Membrane-bound Mg++-activated ATPase activity in epidermal stripsfrom tobacco leaves (Nicotiana tabacum L. Samsun NN) was stimulatedby abscisic acid (ABA) when the strips were floated on ABA solutionin light or in darkness. The optimum ABA concentrations in lightand in darkness were 10–5 M and 10–6 M, respectively.Carbonyl cyanide m-chlorophenylhydrazone (CCCP) and N, N'-dicyclohexylcarbodiimide(DCCD) completely blocked the basal level membrane-bound epidermalATPase activity. ABAinduced membrane-bound epidermal ATPaseactivity was completely inhibited by CCCP, but only partly byDCCD. H+-influx into epidermal strips on a solution in light was lowerthan that in darkness. ABA stimulated H+-influx into epidermalstrips in light and in darkness. CCCP suppressed basal levelH+-influx, whereas DCCD did not. CCCP also suppressed ABA-inducedH+-influx, whereas DCCD did not. Inoue, H. and Katoh, Y. 1987. Calcium inhibitsion-stimulatedstomatal opening in epidermal strips of Commelina communis L.—J.exp. Bot. 38: 142–149. Ca2+ suppressed both the ion-stimulated stomatal opening andH+ extrusion of pre-illuminated epidermal strips isolated fromCommelina communis L. In the absence of Ca2+, the rate of H+release was 18 nmol H+ cm–2 h–1 per epidermal stripunit area in 150 mol m–3 KCL at pH 7?4. Half-maximum inhibitionof stomatal opening was observed with 220 mmol m–3 ofCa2+. The hexavalent dye, ruthenium red, showed concentration-dependentprevention of the inhibition by Ca2+ of the ion-stimulated stomatalopening. The effect of ruthenium red was non-competitive, andthe K1 for the calcium inhibition was found to be 3?6 mmol m–3.The calcium inhibition of H+ extrusion was also prevented byruthenium red. These results suggest that Ca2+ inhibits theactivity of electrogenic H+ translocating ATPase of the guardcell plasma membrane and leads to the suppression of stomatalopening. Cell membrane potentials have been measured both in epidermalstrips and intact leaf sections of Tradescantia virginiana andCommelina communis, and in epidermal cells over green and overalbino mesophyll cells of T. albiflora var. albovittata. Membranepotentials (cell) in strips were considerably lower than thosein intact sections and were insensitive to light and to theabsence or presence of calcium. Their response to external cationlevels was indifferent to ionic species. However, in intactleaf sections incubated with calcium present, membrane potentialsresponded to K+ levels but not to Na+. were more negative thancells in epidermal strips, and responded to changes in illumination. Long-term recordings of cell and vacuolar K+ levels in T. virginianaduring stomatal closure suggest that the fluctuations of cellwere unrelated to K+ movement (which we could not detect) andthus probably to stomatal movement as well. Turgor pressures measured in epidermal cells of intact leafsections of T. virginiana were found to be of the same magnitudeas those previously reported for epidermal strips. It is concludedthat epidermal cells maintain their solute contents during strippingwithout the involvement of an electrophysiological transportsystem. With the possible exception of lateral subsidiary cells,there was no evidence suggesting that ordinary epidermal cellsare capable of osmotic adjustment even when additional KCI wassupplied in the osmoticum. Light stimulates leaf expansion in dicotyledons by increasingapoplastic acidification, cell wall loosening and solute accumulationfor turgor maintenance. Red and blue light enhance growth viadifferent photo-systems, but the cellular location and modesof action of these systems is not known. Here, the effect of red and blue light was studied on transportprocesses in epidermal cells of expanding leaves of the Argenteummutant of Pisum satlvum. Both red and blue light caused extraceiiuiaracidification by isolated epidermal tissue, which was stimulatedby extracellular K+ and inhibited by DCCD at 0.1 mol m–3.Acidification induced by red compared with blue light showeddifferent saturating kinetics in fluence rate-response curves.Under near saturating light conditions the effects of red andblue light were additive. The red light-induced acidificationwas inhibited by far-red light while the blue light-inducedacidification was not. Light caused a hyperpoianzation of themembrane potential in epidermal strips, and stimulated 86Rb+uptake by epidermal protoplasts. These results show that phytochromeand an additional blue light-photoreceptor function in isolatedepidermal cells to promote proton efflux, hyperpolarization,and cation uptake. Weyers, J. D. B. and Paterson, N. W. 1987. Responses of Commelinacommunis stomata in vitro.