Direct radiochemical determinations of juvenile hormone (JH) biosynthesis by corpora allata (CA) isolated from starved and re-fed Periplanteta americana have been employed to elucidate the humoral mechanisms involved in the modulation of reproductive activity in response to food availability. When starvation was initiated in mature adult females at the time of formation of an oötheca the next oötheca was normally deposited 5 to 6 days later, a delay of 2–3 days, and a third oötheca was formed by only 50% of starved females. The terminal oöcytes in the remaining females were either resorbed or maintained in an arrested state. Ovarian development had effectively ceased after 2 weeks of starvation but recommenced within 3 days of re-feeding. The CA of most starved females exhibited 2 activity cycles following food withdrawal. The first peak occurred on day 1 of the starvation period and was coincident with the timing for fed controls. The second peak was delayed by about 2 days and the activity of the CA then declined to the extent that glands from animals starved for more than 11 days were completely inactive. Feeding, after 10 or 16 days starvation, resulted in a resumption of CA activity which was detectable in some animals within 24 hr, and very high rates of JH biosynthesis were found 4 or 5 days later. The results suggest that P. americana can readily and efficiently modulate egg production in response to food supply, and that control is effected through alterations in JH production by the CA. The use of farnesenic acid as a biochemical probe indicates that CA inactivity after long periods of starvation does not arise because malnutrition has caused complete metabolic shut-down in the glands, and that JH biosynthesis is basically modulated at a control point prior to the last two enzymic stages in the pathway.  相似文献

Studies were undertaken to determine whether adult males of Heliothis virescens transfer juvenile hormone (JH) to females during copulation, and an in vitro radiochemical assay was used to determine whether mating causes an allatotropic effect, i.e., stimulation of JH biosynthesis by corpora allata (CA). In vitro, CA from 3-day-old mated females synthesized and released approximately 2.5 times total JH as that of CA from comparably aged virgin females. Of the homologues, JH II exhibited significant increase in mated females; JH I also increased but not significantly. JH III remained similar to that of virgin females. This is the first demonstration of an allatotropic effect of mating in moths. In contrast to the female, CA of virgin males did not produce any JH, but accessory sex glands (ASG) in 3-day-old males synthesized small amounts of JH. Immediately after adult emergence, male ASG contained approximately 1.5 ng JH I and II, which increased by 12 h after emergence and remained at this high level up to 54 h after emergence. JH III was barely detected in ASG. JH in ASG of mated male immediately after uncoupling was depleted almost completely, and 24 h later recovered to levels comparable to that of 54-h-old virgin male. Virgin female bursa copulatrix did not contain any JH, but mated female bursa, immediately after uncoupling, had JH at levels comparable to that observed in virgin male ASG. By 6 h after uncoupling, JH levels decreased dramatically in mated female bursa. These data suggest the transfer of JH to females by the male. Arch. Insect Biochem. Unilateral section of the nervi corporis allati I (NCA-1) of isolated, starved, adult, virgin Periplaneta americana disinhibited oocyte growth during a specific period following their adult emergence. The effect required that the corpus allatum (CA) be free of NCA-1 innervation for 4 days beyond the time the females were 7–8 days old. The onset of this sensitive period corresponds to when most isolated, starved virgins become sexually receptive. The results suggest that NCA-1 inhibition of CA activity, initiated about 7 days, is relieved by mating. When done on sexually receptive, starved virgins, unilateral NCA-1 section was as effective as insemination for stimulating growth and chorionation of the first generation of oocytes. Neural inhibition of juvenile hormone (JH) secretion by the CA may also explain diminished production of oocytes by isolated, fed virgins, for during 30 days following unilateral NCA-1 section they produced 2.6 to 5 times more oothecae than did controls with a single CA removed or after the sham operation. The number of oothecae deposited by fed virgins was similarly increased after bilateral NCA-1 section, but to a lesser extent than when the operation was done on fed, inseminated females of the same age. Specificity of the response of the CA to denervation was substantiated by experiments in which the CA were extirpated and reimplanted, by topically applying C16JH, and by experiments in which the nervus corporis cardiaci 1 and 2 on the right or left side were severed.  相似文献

