European corn borer (ECB; Ostrinia nubilalis (Hubner)) larvae (third instar) fed 0.05% w/w wheat germ agglutinin (WGA) in their diet for 72 h showed very little increase in body weight, whereas weight of control larvae increased nearly fourfold. Light and transmission electron microscopy studies showed that the morphology of the peritrophic membrane (PM) changed within 24 h after ECB larvae fed on the WGA diet. Whereas the PM in the anterior region of the midgut was a thin membranous structure in control larvae, the WGA-fed larvae secreted a multiple-layered and unorganized PM that contained embedded food particles, bacteria, and pieces of disintegrated microvilli. Gold-labeled WGA was localized specifically in the PM and microvilli. The PM of WGA-fed larvae was inundated with dark-staining amorphous structures that, when incubated with anti-WGA, showed heavy WGA localization. The antibody label indicated that most of the ingested WGA was found in the PM, with lesser amounts on the microvillar surface and the least amount within the epithelium. After 72 h, the middle portion of the mesenteron revealed a thin, compact PM in the control larvae, whereas the PM of the WGA-fed larvae was multilayered and discontinuous, which allowed plant cell-wall fragments to penetrate into the microvilli of the epithelium. Scanning electron microscopy of PMs from fifth instar ECB larvae fed the WGA diet revealed a breakdown in the chitinous meshwork by 48 h after initiation of feeding. The endo-PM surface from control larvae was smooth and intact, whereas the PM of WGA-fed larvae showed disintegration of the meshwork and a reduced proteinaceous matrix. This allowed bacteria and food particles to penetrate through the PM into the ectoperitrophic space and directly contact the microvilli. Therefore, WGA, a protein inhibitor of larval growth, interferes with the formation and integrity of the PM, which exposes the brush border to ingested material. This, in turn, appears to stimulate secretion of additional PM layers, the concomitant disintegration of the microvilli, and cessation of feeding.  相似文献

We have demonstrated that larvae of codling moth, Cydia pomonella (L) can successfully complete their first instar when fed apple leaves instead of fruit. Larvae fed leaves after hatching maximized their feeding intensity (about 320 g/larva/day) on day 2. Weight gain revealed a stereotypic sigmoid pattern that peaked on day 3. Although the maximum body weight of larvae fed leaves was 70–85% less than for larvae maintained on apples or on artificial diet, 100% of larvae fed leaves molted to the second instar 3–5 days after hatching. Our investigation revealed a diurnal pattern of leaf ingestion, and neonates' feeding intensity decreased significantly during the scotophase. We also demonstrated that monosodium glutamate (MSG) increased feeding on leaves by codling moth larvae. Depending on the duration of the bioassay, and larval age at time of initial exposure, 0.05 mg/ml and 0.1 mg/ml MSG increased apple leaf consumption by 25–60% over leaves alone. The effect of monosodium glutamate was best demonstrated during the first day following hatching. Exposure to MSG also accelerated molting to the second instar. Larvae exposed to MSG initiated consumption of leaf tissue significantly earlier than control neonates. The feeding stimulatory effect of MSG was not observed if exposure to this chemical was delayed until 3–4 h after hatch.The addition of feeding stimulants to pesticides that act via the alimentary tract may reduce the amount of active ingredients needed to maintain the efficacy of these formulations. Here, we postulate that first instar codling moth larvae are potential targets for treatment with pesticide formulations enhanced with monosodium glutamate.  相似文献

Nectarivorous birds encounter varying nectar concentrations while foraging on different food plants and must adjust their consumption to maintain constant energy intake. We determined how rapidly captive whitebellied sunbirds (Cinnyris talatala) adjust their volumetric intake and feeding patterns after changes in diet concentration. On four consecutive days, birds were fed sucrose diets alternating between a standard diet of 16% w/w and test diets of 2.5, 8.5, 16 or 30% w/w, respectively, for 1.5 h periods. Feeding events were recorded with an infrared photo-detection system and food intake and body mass were monitored continuously by electronic balances interfaced to a computer. Generally, birds demonstrated a measurable increase in feeding frequency and food intake within 10 min after a decrease in sucrose concentration. However, individuals responded differently to the most dilute diet (2.5%): while most increased their food intake, others stopped feeding for a short while, appearing to dislike this diet. Furthermore, the number and duration of feeding events increased in the first 5 min after the switch from 2.5% back to 16%, as the birds attempted to compensate for previous reduced sugar intake. Daily sugar intake was lower when birds alternated between 2.5 and 16% diets than on other test days, but birds were able to maintain body mass, presumably through behavioural adjustments.  相似文献

