Summary A simple viscoelastic film model is presented, which predicts a breakdown electric potential having a dependence on the electric pulse length which approximates the available experimental data for the electric breakdown of lipid bilayers and cell membranes (summarized in the reviews of U. Zimmermann and J. Vienken, 1982,J. Membrane Biol. 67:165 and U. Zimmermann, 1982,Biochim. Biophys. Acta 694:227). The basic result is a formula for the time of membrane breakdown (up to the formation of pores): =(/C)/( m 2 0 2 U 4/24Gh 3+T 2/Gh–1), where is a proportionality coefficient approximately equal to ln(h/20),h being the membrane thickness and 0 the amplitude of the initial membrane surface shape fluctuation ( is usually of the order of unity), represents the membrane shear viscosity,G the membranes shear elasticity modules, m the membrane relative permittivity, 0=8.85×10–12 Fm,U the electric potential across the membrane, the membrane surface tension andT the membrane tension. This formula predicts a critical potentialU c ;U c =(24Gh 3/ m 2 0 2 )1/4 (for = andT=0). It is proposed that the time course of the electric field-induced membrane breakdown can be divided into three stages: (i) growth of the membrane surface fluctuations, (ii) molecular rearrangements leading to membrane discontinuities, and (iii) expansion of the pores, resulting in the mechanical breakdown of the membrane.  相似文献

Summary Nitrogen and phosphorus limitations to growth are common in many loblolly pine (Pinus taeda) stands. Interactions of these nutrients may complicate interpretation of foliar nutrient analysis for predicting response to forest fertilization. Proportions of foliar nutrient concentrations (and the changes in these proportions following fertilization) were examined in 36 semi-mature loblolly pine plantations in the southeastern United States. Mean proportions of nutrient concentrations (NPKCaMg) for non-fertilized stands were 1009.336.517.29.2. Potassium and phosphorus were higher. Nitrogen fertilization generally decreased the PN ratio and enhanced growth, indicating a nitrogen deficiency in most stands under study. Additions of nitrogen and phosphorus together yielded a significant increase in the PN ratio. Several factors that affect the frequency of organogenesis in apple leaf explants were examined for the scion cultivars Empire, Freedom, Golden Delicious, Liberty, McIntosh, and Mutsu and for the rootstocks Malling 7A and Malling 26. The main factors affecting morphogenesis were BA concentration, basal medium, leaf explant origin and maturity, explant orientation, and photosynthetic photon flux. Depending on the genotype, optimal regeneration was obtained using either 22.2 or 31.1 M BA and the N6 basal medium, with the exception of Golden Delicious which regenerated better on MS medium. After 6 weeks, the average number of shoots per segment varied from 5 to 16, and the percentage of regeneration between 70 and 100%, depending on the genotype tested and the maturity of the explant. Regeneration capacity increased dramatically from the tip towards the base of the leaf, and was higher from the middle to the proximal end.Cefotaxime and carbenicillin, two antibiotics commonly used during transformation studies to eliminate Agrobacterium tumefaciens from plant tissue, were tested to determine their effect on morphogenesis. Cefotaxime at a dose of 250 mg 1-1 enhanced regeneration and shoot development, whereas carbenicillin at a dose of 500 mg l-1 induced abundant callus formation and inhibited regeneration. Kanamycin, a widely used selection agent for plant transformation, strongly inhibited regeneration even at very low doses. Anthranilate synthase (AS) is a key enzyme in the biosynthesis of various indole compounds including tryptophan. AS consists of two subunits, and , and converts chorismate to anthranilate. Two or more AS -subunit genes have been identified and characterized in several land plants. Although subunits of AS induced by elicitation have been suggested to play significant roles in secondary metabolism, the biochemical and precise functional properties of individual AS isozymes have remained unclear. We have previously identified and characterized two AS -subunit genes (OASA1 and OASA2) in rice (Oryza sativa). To provide further insight into the enzymatic functions of AS isozymes in rice, we have now isolated rice cDNAs encoding the AS subunits OASB1 and OASB2 and reconstituted AS isozymes in vitro with the wheat germ cell-free system for protein expression. Both OASB subunits conferred glutamine-dependent AS activity on either OASA1 or OASA2, indicating the absence of a marked functional difference between the two subunits in terms of amidotransferase activity. Furthermore, both OASA subunits required assembly with a subunit to achieve maximal enzymatic activity even with NH 4 + as the amino donor. The V max and K i for tryptophan of the OASA1-OASB1 isozyme with glutamine as the amino donor, however, were 2.4 and 7.5 times, respectively, those of OASA2-OASB1, suggesting that AS isozymes containing OASA1 possess a higher activity and are less sensitive to feedback inhibition than those containing OASA2. Our biochemical characterization of reconstituted AS isozymes has thus revealed distinct functional properties of these isozymes in rice.  相似文献

