Gibberellic acid from the fungi has been widely used in agriculture. In this study, more than 20 fungal isolates were screened and Paecilomyces sp. ZB shown to produce more gibberellic acid than other fungal isolates. Cow dung was used as low cost substrate for gibberellic acid production in solid state fermentation (SSF). Carbon, nitrogen and ionic sources stimulated gibberellic acid production in SSF. Lactose emerged as the significant carbon source supporting more gibberellic acid production (731 µg/g). Among the nitrogen sources, glycine appeared to influence the production of more gibberellic acid (803 µg/g). The process parameters were optimized to enhance gibberellic acid production using a two-level full factorial design and response surface methodology. The amount of gibberellic acid production was influenced mainly by moisture and pH of the substrate. Gibberellic acid production was 1312 µg/g under the optimized conditions and the predicted response was 1339 µg/g. The gibberellic acid yield increased twofolds after medium optimization. The extracted gibberellic acid was sprayed on the growing Mung bean plant and it stimulated the growth of the plant effectively. To conclude, cow dung is a new alternative to produce gibberellic acid in SSF.  相似文献

Effects of the application of gibberellic acid on sprouting of tubers or rhizomes were tested in seven species of the genus Dioscorea that are native to the temperate regions of East Asia. The lowest concentrations for significant inhibition of sprouting in these species varied from 0.1–1 µM Application of gibberellic acid at 100 µM inhibited sprouting for more than 500 days at 20 °C. Some responses to the application of gibberellic acid differed between species and between sections of the genus. In D. japonica, the application of gibberellic acid inhibited sprouting of tubers and bulbils while it promoted seed germination.  相似文献

A system for subculture of spinach (Spinacia oleracea L.) roots was established, and differences in regeneration; namely, embryogenic competence, among individuals of the `Nippon' cultivar were examined. Root tissues, excised from seedlings, were grown on medium without growth regulators and subcultured on the same medium and then on medium that contained 10 M naphthaleneacetic acid and 0.1 M gibberellic acid to induce callus formation. Calli were transferred to medium without growth regulators. All explants formed calli. However, the frequency of embryo formation varied among lines. Higher concentrations of gibberellic acid in the callus-induction medium had limited effects on somatic embryogenesis from poorly embryogenic lines. These results indicate that inherent factors are important for somatic embryogenesis in spinach and that the root subculture system is useful for identifying strongly regenerative genotypes among individuals of a single cultivar.  相似文献

The effects of gibberellic acid on the longevity and elongation of variously aged, debladed petioles of Coleus blumei were studied, with particular reference to the hypotheses 1) that auxin increases longevity by increasing growth, and 2) that gibberellic acid acts by increasing the endogenous levels of auxin.

Gibberellic acid, substituted for the leaf blades, significantly decreased longevity of younger petioles, as measured by days or hours to abscission. Gibberellic acid also decreased the longevity resulting from 0.1% indoleacetic acid. This is the opposite of the effect expected if it is increasing auxin levels in the petiole.

In its effect on elongation of younger petioles, however, gibberellic acid did act in the direction expected if it were increasing effective levels of auxin in the petiole. The elongation rate from 0.1% gibberellic acid plus 0.1% indoleacetic acid in lanolin was as large or larger than that for 1.0% indoleacetic acid.

Petioles which were 10 or more weeks old (i.e., at positions 5+ below the apical bud were not affected by 0.1% gibberellic acid in either longevity or rate of elongation, with or without 0.1% indoleacetic acid. Since 1.0% indoleacetic acid increases both longevity and elongation rate of these petioles over 0.1% indoleacetic acid, gibberellic acid is clearly not acting on older petioles as if it were increasing effective auxin levels).

