蝗虫具有材料易得、染色体大、染色体数相对较少等优点, 被普遍用于观察精子发生中减数分裂各个过程。蝗虫精巢制片简单、快速, 但初学者普遍感到辨别分裂相较为困难。鉴于此, 文章以东亚飞蝗(Locusta migratoria manilensis (Meyen))精母细胞的减数分裂过程为例, 介绍如何根据染色体的数目及染色体构象, 识别减数分裂各个时期。此外, 文章也简要描述了东亚飞蝗精母细胞的有丝分裂及精子形成过程, 供初学者参考。  相似文献

Evaluation of the reproductive function of Lama glama is generally considered to be a challenging task due to the difficulty of obtaining representative semen samples. One method that has been proposed for evaluation of testicular function in these animals is histologic examination of testicular needle biopsies. This study was undertaken to examine the safety and efficacy of using needle biopsies to assess testicular function in this species. One randomly selected testicle from each of 16 sexually mature llamas was biopsied with a 14-gauge self-firing biopsy instrument. The llamas were evaluated over a 6-week period with thermography for temperature changes of the scrotum. At the end of the 6-week trial, the llamas were castrated and sections of each testis were fixed in Bouin's solution for histologic examination. Immediately prior to castration, an additional biopsy was taken from each testis to compare the tissue obtained via biopsy with sections from the corresponding testis obtained after castration. A qualitative grading scale was used to compare the seminiferous tubules from each testis. No difference was found between the biopsied and the nonbiopsied testes (P = 0.69). The percentage of normal tubules between the biopsied and the nonbiopsied sides also did not differ (P = 0.70). Furthermore, the percentage of normal seminiferous tubules did not differ between the needle biopsy samples and the corresponding tissue samples obtained at castration (P = 0.48). The number of round seminiferous tubules counted in each biopsy section ranged from 3 to 67. There was no significant difference in the thermographic images of the scrotum between the biopsied and the nonbiopsied testes. This study supports testicular biopsies as a safe and useful procedure in the evaluation of testicular function.  相似文献

The life-cycle of the amoeboflagellate Tetramitus rostratus includes amoeboid, cyst, and flagellate stages. The ultrastructure of these three stages is illustrated, with particular emphasis on flagellate morphology. Amoeba morphology is typical of that of limax amoebas. Cysts, forming from trophic amoebas, are enclosed by a wall made up of two layers: ectocyst (ca. 70 nm), and endocyst (200 nm). The wall apparently forms from precursor material present in vesicles in the pre-cyst stage cytoplasm. Flagellate morphology is characterized by a well-defined top-shaped profile, maintained by microtubules under the plasma membrane. The flagellar apparatus or mastigont consists of four flagella, their basal bodies, sheaves of microtubules associated with two of the basal bodies, and several rhizoplasts (periodicity 20 nm). A deep, microtubule-supported, ventral invagination appears to function as a gullet. A small number of mitotic stages observed in amoeboid and flagellate individuals suggests similarity in the division process in both stages: intranuclear mitotic apparatus, nucleolus persisting through mitosis, no centrioles or basal bodies functioning as centrioles, difficulty in resolving chromosomes. The text compares ultrastructures of several amoeboflagellate organisms and evaluates the phylogenetic significance of those features common to different species. On the basis of this study, Tetramitus most closely resembles Naegleria spp.  相似文献

The possession of a scrotum to contain the male gonads is a characteristic feature of almost all mammals, and appears to have evolved to allow the testes and epididymis to be exposed to a temperature a few degrees below that of core body temperature. Analysis of cryptorchid patients, and those with varicocele suggest that mild scrotal warming can be detrimental to sperm production, partly by effects on the stem cell population, and partly by effects on later stages of spermatogenesis and sperm maturation. Recent studies on the effects of clothing and lifestyle emphasize that these can also lead to chronically elevated scrotal temperatures. In particular, the wearing of nappies by infants is a cause for concern in this regard. Together all of the evidence indirectly supports the view that lifestyle factors in addition to other genetic and environmental influences could be contributing to the secular trend in declining male reproductive parameters. The challenge will be to provide relevant and targeted experimental results to support or refute the currently circumstantial evidence.  相似文献

