bacteria in fresh pork.
Methods and Results:  The different treatment conditions were as follows: (i) treatment with acetic (1%, 2% or 3%) or lactic acid (1%, 2% or 3%) only, (ii) treatment with SC-CO2 at 12 MPa and 35°C for 30 min only and (iii) treatment with 3% acetic or lactic acid followed by treatment with SC-CO2. Within the same organic acid concentration, the lactic and acetic acid treatments had similar reductions. For the combined treatment of lactic acid and SC-CO2, micro-organism levels were maximally reduced, ranging from 2·10 to 2·60 log CFU cm−2 ( E. coli , 2·58 log CFU cm−2; Listeria monocytogenes , 2·60 log CFU cm−2; Salmonella typhimurium , 2·33 log CFU cm−2; E. coli O157:H7, 2·10 log CFU cm−2).
Conclusions:  The results of this study indicate that the combined treatments of SC-CO2 and organic acids were more effective at destroying foodborne pathogens than the treatments of SC-CO2 or organic acids alone.
Significance and Impact of the Study:  The combination treatment of SC-CO2 and organic acids may be useful in the meat industry to help increase microbial safety.  相似文献

Aims:  Brettanomyces / Dekkera bruxellensis is a particularly troublesome wine spoilage yeast. This work was aimed at characterizing its behaviour in terms of growth and volatile compound production in red wine.
Methods and Results:  Sterile red wines were inoculated with 5 × 103 viable cells ml−1 of three B. bruxellensis strains and growth and volatile phenol production were followed for 1 month by means of plate counts and gas chromatography-mass spectrometry (GC-MS) respectively. Maximum population levels generally attained 106–107 colony forming units (CFU) ml−1 and volatile phenol concentrations ranged from 500 to 4000 μg l−1. Brettanomyces bruxellensis multiplication was also accompanied by the production of organic acids (from C2 to C10), short chain acid ethyl-esters and the 'mousy off-flavour' component 2-acetyl-tetrahydropyridine.
Conclusions:  Different kinds of 'Brett character' characterized by distinct metabolic and sensory profiles can arise in wine depending on the contaminating strain, wine pH and sugar content and the winemaking stage at which contamination occurs.
Significance and Impact of the Study:  We identified new chemical markers that indicate wine defects caused by B. bruxellensis. Further insight was provided into the role of some environmental conditions in promoting wine spoilage.  相似文献

1. Groundwater fluxes of nitrogen and dissolved organic carbon (DOC) were investigated in Grape Vine Canyon Stream in the Mojave Desert focusing on the rate of inputs and the fate of groundwater-derived nutrients in the stream. Discharge rates from different ground waters were measured using an end-member mixing model coupled with injections of a conservative solute tracer into the stream channel.
2. In surface water, nitrate concentration averaged 1.13 mg N L–1 and DOC concentration averaged 1.82 mg C L–1.
3. Groundwater discharge into Grape Vine Canyon Stream was derived from three sources. Nitrate concentration varied among the three groundwater sources with mean concentrations of 0.56, 0.94 and 0.08 mg N L–1. DOC, in contrast, did not vary among ground water sources, with an overall average concentration of 2.96 mg C L–1.
4. In the surface stream, nitrate concentration was two-fold greater than the concentration predicted from groundwater input, indicating that in-stream processes generated nitrate. Stream DOC concentration was lower than predicted based upon groundwater input rate. The production of nitrate and loss of DOC suggest that DOC is lost through mineralisation of dissolved organic matter, possibly resulting in the mineralisation of dissolved organic nitrogen to ammonium and subsequent transformation to nitrate via nitrification. In further support of this hypothesised linkage, DOC loss explained 80–89% of the variance in nitrate production in Grape Vine Canyon Stream.  相似文献

1. We estimated uptake of stream water dissolved organic carbon (DOC) through a whole-stream addition of a 13C-DOC tracer coupled with laboratory measurements of bioavailability of the tracer and stream water DOC.
2. The tracer, a leachate of 13C-labelled tree tissues, was added to the head waters of White Clay Creek, Pennsylvania, U.S.A., over a 2-h period and followed 1.27 km downstream to generate mass transfer coefficients for DOC lability classes within the tracer.
3. From the longitudinal 13C uptake curve, we resolved labile and semi-labile DOC classes within the 13C-DOC tracer comprising 82% and 18% of the tracer respectively.
4. Plug-flow laboratory bioreactors colonized and maintained with stream water were used to determine the concentration of stream water DOC fractions that had a similar lability to the labile and semi-labile classes within the tracer and we assumed that stream water DOC and tracer DOC with comparable lability fractions in the bioreactors behaved similarly in the stream, i.e. they had the same mass transfer coefficients.
5. A small fraction (8.6%) of the stream water DOC was labile, travelling 238 m downstream before being taken up. The remaining bioavailable stream water DOC was semi-labile and transported 4.5 km downstream before being taken up. These uptake lengths suggest that the labile DOC is an energy source within a stream reach, while the semi-labile DOC is exported out of the reach to larger rivers and the downstream estuary, where it may provide energy for marine microbial communities or simply be exported to the oceans.  相似文献

