The conformation of a benzodiazepine-like decapeptide corresponding tothe YLGYLEQLLR fragment of a casein has been examined in a sodium dodecylsulfate micellar medium using circular dichroism, two-dimensional1H NMR spectroscopy and restrained molecular dynamicssimulation. Summary A search algorithm, called MEDUSA, is presented which allows the determination of multiple conformations of biomolecules in solution with exchange rate constants typically between 103 and 107 s–1 on the basis of experimental high-resolution NMR data. Multiples of structures are generated which are consistent as ensembles with NMR cross-relaxation rates (NOESY, ROESY), scalar J-coupling constants, and T1p measurements. The algorithm is applied to the cyclic decapeptide antamanide dissolved in chloroform. The characteristic radio-frequency field dependence of the T1p relaxation rates found for the NH protons of Val1 and Phe6 can be explained by a dynamical exchange between two structures.  相似文献

Prostaglandin I2 potentiated the paw swelling induced by carrageenin in rats. Prostaglandin I2 (0.1 μg) showed similar activity to PGE1 (0.01 μg). This potentiating property disappeared in 60 minutes and was completely abolished by diphenhydramine (25 mg kg−1, i.p.). In vascular permeability tests, PGI2 itself (2.5 × 10−10 mol, 88 ng) caused no dye leakage reaction, but PGE1 (2.5 × 10−10 mol, 88.5 ng) caused a significant dye leakage. This effect of PGE1 was statistically significant compared with vehicle- or PGI2-treated group (p<0.05). Prostaglandin I2 potentiated the increased vascular permeability induced by 5-hydroxytriptamine (2.5 × 10−10 mol), bradykinin (5 × 10−10 mol) and histamine (2 × 10−10 to 2 × 10−8 mol). The potentiation was the most evidence in the case of histamine.  相似文献

Summary 1. The decapeptide neurohormone gonadotropin releasing hormone (GnRH) is the first key hormone of the reproductive system. Produced in the hypothalamus, GnRH is released in a pulsatile manner into the hypophysial portal system to reach the anterior pituitary and stimulates the release and synthesis of the gonadotropin hormones LH and FSH. GnRH, a Ca2+ mobilizing ligand, binds to its respective binding protein, which is a member of the seven transmembrane domain receptor family and activates a G-protein (Gq).2. The subunit of Gq triggers enhanced phosphoinositide turnover and the elevation of multiple second messengers required for gonadotropin release and biosynthesis.3. The effect of the rhenium complex cis-[Re2GABA2Cl4]Cl2 on the antioxidant parameters of normal human blood in vitro have been studied. The results suggest that the complex influences various enzymes in the cascade of reactions utilizing active oxygen metabolites. However, the manifestation of this activity varies over the studied concentration range of the complex in the preincubation medium (10–12-10–4 M), so the effects appear to be concentration-dependent. The largest differences in antioxidant parameters in comparison with control were observed for the concentrations 10–8, 10–5, and 10–4 M. Thus, correlations between the peroxidation level, superoxide dismutase (SOD) activity, antioxidant factor (F), and indexes of resistance of erythrocytes for hemolysis (TR) were found.  相似文献

The reaction of desferrioxamine with superoxide has been studied using the pulse radiolysis technique. The decay of O2- was not accelerated in the presence of up to 4 × 10-4 M desferrioxamine at physiological pH. The rate constant was found to be lower than 2 × 104 M-1 S-1. In acid solutions the rate constant of the reaction between desferrioxamine and HO2 was found to be lower than 105 M-1 S-1. The reaction was not studied in alkaline solutions due to the high absorbance of desferrioxamine in the U. V. region. The pK of desferrioxamine was determined to be 9.2 ± 0.05.  相似文献

Summary 16,16-Dimethyl PGE2 (dmPGE2) has previously been shown to protect the in vivo rat liver against CCl4-induced damage. These studies were undertaken to determine if this protection could be demonstrated in vitro where factors of absorption, secretion, and blood flow are not present. Primary hepatocyte cultures were established by perfusing rat liver with collagenase. Hepatocytes were plated at a density of 2×104 cells/cm, allowed 90 min to attach, then stabilized in L15 medium for 18 h. Hepatocytes were then challenged with CCl4 with concomitant exposure to 10−9 to 10−5 M dmPGE2, stearic acid, oleic acid, or ethanol vehicle (0.00001 to 0.1%). After 1 h, challenge was aspirated and cells were stained with 0.04% trypan blue to determine viability. Hepatocytes in the vehicle groups took up more trypan when exposed to CCl4 than those treated with dmPGE2, stearic acid, or oleic acid at concentrations of 10−9 to 10−7 M. At 0.1% ethanol vehicle protected as well as all other treatments. Protection against CCl4 by dmPGE2, stearic, and oleic acids as well as high concentrations of ethanol may occur by altering the metabolism of CCl4.  相似文献

