Inhibition of PHA stimulation of hamster lymph node cells by theophylline, DBcAMP, or indomethacin or PHA stimulation of thymocytes by theophylline or DBcAMP was partially reversed by addition of 10 mM LiCl to the cultures. Addition of LiCl to Con A-stimulated lymphoid cells treated with the same reagents did not alter the inhibition. In contrast, addition of 10 mM LiCl to Con A-stimulated cultures enhanced the inhibition induced by the Na,K ATPase inhibitor, ouabain. Like LiCl, this latter inhibitor was found to be effective in modulating stimulation only if added early in the culture.These data support the hypothesis that LiCl can modulate lymphocyte responsiveness at the level of cyclic nucleotide metabolism, as exemplified by PHA stimulation, or at the level of the Na,K ATPase, exemplified by Con A stimulation. The site of involvement of Li+ ion would appear to be dependent on the biochemical nature of the stimulating signal.  相似文献

Mesophyll cells of leaf slices of bean (Phaseolus vulgaris L.) absorb six to ten times more K+ than Rb+ from 0.1 mM single chlorides of these cations. Absorption of 42K+ from 0.1 mM42KCl is much more inhibited by low concentrations of Rb2SO4 than by K2SO4. The isotherm for K+ absorption is biphasic in the range 0.1–1.1 mM, and K+ is more effective than Rb+ in causing transition from phase 1 to phase 2.  相似文献

The influence of alkali ions on the circadian leaf movements of Oxalis regnellii Mig. was investigated. Ions were given to the oscillating system via the transpiration stream of cut stalks in nutrient medium. Chloride solutions of Rb+, Cs+, Na+ and K+ were tested and the results compared to previously published LiCl-results. The period of the circadian leaf movements was unaffected by a continual addition of Na+ or K+ to the nutrient medium (at least up to 40 mM). Rb+, in the concentration of 2.5 or 5 mM, caused a shortening of the period when applied continuously. Rb+ concentrations up to 60 mM were tested. Cs+ ions caused only lengthenings of the circadian period. Cs+ concentrations up to 40 mM were tested. Cs+ resembled Li+ in producing period lengthenings, but was not as effective as Li+ when compared on a concentration basis. Toxicity of the effective ions was in the following order: Li+Cs+Rb+, Rb+ pulses (50 mM, 4 h) phase-shifted the rhythm and caused advances. A phase response curve was determined and the maximum steady state advances were of the order of 1 h. The dual effect of the Rb+ ions is discussed and is assumed to be due to two counteracting processes, exemplified by Rb+-sensitive ATPase-controlled pumping processes and protein synthesis. For comparison, the effects of Rb+ and Li+ in human depressive disorders is also discussed in relation to their influence on circadian systems. It is emphasized that Rb+ and K+ behave differently and are not interchangeable in their action on circadian systems.  相似文献

Steady state kinetics were used to examine the influence of Cd2+ both on K+ stimulation of a membrane-bound ATPase from sugar beet roots (Beta vulgaris L. cv. Monohill) and on K+(86Rb+) uptake in intact or excised beet roots. The in vitro effect of Cd2+ was studied both on a 12000–25000 g root fraction of the (Na++K++Mg2+)ATPase and on the ATPase when further purified by an aqueous polymer two-phase system. The observed data can be summarized as follows: 1) Cd2+ at high concentrations (>100 μM) inhibits the MgATPase activity in a competitive way, probably by forming a complex with ATP. 2) Cd2+ at concentrations <100 μM inhibits the specific K+ activation at both high and low affinity sites for K+. The inhibition pattern appears to be the same in the two ATPase preparations of different purity. In the presence of the substrate MgATP, and at K+ <5 mM, the inhibition by Cd2+ with respect to K+ is uncompetitive. In the presence of MgATP and K+ >10 μM, the inhibition by Cd2+ is competitive. 3) At the low concentrations of K+, Cd2+ also inhibits the 2,4-dinitrophenol(DNP)-sensitive (metabolic) K+(86Rb+) uptake uncompetitively both in excised roots and in roots of intact plants. 4) The DNP-insensitive (non metabolic) K+(86Rb+) uptake is little influenced by Cd2+. As Cd2+ inhibits the metabolic uptake of K+(86Rb+) and the K+ activation of the ATPase in the same way at low concentrations of K+, the same binding site is probably involved. Therefore, under field conditions, when the concentration of K+ is low, the presence of Cd2+ could be disadvantageous.  相似文献