The effects of diet and different constant temperatures on hemolymph cation concentrations (Na+, K+, Mg2+, Ca2+) have been studied in Morimus funereus larvae collected from natural habitat, fed natural (oak or beech bark) or artificial diet, as well as in larvae reared from hatching on an artificial diet. In the hemolymph of larvae maintained under natural conditions Mg2+ was dominant, whereas Na+ concentration was very low. In their natural diets concentrations of Na+ and K+ were very low, while those of Ca2+ and Mg2+ were high. In larvae continuously reared on an artificial diet, hemolymph Mg2+ concentration was significantly decreased and Na+ concentration increased more than fourfold compared to the results obtained in oak-fed larvae. Na+ and K+ are the dominant cations in the artificial diet. The concentrations of K+ and Ca2+ in the hemolymph of larvae fed natural or artificial diet are nearly identical, suggesting the existence of an internal regulatory mechanism in this insect for these cations. The hemolymph cation concentrations of M. funereus larvae are predominantly dependent upon the diet consumed, much less upon the environmental temperatures. The most stable concentrations of cations were observed in larvae continuously fed an artificial diet and exposed to different constant temperatures. There was much less stability in the hemolymph cation concentration in oak larvae fed either natural or artificial food after their transfer to constant temperatures. The effect of the concentration and structure of dietary sterol on its uptake and distribution in the intestine, hemolymph and fat body was studied in sixth-instar larvae of Heliothis zea. When cholesterol (cholest-5-en-3β-ol) was inoculated per os into the foregut of larvae, it was rapidly taken up by the intestine. Some of the dietary sterol then passed into the hemolymph, primarily via the midgut, during at least the first 9 h after inoculation, while at least 7% of the dose remained associated with the intestine. The amount of dietary sterol per 0.10 g of hemolymph increased until it reached 3–6% of the dose after 9 h. The amount of sterol per 0.10 g of the fat body increased to as much as 5% of the dose after 10 h. As the concentration of sterol in the dose increased from 0.3 to 15 μg/4 μl, the amount of sterol associated with the intestine, hemolymph, and fat body also increased. When cholesterol was inoculated intrahemocoelically, instead of per os, the amount of sterol in the hemolymph decreased, for at least the first 8 h after inoculation, and may have been absorbed, at least in part, by the intestine. The absence of a double bond in cholestanol (5α-cholestan-3β-ol) had no significant effect, at least 5 h after inoculation, on the uptake and distribution of this sterol in the intestine, hemolymph, and fat body of the larva. The results of this study indicate that although larvae of H. zea fed cholestanol have a slower rate of growth than those fed cholesterol, this may be due to differences in the utilization of the two sterols rather than to differences in their uptake by the tissues.  相似文献

Phytophagous insects do not constantly chew their diets; most of their time is spent in a non-feeding quiescent state even though they live on or around their diets. Following starvation, phytophagous insect larvae exhibit enhanced foraging behaviors such as nibbling and walking similar to the sequential behavior that occurs prior to each meal. Although extensive physiological studies have revealed regularly occurring feeding behaviors in phytophagous insects, little has been elucidated regarding the mechanism at the molecular level. Here, we report identification and characterization of a novel 62-amino acid peptide, designated as hemolymph major anionic peptide (HemaP), from the hemolymph of Bombyx mori larvae that induces foraging behaviors. The endogenous HemaP levels are significantly increased by diet deprivation, whereas refeeding after starvation returns them to basal levels. In larvae fed ad libitum, hemolymph HemaP levels fluctuate according to the feeding cycle, indicating that locomotor-associated feeding behaviors of B. mori larvae are initiated when HemaP levels exceed an unidentified threshold. Furthermore, administration of exogenous HemaP mimics the starvation-experienced state by affecting dopamine levels in the suboesophageal ganglion, which coordinates neck and mandible movements. These data strongly suggest that fluctuation of hemolymph HemaP levels modulates the regularly occurring feeding-motivated behavior in B. mori by triggering feeding initiation.  相似文献

