Abstract Potassium (86Rb) influx from 200 mmol m ?3 KCl into dark grown, decapitated maize seedlings 6 d old) was stimulated by nitrate pretreatment. The stimulus was clearly evident by 6h exposure to nitrate and required 12–24 h for maximal expression. Decay of the nitrate-stimulated potassium influx was more than 50% complete within 3 h after transfer to nitrogen-free solutions. The stimulation of potassium influx was entirely accounted for by an increase in the influx component that was resistant to inhibition by presence of 200 mmol m?3 ambient ammonium. In contrast, the component of potassium influx that was sensitive to inhibition by ambient ammonium was unaffected by nitrate pretreatment. Exposure to the glutamine synthetase inhibitor L-methionine-dl-sulphoximine (MSX) during nitrate pretreatment stimulated the resistant component but the sensitive component was nearly eliminated. Pretreatment with ammonium increased the resistant component of potassium influx within 3 h, i.e. before it was increased by nitrate pretreatment, but the sensitive component was concomitantly restricted. The latter recovered partially during extended pretreatment with ammonium. The data indicate that the resistant component responded positively to increases in tissue ammonium concentrations whereas the sensitive component was unaffected by tissue ammonium except at concentrations in excess of 10μmol g?1. Ammonium influx was also stimulated by nitrate pretreatment and to a greater extent than potassium influx. Presence of MSX with nitrate during pretreatment resulted in a further stimulation in ammonium influx. The parallel increases in root ammonium concentrations with the two pretreatments imply that part of the increase in ammonium influx was a consequnce of increased counter-transport with endogenous ammonium.  相似文献

Rapid, Reversible Inhibition of Nitrate Influx in Barley by Ammonium   总被引:18,自引:2,他引:16  
The rate of influx of nitrate into the roots of intact barleyplants was measured over a period of 3–5 min from externalnitrate concentrations of 1–150 mmol m–3, using13N-labelled nitrate as tracer. Ammonium at external concentrationsof 0.005–50 mol m–3 inhibited nitrate influx ina manner which did not conform to a simple kinetic model butincreased approximately as the logarithm of the ammonium concentration.At any particular ammonium concentration, inhibition of nitrateinflux reached its full extent within 3 min of the ammoniumbeing supplied and was not made more severe by up to 17 minpre-treatment with ammonium. On removing the external ammonium,nitrate influx returned to its original rate within about 3min. Potassium at 0.005–50 mol m–3 did not reproducethe rapid effect of ammonium on nitrate influx. Net uptake of nitrate also decreased when ammonium was supplied,over a similar timescale and to a similar extent as nitrateinflux. The decrease in nitrate influx caused by ammonium wassufficient to account for the observed reduction in net uptake,without necessitating any acceleration of nitrate efflux. Depolarization of tomato leaf cells by oligogalacturonide elicitors   总被引:5,自引:0,他引:5  
The electrical potential difference (Em) across the plasma membrane of tomato leaf mesophyll cells consists of a cyanide-sensitive component, presumably produced by an H+-ATPase, and a cyanide-insensitive component. Variation of Em between different batches of tissue is mainly caused by variation in the cyanide-sensitive component. Oligogalacturonide elicitors that induce the synthesis of proteinase inhibitors in tomato seedlings depolarize the Em of tomato leaf mesophyll cells. This depolarization closely resembles that caused by cyanide: they are of similar magnitude and vary in a similar manner with variation in the initial Em of different batches of tissue. Treatments with cyanide and with the elicitors have similar effects on the small depolarization caused by KCl at 10 mol m?3. The results suggest that the elicitors depolarize Em by inhibiting the plasma membrane H+-ATPase, but that the detailed mechanism of inhibition by the elicitors is different from that caused by cyanide.  相似文献

Abstract Salt-stimulated ATPase activity in membrane preparations obtained from roots of Atriplex nummularia Lindl. at pH 5 was not suscep-tible to inhibition by KC1 or NaCl up to 450 mol m-3 but showed a broad peak of activity between 150 and 300 mol m?3. At pH 8 stimulation occurred at 50 mol m?3 but concentrations above 100 mol m?3 depressed activity below the level of the MgATPase activity. By contrast, preparations from roots of Pisum sativum L. at pH 5 showed maximal stimulation at 25 to 50 mol m?3 of NaCl or KC1; concentrations higher than 150 mol m?3 depressed activity below that of MgATPase activity. At pH 8 maximal stimulation was observed at 5 to 10 mol m?3 NaCl or KC1 while the threshold for inhibition was reduced to 15 mol m?3. With increasing salt concentrations the pH profiles for NaCl stimulation of Atriplex ATPase activity (expressed as the difference between treatment and control) showed a progressive displacement of the apparent optimum towards lower pH. The shift was not apparent when stimulation was expressed as a percentage of MgATPase activity. This shift may be accounted for if NaCl stimulated the monovalent salt-activated ATPase activity but simultaneously inhibited MgATPase activity.  相似文献