Skeletal myotubes responded to passive stretch by increased amino acid uptake (as measured with [3H]α-aminoisobutyric acid), increased incorporation of amino acids into total cellular protein and myosin heavy chains, and increased accumulation of total cellular protein and myosin heavy chains. These alterations were preceded by an increase in the uptake of ouabain-sensitive rubidium-86 (86Rb+), a potassium tracer used to measure membrane sodium pump activity (Na+K+ATPase). This stretch-induced stimulation of 86Rb+ uptake resulted from a 60-70% increase in the Vmax of the Na pump with little change in the Km. [3H] ouabain binding studies showed no stretch-induced change in the number of membrane Na pumps, indicating that stretch activates the Na pumps that are already present on the cell surface. Since the stretch-induced increases in amino acid transport and amino acid incorporation into proteins were inhibited by ouabain, Na pump activation may be involved in stretch-induced cell growth of skeletal muscle cells by hypertrophy.  相似文献

Deoxyribokinase, which catalyzes the ATP-dependent phosphorylation of 2-deoxy-d-ribose to 2-deoxy-d-ribose-5-P as the first step in the inducible fermentation pathway for this sugar in Salmonella typhimurium, was purified approximately 600-fold from deoxyribose-grown cells. Apparent Km′s for 2-deoxy-d-ribose and ATP were 0.1 and 0.5 mm, respectively. The enzyme had an absolute requirement for divalent cations which was best satisfied by Mg2+. Optimal activity was obtained in the presence of 0.5 m NH4+ or Cs+. Rb+ and K+ also stimulated enzyme activity whereas Na+ and Li+ inhibited. d-Ribose and 2-deoxy-d-ribitol could replace 2-deoxy-d-ribose as phosphoryl acceptor, and several ribo- and deoxyribonucleotides could replace ATP as phosphoryl donor. Molecular weight determinations gave values of 67,800 for the native enzyme and 33,500 for the dissociated enzyme, suggesting the probable existence of two subunits of similar size.  相似文献

A membrane fraction of sugar beet roots prepared in the presence of dithiothreitol contains (Na++ K++ Mg2+) ATPase activity. This activity was studied in the presence of different concentrations of Tris and histidine. Tris was found to interact with the ATPase in the following way: (1) Tris at 50 mM increases, in the absence of Na and/or K, the activity in an uncompetitive way with respect to MgATP. (2) Concentrations of Tris > 50 mM cause inhibition in the absence of Na and K when the ratio between MgATP and Tris is relatively low. (3) Though Tris at 50 mM stimulates similarly to Na, it can not substitute for Na in the Na + K activation. (4) In the presence of Na and K, Tris acts as a competitive inhibitor. Histidine has little influence on the rate both in the presence and absence of Na and/or K.  相似文献

