Transformation of tobacco leaf discs with the ‘cytokinin’ipt gene yielded several transgenic callus tissue lines, respectiveto the kind of ipt construction present in the A. tumefacienscointegrates. Those calli containing an active ipt gene wereable to grow hormone-autotrophically and showed an increasedendogenous cytokinin level in comparison with controls. Analysisof endogenous IAA level did not allow any quantitative correlationwith the cytokinin content. However, a minimal level of auxinseems to be necessary to obtain hormone-autotrophic growth.Exogenously supplied NAA significantly reduced the endogenouscytokinin content without modifying growth characteristics. The varying chlorophyll content in the different callus lineselicited the study of the ultrastructure of the plastids. Thecontrols contained small plastids, often filled with starchor accumulated vesicles that did not allow observation of theinternal membrane system. The ‘Pssu-ipt’ line, havinga higher cytokinin content, showed plastids with an internalmembrane system consisting of stroma and grana thylakoids, butthis structure was lost during subculture. The seed coats of Digitalis purpurea L. cv. Gloxiniaflora werecultured for indigenous fungi. Alternaria alternata (Fries)was identified as the sole fungus on seeds freshly harvestedfrom unopened capsules, whereas A. alternata, Rhizopus arrhizus(Fischer), Penicillium sp. and other fungi appeared on storedseeds. The appearance of fungi in seed cultures was seasonal,being more frequent in winter and early spring than in summerand autumn. Alternaria grew well on autoclaved seeds, on dehiscentseed coats, or on seed coats separated from the embryos of ungerminatedseeds. Rhizopus did not grow on these but grew weakly on theculture medium from viable seeds. A. alternata appears to functionas a degradation agent for the seed coat subsequent to germination.Neither fungus was found to be essential to germination of Digitalisseeds. Bioassay of the culture medium from germinating seedsshowed that a fungistat effective against both Alternaria andRhizopus is produced coincident with germination. Based on chromatographicanalysis, the fungistat appears to be a cardenolide. Flowers of Digitalis purpurea L. cv. Foxy, take just over aweek to develop from the smallest buds and open. Both the anthersand the stigma are closed when the flower opens, the anterioranthers dehisce 2 days after flower opening and the posterioranthers a day later. The stigma opens 2 days later still andthe corolla remains attached for a further 2 days. The maximum size of the corolla is reached soon after openingand then remains constant. The fresh weight increases throughoutthe attached life of the corolla, whereas the dry weight reachesa maximum at flower opening and decreases during the remainderof the corolla's attached life. The content of antho-cyaninvaries greatly from spike to spike. In some spikes the maximumcontent is at flower opening, in others it is much later, butin all cases the loss of anthocyanin with ageing is very smalland the corollas are abcissed without visible wilting or fading.Maximum protein, RNA and glucose contents occur at, or soonafter, flower opening and the levels decrease in the corollasas they age. Fructose reaches a maximum at the same time asglucose but does not decrease as rapidly, so that fructose becomesthe major reducing sugar in the older corollas. Sucrose cannotbe detected in any flowers. Ethylene production increases steadilywith flower age, but the maximum production is recorded fromflowers which have recently ab-scissed the corolla regardlessof whether this occurs before detachment from the spike or withinthe collection tube.  相似文献

