The baculovirus expression vector, Trichoplusia ni nucleopolyhedrovirus, with the advantage of polyhedral inclusion body formation in recombinant viruses, was used to express the ecdysteroid receptor of the Australian sheep blowfly Lucilia cuprina(LcEcR). pSXIVVI +X3/2 baculovirus transfer vector was chosen for a 2.8kb LcEcR cDNA subcloning since pSXIVVI +X3/2 contains an efficient translational initiation signal (ATG) and it allows the LcEcR cDNA fusion to N-terminal codons in the correct reading frame. The resulting transfer plasmid pSXIVVI +X3-LcEcR was cotransfected into BT1-Tn-5B1-4 cells with the parental virus TnNPV-SVI -G minus polyhedral inclusion body, which expresses β-galactosidase gene. After 3~4 runs of plaque purification, three TnNPV-LcEcR clones were obtained with the LcEcR gene under the dual control of synthetic and XIV promoters. These three TnNPV-LcEcR clones all showed white phenotype when stained with X-gal. Western blot analysis showed 2~3 specific polypeptides with molecular weight ranging from 70~90kD. Three TnNPV-LcEcR clones expressed different level of LcEcR polypeptides in BT1-Tn-5B1-4 cells. The TnNPV-LcEcR-1 clone expressed the highest level of LcEcR polypeptides in BT1-Tn-5B1-4 cells 48~72h post infection.  相似文献

Abstract. The literature relating to the attraction of the sheep blowflies Lucilia sericata and Lucilia cuprina to their ovine hosts is reviewed. The responses of the two species are similar and different components of the behaviour leading to host location and oviposition appear to involve at least two distinct sets of semiochemical cues. Activation, upwind orientation and landing appear to occur in response to putrefactive sulphur-rich volatiles, originating from bacterial decomposition products. Oviposition is elicited primarily by the presence of ammonia-rich compounds; moisture, pheromones and tactile stimuli may also act as oviposition stimuli. There is a pronounced sex difference in the response of Lucilia to semiochemicals with a higher proportion of females attracted than males and a higher proportion of gravid than non-gravid females. While the mechanisms of host location by Lucilia are of intrinsic interest, understanding the responses to semiochemicals is important in the attempt to develop powerful synthetic baits for deployment with the traps or targets used for population sampling or suppression. The literature is discussed with respect to the development of synthetic semiochemical baits.  相似文献

ABSTRACT. Eclosion in Lucilia cuprina (Wiedemann) occurs near dawn. The rhythm of eclosion persists in both darkness and constant light of high intensity (490μW cm-2) with a period close to 24h. The sensitivity to light of the circadian clock controlling eclosion varies greatly according to the stage of the life cycle. During larval life the free running rhythm in darkness can be phase shifted by light pulses of 100μW cm-2 intensity, with the transition from a Type 1 phase response curve to a Type 0, occurring with pulses of between 1 and 8h. Extending the last light period of LD to 24 h followed by constant darkness resets the phase of the rhythm by 12h, a transition from constant light to constant darkness initiates rhythmicity in flies made arrhythmic by being reared from eggs collected from adults maintained in constant light. After pupariation, the rhythm is relatively insensitive to light. Rhythmicity is sometimes induced by a transition from constant light to constant darkness, but the phase of the rhythm is not shifted by extending the last light period of LD before entering constant darkness. Repeated LD cycles applied after pupariation initiate and entrain the rhythm.  相似文献

Field and laboratory populations of the Australian sheep blowfly, Lucilia cuprina (Wiedemann) (Calliphoridae), were surveyed by bioassay for possible resistance to the synthetic pyrethroids, a group of insecticides under development for blowfly control. A normal distribution of LC50 values was found using deltamethrin, the test pyrethroid, with no indication of specific resistance despite widespread use of deltamethrin on sheep to control the sheep body louse, Damalinia ovis (Schrank) (Trichodectidae). There was no cross-resistance to deltamethrin from existing organophosphate (OP) resistance nor from previous use of DDT. Selection with deltamethrin on a combined field strain, CSF85, increased the LC50 gradually over the first twenty generations until it stabilized at approximately 25x that of the unselected CSF85. This laboratory-induced resistance extended to other pyrethroids, cypermethrin (16x), cyhalothrin (25x) and cycloprothrin (10x), and increased the existing resistance of CFS85 to the OP diazinon (11x) and the carbamate, butacarb (83x).  相似文献

