本文建立了紫外谱线组法鉴别地钱、拳卷地钱和粗裂地钱的方法。通过这三种药材的石油醚、氯仿、乙醇和水提取液的紫外谱线比较,发现地钱、拳卷地钱和粗裂地钱的紫外谱线图、最大吸收峰数目及峰位值具有明显差异。该法简单、准确,可用来鉴别地钱、拳卷地钱和粗裂地钱的原药材。  相似文献

采用电镜和细胞化学技术对地钱(Marchantia polymorpha)卵发生过程进行了研究,根据卵发生过程中细胞化学和超微结构特征可将卵发育过程分为幼卵、中期卵和成熟卵3个阶段.幼卵阶段,卵细胞、腹沟细胞及颈沟细胞间有发达的胞间连丝,但卵与腹沟细胞间的胞间连丝很快退化,幼卵细胞内具大量透明的囊泡,均匀分布于细胞质中;卵发育中期,突出特征是卵细胞质内产生嗜锇性的脂滴,位于囊泡中,与此同时,腹沟细胞退化,其细胞质内产生大型囊泡,囊泡内分泌物与卵细胞外的物质类似,呈PAS反应阳性,表明该物质应为多糖类;卵成熟时,腹沟细胞和颈沟细胞完全退化,卵细胞外包被大量粘性多糖类物质,卵细胞核表面不规则,产生明显的核外突,众多的小泡围绕着细胞核,脂滴聚集成簇,卵细胞内其他细胞器不易区分.卵发育过程中,质体不含淀粉粒,线粒体退化,高尔基体相对发达.地钱卵发育的这些特征显著区分于蕨类植物.  相似文献

用显微观察技术对苔类植物地钱(Marchantia polymorpha)生殖托的形态结构进行了研究,本研究的新观察包括:(1)生殖托具有明显的背腹分化,被认为是叶状体为适应有性生殖而高度特化的直立枝;(2)雌托盘的9~11个指状裂瓣中边缘两个稍不同,除了两个边缘裂瓣外的其他裂瓣间具雌苞。托柄具有背腹之分,雌托柄背面具光合组织,并向两侧扩展形成纵沟,雌托柄的腹面具2条被鳞片重叠覆盖相互平行的纵沟,内具假根;(3)与雌托柄不同,雄托柄外观平滑,背面无光合组织及纵沟。观察发现,伞状的雌托能滞留水分,并沿雌托柄的纵沟缓慢释放连续的水流,游动精子沿着纵沟内的水流到达雌托下面的颈卵器。上述观察表明地钱生殖托的结构是对有性生殖的一种适应,这有助于我们理解地钱受精作用的机理。  相似文献

Previously we reported on a suspension culture of chlorophyllous cells of a liverwort, Marchantia polymorpha L., under photoheterotrophic conditions. The chemically defined medium was a modified Murashige and Skoog's medium, which contained, besides glucose, inorganic salts, a growth regulator (2,4-D), and twenty-four organic compounds as micro constituents. Because of this complexity, we undertook a simplification of the medium. Having examined the utilization of the major nutrients and the requirements for the micro constituents, we have succeeded in improving the medium. The new medium contains phosphate at 3.13 mM and only eight out of the twenty-four original micro organic constituents. In this new medium, the cells grow under a well-balanced nutritional condition, with richer chlorophyll and at a higher rate during the exponential phase than in the original medium.  相似文献

对地钱(Marchantia polymorpha)颈卵器发育和卵发生过程进行了显微观察和细胞化学的研究,颈卵器起始于原始细胞,该细胞呈乳突状,经横分裂产生基细胞和顶细胞,顶细胞经3次纵斜向分裂和1次横分裂产生初生细胞,初生细胞是颈卵器内的第一个细胞,经横分裂产生中央细胞和颈沟母细胞,前者产生1个腹沟细胞和1个卵细胞,后者最终产生4个颈沟细胞。颈卵器的成熟表现为颈部显著伸长和腹部膨大,卵细胞成熟时具有不规则的核,细胞质内含有丰富的囊泡和颗粒物,卵细胞周围充满粘性物质,细胞化学研究表明,该粘性物质为多糖,卵细胞质中深染色的颗粒可能为脂类物质,腹沟细胞自产生后就逐渐退化,颈沟细胞的退化迟于腹沟细胞,其数量通常为4个,偶尔可见5个颈沟细胞或具有双核的现象。  相似文献

