Diffrent types of cytoplasmic inclusions were observed in ultrathin sections of plants systemically infected with three different strains of ribgrass mosaic virus (RMV) (tobamovirus group). Tissue from uninoculated plants did not contain such inclusions. Most common were “rounded plates” consisting of layers of aligned virus particles 300 nm long. RMV also induced angled layer aggregates in Capsicum annuum plants. A novel type of inclusion for the tobamovirus group were the abundant spiral aggregates found in Digitalis purpurea, systemically infected with strain D of RMV. In these aggregates the virions become circularly arranged around a center. The orientation of the particles changes in such a way that virions being 300 nm apartare cut in the longitudinal and in the transverse direction respectively.  相似文献

In Lemtal Italian and S.24 perennial ryegrass plants, two isolates of ryegrass mosaic virus (RMV) suppressed the amount of crown rust emerging on leaves inoculated with Puccinia coronata uredospores by up to 75% compared with the amount on virus-free plants. Severity of rust infection on barley yellow dwarf virus (BYDV) infected plants generally did not differ significantly from that on virus-free plants. When both RMV and BYDV were present, rust was restricted in Lemtal plants to a level intermediate between those occurring on plants infected by either virus alone, and in S.24 plants to a level below that obtained with either virus alone. The mean water soluble carbohydrate (WSC) content of Lemtal plants was reduced more than 20% by RMV, but was not significantly altered by BYDV. In S.24 plants the WSC content was increased by 10% by RMV and by 60% by BYDV. Rust reduced the WSC content of healthy and virus-infected plants, the reduction being positively correlated with the level of rust on the sampled leaves. In plants of Lemtal, but not of S.24, the degree of rust infection was positively correlated with the WSC content of leaves from rust-free control plants.  相似文献

Summary Two representative cultivars ofCicer arietinum, the desi-type cv.Annigeri and the kabuli-type cv.ICCV6, were regenerated in vitro and clonally propagated from cotyledonary nodes and meristem tips. The explants were dissected from 1-wk-old seedlings aseptically germinated on WH medium. In both cultivars, all nodes cultured on B5 medium supplemented with 4.4μM 6-benzylaminopurine developed up to seven shoots per node within 3 wk. Meristem tips were much better suited for multiple shoot formation. Cultured on DKW-C-a medium supplemented with 4.4μM 6-benzylaminopurine and 0.05μM indole-3-butyric acid, 96% of the meristem tips produced up to 10 shoots per explant. A new method in improving clonal propagation was subdividing the meristem tips. Doing so, multiple shoot formation was considerably enhanced: up to 90 shoots per original explant could be obtained with cv.Annigeri, and up to 50 with cv.ICCV6. Indole-3-butyric acid proved to be the best rooting factor. From several media tested, the best root induction and development was achieved on WH medium supplemented with 2.5μ M indole-3-butyric acid: 72% rooting with cv.Annigeri and 68% rooting with cv.ICCV6. With both cultivars there were no differences in rooting capacity between shoots of nodal origin and those derived from meristem tips. The plantlets obtained were transferred into soil and kept under greenhouse conditions. The survival frequency was 28% with cv.Annigeri and 23% with cv.ICCV6. R0 plants remained smaller than seed-grown controls and produced only a few fertile seeds. There was no difference between R1 plants and controls in growth, development, and seed set.  相似文献

Mixing the ryegrass mosaic virus (RMV) resistant perennial ryegrass (Lolium perenne) cv. Endura with the susceptible Italian ryegrass (L. multiflorum) cv. RvP decreased infection of RvP wth RMV from 37% when grown alone to 22% when mixed. However, Endura yielded less than RvP and there was no yield benefit from mixing the two cultivars. Mixing red clover (Trifolium pratense) cv. Hungaropoly with RvP had no detectable effect on RMV incidence in RvP but did decrease the incidence of red clover necrotic mosaic virus in Hungaropoly from 9% to 1% and of white clover mosaic virus from 53-5% to 41%. The yield of the mixture was equal to that of RvP grown alone but given nitrogen fertiliser. The numbers of eriophyid mites, including Abacarus hystrix the vector of RMV, on ryegrass leaves were similar in pure and mixed swards. It is concluded that with herbage crops, the common practice of sowing mixtures of species may help control virus diseases.  相似文献

