ABSTRACT. A new diplomonad flagellate, Spironucleus vortens n. sp., is described from the intestinal lumen of the freshwater angelfish, ( Pterophyllum scalare ), bred in Florida. Live organisms are pyriform, and measure 12.5–20.5 μm long by 5.0–11.2 üm wide. Scanning electron microscopy shows that the trophozoite bears two compound lateral longitudinal ridges, each originating posterior to three emerging anterior flagella, and continuing posteriorly to the emergence of the posterior flagellum. Each ridge comprises a broad central part, surrounded by a peripheral ridge. At the opening of the flagellar pocket, the broader right peripheral ridge crosses to the other side of the body, and then back again. The posterior end of the body bears two papillae. Transmission electron microscopy shows that the compound lateral ridges are supported by microtubules, and bear microfibrillar structures in discrete longitudinal plaques. The serendipitous growth of S. vortens in a culture system with lip tumor tissue, facilitated axenic cultivation in a modified TYM medium (trypticase, yeast extract, maltose). The flagellate is now routinely maintained in an axenic TYI-S-33 medium (trypticase, yeast extract, iron serum), and is stabilized in the cryopreserved state. Spironucleus vortens is an aerotolerant anaerobe that can be cultured at 25° C, 28° C and 30° C.  相似文献

ABSTRACT. A hexamitid flagellate Spironucleus barkhanus n. sp., from the lumen of the gut and gall bladder of wild grayling Thymallus thymallus , and from muscle abscesses of farmed Atlantic salmon Salmo salar from Norway, is described by light, scanning and transmission electron microscopy. The flagellate was axenically cultured in trypticase, yeast extract, iron serum medium. Live trophozoites from axenic cultures incubated at 5° C, measure 11–20 x 6–14 μm. The flagellates show a typical bi-radial symmetry. Each recurrent flagellum is almost completely surrounded by a striated lamina. In the posterior end the lamina widens, appearing heart shaped in transverse section. Accompanying each recurrent flagellum are three narrow bands of microtubules, following the longitudinal groove created by the incomplete closure of the striated lamina. The recurrent flagella emerge posterio-medially through cytostome openings halfway surrounded by crescent-shaped ridges, oriented in opposite directions in the two openings. The position and adornment of the cytostome openings, and the arrangement and number of the microtubules accompanying the recurrent flagella, distinguish Spironucleus barkhanus n. sp. from previously described species of Spironucleus .  相似文献

Hole-in-the-head disease is recorded in 11 discus Symphysodon discus Heckel, 1840 and 1 angelfish Pterophyllum scalare Lichtenstein, 1823 obtained from local aquarists within the Southwest of the UK. Spironucleus vortens Poynton et al. 1995, was isolated from the kidney, liver, spleen and head lesions of discus showing severe signs of the disease and from the intestines of all fish. The hexamitid was also recorded from the head lesions of the angelfish. The identity of these flagellates was confirmed as S. vortens on the basis of topographical features seen with the aid of SEM. A modified in vitro culture method was successfully developed for the detection, isolation and long-term maintenance of S. vortens. The flagellate was sub-cultured at 3 to 5 d intervals, new media being supplemented with fresh liver from Oreochromis niloticus (Linnaeus, 1757) free from infection. The results are discussed in relation to S. vortens as the causative agent for hole-in-the-head disease following systemic infection via the digestive tract.  相似文献

Diplomonad flagellates infect a wide range of fish hosts in aquaculture and in the wild in North America, Asia and Europe. Intestinal diplomonad infection in juvenile farmed trout can be associated with morbidity and mortality, and in Germany, diplomonads in trout are commonly reported, and yet are poorly characterised. We therefore undertook a comprehensive study of diplomonads from German rainbow trout Oncorhynchus mykiss, using scanning and transmission electron microscopy, and sequencing of the small subunit (ssu) rRNA gene. The diplomonad was identified as Spironucleus salmonis, formerly reported from Germany as Hexamita salmonis. Our new surface morphology studies showed that the cell surface was unadorned and a caudal projection was present. Transmission electron microscopy facilitated new observations of functional morphology, including vacuoles discharging from the body surface, and multi-lobed apices of the nuclei. We suggest the lobes form, via hydrostatic pressure on the nucleoplasm, in response to the beat of the anterior-medial flagella. The lobes serve to intertwine the nuclei, providing stability in the region of the cell exposed to internal mechanical stress. The ssu rRNA gene sequence clearly distinguished S. salmonis from S. barkhanus, S. salmonicida, and S. vortens from fish, and can be used for identification purposes. A 1405 bp sequence of the ssu rRNA gene from S. salmonis was obtained and included in a phylogenetic analysis of a selection of closely related diplomonads, showing that S. salmonis was recovered as sister taxon to S. vortens.  相似文献

