通过胚拯救, 成功获得鹅观草Roegneria kamoji (2n=6x=42, SSHHYY)和普通小麦中国春Triticum aesti-vum (2n=6x=42, AABBDD)的正反交属间杂种F1, 并对这些杂种F1及其BC1的形态学、减数分裂配对行为、育性和赤霉病抗性进行研究。结果表明, (鹅观草×中国春)F1和(中国春×鹅观草)F1的形态介于双亲之间。杂种F1花粉母细胞减数分裂中期I染色体构型分别为40.33I + 0.78II + 0.03III和40.40I + 0.79II 。杂种F1高度雄性不育, 用中国春花粉与其回交可获得BC1代种子。(鹅观草×中国春) F1×中国春BC1植株的染色体数目主要分布在55~63之间, 单价体较多, 植株高度不育; (中国春×鹅观草)F1×中国春BC1植株染色体数目也主要分布在55~63之间, 但其中部分植株拥有整套小麦染色体且能正常配对、分离, 可形成部分可育花粉粒, 能收到少量自交结实种子。在 (鹅观草×中国春)F1中有1株穗型趋向中国春, 其染色体数目为2n=63, 经染色体分子原位杂交(GISH)检测, 含有42条小麦染色体和21条鹅观草染色体。该杂种F1在减数分裂中期I平均每个花粉母细胞有26.40I+18.30II, 但植株高度雄性不育, 用中国春花粉回交能收到BC1种子。(鹅观草×中国春) F1 (2n=63)×中国春BC1的染色体数目主要分布在40~59之间, 其中的外源染色体已经逐渐减少, 虽然该BC1的穗型已接近中国春, 但仍然高度不育。赤霉病抗性鉴定结果显示, 所有杂种F1及大部分BC1对赤霉病均表现出较好的抗性。  相似文献

Kengyilia melanthera (Keng)J. L. Yang, Yen et Baum and K. kokonorica (Keng)J. L. Yang, Yen et Baum are two hexaploid perennial grasses of the tribe Triticeae native in west China. K. melanthera and K. kokonorica were hybridized with Roegneria kamoji Ohwi(2n=42,StStHHYY) and K. hirsuta (Keng)J. L. Yang, Yen et Baum (2n = 42, PPStStYY) respectively. Chromosome pairing behaviour at metaphase I in the parents and hybrids was studied. Meiotic configurations were 18.20 Ⅰ + 11.74 Ⅱ + 0.09 Ⅲ + 0.01V for R. kamoji×K. melanthera, 1.06Ⅰ + 20.47 Ⅱ for K. hirsuta×K. melanthera, 19.36Ⅰ + 11.26 Ⅱ + 0.04Ⅲ for R. kamoji×K. kokonorica, and2.46Ⅰ + 19.44Ⅱ + 0.14 Ⅲ + 0.06 Ⅳ for K. hirsuta×K. kokonorica. Considering chromosome pairing in the hybrids, as well as morphological characters, K. melanthera and K. kokonorica should be grouped in Kengyilia Yen et J. L. Yang instead of being keeped inRoegneria sect. Paragropyron Keng , or in Elymus L. or Elytrigia Desv.  相似文献

To elucidate the genome relationships in the genus Dasypyrum and the ancestry of tetraploid D. breviaristatum, two cytotypes of D. breviaristatum and D. villosum were reciprocally crossed with one another. Chromosome pairing at the first metaphase of meiosis and fertility were examined in the F1 hybrids and the parental plants. The mean pairing configuration and mean arm pairing frequency in D. villosum-D. breviaristatum (2x) hybrids were 11.12I + 1.44II per cell and 0.107, respectively, and they were almost completely sterile. In D. breviaristatum (4x)-D. breviaristatum (2x) hybrid, up to seven trivalents were formed, and the mean pairing configuration was 3.38I + 3.20II + 3.74III + 0.005IV per cell. The mean arm pairing frequency and relative affinity calculated in that F1 hybrid were 0.915 and 0.641, respectively. Seven bivalents and seven univalents were characteristically formed in D. villosum-D. breviaristatum (4x) hybrids. Based on the present results, we clearly concluded that the genome of diploid D. breviaristatum is distantly related to the genome V of D. villosum, and that these two species have different basic genomes. We, therefore, proposed the symbol Vb for the haploid genome of diploid cytotype of D. breviaristatum. Moreover, we concluded that tetraploid D. breviaristatum is an autotetraploid with doubled sets of the genomes homologous with that of diploid D. breviaristatum, and we proposed the genome constitution VbVb for the haploid genome set of tetraploid cytotype of D. breviaristatum. Furthermore, from the chromosome pairing in the F1 hybrids involving Moroccan and Greek accessions, it was suggested that complicated rearrangements of chromosome structure have occurred in tetraploid D. breviaristatum in its natural populations across the entire distribution area.  相似文献