The effect of platelet activating factor (PAF), a potent lipid mediator of inflammation, was examined in the induction of airway hyperreactivity to known mediators of anaphylaxis. Concentration-dependent contractions of the isolated guinea-pig trachea to PAF (10−7 − 10−5M) were produced and an EC50 value was found to be 7.5 × 10−7M. Pretreatment for 30 min with a known PAF inhibitor, CV-3988 (10−5 or 10−4M), produced significant inhibition of PAF contractions; however, at 10−6M, CV-3988 had no effect. In the presence of meclofenamic acid (10−6M), the concentration-response curve to PAF was shifted significantly upward and to the left. This potentiation could be reversed by pretreating the tissues with the peptidoleukotriene antagonists, FPL 55712 or SK&F 102922 (10−5M). Pretreatment with PAF concentrations having essentially no intrinsic activity (10−8, 10−7) significantly enhanced the contraction of guinea-pig trachea to various concentrations of LTD4 and to certain concentrations of a thromboxane mimic (U-46619). Pretreatment with lyso-PAF failed to potentiate the LTD4 response, while pretreatment with CV-3988 reverse the potentiation by PAF of the lower concentrations of LTD4. However, PAF failed to enhance contractions (with or without the presence of meclofenamic acid) to acetylcholine, histamine, PGD2 or LTC4 (in the presence of serine borate). These results indicate a possible role for PAF as a mediator of airway hyperreactivity.  相似文献

1,25D3 is critical for the maintenance of normal reproduction since reduced fertility is observed in male rats on a vitamin D-deficient diet. Vitamin D-deficient male rats have incomplete spermatogenesis and degenerative testicular changes. In the present study we have examined the ionic involvement and intracellular messengers of the stimulatory effect of 1,25D3 on amino acid accumulation in immature rat testis. 1,25D3 stimulates amino acid accumulation from 10−12 to 10−6 M by increasing the slope to reach a maximum value at 10−10 M, as compared to the control group. No effect was observed at a lower dose (10−13 M). Time-course showed an increase on amino acid accumulation after 15, 30, and 60 min of incubation with 1,25D3 (10−10 M). 1,25D3 stimulated amino acid accumulation in 11-day-old rat testis but not in testis that were 20 days old. Cycloheximide totally blocked the 1,25D3 action on amino acid accumulation. Furthermore, a localized elevation of cAMP increased the stimulatory effect of 1,25D3 and the blockage of PKA nullified the action of the hormone. In addition, 1,25D3 action on amino acid accumulation was also mediated by ionic pathways, since verapamil and apamine diminished the hormone effect. The stimulatory effect of 1,25D3 on amino acid accumulation is age-dependent and specific to this steroidal hormone since testosterone was not able to change amino acid accumulation in both ages studied. This study provides evidence for a dual effect for 1,25D3, pointing to a genomic effect that can be triggered by PKA, as well as to a rapid response involving Ca2+/K+ channels on the plasma membrane.  相似文献

The purpose of this study was to localize vasopressin (VP) V1a receptor in stomach and to characterize the role of VP in the regulation of gastric motility in rats. Double staining was used to locate the V1a receptor in the gastric body of the rat. The contraction of the circular muscle strips of gastric body was monitored by a polygraph. V1a receptor was expressed on the neurons of myenteric plexus of the gastric body. VP (10− 10–10− 6 M) caused a concentration-dependent contractile effect on the circular muscle strips of gastric body in vitro. V-1880 ([deamino-Pen1, O-Me-Tyr2, Arg8]-Vasopressin, 10− 7 M), a V1 receptor antagonist, inhibited the spontaneous contraction of the strips. Tetradotoxin (TTX, 10− 6 M) and V-1880 (10− 7 M) abolished the excitatory effect of VP. Atropine (10− 6 M) partially inhibited VP-induced excitatory effect on the muscle strips but hexamethonium (10− 4 M) did not influence it. These results suggest that V1a receptor was expressed on the neurons of myenteric nerves. The cholinergic nerve was involved in the excitatory effect of VP on the contraction of gastric body.  相似文献

