兔肝细胞膜高密度脂蛋白受体酶联免疫检测法的研究   总被引:6,自引:0,他引:6  
用辣根过氧化物酶标记羊抗人apoAI-IgG,建立了检测兔肝细胞膜HDL受体的酶联免疫吸附检测法.测定时, HDL结合量按抗体-配体-抗配体抗体酶交联物反应制作的标准曲线确定;膜蛋白非特异吸附则用与酶交联的抗体来源相同的同种动物血浆HDL平行抑制试验消除.实验测得正常家兔肝细胞膜HDL受体Kd值为7.17±1.18 mg/L,Bmax值为(622.5±146.1)mg/g(n=7).  相似文献

为寻求一种可替代人肝细胞研究载脂蛋白(apolipoprotein, apo)CⅢ受体的生理模型,并探讨apoCⅢ受体的生理功能及其在内源性高甘油三酯血症发病机制中的作用,首先以125I标记的人apoCⅢ为配体,利用放射性配体结合分析法观察了人肝癌细胞系HepG2细胞是否有apoCⅢ受体存在.结果证实HepG2细胞上存在高亲和力的、可饱和的、特异的apoCⅢ受体结合位点,其受体的亲和力(Kd)和apoCⅢ受体结合容量(Bmax)分别为(9.53±1.03)×10-9 mol/L和(3.28±0.31) μg/g.随后又分别研究了胰岛素及胰高血糖素对HepG2细胞apoCⅢ受体功能的影响,结果表明:胰岛素可使HepG2细胞apoCⅢ受体结合容量(Bmax)显著增加,但对受体的亲和力(Kd)无影响;胰高血糖素可使HepG2细胞apoCⅢ受体亲和力显著下降(即Kd值升高),对受体的结合容量无影响.提示人肝apoCⅢ受体的功能可能受胰岛素及胰高血糖素的不同调节.  相似文献

高胆固醇饲料喂养造成的动脉粥样硬化(As) 模型家兔通过静脉注射人血浆HDL 制剂, 观察HDL 对As家兔肝细胞膜LDL受体活性的影响. 结果发现, 摄取高胆固醇饲料的As 家兔, 其肝细胞膜LDL 受体 Kd 值虽无明显变化但Bmax 值显著减小( P< 0-01 , 与正常对照组比较) ; 注射HDL 制剂后, As 家兔肝细胞膜LDL受体Kd 值仍无明显改变, 但Bmax 值却显著回升( P< 0-01 , 与高脂组比较) . 表明人血浆HDL 具有增加As 家兔肝细胞膜LDL 受体活性的作用.  相似文献

用稳态荧光探针标记技术动态观察内毒素休克大鼠肝亚线粒体质子跨膜转运的变化.发现,休克5 h ATP、NADH和琥珀酸钠所致的9-氨基-6-氯-2-甲氧基吖啶(ACMA)最大荧光淬灭值(ΔAmax)显著低于对照组(P<0.05)、最大荧光淬灭时间(TΔAmax)、半数荧光淬灭时间(T1/2ΔAmax)非常显著延长(P<0.01),肝线粒体质子跨膜转运能力下降;膜脂分子烃链和膜脂深层次流动性下降;线粒体膜PLA2活性增加;血浆脂质过氧化产物MDA和线粒体MDA含量均显著增加.可能膜脂质过氧化和磷脂酶A2的水解是引起内毒素休克肝线粒体质子转运功能改变的重要因素.  相似文献

用[3H]-spiperone作为特异配体,与人的淋巴细胞膜上多巴胺受体进行结合反应,在淋巴细胞浓度1×106个/ml,反应温度37℃,反应时间60min条件下,此结合反应具有低容量和高亲和力的特点。Kd=8.87nmol/L, Bmax=181fmol/l×106个淋巴细胞。用单点法分析,34例正常人特异结合平均值为139(fmol/1×106LC)。此法用血量少重复性优,从而适用于临床研究。  相似文献

为了探讨胆囊收缩素(CCK)受体在中枢神经系统中的信号传递机制,观察了CCK8和CCKA受体拮抗剂L-364,718、CCKB受体拮抗剂L-365,260对大鼠大脑皮质钙调素(CaM)活性的影响.结果表明:a.CCK8对CaM活性的影响具有时间依赖性,15 min达到最高点后逐渐下降;b.CCK8在10-12~10-7 mol/L范围内可刺激CaM活性的增加,超过10-7 mol/L后,逐渐趋于饱和.c.CCKA受体拮抗剂L-364,718、CCKB受体拮抗剂L-365,260均可抑制10-7 mol/L CCK8引起的CaM活性的增加,但两者IC50相差40倍,L-365,260在较低浓度时即能明显拮抗CCK8引起的CaM活性变化.研究结果提示CCK8可能通过CCKB受体引起CaM活性变化,而CaM可能是CCKB受体的重要信号传递机制之一.  相似文献

探讨了马肝醇脱氢酶(HLADH)催化三甲基硅乙酮及其碳结构类似物不对称还原反应动力学.结果表明,在酶浓度低于150 mg/L时,底物浓度与反应初速度的关系符合米氏动力学方程;HLADH催化三甲基硅乙酮不对称还原反应的KmvmaxEa分别为2.67 mmol/L、0.118 mmol/(L·min·mg)和37 kJ/mol, 其碳结构类似物的相应值分别为3.56 mmol/L、0.084 mmol/(L·min·mg)和61 kJ/mol.  相似文献