To understand the function and membrane origin of ionophore-stimulated ATPases, the activity of nigericin-stimulated ATPase was characterized from a low-density microsomal fraction containing sealed vesicles of autonomous tobacco (Nicotiana tabacum Linnaeous cv. Wisconsin no. 38) callus. The properties of KCl-stimulated, Mg-requiring ATPases (KCl-Mg,ATPase) were similar in the absence or presence of nigericin. Nigericin (or gramicidin) stimulation of a KCl-Mg,ATPase activity was optimum at pH 6.5 to 7.0. The enzyme was inhibited completely by N,N′-dicyclohexylcarbodiimide (10 μm), tributyltin (5 μm), and partially by vanadate (200 μm), but it was insensitive to fusicoccin and mitochondrial ATPase inhibitors, such as azide (1 mm) and oligomycin (5 μg/ml). The ATPase was more sensitive to anions than cations. Cations stimulated ATPase activity with a selectivity sequence of NH4+ > K+, Rb+, Cs+, Na+, Li+ > Tris+. Anions stimulated Mg, ATPase activity with a decreasing sequence of Cl = acetate > SO42− > benzene sulfonate > NO3. The anion stimulation was caused partly by dissipation of the electrical potential (interior positive) by permeant anions and partly by a specific ionic effect. Plant membranes had at least two classes of nigericin-stimulated ATPases: one sensitive and one insensitive to vanadate. Many of the properties of the nigericin-sensitive, salt-stimulated Mg,ATPase were similar to a vanadate-sensitive plasma membrane ATPase of plant tissues, yet other properties (anion stimulation and vanadate insensitivity) resembled those of a tonoplast ATPase. These results support the idea that nigericin-stimulated ATPases are mainly electrogenic H+ pumps originated in part from the plasma membrane and in part from other nonmitochondrial membranes, such as the tonoplast.  相似文献

Studies were conducted to characterize ATPase activity associated with purified chloroplast inner envelope preparations from spinach (Spinacea oleracea L.) plants. Comparison of free Mg2+ and Mg·ATP complex effects on ATPase activity revealed that any Mg2+ stimulation of activity was likely a function of the use of the Mg·ATP complex as a substrate by the enzyme; free Mg2+ may be inhibitory. In contrast, a marked (one- to twofold) stimulation of ATPase activity was noted in the presence of K+. This stimulation had a pH optimum of approximately pH 8.0, the same pH optimum found for enzyme activity in the absence of K+. K+ stimulation of enzyme activity did not follow simple Michaelis-Menton kinetics. Rather, K+ effects were consistent with a negative cooperativity-type binding of the cation to the enzyme, with the Km increasing at increasing substrate. Of the total ATPase activity associated with the chloroplast inner envelope, the K+-stimulated component was most sensitive to the inhibitors oligomycin and vanadate. It was concluded that K+ effects on this chloroplast envelope ATPase were similar to this cation's effects on other transport ATPases (such as the plasmalemma H+-ATPase). Such ATPases are thought to be indirectly involved in active K+ uptake, which can be facilitated by ATPase-dependent generation of an electrical driving force. Thus, K+ effects on the chloroplast enzyme in vitro were found to be consistent with the hypothesized role of this envelope ATPase in facilitating active cation transport in vivo.  相似文献

A membrane fraction enriched with a magnesium-dependent, monovalent cation-stimulated ATPase was isolated from red beet (Beta vulgaris L.) storage roots by a combination of differential centrifugation, extraction with KI, and sucrose density gradient centrifugation. This fraction was distinct from endoplasmic reticulum, Golgi, mitochondrial, and possibly tonoplast membranes as determined from an analysis of marker enzymes. The ATPase activity associated with this fraction was further characterized and found to have a pH optimum of 6.5 in the presence of both Mg2+ and K+. The activity was substrate specific for ATP and had a temperature optimum near 40°C. Kinetics with Mg:ATP followed a simple Michaelis-Menten relationship. However the kinetics of K+-stimulation were complex and suggestive of negative cooperativity. When monovalent cations were present at 2.5 millimolarity, ATPase was stimulated in the sequence K+ > Rb+ > Na+ > Li+ but when the concentration was raised to 50 millimolarity, the sequence changed to K+ ≥ Na+ ≥ Rb+ > Li. The activity was not synergistically stimulated by combinations of Na+ and K+. The enzyme was insensitive to NaN3, oligomycin, ouabain, and sodium molybdate but sensitive to N,N′-dicyclohexylcarbodiimide, diethylstilbestrol, and sodium vanadate. Based on the similarity between the properties of this ATPase activity and those from other well characterized plant tissues, it has been concluded that this membrane fraction is enriched with plasma membrane vesicles.  相似文献