The effect of starvation on carbohydrate metabolism in the last instar larvae of the silkworm Bombyx mori was examined. Trehalose concentration in the hemolymph increased slightly during the first 6 h of starvation and decreased thereafter, whereas glucose concentration decreased rapidly immediately after diet deprivation. Starvation-induced hypertrehalosemia was completely inhibited by neck ligation, suggesting that starvation stimulates the release of a hypertrehalosemic factor(s) from the head. The percentage of active glycogen phosphorylase in the fat body increased within 3 h of starvation and its glycogen content decreased gradually. These observations suggest that production of trehalose from glycogen is enhanced in starved larvae. However, hypertrehalosemia during starvation cannot be explained by the increased supply of trehalose into hemolymph alone, as similar changes in phosphorylase activity and glycogen content in the fat body were observed in neck-ligated larvae, in which hemolymph trehalose concentration did not increase but decreased gradually. When injected into larvae, trehalose disappeared from hemolymph at a rate about 40% lower in starved larvae than neck-ligated larvae. The hemolymph lipid concentration increased during starvation, suggesting that an increased supply of lipids to tissues suppresses the consumption of hemolymph trehalose and this is an important factor in hypertrehalosemia.  相似文献

The passage of immunoglobulins in the blood meal into hemolymph prompts development of vaccines against internal antigens of ticks, but little is known about kinetics and specificity of the immunoglobulin uptake. We used capillary feeding of adult Amblyomma americanum hard ticks to introduce compounds into the midgut and then examined the hemolymph after various times for their presence and concentration. Immunoglobulins of different sources, albumin, choramphenicol acetyltransferase, inulin, and mannitol were labeled with (125)I, (14)C, or biotin. With the exception of the carbohydrate inulin, all the compounds entered the hemolymph of tick during capillary feeding. The small molecule mannitol had the highest rate of entry at 9% after 6 h. Among proteins, the entry of immunoglobulin G (IgG) of different species into the hemolymph was greater at 6% after 6 h than for the smaller proteins albumin or choramphenicol acetyltransferase at 1 and 3%, respectively. The entry of denatured IgG was equal to that of nondenatured protein. There was no evidence of degradation of the IgG or of its binding to cells once it entered the hemolymph. A monoclonal IgG antibody labeled with biotin entered the hemolymph and retained its ability to bind to its specific antigen in an immunoassay. Although different proteins entered the hemolymph after capillary feeding, there was evidence of a specific mechanism for immunoglobulin uptake.  相似文献

Anti-vitellin IgG directed againstDermacentor variabilis egg vitellin was used in sodium dodecyl sulfate polyacrylamide (SDS-PAGE) gradient gel immunoblots to detect the presence of vitellin and its precursor, vitellogenin, in the organs of feeding adults and in the immature stages of this tick. Vitellin polypeptides were found in the egg, larvae, nymph, and in the unfed adult stages of both sexes. Vitellin polypeptides were first detected in the ovary of mated females during the rapid-engorgement feeding, period. These polypeptides were also present in the ovaries of ovipositing females, unmated females fed for extended periods, and fed unmated females that were detached from the host and held for 12 h before dissection. The same anti-vitellin antibody was used in immunoblots to monitor the appearance of vitellogenin in the organs and hemolymph of female ticks. Immunoreactive peptides of vitellogenin were found in the fat body, midgut, and hemolymph of pre-rapid-engorging mated and unmated females. These polypeptides were not found in fed males nor in Malpighian tubes of feeding or ovipositing females Our data supported the following conclusions: 1) presence of immunoreactive vitellogenin in the adult female fat body, hemolymph, and midgut was, dependent upon feeding; 2) in mated feeding females, we could not detect the uptake of vitellogenin by the ovary until rapid engorgement; 3) in unmated females, vitellogenesis did not, begin unless prolonged feeding occurred; and 4) during the early developmental stages of this tick, vitellin served as an embryonic nutrient reserve and as a reserve against starvation between feedings.  相似文献

