The changes of ion contents, betaine levels and betaine-aldehyde dehydrogenase (BADH) activity in seedling of halophyte seepweed (Suaeda salsa) were studied under different salinity (NaC1 and KC1). Results showed that the K+ contents in seepweed seedlings grown in NaC1 for 96 h decreased as the external salinity was increased comparing with the controls while the Na+ levels increased substantially. The levels of Na+ in seepweed treated with KC1 fell, but K+ levels rose. The betaine levels and the BADH activity rose with the increase of external NaC1 concentrations and the betaine levels increased as the time of the treatment was lengthened. The effects of KC1 on the betaine levels and BADH activity were similar to these of NaC1. These results illustrated that NaC1 and KC1 can cause a large amounts of betaine accumulation in seepweed, and preliminary data suggest that BADH activity has played a role in betaine accumulation.  相似文献

水分胁迫下山黧豆中ABA及ODAP的积累研究   总被引:4,自引:0,他引:4  
用PEG、PEG+ABA、ABA分别处理15d龄的山黧豆幼苗,取其叶片为实验材料,测定内源ABA、ODAP、MDA和H2O2含量以及几种抗氧化酶活性,结果表明,与对照相比处理材料叶片中ABA和ODAP含量显著增加;外源ABA的加入降低了PEG胁迫引起的MDA和H2O2含量的增加,延缓了PEG胁迫引起的CAT活性的衰减,提高了GR活性.用外源ABA长时间处理山黧豆,发现叶片中ABA含量显著增加,随后出现ODAP的积累;ABA处理初期(0~3d)对叶片中活性氧代谢影响不大,随着ABA处理时间的延长(7~15d),可引起叶片中SOD、POD、CAT、GR活性的降低,MDA、H2O2含量的增加,表明ABA确实可促进ODAP的积累.  相似文献

A correlation between protein kinase phosphorylation and ABA level was studied in Malus sieversii (Ledeb.) Roem. seedlings under water stress. The seedlings were treated with PEG 6000 for imitation of water stress, and the MAPK activity and ABA content in each treatment were then determined. We demonstrated that the increase in the activities of the total protein kinase (TPK) and mitogen-activated protein kinase (MAPK) after treatment with 20% PEG 6000 appeared to result in a high level of ABA. MAPK activity accounted for 76.8% of TPK activity. The activity peaks of TPK and MAPK preceded the highest level of ABA accumulation. It is interesting that the ABA level in roots and leaves of seedlings pretreated with 2 × 10−2 mM exogenous ABA for 20 min following treatment by 20% PEG 6000 was much higher than that of seedlings treated with exogenous ABA only. We analyzed the influence of MAPK inhibitor ITU (5-iodotubercidin) on ABA accumulation in the seedlings of M. sieversii under water stress and showed that 1 μM ITU significantly decreased the ABA level induced by a water loss. However, the phosphoesterase inhibitor PAO (phenylarisine oxide) enhanced ABA accumulation, indicating that the phosphorylated MAPK was correlated to ABA synthesis. Together, these results suggest that MAPK phosphorylation played an important role in ABA accumulation under water stress, and MAPK might mediate the signal transduction of ABA synthesis.  相似文献

Monovalent cations such as Na+ and K+ inhibit the activity of T4 DNA ligase. However, the extent of inhibition varies with the terminal sequence of the duplex DNA used as substrate; in many cases, ligation of DNA is completely inhibited at 200 mM. The activity of the ligase is stimulated by raising the concentration of polyethylene glycol 6000 from 0 to 15% (w/v) when NaC1 and KC1 were both absent. Ligation was reduced as the concentration of NaC1 or KC1 was raised in a mixture containing 5 or 15% PEG 6000. With 10% PEG 6000, both cohesive- and blunt-end ligation of this ligase increased at high concentrations of salt (150-200 mM NaC1, or 200-250 mM KC1). Further, with 10% PEG 6000, inter- and intramolecular ligation occurred at low salt concentrations (0-100 mM NaC1, or 0-150 mM KC1); only linear oligomers were formed by intermolecular ligation at the high concentrations.  相似文献

