To determine the potential role of macrophages as regulators of the immune response, the effect of mouse peritoneal macrophages on transforming mouse spleen lymphocytes was investigated. Mitogen and antigen stimulated lymphocyte transformation, as measured by DNA synthesis, was enhanced by all concentrations of normal macrophages tested, but only by low concentrations of activated macrophages. High concentrations of activated macrophages markedly inhibited lymphocyte transformation. This inhibition occurred whether lymphocyte DNA synthesis was measured by incorporation of [3H]TdR or of 32P. Activated macrophages cultured with lymphocytes within 4 hr of being removed from the peritoneal cavity inhibited lymphocyte transformation. When activated macrophages were cultured alone for 24 or more hours before addition of lymphocytes, enhancement of transformation was noted. Once lymphocytes were exposed to activated macrophages, they could not be induced to undergo transformation in the presence of Con A. Whereas heat-killed activated macrophages, which appeared intact morphologically, lost their capacity to inhibit lymphocyte transformation, macrophages treated with mitomycin C to inhibit DNA synthesis retained this capacity. Syngeneic and allogeneic macrophages had similar inhibitory ability. Supernatants from cultures of many cell types (including normal or activated macrophages, lymphocytes, lymphocytes plus macrophages, and L cells) inhibited [3H]TdR incorporation by both mitogen stimulated lymphocytes and tumor cells. These studies demonstrate the capacity of macrophages to regulate lymphocyte transformation in vitro and suggest a role for these cells as regulators of cell-mediated immunity in vivo.  相似文献

Macrophages from strains 2/N, 13/N, and (2 × 13)F1, guinea pigs have been fractionated by velocity sedimentation and the various subpopulations used as target cells for infection by Leishmania enrietti. The data presented show: (i) that the ability of infected macrophages to support the subsequent growth and replication of the parasite varies according to the cell subpopulation examined, (ii) that different subpopulations differ in their capacity to promote lymphocyte proliferation from lymph node cells of a guinea pig which have recovered from a primary lesion, (iii) that lymphocyte proliferation depends upon presentation of leishmanial antigens in the context of products of the original I-region-coded genes present during the initial (in vivo) infection and, (iv) that in immune animals, changes occur in terms of the ability of the macrophages to promote lymphocyte proliferation, but not apparently in terms of their ability to support parasite growth in vitro.  相似文献

A primary in vitro sensitization system employing a chromium release assay was utilized to investigate reactivity of murine spleen cells toward syngeneic ultraviolet (uv) light induced fibrosarcomas. These tumors are immunologically rejected in vivo when implanted into normal syngeneic mice but grow progressivly when implanted into syngeneic mice that had previously been irradiated with subcarcinogenic levels of uv light. Following appropriate sensitization, spleen cells from both normal and uv irradiated mice are capable of developing cytotoxic lymphocytes in vitro against the uv induced tumors. It was subsequently discovered that in situ uv induced tumors all contained macrophages of host origin that became demonstrable only after enzymatic dissociation of the tumor tissue. These macrophages were immunologically active in vitro as their presence in the stimulator cell population was necessary to achieve an optimum anti-tumor cytotoxic response following in vitro sensitization. Anti-tumor reactivity generated by mixing spleen cells and tumor cells in the absence of tumor derived macrophages could be greatly enhanced by the addition of normal syngeneic peritoneal macrophages. When in vitro anti-tumor reactivity of spleen cells from normal and uv treated mice was compared under these conditions we again found no significant difference in the magnitude of the responses. In addition, the cytotoxic cells generated in response to uv induced tumors appeared to be highly cross reactive with respect to their killing potential. Cross reactive killing was observed between all uv induced tumors tested as well as with a syngeneic benz[a]pyrene (BP) induced fibrosarcoma. No cytotoxicity was observed against normal syngeneic PEC's even through these cells were shown to be susceptible to lysis by anti-H-2k effector cells. It was concluded that: (a) A significant number of host-derived macrophages are present in uv tumor tissue. (b) These macrophages are important for the in vitro generation of tumor specific cytotoxicity. (c) Spleen cells from uv treated mice are capable of recognizing and responding against uv tumor associated antigens in vitro. Cytotoxic effector cells generated in response to uv induced tumors appear to have specificity for tumor associated antigens (TAA) present on all uv tumors tested as well as a syngeneic BP induced tumor. The relationship between in vivo and in vitro reactivity against uv tumors is discussed.  相似文献