In isolated Elodea densa leaves, the relationships between H+ extrusion (-ΔH+), K+ fluxes and membrane potential (Em) were investigated for two different conditions of activation of the ATP-dependent H+ pump. The ‘basal condition’ (darkness, no pump activator present) was characterized by low values of-ΔH+ and K+ uptake (ΔK+), wide variability of the ?ΔH+/ΔK+ ratio, relatively low membrane polarization and Em values more positive than EK for external K+ concentrations (|K+]o of up to 2mol m?3. A net K+ uptake was seen already at [K+]o below 1 mol m?3, suggesting that K+ influx in this condition was a thermodynamically uphill process involving an active mechanism. When the H+ pump was stimulated by fusicoccin (FC), by cytosol acidification, or by light (the ‘high polarization condition’), K+ influx largely dominated K+ and C? efflux, and the ?ΔH+/ΔK+ ratio approached unity. In the range 50 mmol m?3?5 mol m?3 [K+]0, Em was consistently more negative than EK. The curve of K+ influx at [K+]0 ranging from 50 to 5000mmol m?3 fitted a monophasic, hyperbolic curve, with an apparent half saturation value = 0–2 mol m?3. Increasing |K+]0 progressively depolarized Em, counteracting the strong hyperpolarizing effect of FC. The effects of K+ in depolarizing Em were well correlated with the effects on both K+ influx and ?ΔH+, suggesting a cause-effect chain: K+0 influx → depolarization → activation of H+ extrusion. Cs+ competitively inhibited K+ influx much more strongly in the ‘high polarization’ than in the ‘basal’ condition (50% inhibition at [Cs+]/[K+]0 ratios of 1:14 and 1:2, respectively) thus confirming the involvement of different K+ uptake systems in the two conditions. These results suggest that in E. densa leaves two distinct modes of interactions rule the relationships between H+ pump, membrane polarization and K+ transport. Abstract Roots of sterile-grown, intact 6-day-old seedlings of Ricinus communis possess at least two independent active amino acid uptake systems, one for neutral and one for basic amino acids. The kinetics of uptake of L-proline and L-arginine, which were taken as representative substrates for the two systems, are biphasic. At low concentrations (0.01–0.5 mol m?3) Michaelis -Menten kinetics prevail, changing to a linear concentration dependence at higher substrate concentrations (1–50 mol m?3). L-glutamate uptake velocity is linear over the whole substrate concentration range. For comparison the uptake kinetics of nitrate and ammonium were determined as well as interactions among the different nitrogen sources. The Km value for nitrate uptake was 0.4 mol m?3, and for ammonium 0.1 mol m?3. The uptake capacity for nitrate or ammonium was approximately the same as for amino acids. The interaction between the uptake systems for organic and inorganic nitrogen is small. Two hypotheses for the physiological significance of amino acid uptake by roots were considered: (i) Uptake of amino acids from the soil-determination of amino acids in soil and in soil water indicates that they might contribute 15–25% to the nitrogen nutrition of the plant. (ii) Amino acid uptake systems of root cells serve primarily as retrieval of amino acids delivered from the phloem- it was found that 14C L-glutamine, which was delivered to the cotyledon and transported to the root via the phloem, was not lost by the roots, whereas it appeared in the bathing medium if L-glutamine was applied externally to the root to compete for the uptake sites; this suggests that an apoplastic pool of amino acids in the root exists due to their efflux from the phloem.  相似文献