In vitro cultures and endogenous abscisic acid (ABA) analyses with gas chromatography-selected ion monitoring-mass spectrometry (GC-SIM-MS) were used to study the effects of AgNO3 and polyethylene glycol (PEG) on white spruce ( Picea glauca ) somatic embryo maturation and endogenous ABA contents, Normally, in the absence of ABA, white spruce somatic embryos cannot mature. However, AgNO3 and PEG stimulated somatic embryo maturation by increasing the number of cotyledonary embryos. A combination of 100 μ M AgNO3 and 40 g l−1 PEG was the treatment that was most effective in enhancing cotyledonary embryo formation without exogenous ABA. Either AgNO3 or PEG was able to increase endogenous ABA levels of the embryogenic culture but a combination of AgNO3 and PEG was the most effective treatment. Stimulation of cotyledonary embryo formation by AgNO3 occurred only at low ABA levels, while PEG promoted embryo formation at all exogenous ABA concentrations. Germination tests indicated that AgNO3 had no negative effects on embryo germination and conversion while the PEG-treated embryos failed to germinate.  相似文献

Wheat seedlings (Tritium aestivum "No. 1 Yuyuan") with 3 leaves were transplanted to 1/2 strangth Hoagland nutrient solution containing 100 mmol/L NaCl and supplemented with different concentrations of CaCI2, which were mode isosmotic by adding polyethylene glycol (PEG) and aerated by pump. Results showed that the Na+ content of shoots and roots, relative leaf expansion rate, plasma membrane permeability, the levels of membrane lipid superoxidation and the molar percentage of monogalacrosyl diglyceride(MGDG), digalactosyl diglyceride(DGDG) phosphatidyl choline(PC) and phosphatidyl ethanolamine(PE) in membrane lipids of roots increased, the plant dry weight, K+ content, SOD activity and the molar percentage of phosphatidic acid(PA), phospatidyl inositol (PI), phosphatidyl glycerol(PG) and polyphosphoglyceric acid(PPG) decreased in roots. There was no change in sulfolipid(SL). However, the above mentioned salt injury effects were all alleviated by the different Na+/Ca2+ ratios. The maximum alleviation of salt injury effect was at Na+/Ca2+ ratio of I0. As three kinds of free radical scavengers were used to pretreat wheat seedlings prior to NaC1 treatment the malondialdehyde(MDA) content decreased unanimously, but increased with SOD inhibitor sodium diethyldithiocarbamate (DDTC) pretreatment to wheat seedlings. Obviously, the salt injury effects induced by NaC1 was relatied to the extent of superoxidation of membrane [ipids and also to the composkion of membrane of Ca2+ on lipids including their fatty acids as well. On the other hand, the alleviating effect of Ca2+ on NaC1 induced injury in wheat seedlings was also in relation to them.  相似文献

为揭示植物抗旱的调控机理, 对复苏植物旋蒴苣苔(Boea hygrometrica)的一个编码C2结构域小蛋白的基因BhC2DP1进行研究。Real-time PCR和ProBhC2DP1:GUS报告基因检测显示, 该基因只在干旱早期和外源Ca2+处理0.5小时时受诱导表达; 分别施加Ca2+螯合剂EGTA和逆境激素ABA均抑制该基因表达, 但二者同时处理则显著诱导其表达, 表明ABA对该基因转录水平的调控是Ca2+依赖型的。过表达BhC2DP1的拟南芥(Arabidopsis thaliana)对ABA的敏感性增强, EGTA处理可消除其与野生型的差异, 表明Ca2+是BhC2DP1蛋白参与ABA反应所必需的。综上所述, ABA和Ca2+信号途径的精细调控可能是决定干旱诱导旋蒴苣苔中BhC2DP1基因表达时间、丰度和功能的重要机制。  相似文献