Coloured, odour-baited, sticky targets, designed to catch the sheep blowfly Lucilia sericata Meigen are described and the results of field trials are presented. The number of L. sericata caught by the targets was positively related to target surface area and was dependent on target colour. The performance of the coloured targets is described by a model in which catch is related negatively to reflectivity in the 300-450 nm (ultra-violet/blue) band of the spectrum and positively to the reflectivity in the 450-580 nm (blue-green-yellow) band. Of the Lucilia blowflies caught by the targets, 13-16% were L.sericata and 72-75% of the L.sericata were female. The number of L.sericata caught appeared to be positively related to the availability of sheep over a relatively localized area and showed a marked decline following insecticidal dipping of sheep. The further development of targets and the implications of the results for L.sericata control are discussed.  相似文献

Abstract  Several genes on chromosomes IV and VI have a significant influence on high-level resistance to diflubenzuron in a strain of the Australian sheep blowfly from Tara, Queensland. Low-level resistance to cyromazine in the same strain is due to genes on these chromosomes with a gene (gene complex) in the sv marker region of chromosome IV being particularly important. For both insecticides, genetic background influences resistance status. If the results of the Tara strain prove typical for those of other populations, resistance to diflubenzuron in the Australian sheep blowfly has potentially significant consequences for woolgrowers.  相似文献

Abstract. The quantity of specific antibody ingested by larvae of Lucilia cuprina and its fate after ingestion were studied in larvae grown on sheep and on an artificial diet. Larvae grown to late first or early second instar on sheep vaccinated with horse myoglobin contained 66% less specific antibody detected by enzyme linked immunosorbent assay than larvae grown to a similar stage on an artificial diet containing 75% serum from the same sheep. A similar result was obtained when larvae were grown to mid-third instar. Larvae grown on sheep to first or second instar contained approximately the same quantity of specific antibody per unit weight of larvae as those grown to third instar. Larvae grown on diet to third instar contained 22% less specific antibody per unit weight than those grown to first or second instar. In larvae grown on diet to late third instar, ingested diet retained 91 ± 12% of its original specific antibody activity in the crop, 50 ± 11% in the anterior midgut, 8 ± 2% in the posterior midgut and 13 ± 6% in the hindgut. The mean concentration of total immunoglobulin detectable in the haemolymph of individual third instar larvae grown on diet was 1.7 ± 2.8 ug/ml. Assays of specific antibody in the haemolymph of similarly reared larvae indicated that all or most of this immunoglobulin remained functional. The implications of the quantities and distribution of ingested functional antibody found in feeding larvae of L.cuprina are discussed in relation to the possibility of vaccinating sheep against these larvae and the selection of likely internal targets as sources of potential protective antigens.  相似文献

ABSTRACT. Under laboratory light: dark cycles, the flight activity of adult Lucilia cuprina (Wied.) was low during darkness and uniformity high during light. This pattern persisted as a rhythm both in constant darkness and in constant light of intensity up to 1lx, with a period of approximately 22 h in each. Light pulses of 15 min at l00lx applied to the free-running rhythm in constant darkness generated phase shifts of up to 60°, 12-h light pulses of the same intensity generated maximal (180°) phase shifts. The phase response curves had shapes similar to those of a number of other insect rhythms. When exposed to light periods (70 lx) of greater than 12 h followed by constant darkness, the rhythm reinitiated at the light-dark transition from a constant phase equivalent to that at the time of the light-dark transition in the LD 12:12 cycle.  相似文献