The direct immobilization of glucose oxidase (GOD) on TiO2/SiO2 nanocomposite and its application as glucose biosensor were investigated. The room-temperature phosphorescence of TiO2/SiO2 nanocomposite can be quenched by hydrogen peroxide (H2O2). The detection of glucose may be accomplished by monitoring the formation of hydrogen peroxide which generated in the oxidation process of glucose with the catalysis of GOD. To our surprise, by using a 96-hole polyporous plate accessory of fluorescence spectrophotometer, the biosensor exhibits excellent linear response to glucose concentrations ranging from 1.0 × 10−9 to 1.0 × 10−2 M with a detection limit of 1.2 × 10−10 M. The TiO2/SiO2 nanocomposite can be used as both supporting material and signal transducer. The phosphorescence intensity and color of the biosensor change obviously and even could be observed with naked eyes by continuous addition of glucose. Based on the room-temperature phosphorescence of TiO2/SiO2 nanocomposite, a new method of solid substrate-room-temperature phosphorimetry (SS-RTP) for glucose determination is proposed. A glucose biosensor was fabricated with wide determination concentration range, low detection limit, high sensitivity, and fast response time. And the biosensor has been successfully applied to the determination of glucose in human blood serum. The coacervation of GOD enzyme and its interaction with TiO2/SiO2 nanocomposite enlarge the surface area and enhance the chemical stability of GOD. The nice biocompatibility, large surface area, good chemical stability and nontoxicity of the TiO2/SiO2 nanocomposite have made this material suitable for functioning as biosensor.  相似文献

Three group 10 complexes containing nido-carborane diphosphine, [NiCl(PPh3){7,8-(PPh2)2-7,8-C2B9H10}] (1), [PdCl(PPh3){7,8-(PPh2)2-7,8-C2B9H10}] · 1.25CH2Cl2 (2) and [PtCl(PPh3){7,8-(PPh2)2-7,8-C2B9H10}] · 2.5CH2Cl2 (3) have been synthesized by the reactions of [M(PPh3)2Cl2] (M = Ni, Pd, Pt) with closo carborane diphosphine 1,2-(PPh2)2-1,2-C2B10H10 in ethanol. For complex 3, it could also be obtained under solvothermal condition. All three complexes were characterized by elemental analysis, FT-IR, 1H and 13C NMR spectroscopy and X-ray structure determination. Single crystal structures show that their structures are similar to each other. In each complex, the nido [7,8-(PPh2)2-7,8-C2B9H10], which resulted from the degradation of the initial closo ligand 1,2-(PPh2)2-1,2-C2B10H10 during the reaction process, was coordinated bidentately through the P atoms to M(II) ion, and this resulted in a stable five-membered chelating ring between the bis-diphosphine ligand and the metal. The coordination mode of the metal can be described as a slightly distorted square-planar, in which the remaining two positions were occupied by one Cl and one PPh3 group.  相似文献

Several amino terminal fragments of the emerimicins (Ac-Phe1-Aib2-Aib3-Aib4-Val5-Gly6-Leu7-Aib8-Aib9-Hyp10-Gln11-D-Iva12-Hyp13-Ala/Aib14-Phol15) ranging in length from five to ten residues have been synthesized. Nuclear magnetic resonance studies have been carried out on the 1-5, 6-10, 1-6, 1-7, 1-8, 1-9, and 1-10 fragments. The number of solvent-shielded NH groups in CDCl3 solutions for 1-5, 1-6, 1-7, 1-8, 1-9, and 1-10 indicate that 310-helical structures are favored in this solvent. In (CD3)2SO, an additional NH group, assigned to Aib(3) NH is solvent exposed in the fragments longer than six residues, suggesting partial unfolding of the N-terminal β-turn or transition to an α-helical conformation. The data for fragment 6-10 are consistent with a conformation having a single Leu-Aib β-turn. Infrared studies suggest an increase in the number of intramolecular hydrogen bonds with increasing peptide chain length. Appreciable mitochondrial uncoupling activity is observed for peptides with a chain length of at least seven residues. The order of efficiencies of the fragments is 1-7 < 1-8 ~ 1-10 < 1-9, with the decapeptide exhibiting anomalously low uncoupling activity.  相似文献