Protocols were established for achieving plant regeneration from stem internode, callus, and cell suspension cultures of Solanum lycopersicoides Dun. Two accessions of S. lycopersicoides exhibited different responses as to callus formation on various media, requirement of gibberellic acid for shoot regeneration, and ability to grow in suspension culture. The optimum medium for initiation and maintenance of cell suspension cultures was Murashige and Skoog [9] medium with 15 mg l NAA. For shoot regeneration, of three cytokinins tested, zeatin was found most effective relative to number, rapidity of response and overall quality of shoots. The present work describes the changes that take place in phytohormone contents in germinating chickpea (Cicer arietinum cv. Aziziye-94) seeds in response to heavy metal stress. For this aim, endogenous abscisic acid (ABA), gibberellic acid (GA3), zeatin (Z) and zeatin riboside (ZR) contents were followed for 24, 48 and 72 h in chickpea seeds germinating at the concentrations of 0.1, 1.0 and 5.0 mM Pb or 0.1, 1.0 and 10 mM Zn. The results showed that Pb and Zn significantly delayed and impeded the germination of chickpea seeds. The negative effect of Pb on germination was higher than that of Zn. Further, Pb increased ABA and Z contents while decreased GA3 content in the germinating seeds. The high concentrations of Zn (1.0 and 10 mM) decreased contents of Z, ZR and GA3 while 0.1 mM Zn increased the content of the same hormones. The ABA content was enhanced by Zn in all concentrations used.  相似文献

Method for simultaneous measurement of gibberellic acid was applied using capillary zone electrophoresis. Gibberellic acid was identified in extracts of apical part of thallus of Chara vulgaris L. The amount of gibberellins measured on the basis of activity determined by the micro-drop bioassay (59.8 mg·kg−1; with gibberellic acid as a standard) was comparable with that estimated by capillary electrophoresis (54.9 mg·kg−1).  相似文献

Three cell lines of Taxus brevifolia Nutt. with differing growth rates were used to assess the effects of basal salt mixtures, carbohydrates, organic nitrogen additives, vitamin formulations, and plant growth regulators on callus growth. Gamborg's B5 major salts provided significantly better growth than all other salt formulations tested. The greatest biomass was obtained with 1% total carbohydrate. The best carbohydrate combination, 0.5% fructose + 0.5% sucrose, was significantly better than all other combinations of carbohydrates tested. A complex vitamin mixture was significantly better than any one previously published vitamin formulation. Greatest rates of callus growth were obtained with 4.14 M (1 mg l-1 picloram, 0.46 M (0.1 mg l-1 kinetin, and 0.38 M (0.1 mg l-1) abscisic acid or 0.29 M (0.1 mg l-1 gibberellic acid. Our final medium, TM5, is superior to published methods for the general callus culture of T. brevifolia. 筛选出赤霉酸产生菌UF456的发酵培养基配方,研究了各种碳源、氮源、碳氮比和不同微量元素对赤霉酸发酵的影响.结果表明:最佳碳源为玉米淀粉,含量为12.5%;最佳氮源为黄豆饼粉和花生饼粉,含量分别为0.3%和1.2%;微量元素Fe^(2+)、Cu^(2+)和Zn^(2+)对赤霉酸发酵具有促进作用,其中Fe^(2+)尤为重要,三种元素不同的组合对赤霉酸发酵的促进作用都具有累加效应.  相似文献

