The objectives of this study were to examine long-term effects of feeding forage rape (Brassica napus L.) on methane yields (g methane per kg of feed dry matter intake), and to propose mechanisms that may be responsible for lower emissions from lambs fed forage rape compared to perennial ryegrass (Lolium perenne L.). The lambs were fed fresh winter forage rape or ryegrass as their sole diet for 15 weeks. Methane yields were measured using open circuit respiration chambers, and were 22-30% smaller from forage rape than from ryegrass (averages of 13.6 g versus 19.5 g after 7 weeks, and 17.8 g versus 22.9 g after 15 weeks). The difference therefore persisted consistently for at least 3 months. The smaller methane yields from forage rape were not related to nitrate or sulfate in the feed, which might act as alternative electron acceptors, or to the levels of the potential inhibitors glucosinolates and S-methyl L-cysteine sulfoxide. Ruminal microbial communities in forage rape-fed lambs were different from those in ryegrass-fed lambs, with greater proportions of potentially propionate-forming bacteria, and were consistent with less hydrogen and hence less methane being produced during fermentation. The molar proportions of ruminal acetate were smaller and those of propionate were greater in forage rape-fed lambs, consistent with the larger propionate-forming populations and less hydrogen production. Forage rape contained more readily fermentable carbohydrates and less structural carbohydrates than ryegrass, and was more rapidly degraded in the rumen, which might favour this fermentation profile. The ruminal pH was lower in forage rape-fed lambs, which might inhibit methanogenic activity, shifting the rumen fermentation to more propionate and less hydrogen and methane. The significance of these two mechanisms remains to be investigated. The results suggest that forage rape is a potential methane mitigation tool in pastoral-based sheep production systems.  相似文献

The performance of a mesophilic two-stage system generating hydrogen and methane continuously from sucrose (10-30 g/L) was investigated. A hydrogen-generating CSTR followed by an upflow anaerobic filter were both inoculated with anaerobically digested sewage sludge, and ORP, pH, gas output, %H(2), %CH(4) and %CO(2) monitored. pH was controlled with NaOH, KOH or Ca(OH)(2). Using NaOH as alkali with 10 g/L sucrose, yields of 1.62 +/- 0.2 mol H(2)/mol hexose added and 323 mL CH(4)/gCOD added to the hydrogen and methane reactors respectively were achieved. The overall chemical oxygen demand (COD) reduction was 92.6% with 0.90 +/- 0.1 g/L sodium and 316 +/- 40 mg/L residual acetate in the methane reactor. Operation at 20 g/L sucrose and NaOH as alkali led to impaired volatile fatty acid (VFA) degradation in the methane reactor with 2.23 +/- 0.2 g/L sodium, 1,885 mg/L residual acetate, a hydrogen yield of 1.47 +/- 0.1 mol/mol hexose added, a methane yield of 294 mL/gCOD added and an overall COD reduction of 83%. Using Ca(OH)(2) as alkali with 20 g/L sucrose gave a hydrogen yield of 1.29 +/- 0.3 mol/mol hexose added, a methane yield of 337 mL/gCOD added and improved the overall COD reduction to 91% with residual acetate concentrations of 522 +/- 87 mg/L. Operation at 30 g/L sucrose with Ca(OH)(2) gave poorer overall COD reduction (68%), a hydrogen yield of 1.47 +/- 0.2 mol/mol hexose added, a methane yield of 138 mL/gCOD added and residual acetate 7,343 +/- 715 mg/L. It was shown that sodium toxicity and overloading are important issues for successful anaerobic digestion of effluent from biohydrogen reactors in high rate systems.  相似文献