Adoptive immunotherapy using MHC-nonrestricted-lymphocytes, peripheral blood T cells and NK cells was devised. Peripheral blood mononuclear cells (3 x 107) were selected by immobilization to anti-CD3 monoclonal antibody for 4 days and cultured for 2 weeks in the presence of IL-2. Thereafter they were reactivated by 500 U/ml of IFN- and 1000 U/ml of IL-2 for 1 hour. Enhancement of NK and LAK activities was confirmed. Peripheral blood T cells proliferated in response to immobilized anti-CD3 antibody (3% to 30%). Approximately 6 x 109 BRM-activated killer (BAK) cells composed of CD56+ T cells and CD56+ NK cells, were dispensed to cancer patients via intravenous drip infusion. Nine patients were treated with BAK cells every 2 weeks or every month on an outpatient basis. During the course of adoptive immunotherapy, the crossed affinity immunoelectrophoresis (CAIE) pattern of serum immunosuppressive acidic protein (IAP) was analysed. Both the production and glycosylation pattern of IAP is changed in response to tumor enlargement and may therefore act as a marker of the disease progression. During the course of BAK therapy, the glycosylation IAP pattern of 6 patients changed from tumor (T) to normal (N). In addition, the performance status of all patients was maintained at 90–100% of the Karnofsky scale and any side effects including fever were not observed during treatments with BAK cells. Moreover, the overall quality of life (QOL) of the patients, scored at the Face scale was favorable. In addition, blood levels of activated T cells producing IFN- were assayed as an indication marker of BAK therapy. The normal range of IFN- producing T cells comprised 6.9 ± 0.9% of peripheral blood mononuclear cells (PBMC), according to a single cell FACScan analyses of PBMCs derived from normal individuals. IFN- producing T cells of Patients No. 8 and 9, who received extensive chemotherapy before initiation of BAK therapy, comprised only 0.2% and 2% of PBMC, respectively. These patients died 3 and 6 months after beginning BAK therapy. Peripheral blood T cells of Patients Nos. 1–7 proliferated in response to immobilized anti-CD3 antibody and the frequency of IFN- producing T cells in PBMC preparation of these patients were over 3% before initiation of BAK therapy. Since our data show a positive correlation between survival time and initial T cell counts, a low frequency of these cells may contraindicate BAK therapy.  相似文献

Cross-correlated relaxation rates involving the C-H dipolar interaction and the carbonyl (C) chemical shift anisotropy (CSA) have been measured using two complementary 3D experiments. We show that the protein backbone angle can be directly refined against such cross-correlated relaxation rates (H C,C) and the three-bond H/D isotope effect on the C chemical shifts (3C (ND)). By simultaneously using both experimental parameters as restraints during NMR structure calculations, a unique value for the backbone angle is defined. We have applied the new refinement method to the -Spectrin SH3 domain (a -sheet protein) and to the Sgs1p HRDC domain (an -helical protein) and show that the quality of the NMR structures is substantially improved, judging from the atomic coordinate precision and the Ramachandran map. In addition, the -refined NMR structures of the SH3 domain deviate less from the 1.8 Å crystal structure, suggesting an improved accuracy. The proposed refinement method can be used to significantly improve the quality of NMR structures and will be applicable to larger proteins.  相似文献

Résumé Nous avons fait élever des larves d'Anergates atratulus par des ouvrières deMyrmica laevinodis à 22°C. Pour y parvenir, il n'est pas utile de faire hivernerensemble les larves d'Anergates et les ouvrières deMyrmica. La présence de larves autochtones n'empêche pas lesMyrmica d'élever des larves d'Anergates. Dans toutes les expériences lesMyrmica ont été soumises au fridavant de recevoir des larves d'Anergates. Aucune reine deMyrmica n'a été utilisée dans ces expériences.Sur les 64 larves d'Anergates que nous avons utilisées, 38 se sont transformées en imagos. C'est au début de l'adoption et au moment des métamorphoses que périrent la plupart des 26Anergates perdus. Les femelles vécurent en général 2 ou 3 jours et cherchèrent très tôt à quitter le nid natal. Les mâles vécurent 2 à 3 semaines.
Summary Larvae ofAnergates atratulus were experimentally reared by workers ofMyrmica laevinodis, at 22°C. An overwintering of both larvae ofAnergates and workers ofMyrmica is not necessary for the success of that experiment. The presence of larvae ofMyrmica does not keep theMyrmica from rearing larvae ofAnergates. The workers ofMyrmica have been cooled, in all the experiments, before receiving larvae ofAnergates. No queen ofMyrmica have been used in that experiments.38 of the 64 larvae ofAnergates used became imagos. Most of the 26 lostAnergates died at the beginning of the adoption and during the metamorphosis. The females lived generally 2 or 3 days and tried, very early, to leave their native nest. The males lived 2 or 3 weeks.