SUMMARY. 1. In laboratory experiments, 9.7–25.7% of dissolved organic carbon (DOC) in groundwater (at concentrations of 18.7–24.8 mg 1-−1) was immobilized after perfusion through 8-cm-deep (22-cm-diameter) cores of stony stream-bed substratum.
2. This represented immobilization rates of 7.1–23.5 mg m−2 h−1 or, extrapolated across the year, potential immobilization rates within the stream bed of 62.2–205.9g m−2 yr−1. Actual rates in the entire stream bed were probably higher because perfusion through the experimental cores did not reduce groundwater DOC concentrations to levels measured in the adjacent stream.
3. Natural concentrations of dissolved free amino acids (DFAAs) in groundwater were generally unchanged following perfusion through the cores, suggesting the maintenance of a dynamic equilibrium in their concentrations.
4. Selective enrichments of amino acids in groundwater (up to 20-fold) were entirely immobilized following perfusion, indicating their rapid retention and flux in this environment. Thus, immobilization of the bulk DOC in stream-bed cores probably did not reflect net reductions in dissolved free, low-molecular-weight material, with higher molecular weight, more 'refractory' material being immobilized instead.
5. We conclude that groundwater can contribute substantial amounts of DOC, both high and low molecular weight, to a stream ecosystem. The stream bed is the site at which much of this material could be initially immobilized and made available to the stream trophic structure.  相似文献

Aims:  To isolate and identify a benefic bacterium, Bacillus subtilis E20, from natto (fermented soybeans), and incorporate it into shrimp feed to promote shrimp growth performance.
Methods and Results:  A protease-producing bacterium, E20, isolated from natto was identified as B. subtilis by an API 50 CHB kit and the 16S rDNA sequence. B. subtilis E20 was able to grow at a broad range of temperatures (10–50°C), pH values (5–10), and NaCl levels (0–9%). The best culture conditions for B. subtilis E20 to produce the protease were 40°C, a pH of 6–8 and 0% NaCl. No shrimp died after being injected with B. subtilis E20 [up to 109 colony-forming units (CFU) per shrimp]. Bacillus subtilis E20 was incorporated in diets at the levels of 0 (control), 106, 107, and 108 CFU kg−1 for shrimp grow-out culture, and results showed that after feeding on B. subtilis E20-containing diets (108 CFU kg−1 of diet), shrimp had excellent growth performance and production compared to the control because protease activities in the digestive tract were improved by B. subtilis E20.
Conclusions:  Bacillus subtilis E20 isolated from natto is a great protease producer and is able to improve shrimp growth performance through increasing the digestibility of food.
Significance and Impact of the Study:  Results suggest that B. subtilis E20 is a potential candidate for use as a probiotic to improve shrimp growth performance, and consequently reduce feed costs.  相似文献

Eragrostis pilosa (Linn.) P Beauv., a C4 grass native to east Africa, was grown at both ambient (350 μmol mol−1 and elevated (700 μmol mol−1) CO2 in either the presence or absence of the obligate, root hemi-parasite Striga hermonthica (Del.) Benth. Biomass of infected grasses was only 50% that of uninfected grasses at both CO2 concentrations, with stems and reproductive tissues of infected plants being most severely affected. By contrast, CO2 concentration had no effect on growth of E. pilosa , although rates of photosynthesis were enhanced by 30–40% at elevated CO2. Infection with S. hermonthica did not affect either rates of photosynthesis or leaf areas of E. pilosa , but did bring about an increase in root:shoot ratio, leaf nitrogen and phosphorus concentration and a decline in leaf starch concentration at both ambient and elevated CO2. Striga hermonthica had higher rates of photosynthesis and shoot concentrations of soluble sugars at elevated CO2, but there was no difference in biomass relative to ambient grown plants. Both infection and growth at elevated CO2 resulted in an increase in the Δ13C value of leaf tissue of E. pilosa , with the CO2 effect being greater. The proportion of host-derived carbon in parasite tissue, as determined from δ13C values, was 27% and 39% in ambient and elevated CO2 grown plants, respectively. In conclusion, infection with S. hermonthica limited growth of E. pilosa , and this limitation was not removed or alleviated by growing the association at elevated CO2.  相似文献

Abstract: A continuous dual 13CO2 and 15NH415NO3 labelling experiment was undertaken to determine the effects of ambient (350μmol mol-1) or elevated (700μmol mol-1) atmospheric CO2 concentrations on C and N uptake and allocation within 3-year-old beech ( Fagus sylvatica L.) during leafing. After six weeks of growth, total carbon uptake was increased by 63 % (calculated on total C content) under elevated CO2 but the carbon partitioning was not altered. 56 % of the new carbon was found in the leaves. On a dry weight basis was the content of structural biomass in leaves 10 % lower and the lignin content remained unaffected under elevated as compared to ambient [CO2]. Under ambient [CO2] 37 %, and under elevated [CO2] 51 %, of the lignin C of the leaves derived from new assimilates. For both treatments, internal N pools provided more than 90 % of the nitrogen used for leaf-growth and the partitioning of nitrogen was not altered under elevated [CO2]. The C/N ratio was unaffected by elevated [CO2] at the whole plant level, but the C/N ratio of the new C and N uptake was increased by 32 % under elevated [CO2].  相似文献

1. Inorganic carbon availability influences species composition of phytoplankton in acidic and highly alkaline lakes, whereas the overall influence on community photosynthesis and growth is subject to debate.
2. The influence of total dissolved inorganic carbon (DIC) and free CO2 on community photosynthesis was studied in six Danish lakes during the summer of 1995. The lakes were selected to ensure a wide range of chlorophyll a concentrations (1–120 μg l–1), pH (5.6–9.6) and DIC concentration (0.02–2.5 m m ). Photosynthesis experiments were performed using the 14C technique in CO2-manipulated water samples, either by changing the pH or by adding/removing CO2.
3. Lake waters were naturally CO2 supersaturated during most of the experimental period and inorganic carbon limitation of photosynthetic rates did not occur under ambient conditions. However, photosynthesis by phytoplankton in lakes with low and intermediate DIC concentrations was seriously restricted when CO2 concentrations declined. Similarly, photosynthesis was limited by low CO2 concentrations during phytoplankton blooms in the hardwater alkaline lakes.  相似文献