Parastrachia japonensis adults in diapause live mostly in aggregated conditions and can survive more than 1 year on only water. In this study, we demonstrated that diapausing adults had a high tendency to form clusters with no sexual bias. When 3-40 insects were enclosed in chambers of equal volume used to measure respiration, oxygen consumption was reduced to nearly half that when a single individual was enclosed. However, this reduction in metabolic rate was lost when the bugs were prevented from having physical contact with other individuals, partly lost by being enclosed with dead individuals, totally lost with the ones being washed with dietylether, and not influenced by humidity. No such effect of aggregation on respiration was found in adults in the reproductive stage, nor in two other diapausing bugs, Erthesina fullo and Plautia crossata, which hibernate in groups. From these results, we concluded that the reduction in oxygen consumption in P. japonensis was due mostly to physical contact with other individuals and partly to chemical cue that functioned to settle them down and resulted in a greatly reduced respiration rate. This trait is an effective strategy that allows diapausing adults to conserve energy and prolong survival.  相似文献

Three new unusual compound chromosomes have been synthesized in Drosophila melanogaster. They consist of two homologous autosomes joined together in the new order: right arm, left arm, centromere, left arm, right arm, for each of the two major autosomes, and one in which chromosomes 2 and 3 have been combined in the order: right arm of 2, left arm of 2, centromere, left arm of 3, right arm of 3. The attachments of the autosomal arms were accomplished by obtaining chromosome breaks at or very close to the ends of the left arms of the autosomes such that no essential chromosome material has been removed; the compounds derived from them are therefore referred to as entire compounds. These large chromosomes are recovered in progeny with frequencies lower than expectation partly because of zygote mortality associated with these chromosomes, and partly because of a failure of spermiogenesis.  相似文献

On the cause of the mammalian descent of the testes (Descensus testiculorum) Two explanations have been offered for the descent of the testes in mammals, both of which are frequently cited in the literature. Moore and co-workers argued that the phylogenetic rise of body temperature caused the translocation of the testes. According to Portmann , the descent of the testes was due to the evolution of the scrotum as a signal. However, both the sensibility of the extra-abdominal testes to temperature elevations and the optical effects of the scrotum can be interpreted as a consequence of testicular descent rather than as its cause. The hypothesis presented in this paper suggests a new adaptive explanation for the descent of the testes in mammals and regards its development as an example of evolutionary compromise. Obvious disadvantages such as reduced protection of the extra-abdominal testes, “perforation” of the inguinal abdominal wall, and an increased loss of heat from the body core have to be outbalanced by a strong selective advantage. This advantage is seen in the development of a completely new, fast mode of locomotion - the gallop. The strong flexions and extensions of the vertebral column during gallop should cause intense fluctuations of intra-abdominal pressure. Fluctuations of intra-abdominal pressure severely impede continuous flow of blood in the abdominal veins. Periodically reduced venous drainage resulting in fluctuations of intra-testicular pressure would impair the process of spermiohistogenesis, which is dependent on an absolutely constant pressure within the testis. Thus, it is the displacement of the pressure sensitive testes out of the abdominal cavity that allows for the evolution of a fast mode of locomotion accompanied by strong fluctuations of intraabdominal pressure. In the course of the phylogenetic translocation of the testes increasing specializations of the testicular blood vessels occur. In mammals possessing a scrotum the return flow of venous blood from the extra-abdominal testes to the abdominal cavity is supported by utilizing the energy of the arterial pulse (‘peripheral arterial pump’). A model for the successive stages of the descent of the testes is illustrated in Figures 7–10. The morphological changes related to the phylogenetic descent of the testes, such as the specialization of the testicular blood vessels, the forming of a cremasteric sack in the inguinal region, and the differentiation of the inguinal or perineal integument into a scrotum can all be interpreted as serving one purpose: they aid in maintaining a constant intra-testicular pressure in spite of increasing fluctuations of intra-abdominal pressure and venous blood flow during the evolution of the gallop. Although the blood vessels of the spermatic chord basically serve the same functions, they show markedly different specializations in Marsupials and Eutherians. This indicates that the descent of the testes has occurred independently, at least, in these two groups. The explanation put forward here postulates a causal relationship between the mode of locomotion and the position of the testes. Mammals possessing testes wnich reside permanently within the abdominal cavity (‘Testiconda’) cannot gallop, whereas mammals with the ability to gallop must have (periodical or permanent) extraabdominal testes (‘Testiphaena’).  相似文献