Juvenile hormones (JHs) are synthesized by the corpora allata (CA) and play a key role in insect development. A decrease of JH titer in the last instar larvae allows pupation and metamorphosis to proceed. As the anti-metamorphic role of JH comes to an end, the CA of the late pupa (or pharate adult) becomes again “competent” to synthesize JH, which would play an essential role orchestrating reproductive maturation. In the present study, we provide evidence that ecdysis triggering hormone (ETH), a key endocrine factor involved in ecdysis control, acts as an allatotropic regulator of JH biosynthesis, controlling the exact timing of CA activation in the pharate adult mosquito. Analysis of the expression of Aedes aegypti ETH receptors (AeaETHRs) revealed that they are present in the CA and the corpora cardiaca (CC), and their expression peaks 4 h before eclosion. In vitro stimulation of the pupal CA glands with ETH resulted in an increase in JH synthesis. Consistent with this finding, silencing AeaETHRs by RNA interference (RNAi) in pupa resulted in reduced JH synthesis by the CA of one day-old adult females. Stimulation with ETH resulted in increases in the activity of juvenile hormone acid methyltransferase (JHAMT), a key JH biosynthetic enzyme. Furthermore, inhibition of IP3R-operated mobilization of endoplasmic reticulum Ca2+ stores prevented the ETH-dependent increases of JH biosynthesis and JHAMT activity. All together these findings provide compelling evidence that ETH acts as a regulatory peptide that ensures proper developmental timing of JH synthesis in pharate adult mosquitoes.  相似文献

The cessation of juvenile hormone (JH) production is a key endocrine event that halts ovarian development and hence initiates diapause in females of the mosquito, Culex pipiens. The shutdown in endocrine activity of the corpora allata (CA), the source of JH, was manifested in the smaller size of CA in females reared under short daylengths (diapause) compared to those reared under long daylengths (nondiapause), as well as in low expression of the mRNA encoding allatotropin, the neuropeptide that promotes JH biosynthesis in the CA. Genes encoding both allatotropin and allatostatin were identified in C. pipiens, but only expression levels of allatotropin differed in the two types of females. Knockdown of allatotropin mRNA using RNA interference in females programmed for nondiapause resulted in a cessation of ovarian development akin to diapause. This arrest in development could be reversed with an application of JH. Our results thus suggest that suppression of allatotropin is a critical link in regulating the shutdown of the CA during diapause.  相似文献

Juvenile hormone (JH) synthesis by the corpora allata of gregarious and solitarious phase females of Schistocerca gregaria was determined in vitro during the penultimate and last stadia as well as during the first gonotrophic period of adults. Generally, the corpora allata of solitarious females showed higher rates of JH synthetic activity. In addition, in adult females there was a temporal difference between the corpora allata activities of gregarious and solitarious locusts, the latter exhibiting relatively higher rates of JH synthesis early in the first gonotrophic period. The corpus allatum volumes of solitarious females were also generally larger than those of their gregarious counterparts; there was no synchrony between fluctuations in JH synthetic activity and changes in corpus allatum volume in either phase.The early onset of relatively high JH synthetic rates in solitarious females was correlated with the early detection, by rocket immunoelectrophoresis, of vitellogenin in the haemolymph and vitellin in the oöcytes. Vitellogenin appeared in the haemolymph on day 4 in solitarious females and on day 6 in gregarious females and vitellin appeared in the oöcytes on days 6 and 8 respectively. Oöcyte length at which vitellogenesis was first detected was 1.8 mm for gregarious and 1.3 mm for solitarious females. However, despite the accelerated onset of both vitellogenin synthesis and uptake, oöcyte maturation time of solitarious females was longer. In both gregarious and solitarious females, vitellogenin titres increased until oöcytes reached a length of about 4 mm and declined thereafter. Vitellin content of ovaries increased proportionately to oöcyte growth until they attained a length of 5.0 mm. The subsequent increase in length of oöcytes to maturity is attributed to postvitellogenic growth, possibly by hydration.  相似文献

Juvenile hormone analogs are known to inhibit the production of juvenile hormone (JH) by the corpora allata (CA). However, the mechanism of this inhibition remains undefined. We have used two JH mimics, fenoxycarb and pyriproxyfen, to examine the mechanism of suppression in the cockroach, Diploptera punctata. Denervation experiments demonstrated the importance of nervous connections between the brain and CA for the inhibition of JH biosynthesis by fenoxycarb. Fenoxycarb treatment alters the sensitivity of CA to allatostatin treatment in vitro. Suppression of JH biosynthesis by fenoxycarb following denervation of the CA showed that innervation was in part responsible for the inhibition. Similarly, maximal inhibition by Dippu-AST7 requires intact nervous connections between the brain and CA, particularly during rapid vitellogenesis. qPCR analysis of brain, CA, ovary and midgut extracts revealed that both allatostatin and its receptor Dippu-ASTR2 show increased levels of expression following topical fenoxycarb treatment, particularly in brain tissue on days 4 and 5 of the first gonadotrophic cycle and in CA on day 4. The correlation between inhibition of JH biosynthesis and increased expression of AST and ASTR2 in brains and CA, together with increased sensitivity of CA to allatostatin in vitro, suggests that allatostatin may be one of the effectors by which fenoxycarb inhibits JH biosynthesis.  相似文献