The exogenous ABA in different concentrations was used to spray Zea mays L. (cv. Luyu 11) seedlings under different salinity. The growth status, osomotic potential and osmotic regulatory activity, the contents of Na+ and K+, soluble sugars and proline were determined at different time after treatment. The results were shown as follows: The exogenous ABA gave an increase in salt resistance and dry weight of corn seedlings under different salinity, this increase was as a result of inhanced osmotic adjustment activity and exclusion of sodium ions from the shoot through the action of exogenous ABA, resulting an accumulation of sodium in the roots. Moreover, the decrease of osmotic potential and the increase of osmotic regulatory activity were for reasons of enhancement of organic osmotors, instead inorganic, e.g. Na+ and K+ osmotors. The mechanism of sodium exclusion from the shoot was also discussed.  相似文献

本文比较研究了等渗NaCl和KCl胁迫下,高粱幼苗生长及叶片离子含量、质膜相对透性和有关气体交换参数的变化。结果表明,在低浓度NaCl和KCl胁迫7天时,高粱生长、含水量和质膜相对透性与对照相比没有明显变化,而净光合速率、蒸腾速率和气孔导度已明显下降,叶肉细胞间隙CO2浓度明显增加。NaCl胁迫下叶片Na+含量成倍增加,而K+和Ca2+含量无明显变化。KCl胁迫时叶片K+含量明显增加,Ca2+含量明显下降,而Na+含量没有明显变化。随着NaCl或KCl浓度的增加,幼苗生长和叶片含水量明显下降,质膜透性和细胞间隙CO2浓度明显增加,净光合速率、蒸腾速率和气孔导度进一步下降。 NaCl胁迫下叶片Na+含量进一步增加,K+和Ca2+进一步下降,而KCl胁迫下叶片K+含量进一步 增加,Na+和Ca2+含量进一步下降。KCl对高粱生长抑制、质膜透性、Ca2+含量下降及光合气体交换参数的影响均明显大于等渗的NaCl。  相似文献

玉米液泡膜焦磷酸酶基因ZmVPP1的克隆及逆境下的表达分析   总被引:1,自引:0,他引:1  
采用同源克隆的方法在玉米中得到一个与拟南芥耐盐基因AVP1类似的基因.BLAST分析表明,该基因编码蛋白属于质子泵焦磷酸酶家族(H_PPase superfamily)中的Ⅰ型VPP,将其命名为ZmVPP1.ZmVPP1包舍一个2301bp的开放阅读框,编码766个氨基酸残基.蛋白比对结果表明,该蛋白在不同植物中相当保守.实时荧光定量PCR检测发现,ZmVPP1基因在成熟叶片中高丰度表达,在生殖器官中表达量较少.脱水、高盐、低温等逆境胁迫条件和ABA处理下的表达分析表明,ZmVPP1基因受逆境的诱导表达,属于不依赖于ABA的途径.由此推断,ZmVPP1可能参与玉米对Na+的隔离,从而起到耐盐的作用.  相似文献

Tanshinones, a group of active ingredients in Salvia miltiorrhiza, are derived from at least two biosynthetic pathways, which are the mevalonate (MVA) pathway in the cytosol and the 2-C-methyl-d-erythritol-4-phosphate (MEP) pathway in the plastids. Abscisic acid (ABA) and methyl jasmonate (MJ) are two well-known plant hormones induced by water stress. In this study, effects of polyethylene glycol (PEG), ABA and MJ on tanshinone production in S. miltiorrhiza hairy roots were investigated, and the role of MJ in PEG- and ABA-induced tanshinone production was further elucidated. The results showed that tanshinone production was significantly enhanced by treatments with PEG, ABA and MJ. The mRNA levels of 3-hydroxy-3-methylglutaryl co-enzyme A reductase (HMGR), 1-deoxy-d-xylulose 5-phosphate reductoisomerase (DXR) and 1-deoxy-d-xylulose 5-phosphate synthase (DXS), as well as the enzyme activities of HMGR and DXS were stimulated by all three treatments. PEG and ABA triggered MJ accumulation. Effects of PEG and ABA on tanshinone production were completely abolished by the ABA biosynthesis inhibitor [tungstate (TUN)] and the MJ biosynthesis inhibitor [ibuprofen (IBU)], while effects of MJ were almost unaffected by TUN. In addition, MJ-induced tanshinone production was completely abolished by the MEP pathway inhibitor [fosmidomycin (FOS)], but was just partially arrested by the MVA pathway inhibitor [mevinolin (MEV)]. In conclusion, a signal transduction model was proposed that exogenous applications of PEG and ABA triggered endogenous MJ accumulation by activating ABA signaling pathway to stimulate tanshinone production, while exogenous MJ could directly induce tanshinone production mainly via the MEP pathway in S. miltiorrhiza hairy roots.  相似文献