We document the occurrence of ``sex ratio' meiotic drive in natural populations of Drosophila testacea. ``Sex ratio' males sire >95% female offspring. Genetic analysis reveals that this effect is due to a meiotically driven X chromosome, as in other species of Drosophila in which ``sex ratio' has been found. In contrast to other drosophilids, the ``sex ratio' and standard chromosomes of D. testacea do not differ in gene arrangement, implying that the effect may be due to a single genetic factor in this species. In all likelihood, the ``sex ratio' condition has evolved independently in D. testacea and in the Drosophila obscura species group, as the loci responsible for the effect occur on different chromosomal elements. An important ecological consequence of ``sex ratio' is that natural populations of D. testacea exhibit a strong female bias. Because D. testacea mates, oviposits, and feeds as adults and larvae on mushrooms, this species provides an excellent opportunity to study the selective factors in nature that prevent ``sex ratio' chromosomes from increasing to fixation and causing the extinction of the species.  相似文献

To study chromosomes in bull testes and their behavior at meiosis, a prefixation treatment with hypotonic solutions was used. After fixation, the material was squashed and stained with Feulgen's nucleal reaction. When needed, additional staining with iron-hematoxylin was feasible. The hypotonic Tyrode solutions of 10 to 20% normal strength were prepared according to A. Hughes (1952); the time of pretreatment was, in general, 15-30 min; fixation was made in glacial acetic acid-absolute alcohol (1:3) and Feulgen squash technic was carried out according to Schedule 5 of Darlington and La Cour (1947). Morphologic details were well preserved and photomicrographs of well-spread chromosomes in one plane were obtained easily.  相似文献

It is well known that there is a strong influence of fixation, i.e., acetic methanol versus formaldehyde, on the chromosome morphology at stages of the first meiotic division. In this study the influence of both these types of fixation on the morphology of mitotic chromosomes was examined in human lymphocytes. After methanol-acetic acid (3:1) fixation, the chromosomes show the "classical" condensed shape in which it is not always possible to recognize the two sister chromatids. These chromosomes are accessible to the conventional G-, R-, and C-banding techniques. After formaldehyde fixation at a relatively high pH, the chromosomes are thinner and longer (two to six times) when compared with chromosomes following methanol-acetic acid fixation. They show a scaffold-like morphology, sometimes with a halo of thin material around it. In all cases the two sister chromatids could be recognized. This chromosome structure could be easily stained with silver, Giemsa, 4,6-diamino-2-phenyl-indole (DAPI), and fluorescein isocyanate isomere 1 (FITC). The results obtained following these stainings gave no indication to any specific chemical composition of a probable central scaffold. The scaffold-like structures were not accessible to G-, R-, or C-banding techniques. The only effect observed following these banding techniques was the disappearance of the halo of thin material around the central scaffold-like structure.  相似文献

Rats were surgically made bilaterally cryptorchid and after 4-8 days the testes were returned to the scrotum. After 70 days fertility was tested by pairing with females. Fertility was restored in 5/6 rats with testes cryptorchid for 4 or 5 days, but only 2/9 were fertile when the duration of cryptorchidism was 6-8 days. The sterility was due to irreversible degeneration of the spermatogonial stem cells. The testes of infertile males were smaller and lighter than those of fertile males and the seminiferous tubule diameters were reduced.  相似文献