Abstract. In order to develop and evaluate control strategies for blowfly strike, a greater understanding of the epidemiology is essential. A postal survey of sheep farmers yielded information about ten farm and management factors and their relationship to blowfly strike prevalence. The risk of a farm reporting at least one case of blowfly strike increased as flock size and stocking density increased (adjusted odds ratio of 1.13 for an increase in flock size of 100 sheep and 1.38 for an increase in stocking density of ten sheep per hectare). As farm altitude increased, the risk of blowfly strike decreased (adjusted odds ratio 0.67 for an increase in farm altitude of 100 m). The risk of high strike prevalence (more than 2% of sheep struck) decreased as both farm altitude and flock size increased. High strike prevalence was also associated with on-farm sheep carcase disposal (odds ratio 1.35). Farmers in the south-west of England were more likely to report at least one case of blowfly strike and high strike prevalence compared to all other regions.  相似文献

Abstract A deterministic simulation model is used to explore the potential for control of the sheep blowfly, Lucilia sericata , using odour-baited targets. The simulations show that the ambient temperatures of 15oC or 20oC, 13% and 24% of the females present in a population would need to be killed per day, respectively, to prevent the population growing from its initial spring density during the summer blowfly season. In contrast, if both sexes could be sterilized, so that sterilized males were also able to disrupt the fertility of unsterilized females, only 6.5–13% of both sexes would need to be attracted and sterilized at 15oC or 20oC respectively. To examine whether these numbers of individuals could be attracted by targets baited with liver and sodium sulphide, mark-release-recapture studies in sheep pastures were used to quantify sticky target efficiencies. The percentage of the Lsericata population caught on any one day was shown to be positively related to average ambient temperature. At 15oC each sticky target caught approximately 13% and at 20oC, 41% of the females within a 20 m radius. The results indicate that, at ambient temperatures between 15 and 20oC, if distributed at a density of approximately 5/ha, targets baited with liver and sodium sulphide could effectively suppress a population of L.sericata. Lower densities of targets would be required if they were able to sterilize both sexes rather than kill or if more attractive synthetic semiochemical baits could be developed.  相似文献

Larval cultures of the flesh-fly Sarcophaga argyrostoma maintained in circadian ‘resonance’ experiments produced a high incidence of pupal diapause when the period of the light cycle was close to (T) 24, 48 or 72 hr, but a low incidence of diapause at T 36, 60 or 84 hr. Cultures pre-programmed for diapause by exposing pregnant females to long nights indicated the induction of non-diapause development at T 36, 60 and 84, whereas cultures pre-programmed for diapause-free development by exposing females to continuous light indicated the induction of diapause at T 24, 48 and 72.Raising the temperature reduced the heights of the diapause peaks whereas lowering the temperature raised them. With progeny from long-night-reared flies the lowest temperature tested (18°C) produced a result indistinguishable from an ‘hour-glass’ response, warning that ‘negative’ resonance experiments may merely indicate non-permissive conditions for demonstrating the involvement of circadian rhythmicity in insect photoperiodism.The results of the ‘resonance’ experiments and the effects of temperature are interpreted in terms of a multioscillator ‘external coincidence-photoperiodic counter’ model for the clock.  相似文献

Chitin synthesis in third-instar Lucilia cuprina larvae cultured at 23 °C was investigated using in vivo and in vitro systems, the latter with whole and with homogenized integuments. Synthesis was at a maximum between 24 and 48h after ecdysis from the second instar. Chitin was deposited in layers, and labeled GlcNAc was rapidly cleared from the hemolymph. In in vitro homogenate systems, the rapid conversion of UDP-([14C]GlcN)Ac to ([14C]GlcN)Ac and its 1-phosphate derivative contributed to the low incorporation of this precursor into chitin. The extent of the conversion was reduced by the addition of KCN or phenylthiourea. In in vivo and in vitro tissue systems the level of incorporation of ([14C]ClcN)Ac was higher than that of UDP-([14C]GlcN)Ac. However, in in vitro homogenate systems there was no difference unless UTP was added when the level of incorporation of only ([14C]GlcN)Ac was increased (by a factor of 9). Incorporation of UDP-([14C]GlcN)Ac, but not that of ([14C]GlcN)Ac, was decreased when larvae were deprived of food. Soluble oligosaccharides were detected in in vitro homogenate systems. They were formed during chitin synthesis and may represent newly initiated chitin chains. A reappraisal of current ideas on chitin synthesis in insects is needed.  相似文献