— J. exp. Bot. 38: 631–641. Analysis of the kinetics of movements of Commelina communisL. stomata in vitro revealed a sequence of opening and closingphases dependent on the incubation medium used and the physiologicalstate of the plant material. In buffer containing 50 mol m –3KC1 the sequence of aperture changes appeared to be influencedby equilibration of cell water potentials with that of the mediumand by solute fluxes (dependent and independent on metabolicactivity). The results indicate that the stomatal aperture afterseveral hours of incubation may not always provide a reliablequantitative estimate of the ability of the stomata to operate.As a consequence, modifications are suggested to the ways inwhich experiments using epidermal strips are carried out andreported. Auxin-induced and 1-aminocyclopropane-1-carboxylic acid (ACC)-dependentethylene production in mung bean (Vigna radiata [L] Wilczek)hypocotyl sections, from which epidermis had been removed, wasinvestigated. Ethylene production in hypocotyl sections withoutepidermis was induced by treatment with IAA, and also occurredfrom exogenously supplied ACC in the presence of 0.2 M mannitol.Isolated epidermal strips alone failed to produce substantialamounts of ethylene in response to IAA or from exogenous ACC.3,4-[14C]-Methionone was incorporated into both ACC and ethylenein peeled sections treated with IAA, but not in the isolatedepidermal strips. Radioactive ACC, however, was detected inthe epidermal strips separated from the unpeeled sections previouslyfed with 3,4-[14C]-methionine in the presence of IAA. We concludethat the Site of auxin-induced ethylene production is not inthe epidermis, but in other hypocotyl cells, and that epidermalcells lack the activity which converts ACC to ethylene. Cell-wall synthesis in guard cells of Vicia faba L. was examinedusing sonicated epidermal strips incubated with [14C]glucose.The cell walls of the guard cells incorporated [14C]glucoseat a lower level in the dark than in the light. Stomatal aperturein the epidermal strips was reduced by application of 1 µmabscisic acid (ABA) in the light but not in the dark. The ABAtreatment reduced the incorporation of [14C]glucose into thecell walls especially in the light. Fractionation of the labeledcell-wall components revealed that ABA inhibited the synthesisof pectic substances and cellulose, but did not affect hemicellulosesynthesis. Microautoradiographs of the cell-wall fraction ofthe epidermal strips showed that a large amount of radioactivitywas distributed at both ends of the guard cells in the absenceof ABA and that removal of pectic substances from the cell-wallfraction resulted in uniform distribution of the radioactivityin the cell walls of the guard cells. These results indicatedthat the synthesis of pectic substances was active at both endsof the guard cells and was inhibited by ABA. Measurement ofspecific activities of neutral sugars in the guard-cell wallsshowed that polymers composed of galactose underwent activeturnover and that synthesis of glucans was inhibited by ABA.These results revealed a strong correlation between the stomatalmovement and the synthesis of pectic substances and cellulosein the guard cells, suggesting that the cell-wall metabolismin the guard cells may play a role in the regulation of stomatalmovement. Tentoxin and, to a lesser extent, dihydrotentoxin (both at 10mmol m–3) reduce stomatal opening in epidermal stripsof Commelina communis in the light but not in darkness. Thiseffect was significantly greater in normal air than in CO2-freeair. Fusicoccin overcame the tentoxin effect. However, tentoxindid not inhibit stomatal opening in the light in epidermal stripsof Paphiopedilum harrisianum, a species which lacks guard cellchloroplasts. It is concluded that tentoxin exerts its actionon stomata not by an ionophorous effect in the plasmalemma ofguard cells but by the inhibition of photophosphorylation intheir chloroplasts. The effects of DCMU and tentoxin on guardcells are discussed in terms of their effects on chloroplastsand the extent to which energy is supplied from this organelleduring stomatal opening in the light. The results indicate thatneither photophosphorylation nor non-cyclic electron transportin guard cell chloroplasts are