大鹅观草( Roegneria grandis Keng) 是分布于我国陕西的一种多年生四倍体植物。为了探索大鹅观草与鹅观草( Roegneria kamoji Ohwi) 间的物种生物学关系, 通过对其进行远缘杂交,幼胚离体培养, 合成了远缘杂种; 并对亲本及杂种F1 代花粉母细胞减数分裂中期Ⅰ染色体配对行为及形态学进行了研究。结果表明杂种F1 减数分裂染色体配对平均构型为: R- kamoji ×R-grandis10-26 Ⅰ+ 12-37 Ⅱ(c- 值= 0-74) , 大鹅观草与鹅观草所含的SY 染色体组间存在较大的同源性分化; 杂种穗部特征大多数介于双亲之间。  相似文献

Morphological comparison, cytogenetic study and fertility analysis of Hystrix duthiei (2n = 28) from China, Hystrix longearistata (2n= 28) from Japan and their artificial hybrids were carried out. Morphologically H. duthiei was similar to H. longearistata. H. longearistata had longer lemma awn, wider leaf and 2~3 florets per spikelet, while H. duthiei had 1~2 florets per spikelet. These two taxa can be easily crossed. Fl hybrids showed very high degree of bivalent pairing (13~14 bivalents) at the metaphase- I of meiosis. No multivalents were found. The fertility of pollen and seed set of the parents were normal, while the Fl hybrids were of only partial fertility. H. longearistata was closely related to H. duthiei. They should be included in the same species. Because of the differences of their distributions and habitats, some morphological divergency and a little sterility barrier have had appeared between them. It is reasonable to treat Hystrix longearistata as a sub-species of Hystrix duthiei .  相似文献

鹅观草广泛分布于我国各地及日本和朝鲜等地,具有丰富的遗传变异,种内变异类型丰富,一些居群间存在明显的生殖隔离。本研究利用人工杂交、形态学、繁育学及染色体组分析方法对来自四川雅安、四川宜宾新文、四川宜宾翠屏山、青海、新疆、浙江及山东的7个鹅观草居群进行了遗传分化研究。结果显示:(1)不同居群在株高、穗长、外稃长等表型性状存在差异,各居群间杂种F1表型性状普遍优于两亲本居群,但是颖宽个别性状劣于亲本。(2)各居群间杂种F1染色体配对水平总体较高,但也存在差异;二价体数目变异范围为19.77~20.91,单价体数目变异范围为0.18~2.46,还有低频率的三价体、四价体出现。(3)杂种F1的育性为78.9%~89.67%,自然结实率43.05%~77.98%,其中浙江与四川宜宾翠屏山居群的杂种F1的自然结实率最低(43.05%)。以上结果表明,7个鹅观草居群间存在一定程度的分化,且与各居群的地理分布呈正相关;四川宜宾与浙江居群之间存在一定的生殖隔离,遗传变异较大。  相似文献