A radiochemical assay which fulfills the required validation criteria has been used for quantification of the in vitro biosynthesis of JH III by the corpora allata of adult females of Blattella germanica throughout the 7 days of the first reproductive cycle. The presence of JH III has been independently confirmed by HPLC and mass spectrometry. Results indicate that rates of JH release increase repidly from day 3 to day 6, which is correlated with oocyte growth. The highest levels of JH release (2.58 ± 1.11 pmol/hr per pair) were obtained from day-6 females. The time course of JH production by CA from day-6 females showed that CA released JH at a linear rate for at least 9 hr. From these results, it can be concluded that titers at high production ages and linearity ranges are satisfactory enough to be used in studies on the regulation of JH production in this species.  相似文献

The synthesis of juvenile hormone III (JH III) by the isolated corpora allata (CA) of Aedes aegypti adult female was studied using an in vitro radiochemical assay. We dissected the corpora allata-corpora cardiaca (CA-CC) complex attached to a piece of aorta. The complex was left connected to the intact head capsule to facilitate the visualization and transfer of the glands. A linear increase in the cumulative amount of biosynthesized JH III was found for at least the first 6 h of incubation; approximately 45% of the synthesized JH III was present in the medium. There was a dependence of JH III synthesis on exogenous methionine supply. Using reversed phase high performance liquid chromatography two major labeled products biosynthesized by the CA were separated. They co-migrated with JH III and methyl farnesoate (MF). The identity of the biosynthesized JH III was confirmed by gas chromatography-mass spectrometry. JH III synthesis was only 2.0 fmol/pair gland/h immediately after adult emergence, but increased to 32.6 fmol/ pair gland/h 18 h later in sugar-fed females. Two days after emergence, the CA biosynthetic activity slowly started to decrease, and reached values of around 5.3 fmol/pair gland/h by one week after emergence. Synthesis of JH was similar from either sugar-fed females mated or unmated. A blood meal resulted in a decrease of JH III synthesis in CA from mated females by 12 h after feeding and from virgin females by 24 h after feeding. JH III biosynthesis remained low for at least 96 h in mated females, but was back to higher levels 72 h after feeding in virgin females. Rates of JH III biosynthesis closely reflected the hemolymph levels of JH III both after emergence and after a blood meal described by Shapiro et al. (1986). The activity of the CA in Aedes aegypti females seems to be regulated by developmental changes and nutritional signals, and to be independent of mating stimulus.  相似文献

Abstract.  The reproductive physiology of social insects is often highly responsive to social stimuli from nestmates, but the mechanisms underlying this sensitivity are usually poorly understood. The effect of varied social conditions on the endocrinology and ovarian maturation in maturing female primary reproductives of the dampwood termite Zootermopsis angusticollis Hagen is studied to better understand their developmental responses to conspecifics. Newly dealate queens are paired with a reproductive male, with another maturing female, or are allowed to mature in isolation. Developmental responses are tracked by monitoring rates of juvenile hormone (JH) production by corpora allata in vitro , haemolymph ecdysteroids titres and ovarian development after 5, 10, 15 and 30 days of maturation. Significant declines in rates of JH production rates are observed by days 5, 10 and 30, respectively, for females paired with males, left to mature in isolation or paired with a female. Ecdysteroid titres increase by day 5 and stay elevated in females nesting with males. By contrast, for CA from females that are isolated or paired with another female, the titres decline and stay low until at least day 15, but increase significantly by day 30. These hormonal differences correspond to differential rates of physiological maturation, with more rapid ovarian development occurring in females paired with a male, than in those nesting alone or with another female. The results suggest that JH and ecdysteroids modulate the ovarian response of alates to stimuli from nestmates during this period of maturation, and that JH and ecdysteroid titres may be regulated independently during this period of development.  相似文献