essential for stomatal opening. Measurements were made of changes in stomatal pore widths inepidermal strips of leaves ofVicia faba and Commelina communis.Strips were incubated in dilute KCI solutions (1 and 10 molm–3) flowing through a perfusion chamber on the stageof a microscope and kept for 4 d in continuous light. Circadianrhythms of stomatal apertures were detected in both species.Although the amplitude was small it was statistically significant.It is concluded that at least partof the mechanism for the stomatalrhythm resides in the epidermis, probably in the guard cells. The effects of abscisic acid (ABA) on the size of the apertureof stomata on epidermal strips of Vicia faba were studied inincubation media with different pH values. The osmotic potentialof guard cells, as determined by the limiting plasmolysis method,was higher at pH 4.0 than at pH 6.0, although the size of thestomatal apertures was almost identical at both pH values. AtpH 4.0, ABA effectively caused stomatal closure but had onlya small effect on the osmotic potential, whereas, at pH 6.0,ABA significantly increased the osmotic potential. ABA promotedthe efflux of Cl and malate from epidermal strips intothe incubation medium, an effect which was more marked at pH6.0, with a concomitant efflux of K+ to balance the charge onthe exported anions. Membrane-bound Mg++-activated ATPase was separated from thelower epidermis of tobacco leaves (Nicotiand tabacum L. SamsunNN) on stepwise sucrose density gradient centrifugation. Membrane-bound epidermal ATPase was localized in the interfaceof densities in sucrose of 1.12 to 1.16 in the sedimentary fractionbetween 1,500?g to 10,000?g from the homogenate of the lowerepidermis. The epidermal ATPase activity was activated by divalentcations (Mg++>Mn++Co++>Fe++>Zn++>Ca++) and furtherstimulated by KCl by ca. 20%. The pH optimum for Mg++-activationof the epidermal ATPase was ca. 6.0. The enzyme hydrolyzed ATPmore rapidly than other nucleoside triphosphates. The optimumtemperature for activation of the epidermal ATPase activitywas ca. 40?C. 50% of the epidermal ATPase activity was lostin 18 min at 55?C and in 2.5 days at 2.5?C. The apparent Kmvalue of the epidermal ATPase was 4.7?10–4 M and Vmaxwas 65.4 nmoles Pi/mg protein/min. The epidermal ATPase wasstrongly inhibited by N, N'-dicyclohexylcarbodiimide (DCCD)in vitro whereas oligomycin, carbonyl cyanide m-chlorophenylhydrazone(CCGP), indoleacetic acid (IAA) and abscisic acid (ABA) wereinsensitive to the epidermal ATPase activity. The effects of H$ and fusicoccin (FC) on stomatal opening inthe dark were investigated using epidermal strips of Commelinacommunis and Vicia faba cv. Ryosai Issun. Citrate-phosphatebuffer induced maximal opening of stomata at pH 3.0 when testedover the range of 2.7 to 5.0. HCl at 1 mM also induced stomatalopening without appreciable accumulation of K$ in the guardcells. After 4 hr treatment with 10 µM FC, stomata openedwith concomitant accumulation of K$ in the guard cells, although1–2 hr treatment caused opening without concomitant K$increase. The effect of glucose on flower bud formation was studied inthin-layer tissue cultures of epidermal strips from flower stalksof Nicotiana tabacum L. cv. Samsun. A minimum concentration of 30 mol m–3 glucose in the MS-mediumcontaining 1.0 mmol m–3 of both NAA and BA was necessaryfor flower bud formation. With 150 mol m–3 glucose a minimumstay of 10 d was required for optimal flower bud formation. Withholding glucose for a limited period at different time intervalsafter the onset of culture caused a delay in flower bud formationand did not affect previous development on glucose. The resultsindicated that competence for flower bud initiation is not restrictedto the early stage of culture. The process may start at anytime later at the appropriate glucose concentration. However,for both optimal initiation and further development of flowerbuds the presence of a metabolizable sugar is required. The ionophores benzo-18-crown-6 (18-C-6), t-butylbenzo-18-crown-6(TBB) and di-t-butyldibenzo-30-crown-10 (30-C-10) were testedfor their effects on potassium ion absorption in onion rootsegments, and in wheat and mung bean seedlings. Potassium uptake,efflux and transport were progressively reduced in onion rootsegments and seedlings by 18-C-6 over the range 0.1–1.0mM. The effects of TBB (up to 0.3 mM) were more severe but otherwisegenerally similar to those of 18-C-6 in seedlings. Both