通过远缘杂交,结合杂种幼胚离体培养,获得了节节麦(Aegilops tauschii,2n=14,DD)和簇毛麦(Dasypyrum villosum,2n=14,VV)的属间杂种F1。对杂种F1花粉母细胞减数(PMC)分裂中期Ⅰ (MⅠ)染色体配对行为进行观察发现,“节节麦×簇毛麦”杂种F1平均每PMC有1.25个棒状二价体, 染色体的平均构型为2n=14=11.49Ⅰ+1.25Ⅱ (Xta=1.25), 大部分被观察的细胞出现1~5个二价体, 表明节节麦D染色体与簇毛麦V染色体间具有相对较高的部分同源配对, D和V染色体之间存在一定的部分同源性。F1植株高度自交不育,经染色体加倍处理后能够自交结实。Abstract: ‘Aegilops tauschii×Dasypyrum villosum’ F1 hybrids were obtained by the combination of hybridization and embryo culture in vitro. Chromosome pairing behavior in meiosis of the hybrid F1 was carried out. Results showed that in an average , 1.25 rod bivalents were observed in one PMC, meiotic configuration was 2n=14=11.49Ⅰ+1.25Ⅱ(Xta=1.25) and most of PMCs possessed 1~5(rod) bivalens, indicating that the relatively high homeology was detected between the D genome of Ae.tauschii and the V genome of D.villosum. The morphological differences between F1 hybrids and their parents were significant. F1 plants were highly self-sterile, but partially self-fertile after treated by chromosome doubling technique.  相似文献

In order to investigate morphological variations of Elymus sibiricus L., E. nutans Griseb., and E. burchan-buddae (Nevski) Tzvelev [=Roegneria nutans (Keng) Keng], and to explore their systematic relationships, six morphological characters were measured and compared between the Elymus species. Interspecific hybridizations between E. nutans and E. sibiricus, and E. burchan-buddae were carried out with the aid of embryo rescue. Chromosome pairing behaviour was also analysed at metaphase- I of meiosis in pollen mother cells of F1 hybrids. Morphologic characters, such as length of spikes and length of glumes varied considerably between different individuals of each species, whereas length and width of paleas were quite stable and different between species. The interspecific hybrids were completely sterile and their meioses were irregular. Meiotic configuration of E. nutans × E. burchan-buddae was 7.70I +13.40Ⅱ+0.06Ⅲ+0.08 IV, whereas that of E. sibircus×E. nutans was 11.98I+9.61Ⅱ+0.64Ⅲ+0.39Ⅳ+0.01V. It is concluded from the morphological and cytological study that (1) it is possible to identify the three Elymus species using the palea character, in addition to other traditionally applied characters; (2) Elymus nutans and E. burchan-buddae have a comparatively high genomie relationship, while E. nutans and E. sibircus have a relatively low genomic affinity to one another; and (3) a certain chromosome pairing regulator was presented in the hybrid between E. burchan-buddae andE. nutans.  相似文献

Background and Aims

Lady''s slipper orchids (Paphiopedilum) are of high value in floriculture, and interspecific hybridization has long been used for breeding improved cultivars; however, information regarding the genome affinities of species and chromosome pairing behaviour of the hybrids remains almost unknown. The present work analyses the meiotic behaviour of interspecific hybrids by genomic in situ hybridization and cytologically evaluates the genomic relationships among parental species.

Methods

Eight interspecific F1 hybrids of Paphiopedilum species in various subgenera or sections were investigated in this study. The chromosome behaviour in meiosis of these interspecific hybrids was analysed and subjected to genomic in situ hybridization and fluorescent in situ hybridization.

Key Results

Genomic in situ hybridization was demonstrated as an efficient method to differentiate between Paphiopedilum genomes and to visualize the chromosome pairing affinities in interspecific F1 hybrids, clarifying the phylogenetic distances among these species. Comparatively regular chromosome pairing observed in the hybrids of P. delenatii × P. bellatulum, P. delenatii × P. rothschildianum and P. rothschildianum × P. bellatulum suggested high genomic affinities and close relationships between parents of each hybrid. In contrast, irregular chromosome associations, such as univalents, trivalents and quadrivalents occurred frequently in the hybrids derived from distant parents with divergent karyotypes, such as P. delenatii × P. callosum, P. delenatii × P. glaucophyllum, P. rothschildianum × P. micranthum and P. rothschildianum × P. moquetteanum. The existence of multivalents and autosyndesis demonstrated by genomic in situ hybridization in this study indicates that some micro-rearrangements and other structural alterations may also play a part in differentiating Paphiopedilum species at chromosomal level, demonstrated as different chromosome pairing affinities in interspecific hybrids.