Potassium influx into roots of dark-grown decapitated maize seedling (Zea mays L., cv Pioneer 3369A) was examined in presence and absence of ambient ammonium and at various root potassium concentrations. Six-day old seedlings which were dependent on the endosperm reserves for their energy source were exposed to KCl (labeled with 86Rb) ranging from 5 to 200 micromolar. At both low (13 micromoles per gram fresh weight) and high (100 micromoles per gram fresh weight) root potassium concentration, isotherms indicated two potassium influx systems, one approaching saturation at 50 to 100 micromolar potassium and an additional one tentatively considered to be linear. A mixed-type inhibition by ammonium for the low-concentration saturable system was indicated by a concomitant decrease in Vmax and increase in Km. High root potassium concentration decreased Vmax but had little effect on Km of this system. The rate constant for the second quasilinear system was decreased by ambient ammonium and by high root potassium status. Transfer of high potassium roots to potassium-free solutions resulted in an increase in influx within 2 hours; by 24 hours influx significantly exceeded that of roots not previously exposed to potassium. In roots of both low and high root potassium concentrations, potassium influx was restricted progressively as ambient ammonium increased to about 100 micromolar, but there was little further inhibition as ammonium concentrations increased beyond that to 500 micromolar. The data imply that potassium influx has two components, one subject to inhibition by ambient ammonium and one relatively resistant.  相似文献

13NO3 influx into the roots and in vivo nitrate reductase activity (NRA) in the roots and leaves have been measured in trembling aspen (Populus tremuloides Michx.) and lodgepole pine (Pinus contorta Dougl.) seedlings after exposure to either 0·1 or 1·5 mol m–3 NO3 for varying periods up to 20 d. Both NO3 influx and NRA were inducible in these species and, in trembling aspen, peak induction of nitrate influx and NRA were achieved within 12 h, compared to 2–4 d for influx and 4–12 d for NRA in lodgepole pine. In trembling aspen, ≈ 30% of the total 13N absorbed during a 10 min influx period followed by 2 min of desorption was translocated to the shoot. In lodgepole pine, by contrast, translocation of 13N to the shoot was undetectable during the same time period. Root NRA as well as NO3 influx from 0·1 mol m–3 NO3 were substantially higher in trembling aspen than in lodgepole pine at all stages of NO3 exposure, i.e. during the uninduced, the peak induction, and steady-state stages. In order to examine whether the lower rates of NO3 influx and NRA were related to proportionately fewer young (unsuberized) roots in lodgepole pine, we determined these parameters in young and old (suberized) roots of this species separately. Induction of influx and NRA were initially greater in young roots but at steady-state there were only minor differences between the young and the old roots. However, even the elevated initial rates in the young roots of lodgepole pine were substantially lower than those of aspen. In pine, influx at 1·5 mol m–3 NO3 was ~ 6-fold higher than at 0·1 mol m–3 NO3 and appeared to be mostly via a constitutive system. By contrast, in aspen, steady-state influxes at 0·1 and 1·5 mol m–3 were not significantly different, being similar to the rate attained by pine at only the higher [NO3]. In aspen, leaf NRA was ~ 2-fold higher than that of roots. In lodgepole pine NRA of the needles was below the detection limit. These results show that trembling aspen seedlings are better adapted for NO3 acquisition and utilization than lodgepole pine seedlings.  相似文献

The effect of ambient ammonium (0.5 millimolar [14NH4]2SO4) added to a nutrient solution containing 1.0 millimolar K15NO3, 99 atom per cent 15N, upon [15N]nitrate assimilation and utilization of previously accumulated [14N]nitrate was investigated. Corn seedlings, 5-day-old dark-grown decapitated (experiment I) and 10-day-old light-grown intact (experiment II), which had previously been grown on K14NO3 nutrient solution, were used. In both experiments, the presence of ambient ammonium decreased [15N]nitrate influx (20% after 6 hours) without significantly affecting the efflux of previously accumulated [14N]nitrate. In experiment I, relative reduction of [15N]nitrate (reduction as a percentage of influx) was inhibited more than was [15N]nitrate influx. Nevertheless, in experiment I, where all reduction could be assigned to the root system, the absolute inhibition of reduction during the 12 hours (13 micromoles/root) was less than the absolute inhibition in influx (24 micromoles/root). The data suggest that the influence of ammonium on [15N]nitrate influx could not be totally accounted for by the decrease in the potential driving force which resulted from restricted reduction; an additional impact on the influx process is indicated. Reduction of [15N]nitrate in experiment II after 6 hours accounted for 30 and 18% of the tissue excess 15N in the control and ammonium treatments, respectively. Relative distribution of 15N between roots and exudate (experiment I), or between roots and shoots (experiment II) was not affected by ammonium. On the other hand, the accumulation of [15N]nitrate in roots, shoots, and xylem exudate was enhanced by ammonium treatment compared to the control, whereas the accumulation of reduced 15N was inhibited.  相似文献