A model is presented for analysis of mark-recapture data of mobile insects which, unlike the Lincoln Index, does not require marked individuals to remain within the sampling area or to mix uniformly with the wild population. The model assumes a single or multiple releases of marked insects from the centre of the sampling area and that captured individuals are not returned to the population. Dispersal rates of marked insects are estimable from serial recaptures and, for catches that are either unaffected by or have been corrected for weather effects, the model also provides estimates of mortality and age-dependent trappability. Application of the model is illustrated using mark-recapture data for adults of the Australian sheep blowfly Lucilia cuprina.  相似文献

Abstract. A treated-surface test was used in a bioassay to detect insecticide resistance to the organophosphorus compounds diazinon and propetamphos, in larvae of field strains of Lucilia cuprina in New Zealand during 1990/91 and 1991/92 (December-April). Mean LC50s for both diazinon and propetamphos showed a significant increase between seasons although this is not necessarily indicative of a trend. Mean LC50s for diazinon increased from 1.16 to 1.60 mg/1 (P< 0.001) and for propetamphos from 0.49 to 0.59 mg/1 (P< 0.005) in pooled populations. Diazinon and propetamphos resistance was significantly correlated in both seasons, suggesting side-resistance between the two insecticides.  相似文献

Understanding the spatial scale and temporal pattern of disease incidence is a fundamental prerequisite for the development of appropriate management and intervention strategies. It is particularly critical, given the need to understand the elevated risks linked to climate change, to allow the most likely changes in the distribution of parasites and disease vectors to be predicted under a range of climate change scenarios. Using statistical models, the spatial distribution and climatic correlates of a range of parasites and diseases have been mapped previously, but their development into dynamic, predictive tools is less common. The aim of the work described here, was to use a species distribution model to characterise the environmental determinants of the monthly occurrence of ovine cutaneous myiasis (blowfly strike) by Lucilia sericata, the most frequent primary agent of northern European myiasis, and to then use this model to describe the potential spatial changes that might be expected in response to predicted climate change in Great Britain. The model predicts that the range of elevated temperatures predicted by current climate change scenarios will result in an increase in the risk of strike and an elongated blowfly season. However, even for the most rapid warming scenario predictions over the next 70 years, strike is not predicted to occur throughout the winter. Nevertheless, in this latter case, parts of central and southern England are likely to become too hot and dry for strike by L. sericata, to persist in mid-summer. Under these conditions, it is possible that other, more pathogenic Mediterranean agents of myiasis, such as Wolfhartia magnifica, could potentially replace L. sericata. Where the phenology of strike is altered by climate change, as predicted here, significant changes to the timing and frequency of parasite treatments and husbandry practices, such as shearing, will be required to manage the problem. The results suggest that the modelling approach adopted here could be usefully applied to a range of disease systems.  相似文献