The sulfate-reducing Archaeoglobus fulgidus contains a number of enzymes previously thought to be unique for methanogenic Archaea. The purification and properties of two of these enzymes, of formylmethanofuran: tetrahydromethanopterin formyltransferase and of N 5,N 10-methylenetetrahydromethanopterin dehydrogenase (coenzyme F420 dependent) are described here. Hydrogen peroxide (H2O2)-induced aggregation of calf platelets and its modification by agents with specific properties were characterized employing a spectrophotometric assay. An Arrhenius activation energy of 20 ± 1 kcal/mol was found in the temperature range of 25‡-36‡C. Rate inhibition occurred on either side of this temperature range, and under anaerobic conditions. Exogenous Ca2+ ions were not required but Ca2+ ions, at 1 mM-concentration, optimally increased rates and extent of aggregation at suboptimal H2O2 concentrations but only extent of aggregation at optimal H2O2 concentrations. Ba2+, Sr2+, Cd2+, Mn2+ and Ni2+ ions (1 mM) and Zn2+, Pb2+ and Hg2+ ions (10 mM) were inhibitory. The cyclo-oxygenase inhibitor, indomethacin (10-30 mM) exerted only mild inhibition by a competitive mechanism. Another cyclo-oxygenase inhibitor, aspirin, functioned to increase aggregation. Ligands acting directly at the prostaglandin H2/thromboxane A, receptor (5Z. 9, 11, 13E, 15(S) 15-hydroxy 9(11) epoxy methano prosta 5, 13-dien-1-oic acid, pinane thromboxane A2, arachidonic acid, eicosapentaenoic acid, and N-ethylmaleimide) functioned as competitive inhibitors. Another platelet-activating sulphydryl reagent, thimerosal, also inhibited competitively while the protein kinase C inhibitor, sphingosine, and the protein kinase C modulator, Zn2+ ions, inhibited by different mechanisms. The results indicate direct action of H2O2 at the prostaglandin H2/thromboxane A2 receptor, possibly its sulphydryls, to activate the protein kinase C pathway, independently of cyclo-oxygenase products. The results underscored the power of the kinetic approach for investigating mechanisms of platelet activation.  相似文献

Methods including spectroscopy, electronic chemistry and thermodynamics were used to study the inclusion effect between γ-cyclodextrin (CD) and vitamin K3(K3), as well as the interaction mode between herring-sperm DNA (hsDNA) and γ-CD-K3 inclusion complex. The results from ultraviolet spectroscopic method indicated that VK3 and γ-CD formed 1:1 inclusion complex, with the inclusion constant Kf = 1.02 × 104 L/mol, which is based on Benesi–Hildebrand's viewpoint. The outcomes from the probe method and Scatchard methods suggested that the interaction mode between γ-CD-K3 and DNA was a mixture mode, which included intercalation and electrostatic binding effects. The binding constants were K θ25°C = 2.16 × 104 L/mol, and Kθ37°C = 1.06 × 104 L/mol. The thermodynamic functions of the interaction between γ-CD-K3 and DNA were ΔrHmθ = ?2.74 × 104 J/mol, ΔrSmθ = 174.74 J·mol?1K?1, therefore, both ΔrHmθ (enthalpy) and ΔrSmθ (entropy) worked as driven forces in this action.  相似文献

This work reports a novel signal amplification strategy based on three-dimensional ordered macroporous C60-poly(3,4-ethylenedioxythiophene)-1-butyl-3-methylimidazolium hexafluorophosphate (3DOM C60-PEDOT-[BMIm][BF6]) for ultrasensitive detection of thrombin by cascade catalysis of Au-PEDOT@SiO2 microspheres and alcohol dehydrogenase (ADH). Au-PEDOT@SiO2 microspheres were constructed not only as nanocarriers to anchor the large amounts of secondary thrombin aptamers but also as nanocatalysts to catalyze the oxidation of ethanol efficiently. Significantly, the electrochemical signal was greatly enhanced based on cascade catalysis: First, ADH catalyzed the oxidation of ethanol to acetaldehyde with the concomitant generation of NADH in the presence of β-nicotinamide adenine dinucleotide hydrate (NAD+). Then, gold nanoparticles (AuNPs) as nanocatalysts could effectively catalyze NADH to produce NAD+ with the help of PEDOT as redox probe. Under the optimal conditions, the proposed aptasensor exhibits a linear range of 2 × 10−13 to 2 × 10−8 M with a low detection limit of 2 × 10−14 M for thrombin detection and shows high sensitivity and good specificity.  相似文献

