南昌霉素高产菌株的链霉素抗性基因突变诱变筛选研究   总被引:10,自引:0,他引:10  
通过对链霉素对南昌霉素(Nanchangmycin)产生菌NS-41-80菌株孢子的致死浓度测定基础上,采用诱变剂甲基磺酸乙酯(EMS)的不同诱发剂量对菌株孢子进行诱变处理,诱变处理的孢子涂布在含链霉素(10ug/mL)致死浓度的高氏平板上,获得了大量的链霉素抗性基因(str)突变株。然后从3,000株链霉素抗性基因(str)突变株中通过初筛获得比诱变出发菌株产素能力提高20%以上的菌株202株,再进一步通过摇瓶复筛,获得比出发菌株产素能力分别提高100%,200%,300%高产菌株为48株,7株和1株,分别为复筛菌和初筛菌株的23.76%和1.60%,3.46%和0.23%,0.5%和0.03%,将产素能力提高240%以上5个菌株连同出发菌株连续3批次进行摇瓶发酵结果,5个突变株的产素能力均比出发菌株的产素能力提高57%-96.4%,其中突变株80-5.3-165菌株摇瓶发酵单位达6,000ug/mL以上,3批次摇瓶平均发酵单位达5,855ug/mL,建立了南昌霉素高产菌株的链霉素抗性基因突变诱变快速高效的筛选方法。  相似文献

氮离子注入选育阿维拉霉素高产菌株的研究   总被引:3,自引:0,他引:3  
以提高产绿链霉菌(Streptomyces viridochromogenes)SV-1产阿维拉霉素(Avilamycin)产量为目的,采用低能氮离子注入技术,辅之以链霉素抗性筛选法进行诱变选育研究。结果表明,“马鞍”区域即注入剂量范围在3×10~(15)~5×10~(15)ions/cm~2诱变效果最佳,菌株的抗药性突变与产量突变密切相关,链霉素抗性筛选法具有可行性。在摇瓶条件下,最终获得稳定性良好,阿维拉霉素产量达到83.5mg/L,较出发菌株提高195%的突变株SVT-45。实验表明,离子注入技术是一种有潜力的微生物诱变育种新方法。  相似文献

以林肯链霉菌9502(Streptomyces lincolnensis9502)为出发菌株,进行NTG诱变处理,并用高效的琼脂块培养法对菌株进行筛选,得到产林肯霉素相对效价提高35.4%的变异株9502-7。对9502-7菌株孢子采用紫外线处理,得到变异高产菌株9502-7-12,其相对效价较出发菌株提高50%以上。  相似文献

可利霉素是通过基因工程定向育种技术获得的新型大环内酯类抗生素,是国家一类新药.[目的]为满足工业化生产需要,其工程菌株的发酵水平有待提高.[方法]多种常规诱变技术交替处理和高通量筛选方法选育可利霉素高产菌株,处理方法包括原生质体紫外诱变、DES(硫酸二乙酯)诱变、紫外光复活诱变、缬氨酸抗性筛选和正突变菌株的富集.[结果]高产菌株WSJ-1-7-49-133-82-43的摇瓶生物效价比出发菌株WSJ-1-7-49提高56%,500L中试发酵罐突变菌株效价较出发株高61%.[结论]说明多轮常规诱变育种结合高通量的筛选方法可以用于工业生产菌株的高效筛选.  相似文献

应用链霉素抗性筛选法,将经过紫外线诱变处理的纳他霉素生产菌——褐黄孢链霉菌(Streptomyces gilvosporeus)ATC13326的孢子涂布在含有链霉素最小抑制浓度(0.6μg/mL)的培养基平板上,获得了122株链霉素抗性突变株。其中纳他霉素产量高于出发菌株的有13株,产量阳性效率达到10.6%,同时获得了产抗生素能力为出发菌株1.46倍的突变株SG-56。  相似文献

利用氮离子注入对链霉菌的诱变效应,筛选高产恩拉霉素的变异菌株。利用不同剂量的氮离子对杀真菌放线菌S.fungicidicus NL629-3菌株进行诱变处理,研究低能氮离子注入对其存活率及产恩拉霉素能力的影响。低能氮离子注入剂量在60×1013ions/cm2时对链霉菌的诱变效应显著,试验得到了5株恩拉霉素产量较高的突变菌株,其中N3-643菌株经连续传代4次,遗传稳定性较好,其摇瓶发酵水平较对照提高了41%,放大发酵生产后平均发酵水平提高25.8%。离子注入诱变是获得高产恩拉霉素突变菌株的有效方法。  相似文献