The plastids of young dark-grown bean leaves, exposed to periodiclight are agranal, devoid of chlorophyll b and contain primarythylakoids and chlorophyll a. Transfer of these plants to continuousillumination results in synthesis of new chlorophyll a, chlorophyllb and grana. This study was done in order to study whether andhow the grana are formed from preexisting primary thylakoids.14C--aminolevulinic acid was used to label the chlorophyll aof the primary thylakoids, and its fate was studied after transferof the plants to continuous light. It was found that chlorophyll b and grana become 14C-labelled.The total radioactivity of chlorophyll b per bean increasedwith the parallel decrease of that of chlorophyll a. All subchloroplastfractions, obtained after digitonin disruption of chloroplasts,contained chlorophyll a of equal specific radioactivity. Thespecific radioactivity of chlorophyll b was lower than thatof chlorophyll a, and, in addition, it was lower in the granathan in the stroma lamellae fraction. The data suggest that chlorophyll b is formed from chlorophylla; the grana are formed by stacking of preexisting primary thylakoids;chlorophyll b is synthesized faster in the grana than the stromalamellae; the newly formed chlorophyll a molecules are distributedat random throughout the developing photosynthetic membraneand not on specific growing sites. Regreening of senescent Nicotiana leaves. II. Redifferentiation of plastids   总被引:4,自引:0,他引:4  
Single senescent leaves attached to decapitated shoots of Nicotiana rustica L. regreened, especially when treated with cytokinin. Regreening caused an increase in leaf thickness, due to cell expansion. Senescent leaf plastids (gerontoplasts) were smaller than green chloroplasts, with degenerated membrane systems and stroma, and larger plastoglobuli. At advanced senescence, micrographs showed disintegrating gerontoplasts, reduced numbers of plastids were counted, and regreening became variable. The redevelopment of grana and stroma in regreening plastids was accelerated by cytokinin. All plastids in regreening leaves were identifiable as redifferentiating gerontoplasts because of their content of plastoglobuli and starch. Immunogold labelling showed significant association of POR with etioplasts in cotyledons, but with mature plastids in regreening leaves. No proplastids or dividing chloroplasts were observed in regreening leaves. Plastids numbers declined during senescence and did not increase again during regreening. Seeds of lettuce (Lactuca saliva L. cv. Grand Rapids) were incubatedin water or in gibberellin A3 (GA). Some ultrastructural differencesbetween the cotyledons of 145 hour-old seedlings resulting fromthese two treatments are reported. The cotyledons of GA-treatedseedlings contained many plastids with poorly developed grana,while the control cotyledons contained only well-developed plastids.The frequency of grana in developed chloroplasts was also reducedby the hormone treatment. Effects of GA on the nucleus includeda decrease in heterochromatin and an increase in nucleolar volume.After GA treatment, the nucleolar matrix had a largely fibrillarappearance in contrast to the predominantly granular structureof the control nucleoli. Ultrastructural changes are discussedin relation to the GA-induced variations in the biochemicalcomponents previously reported.  相似文献

The proteins from both grana and stroma lamellae of maize (Zea mays) mesophyll plastids and from maize bundle sheath plastid membranes have been compared by electrophoresis in sodium dodecyl sulfate-polyacrylamide gels using a discontinuous buffer system. Peptide differences between grana and stroma lamellae were essentially quantitative and not qualitative. Bundle sheath plastid membrane peptides more closely resembled those of the ultrastructurally similar stroma lamellae. However, bundle sheath membranes contained several peptides not apparent in the stroma lamellae.  相似文献

The cytochrome bf complex was isolated from spinach thylakoids,and also from separated grana and stroma lamellae vesicles,by a procedure involving NaBr washing, detergent treatment andcentrifugation in sucrose gradients. The resulting complex fromall three types of membranes, were almost completely devoidof chlorophyll and carotenoids. The complexes have kinase activitytowards histone III-S and contain a 64 kDa protein claimed tobe a kinase. Electrophoretic analyses indicate that the complexesare in dimeric form and composed of six polypeptides with molecularmasses of 34/33, 23, 20, 17, 12 and 4 kDa. The complexes containtwo moles cytochrome b6 per mole cytochrome f and one mole RieskeFeS. The 17 kDa and 4 kDa polypeptides are the so called subunit4 and 5 respectively. The 12 kDa protein was identified as plastocyaninby immunoblotting. Plastocyanin and the 4 kDa protein were presentin the cytochrome bf complex even after a second repeated sucrosedensity gradient centrifugation. The sucrose gradient sedimentation pattern was different forthe grana and stroma lamellae complexes. The complex from thestroma lamellae arrives at a higher density than the grana complex.Furthermore, the gradient centrifugation diagram of the stromalamellae consists of one main peak while the diagram of thegrana complex shows two peaks. There is significantly more plastocyaninand 4 kDa protein in the bf complex isolated from stroma lamellaethan from grana. In addition there is a 15 kDa protein in thecomplex isolated from the grana vesicles. Immunoblot analysisafter crosslinking indicated that the 4 kDa protein and theplastocyanin are associated in the cytochrome bf complex. Theoxidoreductase activity is higher (about 50%) in the cytochromebf complex from the grana than from the stroma lamellae fraction.We suggest that a difference in composition of the cytochromebf complex between the two membranes might be important in theregulation of cyclic and non cyclic electron flow. 1Present address: Department of Plant Physiology II, Universityof Warsaw, 00 927 Warsaw, Poland  相似文献