The gemma and gametophyte of Marchantia #olymorpha were propagated in vitro. Dedifferentiation and redifferentiation as well as the media used and cultural conditions reguired were described. Since the differentiation of bryophytes was very difficult, it was necessary to culture the tissue through initiation of partial dedifferentiation on MS agar medium supplemented with 1 mg/1 2,4-D and 3% sucrose, and then subsequently the tissue was transplanted onto 1/2 Knop agar medium with addition of 4–8 mg/1 2,4-D, 0.25–0.5 mg/1 BA and Fe salt of MS medium. The formed calli were visual but still contained rhizoid, in this stage. The small calli finally were subcultured in white agar medium supplemented with mixture of pyruvic acid, citric acid and fumaric acid (5 mmol/1); 1 mg/1 2,4-D and 4% glucose. They could be differentiated thoroughly into normal tissue. The time of the total process for differentiation requied as long as 10 months. The redifferentiation and regeneration of thalli were far easier than those of higher plants even if they were transplanted onto MS phytohormone-free medium.  相似文献

Bioassay-guided separation of the antifungal constituents of the Chinese liverwort Marchantia polymorpha L. (Marchantiaceae) led to the isolation of seven bis[bibenzyl]-type macrocycles. On the basis of NMR and MS analyses, the three new compounds plagiochin E (1), 13,13'-O-isoproylidenericcardin D (4), and riccardin H (7) were identified, together with four known compounds: marchantin E (2), neomarchantin A (3), marchantin A (5), and marchantin B (6). 哺乳动物卵母细胞成熟过程需要进行两次连续的不对称分裂,最终形成体积差异巨大的子细胞:大体积的卵母细胞和两种体积较小的极体。不对称分裂现象是哺乳动物卵母细胞减数分裂的典型特征,不对称分裂后的卵母细胞是高度极化的细胞。精卵结合后,细胞重新恢复了对称分裂,但是在卵母细胞减数分裂过程中形成的极性特征却得以保留并影响早期胚胎的极性。本文对近年来在哺乳动物卵母细胞不对称分裂方面的相关研究展开综述,从细胞质不对称分裂和细胞核不对称分裂两个方面对染色体、细胞骨架在哺乳动物卵母细胞不对称分裂中的作用、细胞器在哺乳动物卵母细胞成熟过程中的重组分配、染色体非随机分离等过程进行介绍,旨在从细胞和分子水平阐述哺乳动物卵母细胞不对称分裂的主要机制。  相似文献

Aluminum (Al) induces agricultural problems limiting crop productivity in acid soils. Since Al causes morphological changes in roots, and because microtubules (MTs) play important roles in determination of tissue morphology, we investigated whether Al affects the arrangement of MTs in maize root meristem using immunolocalization techniques. When seedling roots were treated with 50 μM Al, the orientations of MTs were dramatically altered in a population of cells located in the protoderm and the two outer layers of cortex: interphase cortical MT arrays lost their normal transverse organization and became random or longitudinal; the preprophase band of MTs, mitotic spindle, and phragmoplast developed at planes 90° rotated compared to their counterparts in controls. These changes in MT orientation resulted in the change of the division plane from transverse to longitudinal, producing daughter cells positioned side by side instead of above and below. The rotation of the otherwise normal MT arrays and the division plane in Al-treated roots indicates that Al interferes with the normal polarity sensing mechanism, which may contribute to the reduced axial growth of the Al-treated roots.  相似文献

Drosophila larval brain neuroblasts divide asymmetrically to balance between self‐renewal and differentiation. Here, we demonstrate that the SCFSlimb E3 ubiquitin ligase complex, which is composed of Cul1, SkpA, Roc1a and the F‐box protein Supernumerary limbs (Slimb), inhibits ectopic neuroblast formation and regulates asymmetric division of neuroblasts. Hyperactivation of Akt leads to similar neuroblast overgrowth and defects in asymmetric division. Slimb associates with Akt in a protein complex, and SCFSlimb acts through SAK and Akt to inhibit neuroblast overgrowth. Moreover, Beta‐transducin repeat containing, the human ortholog of Slimb, is frequently deleted in highly aggressive gliomas, suggesting a conserved tumor suppressor‐like function.  相似文献

Over 60 macrocyclic bis-bibenzyls have been isolated from hepatics. They possess various biological activities such as antimicrobial, antifungal, muscle relaxant and antitumor activities, as well as inhibitory activity against DNA polymerase β and HIV. To explore the feasibility of obtaining novel bis-bibenzyls through laboratory culture, in this study, we compare the qualitative and quantitative contents of axenically farmed and field-collected specimens of Marchantia polymorpha subsp. ruderalis (Mpr). We analyzed methanol extracts of Mpr by LC–MS, and confirmed the identity of isolable compounds by NMR. We found that extracts from natural populations of Mpr contained the known bis-bibenzyl marchantin A as a major component, while the minor components do not correspond to other known bis-bibenzyls. In contrast, the chromatograms of axenically farmed Mpr extracts were highly complex, containing numerous UV active compounds. The only similarity between natural and farmed populations of Mpr was the presence of marchantin A. In the farmed Mpr production of marchantin E, G and/or C, and dehydromarchantin A is suggested. Thus, the axenically farmed liverworts produce numerous metabolites not seen in natural populations, opening up the possibility of directing biosynthesis to either increase production of known compounds or generate new compounds through non-natural precursor feeding.  相似文献