In a field experiment at Hurley, small areas of old ryegrass sward, known to be infected with ryegrass mosaic virus (RMV) were partially surrounded by newly-sown Italian ryegrass. The experiment was harvested by mowing on five occasions. Samples of Italian ryegrass taken when it was 11 months old showed a very marked increase in the levels of RMV close to the areas of old sward. Thus, 84% of the Italian ryegrass plants bordering the old sward to the north contained RMV, compared with 63% of plants only 2.7 m away. The corresponding values to the south were 68% and 39%. The much higher values encountered to the north are presumed to be caused by the movement of the mite vector of RMV (Abacarus hystrix) in relation to the prevailing wind. Two different mowers were used, a rotary mower and a reciprocating knife mower. We could demonstrate no increase in the level of RMV as a result of mowing from the infected sward and on to the new sowing and could demonstrate no difference in the infection levels as a result of using the different mowers. We conclude that in mown swards migration by the mite is responsible for all or nearly all of very local spread of RMV.  相似文献

The influence of ryegrass mosaic virus (RMV) on yield of perennial ryegrass swards was examined under two harvesting frequencies and at two levels of nitrogen fertilizer, in a field trial planted with a clone of ryegrass, cv. S 321, using small plots established with proportions of healthy and infected plants. RMV reduced the height and the number of tillers of plants grown in pots and reduced the height also of plants in the field. The swards were planted in August and at a harvest 8 wk later RMV reduced the yield from 2–12 to 1–52 t dry matter (d.m.)/ha. In the first full harvest year, RMV caused only a small reduction in yield where no N fertilizer was applied and the application of a very small amount of fertilizer would have recouped the loss. On the other hand RMV severely restricted the increase in yield resulting from heavy dressings of the fertilizer. Thus where 400 kg N/ha was applied, RMV reduced the total annual yield from 18-6 to 13-8 t d.m./ha. The effect of the virus was especially great both in the Spring, the period of maximum herbage production, and when the level of fertilizer was high.  相似文献

Perennial ryegrass plants collected from fields and Italian ryegrass plants grown from seed were selected for resistance to infection by ryegrass mosaic virus (RMV) by repeated manual inoculation. Two of 108 perennial ryegrass plants and one of 150 Italian ryegrass plants were symptomless after seven and nine inoculations respectively. These three plants were propagated vegetatively. Plants of the two perennial ryegrass clones showed no symptoms after further manual inoculations with the initial isolate of RMV, or with an inoculum from infected plants collected from several fields, or after inoculation by viruliferous mites. Electron microscopy and back tests indicated that the plants were virus free. Some plants of the selected Italian ryegrass clone became infected after a further inoculation with mites or sap, but fewer than similarly inoculated unselected plants.  相似文献

Methods previously established for the propagation of cassava plants free from cassava mosaic disease have been applied to Nigerian clones. Meristem tips from diseased plants subjected to heat treatment for not less than 30 days at 35°–38°C were cultured on modified Murashige-Skoog medium. Concentration ranges of benzyladenine in combination with α-naphthalene acetic acid and gibberellic acid were investigated and, at optimal levels, 36% of the meristems regenerated. Regenerants, with callus and shoots only, were rooted with 80% efficiency by sub-culturing following a dip in a hormone rooting powder. All plants raised from heat-treated meristems were free of the disease as judged by visual inspection of the leaves, rooted explants and assay for the suspected pathogenic agent of the disease.  相似文献

Several Agropyron species were tested for new sources of resistance to barley yellow dwarf virus (Bydv ) and wheat streak mosaic virus (WSMV). With BYDV strain PAV, 11 of the 17 Agropyron species showed no virus transmission when plants were given access feed by viruliferous Rhopalosiphum padi. Similar trials with BYDV strain RMV (vectored by R. maidis) indicated that all plants, except susceptible control plants, remained virus free. Virus status was confirmed by enzyme-linked immunosorbent assays. When plants were mechanically inoculated with WSMV, 11 Agropyron species failed to express symptoms, while five other species showed a segregating response or had some accessions segregating and some resistant. Test results suggest that resistance to BYDV and WSMV in Agropyron species does not appear to be correlated with any specific genome of Agropyron species although most of the Agropyron species containing S genome were resistant to BYDV and WSMV.  相似文献

After exposure to infection in the field, the proportion of plants showing distinct symptoms of ryegrass mosaic virus (RMV) was less in perennial than in Italian ryegrass. The perennial ryegrass cv. Mascot had a smaller proportion of plants with symptoms than cv. S.23. Far milder symptoms were induced in test plants by RMV from naturally infected perennial ryegrass plots than from Italian ryegrass plots. Within perennial ryegrass, RMV from cv. Mascot caused milder symptoms than that from cv. S.23. Severe RMV isolated from Italian ryegrass cv. Trident (RMVT) became milder after one passage through cv. Mascot, although not as mild as RMV obtained from field plots of cv. Mascot (RMVM). Families from two highly resistant perennial ryegrass clones and two randomly selected clones of cv. S.23 crossed in all possible combinations varied in symptom severity when inoculated with RMVT but not when inoculated with RMVM. Families inoculated with RMVT also yielded virus which varied in the severity of symptoms induced in test plants, families with severe symptoms yielding severer virus. Thus, much of the variation in the resistance of these clones could be attributed to infection with RMV of differing severity. Resistance was controlled by several genes which were mainly additive in their effect.  相似文献