Spironucleus vortens were cultivated in either an artificial medium at different temperatures, or in medium at various pH conditions or supplemented with different bile concentrations at 25 degrees C. Temperature, pH and bile requirements for the optimal growth of the parasite were determined. Parasites multiplied quickly at 28 and 31 degrees C and reached maximum numbers on Day 4 of cultivation, whereafter they did not survive. At 25 degrees C, parasites survived longer than those at 28 and 31 degrees C with no difference in multiplication rate during the exponential phase. The longest survival period was seen at 22 degrees C, although the growth rate of the parasite was not as high as those at 25 degrees C. At a higher temperature of 37 degrees C, no parasites were observed alive after the second day of cultivation. Optimal pH range for the parasite's growth was 6.5 to 7.5, with the highest cell number at pH 7.5. Parasites survived longest (15 d) at pH 6.0, although the maximum number of cells was lower than those at the optimal pH. Parasites were dead within 24 h at pH levels above 8.5 or below 5.5. All cultures supplemented with either bovine or fish bile yielded numbers of parasites lower than cultures with no bile. In addition, parasite growth was significantly suppressed in medium supplemented with higher concentrations of bile. These results indicate that the optimal condition for the in vitro cultivation of S. vortens is 25 degrees C and pH 6.5 to 7.5 without supplementation with bile.  相似文献

This is believed to be the 1st confirmed report of Spironucleus from a marine fish. Organisms swimming in the rectal lumen were broadly pyriform to elongate, and measured 10.5-18.6 microns long and 3.2-13.3 microns wide; other elongate organisms were attached to the rectal epithelium, via apical extensions appearing continuous with the microvilli. The posterior end of the body was extended into a caudal projection, on either side of which was a posteriolateral ring-shaped protrusion or torus, with a recurrent flagellum emerging from its centre. A symmetrical system of microtubules and lamellae, forming a "V" in protargol impregnated specimens, supported the flanges of the body surrounding the tori, the tori themselves and the caudal projection. Supranuclear microtubules were an inverted V to U shape in transverse section, and an electron dense band accompanied the cytostomes. Lightly staining homogenous cytoplasm was usually present in the anterior part of the body, the remainder being highly vacuolated with numerous dark granules. In swimming organisms, rough endoplasmic reticulum (RER) was present around the nuclei and cytostomes, and bacteria were occasionally seen in the cytoplasm. In "attached" organisms, RER was reduced, and bacteria were absent. Hexamita salmonis Moore from Salvelinus fontinalis was studied by light and scanning electron microscopy for comparison; its cytoplasm was not highly vacuolated. A case of intracellular systemic infection with the diplomonad flagellate Spironucleus barkhanus in farmed Arctic char Salvelinus alpinus is described. The parasites were widely disseminated throughout the vasculature and in most organs. Aggregates of the parasites were seen within well-defined structures regarded as host cells in capillaries and sinusoids of the liver, spleen and head kidney. Intracellular infection with Spironucleus spp. has never previously been reported. The prevalence of infection and mortality in the affected farm was low. In contrast to systemic spironucleosis in farmed Atlantic salmon, and despite huge numbers of flagellates in the vasculature, the tissues of the organs were remarkably unaffected. The relatively few gross and histopathological lesions may indicate that Arctic char are more tolerant to this parasite than Atlantic salmon.  相似文献