目的:获得博安霉素高产菌株,同时比较了铜蒸汽激光与妥布霉素抗性及二者复合诱变的选育效果。方法:采用铜蒸汽激光辐照30 min与妥布霉素100 r/m L抗性处理及其复合诱变选育博安霉素产生菌轮枝链霉菌(S.verticillus)B-31。结果:在复合诱变组中,获得一株高产突变株GB-160,经发酵罐应用后,发酵单位较出发菌株提高1.5倍,并且遗传性能稳定。结论:该方法能有效获得抗生素高产优质菌株。在医药生物工程中,具有较高的实用价值,为其它药物微生物选育提供借鉴。  相似文献

This study aimed to obtain strains with high glyphosate-degrading ability and improve the ability of glyphosate degradation enzyme by the optimization of fermentation conditions. Spore from Aspergillus oryzae A-F02 was subjected to ultraviolet mutagenesis. Single-factor experiment and response surface methodology were used to optimize glyphosate degradation enzyme production from mutant strain by liquid-state fermentation. Four mutant strains were obtained and named as FUJX 001, FUJX 002, FUJX 003, and FUJX 004, in which FUJX 001 gave the highest total enzyme activity. Starch concentration at 0.56%, GP concentration at 1,370?mg/l, initial pH at 6.8, and temperature at 30°C were the optimum conditions for the improved glyphosate degradation endoenzyme production of A. oryzae FUJX 001. Under these conditions, the experimental endoenzyme activity was 784.15?U/100?ml fermentation liquor. The result (784.15?U/100?ml fermentation liquor) was approximately 14-fold higher than that of the original strain. The result highlights the potential of glyphosate degradation enzyme to degrade glyphosate.  相似文献

Application of random effects to the study of resource selection by animals   总被引:5,自引:0,他引:5  
1. Resource selection estimated by logistic regression is used increasingly in studies to identify critical resources for animal populations and to predict species occurrence. 2. Most frequently, individual animals are monitored and pooled to estimate population-level effects without regard to group or individual-level variation. Pooling assumes that both observations and their errors are independent, and resource selection is constant given individual variation in resource availability. 3. Although researchers have identified ways to minimize autocorrelation, variation between individuals caused by differences in selection or available resources, including functional responses in resource selection, have not been well addressed. 4. Here we review random-effects models and their application to resource selection modelling to overcome these common limitations. We present a simple case study of an analysis of resource selection by grizzly bears in the foothills of the Canadian Rocky Mountains with and without random effects. 5. Both categorical and continuous variables in the grizzly bear model differed in interpretation, both in statistical significance and coefficient sign, depending on how a random effect was included. We used a simulation approach to clarify the application of random effects under three common situations for telemetry studies: (a) discrepancies in sample sizes among individuals; (b) differences among individuals in selection where availability is constant; and (c) differences in availability with and without a functional response in resource selection. 6. We found that random intercepts accounted for unbalanced sample designs, and models with random intercepts and coefficients improved model fit given the variation in selection among individuals and functional responses in selection. Our empirical example and simulations demonstrate how including random effects in resource selection models can aid interpretation and address difficult assumptions limiting their generality. This approach will allow researchers to appropriately estimate marginal (population) and conditional (individual) responses, and account for complex grouping, unbalanced sample designs and autocorrelation.  相似文献

本文以实验室筛选得到的翰逊德巴利酵母菌(Debaryomyces hansenii)DH01为出发菌株,利用氯化锂诱变选育3-羟基丙酸(3HP)产量高并且可以用于实际生产的突变株。经过两轮氯化锂诱变,筛选得到一株产酸量较高的菌株wt06。在培养温度为28℃,120 r/min的条件下,经过48 h培养后,该菌株3HP的产量最高达到23.70 g/L,是原始菌株产量的5.44倍,且该菌株具有良好的遗传稳定性。  相似文献

Firefly luciferase covers a wide range of applications. One common usage of the bioluminescence assay is the measurement of intracellular concentration of adenosine triphosphate (ATP) for cell viability. However, inhibition of the enzyme reaction by chemicals in the assay has so far limited the application of luciferase for high production volume (HPV) chemical testing. The objective of this research was to obtain a mutant luciferase with increased stability to inhibition by HPV chemicals, yet retaining specific activity comparable to, or better than, wild-type luciferase. The enzymatic properties of the wild-type luciferase were improved by random mutagenesis and colony-level screening. In this paper, the detailed process of creating mutant luciferases for testing the toxicity of HPV chemicals is described. As a result, two mutant luciferases were created, with different degrees of improved tolerance to inhibition by chloroform and other HPV chemicals.  相似文献