The effects of light and NO3- nutrition on 15NO3- influx in roots were investigated in young, 19-d-old, induced tomato plants grown at a constant air and solution temperature of 20C. Nitrate influx was measured by 15N accumulation for 5 min, on plants exposed to a wide range of exogenous concentrations, from 10 x 10-3 to 30 mol m-3. Influx kinetics, fitted to the data following a non-linear procedure, showed multiphasic patterns. The best fits were obtained when three pure and non-additive Michaelis-Menten kinetics were applied, with phase transitions at approximately 0.8 and 4 mol m-3. In plants grown at 3.0 mol m-3 NO3-, the asymptotic maximum influx rate (Imax) of each phase declined during the night until 24 h darkness. At the end of the day period, about a 2-fold enhancement of Imax was observed when plants were pretreated for 3 d with 0.2 instead of 3.0 mol m-3 NO3-. The influx rates measured at any given NO3- concentration and the Imax for any phase showed a negative non-linear correlation with plant nitrate concentration. Furthermore, the results suggest the existence of a set point, approximately 66 mol m-3 plant nitrate, for which influx is null at any given solution nitrate concentration. A model using modified Michaelis-Menten kinetics is proposed to predict the influx rate as a function of both solution and plant NO3- concentrations.  相似文献

The electrical response of nitrate-grown maize (Zea mays L.) roots to 0.1 millimolar nitrate was comprised of two sequential parts: a rapid and transient depolarization of the membrane potential, followed by a slower, net hyperpolarization to a value more negative than the original resting potential. The magnitude of the response was smaller in roots of seedlings grown in the absence of nitrate, but, within 3 hours of initial exposure to 0.1 millimolar nitrate, increased to that of nitrate-grown roots. Chloride elicited a separate electrical response with a pattern similar to that of the nitrate response. However, the results presented in this study strongly indicate that the electrical response to nitrate reflects the activity of a nitrate-inducible membrane transport system for nitrate which is distinct from that for chloride. Inhibitors of the plasmalemma H+-ATPase (vanadate, diethylstilbestrol) completely inhibited both parts of the electrical response to nitrate, as did alkaline external pH. The magnitude of the initial nitrate-dependent, membrane potential depolarization was independent of nitrate concentration, but the subsequent nitrate-dependent hyperpolarization showed saturable dependence with an apparent Km of 0.05 millimolar. These results support a model for nitrate uptake in maize roots which includes a depolarizing NO3/H+ symport. The model proposes that the nitrate-dependent membrane potential hyperpolarization is due to the plasma membrane proton pump, which is secondarily stimulated by the operation of the NO3/H+ symport.  相似文献

Ricinus communis L. was grown under limiting N supply in quartz sand culture, fed with 0.2, 1 or 5 mol m?3 NO3?, or in liquid culture with 0.022, 0.05 or 0.5 mol m?3 NO3?. Some of the plants were infected with Cuscuta reflexa Roxb. As occurred for the host, dry matter production and growth of C. reflexa were severely depressed with decreasing N supply to the host. When parasitized by C. reflexa, the shoot and root dry weight of Ricinus was diminished at all levels of N nutrition, but the total dry weight of host plus parasite was almost the same as that of uninfected Ricinus. In contrast to the situation in Lupinus albus (Jeschke et al. 1994b), infection by Cuscuta resulted in increased tissue N levels in the host and the N content of the system Ricinus plus C. reflexa was the same or even somewhat larger than that of uninfected plants. This indicated a sink-dependent stimulation of nitrate uptake. As a result of decreased root weights, nitrate uptake g?1 FW was stimulated by 80, 60 or only 40% at 0.2, 1 or 5 mol m?3 nitrate supply. Increased nitrate uptake was reflected, particularly at low N supply, in xylem transport; xylem sap nitrate concentrations were substantially elevated, while those of amino acids were decreased in parasitized plants. This indicated an inhibition of nitrate assimilation in roots of parasitized plants under limiting N supply. Besides these effects on N relations, C. reflexa induced a substantial sink-dependent stimulation of net photosynthesis in host leaves and a concomitant increase in stomatal opening and transpiration. This stimulation depended on the relative sink size induced by Cuscuta, on nitrogen nutrition and on leaf age, indicating that delayed senescence of leaves contributes to the overall effects of Cuscuta on its host. The Cuscuta-induced inhibition of nitrate assimilation in the roots and the increase in nitrate uptake suggest that nitrate reduction was shifted towards the leaves in the presence of C. reflexa. The stimulating effects of C. reflexa in the Ricinus-Cuscuta association are compared with the strongly inhibitory effects occurring in the tripartite association L. albus–Rhizobium–Cuscuta reflexa.  相似文献