Sensitization of guinea pigs to 2,4-dinitrofluorobenzene is accompanied by increases in alpha globulins determined electrophoretically. During sensitization, lymphocyte responses were measured in vitro by mitogen induced 3H-thymidine uptake in whole blood cultures and in vivo by dermal skin reactivity. Following 5 days of dinitrofluorobenzene sensitization alpha globulins were elevated and lymphocyte transformation to phytohemagglutinin, pokeweed mitogen, and concanavalin A was significantly suppressed. When the alpha globulins returned to normal levels following sensitization, lymphocyte responses returned to pretreatment values. Antigen induced lymphocyte responsiveness was also suppressed concomitant with elevations in alpha globulins. Tuberculin sensitive guinea pigs responded poorly to PPD in vitro and in vivo during DNFB sensitization. It is suggested that increases in alpha globulins detected during the development of cellular immunity are associated with immunosuppression.  相似文献

Intratumor host cells of methylcholanthrene-induced fibrosarcoma(s) were shown to enhance the in vivo outgrowth of syngeneic homologous tumors (MCIA, Mc2A, Mc2B) but not two heterologous T-lymphomas (EL4 and TLX9) in the Winn adoptive transfer assay. This enhancing activity was not restricted only to the latent period of tumor growth but was also observed during the period of active in vivo tumor proliferation. Tumor enhancement was mediated by a population of cells adherent to nylon wool and glass and insensitive to irradiation (with 850 rads) or to treatment with anti-Thy 1.2 serum and complement. Macrophages from peritoneal exudates of normal mice, used as control host cell population, showed similar tumor-enhancing activity. These findings suggest that tumor infiltrating host cells, predominantly macrophages appear to be the cell type responsible for tumor enhancement and active promotion of tumor growth (in vivo).  相似文献

The capacity of spleen cell populations enriched for T and B lymphocytes by a physical adherence column method to respond in vitro to phytomitogens and allogeneic lymphocytes was determined. Column filtrate cells (T lymphocytes) responded well to phytohaemagglutinin- and mitomycin-C-treated allogeneic spleen cells, but poorly to pokeweed mitogen. Adherent cell populations from the column (B and some T lymphocytes) responded well to pokeweed mitogen, but poorly to phytohaemagglutinin- and mitomycin-C-treated allogeneic cells.Purified peripheral T lymphocytes prepared from normal mouse spleen by the column method reconstituted the depleted in vitro antibody response to the thymic-dependent SRBC antigen of all B lymphocyte sources tested, namely, spleen cells from congenitally athymic mice, neonatally thymectomized mice, and adult thymectomized mice which had been reconstituted with bone marrow, and a lymphocyte population prepared by incubating spleen cells with anti-θ serum and complement. When transferred with sheep erythrocytes to congenitally athymic mice, purified peripheral T cells restored the in vivo IgM and IgG responses of these animals. These results confirm that the column filtrate is a thymus derived subpopulation of cells capable of cell-mediated immunity and cooperation with B lymphocytes in humoral immunity both in vitro and in vivo.  相似文献

The in vivo susceptibility of several inbred strains of mice to the Y and CL strains of Trypanosoma cruzi was compared to the in vitro ability of spleen cells from infected mice to generate factor(s) able to activate macrophages to a trypanocidal state. Spleen cells from resistant immune mice generate higher levels of the factor(s) and do so at earlier times during infection than those of susceptible mice. The spleen cells capable of generating the in vitro factor(s) are also capable of conferring resistance upon passive transfer. Removal of immunoglobulin-bearing cells from the immune spleen cell population did not affect either transfer of protection in vivo or generation of the factor(s) in vitro. The cellular basis underlying the differences between susceptible and resistant mouse strains has not yet been determined.  相似文献