We study short-range correlations in DNA sequences with methods from information theory and statistics. We find a persisting degree of identity between the correlation patterns of different chromosomes of a species. Except for the case of human and chimpanzee inter-species differences in this correlation pattern allow robust species distinction: in a clustering tree based upon the correlation curves on the level of individual chromosomes distinct clusters for the individual species are found. This capacity of distinguishing species persists, even when the length of the underlying sequences is drastically reduced. In comparison to the standard tool for studying symbol correlations in DNA sequences, namely the mutual information function, we find that an autoregressive model for higher order Markov processes significantly improves species distinction due to an implicit subtraction of random background.  相似文献

本文研究了五种稻蝗Oxya chinensis、O. shanghaiensis、O. adentata、O. hyla intricara、O. agavisa的染色体核型和C带带型.玻片制备采用压片法,BSG法C带处理.结果表明:五种稻蝗染色体核型差异较小,具有属的保守性,但在C带方面则显示出一定的差别,种间染色体带型的差异大小与形态学上的差异程度具明显的正相关.其中小稻蝗和山稻蝗各自具有独特的带型结构,与其余三种显然不同,而中华稻蝗、上海稻蝗、无齿稻蝗虽在主要标志性染色体上带纹趋于一致,但在一些细微结构和统计数据方面存在一定差异.本文对五种稻蝗减数分裂中期Ⅰ的染色体数目和交叉定位也作了统计和比较,在染色体行为分析方面做了初步工作.据此,本文讨论了五种稻蝗的染色体分类问题.  相似文献

In Nysius californicus (family Lygaeidae, subfamily Orsillinae), a pest commonly known as the seed bug, the chromosome complement is 2n = 16 (12A + 2m + XY), testes are formed by seven seminiferous tubules covered by an orange-colored membrane, and spermatogenesis is cystic. At prophase, sex chromosomes are heteropycnotic and autosomes usually show a chiasma. At metaphase, sex chromosomes along with microchromosomes may be seen located at the center of a ring formed by the remaining autosomes. A characteristic specific of N. californicus was the presence of nucleolar material observed from the cystic cell to the completely differentiated spermatozoon. Variations in size, shape and location of the nucleolar material occur during this process, denoting a variable degree of activity in the different stages.  相似文献

The organisation of cytokeratin filaments in mitotic HeLa cells has been analysed by immunofluorescence microscopy using a monoclonal antibody which recognises proteins with apparent subunit molecular weights of 52 kDa and 57 kDa and which binds exclusively to cytokeratin-type filaments. Mitotic cells were prepared for microscopic analysis by hypotonic swelling, centrifugation onto glass slides, brief pre-extraction with 0.1% Triton X-100 and fixation in 80% ethanol. This procedure gave particularly good resolution of intermediate filaments and preservation of chromosome morphology. In prometaphase-metaphase cells the antigen was present in an anastomosing filament network which completely or partially enclosed the chromosomes, in filament fragments and in cytoplasmic aggregates. The epichromosomal filament network was absent from cells in anaphase or later stages of mitosis. In these cells non-filamentous antigen was often located in a narrow band defining the periphery of individual chromosomes and in variable numbers of cytoplasmic filaments or fragments. The results suggest that extensive disaggregation and reformation of cytokeratin filaments occurs during mitosis and that disaggregated cytokeratin proteins are frequently located adjacent to mitotic chromosomes.  相似文献