Untreated excised segments of the hypocotyls of dark grown cabbage seedlings are always systemically infected when inoculated with the conidia of the obligate parasite Peronospora parasitica (Downy mildew). Cabbage hypocotyl elongation is promoted by 10–4 M indolylacetonitrile (IAN) and this elongation is inhibited by 100 g mL–1 chloramphenicol (CAM). The fungus remains localized in 5–8 day old hypocotyl segments exposed to CAM, but this inhibition is reversed by IAN. Indol-3-acetic acid (IAA) has the same effect as IAN. Both gibberellic acid and kinetin inhibit systemic infection. Conidial spore germination is not reduced by the CAM concentration used in these experiments. Essential oils were produced by flowers of Hyacinthus orientalis L. that had been regenerated in vitro. The production of these oils was affected by the concentration of gibberellic acid and sucrose in the medium and by temperature. The highest concentration of essential oils was obtained when regenerated flowers were cultured in vitro for 3 weeks at 20 °C on Murashige and Skoog's medium that contained 30 g/l sucrose plus 1 mg/l gibberellic acid, whereas the highest amount of essential oils was obtained after a culture period of 3 weeks at 25 °C. The composition of essential oils from flowers that had been regenerated in vitro was compared with that from flowers grown in the field. Essential oils detected by gas-liquid chromatography included nine components in the case of the regenerated flowers and six and ten components in the case of stage 3 and stage 4 flowers grown in the field, respectively. There were four common components, namely, 1-hepten-3-ol, benzyl alcohol, phenethyl alcohol and cinnamyl alcohol. In the regenerated flowers, a single component, phenethyl alcohol, was a major constituent (75%), whereas two compounds, phenethyl alcohol (stage 3, 55%; stage 4, 48%) and cinnamyl alcohol (stage 3, 23%; stage 4, 29%) were the major constituents in the case of flowers grown in the field. An efficient and simple method for high frequency plant regeneration from immature cotyledons of mungbean is described. Immature cotyledons isolated from embryos, one week prior to harvest were cultured on MS medium with combinations of growth regulators such as benzyladenine (1 or 2 mg l−1), thidiazuron (0.1 or 0.5 mg l−1), gibberellic acid (0.1 mg l−1) and indole-3-acetic acid (0.1 or 0.5 mg l−1). A large number of greenish shoot primordia were initiated from the entire surface of the cotyledons in some of the growth regulators. Medium supplemented with benzyladenine (2 mg l−1) in combination with indole-3-acetic acid (0.5 mg l−1) produced the best response. On subculture to the same medium, well developed shoots were obtained. Addition of 0.5% activated charcoal to the shoot initiation medium completely inhibited initiation of shoot primordia. The shoot buds could be rooted on medium supplemented with 0.1 mg l−1 indole butyric acid and plants transferred to soil. An orthogonal experimental design L9 (3(4)) was used to investigate effects of temperature, pH, C:N ratio (glucose-C, NH4Cl-N) and concentrations of rice flour on production of gibberellic acid by Gibberella fujikuroi in 3.5 l fluidized bioreactors. The gibberellic acid production in a fluidized bioreactor could reach 3.90 g l(-1), more than 3-times greater than previously reported for submerged and solid fermentations. pH, rice flour concentration and C:N ratio were the factors that most influenced the production of gibberellic acid; pH being the most important. The response surface of gibberellic acid production to changes in pH and C:N ratio or rice flour concentration indicated that greatest production was found with a C:N ratio of 36.8 and pH 5 while the optimum concentration for rice flour was 2 g l(-1) and production increased with increased pH. The effect of temperature on the production of gibberellic acid was also significant and greatest production was at 30 degrees C.  相似文献

The effect of gibberellic acid on the secretion of proteins from barley (Hordeum vulgare L.) aleurone layers has been investigated for its suitability as a gibberellin bioassay. Concentrations from 10–4 g/ml to 100 g/ml of GA3 resulted in the release of proportionally increasing amounts of total protein. The release of proteins is not affected by indoleacetic acid and kinetin. Stimulation of Aspergillus niger in submerged culture using a commonly known precursor, mevalonic acid (MVA), was investigated in terms of growth and gibberellic acid production. An in vitro method for obtaining gingseng inflorescences directly from explants of gingseng (Panax ginseng) is reported. Isolated shoot-buds of somatic embryo-derived plantlets ginseng were used as explants and incubated in B5 medium supplemented with 1 mg l−1 benzyladenine (BA) and 1 mg l−1 gibberellic acid (GA3). About 15% of the buds flowered directly without developing vegetative organs. Cytokinin was found to be the key factor for inducing these isolated buds to proliferate and flower, but both these processes also occurred when benzyladenine (BA) was replaced by thidiazuron (TDZ). The optimal concentration of TDZ for obtaining the best ratios of bud proliferation and total flowering was 0.1 mg l−1, while the highest number of vegetative shoots was obtained in medium supplemented with 1 mg l−1 GA3 and 0.1 mg l−1 TDZ. The explant elongated abnormally in the presence of 10 mg l−1 GA3. Although a low concentration (1 mg l−1) of NAA increased the bud proliferation ratio in the medium supplemented with 0.1 mg l−1 TDZ and 1 mg l−1 GA3, a high concentration (5 mg l−1) of NAA reduced the bud proliferation ratio and inhibited the flowering.  相似文献