The roles of K+ uptake and loss in the salinity response of the wild type and the salt-tolerant mutant stl2 of Ceratopteris richardii were studied by measuring Rb+ influx and loss and the effects of Na+, Mg2+, Ca2+ and K+-transport inhibitors. In addition, electrophysiological responses were measured for both K+ and Rb+ and for the effects of Na+ and NH4+ on subsequent K+-induced depolarizations. stl2 had a 26–40% higher uptake rate for Rb+ than the wild type at 0.5–10 mol m?3 RbCl. Similarly, membrane depolarizations induced by both RbCl and KCl were consistently greater in stl2. In the presence of 0–180 mol m?3 NaCl, stl2 maintained a consistently greater Rb+ influx than the wild type. stl2 retained a greater capacity for subsequent KCl-induced depolarization following exposure to NaCl. Five mol m?3 Mg2+ decreased Rb+ uptake in stl2; however, additional Mg2+ up to 40 mol m?3 did not affect Rb+ uptake further. Ca2+ supplementation resulted in a very minor decrease of Rb+ uptake that was similar in the two genotypes. Tetraethylammonium chloride and CsCl gave similar inhibition of Rb+ uptake in both genotypes, but NH4Cl gave substantially greater inhibition in the wild type than in stl2. NH4Cl resulted in a greater membrane depolarization in the wild type and the capacity for subsequent depolarization by KCl was markedly reduced. stl2 exhibited a higher Independent loss of Rb+ than the wild type, but, in the absence of external K+, loss of Rb+ was equivalent in the two genotypes. Since constitutive K+ contents are nearly identical, we conclude that high K+ influx and loss exact a metabolic cost that is reflected in the inhibition of gametophytic growth. Growth inhibition can be alleviated by reduced supplemental K+ or by treatments that slightly reduce K+ influx, such as moderate concentrations of Na+ or Mg2+. We propose that high throughput of K+ allows maintenance of cytosolic K+ under salt stress and that a high uptake rate for K+ results in a reduced capacity for the entrance and accumulation of alternative cations such as Na+ in the cytosol.  相似文献

Summary Absorption of nitrate and ammonium was studied in water culture experiments with 4 to 6 weeks old plants of barley (Hordeum vulgare L.), buckwheat (Fagopyrum esculentum L. Moench) and rape (Brassica napus L.). The plants were grown in a complete nutrient solution with nitrate (5.7±0.2 mM) or nitrate (5.6±0.2 mM) + ammonium (0.04±0.02 mM). The pH of the nutrient solution was kept at 5.0 using a pH-stat. It was found that phosphorus deficiency reduced the rate of nitrate uptake by 58±3% when nitrate was the sole N source and by 83±1% when both nitrate and ammonium were present. The reduction occurred even before growth was significantly impeded by P deficiency. The inhibition of the uptake of ammonium was less,i.e. ammonium constituted 10±1% of the total N uptake in the P sufficient plants and 30±5% in the P deficient plants. The reduction of nitrate absorption greatly decreased the difference between the uptake of anions and cations. It is suggested that P deficiency reduced the assimilation of NO 3 into the proteins, which might cause a negative feedback on NO 3 influx and/or stimulate NO 3 efflux.  相似文献