地钱(Marchantiapolymorpha)作为雌雄异株的苔纲植物,具有对重金属胁迫敏感的特性,且种群中雌雄比例差异较大,是研究重金属环境下植物性二态的理想对象之一。为探究雌雄地钱转录组对重金属镉的响应机制差异,利用高通量测序和加权基因共表达网络分析(WGCNA),鉴定了在镉胁迫条件下与不同性别地钱有关的模块和基因。结果表明,镉胁迫响应转录因子雌雄差异主要集中于bHLH、 ERF和MYB家族。关键差异响应基因包括MARPO_0147s0038、MARPO_1326s0001和MARPO_0015s0058。差异响应通路雌性主要集中在信号转导通路,雄性则集中于类黄酮生物合成。研究结果有助于揭示雌雄异株地钱对镉胁迫的反应过程和响应机制,可为理解雌雄异株植物对重金属胁迫的性别特异性响应机制提供参考。  相似文献

Very high enantioselectivities were observed in the hydrolysis of racemic 1,2- and 1,3-diacetoxycyclohexanes and their derivatives by cultured cells of Marchantia polymorpha in suspension. Asymmetry was also induced in the hydrolyses of meso-1,2- and 1,3-diacetates to the corresponding monoacetates. These findings indicate that these cultured cells hydrolyze the acetoxyl groups enantioselectively at the stereogenic center of R-configuration. Asymmetric cell division is one of the most elegant biological systems by which cells create daughter cells with different functions and increase cell diversity. In particular, PAR polarity in the cell membrane plays a critical role in regulating the whole process of asymmetric cell division. Numerous studies have been conducted to determine the underlying mechanism of PAR polarity formation using both experimental and theoretical approaches in the last 10 years. However, they have mostly focused on answering the fundamental question of how this exclusive polarity is established but the precise dynamics of polarity domain have been little notified. In this review, I focused on studies on the shape, length, and location of PAR polarity from a theoretical perspective that may be important for an integrated understanding of the entire process of asymmetric cell division.  相似文献

A strong correlation between centrosome age and fate has been reported in some stem cells and progenitors that divide asymmetrically. In some cases, such stereotyped centrosome behaviour is essential to endow stemness to only one of the two daughters, whereas in other cases causality is still uncertain. Here, we present the different cell types in which correlated centrosome age and fate has been documented, review current knowledge on the underlying molecular mechanisms and discuss possible functional implications of this process.  相似文献

The methanol and chloroform extracts of Marchantia polymorpha were tested against three Gram-negative bacterial strains, viz. Xanthomonas oryzae pv. oryzae, Salmonella enterica and Pasturella multocida and four fungal strains, viz. Tilletia indica Mitra, Fusarium oxysporum f. sp. lini, Sclerotium rolfsii Sacc. and Rhizoctonia solani Kuhn. by using disc diffusion and micro broth dilution techniques. Both the extracts showed unique activity against X. oryzae and P.multocida [per cent inhibition (PI) 11.58 and 12.55, minimum inhibitory concentration (MIC) 2.50 and 1.25 μg/mL, minimum bactericidal concentration (MBC) 2.75 and 1.25 μg/mL, respectively] but for fungi, it was shown against S.rolfsii and F. oxysporum [PI 32.65 and 33.44, MIC 2.50 and 0.65 μg/mL, minimum fungicidal concentration (MFC) 4.50 and 0.65 μg/mL, respectively].The extracts possessed antimicrobial activity with different potency against variety of micro-organisms pathogenic to plants as well as animals. Some extracts were fungistatic and bacteriostatic (methanol extract against R. solani and chloroform extract against F. oxysporum and methanol extract for bacteria, respectively), while rest showed fungicidal/bactericidal potential. The results suggest the potential of M. polymorpha for developing a broad spectrum antimicrobial formulation in future.  相似文献

Plants precisely coordinate the balance between cell proliferation and differentiation to ensure the continuous development. In Arabidopsis thaliana, members of glycogen synthase kinase 3 (GSK3) family, which are highly conserved serine/threonine protein kinases among eukaryotes, play important roles in regulating cell proliferation and differentiation during various developmental processes. However, functional roles of GSK3s in the plant lineages except angiosperms remain to be elucidated. Here, we utilized a model liverwort, Marchantia polymorpha, for studies of GSK3, because it has a single GSK3-like kinase, MpGSK. When M. polymorpha was treated with a chemical compound, bikinin, which is known as a specific inhibitor for GSK3-like kinases, growth and morphologies were altered with an expansion of the meristematic region. Similarly, Mpgsk loss-of-function mutants accumulated undifferentiated cell mass with no differentiated tissues. By contrast, overexpression of MpGSK reduced the size of the meristem region. These results suggest that MpGSK plays important roles as a regulator for the balance between cell differentiation and proliferation in M. polymorpha.  相似文献