A high molecular weight protein was found in pea ( Pisum sativum L. ) seedling by means of Westem-blotting, and it consisted of a ct (60.4 kD) and α β (65.5 kD) subunits. The protein had low ATPase activity. Its expression could be enhanced by 3 to 4 folds under heat-shock stress, but was not affected by exogenous ABA. The results of localization and 35S-Met labeling showed that it was a cytoplasmic protein and its synthesis was not inhibited by chloramphenicol.  相似文献

Effect of ABA on the activity of mitochondrial membrane bound Na+-K+ATPase during isolation of mitochondria from soybean cotyledons, there was an increased activity of the mitochondrial membrane bound Na+-K+ATPase if abscisic acid (ABA) was added to the medium when soybean seedling were grown at 27 ℃ or 16℃, 40 mol/L ABA could change the turning point temperature of Arrhenius the activation energies (Ea) of Na+-K+ATPase from 36.6℃ or 22.7℃ decreased to 30. 3℃ or 17.8℃ respectively. The Km value and S0.5 value for this enzyme with ABA was higher than that without ABA. Hill coefficient (n) of this enzyme with ABA was 1.01 and without ABA was 1.89. The o2 uptake of mitochondria also increased. These results showed that the temperature of phase transition of mitochondrial membrane were decreased by ABA treatment.  相似文献

Embryogenic tissues of white spruce [Picea glauca (Moench) Voss] remain in an early developmental stage while cultured on 2,4-dichlorophenoxyacetic acid and N6-benzyladenine, but develop to cotyledonary embryos when these phytohormones are replaced by abscisic acid. Twenty-eight cDNAs were isolated from cotyledonary embryos by differential screening against immature embryo and non-embryonic tissues. Temporal expression patterns of these cDNAs during ABA-stimulated somatic embryo development were observed. This showed that clones could be allocated to various groups, including embryo-abundant, embryo-maturation-induced, and those whose expression was modulated during embryo development, germination or in non-embryogenic tissues. Expression corresponding to these cDNA clones showed that there were various responses to exogenous ABA or polyethylene glycol during a period of 48 h. 采用荧光动力学的方法来区分盐胁迫中的渗透因素和离子因素。用五种等渗Hogland培养液(分别含(NaCl,KCl,NaNO3,KNO3和PEG)对冬小麦处理两星期。结果,与对照相比,NaCl处理引起PSII受体侧电子库(CA/Fo)变小,PSII活性(Fv/Fo)、原初光能转化效率(Fv/Fm)、量子产量(Yield)与荧光化学猝灭系数(qP)下降,但使QB-非还原性PSII反应中心含量增加。然而,等渗的PEG处理并不产生类似的伤害。这表明渗透因素不是盐胁迫对光合作用造成伤害的主要原因。同时,KNO3处理对光合作用不产生伤害。由于NaCl和NaNO3处理均造成受体侧电子库变小,PSII活性和原初光能转化效率下降,并使QB-非还原性PSII反应中心增加,而等渗的PEG和KCl处理并不产生类似的伤害,这暗示Na+可能是盐胁迫影响光合作用的主要毒害离子。  相似文献