Seedlings of barley (Hordeum vulgare L. cvs Salka and Zita), buckwheat (Fagopyrum esculentum Moench) and rape (Brassica napus L. ssp. napus cv. Line) were raised at 8 or 10 different extenral P concentrations in the range 0–2000 μM. Apart from P, the nutrient solutions were complete. Phosphate influx in roots of different P status was determined by use of a nutrient solution containing 0.1 mM32P-labelled phosphate. A double labelling technique was used for simultaneous determination of influx (33P) and efflux (32P) of phosphorus by roots of barley and rape with three selected P levels. Flux determinations were also done in presence of a metabolic uncoupler (2,4-dinitrophenol) and a protein synthesis inhibitor (cycloheximide). Influx of phosphate was maximal at a certin optimal P level of the roots and decreased at both lower and higher P levels. Maximum phosphate influex [μmol (g root)-?1 h?1] were: rape 4,4, buckwheat 2.2, barley cv. Salka 1.6, barley cv. Zita 1.5. Both Hill plots and plots of the untransformed decreasing phosphate influx vs root P concentrations above the optimal were linear and had high correlation coefficients. The Hill coefficient varied between -3.1 and -4.2. The decrease of phosphate influx from the maximum to the lowest value at the highest P concentration of the root was 60–70%. Hence, phosphate influex appeared to be regulated through negative feedback by the internal level of phosphorous in the roots. The regulation mechanism seems bascially similar for the three species and may be of an allosteric type. P efflux from roots of low and optimal (with regard to P influx) P status was 15–20% of the simultaneous P influx. Contary to P influx, P efflux increased at high P status and almost eliminated (barley) or halved (rape) net P uptake. 2,4-Dinitrophenol reduced both P influx and P efflux by low P roots and gave linearly increasing P efflux with increasing root P status. This indicates that P efflux partly occurred by counter transport and ion exchange at the uptake sites, partly by passive P efflux along an electrochemical potential gradient. Phosphate influx was not affected by inhibition of barley root growth with cycloheximide, but P efflux increased considerably.  相似文献

Measurements were made of the influx of 45Ca into internodal cells of Chara corallina in solutions containing high concentrations of NaCl. Increasing salinity in the range 4–100mol m?3 NaCl resulted in a doubling of Ca2+ influx at the plasmalemma. A time-course of Ca2+ influx in 50 mol m?3 NaCl, 0.5mol m?3 CaCl2 showed that while influx at the plasmalemma increased only 1.5-fold, influx to the vacuole increased by up to 15-fold. This was interpreted as being due to inhibition of active Ca2+ efflux from the cell. The stimulation of Ca2+ influx by increasing salinity appeared to be principally a response to reduced turgor since similar stimulations were obtained when turgor was reduced by NaCl, Na2SO4 or mannitol. When cells were plasmolysed Ca2+ influx increased by 10–20-fold. The increased permeability was relatively specific for Ca2+ and was inhibitable by La3+. Survival of cells in high salt conditions was increased by 30 mmol m?3 La3+, which inhibited Ca2+ influx. Paradoxically, survival can also be extended by increasing external Ca2+ which leads to a higher influx. Therefore, it seems unlikely that the ameliorative effect of Ca2+ on the sensitivity of plants to high NaCl is mediated by Ca2+ entry across the plasmalemma. It seems more likely that the principal role of Ca2+ under these conditions is exerted externally through the control of membrane voltage and permeability.  相似文献

Nitrate-selective microelectrodes have been made using a quaternary ammonium sensor, methyl-tridodecylammonium nitrate, in a Polyvinylchloride matrix. These electrodes showed a log-linear response from 0.1 to 100 mol · m?3 nitrate with a typical slope of 55.6 mV per decade change in nitrate concentration. The only physiologically significant interfering anion was chloride but the lower limit of nitrate detection was 0.5 mol · m?3 in the presence of 100 mol · m?3 chloride which means this interference will not be important in most physiological situations. These microelectrodes were used to measure nitrate concentrations in internodal cells of Chara corallina cultured under low nitrate and nitrate-replete conditions for 6 to 30 weeks. Cells maintained in low nitrate only showed measurements which were less than the detection limit of the electrodes, while cells grown under nitrate-replete conditions showed two populations of measurements having means of 1.6 and 6.2 mol · m?3. Chemical analysis of the high-nitrate cells indicated that they contained a mean nitrate concentration of 5.9 mol · m?3. As vacuolar nitrate concentration would dominate this whole-cell measurement, it is concluded that the higher concentration measured with the electrodes represents vacuolar nitrate concentration and the lower value represents the cytoplasmic concentration. This intracellular distribution of nitrate could only be achieved passively if the electrical potential difference across the tonoplast is between +25 and + 35 mV.  相似文献