Summary Effects of increasing rates of lime and phosphorus addition on concentrations of available nutrients in soil and on P, Al and Mn uptake by two pasture legumes, lotus (Lotus pedunculatus Cav.) and white clover (Trifolium repens L.), were studied in a pot experiment using a highly leached acid (pH 4.2) soil.Liming resulted in an increase in exchangeable Ca and thus in percentage base saturation, with concomitant decreases in levels of exchangeable Al, Fe and Mn. The relationship between exchangeable Ca and Al was linear and negative with a gradient of almost unity. Liming had no consistent effect on measured CEC values. Increasing lime rates significantly reduced concentrations of Mg, K and Na in saturation paste extracts but had no effect on exchangeable Mg, K and Na levels.With increasing lime additions, available phosphate indicesviz water soluble, resin-, Morgan-and Williams-extractable all decreased significantly, Truog-extractable was unaffected, while Brayextractable generally increased. Fractionation revealed that lime additions caused a decrease in easily soluble, Fe-bound and to a lesser extent Ca-bound phosphate fractions, had no effect on reductant soluble phosphate, but resulted in an increase in the Al-bound fraction. P uptake and yield of both legumes increased with lime and P additions.Correlations between available phosphate indices and yield of both legumes were weak or nonsignificant. However, high, significant positive correlation coefficients were found between available phosphate and plant uptake of P. Indices of available Al and Mn were not generally significantly correlated with plant uptake of Al or Mn but significant negative correlations were found between available Al and Mn and yield of both species.  相似文献

Summary The enzymatic synthesis of a peptide compound was carried out successfully in homogeneous organic solvent.Solid Thermolysin was found to catalyze the synthetic reaction of N-benzyloxycarbonyl-L-aspartyl-L-phenylalanine methyl ester (Z-APM; a precursor of sweetner Aspartame) from N-benzyloxycarbonyl-L-aspartic acid (Z-L-Asp) and L-phenylalanine methyl ester (L-PheOMe) in a 98 percent organic medium (ethylacetatebenzenemethanolwater=5029192). The dissolution of enzyme was not observed. The optimal pH shifted to acidic side by 1.0 pH unit, compared with that in aqueous medium. The enzymatic activity of solid thermolysin with an average size of 3.4×9.5 m was determined to be 0.18 moles-product/(mg-solid)·h under the initial concentrations of L-PheOMe of 0.1M and Z-L-Asp of 0.05M, and at pH 6.0 and 40°C.  相似文献

Quantification of chlorophyll (Chl) fluorescence is a versatile tool for analysing the photosynthetic performance of plants in a non-intrusive manner. A pulse-amplitude modulated fluorometer was combined with a CNC router for the automated measurement of the effective quantum yield of photosystem 2 (2) of Arabidopsis thaliana plants. About 90 000 individual plants representing 7 500 lines derived from En-transposon and T-DNA mutagenised Arabidopsis populations were screened for mutants with altered 2. Forty-eight recessive 2 mutations were identified of which most exhibit also altered pigmentation and increased photosensitivity. For three 2 mutants the corresponding mutated genes were identified that code all for chloroplast-located proteins. Comparison of the 2 mutant screen with other screening methods based on the measurement of Chl fluorescence shows that the 2 mutants identified are different to mutants identified by high Chl fluorescence. Some 2 mutants, on the contrary, are common to mutants identified by screens based on non-photochemical quenching.  相似文献