Conclusions

The results indicate that genome homology and the interaction of genetic factors, but not chromosome number nor karyotype similarity, determine the chromosome pairing behaviour in Paphiopedilum hybrids.  相似文献

Intergeneric crosses were made betweem Psathyrostachys huashanica (2n=14, NN)and two Roegneria species, namely, R. ciliaris (2n=28, SSYY), and R. tsukushiensis (2n=42, SSHHYY). Two combinations of P. huashanica crossed with R. ciliaris and R. tsukushiensis produced adult hybrid plants. Although completely sterile, the hybrid plants developed rather vigorously, and were morphologically intermediate between the two parents.Two spikelets per node in part were observed in hybrids, which evidently came from P. huashanica. The chromosome configurations of R. ciliaris × P. huashanica and R. tsukushiensis × P. huashanica were 20.73 I+0.318 II, 24.80 I+1.578 II+0. 012 III, respectively. Polypolar division was found at anaphase I in meiosis of two hybrids. Abnormal meiosis in two hybrids was observed. The chromosome pairing indicates that there is only a little chromosome homoeology between “N” genome of P. huashanicaand “S”, “Y” or “H” genomes of R. ciliaris and R. tsukushiensis.  相似文献

The successful production of a large number of artificial hybrids betweenDahlia species based on x = 16 has allowed a detailed study of their genomic relationships. Chromosome behaviour in these artificial hybrids was extremely similar to that observed in parental species suggesting that there is a considerable degree of homology between the genomes of theseDahlia species. Using GISH it can be demonstrated that in these hybrids bivalent formation involved pairing only between parental genomes. The ability of GISH to differentiate between parental genomes in artificial hybrids was variable, indicating that molecular divergence of highly repeated sequences has accompanied the evolution of these species. However, the extent of chromosome pairing and chiasma formation in the hybrids does not reflect the differences that can be detected by GISH. Seyeral of the new hybrid combinations have resulted in horticulturally interesting plants.  相似文献

Summary The meiotic behaviour of F1 hybrids of hexaploid Triticale that differed in their genotypic or chromosomic constitution, and diploid rye, was investigated. Meiotic analysis were done by Feulgen and C-banding staining methods. A differential desynaptic effect in the hybrids was detected and explained in terms of genetic differences in pairing regulators. The high homoeologous pairing (A-B wheat chromosomes and wheat-rye chromosomes) observed in the hybrids can be explained in terms of an inhibition of the effect of a single dose of thePh allele of the 5B chromosome produced by two doses of the 5R chromosome. The higher homoeologous pairing detected in the hybrid 188 x Canaleja could be the overall result of the balance between thePh diploidizing system (1 dose), the pairing promoter of the 5R chromosome (2 doses) and that of the 3D chromosome (1 dose coming from the parental line Triticale with the substitution 3R by 3D).  相似文献

Genomic in-situ hybridization (GISH) was used to determine the amount of wheat-rye chromosome pairing in wheat (Triticum aestivum) x rye (Secale cereale) hybrids having chromosome 5B present, absent, or replaced by an extra dose of chromosome 5D. The levels of overall chromosome pairing were similar to those reported earlier but the levels of wheat-rye pairing were higher than earlier determinations using C-banding. Significant differences in chromosome pairing were found between the three genotypes studied. Both of the chromosome-5B-deficient hybrid genotypes showed much higher pairing than the euploid wheat hybrid. However, the 5B-deficient hybrid carrying an extra chromosome 5D had significantly less wheat-rye pairing than the simple 5B-deficient genotype, indicating the presence of a suppressing factor on chromosome 5D. Non-homologous/non-homoeologous chromosome pairing was observed in all three hybrid genotypes. The value of GISH for assessing the level of wheat-alien chromosome pairing in wheat/alien hybrids and the effectiveness of wheat genotypes that affect homoeologous chromosome pairing is demonstrated.  相似文献