To determine whether macrophages can discriminate in an immunologically specific manner between the intracellular pathogens which they inhibit or kill, unelicited peritoneal macrophages from mice infected with either of two related but antigenically dissimilar protozoa were challenged with these protozoa in vitro. Experimental conditions were varied in an attempt to establish a state in vivo in which macrophage specificity might be demonstrated. No differences could be discerned between the ability of macrophages from three different strains of mice infected with the protozoa to kill Besnoitia and Toxoplasma. The effect of macrophages on Toxoplasma as compared with Besnoitia did not evolve or vary during development, expression, or decline of an immune response, i.e., with varying times after infection of mice as well as with varying times after treatment of mice with irradiated Toxoplasma. The route of infection could not be shown to confer specificity on macrophages, as subcutaneous and intraperitoneal inoculation of Toxoplasma did not lead to differential ability of macrophages to inhibit or kill the protozoa. The different strains of protozoa used for infection of mice did not affect the ability of peritoneal macrophages from Besnoitia- and Toxoplasma-infected mice to inhibit multiplication of or kill Besnoitia and Toxoplasma comparably in vitro. Peritoneal macrophages of mice treated with Corynebacterium parvum kill both organisms efficiently. These macrophages were employed to determine whether stimulation of macrophages by treatment of mice with a substance unrelated to the protozoa would produce activated macrophages. Uninfected mice and mice infected with either Besnoitia or Toxoplasma were challenged with varying doses of the protozoa in parallel with examination of macrophages from the same groups of mice in vitro to determine whether the presence of stimulated macrophages in the peritoneal cavity was necessary for protection against Toxoplasma and Besnoitia, and if so if their presence was sufficient for protection. Only mice with activated peritoneal macrophages were protected. However, protection was greater when the primary infection was with the same organisms used for challenge at a time when macrophages inhibited or killed both protozoa efficiently in vitro. The possible role of other effector cells, subpopulations of macrophages of different functional abilities in various sites, and antibody or other lymphocyte products acting in concert with macrophages as factors which may explain the differences observed between in vivo protection and in vitro capacity to inhibit or kill the protozoa are discussed.  相似文献

RNA from antigenically stimulated peritoneal macrophages is immunogenic in vitro1–4. The studies of Fishman and Adler5–6 suggest that the peritoneal macrophage population consists of at least three functionally distinct subpopulations. Although most peritoneal macrophages act as scavenger cells7, a second population—possibly less than one cell per 1,000—consists of cells that produce but do not necessarily secrete antibody8,9 and respond to antigen by synthesizing informational RNA. On transfer to normal lymphoid cells, this RNA elicits IgM antibody with the allotypic specificity of the macrophage donor10. A third type of macrophage gives rise to the RNA-antigen complex responsible for the in vitro synthesis of IgG antibody with the allotypic specificity of the lymphocyte donor10.  相似文献

MRL/lpr mice possess an autosomal recessive gene, lpr, which is associated with lymphoproliferation and acceleration of autoimmune disease. Lymphoproliferation has been ascribed to a single gene defect predominantly affecting the T-lymphocyte component of the immune system. MRL/++ mice do not possess the lpr autosomal recessive mutation and do not develop early lymphadenopathy. T-lymphocyte functional activity was studied in these mice using the polyclonal T-cell mitogens PHA and Con A. Our results indicated a significant suppression of the spleen and lymph node response of MRL/lpr mice to these polyclonal mitogens as compared to the MRL/++ response noted as early as 6 weeks of age. In addition, there was a progressive decline in the MRL/lpr spleen and lymph node cell mitogenic responses with increasing age. Spleen and lymph node cells from 20-week MRL/lpr mice were also relatively unresponsive in the mixed lymphocyte culture reaction as compared to cells from MRL/ ++ or BALB/c mice. The in vitro proliferative response of the MRL mice was further examined with respect to possible accessory cell modulation by both macrophages and T cells. It was found that in 20-week MRL/lpr lymph nodes a significant degree of suppression of lymphocyte proliferation could be mediated by the MRL/lpr T cell. Increased lymphocyte proliferation to a mitogenic signal could only be demonstrated in those MRL/lpr mice 3 weeks of age.  相似文献