A microsomal (Na++ K++ Mg2+)ATPase preparation from sugar beet roots was used. The activation by simultaneous addition of Na+ and K+ at different levels was examined in terms of steady state kinetics. The observed data can be summarized in the following way: 1. The apparent affinity between the enzyme and the substrate MgATP depends on the ratio between Na+ and K+. At low Na+ concentration (below 5 mM), the apparent Km decreases with increasing concentrations of K+ (1–20 mM). At 5 mM Na+, the K+ level does not change the apparent Km, while at Na+ levels above 10 mM, the apparent Km between enzyme and substrate increases with increasing concentration of K+. 2. When the MgATP concentration is kept constant, homotropic cooperativity (concerning one type of ligand) and heterotropic cooperativity (concerning different types of ligands) exist in the activation by Na+ and K+. The Na+ binding is cooperative with different Km values and Hill coefficients (n) in the presence of low and high concentration of K+. At low Na+ level (< 5 mM). a negative cooperativity exists for Na+ (nNa < 1) which is more pronounced in the presence of high [K+]. When the concentration of Na+ is raised the negative cooperativity disappears and turns into a positive one (nNa > 1). Only K+ binding in the presence of low [Na+] shows cooperativity with a Hill coefficient that reflects changes from negative to positive homotropic cooperativity with increasing concentrations of K+ (nK < 1 → nK > 1). In the presence of [Na+] > 10 mM, the changes in nk are insignificant. 3. A model is proposed in which one or two different K sites and one or two Na sites control the catalytic activity, with multiple interactions between Na+, K+ and MgATP. 4. In the presence of Na+ (< 10 mM), K+ is probably bound to two K sites, one of which translocates K+ through the membrane by an antiport Na+/K+ mechanism. This could be connected with an elevated K+ uptake in the presence of Na+ and could therefore explain some field properties of sugar beets.  相似文献

The addition of LiCl stimulated the (Na++K+)-dependent ATPase activity of a rat brain enzyme preparation. Stimulation was greatest in high Na+/low K+ media and at low Mg. ATP concentrations. Apparent affinities for Li+ were estimated at the α-sites (moderate-affinity sites for K+ demonstrable in terms of activation of the associated K+-dependent phosphatase reaction), at the β-sites (high-affinity sites for K+ demonstrable in terms of activation of the overall ATPase reaction), and at the Na+ sites for activation. The relative efficacy of Li+ was estimated in terms of the apparent maximal velocity of the phosphatase and ATPase reactions when Li+ was substituted for K+, and also in terms of the relative effect of Li+ on the apparent KM for Mg· ATP. With these data, and previously determined values for the apparent affinities of K+ and Na+ at these same sites, quantitative kinetic models for the stimulation were examined. A composite model is required in which Li+ stimulates by relieving inhibition due to K+ and Na+ (i) by competing with K+ for the α-sites on the enzyme through which K+ decreases the apparent affinity for Mg·ATP and (ii) by competing with Na+ at low-affinity inhibitory sites, which may represent the external sites at which Na+ is discharged by the membrane NA+/K+ pump that this enzyme represents. Both these sites of action for Li+ would thus lie, in vivo, on the cell exterior.  相似文献

Addition of LiCl (1–25 mM) to serum-free cultures of MHA hamster thymocytes, lymph node cells, or splenocytes stimulated with concanavalin A had a biphasic effect on [3H]thymidine incorporation. These concentrations of LiCl enhanced stimulation of [3H]thymidine incorporation by suboptimal levels of concanavalin A but inhibited stimulation of optimal and supraoptimal concentrations of concanavalin A. This effect was specific for Li+ since it was not observed when similar concentrations of Na+, K+, or Mg2+ were added to cultures stimulated by concanavalin A. The inhibitory effect of LiCl on concanavalin A stimulation was not reversed by addition of Na+, Ca2+, Mg2+, or Ca2+ + Mg2+ to the cultures. Significant reversal of LiCl inhibition of stimulation was observed when KCl was added to the cultures. However none of the ions tested blocked the Li-induced enhancement of [3H]thymidine incorporation in the presence of suboptimal concentrations of concanavalin A.  相似文献