The feasibility of altering the chain length specificity of industrially important Rhizomucor miehei lipase was investigated by randomly mutating Phe94 in the protein groove which is responsible for accommodating the acyl chain of the substrate. The recombinant lipase was initially expressed in E. coli. Individual colonies were selected, grown, and the DNA sequence of the lipase gene determined. Fourteen of the 19 possible mutants were identified and each of these was transformed into Pichia pastoris which expresses the enzyme extracellularly. The yeast was grown and the supernatants assessed in several assays with long and short chain substrates. Based on this preliminary screen, one mutant, Phe94Gly, was selected and purified to homogeneity for further analysis. It was found that the substitution of phenylalanine 94 with glycine led to an enzyme which was about six times less active against resorufin ester but displayed 3-4 times higher activity with short chain substrates such as butyric acid esters. The observed alteration to the enzyme specificity was rationalised using the available 3D structure of the lipase.  相似文献

为获得高产菊粉酶的黑曲霉菌株,以Aspergillus niger YH-1为出发菌株,经过亚硝基胍(NTG)诱变,以高温高菊芋粉相结合的方式进行梯度驯化,选育出一株产菊粉酶菌株YH-3,并运用响应面实验方法对该菌株的培养基进行优化。确定了最佳培养基组成:菊芋粉25.2 g/L、豆饼粉40 g/L、蔗糖酯4.9 g/L、NaCl 5.5 g/L。发现内切菊粉酶活力(I)由60.9 U/mL提高到165.0 U/mL,比出发菌株提高了1.7倍。研究证明蔗糖酯对于黑曲霉YH-3发酵产菊粉酶是一种有效的促进剂。  相似文献

以果胶酶产生菌黑曲霉EIM6为出发菌株,初始果胶酶活为14 539 U/mL,经紫外诱变反复处理,摇瓶复筛和遗传稳定性试验,最终获得一株果胶酶高产菌株EIMU2。EIMU2菌株的形态特征发生了明显的改变,相较于原出发菌株EIM6,孢子色泽更黑,孢子团也较出发菌株大,菌丝与孢子上凝结有更多的液珠。复筛后EIMU2酶活为32 161 U/mL,较原出发菌株提高了1.212倍。进一步通过响应面法对EIMU2菌株的液体发酵培养条件进行优化。优化后的培养条件为甜菜渣1.83%,花生饼粉1.69%,(NH_4)_2SO_4 0.5%,K_2HPO_4 0.3%,CaCO_3 0.2%,MgSO_4 0.15%(w/v),接种量6%(v/v),装液量21.36 mL。优化的突变菌株产酶活性进一步提高至98 794.3 U/mL,提高了2.07倍。  相似文献

A mutant producing a pyranose oxidase, which has a higher thermal stability and lower Km values for d-glucose and 1,5-anhydro-d-glucitol than those of the wild type enzyme, was obtained. A single amino acid substitution, Lys for Glu at position 542, had occurred. This altered enzyme, E542K, was not only stable at 55°C, which was 5°C higher than the wild-type enzyme, but was stable in alkaline solution at pH 8.0–11.0. Km values of E542K for d-glucose and 1,5-anhydro-d-glucitol were 0.7 mM and 14.3 mM, respectively, in contrast with 1.4 mm and 35.3 mM for the wild-type enzyme. A little effect was observed in kcat values, and improvement in reactivity was mainly due to the decreases in Km values. This altered pyranose oxidase is useful for food analysis and diagnosis.  相似文献

中国被毛孢(Hirsutella sinensis)是天然冬虫夏草[Cordyceps sinensis(Berk)Sacc.]的真正无性型,其生长温度低(低于20℃),生长缓慢(发酵周期长达20~30d),对人工发酵的技术条件要求很高,因而大大增加了工业化发酵生产的成本和技术难度。为了解决上述问题,实验中从天然冬虫夏草子座中分离到中国被毛孢菌株BS-1,并对其进行了原生质体紫外诱变处理。经过粗筛和精筛,从中选出一株可常温下(25~28℃)发酵、生长迅速(发酵周期4~5 d)、有效成分(甘露醇和腺苷)含量显著高于原始菌株的诱变株BS-5962。通过连续10代PDA斜面继代培养和摇瓶试验,以及连续3批的1t罐的中试试验,检查结果表明所得诱变株性状优异而稳定。可为降低中国被毛孢的液体深层发酵成本和技术难度提供一种有效的解决方案。  相似文献