Abstract Growth-chamber cultivated Raphanus plants accumulate nitrate during their vegetative growth. After 25 days of growth at a constant supply to the roots of 1 mol m?3 (NO?3) in a balanced nutrient solution, the oldest leaves (eight-leaf stage) accumulated 2.5% NO?3-nitrogen (NO3-N) in their lamina, and almost 5% NO3-N in their petioles on a dry weight basis. This is equivalent to approximately 190 and 400 mol?3 m?3 concentration of NO?3 in the lamina and the petiole, respectively, as calculated on a total tissue water content basis. Measurements were made of root NO?3 uptake, NO?3 fluxes in the xylem, nitrate uptake by the mesophyll cells, and nitrate reduction as measured by an in vivo test. NO?3 uptake by roots and mesophyll cells was greater in the light than in the dark. The NO?3 concentration in the xylem fluid was constant with leaf age, but showed a distinct daily variation as a result of the independent fluxes of root uptake, transpiration and mesophyll uptake. NO?3 was reduced in the leaf at a higher rate in the light than in the dark. The reduction was inhibited at the high concentrations calculated to exist in the mesophyll vacuoles, but reduction continued at a low rate, even when there was no supply from the incubation medium. Sixty-four per cent of the NO?3 influx was turned into organic nitrogen, with the remaining NO?3 accumulating in both the light and the dark.  相似文献

Role of sugars in nitrate utilization by roots of dwarf bean   总被引:4,自引:0,他引:4  
Nitrate uptake and in vivo, nitrate reductase activity (NRA) in roots of Phaseolus vulgaris, L. cv. Witte Krombek were measured in nitrogen-depleted plants of varying sugar status, Variation in sugar status was achieved at the start of nitrate nutrition by excision, ringing, darkness or administration of sugars to the root medium. The shape of the apparent induction pattern of nitrate uptake was not influenced by the sugar status of the absorbing tissue. When measured after 6 h of nitrate nutrition (0.1 mol m?3), steady state nitrate uptake and root NRA were in the order intact>dark>ringed>excised. Exogenous sucrose restored NRA in excised roots to the level of intact plants. The nitrate uptake rate of excised roots, however, was not fully restored by sucrose (0.03–300 mol m?3). When plants were decapitated after an 18 h NO3? pretreatment, the net uptake rate declined gradually to become negative after three hours. This decline was slowed down by exogenous fructose, whilst glucose rapidly (sometimes within 5 min) stimulated NG?3 uptake. Presumably due to a difference in NO3? due to a difference in NO3? uptake, the NRA of excised roots was also higher in the presence of glucose than in the presence of fructose after 6 h of nitrate nutrition. The sugar-stimulation of, oxygen consumption as well as the release of 14CO2 from freshly absorbed (U-14C) sugar was the same for glucose and fructose. Therefore, we propose a glucose-specific effect on NO3? uptake that is due to the presence of glucose rather than to its utilization in root respiration. A differential glucose-fructose effect on nitrate reductase activity independent of the effect on NO3? uptake was not indicated. A constant level of NRA occurred in roots of NO3? induced plants. Removal of nutrient nitrate from these plants caused an exponential NRA decay with an approximate half-life of 12 h in intact plants and 5.5 h in excised roots. The latter value was also found in roots that were excised in the presence of nitrate, indicating that the sugar status primarily determines the apparent rate of nitrate reductase decay in excised roots.  相似文献

Studies of uptake of ionic sources of N by two hydroponically grown rice (Oryza sativa L.) cultivars (paddy‐field‐adapted Koshihikari and dryland‐adapted Kanto 168) showed that the magnitude of the nitrogen isotope fractionation (?) for uptake of NH4+ depended on the concentrations of NH4+ and cultivar (averaging –6·1‰ for Koshihikari and –12·0‰ for Kanto 168 at concentrations from 40 to 200 mmol m?3 and, respectively, –13·4 and –28·9‰ for the two cultivars at concentrations from 0·5 to 4 mol m?3). In contrast, the ? for uptake of NO3? in similar experiments was almost insensitive to the N concentration, falling within a much narrower range (+3·2‰ to –0·9‰ for Koshihikari and –0·9‰ to –5·1‰ for Kanto 168 over NO3? concentrations from 0·04 to 2 mol m?3). From longer term experiments in which Norin 8 and its nitrate‐reductase deficient mutant M819 were grown with 2 or 8 mol m?3 NO3? for 30 d, it was concluded that the small concentration‐independent isotopic fractionation during absorption of this ion was not related to nitrate reductase activity.  相似文献