In the presence of an iso-osmotic concentration (0.4 M) of LiCl, the exit of cellular K+ and concomitant entry of Li+ in the marine bacterium, Vibrio alginolyticus, were enhanced by an increase in the medium pH, with an optimum at about pH 9.6. In addition to alkaline pH, the K+ exit in the NaCl medium required the presence of a weak base such as diethanolamine, ethanolamine or methylamine, which is permeable to the membrane in its unprotonated form. No net entry of Na+ was detected in this case and the amine accumulated in exchange for K+. The K+ exit observed at alkaline pH could be explained by the function of a K+/H+ antiporter. Once the cells were loaded with the amine, their exposure to the NaCl medium in the absence of loaded amine induced the entry of Na+. In RbCl or CsCl medium, fast entry of Rb+ or Cs+ and exit of K+ were observed at neutral pH (7.5), and the rate of K+ exit increased with the medium pH. From these results, we established a simple method for the replcement of cellular cations with a desired cation (Li+, Na+, K+, Rb+ or Cs+). The present method was found to be applicable also to Escherichia coli.  相似文献

Maximum ATPase activities in the cell wall fraction of English ryegrass (Lolium perenne L.) roots were stimulated by foru discrete millimole ratios of (Na++ K+); 40:0, 35:5, 5:35, and 0:40. The optimal pH for stimlation was found to be 6.5. Contrary to data in the literature, Mg2+ inhibited all stimulatory ratios of (Na++ K+) when plants were cultured on an adequate nutrient solution. When grown on a dilute solution, Mg2+ enhanced (Na++ K+)-stimulated ATPase activity in this membrane preparation. The single optimal combined concentration of (Na++ K+) for all stimulatory ratios was 40 MM. The ratios of (Na++ K+) which stimulated ATPase activity in the cell wall fraction varied with position along the root axis such that all rarely existed simultaneously nor did any exist in the terminal millimetre of the root. Both cell wall and microsomal fractions showed stimulation by (Na++ K+) at all the above ratios indicating the possible presence of plasma membrane fragments in both fractions. Only the 35:5 ratio was stimulations were found in the supernatant. Implications of ion-stimulated ATPase involvement in ion transport were drawn from the appearance of ATPase activity at a 40:0 ratio of (Na++ K+) and the disappearance of stimulations at 35:5, 5:35, and 0:40 ratios when plants were moved from a strong (35 mM total concentration) to a dilute (0.75 mM) nutrient solution.  相似文献

Internal Cs+, Na+, Li+, and, to a lesser degree, Rb+ interfere with outward current through the K pores in voltage clamped squid axons. Addition of 100 mM NaF to the perfusion medium cuts outward current for large depolarizations about in half, and causes negative conductance over a range of membrane voltages. For example, suddenly reducing membrane potential from +100 to +60 mv increases the magnitude of the outward current. Internal Cs+ and, to a small extent, Li+, also cause negative conductance. Na+ ions permeate at least 17 times less well through the K pores than K+, and Cs+ does not permeate measurably. The results strongly suggest that K pores have a wide and not very selective inner mouth, which accepts K+, Na+, Li+, Cs+, tetraethylammonium ion (TEA+), and other ions. The diameter of the mouth must be at least 8 A, which is the diameter of a TEA+ ion. K+ ions in the mouths probably have full hydration shells. The remainder of the pore is postulated to be 2.6–3.0 A in diameter, large enough for K+ and Rb+ but too small for Cs+ and TEA+. We postulate that Na+ ions do not enter the narrower part of the pore because they are too small to fit well in the coordination cages provided by the pore as replacements for the water molecules surrounding an ion.  相似文献