Fermentative production of hydrogen from a wheat flour industry co-product   总被引:3,自引:1,他引:2
The global flour industry produces 96 million ton/year of wheatfeed, which is mainly used for animal feed. This co-product is high in carbohydrates and potentially a significant substrate for biohydrogen production. A 10 l bioreactor, inoculated with sewage sludge, was operated on wheatfeed (10 g l(-1)) at pH 5.5 and 35 degrees C in batch and semi-continuous mode (15 h hydraulic retention time (HRT)). Wheatfeed hydrolysate was also investigated in continuous mode (15 h HRT). NaOH-H2O2 treatment of 25 g l(-1) wheatfeed resulted in hydrolysate containing on average 8.1 g l(-1) total sugar. Hydrogen yields of 64 and 56 m3 H(2) per ton dry weight were produced from wheatfeed in batch and 56 m3 H2 per ton dry weight of wheatfeed in semi-continuous mode. Hydrogen yields from hydrolysate were only 22 and 31 m3 H2 per ton dry weight, (or 0.9 mol H2 per mol hexose degraded, assuming all sugar is hexose). Fermentation of unhydrolysed wheatfeed is therefore recommended. It is calculated that approximately 264 m3/ton of CH4 can be produced from a subsequent anaerobic digestion stage. The biohydrogen produced (diesel equivalents) from the 1.2 million ton/year of wheatfeed in the UK would be more than twice that required for transportation by the UK flour industry.  相似文献

Summary Root hair production by young plants of lucerne, maize and perennial ryegrass grown in a sandy loam was assessed by examining roots growing at a soil-glass interface. Results are given for the percentage frequency distribution of root hair lengths and the numbers of root hairs produced per mm root. The mean lengths of root hairs observed on lucerne, maize and perennial ryegrass roots were 0.35, 0.90 and 1.12 mm respectively. Lucerne produced an average of 105 root hairs per mm of root, whereas maize produced 161 and perennial ryegrass produced 88. The total length of root hairs per mm length of root was estimated to be 37, 146 and 99 mm for lucerne, maize and perennial ryegrass resp. Letcombe Laboratory  相似文献

Information on the interaction between mixed populations in the rumen and plant phenolics is required to fully elucidate the limitations of phenolic compounds on forage digestibility. The objective of this study was to examine the degradation of Italian ryegrass (Lolium multiflorum L.) hay incubated with mixed ruminal populations in consecutive batch culture (CBC) with or without phenolic acids or phenolic compounds extracted from plant cell walls. Each CBC consisted of a series of 10 cultures (3 replicates per culture) inoculated (10%, vol/vol) in sequence at 48-h intervals with microbial suspension from the previous set of cultures. All cultures were grown on a semidefined medium containing Italian ryegrass hay, and each CBC was initiated with an inoculum from the rumen. Rumenlike fermentation characteristics were maintained in control CBCs by repeated inoculum transfer. Treatment CBCs were transferred as described above, but cultures 5, 6, and 7 were incubated in the presence of trans-p-coumaric, cis-p-coumaric, or trans-ferulic acid or phenolics extracted from the cell walls of maize stem or barley straw. Mean apparent dry matter disappearance in control CBC cultures was 495 mg per g of hay, whereas the presence of phenolics reduced the initial dry matter disappearance by 6.3 to 25.6%. trans-p-Coumaric acid and, to a lesser extent, the phenolics from cell walls of maize stem were the most inhibitory compounds for dry matter disappearance and for the production of volatile fatty acids; trans-p-coumaric acid altered the molar ratio of acetate/propionate/butyrate. The CBC further showed variations in the ability of the rumen microbial population to adapt to phenolic compounds.  相似文献

An artisanal static process for protein enrichment of cassava by solid-state fermentation, developed in laboratory and tested on pilot units in Burundi (Central Africa), provides enriched cassava containing 10.7% of dry matter protein versus 1% before fermentation. Cassava chips, processed into granules of 2-4-mm diameter, are moistened (40% water content) and steamed. After cooling to 40 degrees C, cassava is mixed with a nutritive solution containing the inoculum (Rhizopus oryzae, strain MUCL 28627) and providing the following per 100 g dry matter: 3.4 g urea, 1.5 g KH(2)PO(4), 0.8 g MgSO(4).7H(2)O, and 22.7 g citric acid. For the fermentation, cassava, with ca. 60% moisture content, is spread in a thin layer (2-3 cm thick) on perforated trays and slid into an aerated humidified enclosure. The incubation lasts +/- 65 h. The production of protein enriched cassava is 3.26 kg dry matter/m(2) tray. The effects of the variation of the nutritive solution composition and the inoculum conservation period on the protein production are equally discussed.  相似文献