Spironucleus salmonicida is a diplomonad flagellate known to cause systemic infections in farmed salmonids. In northern Norway, outbreaks of spironucleosis in farmed Atlantic salmon Salmo salar have been a recurring problem. Common to all these outbreaks was the origin of smolts: all came from the same farm. In the present study, wild Arctic char Salvelinus alpinus and brown trout Salmo trutta were sampled from the lakes used as a water source for the smolt supplier. In addition, smolt and three-spined sticklebacks Gasterosteus aculeatus were sampled from the smolt farm. Bile and intestinal contents from the sampled fish were examined by light microscopy and PCR. Spironucleus salmonicida was identified in both wild Arctic char and brown trout from the lakes used as water sources by the smolt farm, suggesting that the farmed fish were exposed to this pathogen before transfer to the sea. Spironucleus barkhanus and Spironucleus salmonis were also identified in the sampled fish. The present study also demonstrated that infections with multiple Spironucleus species are present in wild salmonids. No indications of disease related to diplomonad infections were observed in the wild fish, suggesting that wild salmonids are reservoir hosts of Spironucleus salmonicida.  相似文献

Eggs of Ascaris lumbricoides, A. suum, Toxocara canis, and T. mystax were examined by scanning electron microscopy (SEM). All species under study exhibited pronounced surface ridges. The ridges formed distinctive patterns in T. canis and T. mystax. In the Ascaris species, the ridges are similar except that they are more pronounced in the eggs of A. suum. Operculumlike structures were observed only in Ascaris. Correlation of data from SEM with previously reported transmission electron microscopy suggests that the surface ridges seen in Ascaris eggs are formed by the chitinous layer of the shell.  相似文献

A thermophilic facultative bacterial isolate was recovered from 3.2km depth in a gold mine in South Africa. This isolate, designated GE-7, was cultivated from pH 8.0, 50 degrees C water from a dripping fracture near the top of an exploration tunnel. GE-7 grows optimally at 65 degrees C and pH 6.5 on a wide range of carbon substrates including cellobiose, hydrocarbons and lactate. In addition to O(2), GE-7 also utilizes nitrate as an electron acceptor. GE-7 is a long rod-shaped bacterium (4-6microm longx0.5microm wide) with terminal endospores and flagella. Phylogenetic analysis of GE-7 16S rDNA sequence revealed high sequence similarity with G. thermoleovorans DSM 5366(T) (99.6%), however, certain phenotypic characteristics of GE-7 were distinct from this and other previously described strains of G. thermoleovorans.  相似文献

Rapid freezing and substitution with fixative prior to scanning electron microscopy was used to demonstrate the pattern of beat and recovery of the cilia of free swimming miracidia of Fasciola hepatica. There were stages of dexioplectic metachronal co-ordination and the power stroke was approximately 15 degrees anticlockwise from the anterior-posterior axis. Around the circumference of the body of the miracidia there were approximately 12 metachronal waves of power and recovery. Free-swimming cercariae were recorded by time-lapse photography and, after conventional fixation, by scanning electron microscopy. Cercarial tail-beats were to the posterior of the body in the lateral plane at a rate of 8 Hz. The tail has paired lateral ridges positioned to act as leading edges. There is an array of 32 sensory papillae on the mid-ventral surface of the tail. The tegument of the most distal part of the tail is described: it is free of sensory endings and the surface shows a spiral pattern.  相似文献

The seed morphology of 15 species ofSagina from North America, Europe, and eastern Asia was studied, utilizing the scanning electron microscope, to determine the significance of seed coat features as taxonomic characters. There is variation in particular surface features within some members of the genus. However, superficial ridges and tubercules exhibit patterns very helpful in determining relationships among some species. Two basic seed types occur in the genus and are diagnostic at the sectional level. The saginoid seed, characteristic of sectionSagina, is obliquely triangular in outline, is grooved along the two dorsal ridges, and has slightly concave lateral surfaces. The crassuloid seed, characteristic of sectionMaxima, is obliquely reniform in outline, lacks dorsal grooves, and has shallowly convex lateral surfaces.  相似文献