Abstract: The effect of crude methanolic extracts of Adhatoda vasica leaves on the feeding and performance of Spodoptera littoralis larvae was investigated in the laboratory. Feeding on fresh leaves resulted in 100% mortality of larvae after 26 days of unsubstantial growth. The extract exhibited strong antifeedant and toxic activity against the larvae when applied either on leaf discs or incorporated into artificial diet. Under choice conditions the antifeedant index calculated over 72 h for neonate larvae increased significantly (from 71.5 ± 3.2 to 92.1 ± 4.2) as the concentration of extract in the treated diet increased from 200 to 1000 ppm. Consumption by the sixth instar larvae of leaf discs dipped in 0.01, 0.1 and 0.2% extract solutions was significantly lower than consumption of control discs in both choice and no-choice tests. The latter two concentrations deterred feeding by 63.4 and 90.4%, respectively, under choice conditions, while only the 0.2% extract solution deterred feeding by 56.8% in the no-choice test. Toxicity of the extracts was manifested by a high mortality, reduced growth rates, and low weight gain by larvae fed on diets containing 200–2000 ppm of the extract. No larvae survived to pupation under the latter concentration. The time to pupation increased from 15.8 ± 0.4 to 37.9 ± 4.1 days as the extract concentration in diet increased from 0 to 1000 ppm. When fed to the fifth instar larvae, the crude extract significantly reduced consumption, growth, utilization of ingested and digested food, and approximate digestibility. The consumption-dependent growth efficiency of animals fed on extract-free diet was significantly higher than the growth efficiency of animals fed on extract-containing diets, suggesting both antifeedant and toxic activities of the extract.  相似文献

The quantity of free and esterified sterols in the whole body, intestine, hemolymph, fat body, and frass of 6th-instar larvae of H. zea, fed cholesterol or cholestanol, was measured in order to determine if there was a difference in the utilization of these two molecules. The principal sterol in the tissues of the larvae was cholestanol or cholesterol, when they were fed diet containing these two molecules, respectively; there was little, if any, metabolism of dietary cholestanol to cholesterol. There was little or no difference in the amount of total sterol in the whole body, tissues, or frass of larvae fed the two different diets, indicating that the absence of a Δ5-bond in cholestanol does not prevent the uptake or distribution of this sterol to various tissues. However, the relative percentage of steryl ester was significantly higher in prepupae reared on a diet containing cholestanol instead of cholesterol (6–7-, 4-, 13-, 4-, and 2-fold increase, for the whole body, intestine, hemolymph, fat body, and frass, respectively). The average percentage of total sterol that was esterified in the tissues was greater in the fat body (10.8 ± 15.4 and 44.2 ± 12.3%, respectively, for larvae fed cholesterol and cholestanol) than in the hemolymph (0.5 ± 0.1 and 6.3 ± 0.8%) and intestine (1.2 ± 0.1 and 4.7 ± 1.1%). The percentage of sterol that was esterified in the frass of larvae was large (26.9 ± 3.7 and 48.2 ± 0.5%, respectively, for larvae fed cholesterol and cholestanol). Therefore, the fact that larvae of H. zea fed cholestanol, instead of cholesterol, contain this saturated molecule as their principal tissue sterol and preferentially esterify it may explain, at least in part, why their rate of growth on cholestanol is slower than on cholesterol.  相似文献