Between pH 4–10, basal ATPase activity, measured in the absence of mineral ions, was 10 to 100 times higher in the final cytoplasmic supernatant from potato tuber homogenates than in the membraneous fractions (purified plasmalemma, purified mitochondria and microsomes). The soluble ATPase was slightly inhibited, whereas the membrane-bound ATPases were all stimulated by Mg2+ ions. A further stimulation by Na+ or K+ ions was only observed in purified plasmalemma or mitochondria, at alkaline pH (7.5–9.5). At a fixed (Na++ K+) concentrations (80 mM), this last stimulation was much greater in purified mitochondria (350%) than in plasmalemma (33%); it also increased with (Na++ K+) concentrations up to 200 mM in mitochondria whereas, in plasmalemma, it was roughly constant for monovalent ion concentrations between 20 and 200 mM. General properties of the plasma membrane-bound ATPase have been determined, i.e. substrate specificity, activity variations with quantity of substrate, temperature, pH, etc. Divalent cations stimulated strongly the ATPase in the following order: Mn2+ > Mg2+ > Ca2+. The maximum ATP hydrolysis velocity for that part of ATPase activity which is strictly dependent on Mg2+ ions was 3.85 μmol × mg?1 protein × h?1. This plasma membrane ATPase was not sensitive to ouabaïn or to oligomycin.  相似文献

At concentrations of 10-?3M, Li+ inhibits the recovery of solute uptake capacity of Acer pseudoplatanus L. cell suspension cultures after gas-shock (i.e. after rapid exchange of the atmosphere in the culture flasks for ambient air). It also reduces solute uptake capacity of cells having already attained high rates of uptake during recovery from gas-shock. The effects of Li+ are much greater in cells which have been cultivated in 7 mM K+ solution than in cells cultivated with higher K+ levels (19 mM). Increasing K+ concentration during recovery reverses the effect of 10–3M Li+ and, with sufficiently high concentrations of K+ (≥ 10-?2M) during recovery, the solute uptake capacity of the fully recovered cells can even become greater than that of the control, at least for the low values of substrate concentration (here sulphate 10-?5M). Since Li+ does not affect the time course of solute uptake measured over 15–20 min, it is thought that it interacts with the synthesis and turnover of the solute uptake machinery of the Acer pseudoplatanus cells. Thermodynamic analysis of the flux data also supports the hypothesis that Li+ inhibits the biosynthesis of specific sites of solute permeation, but it does not rule out the possibility that K+ interferes rather on the forces acting on the transport of the considered solutes than on the catalytic structures of permeation.  相似文献

The (K+,Mg2+)-ATPase was partially purified from a plasma membrane fraction from corn roots (WF9 × Mol7) and stored in liquid N2 without loss of activity. Specific activity was increased 4-fold over that of the plasma membrane fraction. ATPase activity resembled that of the plasma membrane fraction with certain alterations in cation sensitivity. The enzyme required a divalent cation for activity (Co2+ > Mg2+ > Mn2+ > Zn2+ > Ca2+) when assayed at 3 millimolar ATP and 3 millimolar divalent cation at pH 6.3. When assayed in the presence of 3 millimolar Mg2+, the enzyme was further activated by monovalent cations (K+, NH4+, Rb+ Na+, Cs+, Li+). The pH optima were 6.5 and 6.3 in the absence and presence of 50 millimolar KCl, respectively. The enzyme showed simple Michaelis-Menten kinetics for the substrate ATP-Mg, with a Km of 1.3 millimolar in the absence and 0.7 millimolar in the presence of 50 millimolar KCl. Stimulation by K+ approached simple Michaelis-Menten kinetics, with a Km of approximately 4 millimolar KCl. ATPase activity was inhibited by sodium orthovanadate. Half-maximal inhibition was at 150 and 35 micromolar in the absence and presence of 50 millimolar KCl. The enzyme required the substrate ATP. The rate of hydrolysis of other substrates, except UDP, IDP, and GDP, was less than 20% of ATP hydrolysis. Nucleoside diphosphatase activity was less than 30% of ATPase activity, was not inhibited by vanadate, was not stimulated by K+, and preferred Mn2+ to Mg2+. The results demonstrate that the (K+,Mg2+)-ATPase can be clearly distinguished from nonspecific phosphohydrolase and nucleoside diphosphatase activities of plasma membrane fractions prepared from corn roots.  相似文献