AIMS: To examine the production of gibberellic acid by selected morphological mutants of Gibberella fujikuroi in liquid cultures. METHODS AND RESULTS: Mutants of G. fujikuroi having different morphological characteristics were selected after UV irradiation. The production of gibberellic acid by mutants that had different hyphal lengths was examined in shake flasks in media with different concentrations of nutrients as well as different volumes of the medium. Fed-batch fermenter study was performed to evaluate the mutant Mor-25 for growth and production of gibberellic acid. The broth was analysed by high performance liquid chromatography for fusaric acid, the common mycotoxin produced by strains of Fusarium. A variety of morphological mutants having different mycelial and soluble pigmentation as well as colony morphologies were generated from G. fujikuroi upon exposure to UV radiation. A nonpigmented mutant (Car-1) was selected as intermediate parent and later, mutants Mor-1 and Mor-25 were selected based on their distinct morphology. The colonies on regeneration agar plates were small, compact and dry. In liquid medium, mutant Mor-25 grew in a micro-pelleted form and the mycelium had short, highly branched hyphae, curly at tips with thick, swollen cells. Mutant Mor-25 grew rapidly in a low-cost medium containing defatted groundnut flour, sucrose and salts. In media with higher nutrient concentrations as well as larger volumes, it produced twofold more gibberellic acid than the parent. Fusaric acid, the common mycotoxin, was absent in the fermentation broth of mutant Mor-25. The mutants have been deposited in National Collection of Industrial Microorganisms (NCIM), National Chemical Laboratory, Pune, India under following culture collection numbers (Car-1, NCIM 1323; Mor-1, NCIM 1322; and Mor-25, NCIM 1321). CONCLUSIONS: Growth of unpigmented, morphological mutants of G. fujikuroi that led to lower viscosity in fermentation broth resulted in increased production of gibberellic acid. SIGNIFICANCE AND IMPACT OF THE STUDY: The use of morphological mutants that have lower viscosity in liquid cultures for gibberellic acid production is not reported earlier. Similar mutants can be useful for other types of fungal fermentations also.  相似文献

Benzyladenine and gibberellic acid sprays (100 mg/l) onEuphorbia lathyris L. plants were absolutely successful (100% of the plants) in inducing a change in its normal decussate phyllotaxis. Benzyladenine produced a change to tetracussate, tricussate and bijugate, and gibberellic acid to spiral phyllotaxis. Benzyladenine and gibberellic acid treatments resulted in a significant increase in apex diameter (72.8% and 19.1% respectively). CCC, Ancymidol, Alar and Glyphosine did not alter decussate phyllotaxis.  相似文献

The regeneration of plants from leaf explants of a number of potato cultivars using a number of published one-, two- and three-step methods was assessed. Gibberellic acid at 10-4 Mxxx was optimal for enhancement of growth, O2 evolution, photosystem II and I and the activity of glycollate dehydrogenase of Anacystis nidulans. A stimulatory effect was observed on photosystem II. Other concentrations of gibberellic acid were inhibitory to O2 evolution and photosystem I. Syntheses of phycocyanin, phycoerythrin and -carotene were significantly enhanced after 48 h incubation with gibberellic acid at 10-3 Mxxx but the chlorophyll content began to increase 3 h after adding 10-4 Mxxx gibberellic acid.The author is with the Department of Biological Sciences, Faculty of Science, University of Science and Technology, Irbid, Jordan.  相似文献