In order to replace the conventional chemical pesticides, extensive researches have been done on entomopathogenic fungi. Entomopathogenic fungus Beauveria bassiana is an important biocontrol agent against major economic pests and is being employed in Integrated pest management (IPM) along with synthetic pesticides. Cabbage aphid Brevicoryne brassicae L. is one of the important pests of Brassicaceae family. Therefore, in this research, the virulence isolate of B. brassicae (IRAN 429C) was investigated on adults of cabbage aphid under laboratory conditions. The experiments were conducted at 25 ± 2 °C, 60 ± 10 R. H. and a photoperiod of 16:8 (L: D). After preliminary experiments, the adult aphids were treated with fungal concentrations of 1 × 103 to 1 × 107 spores/ml. Probit analysis was conducted to calculate LC50 and LC95 values for the isolate. Positive correlation was observed between concentrations and pest mortality. LC50 and LC95 values calculated for IRAN 429C isolate are 2.04 × 105 and 1.82 × 108, respectively. The mortality was counted one day after the treatment and then continued for 14 days. Cumulative mortality for 14 days after treatment varied from 54% for IRAN 429C at low concentration (103 conidia/ml) to 83% at high concentration (107 conidia/ml). The lowest LT50 was obtained at 7.67 days for IRAN 429C isolate at concentration 1 × 107 spore/ml. According to the insecticidal activity of mentioned fungi on cabbage aphid, it can be used in biocontrol programmes of B. brassicae.  相似文献

The diamondback moth, Plutella xylostella (L.) (Lepidoptera: Plutellidae) is one of the most important pests of cruciferous plants throughout the world. In recent years, this insect has been a serious pest for cabbage fields in Tehran province. Resistance of P. xylostella to all main groups of insecticides has been recorded and it is ranked in the 20 most resistant pest species reported up to now. According to many researchers, to eliminate the problem of pest resistance to chemical pesticides, an integrated pest management programme should be used. In line with this, the uses of microbial control agents (MCAs) are discussed. The bacterium, Bacillus thuringiensis (Bt) is one of microbial control agents of pests. It is characterised by its ability to produce proteic crystalline inclusions during sporulation. Cry1 protein has insecticidal activity and is highly specific to certain insects and not toxic to unrelated insects, plants or vertebrates. In this work, the pathogenicity of some Bt isolates, including Dipel, 20, 29, 79 and 87, was tested against P. xylostella and the lethal concentrations (LC50) of their crystal proteins to P. xylostella third larval instar was determined. The experiment was designed in factorial in randomised complete design with 5 treatments (different concentrations including 104, 105, 106, 107, 108 CFU/ml and 5 replications and with 10 third larval instars. Spore–crystal complex was applied to the surface of natural diets (cabbage leaves) and the mortality of P. xylostella larvae was assessed 120?h after exposure of Bt toxin in each treatment. Results showed that percentage of survival was significantly higher for control treatment. Results also showed that after 5?days, LC50 for isolates of Dipel, 20, 29, 79 and 87 were equal to 1?×?106, 1?×?105, 5?×?105, 4?×?105 and 1?×?104 CFU/ml, respectively. LT50 were equal to 93.71, 48.04, 71, 40.49 and 75.28?h. Of and most the percentage larval mortality relate to attendance 87 and also at least percentage mortality is related to the groom Dipel.  相似文献

The objective of this study was to evaluate the human NCI-N87 cell line as a model for gastric permeability drug studies under pH conditions of the stomach. The optimal conditions that led NCI-N87 cells to form a typical differentiated gastric epithelial barrier were a seeding density of 2.5 × 105 cells/cm2 on porous inserts and growth in serum-complemented RPMI-1640 medium until 18–27 days post-confluency. The resulting cell monolayers showed moderately high transepithelial electrical resistance (TEER) values of about 500 Ω cm2, cells of polygonal morphology expressing E-cadherin and ZO-1 proteins at their contact surfaces, and production of mucus clusters. The monolayers withstood apical pH of 7.4 down to 3.0 with the basal pH fixed at 7.4. The apparent permeability coefficients (Papp) of model compounds were evaluated in the apical-to-basolateral and basolateral-to-apical directions under different pH gradients. The monolayers were impermeable to the integrity marker Lucifer Yellow (low Papp of 0.3–1.1 × 10−6 cm/s). The furosemide Papp (0.4–1.5 × 10−5 cm/s) were slightly dependent on pH but remained moderate. The caffeine Papp (4.2–5.0 × 10−5 cm/s) were higher and insensitive to pH changes. The NCI-N87 cell line provides a useful in vitro tool to assess gastric drug permeability and absorption under physiologic conditions prevailing in the human stomach.  相似文献