Macrophage tumoricidal activity relies, mainly, on the release of Tumor Necrosis Factor alpha (TNFα) and/or on reactive oxygen or nitrogen intermediates. In the present work, we investigated the cytotoxic activity of resident peritoneal macrophages against L929 fibrosarcoma cell line in vitro and in vivo. Resident macrophages lysed L929 cells in a mechanism independent of TNFα and cell-to-cell contact. The cytotoxic activity was largely dependent on nitric oxide (NO) release since treatment with L-NAME (NOS inhibitor) inhibited L929 cells killing. Macrophages from mice with targeted deletion of inducible NO synthase (iNOS) together with L929 cells produced less NO and displayed lower, but still significant, tumoricidal activity. Notably, NO production and tumor lysis were abolished in co-cultures with macrophages deficient in Interferon Regulatory Factor, IRF-1. Importantly, the in vitro findings were reproduced in vivo as IRF-1 deficient animals inoculated i.p with L929 cells were extremely susceptible to tumor growth and their macrophages did not produce NO, while WT mice killed L929 tumor cells and their macrophages produced high levels of NO. Our results indicate that IRF-1 is a master regulator of bi-directional interaction between macrophages and tumor cells. Overall, IRF-1 was essential for NO production by co-cultures and macrophage tumoricidal activity in vitro as well as for the control of tumor growth in vivo.  相似文献

Peritoneal macrophages from Schistosoma mansoni-infected mice are activated both for nonspecific tumor cytotoxicity and for killing of skin-stage schistosomula in vitro. In the current study, mechanisms for induction of macrophage tumoricidal and schistosomulacidal activity have been compared. Examination of macrophages activated in vivo by BCG infection or C. parvum treatment, or in vitro by exposure to lymphokine prepared from antigen-stimulated BCG-immune spleen cells, showed that these effector functions were closely linked. Indeed, fractionation of lymphokine-rich supernatant fluids by Sephadex G-100 gel filtraction showed that activities responsible for induction of schistomula killing by inflammatory macrophages and for induction of tumoricidal activity cochromatographed as a single peak in the 50,000 MW region. Thus, development of macrophage-mediated cytotoxicity against these two extracellular (tumor cell or helminth) targets was coincident in several cell populations activated in vivo or in vitro. However, activation for tumoricidal and schistosomulacidal capacity appeared to be quantitatively dissociated in macrophages from mice with chronic schistosomiasis; those cells demonstrated low, yet significant, levels of larval killing (13 to 15 those of BCG or lymphokine-activated cells) but maximal levels of tumor cell cytotoxicity. Furthermore, cytotoxicity by peritoneal cells from S. mansoni-infected mice was not increased in vitro by exposure to lymphokine. Identification of this functional alteration in S. mansoni-activated cells may help to clarify the role of macrophages in the partial immunity against challenge infection which is demonstrated by mice with chronic primary S. mansoni infection.  相似文献