The genome of Tribolium castaneum encodes two allatostatin [AS type B; W(X)6Wamide and AS type C; PISCF‐OH] and one allatotropin (AT) precursor, but no AS type A (FGLamide) (Tribolium Genome Sequencing Consortium, 2008: Nature 452:949–955). Here we studied the activity (in vitro) of peptides derived from these precursors on the synthesis/release of juvenile hormone (JH) III. The corpora cardiaca‐corpora allata (CC‐CA) complexes of adult females of another tenebrionid beetle, the mealworm Tenebrio molitor, were used. Incubating the gland complexes in a medium containing Trica‐AS B3 peptide, we showed that the peptide has allatostatic function in T. molitor. The activity of the type C AS depended on the age of the test animals and their intrinsic rate of JH III biosynthesis. The Trica‐AS C peptide inhibited the JH release from CA of 3‐day‐old females with a high intrinsic rate of JH synthesis, but activated JH release from the CA of 7‐day‐old females with a lower intrinsic rate of JH production. The allatotropin peptide (Trica‐AT) also activated the JH release from the CA of 7‐day‐old females in a dose‐dependent and reversible manner. Unexpectedly, a type A AS derived from the precursor of the American cockroach Periplaneta americana (Peram‐AS A2b) inhibited the JH release from the CA of younger and older females in the concentration range of 10?8 to 10?4 M, and the effects were fully reversible in the absence of peptide. These data suggest a complex role of allatoactive neuropeptides in the regulation of JH III biosynthesis in beetles. © 2010 Wiley Periodicals, Inc.  相似文献

We have observed changes with time in the fine structure of corpora allata (CA) during a known cycle of increasing and decreasing juvenile hormone (JH) synthesis in late embryos of Diploptera punctata. A previous report showed that rates of JH release were relatively low in 28-day-old embryos, but CA activity subsequently rose linearly to a peak on about day 42, and thereafter steadily declined to a low level on day 64 just before birth (Holbrook et al., 1997). We now show that, regardless of rate of JH synthesis, CA cells are large and replete with organelles which nevertheless exhibit variable morphology in embryos of different age. Highly active CA cells on day 40 contain abundant ring-form mitochondria, whereas CA cells of low activity on days 28 and 64 contain mitochondria that are rod-shaped or globular. Mitochondrial cristae were scarce and indistinct on day 28 but numerous and well developed on day 64. Endoplasmic reticula (ER) are rare on day 28 and appear in increasing numbers when CA activity rises. On day 40, ER are abundant and often exhibit a whorl-like appearance which is not observed on day 28. After day 44, when biosynthetic activity is declining, whorls of ER gradually decrease in number and are ultimately replaced by vesicular smooth ER on day 64. Neurosecretions are found only after day 38, by which time rates of JH synthesis have increased substantially from those of day 28. Except for membranous autophagic vacuoles, which are frequently found when ER whorls disintegrate as rates of JH synthesis decline toward birth, most autophagic vesicles such as multivesicular vesicles and dense bodies occur only sporadically among CA cells at all examined ages. We conclude that synchronous autophagy of exhausted organelles, which results in atrophy of CA cells and long-term arrest of JH synthesis in adult females of D. punctata, does not occur in embryos. The slow cyclic change in rate of JH synthesis in embryonic CA is most likely due to asynchronous autophagic activity and to alterations in certain unique features of intracellular organelles.  相似文献

In a previous study [ McNeil et al. (1996) Archives of Insect Biochemistry and Physiology, 32, 575–584], patterns of sexual maturation and Juvenile Hormone (JH) biosynthesis were compared in virgin females from migratory (North American) and non‐migratory (Azorean) populations of the true armyworm moth, Pseudaletia unipuncta Haworth (Lepidoptera: Noctuidae). Sexual maturation occurred at a significantly earlier age after emergence in the non‐migrant population, and the rates of biosynthesis of JH in vitro suggested that lower titres of JH may be required to initiate the onset of calling behaviour (pheromone emission) and ovarian development in Azorean females. To examine the physiological differences in the reproductive biology of migratory and non‐migratory populations in greater detail, the haemolymph titres of JH and JH esterase activity were compared in virgin females as a function of age. In addition, the effects of mating on JH biosynthesis in vitro, JH titres, JH esterase activity and egg production were measured in the two populations. As expected, JH titres rose more rapidly after emergence in Azorean females than in their North American counterparts but, contrary to our prediction, the maximum levels were also higher in the non‐migrant population. Activity of JH esterase was much higher in Azorean females on the day of emergence. However, by the second day both populations had similar activity levels (about 17 nmol JH/min/ml) and exhibited a similar age‐related decline in subsequent days. Mating did not affect the rate of JH biosynthesis in vitro but resulted in a significant increase in the titres of JH in the haemolymph of both populations. The maximum titre (a five‐fold increase) occurred within 24 h of mating in Azorean females. In North American individuals the increase was greater (seven‐fold) but did not occur until 48 h after mating. No difference in the activity of JH esterase was observed between mated and virgin North American females. By contrast, while there was an age‐related decline in the activity of JH esterase in mated Azorean females, as seen in both North American groups, activity levels in virgin females remained constant with age. In all females, mating resulted in a significant increase in egg production within 24 h. The Azores is a volcanic archipelago, so these non‐migratory populations were probably founded by immigrants originating from migratory continental populations. It is clear from our results that the change from a life history that includes migration to a non‐migratory one involved more than just a temporal shift in the timing of the production of JH. Furthermore, the interpopulation differences in titres of JH and mating‐induced changes reported here cannot be fully explained by the observed differences in the patterns of activity of JH esterase and JH biosynthesis in vitro.  相似文献