ionophoresreduced growth, and at the highest concentrations, resultedin root potassium ion content falling below initial values after48 h treatment. The effects of 30-C-10 were evident at muchlower concentrations, inhibition of net potassium uptake occurringabove 10–4 M. Between 10–4 and 10–4 M 30-C-10,however, a modest but significant stimulation of potassium uptakewas observed in onion roots and seedlings; growth of seedlingswas largely unaffected. The reductions in potassium absorptionwere attributed to the promotion, by the ionophores, of facilitateddiffusion down the electrochemical diffusion gradient, counteringthe efficiency of the potassium ion influx pump. Stimulationof uptake at certain concentrations of 30-C-10 was consideredmore likely to be due to an inhibition of passive potassiumefflux, rather than a stimulation of active influx. The importanceof stability constants, bonding and lipophilicity, in determiningthe relative effectiveness of the ionophores, is discussed. Potassium channels of Chara corallina have an activation energyof 36±1 kJ mol–1 and 50±2 kJ mol–1at temperatures higher and lower than 15°C respectively.The fast time variant conductance property of potassium channelsis insensitive to sodium and magnesium ions and is depressedby the presence of calcium, potassium and tetraethylammoniumions. Using excised low-salt roots of barley and Atriplex hortenslsthe transport of endogenous potassium through the xylem vesselswas studied It was enhanced by nitrate and additionally by sodiumions which apparently replaced vacuolar potassium which wasthen available in the symplasm of root cells for transport tothe shoot Vacuolar Na/K exchange also has been investigatedby measurements of longitudinal ion profiles in single rootsof both species. In Atriplex roots a change in the externalsolution from K+ to Na+ induced an exchange of vacuolar K+ forNa+, in particular in the subapical root tissues and led toincreased K+ transport and loss of K+ from the cortex. In inverseexperiments a change from Na+ to K+ did not induce an exchangeof vacuolar Na+; merely in meristematic tissues Na+—apparentlyfrom the cytoplasm—was extruded in exchange for K+. Inroots of barley seedlings without caryopsis, as in excised roots,a massive exchange of K+ for Na+ was observed in the continuouspresence of external 1.0 mM Na and 0.2 mM K. This exchange alsowas attributed to the vacuole and was most pronounced in theyoung subapical tissues. It did not occur, however, in the correspondingtissues in roots of fully intact barley seedlings. In these,the young tissues retained a relatively high K/Na ratio alsoin their vacuoles. Similarly, contrasting results were obtainedwith intact and excised roots of Zea mays L. Based on theseresults a scheme of the events that lead to selective cationuptake in intact barley roots is proposed. In this scheme acrucial factor of selectivity is sufficient phloem recirculationof K+ by the aid of which K+ rich cortical cells are formednear the root tip. When matured these cells are suggested tomaintain a high cytoplasmic K/Na ratio due to K+ dependent sodiumextrusion at the plasmalemma and due to recovery of vacuolarK+ by Na/K exchange across the tonoplast. Epidermal strips were taken from plants of Commelina communisat opposite phases of an entrained circadian rhythm, at timeswhich corresponded to the middle of the day phase and the middleof the night phase. Earlier observations of much reduced openingin response to external stimuli in the night phase were confirmed. Evidence is now presented that uptake or retention of specificions by the guard cells is affected by the phase of the rhythm.The overt expression of the rhythm was reduced if the epidermiswas presented with potassium iminodiacetate instead of potassiumchloride. Iminodiacetate is a non-absorbable anion, and thisresult suggested that chloride uptake is important for the fullexpression of the rhythm. The use of an anion channel inhibitor(DIDS) gave strong support for this conclusion. Reduced uptakeof chloride would explain an earlier observation that formationof malate is greater during the night phase. When the guard cells were presented with sodium chloride insteadof potassium chloride there was still an overt expression ofthe rhythm, but this was of reduced magnitude. This suggeststhat potassium movements might contribute to the rhythm, andstudies of ionic fluxes using 86Rb as a tracer showed greatereffluxes during the night phase. It