The floral nectary of the foxglove (Digitalis purpureaL.), locatedat the base of the ovary, was examined by: scanning electronmicroscopy; quantitative bright-field microscopy via computer-aided3-D reconstruction from serial sections; morphometric procedures;transmission electron microscopy and measurement of nectar effluxunder different experimental conditions. Time-lapse video recordingvia a microscope with incident light clearly showed that thenectar escaped from the apertures of modified stomata. The volumeflux via individual stomatal apertures was 0.31±0.1 nlmin-1; therefore only a fraction of the total number of stomataper nectary (115±8) would be sufficient to dischargethe amount of nectar reported in previous publications. Thestomatal apertures are continuous with intercellular spacestraversing the small-celled nectariferous tissue. The latteris vascularized only by phloem, whose termini consists of rowsof slender cells. These sieve-like cells are surrounded by moreor less isodiametrical sheath cells with dimensions similarto the secretory cells. The flowers of Digitalis purpurea respond to pollination by rapid corolla abscission without any loss of corolla turgor, nor any significant loss of corolla constituents, relative to the corollas of unpollinated flowers of a similar age. The corollas of unpollinated flowers too eventually abscise, 6 d after the stigma opens, however, they do so with only a minimal loss of fresh weight or corolla constituents. Pollination causes an increase in ethylene production detectable within 1 h. Increased ethylene production occurs initially only from the upper portion of the style, later from the lower portion, and lastly, between 23 and 48 h after pollination, from the ovary plus calyx. The pollination response can be induced by exogenous ethylene, the degree of weakening of the corolla abscission zone being dependent upon the concentration and duration of the exposure period and on the stage of flower development. The regulation of ethylene biosynthesis and its involvement in the control of pollination-induced corolla abscission are discussed.  相似文献

The effects of different drying methods on desiccation toleranceand longevity in seeds of foxglove (Digitalis purpurea L.) wereassessed from just prior to mass maturity (when seeds have attainedmaximum dry weight), and at intervals during the post-abscissionphase of development. Tolerance of drying under seed conservationconditions (15% relative humidity, RH, and 15 °C), was acquiredclose to mass maturity at 36 d after flowering (DAF). Increasesin desiccation tolerance were induced when drying was delayedfor 4 d by placing seeds in a near-saturated atmosphere (approx.100% RH), or if seeds were pre-dried for 7 d at either approx.32% or approx. 73% RH. Irrespective of the drying treatment, seed longevity increasedthroughout the sampling period, i.e. beyond the point of massmaturity and throughout the post-abscission phase, up to thepoint of incipient natural dispersal. At each developmentalstage, delayed drying or pre-drying led to an increase in seedlongevity under controlled ageing conditions compared with seedsdried directly under seed conservation conditions. Increasesin longevity were apparent as increases in the estimates forthe intercept of transformed seed survival curves (Ki) and forthe standard deviation of the normal distribution of seed lifespans,and also in the mean time to death of individuals in storage,consistent with a continuation of ripening events. The differentiation of chromoplasts and amyloplasts has beenfollowed in developing petals of Ranunculus acris, R. ficariaand R. repens from small bud stages through anthesis. Theseare typical of Ranunculus species with glossy yellow petals.The plastids in the adaxial epidermis of the glossy part developmany globules which enlarge and coalesce with the eventual completebreakdown of plastid structure. At anthesis only a few cytoplasmicremnants remain in the dispersed pigment in these cells whichpartially collapse with folding of the anticlinal walls. Inthe non-glossy proximal part of the petal the adaxial epidermalcells have more convex outer walls and the chromoplast developmentdoes not progress beyond a stage found in the glossy regionjust before the petal tips emerge from the bud: they have largeperipheral globules with irregular tubular lamellae betweenthem and in the central stroma. In the abaxial epidermis theplastids produce small globules and retain some grana; somehave small starch grains and differ little from the plastidsin the mesophyll. The starch layer beneath the adaxial epidermisin the glossy region is formed from a single embryonic celllayer. Amyloplasts differentiate rapidly in these cells andall plastid structure has disappeared before the flower budopens leaving the sloping palisade cells packed with starchgrains. Developing capsules of foxglove (Digitalis purpurea L.) weredetached at 4-d intervals between 12 and 28 d after flowering(DAF) and attached to canes within a natural foxglove standsuch that they were experiencing field conditions identicalto those experienced by normally developing, on-plant capsules.Seeds were subsequently harvested at 4-d intervals until 40total d after flowering (tDAF). Capsule detachment resultedin the cessation of dry matter accumulation; the mean dry weightof seeds from 12, 16, 20, 24, and 28 DAF-detached capsules was21, 32, 51, 60, and 79% respectively, of the mean dry weightof seeds during the post-abscission phase of normal, on-plantdevelopment. Nonetheless, seeds from detached capsules acquiredthe ability to germinate at harvest and tolerance to dryingunder seed conservation conditions (15% relative humidity and15 °C). The capability to withstand storage also arose followingcapsule detachment. Seed longevity increased the longer theperiod of detachment but, in the earlier-detached capsules (12,16, and 20 DAF) longevity subsequently declined. Only seedsfrom later detached capsules (24 and 28 DAF) acquired longevitieswhich were comparable with seeds from on-plant capsules, however,no seeds from detached capsules were as long lived as seedsfrom on-plant capsules harvested at 40 DAF. Structural and immunochemical studies were used to determinethe photosynthetic potential of the dodder (Cuscuta pentagona)chloroplast. Ultrastructural studies revealed that thylakoidmembranes of pre-parasitic phase Cuscuta pentagona are almostall organized into long, overlapping grana stacks of mainlytwo to five thylakoids with little space between adjacent stacks.Immunoblots reveal chloroplast proteins associated with PSIand II, as well as cytochrome f and plastocyanin. Stromal extractscontained immmunologically-detectable RuBisCO and phosphoribulokinase.Cytochemical localizations of the oxidizing side of PSI showedproduct localization on the lumen side of the thylakoid. Immunocytochemicallocalizations of RuBisCO reveal exclusive labeling in the stroma,whereas antibodies to the PSII proteins, light-harvesting Chla/b complex and the oxygen-evolving complex of PSII, are concentratedover the thylakoids. A limited capacity for CO2 fixation wasfound in seedlings by monitoring CO2 exchange rates in the presenceand absence of atrazine. These data indicate that the chloroplastfrom this species of dodder contains a number of the proteinsrequired for a successful fixation of CO2 and the proteins inthe thylakoids are organized much like other higher plants,with the exception of the large percentage of the thylakoidsorganized into grana. Summary The plastid ontogenesis inOlea europaea tapetum has been studied. Tapetum plastids start their development as proplastids and differentiate into elaioplasts. At the end of their development the tapetal cells degenerate and are substituted by roundish lipidic masses which will later form an exine coating (Pollenkitt). During their ontogenesis the plastids are characteristically associated with membrane outlines of mostly smooth ER, which appear to be correlated with lipid accumulation inside the plastids.  相似文献