A field plot experiment was set up on a sandy loam soil of SW England in order to determine the efficiency of nitrogen use from different cattle manures. The manure treatments were low and high dry matter cattle slurries and one farmyard manure applied at a target rate of 200 kg total N ha(-1)year(-1), and an untreated control. There were three different cropping systems: ryegrass/clover mixture, maize/rye and maize/bare soil, which were evaluated during 1998/99 and 1999/00. Measurements were made of N losses, N uptake and herbage DM yields. Result showed that manure type had a significant effect on N utilisation only for maize. N balances were negative in maize (approximately -247 to -10 kg N) compared to grass (approximately 5-158 kg N). Agronomic management was more important than manure type in influencing N losses, where soil cultivation appeared to be a key factor when comparing maize and grass systems.  相似文献

Hydrogen was produced from various marine macro-algae (seaweeds) through anaerobic fermentation using an undefined bacterial consortium. In this study, anaerobic fermentation from various marine macro-algae for Ulva lactuca, Porphyra tenera, Undaria pinnatifida, and Laminaria japonica was studied. From this analysis Laminaria japorica was determined to be the optimum substrate for hydrogen production. When L. japornica was used as the carbon source for enhanced hydrogen production, the optimum fermentation temperature, substrate concentration, initial pH, and pretreatment condition were determined to be 35°C, 5%, 7.5, and BT120 (Ball mill and thermal treatments at 120°C for 30 min), respectively. In addition, hydrogen production was improved when the sludge was heat-treated at 65°C for 20 min. Under these conditions, about 4,164 mL of hydrogen was produced from 50 g/L of dry algae (L. japonica) for 50 h, with a hydrogen concentration around 34.4%. And the maximum hydrogen production rate and yield were found to be 70 mL/L·h and 28 mL/g dry algae, respectively.  相似文献

Perennial ryegrass and perennial ryegrass/white clover permanent dairy pastures are compared with respect to productivity, environmental impact and financial costs in nitrate vulnerable zones (NVZ) in the UK. With appropriate management, and utilisation of recommended perennial ryegrass and white clover cultivars, white clover is likely to stabilise at around 20% of total dry matter production in a mixed pasture. Plant dry matter production and milk production from a perennial ryegrass/white clover pasture are likely to be similar to that from a perennial ryegrass pasture receiving 200 kg N ha−1 annum−1 and around 70% of that obtained with perennial ryegrass supplied with 350–400 kg N ha−1 annum−1. Nitrate, phosphorus and methane losses from the system and decreases in biodiversity relative to a grazed indigenous sward are likely to be similar for a perennial ryegrass/white clover pasture and a perennial ryegrass pasture receiving 200 kg N ha−1 annum−1: nitrate leachate from both systems is likely to comply with European legislation. Greenhouse gas emissions resulting from nitrogen (N) fertiliser production would be avoided with the perennial ryegrass/white clover pasture. Within NVZ stocking rate restrictions, white clover can provide the N required by a pasture at a lower financial cost than that incurred by the application of N fertiliser.  相似文献