The juvenile hormones (JHs) play a central role in insect reproduction, development and behavior. Interrupting JH biosynthesis has long been considered a promising strategy for the development of target-specific insecticides. Using a combination of RNAi, in vivo and in vitro studies we characterized the last unknown biosynthetic enzyme of the JH pathway, a fatty aldehyde dehydrogenase (AaALDH3) that oxidizes farnesal into farnesoic acid (FA) in the corpora allata (CA) of mosquitoes. The AaALDH3 is structurally and functionally a NAD+-dependent class 3 ALDH showing tissue- and developmental-stage-specific splice variants. Members of the ALDH3 family play critical roles in the development of cancer and Sjögren–Larsson syndrome in humans, but have not been studies in groups other than mammals. Using a newly developed assay utilizing fluorescent tags, we demonstrated that AaALDH3 activity, as well as the concentrations of farnesol, farnesal and FA were different in CA of sugar and blood-fed females. In CA of blood-fed females the low catalytic activity of AaALDH3 limited the flux of precursors and caused a remarkable increase in the pool of farnesal with a decrease in FA and JH synthesis. The accumulation of the potentially toxic farnesal stimulated the activity of a reductase that converted farnesal back into farnesol, resulting in farnesol leaking out of the CA. Our studies indicated AaALDH3 plays a key role in the regulation of JH synthesis in blood-fed females and mosquitoes seem to have developed a “trade-off” system to balance the key role of farnesal as a JH precursor with its potential toxicity.  相似文献

In a Japanese population of Locusta migratoria, adult females become reproductively inactive under crowding and long days (LD) and reproductively active under crowding and short days (SD). The identity and titre of ecdysteroids in the haemolymph and ovaries from adult females reared under SD and LD were investigated by RIA/HPLC. The effects of exogenous juvenile hormone (JH) III treatments on the termination of such reproductive arrest and ecdysteroid contents in LD females were also examined. In general, ecdysteroid titres in both haemolymph and ovaries were significantly higher in reproductively active SD females than in reproductively inactive LD females. A clear difference was also observed in oocyte growth between SD and LD individuals. JH III applications (three consecutive topical applications, 150 μg per insect per day from day 3) stimulated ovarian development in LD females and significantly increased the haemolymph and ovarian ecdysteroids to a level comparable to that of reproductively active SD adult females.  相似文献

Juvenile hormone (JH) esterase was characterized from the plasma of adult females of the cabbage looper, Trichoplusia ni, and compared with that present in 4th and 5th instar larvae. Ester hydrolysis was the principal route of JH metabolism. Gel filtration of plasma resolved a single peak of JH esterase which was distinct from that of the α-naphthyl acetate (α-NA) esterase activity. The JH esterase apparent molecular weight was 62,000 in prepupae and virgin, female adults and 69,000 in 2-day-old 4th instar larvae. Broad range isoelectric focusing of plasma of prepupae and adults resolved a major peak of activity at pH 5.5 with a minor peak of activity at pH 6.1 and in 4th instar larvae at pH 5.45 and 5.8, respectively. By this method JH esterase was resolved from the α-NA esterase activity. The plasma of prepupae and adults metabolized JH I at about twice the rate of JH III. JH esterase activity from adult plasma was more stable than the α-NA esterase activity. Adult JH esterase activity was insensitive to inhibition by O,O-diisopropyl phosphorofluoridate in contrast to that of the α-NA esterase activity. Mated females oviposited 8 times more eggs than virgin females to 10 days after emergence. The total haemolymph protein content of virgin females remained high throughout the period of study whereas mated females showed a significant decline beginning on day 4. JH esterase activity remained unchanged in virgins whereas it declined drastically in mated females. The α-NA esterase activity declined to low levels shortly after emergence in both groups. JH and α-NA esterase activity was not affected by the application of the juvenoid, (RS)-methoprene. The present study provides evidence of a functional role for JH esterase in JH metabolism and reproduction in adult T. ni. JH esterases in the adult were identical to that of prepupae by the methods described above.  相似文献