The objective of the present research was to quantify the seminiferous epithelium cells, spermatogenesis efficiency and characterize the ultrastrucure of Sertoli cells in goats. Eighteen goats were used and divided into three groups: Group I - goats without bipartition of the scrotum; Group II - animals with bipartition of the scrotum in up to 50% of the testicular length; Group III - goats with bipartition of the scrotum in more than 50% of the testicular length. The goat testes in Group III had a greater number of primary spermatocytes (25.37 ± 4.55 cells per cross sections), spermatids (112 ± 15.12 cells per cross sections), and Sertoli cells (9.46 ± 1.74 cells per cross sections) than the animals in Groups I and II (P<0.05). The spermatogenic mitotic, meiotic, and general efficiency were greater in animals in Group III (1.25 ± 0.28; 5.12 ± 1.63; 6.44 ± 1.96) when compared to those in Groups I and II. Sheet-like processes originated from the Sertoli cell body as simple and smooth structures which involved almost all the surface of germ cells. Slender cord-like processes originated from Sertoli cells and also from the sheet-like processes. The relative frequency of the cycle stages showed differences among the groups of goats studied, and the highest frequency was in Stage 3 (20.68% for goats in Group I, 21.15% for those in Group II, and 16.89% for the animals in Group III). In conclusion, goats with bipartition of the scrotum have a greater number of germ and Sertoli cells per cross section of seminiferous tubule, that indicated a greater sperm production when compared to the other groups, and the ultrastructure of the Sertoli cell process did not present any relationship with bipartition of the scrotum.  相似文献

The study of spermatogenesis in Hymenoptera is limited to few taxa due to limitations to access the material in the right stage for analysis. The information available up to now states that the gametogenesis in males involves a modified meiosis, similar to mitosis. This is due to the haplodiploid sex determination system present in the order, where females are diploid and males are haploid. In the present work, we examined the spermatogenesis process in the parasitoid Diachasmimorpha longicaudata (Ashmead) (Hymenoptera: Braconidae). We performed squash preparations from testes of individuals from the third larval instar until the adult stage. Some cytological traits, such as the presence of a monopolar spindle, were analysed in detail by means of histological semi‐thin sections. The spermatogenesis in this species comprises an abortive first division with the formation of an anucleated cytoplasmic bud and a nucleated cell, and an equal second division of the nucleated cell, leading to the generation of two mature spermatozoa. The results presented herein provide more information about the process of spermatogenesis in Hymenoptera, which has not been studied in depth in parasitic species.  相似文献

A schedule is given which produced excellent results for somatic chromosome counts on some British species of Galium, These species present great difficulties owing to the small size of the chromosomes (ca. 1.5μ long), the large somatic numbers (up to 96) and the slenderness of the root tips. Special features of the schedule, which is the result of much experiment with various technics, are: fixation with Belling's Navashin type fixative, which was quite the best tried; the modification of Randolph's card mount method to overcome the difficulty of the small diameter of the root tips by mounting together a number from the same plant, so that they can be embedded and sectioned almost as easily as much larger root tips; staining with dilute (0.1%) crystal violet, the most critical stain used, after mordanting with 1% aqueous chromic acid to intensify the stain in the small chromosomes; and the addition of an extra stage of differentiation in absolute alcohol diluted with xylol to remove strands of stain, which are often left in the cytoplasm between the chromosomes, since clove oil, usually the last differentiating fluid used, differs from alcohol in removing crystal violet more rapidly from the chromosomes than from the cytoplasm. It is suggested that this schedule will be valuable in chromosome counts of other plants where similar difficulties arise.  相似文献

Homoeologous relationships of rice, wheat and maize chromosomes   总被引:34,自引:0,他引:34  
A set of cDNA clones, which had previously been mapped onto wheat chromosomes, was genetically mapped onto the chromosomes of rice. The resulting comparative maps make it possible to estimate the degree of linkage conservation between these two species. A number of chromosomal rearrangements, some of which must have involved interchromosomal translocations, differentiate the rice and wheat genomes. However, synteny of a large proportion of the loci appears to be conserved between the two species. The results of this study, combined with those from a recently published comparative map of the rice and maize genomes, suggest that rice, wheat and maize share extensive homoeologies in a number of regions in their genomes. Some chromosomes (e.g. chromosome 4 in rice, chromosomes 2 and 2S in wheat and maize, respectively) may have escaped major rearrangement since the divergence of these species from their last common ancestor. Comparative maps for rice, wheat and maize should make it possible to begin uniting the genetics of these species and allow for transfer of mapping information (including centromere positions) and molecular marker resources (e.g. RFLP probes) between species. In addition, such maps should shed light on the nature of chromosome evolution that accompanied the radiation of grasses in the early stages of plant diversification.  相似文献