【目的】明确桃蛀螟Conogethes punctiferalis幼虫取食Cry1Ab蛋白后体内CYP6AE76的过表达及对Cry1Ab蛋白有解毒作用。【方法】分析桃蛀螟CYP6AE76序列特征;利用RT-qPCR检测CYP6AE76在不同发育阶段(1-5龄幼虫)和4龄幼虫不同组织(头、中肠、血淋巴和脂肪体)以及4龄幼虫取食含有Cry1Ab蛋白(LC50=1.08 ng/cm2)的人工饲料3 d存活的幼虫中肠和血淋巴中的表达量;利用RNAi饲喂法沉默桃蛀螟4 龄幼虫CYP6AE76后检测中肠中CYP6AE76的表达量,并统计120 h后幼虫体重并计算幼虫存活率;利用RNAi饲喂法沉默桃蛀螟初孵幼虫CYP6AE76后饲喂含1.08 ng/cm2 Cry1Ab蛋白的饲料,7 d后统计幼虫体重并计算幼虫存活率。【结果】桃蛀螟CYP6AE76基因开放阅读框长1 572 bp,编码524个氨基酸,分子量约为60.34 kD,属于CYP6家族基因。发育表达谱结果表明, CYP6AE76基因在桃蛀螟整个幼虫阶段均有表达且在1龄幼虫期表达量最高,随着幼虫龄期增大而表达量降低;组织表达谱结果表明,CYP6AE76在4龄幼虫中肠中表达量最高。4龄幼虫取食含有Cry1Ab蛋白(1.08 ng/cm2)的人工饲料后,CYP6AE76在中肠和血淋巴中的表达量相比对照显著上调。通过RNAi沉默CYP6AE76后,桃蛀螟初孵幼虫再取食含有Cry1Ab蛋白的人工饲料后体重显著降低。【结论】CYP6AE76可能参与对桃蛀螟幼虫摄入的Cry1Ab蛋白的解毒。  相似文献

A 171,000 M(r )polypeptide of Podisus maculiventris (Say) (Heteroptera: Pentatomidae) that constituted 16% of the protein in eggs also constituted up to 25% of the protein in hemolymph of fed females. It was identified as the major or sole apoprotein of vitellogenin. Eggs contained major polypeptides of 171, 106, and 51 kDa. The hemolymph polypeptide was identified with a polypeptide (vitellin) in egg extracts by comparing molecular weights, specificity of occurrence in fed females, and immunological reactivities. Females, starved for 5 days after eclosion to assure complete previtellogenic development, produced vitellogenin within a day after feeding on larval Galleria mellonella, and within 4 days after feeding on an artificial diet. Appearance of vitellogenin preceded ovarian growth by 2-3 days. Two monoclonal antibodies raised against egg proteins of P. maculiventris were selected for their strong reaction against egg extract and female hemolymph and null reaction against male hemolymph. Only one 170-kDa band in egg and hemolymph reacted with the antibodies on denaturing Western blots. These monoclonal antibodies are being used to develop an enzyme-linked immunosorbent assay (ELISA) to quantitate reproductive response of females to diets of differing quality.  相似文献

Hybridoma cell lines secreting monoclonal antibodies (MAbs) specific to a 31,000 molecular weight viral protein or a 31,000 molecular weight polyhedrin protein of Lymantria dispar nuclear polyhedrosis virus (LdNPV) were developed. The two polypeptides were shown to be different by comparing their amino acid compositions. Immuno-electron microscopy was used to verify specific binding of the MAbs to their respective targets. Specific MAbs were used to develop an ELISA procedure to monitor the development of LdNPV virus and polyhedrin in vivo. Results indicated that in hemolymph of larvae fed 10(6) polyhedral inclusion bodies, the concentration of virus began to increase 16 h after inoculation and continued to increase for the next 5 days. By 36 h, the concentration of polyhedrin increased and was maintained at a high level in the later stages of infection. One-third of this group of infected larvae survived the infection. In these individuals, the concentrations of virus and polyhedrin declined to a low level 5 days after infection. This suggests the presence of a host mechanism for clearing the virus from the hemolymph.  相似文献

Scolus secretions and hemolymph of caterpillars of Saturnia pyri fed with two different foodplants (Crataegis monogyna, Prunus spinosa) were chemically analyzed and their chemical similarities determined. The secondary-compound patterns obtained for the two body fluids showed no significant differences when compared between the two groups of alternatively fed last-instar larvae. Thus, the composition of these fluids of full-grown caterpillars is not influenced by the larval diet. However, younger larvae on P. spinosa revealed a diversity of compounds differing significantly from that of larger caterpillars fed with either C. monogyna (both body fluids) or P. spinosa (hemolymph only). This indicates that, on the one hand, the hemolymph composition is adapted to the changing physiological requirements of the given instars whereas, on the other hand, the defensive mixtures remain unaltered in the late larval instars due to a constant spectrum of potential enemies.  相似文献