本文研究了禾本科小麦族鹅观草属的3个种:缘毛鹅观草(Roegneria pendulina Neuski 2n=4x=28),纤毛鹅观草(R. ciliaris (Trin.) Nevski 2n=4x=28)和鹅观草(R. kamoji Ohwi 2n=6x=42)及其种间杂种的形态变异和染色体配对行为。各杂种F_1的减数分裂染色体配对数较高,但杂种高度不育。在杂种减数分裂中还观察到一定频率的多价体形成。以上结果充分表明该3种植物享有两个共同的基本染色体组,在S和Y染色体组之间发生过染色体相互易位,缘毛鹅观草的染色体组可拟定为S~PY~P。  相似文献

Two closely related species of Gibasis, G. karwinskyana and G. consobrina, and their F1 hybrids were studied cytologically at the diploid and tetraploid level. Despite similarity in their basic karyotype, pairing was extremely limited in the diploid hybrid and almost exclusively autosyndetic in the tetraploid, except for multivalent formation due to interchange heterozygosity. The analysis was considerably facilitated by the use of C-banding techniques at meiosis, by which the chromosomes of each species could be readily identified. In the parents, quadrivalents were formed between homologous but non-identical chromosomes, which also formed autosyndetic bivalents in the hybrids. Meiotic pairing in the hybrids was unaffected by polytypy for C-bands among different populations of the parental species.  相似文献

The evolutionary consequences of hybridization ultimately depend on the magnitude of reproductive isolation between hybrids and their parents. We evaluated the relative contributions of pre-and post-zygotic barriers to reproduction for hybrid formation, hybrid persistence and potential for reproductive isolation of hybrids formed between two Rhododendron species,R. spiciferum and R. spinuliferum. Our study established that incomplete reproductive isolation promotes hybrid formation and persistence and delays hybrid speciation.All pre-zygotic barriers to reproduction leading to hybrid formation are incomplete: parental species have overlapping flowering; they share the same pollinators;reciprocal assessments of pollen tube germination and growth do not differ among parents. The absence of post-zygotic barriers between parental taxa indicates that the persistence of hybrids is likely. Reproductive isolation was incomplete between hybrids and parents in all cases studied, although asymmetric differences in reproductive fitness were prevalent and possibly explain the genetic structure of natural hybrid swarms where hybridization is known to be bidirectional but asymmetric. Introgression, rather than speciation, is a probable evolutionary outcome of hybridization between the two Rhododendron taxa. Our study provides insights into understanding the evolutionary implications of natural hybridization in woody plants.  相似文献

利用普通小麦与6x小簇麦杂交转移簇毛麦有利基因的过程中,发现由小麦品系J-11为亲本的杂种F1花粉具有高可育的特性,研究结果表明,它可能是由小麦品系J-11核基因组上所携带的基因与簇毛麦染色体相互作用影响杂种F1减数分裂四分体时期的行为而形成的,导致其花粉育性及结实性的增高和外源染色体传递行为的不同。结果指出了可能存在一类影响远缘杂种减数分裂四分体时期行为的新基因,这种基因与外源染色体的相互作用对控制远缘杂种结实性及其外源基因的转移具有重要意义。  相似文献

Cytological studies were carried out on two wild species (L. hirsutum and L. pennellii) and the cultivated species (L. esculentum) of tomato and their F1 hybrids. Both parents and hybrids show a diploid chromosome number of 2n=24. The meiotic behaviour of the cultivated species showed a high degree of chromosome homology resulting in a high level of chiasmata frequency per bivalent. In contrast, the two wild species showed a slight increase in uniyalent frequency and a decrease in bivalent formation and chiasmata frequency. The meiotic behaviour of the hybrids showed a high level of univalents and low levels of bivalents as well as trivalents. Highly significant decreases in chiasmata frequency and increases in meiotic abnormalities, especially in the L. esculentum X L. pennellii hybrid, also were detected. The high meiotic irregularity and low chiasmata frequency recorded in the second hybrid indicated the disharmony and difference between its parental genomes and also served to predict its sterility. With regard to degree of pairing recorded in the hybrids, there is a possibility that sterility in such cases may refer to genetic factors in addition to the previously mentioned reasons. Pollen fertility showed no great difference between L. esculentum and L. hirsutum and their F1 hybrid, but a significant decrease was recorded in the L. esculentum X L. pennellii hybrid, which was clearly associated with high meiotic irregularity, low chiasmata frequency and chromosome association.  相似文献