检测以维甲酸(RA), 1, 25-二羟基维生素D3(1,25(OH)2VD3)诱导2d、佛波酯(PMA)诱导6h的人大肠癌细胞CCL229的蛋白激酶C(PKC)及其抑制剂活性.结果显示:诱导后PKC总活性升高(P<0.05); RA, 1, 25(OH)2VD诱导引起细胞质PKC活性增加,PMA诱导后细胞膜PKC比率(细胞膜活性/总活性)显著升高(P<0.01);诱导后PKC抑制剂活性均降低,其中1, 25 (OH)2VD3组与对照组有显著差异(P<0.05);提示PMA引起PKC从细胞质向细胞膜转移,不同药物诱导后PKC及其抑制剂活性出现不同的相对均衡关系.  相似文献

Background Elevated plasma homocysteine and amyloid β (Aβ) have been associated with Alzheimer’s disease (AD). We investigated the cross-sectional association between these biomarkers. Methods We used linear regression to relate plasma homocysteine and Aβ adjusting for age, gender, creatinine, APOE-ε4, and ethnic group in 327 persons aged 78 ± 6.6 years. Results Plasma homocysteine correlated with age, serum creatinine, plasma Aβ40 and Aβ42, and was inversely correlated with serum vitamin B12, and folate. Aβ42, but not Aβ40, was related to later development of dementia. Homocysteine was related to higher Aβ40 levels (coefficient = 2.0; P < 0.0001) and this association was attenuated after adjustment for creatinine (coefficient = 1.0; P < 0.0001). The crude association between homocysteine and Aβ42 was weaker (coefficient = 0.5; P = 0.01) and became non-significant after adjustment for creatinine (coefficient = 0.4; P = 0.06). These associations were unrelated to ethnicity, the presence of APOE-ε4 or dementia. Analyses by quartiles of homocysteine showed that these association were driven primarily by the fourth quartile. Conclusions Plasma homocysteine is directly related to Aβ40. The association with Aβ42 is not significant. These results seem to indicate that homocysteine is related to aging but not specifically to AD. Special issue dedicated to John P. Blass.  相似文献

This paper investigates the design of four different configurations of plasma reflectarrays. The results provide insight into different radiation characteristics of plasma reflectarrays. The proposed unit cell consists of a cubic glass box filled with argon gas energized with applied AC voltage. The reflectarray reflection coefficient phase variation is achieved by varying the plasma frequency of the energized gas. Four plasma reflectarrays for satellite applications at 12 GHz are proposed (centre feed centre beam, centre feed offset beam, offset feed centre beam, and finally, offset feed offset beam). The finite integration technique is used to analyse the plasma reflectarray.  相似文献

Fibronectin (FN) is a glycoprotein (disulfite-bonded dimer of 200 to 220 Kd submits) found in a soluble form in blood (concentration 250--500 microg/ml), it can be removed from it by cryoprecipitation and affinity chromatography on gelatin or heparin-agarose. It is also found in an insoluble fibrillar form as a component of connective tissue matrix like collagen, proteoglycans... FN fundamentally forms molecular complexes with collagen, fibrinogen or fibrin, heparin, activated factor XIII, bacteria, cellular membranes..., these various proteins binding with now well known functional "domains" on subunits. Thus FN mediates adhesion of cells to cells as well to biomaterials or tissue, cell migration and chemotactic activity, tissue stromal organization... The transformed cultured cells in presence of oncogen virus loose ability to secrete FN which contribute to their invasive tendency. FN also interacts with hemostatic and fibrinolytic systems, as component of the subendothelium (secreted, like Willbrand factor, by endothelial cells) and of platelet alpha-granules released by stimulated platelets. FN could then provoke platelet spreading on the subendothelium surface after collagen-platelet adhesion, triggered by Willebrand factor, has happened. FN is a part of the fibrinous clot. It participates in anchorage of the clot to subendothelium and mediates its colonisation by fibroblasts, first step to wound reparation. Lastly FN probably has an important role in organism defence. It acts as a non-immunological opsonin, promoting phagocytosis by RES macrophages of bacteria, cellular or fibrin fragments, immune complexes... present in blood. Plasmatic FN concentration is strongly decreased in several ill patients following major trauma, extensive burns, shock, sepsis, with or not evidence of DIVC, of respiratory distress... SABA and various other authors have obtained good results after injections of FN (as cryoprecipitates or concentrated fractions). It is yet necessary to confirm therapeutic role of FN.  相似文献

The proteomic analysis of plasma samples represents a challenge as a result of the presence of highly abundant proteins such as albumin. To enable the detection of biomarkers, which are commonly low-abundance proteins, in complex blood fluids, it is necessary to remove high-abundance proteins efficiently. Moreover, there is a range of about 10 orders of magnitude for the abundance of different protein species in serum. Here, we describe for the first time a study of reptilian albumin depletion using resins usually used in mammalian plasma depletion procedures. We performed the depletion of albumin from Bothrops jaraca plasma using the HiTrap Blue high-performance column (GE Healthcare Life Sciences, Piscataway, NJ, USA) and the kit Albumin & IgG Depletion SpinTrap column (GE Healthcare Life Sciences). In addition, proteomic approaches were used to analyze reptilian plasma. Our results showed that B. jararaca albumin bound to both columns, but those interactions were not enough to remove a large amount of albumin to reach an enrichment of low-abundance proteins. Although the depletion techniques used in this work were not the best to remove B. jararaca plasma albumin, our present work highlights the similarity between B. jararaca and mammalian albumin, contributing to the knowledge of comparative hemostatic proteins.  相似文献