The induction of B cell unresponsiveness with hapten-modified syngeneic murine lymphoid cells (hapten-modified self, HMS) can be achieved in vivo and in vitro. Tolerance in vivo in mice required a latent period of 3 to 4 days. Moreover, B cell unresponsiveness could not be induced by HMS in athymic nude mice, although their nu/+ littermates were rendered hyporesponsive by HMS. Pretreatment of normal mice with cyclophosphamide (cyclo) prevented their susceptibility to tolerance induction by haptenated lymphoid cells. Nude mice became sensitive to HMS-induced suppression if they were first reconstituted with spleen cells from normal (but not cyclo-treated) donors.Interestingly, labeling of H-2 antigens was not necessary for tolerance induction by HMS since haptenated teratoma cells (lacking H-2) were tolerogenic in normal recipients.In contrast, suppression of the in vitro response to haptenated flagellin occurred equally well with nude, nu/+ and anti-Ly 2 + C-treated spleen cells. These data suggest that cyclo-sensitive modified self-reactive (T) cells may regulate the immune response and mediate tolerance to HMS in vivo. However, the in vitro “blockade” of B cell reactivity may be directly mediated on hapten-specific PFC precursors.  相似文献

The question addressed in this report was whether immunological processes which culminate in in vivo expression and augmentation as well as suppression of delayed effector cell activity are mirrored by events that can be quantified in vitro. For this purpose the previously characterized murine model of delayed hypersensitivity (DHS), which employs SJL/J mice immunized with aggregated human γ-globulin (AHGG) in complete Freund's adjuvant (CFA) was employed. The results indicated that lymph node cells (LNC) from cyclophosphamide (CY)-pretreated, AHGG-CFA immunized mice expressed increased proliferation in the presence of HGG and concanavalin A (Con A) but decreased LPS responsiveness compared with LNC from equivalently immunized but non-CY-treated animals. It was also found that LNC from CY-treated, AHGG-CFA immunized mice that were pretreated with aqueous HGG (aqHGG), but not aqueous bovine serum albumin (aqBSA), evidenced a markedly decreased capacity to proliferate in the presence of HGG compared with LNC from equivalent animals that were not pretreated with aqHGG. This suppressive effect was not attributable to antibody production. These findings support the conclusion that in vitro quantitation of antigen-induced proliferation by LNC from HGG-DHS mice appears to correlate with modulatory effects which are observed in in vivo expression of DHS responses.  相似文献

Rhadinoviruses establish chronic infections of clinical and economic importance. Several show respiratory transmission and cause lung pathologies. We used Murid Herpesvirus-4 (MuHV-4) to understand how rhadinovirus lung infection might work. A primary epithelial or B cell infection often is assumed. MuHV-4 targeted instead alveolar macrophages, and their depletion reduced markedly host entry. While host entry was efficient, alveolar macrophages lacked heparan - an important rhadinovirus binding target - and were infected poorly ex vivo. In situ analysis revealed that virions bound initially not to macrophages but to heparan+ type 1 alveolar epithelial cells (AECs). Although epithelial cell lines endocytose MuHV-4 readily in vitro, AECs did not. Rather bound virions were acquired by macrophages; epithelial infection occurred only later. Thus, host entry was co-operative - virion binding to epithelial cells licensed macrophage infection, and this in turn licensed AEC infection. An antibody block of epithelial cell binding failed to block host entry: opsonization provided merely another route to macrophages. By contrast an antibody block of membrane fusion was effective. Therefore co-operative infection extended viral tropism beyond the normal paradigm of a target cell infected readily in vitro; and macrophage involvement in host entry required neutralization to act down-stream of cell binding.  相似文献

Bestatin, a new competitive aminopeptidase-inhibitor of dipeptide nature, was shown to enhance markedly activation of peripheral blood lymphocytes by concanavalin A (Con A). More than 40 percent stimulation over the control (the culture with Con A only) was observed at 50 μg/ml of bestatin, and this stimulatory effect was most predominant at an early stage of lymphocyte blasto-genesis by Con A: bestatin had most effect when added to the culture simultaneously with Con A and no appreciable effect when added 44 h after Con A. The effect of bestatin on T lymphocyte activation in vitro is discussed in relation to its in vivo enhancing effect on cell-mediated immunity and a role in lymphocyte blastogenesis of some proteolytic activities possibly located at the cell surface is emphasized.  相似文献