The behavior of the basic nutrients (NO3, PO4, SiO2) was studied in the estuary of the Razdolnaya River in low and high water, the flow was 4.3 × 106 m3/day and 10.8 × 106 m3/day, respectively. It was shown that within the limits of the euphotic zone the nutrients were characterized by a pronounced nonconservative behavior caused by their removal by phytoplankton in primary production. It was determined that phytoplankton removal of nutrients occurred with ratios C : NO3 : P : Si = 105 : 18 : 1 : 37 and C : NO3 : P : Si = 93 : 11 : 1 : 29 at a respective ratio P : NO3 : Si = 1 : 22 : 140 in low water and P : NO3 : Si = 1 : 17 : 120 in high water. It was also determined that the maximum rate of nutrient removal was 4 times higher in the high water than in the low water. The maximum value of primary production of phytoplankton was 2.5–4.0 gC/m2 day. The estuary area of the Razdolnaya River was specified by rather high production. Summary The inheritance of resistance to whitebacked planthopper Sogatella furcifera (Horvath) was studied in 21 rice varieties. Reactions of F1; F2 and F3 progenies of the crosses of 21 resistant varieties with the susceptible variety TN 1 revealed that a single dominant gene governs resistance in Mushkan 41, Santhi, Siahnakidar 195, SM2-34, Tirisurkh 251, Zirijowaian 245, 18, 24A, 39, 76 S, 78, 180, 213 B, 267, 293, CI 6037-4, NP97, S39 JKW and Bansphul. In varieties 65 and 274 A, resistance is governed by one dominant and one recessive gene which segregate independently of each other. Tests for allelism with the Wbph 1 gene originally identified in N 22 revealed that the dominant gene present in all the test varieties is the same as Wbph 1. Further studies are required to determine the allelic relationships of the recessive gene found in varieties 65 and 274 A.  相似文献

Summary Over sixty EMS induced mutations affecting gene lamB, presumably the structural gene for the receptor in Escherichia coli K12, were examined for growth of host range mutants and effect of nonsense suppressors. By the first criterion the mutations could be grouped in three classes. Bacteria with class I mutations allow growth of mutants with extended host range (noted h) of the type already described (Appleyard, Mac Gregor and Baird, 1956). Bacteria with class II mutations allow growth of h mutants with still more extended host range (noted hh *). No host range mutants of could be found which would grow on bacteria with class III mutations. Using nonsense suppressors it was found that class I and II consist of missense mutations, while class III consists of nonsense mutations. Exceptions are likely to exist (especially in class III) but were not found among the mutations tested. These observations are briefly discussed in terms of outer membrane protein integration and of phage receptor interaction.  相似文献

The frequency of two common disease-associated mutations in the arylsulphatase A (ASA) gene, and of a mutation causing ASA pseudodeficiency, have been established in metachromatic leukodystrophy patients diagnosed in our laboratory. A total of 37 mutant genes have been analysed. The GA change destroying the splice donor site of exon 2 is generally associated with more severe disease and was found in 43.2% of mutant ASA genes. The CT transition causing a proline to leucine substitution at position 426 in exon 8 (P426L) is associated with later onset disease, and was found in 16.2% of mutant genes. The AG transition leading to loss of a polyadenylation signal associated with ASA pseudodeficiency was present at a frequency of 7.5% in the patients and heterozygotes studied.  相似文献

Inhibition of spore germination of selected isolates ofCochliobolus carbonum was studied with increasing concentrations of thiram. Isolate differences were detected for the TMTD polymodal curve. This difference was shown to be heritable and independent of tolerance to thiram as indicated by vegetative growth. Life on Earth is essentially nucleic acids (NAs) influencing peptide synthesis such that NA replication is favored. It is proposed that the ability to synthesize polypeptides evolved gradually — one peptide bond at a time. The proposed evolution of the peptide synthesis apparatus begins with a transfer NA (tNA) which catalyzes the transfer of activated amino acids to accessible amino groups in its environment. The resulting capped molecules (with single amino acid caps) in turn favor NA replication. The proposed evolution of the peptide synthesis apparatus from the tNA onward is characterized by a progressive increase in the number of amino acids per cap: two tNAs jointly produce a dipeptide cap, three tNAs jointly produce a tripeptide cap, etc. Messenger NAs evolve because they can specify the composition and sequence order of the peptide caps. Lastly, ribosomal NAs evolve. The origin, expansion, and standardization of the genetic code are discussed. It is proposed that the present triplet code evolved by a process of codon length refinement, and that originally codons of varying lengths were allowable, as were unassigned bases between codons. An environmental supply of activated compounds for early evolving entities is proposed. An environmental NA replication process via single template-directed bond formation events is proposed. An environmental retention and redistribution process is proposed to have acted as a functional substitute for the cell wall and cell division of early evolving entities.  相似文献