In the Savio strain of Locusta migratoria an imaginai diapause is induced by long daylength. In diapausing females, the haemolymph level of juvenile hormone (JH) was undetectable during the first 3-wk of imaginai life and later rose only slightly to about 20 ng/JH3IR per ml. Only peripheral cells of the corpora aliata (CA) were active. In nondiapausing animals, or after the termination of diapause, the JH level was high (140–200 ng/ml) and the ultrastructure of the gland exhibited signs of activity. CA severance in 3-wk-old diapausing females terminated diapause as a result of activation of the CA. CA disconnection in the fifth larval instar or at the imaginai moult in long daylength animals did not break diapause and the CA stayed inactive. The lateral cells of the protocerebrum exert a jdual effect: at the end of larval life they bring about CA maturation and render them active, whereas during the imaginai diapause they inhibit CA activity. The median neurosecretory cells of the pars intercerebralis support CA activity during vitellogenesis.  相似文献

Abstract. The regulation of Juvenile Hormone (JH) HI biosynthesis and release by the corpora allata (CA) was studied in final instar male and female larvae of the earwig, Euborellia annulipes , using a radiochemical assay in vitro. In males, maximal biosyntiiesis of JH IH occurred on day 1, then declined to virtually undetectable levels for the following 12 days of the stadium, and finally increased on days 14–16. In females, peaks of biosynthesis were detected on days 0–1 and on day 12. A further investigation of the 12-day-old larvae demonstrated mat in nonmoulting males and females, JH UJ biosynthesis was undetectable. However, for males and females undergoing ecdysis, the biosynthesis of JH III was detected and quantified.
The addition of 60 μM farnesoic acid to the incubation medium significantly increased the production of JH III by CA taken from females from day 8 until the end of the stadium. Glands from 12-day old females that had initiated ecdysis were stimulated by farnesoic acid. By contrast, we could detect no stimulation of production of JH III by farnesoic acid in CA taken from males, even very late in the stadium. CA from newly emerged adult males and females were more active than those of larvae, and were greatly stimulated by farnesoic acid. CA from females immediately after emergence were stimulated significantly more by farnesoic acid man were glands from newly emerged males. We investigated the effect of fifteen 1,5-disubstituted imidazoles (1,5-dis) on juvenile hormone III (JH III) and methyl farnesoate (MF) biosynthesis by the corpora allata (CA) of the mosquito Aedes aegypti in vitro. Four compounds (TH-35, TH-83, TH-62 and TH-28) significantly decreased JH biosynthesis in the CA dissected from 3-day old sugar-fed females. The decrease of JH synthesis was not always associated with increased MF. TH-30 and TH-83 increased MF levels, while TH-85 and TH-61 significantly decreased MF levels. Five compounds (TH-26, TH-60, TH-83, TH-35 and TH-30) significantly inhibited JH biosynthesis in the CA dissected from females 15 h after a blood meal. Four 1,5-dis (TH-30, TH-26, TH-28 and TH-66) caused MF increases in CA from blood-fed females. 1,5-Disubstituted imidazoles had higher inhibitory activity on JH synthesis when substituted at position 5 by a 3-benzyloxyphenyl group and at position 1 by a benzyl group (such as TH-35). Inhibition of JH and MF biosynthesis by TH-35 was age-dependent and influenced by nutritional status; inhibition differed when evaluated in the CA dissected from sugar-fed females at different days after emergence and in the CA dissected from females at different hours after a blood meal. Inhibition was always higher when the CA was more active. The addition of TH-35 significantly reduced the stimulatory effect of Aedes-allatotropin and farnesoic acid on JH synthesis. This is the first report of an inhibitory effect of 1,5-disubstituted imidazoles on JH synthesis in Diptera.  相似文献