The development of the dimorphic chloroplasts of Zea mays L. in adult foliage leaves is described, and a method of correlating ultrastructural stages by means of leaf chlorophyll is presented. In addition, the developmental changes in chlorophyll a/b ratio are discussed. Both the mesophyll and the bundle sheath plastids contain small grana at the earliest stages of plastid development. As the plastids enlarge, the mesophyll grana stacks increase in both length of the appressed membrane and in the number of thylakoids per granum. Initially, the grana stacks in the bundle sheath plastids also enlarge, but as the plastids approach full size, most of the membrane appression is lost. However, the remaining areas of appression in the bundle sheath plastids show an increase in the number of thylakoids in each small granum.  相似文献

The leaves of Acer negundo L. var. odessanum (H. Rothe), if permanently exposed to strong sunlight, do not green, but remain yellow and finally become bleached. In yellow leaves the plastids contain single thylakoids and no grana. In plastids of bleached leaves, however, only vesicles are present. The concentration of chlorophylls and photosynthetic activity are much lower in those leaves than in the green ones. If the illumination is reduced (e.g. by shading) both the yellow and the bleached leaves become greenish, and even fully green after a few days at a sufficiently low light intensity. The plastids of yellow-green leaves contain small grana. In dark green leaves the thylakoid system of the chloroplasts is normally developed forming true grana, regardless of whether the leaves were originally green, or became green by shading the yellow or bleached ones. Their pigment concentration and photosynthetic activity are also normal. If green leaves are exposed to sunlight they do not yellow or bleach. During a 3-week period the structure of the thylakoid system did not perceivably change, with the exception that large plastoglobules formed in the stroma.  相似文献

Pieces of callus obtained from seedlings of Digitalis purpureawere grown on solid Murashige-Skoog's medium supplemented with1 mg liter–1 BA and 0.1 mg liter–1 IAA or NAA, withor without phenobarbital (40 mg liter–1). The replacementof the natural auxin IAA by the synthetic auxin NAA increasedcallus growth and inhibited organogenesis, whereas the additionof phenobarbital had the opposite effect. Morphometric measurementsrevealed a high ratio of vacuole to cytoplasm (v/v) in calluscells. This ratio was affected by the different treatments inthe same way as the fresh weight. The activity of mitochondrialcytochrome P450scc (the enzyme that provides the precursor,pregnenolone, for the biosynthesis of cardenolide in foxgloveplants) was detected in the relevant fraction of callus grownunder all experimental conditions, and its activity was increasedby the addition of phenobarbital. The different treatments testedincreased the cardenolide content and quantifiable amounts ofdigitoxin were detected in all callus tissues. It is of specialinterest that phenobarbital added to the culture medium increasedthe accumulation of digitoxin. The mechanism affecting the developmentand production of cardenolide in callus tissues of D. purpureaby phenobarbital and the replacement of IAA by NAA is discussed.