The effects of light wavelength on photoperiodic clock were determined in the migratory male blackheaded bunting (Emberiza melanocephala). We constructed an action spectrum for photoperiodic induction (body fattening, gain in body mass, and gonadal recrudescence) by exposing birds for 4.5 weeks to 13 h light per day (L:D = 13:11 h) of white (control), blue (450 nm), or red (640 nm) color at irradiances ranging from 0.028 to 1.4 W m?2. The threshold light irradiance for photoinduction was about 10-fold higher for blue, compared to red and white light. Phase-dependent effects of light wavelength on the photoperiodic clock were further examined in the next two sets of skeleton photoperiods (SKPs). In the first set of SKPs, birds were exposed for four weeks to asymmetrical light periods (L:D:L:D = 6:6:1:11 h) at 0.25 ± 0.01 W m?2; two light periods applied were of the same (450 nm: blue:blue, B:B; 640 nm, red:red, R:R) or different (blue:red, B:R or red:blue, R:B) wavelengths, or of white:white (W:W, controls). Photoperiodic induction occurred under R:R and B:R, but not under B:B and R:B light conditions; the W:W condition induced an intermediate response. The second set of SKPs used symmetrical light periods (L:D:L:D = 1:11:1:11 h), and measured effects also on the activity rhythm. Birds were first exposed to one of the four SKPs (R:R, B:B, R:B, or B:R) for three weeks, subsequently were released into dim constant light (LLdim; ?0.01 W m?2, the night light used in an L:D cycle) for two weeks, and then were returned to respective SKPs for another three weeks. Activity was greater in the R:R compared to B:B, and in B:R compared to R:B light condition. Zugunruhe (intense nighttime activity, indicating migratory restlessness in a caged situation) developed under the R:R and B:R, but not the B:B and R:B, light condition. Under LLdim, all birds free-ran with a period >24 h, the Zugunruhe had a circadian period longer than the daytime activity, and the re-entrainment to SKPs was influenced by the position of light periods relative to circadian phase of the activity rhythm. Photoperiodic induction at the end of 8 weeks was found in the R:R and B:R, but not in B:B, light conditions; in the R:B condition only one bird had initiated testes. Taken together, these results suggest that in the blackheaded bunting, the circadian photoperiodic clock is differentially responsive to light wavelengths; this responsiveness is phase-dependent, and the development of Zugunruhe reflects a true circadian function. Wavelength-dependent response of the photoperiodic clock could be part of an adaptive strategy in evolution of the seasonality in reproduction and migration among photoperiodic species under wild conditions.  相似文献

This review examines possible role(s) of circadian ‘clock’ genes in insect photoperiodism against a background of many decades of formal experimentation and model building. Since ovarian diapause in the genetic model organism Drosophila melanogaster has proved to be weak and variable, recent attention has been directed to species with more robust photoperiodic responses. However, no obvious consensus on the problem of time measurement in insect photoperiodism has yet to emerge and a variety of mechanisms are indicated. In some species, expression patterns of clock genes and formal experiments based on the canonical properties of the circadian system have suggested that a damped oscillator version of Pittendrigh's external coincidence model is appropriate to explain the measurement of seasonal changes in night length. In other species extreme dampening of constituent oscillators may give rise to apparently hourglass-like photoperiodic responses, and in still others there is evidence for dual oscillator (dawn and dusk) photoperiodic mechanisms of the internal coincidence type. Although the exact role of circadian rhythmicity and of clock genes in photoperiodism is yet to be settled, Bünning's general hypothesis (Bünning, 1936) remains the most persuasive unifying principle. Observed differences between photoperiodic clocks may be reflections of underlying differences in the clock genes in their circadian feedback loops.  相似文献

Summary In 1979–80 a field trial of a compound chromosome (CC) strain of the Australian sheep blowfly Lucilia cuprina was conducted in the isolated Brindabella Valley, N.S.W. New genetic material was introduced into the strain before release by inducing 104 new CC elements by irradiation of recently captured field strains, and combining the resulting strains. Weekly releases, averaging 1.1 million larvae per week, were begun in November 1979 and continued to May 1980. Field-inseminated females were trapped weekly and their genotypes and those of their mates were determined through genetic testing. The proportion of wild X wild matings declined from 16% in December 1979 to 1% in April 1980. During this period the proportion of CC X CC matings rose from 50% to 90%. Larvae sampled from infested sheep had compound chromosomes, indicating that compound chromosomebearing females can successfully oviposit in the field. Trapping of flies resumed at the start of the 1980–81 season, without further releases. Progeny tests revealed the presence of both CC and wild flies. The proportions of CC X CC matings among field-inseminated females were 90% in October, 44% in November, nil in December, and 12% in January. No CC X CC matings were detected in 33 field-inseminated females trapped and tested during April, and 70 tested males reared from myiasis samples in April 1981 proved to be wild type. These results indicate that the CC strain overwintered in the field and strongly suggest that it bred in the field for at least one generation following the spring emergence before being eliminated from the population.  相似文献