Peanut (Arachis hypogaea L.) plants (cvs Florunner and Pronto) were inoculated at the two-leaf stage with peanut mottle virus (PMV) to obtain PMV-infected plants. Shoot-tips from plants grown in the glasshouse (27°C) or from plants maintained at 35°C were used for tip culture. In some experiments ribavirin was added to the culture medium at 5 mg/1, 10 mg/1, 15 mg/1 or 20 mg/1. Plants regenerated from meristems or shoot-tips taken from virus-infected plants were not virus-free. After 45 days at 35°C, foliar tissue of 93 % of Florunner and 95 % of Pronto plants tested negative for PMV by enzyme-linked immunosorbent assay (ELISA). When shoot-tips from the plant that tested negative by ELISA were used for tip culture, no virus-free plants were obtained. No virus-free plants were obtained from tips cultured on medium supplemnted with ribavirin. However, when tip culture, thermotherapy and chemotherapy were combined; 80 % of Florunner and 100 % of Pronto plants were found negative for PMV.  相似文献

Ryegrass mosaic virus (RMV) was reliably detected in both perennial (S24) and Italian (S22) ryegrass, by enzyme-linked immunosorbent assay (ELISA) when plants had been infected for 8 wk. ELISA detected more infections in field-grown perennial ryegrass cv. Premo than either visual assessment or electron microscopy. However, with plants of Italian ryegrass cultivars only recently infected with RMV, positive reactions were more difficult to separate from the reactions of RMV-free plants, which varied considerably with cultivar, some giving high absorbance values. Immunosorbent electron microscopy showed that the RMV antiserum also contained antibodies to ryegrass seed-borne virus (RGSV), suggesting that these high values were caused by RGSV infection in the material tested.  相似文献

Indian citrus ringspot virus (ICRSV) is known to cause serious disease problem in Kinnow (Citrus nobilis Lour × C. deliciosa Tenora) plants. This work reports the elimination of ICRSV by using thermotherapy coupled with shoot tip grafting in vitro. Nodal segments from infected mother plants (indexed by indirect ELISA and RT-PCR) were treated both in water bath and moist hot air at different temperatures viz. 40, 45 and 50°C for 30, 60 and 120 min and cultured on MS medium containing 2-iP (1 mg/l) and malt extract (800 mg/l). Shoot tips were excised from the nodal sprouts and grafted on to rough lemon (C. jambhiri) rootstock under aseptic conditions. Water bath treatment was found to be more effective as compared to moist hot air treatment as maximum number of ICRSV free plants (36.84%) were obtained by grafting the tips (0.7 mm) taken from the nodal segments treated at 50°C in water bath for 2 h. In an alternate treatment regime, 1-year-old infected plants were kept at various temperatures viz.36, 38 and 40°C in a thermotherapy chamber. Maximum of 60% ICRSV free plants were obtained by grafting the tips (0.7 mm) from the plants placed at 40°C followed by the plants placed at 38°C (59.09%) and the least was observed in case of the plants placed at 36°C (40.74%). Only those plants/plantlets were considered virus free, which showed negative reaction both in Indirect ELISA and RT-PCR.  相似文献

The witches' broom disease of sweet potato has so far been reported only from Asia and the South Pacific. To insure safe distribution of germplasm it is important that vegetative material introduced from regions in which this disease is present has to be free of this pathogen. Meristem tip culture was found to be effective in eliminating MLO from infected sweet potato tissue. On the other hand, all plantlets originating from 1 cm long stem tips immediately below the meristems developed typical witches' broom symptoms as early as 5 weeks after excision of the stem tips. A high percentage of stem tips originating from cuttings previou.sly treated with 25, 50 and 100 mg tetracycline for 3 days developed into healthy plants. Similarly, more than 90 % of plants derived from stem tips excised from cuttings which had received a one-month heat treatment at 37°C remained without symptoms for 9 months.  相似文献