Apoptotic cell removal (efferocytosis) is an essential process in the regulation of inflammation and tissue repair. We have shown that monocyte chemoattractant protein-1/CC chemokine ligand 2 (MCP-1/CCL2) enhances efferocytosis by alveolar macrophages in murine bacterial pneumonia. However, the mechanism by which MCP-1 exerts this effect remains to be determined. Here we explored that hypothesis that MCP-1 enhances efferocytosis through a Rac1/phosphatidylinositol 3-kinase (PI3-kinase)-dependent mechanism.We assessed phagocytosis of apoptotic cells by MCP-1 treated murine macrophages in vitro and in vivo. Rac activity in macrophages was measured using a Rac pull down assay and an ELISA based assay (GLISA). The downstream Rac1 activation pathway was studied using a specific Rac1 inhibitor and PI3-kinase inhibitor in in vitro assays.MCP-1 enhanced efferocytosis of apoptotic cells by murine alveolar macrophages (AMs), peritoneal macrophages (PMs), the J774 macrophage cell line (J774s) in vitro, and murine AMs in vivo. Rac1 activation was demonstrated in these cell lines. The effect of MCP-1 on efferocytosis was completely negated by the Rac1 inhibitor and PI3-kinase inhibitor.We demonstrated that MCP-1 enhances efferocytosis in a Rac1-PI3 kinase-dependent manner. Therefore, MCP-1-Rac1-PI3K interaction plays a critical role in resolution of acute lung inflammation.  相似文献

Enterohemorrhagic Escherichia coli (EHEC) are food-borne pathogens that can cause serious infections ranging from diarrhea to hemorrhagic colitis (HC) and hemolytic-uremic syndrome (HUS). Translocation of Shiga-toxins (Stx) from the gut lumen to underlying tissues is a decisive step in the development of the infection, but the mechanisms involved remain unclear. Many bacterial pathogens target the follicle-associated epithelium, which overlies Peyer''s patches (PPs), cross the intestinal barrier through M cells and are captured by mucosal macrophages. Here, translocation across M cells, as well as survival and proliferation of EHEC strains within THP-1 macrophages were investigated using EHEC O157:H7 reference strains, isogenic mutants, and 15 EHEC strains isolated from HC/HUS patients. We showed for the first time that E. coli O157:H7 strains are able to interact in vivo with murine PPs, to translocate ex vivo through murine ileal mucosa with PPs and across an in vitro human M cell model. EHEC strains are also able to survive and to produce Stx in macrophages, which induce cell apoptosis and Stx release. In conclusion, our results suggest that the uptake of EHEC by M cells and underlying macrophages in the PP may be a critical step in Stx translocation and release in vivo. A new model for EHEC infection in humans is proposed that could help in a fuller understanding of EHEC-associated diseases.  相似文献

The cell interactions involved in azobenzenearsonate-N-acetyl-tyrosine (ABA-tyr)-induced delayed hypersensitivity in the guinea pig were studied by in vitro blastogenesis. The ABA-sensitive lymphocyte was demonstrated to be a T lymphocyte and its presence in peritoneal exudate cells was shown to be much higher than spleen or lymph node populations. The secondary response of ABA-sensitized lymphocytes to ABA-tyr in culture is dependent on the presence of an accessory cell, with both splenic and peritoneal macrophages being equally effective. ABA coupled directly to macrophages as an immunogen induced strong responses to itself and not to ABA-tyr-pulsed macrophages or ABA-tyr in solution. The reverse was true in animals, immunized with ABA-tyr. ABA conjugated to thymocytes, L2C leukemia cells, and guinea pig erythrocytes however, did not elicit significant responses. The results obtained in animals immunized with ABA- or ABA-tyr-modified cells was similar whether or not CFA was used. The difference in specificity shown between ABA-coupled and ABA-tyr-pulsed macrophages favors a single receptor hypothesis for T-cell recognition.  相似文献