The cuticle micromorphology of the leaves of the monospecific genus Saxegothaea (Podocarpaceae) was studied by scanning electron microscopy. The external and internal features of the adaxial and abaxial surfaces were characterized. The leaves are hypostomatic. The external adaxial cuticle is rugose with irregular ridges and shallow trench‐like structures that do not correspond to any feature of the inner cuticle surface. The external abaxial cuticle has densely crowded stomata arranged in two bands. The stomata are sunken with pronounced, interrupted Florin rings. Stomatal plugs were not observed. Internally, the adaxial epidermal cells are usually rectangular to square; the abaxial epidermal cells are mainly restricted to the midrib and margins and narrowly rectangular; any among the stomata are irregularly shaped. The stomata are nearly all in direct contact. They show unusual features, including an extra pair of cuticular flanges between the guard cell flanges and those of the lateral subsidiary cells, and ‘bridges’ of lateral subsidiary cell tissue extending polewards above the polar extensions to unite with those at their tips. Neither of these features has been reported previously in Podocarpaceae. The results are discussed in the light of recent phylogenetic studies. Seven chemotherapeutic agents (dimetridazole, metronidazole, pyrimethamine, albendazole, fenbendazole, mebendazole and magnesium sulfate) were examined for growth inhibition on the cultivation of Spironucleus vortens. Dimetridazole and metronidazole were effective in inhibiting the parasite's growth. At concentrations of 1 microgram ml-1 or higher, both dramatically decreased numbers of parasites. At 24 h exposure, 33% of parasites were inhibited when exposed to dimetridazole or metronidazole at concentrations of 2 and 4 micrograms ml-1, respectively. Dimetridazole at 4 micrograms ml-1 or higher concentrations decreased the number of organisms to 50% or less after 48 h exposure. During the same period of time, the numbers of parasites decreased to 50% or less when exposed to metronidazole at 6 micrograms ml-1 or higher. Pyrimethamine at concentrations of 1 to 10 micrograms ml-1 was not effective in inhibiting the parasite's growth. Albendazole and fenbendazole at concentrations of 0.1 and 0.5 microgram ml-1 were similar in inhibiting the growth of the organism. Both compounds suppressed parasite growth at concentrations of 1.0 microgram ml-1 or higher after 24 h exposure. Mebendazole inhibited the parasite's growth at concentrations of 0.5 microgram ml-1 or higher. At 72 h exposure, 45 to 50% of the parasites were inhibited when exposed to mebendazole at concentrations higher than 0.5 microgram ml-1. Magnesium sulfate at concentrations of 70 mg ml-1 or higher also suppressed the growth of parasites after 24 h exposure. These results indicate that dimetridazole, metronidazole and mebendazole are the most effective chemotherapeutic agents in vitro at inhibiting the growth of S. vortens.  相似文献

The legless locomotion of snakes requires specific adaptations of their ventral scales to maintain friction force in different directions. The skin microornamentation of the snake Corallus hortulanus was studied by means of scanning electron microscopy and the friction properties of the skin were tested on substrates of different roughness. Skin samples from various parts of the body (dorsal, lateral, ventral) were compared. Dorsal and lateral scales showed similar, net-like microornamentation and similar friction coefficients. Average friction coefficients for dorsal and lateral scales on the epoxy resin surfaces were 0.331 and 0.323, respectively. In contrast, ventral scales possess ridges running parallel to the longitudinal body axis. They demonstrated a significantly lower friction coefficient compared to both dorsal and lateral scales (0.191 on average). In addition, ventral scales showed frictional anisotropy comparing longitudinal and perpendicular direction of the ridges. This study clearly demonstrates that different skin microstructure is responsible for different frictional properties in different body regions.  相似文献

Lagenophrys dennisi, a peritrich ciliate, was observed attached to the exoskeleton of the crayfish Cambarellus patzcuarensis in Lake Pátzcuaro, Michoacán, Mexico. Lagenophrys dennisi presents a hemispheroidal, suboval or oval lorica in dorsal view, the distinctive lorica aperture consists of a pair of lips highly arched, unthickened, and smooth. Comparison of morphometric characters of the ciliate with previous records is made. Structures such as a "V"-shaped lorica suture, collar ridges, and myoneme are proposed for species identification. An anterior crescentic thickening on the dorsal surface of the lorica was observed under the scanning electron microscope. Lagenophryids were associated with 11 of 13 body parts with antennules and rostrum showing the highest prevalence. Lagenophrys dennisi was also found attached to submerged glass slides. This study represents the first record of L. dennisi on C. patzcuarensis and the first record of its presence in Mexico.  相似文献