The effect of Hg treatment on hemolymph and tissue ferritin in the wax moth Galleria mellonella was examined by western blotting. At 48 h after feeding HgCl2, the level of hemolymph ferritin increased approximately 1.8‐fold over that of control insects that were not fed HgCl2, while there was a small increase in tissue ferritin. Time series experiments showed that tissue ferritin had a typically saturated pattern, with a maximum level from 24 to 72 h, although it decreased 12 h following HgCl2 feeding, while hemolymph ferritin first decreased but subsequently increased. Tissue ferritin in the fat body, gut and Malpighian tubules, the main tissues of ferritin expression, was upregulated over time following treatment with Hg, and in particular, tissue ferritin in the gut increased by a large amount at 12–48 h. The results suggest that in G. mellonella, the ferritin‐inducible mechanisms following treatment with HgCl2 are different for hemolymph and tissue ferritin, as are their biochemical properties.  相似文献

IAE5 (EbPXG5)、Bt菌株Bt8010、Bt8010+肠道总菌群和Bt8010+EbPXG5的人工饲料,以及分别取食无菌人工饲料与含EbPXG5上清、EbPXG5菌体破碎液、Bt8010+EbPXG5上清、Bt8010+EbPXG5菌体破碎液和Bt8010的人工饲料不同时间后的存活率,分析肠道细菌对小菜蛾Bt敏感性的影响;利用平板培养技术,测定分别取食含EbPXG5, Bt8010和Bt8010+EbPXG5人工饲料的小菜蛾3龄幼虫肠道和血淋巴中EbPXG5和Bt8010的丰度以及EbPXG5对Bt8010的体外抑制效应,分析肠道细菌对Bt8010在小菜蛾肠道中增殖以及入侵血腔的影响;利用扫描电镜观察小菜蛾3龄幼虫无菌肠道组织的内壁形态以及EbPXG5, Bt8010和EbPXG5+Bt8010分别处理的肠道组织的内壁形态,揭示肠道细菌对肠道内壁的保护功能。【结果】与取食无菌人工饲料的对照组相比,取食含肠道总菌群饲料和含EbPXG5饲料的小菜蛾3龄幼虫存活率没有显著差异,但取食含Bt8010+EbPXG5和含Bt8010+肠道总菌群饲料的3龄幼虫在24, 36, 48和60 h时存活率均显著高于取食含Bt8010人工饲料的;取食含EbPXG5上清和含EbPXG5菌体破碎液饲料的3龄幼虫存活率与对照组相比无差异,取食含Bt8010+EbPXG5上清和含Bt8010+EbPXG5菌体破碎液饲料的3龄幼虫存活率与取食含Bt8010饲料的相比也无差异。分别取食含EbPXG5和Bt8010+EbPXG5人工饲料的小菜蛾3龄幼虫肠道中EbPXG5菌株的丰度均没有显著差异,但取食含Bt8010+EbPXG5饲料24, 36和48 h的小菜蛾3龄幼虫肠道内Bt8010丰度显著低于取食含单一Bt8010饲料;血淋巴中,取食含Bt8010+EbPXG5饲料的小菜蛾,36 h和48 h的EbPXG5丰度高于取食含单一EbPXG5饲料的,同时取食含Bt8010+EbPXG5饲料的血淋巴中Bt8010丰度显著低于取食含单一Bt8010饲料的。牛津杯抑菌圈试验表明小菜蛾肠道细菌EbPXG5在体外对Bt8010菌株无抑制作用。扫描电镜观察结果发现,Bt8010会破坏小菜蛾幼虫肠道形成孔洞,同时介导Bt8010及其他细菌穿越肠道屏障进入血淋巴;肠杆菌EbPXG5能定殖于小菜蛾肠腔内壁,减弱Bt8010对肠道内壁的破坏,降低Bt8010在小菜蛾肠道和血淋巴中的丰度。【结论】肠道细菌EbPXG5在保护小菜蛾,降低其对Bt敏感性方面起一定作用,推测该菌通过竞争生态位和保护肠道内壁等方式减弱病原体的定殖和入侵,从而降低宿主对Bt的敏感性。该结果对于促进小菜蛾的生物防治和综合治理具有重要的参考意义。  相似文献