Experiments were conducted to study the distribution of three selectedpolychlorinated biphenyl (PCB) congeners within the microbial food web attwo different nutrient levels; control and nutrient enriched. The objectivewas to quantify the uptake of PCBs through grazing by protozoa. The14C-PCBs tested were 4-chlorobiphenyl (IUPAC # 3),2,2,5,5-tetrachlorobiphenyl (IUPAC # 52), and2,2,4,4,5,5-hexachlorobiphenyl (IUPAC # 153). EachPCB was incubated in triplicate seawater samples at 20 idref;Cover one week. Daily, samples were separated into four fractions; <0.2µm (dissolved), 0.2-2 µm (bacteria), 2-10 µm(flagellate), and > 10 µm (microplankton; phytoplankton andprotozoa) by selective filtration. Of the PCB fraction that initiallyadsorbed to particles, 60–100% was associated to the bacterialfraction and 0–5% to the microplankton fraction. The totaluptake was highest in the nutrient enriched samples, but when normalized tothe carbon biomass the concentration was lower or equal to the control inall particle fractions. The recovery of the PCBs in the particulatefractions depended on the degree of chlorination, as the highest values wereobserved for the 2,2,4,4,5,5-hexachlorobiphenyl and thelowest for the 4-chlorobiphenyl. The concentrations in the bacterial andflagellate fractions decreased over the first 48–96 hours whilst theconcentration increased in the highest trophic level (>10 µmfraction). Approximately 75% of the increase in concentration of the2,2,4,4,5,5-hexachlorobiphenyl in the > 10 µmfraction was estimated to be the result of bacterivory. Our results indicatethe microbial food web can contribute to a rapid uptake of higherchlorinated PCBs, particularly in oligotrophic ecosystems where thebacterial biomass dominates.  相似文献

In vitro culture of pineapple (Ananas comosus) was studied to determine the efficiency of axillary bud culture for rapid propagation of several cultivars. The technique used maximizes the success rate of various steps in the production of pineapple plants. Rapid mass multiplication of plantlets started 9 months after explanting with a significant log phase. The number of plantlets obtained from the culture of a single bud by the thirteenth month ranged from 210 to 380 for Perolera; 300 to 350 for PR-1-67; and 40 to 85 for Smooth Cayenne. The method permits culture of a range of pineapple cultivars. Little morphological variation was observed in young regenerated plants.  相似文献

Summary We have mapped and sequenced the globin gene and seven surrounding Alu repeat sequences in the orangutan globin gene cluster and have compared these and other orangutan sequences to orthologously related human sequences. Noncoding flanking and intron sequences, synonymous sites of , , and globin coding regions, and Alu sequences in human and orangutan diverge by 3.2%, 2.7%, and 3.7%, respectively. These values compare to 3.6% from DNA hybridizations and 3.4% from the globin gene region. If as suggested by fossil evidence and molecular clock calculations, human and orangutan lineages diverged about 10–15 MYA, the rate of noncoding DNA evolution in the two species is 1.0–1.5×10–9 substitutions per site per year. We found no evidence for either the addition or deletion of Alu sequences from the globin gene cluster nor is there any evidence for recent concerted evolution among the Alu sequences examined. Both phylogenetic and phenetic distance analyses suggest that Alu sequences within the and globin gene clusters arose close to the time of simian and prosimian primate divergence (about 50–60 MYA). Synopsis Lead capture techniques have been investigated for demonstrating phosphatase activity, with the aid of the electron microscope, in subcellular fractions isolated from rat liver. Controls with no exposure to the enzyme substrate showed little staining (lead phosphate deposits). Results with preparations where the enzyme incubation period had been prolonged, and the lead phosphate deposits were heavy (over staining), were also reassuring: the staining did not tend to become generalized. Particularly satisfactory results were obtained when the tissue fractions were used in the form of unfixed suspensions for incubation with the substrate, this being performed with the minimum temperature and time needed to obtain representative staining of the elements present. Thus, for 5-nucleotidase in plasma-membrane fragments there is good staining after only 10 min at 2°C. The favoured concentration of lead ions is 2mm. When the reaction period has elapsed, glutaraldehyde is added, and the tissue material repelleted; only then should osmium tetroxide be added. Glucose-6-phosphatase staining, particularly in rough microsomes, showed a time-lag which could be abolished by prior sonication or treatment with deoxycholate. In general, the cytochemical findings tallied well with the biochemical characterization of the various fractions examined, including zonalrotor fractions containing smooth vesicles the origin of which could not be established merely by morphology.A.