is concluded that the circadian rhythm in stomatal openingmay be the result of a varying ability of the guard cells toaccumulate or retain both chloride and potassium ions. A rapid and convenient procedure was developed for isolatingguard cell protoplasts (GCPs) from epidermal strips of Viciafaba L. The mean rates of O2 uptake in the dark and evolutionin light of the isolated GCPs were 200 and 290 µmol O2mg–1 Chl h–1, respectively, showing net O2 evolutionin light. Photosynthetic O2 evolution was suppressed completelyby 5 µM DCMU. Stomatal conductances were measured in ageing leaves of Commelinacommunis L. as plants developed; stomatal responses to CO2 andabscisic acid (ABA) in epidermal strips of C. communis takenfrom ageing leaves of developing plants and in epidermal stripsfrom the same-aged leaves (the first fully-expanded leaf) ofdeveloping plants were also monitored. Stomatal behaviour wascorrelated with parallel measurements of photosynthesis andleaf ABA concentrations. Stomatal conductance in intact leavesdecreased from a maximum of 0-9 cm s– 1 at full leaf expansionto zero about 30 d later when leaves were very senescent. Conductancesdeclined more slowly with age in unshaded leaves. Photosynthesisof leaf slices also declined with age from a maximum at fullleaf expansion until about 30 d later when no O2 exchange wasdetectable. Exogenously applied ABA (0.1 mol m– 3) didnot affect respiration or photosynthesis. In epidermal stripstaken from ageing leaves the widest stomatal apertures occurredabout 10 d after full leaf expansion (just before floweringbegan) and then decreased with age; this decrease was less dramaticin unshaded leaves. The inhibitory effects of ABA on stomatalopening in epidermal strips decreased as leaves aged and wasgreater in the presence of CO2 than in its absence. When leaveswere almost fully-senescent stomata were still able to open.At this stage, guard cells remained healthy-looking with greenchloroplasts while mesophyll cells were senescing and theirchloroplasts were yellow. Similar data were obtained for stomatain epidermal strips taken from the same-aged leaves of ageingplants. The inhibitory effects of ABA on stomatal opening alsodecreased with plant age. In ageing leaves both free and conjugated ABA concentrationsremained low before increasing dramatically about 30 d afterfull leaf expansion when senescence was well advanced. Concentrationsof free and conjugated ABA remained similar to each other atall times. It is concluded that the restriction of stomatal movements inintact leaves as the leaves and plants age is due mainly toa fall in photosynthetic capacity of the leaves which affectsintracellular CO2 levels rather than to an inherent inabilityof the stomata to function normally. Since stomatal aperturein epidermal strips declines with plant and leaf age and stomatabecome less responsive to ABA (while endogenous leaf ABA levelsremain fairly constant until leaf senescence) it is suggestedthat some signal, other than ABA, is transmitted from the leafor other parts of the plant to the stomata and influences theirbehaviour. Plasmolytic disruption of plasmodesmata interconnecting metaphloemsieve element-companion cell complexes with small and largephloem parenchyma cells in the elongating region of internode2 ofPhaseolus vulgarisL. seedlings did not affect accumulationof phloem-imported14C-photosynthates and86rubidium. The membrane-impermeantdye, 5(6) carboxyfluorescein, loaded into leaf phloem as themembrane-permeant diacetate ester, was found not to move radiallyout of the importing sieve elements in the internode elongationregion. In contrast, the apoplasmic tracer, Calcuofluor White,rapidly moved laterally throughout all tissues of the elongationzone. Hexoses, sucrose and potassium were identified as themain osmotica in internode apoplasmic sap. Label asymmetry in[14C](fructosyl)sucrose was retained on accumulation by excisedstem segments. Uptake of [14C]sucrose and86rubidium by stemsegments exhibited saturation kinetics. Sucrose uptake was inhibitedby the slowly penetrating sulphydryl reagent, para-chloromercuribenzenesulphonicacid.In vitrorates of sucrose uptake, at apoplasmic concentrations,corresponded to its predictedin vivorate of delivery to thestem ground tissues from mature sieve elements when respiratorylosses were assumed to be confined to the stem phloem. For potassium,the total delivery rate could be accounted for by itsin vitrorateof uptake.