Regrowth after cutting and the distribution of nitrogen (N),phosphorus (P) and potassium (K) in different plant organs ofwhite clover and perennial ryegrass growing in pure or mixedswards were investigated under field conditions in a soil witha low-to-moderate availability of P and K. In all treatments, white clover constituted more than 70% of the above-ground biomassin the mixed swards. The petioles were the dominant pool ofdry matter throughout regrowth and contained the greatest amountsof N, P and K. Increased supply of P and K increased the growthof ryegrass, but not that of white clover in the mixed swards. The increased competition from ryegrass led to a decline inthe yield of white clover laminae as well as in the N contentper unit of dry matter in laminae, petioles and stolons. TheP content of all white clover organs also declined followingP application to the mixed swards, whereas K application increasedtheir K contents. In the pure swards of ryegrass and white clover, yields and contents of N, P and K in the dry matter were eithernot affected or increased following P and K application. Itwas concluded that commonly-used defoliation heights may remove80% or more of the nutrient and dry matter pools located inthe petioles but the remaining quantities of dry matter andnutrients in the petioles will normally exceed the correspondingquantities in the stolons. A computer simulation model of the turnover of organic matter in soil was adapted to simulate the change in soil organic C and N contents of soil during several years following annual additions of farm slurry to maize fields. The model proved successful in estimating the build-up of both C and N in soil and the leaching of N to ground-water in response to applications of slurry ranging from 50 to 300 tons per hectare per year. The model was then used to estimate the build-up of organic matter in soil under crops of fodder maize that were grown using the excess of manure produced during the last 20 years in the Netherlands. The build-up of organic matter from these applications was estimated to lead to about 70 kg extra nitrogen mineralized ha-1 yr-1. As a result of legislation manure applications have decreased and are expected to decrease further in the immediate future. Calculations suggest that after 10 years of manure applied at rates no longer exceeding the amount needed to replace the phosphorus removed by crops, the extra mineralization of N will still be between 45 and 60 kg ha-1 yr-1. If manure applications cease altogether then the extra mineralization will be about 25–30 kg N ha-1 yr-1.  相似文献

Hydrogen was successfully produced by fermenting primary sewage sludge which had been both heat treated and digested with a commercially available enzyme preparation. When either heat treatment or enzymatic digestion were not used, no hydrogen was produced during fermentation. Heat treated mesophilic anaerobic sludge was used as an inoculum rather than a pure microbial culture. Fermentation was conducted at pH levels ranging from of 4.5 to 7.0. When fermentation took place at pH 5.5 a peak hydrogen production rate of 3.75 ml min(-1) was observed. At this pH the hydrogen yield was 0.37 mol H(2)mol(-1) carbohydrate, equivalent to 18.14L H(2)kg(-1) dry solids.  相似文献

Hydrogen and volatile fatty acids (VFAs) were coproduced from marine macroalgae by anaerobic fermentation using a microbial community. The hydrogen and VFAs production were characterized based on inoculum heat-treatment, methanogen inhibitor addition, operating temperature, and in-situ extraction of VFAs. Maximum hydrogen of 179 mL/g-VS and VFAs concentration of 9.8 g/L were produced from 35 g/L of S. japonica within 5 days of anaerobic fermentation. Hydrogen and VFAs yields were well-correlated with carbohydrate content of substrate. Inoculum heat-treatment significantly improved hydrogen production while the VFAs productivity was affected adversely. The addition of methanogen inhibitors also enhanced the hydrogen production, but the effect on VFAs production was dependent on the type of inhibitor used. Low temperature (25°C) was found to be favorable for high hydrogen and VFAs yield, while high temperature (40°C) and programmed-temperature (35 ~ 45°C) were more favorable for hydrogen and VFAs productivity. Clostridium sp. content was found to be the most abundant at 25°C. An extractive fermentation with anion-exchange resin was tested to recover the VFAs and to control the pH during the anaerobic fermentation.  相似文献

Summary Significant differences in total dry matter yields of shoots and roots were found between 11 ryegrass (Lolium) cultivars grown in a glasshouse. Although shoot yield varied significantly between individual cultivars there was no overall difference between the annual and perennial cultivars; whereas for roots, the yields of the perennial plants were much smaller than those of the annual types. Water use (g H2O g total DM–1) also varied significantly between cultivars. However, there was no relationship between efficient water use and dry matter production. No significant differences were found in shoot composition between the cultviars for nitrogen, phosphorus, and potassium; however, concentrations of sulphur, magnesium, calcium, and sodium varied significantly. Sodium concentrations were generally higher in the annual compared to the perennial cultivars. For roots only nitrogen, phosphorus, and sulphur differed significantly between cultivars. Of the elements only calcium in the shoots was shown to be related to water use. Thus cultivars which were low users of water also had significantly lower calcium concentrations in their shoots. Water use appeared to affect the absorption of calcium by the root to a far greater extent than the transport from roots to shoot. An apparent relationship between magnesium concentration in the shoots and water use was shown to be due to the close association of magnesium with calcium in the plant.  相似文献