Females ofSpodoptera littoralis Boisd. (Lepidoptera: Noctuidae) with different feeding experiences during their larval development were tested for their ovipositional response to methanol extracts of larval frass and semisynthetic diets. The effect of the following frass, diet and diet component extracts was tested: (a) frass fromS. littoralis orAgrotis segetum larvae fed on a potato-based diet; (b) frass fromS. littoralis larvae fed on a wheat germ-based diet; (c) potato and wheat germ-based diets; and (d) potatoes and wheat germ. Ovipositing females without prior experience of the potato diet were deterred by extracts of: (1) larval frass from either species fed on potato diet; (2) the potato-based diet; (3) potato. Also females with experience of the potato diet during only a part of their larval development were deterred from oviposition by frass of larvae reared on the potato diet and by the diet itself. However, for females reared on the potato diet for their entire larval development, oviposition was no longer deterred by either of the three extracts listed above. Extracts of: (1) frass from larvae of either species reared on wheat germ diet: (2) the wheat germ diet; or (3) wheat germ did not significantly affect oviposition. Females with ablated antennae were still deterred by frass extracts from larvae fed on potato diet, when they had been reared on the wheat germ diet. In feeding experiments, larvae of larval stage one and of larval stage three-four reared on either of the two diets preferred to feed on the wheat germ diet. However, the preference was significantly stronger for larvae with no prior contact with the potato diet. The effect of larval experience on the loss of oviposition-deterring activity by extracts of larval frass, diets and diet components is discussed in view of induction and selection.  相似文献

This study of larval Drosophila melanogaster examined the effects of injecting the prototypical organic cation tetraethylammonium (TEA) into the hemocoel or adding TEA and/or other organic cations to the diet. Mortality, hemolymph TEA levels, and Malpighian tubule TEA secretion rates were measured. The LD50 for dietary TEA was 158.4 mM and mortality increased if competitive inhibitors of organic cation transporters were also included in the diet. Mortality increased from 24% on TEA (100 mM) alone to 83 and 67% when the diet contained both TEA and quinidine (10 mM) or cimetidine (100 mM), respectively. TEA-selective microelectrode measurements indicated that hemolymph TEA concentration was approximately 3% of that in the diet for larvae maintained on TEA-enriched diet for 24 h. Malpighian tubules isolated from larvae exposed to dietary TEA excreted more TEA than did tubules from controls fed a TEA-free diet. However, the rate of decline of hemolymph TEA concentration following ingestion or injection of TEA into the hemocoel was greater than that explicable by rates of active transport by the gut and Malpighian tubules (MTs). We propose that TEA concentrations in the hemolymph are reduced not only by active transport across the MTs and gut, but also by diffusion into the gut. The latter pathway is particularly important when larvae previously maintained upon TEA-enriched diet are transferred to a TEA-free diet. The ingestion of TEA-free food not only clears the gut lumen, but also creates a TEA-free compartment into which TEA may passively diffuse from the hemolymph.  相似文献

The release of lipophorin and total protein was examined from the fat body of nondiapause and diapause larvae of the southwestern corn borer, Diatraea grandiosella, incubated in vitro in Grace's medium. The characteristics of the released lipophorin were compared to those of the high-density lipophorin present in the hemolymph of nondiapause and diapause larvae. Over a 4 h incubation period, the fat body of nondiapause larvae released about 1.5 times more total protein and 2 times more lipophorin per mg dry weight than did that of diapause larvae. Lipophorin isolated from the medium in which fat bodies of nondiapause and diapause larvae had been incubated and from the plasma of nondiapause and diapause larvae had similar mean densities of 1.115, 1.112, 1.117 and 1.119 g/ml, respectively. Although the lipid classes detected in lipophorin isolated from the fat body incubation medium and hemolymph were identical, more polar lipids and less diacylglycerol were associated with lipophorin isolated from fat body incubation medium then were associated with lipophorin isolated from the hemolymph. Sterols accounted for about 11% of the total lipids of lipophorin isolated from the fat body incubation medium, whereas they accounted for about 20% of the total lipids of lipophorin from hemolymph. We conclude that the fat body of feeding nondiapause larvae and nonfeeding diapause larvae releases high-density lipophorin.  相似文献