In 1983, cereal plants showing symptoms of barley yellow dwarf virus (BYDV), collected from 15 localities in Sweden, were tested for BYDV using enzyme-linked immunosorbent assay (ELISA). Antisera against two Swedish isolates of BYDV were used, a mild isolate (27/77) transmitted specifically by Sitobion avenae and a severe one (39/78) transmitted mainly by Rhopalosiphum padi. No virus was detected in 57 of 607 plants of oats and barley tested. Of the 550 plants in which virus was detected, 366 were infected with viruses similar to isolate 27/77, 116 with viruses similar to 39/78 and the remaining 68 reacted strongly with both antisera. When tested, the latter isolates were shown to be mixtures. Thirty-nine selected samples were also tested with antisera against the USA isolates RPV, RMV, MAV and PAV, and for transmission by S. avenae and R. padi. Twenty-six of these samples were transmitted specifically by S. avenae, one was transmitted only by R. padi and the remaining 12 samples were shown to be infected with a mixture of an S. avenae-specific isolate and one transmitted mainly by R. padi. Antisera against PAV and MAV each detected all isolates tested and the results were very similar to those with the antisera to the 39/78 and 27/77 isolates, respectively. None of the field isolates reacted with antisera against RMV or RPV. It was concluded that 1983 was an epidemic year for BYDV in Sweden and that isolates specifically transmitted by S. avenae predominated. Symptoms of infection by these isolates on oat plants ranged from mild to severe.  相似文献

In order to develop transgenic plants via the biolistic gun method regenerable embryogenic tissues are required. Meristem shoot tips of 19 cultivars of cotton were cultured on several media formulations and assessed for shoot and root development. The best shoot development was observed on media containing 0.46 mM kinetin while rooting was observed on media containing 2.68 mM NAA and 0.46 mM kinetin. No intervarietal variability was observed. A complete protocol was developed from meristem tip culture to field transfer. This methodology is simple and replaces the existing protocols for meristem tip culture of cotton. The Pineapple mealybug wilt-associated virus (PMWaV) provokes a disease that causes considerable losses to pineapple growers due to the reduced leaf tissue turgescence, resulting in leaf necrosis and death in severe cases. In this work, we describe the comparison of shoot tip culture and cryotherapy methods to eradicate the pineapple wilt disease-associated ampeloviruses. Plants from the accessions Ananas comosus var. comosus (AGB-009), var. bracteatus (AGB-119), and var. parguazensis (AGB-376) were indexed by RT-PCR, confirming mixed infections of PMWaV-1, PMWaV-2, and PMWaV-3. The accessions were cultured in vitro and their shoot tips treated by cryotherapy following a droplet vitrification protocol. The regeneration rate from shoot tip culture was 93% for AGB-376 and 100% for the other two accessions. After freezing, AGB-376 had 100% regeneration subsequent to exposure to PVS2 for 45 min, followed by 95% for AGB-009, while for AGB-119 the optimal exposure time was 60 min, with plant regeneration from nearly 80% of the shoot tips. For the accessions AGB-009 and AGB-376, all the recovered plants were virus free by the two methods, while 50% of the plants from accession AGB-119 remained infected. These results indicate that shoot tip culture alone or in association with cryotherapy is a promising routine method for virus removal from pineapple plant tissues and is useful to ensure that backup reserves of pineapple germplasm, conserved by in vitro bank and cryobank, are formed with virus-free plants.

The effect of rate of nitrogen applied to three Lolium perenne-based field swards on the development of RMV symptoms and on the impact of the virus on the growth and composition of the swards was examined over two growing seasons. Glasshouse experiments investigated the competition between Lolium spp. in relation to RMV infection and rate of nitrogen. In general, there was a higher incidence of RMV symptoms at the higher rates of nitrogen but the relationship was not consistent in all experiments. RMV infection only significantly reduced the cumulative total yield of the sward that contained the very susceptible Lolium multiflorum cv. Barverdi and was treated with the higher rate of nitrogen. However, yield losses were consistently greater at the higher rates of nitrogen. RMV altered the relative yields of sward components and the effect was greatest after an extended period of severe frost at the end of 1995. There is evidence that the frost severely restricted the yield of RMV-infected L. multiflorum tillers in 1996. Where RMV reduced the yield of susceptible species, there was significant compensatory growth by less susceptible companion species, i.e. L. perenne and Phleum pratense, but the compensation was generally incomplete, resulting in reduced yields for virus-infected swards. RMV had no significant effect on the overall quality of the three field swards but for cv. Barverdi grown in the glasshouse it significantly reduced the percentage organic matter and water soluble carbohydrate, and D value. The reductions were greater at the higher rates of nitrogen.  相似文献

An efficient protocol forin vitro micropropagation of seven indica rice varieties was developed from meristem culture. Meristem (leaf base) was isolated from different age of seedlings and cultured on MS medium without hormones and supplemented with different concentrations of NAA and BAP. Regeneration of plantlets from meristem was observed within five days of culture. The meristem isolated from 4-day old seedlings gave highest regeneration on hormone free MS medium. Histological study of meristem (leaf base) from 4-day old seedlings confirmed the presence of meristematic cells. Regenerated plants were multiplied on MS medium supplemented with 0.05 mg/L NAA and 5 mg/L BAP. An average of five plants were obtained from single regenerated meristem. The plants regenerated from meristem showed morphological uniformity.  相似文献