The majority of New Zealand dairy goat farmers utilise cultivated green-fed fodder dominated by perennial ryegrass (Lolium perenne L.) and white clover (Trifolium repens L.), but evidence from other ruminant species suggests that milk production may be improved when using a more diverse array of species within the green fodder. The aim of this experiment was to determine whether feeding lactating dairy goats a mixed-species green fodder (MF, consisting of perennial ryegrass, timothy (Phleum pratense L.), prairie grass (Bromus willdenowii Kunth), white clover, red clover (Trifolium pratense L.), lucerne (Medicago sativa L.), chicory (Cichorium intybus L.) and plantain (Plantago lanceolata L.) improves dietary intake, milk yield and composition compared with a standard ryegrass and white clover green fodder (SF). Thirty-six mid-lactation goats were housed indoors in pairs and split into two groups (A and B). The trial was split into three periods – firstly a uniformity period of 6 days, in which all goats were fed a combination of both green fodder types, followed by two treatment periods (P1 and P2) of 12 days, respectively. For P1, group A was fed MF and group B was fed SF, and then the group diets were switched for P2. Goats fed MF had 13% greater dry matter intake and 7% greater milk yield than goats fed SF. In addition, the milk protein and fat concentration of goats fed MF were 4% greater than for those fed SF, whereas there was no effect on milk lactose concentration. There was no treatment effect on the levels of protein, glucose, urea or non-esterified fatty acids in the blood of the goats. An effect of green fodder type on milk fat profile was demonstrated, with proportions of pentadecylic acid (C15:0), cis-vaccenic acid (C18:1 c11), linoleic acid (C18:2 n6) and α-linolenic acid (C18:3 n3) being increased in response to MF consumption. In contrast, iso-C15 and iso-C17 proportions were lesser. In summary, this study demonstrated that goats fed MF increased green fodder intake and milk production compared with goats fed SF. The green fodder type affected the fatty acid profile of goat’s milk, with MF increasing the levels of beneficial polyunsaturated omega fatty acids (linoleic and α-linolenic acids).  相似文献

Lactobacillus intermedius B-3693 was selected as a good producer of mannitol from fructose after screening 72 bacterial strains. The bacterium produced mannitol, lactic acid, and acetic acid from fructose in pH-controlled batch fermentation. Typical yields of mannitol, lactic acid, and acetic acid from 250 g/L fructose were 0.70, 0.16, and 0.12 g, respectively per g of fructose. The fermentation time was greatly dependent on fructose concentration but the product yields were not dependent on fructose level. Fed-batch fermentation decreased the time of maximum mannitol production from fructose (300 g/L) from 136 to 92 h. One-third of fructose could be replaced with glucose, maltose, galactose, mannose, raffinose, or starch with glucoamylase (simultaneous saccharification and fermentation), and two-thirds of fructose could be replaced with sucrose. L. intermedius B-3693 did not co-utilize lactose, cellobiose, glycerol, or xylose with fructose. It produced lactic acid and ethanol but no acetic acid from glucose. The bacterium produced 21.3 +/- 0.6 g lactic acid, 10.5 +/- 0.3 g acetic acid, and 4.7 +/- 0.0 g ethanol per L of fermentation broth from dilute acid (15% solids, 0.5% H(2)SO(4), 121 degrees C, 1 h) pretreated enzyme (cellulase, beta-glucosidase) saccharified corn fiber hydrolyzate.  相似文献

The balance of nitrogen and nitrogen compounds in Pleurotus ostreatus, cultivated on waste materials, corn straw and maize residues, was investigated. The results show that this organism has a considerable ability to fix atmospheric nitrogen, fixing 312 g of total nitrogen per 100 kg dry weight. When recalculating with respect to a decrease of the substrate during growth of the organism a value of 9.7 mg per 1 g of the decrease in material is obtained. Fruiting bodies, as well as the produced substrate contain 17-19 amino acids. In the produced substrate the content of the protein nitrogen represents only 30% of the total. It can concluded that Pleurotus ostreatus yields a suitable raw material in the food industry and possibly also a fodder based on the basis of solid, cellulose-containing wastes.  相似文献
