Sweetness is one of the key factors determining peach fruit quality. To better understand the molecular basis of gibberellic acid (GA) and 1-naphthaleneacetic acid (NAA) interference with sugar biosynthesis, a middle-late maturing commercial cultivar, ‘Jinxiu’ yellow peach fruit, was treated with three different concentrations of GA4+7 and four of NAA. Fruit weight, firmness, total soluble solids, different sugar contents and the expression level of sugar-related genes were evaluated. The results showed that maximum increase in cv. ‘Jinxiu’ peach fruit size and sucrose content was with 1.25 mM GA4+7, compared to control fruits and the other treatments during the ripening stages. The sucrose-phosphate synthase gene (PpSPS2) which had a high level of expression and positive correlation with sucrose content was significantly regulated by 1.25 mM GA4+7 in the final ripening stages. 0.5 mM NAA treatments significantly reduced the sucrose content and fruit size. Ninety percent of the fruits were deformed or dropped from the trees with treatments of 1 mM NAA and 2 mM NAA in the early development period. The crosstalk of different phytohormones and the related genes will be further investigated to get an insight into the inherent association between hormone control and sugar accumulation.

Effect of GA3 on postharvest ripening in strawberry fruit was evaluated through different biochemical parameters. Strawberry slices at different ripening stages were incubated with GA3. A significant decrease on respiratory activity depending on GA3 concentration was obtained. Also GA3 was applied to whole and deachened fruit at white and green ripening stages. Our results show that GA3 has an inhibitory effect on strawberry fruit ripening, evidenced by a decrease in the respiratory activity and a delay in anthocyanin synthesis and chlorophylls degradation.  相似文献

In Citrus, gibberellic acid (GA3) applied at the floral bud inductive period significantly reduces flowering intensity. This effect is being used to improve the fruit set of parthenocarpic cultivars that tend to flower profusely. However, the molecular mechanisms involved in the process remain unclear. To contribute to the knowledge of this phenomenon, adult trees of ‘Salustiana’ sweet orange were sprayed at the floral bud inductive period with 40?mg?L?1 of GA3 and the expression pattern of flowering genes was examined up to the onset of bud sprouting. Trees sprayed with paclobutrazol (PBZ, 2,000?mg L?1), a gibberellin biosynthesis inhibitor, were used to confirm the effects, and untreated trees served as control. Bud sprouting, flowering intensity, and developed shoots were evaluated in the spring. GA3 significantly reduced the number of flowers per 100 nodes by 72% compared to the control, whereas PBZ increased the number by 123%. Data of the expression pattern of flowering genes in leaves of GA3-treated trees revealed that this plant growth regulator inhibited flowering by repressing relative expression of the homolog of FLOWERING LOCUS T, CiFT, whereas PBZ increased flowering by boosting its expression. The activity of the homologs TERMINAL FLOWER 1, FLOWERING LOCUS C, SUPPRESSOR OF OVEREXPRESSION OF CONSTANS 1, and APETALA1 was not affected by the treatments. The number of flowers per inflorescence, in both leafy and leafless inflorescences, was not altered by GA3 but increased with PBZ; the latter paralleled LEAFY relative expression. These results suggest that GA3 inhibits flowering in Citrus by repressing CiFT expression in leaves.  相似文献

Four different plant growth regulators, gibberellic acid (GA3), naphthaleneacetic acid (NAA), benzyladenine (BA) and abscisic acid (ABA), were individually mixed in a lanolin paste and applied to immature fruit on grapefruit trees beginning soon after fruit set. The treated fruit was allowed to mature on the tree. Application of 1000 ppm GA3 in this manner generally increased fruit size, decreased the concentration of the total acid in the juice and decreased the concentration of naringin in the juice sacs compared to that of the controls. GA3 increased the total soluble solids (brix) in the juice in some experiments. Treatment of fruit with 1000 ppm ABA and BA significantly decreased the size of the fruit and increased the naringin concentration, but had variable effects on the soluble solids content and the acid content. Early foliar spray treatments containing gibberellic acid(GA3) significantly lower the concentration ofthe bitter flavonoid naringin in fruit tissues. Sprayscontaining a surfactant and different levels ofGA3 (5, 50, 100, and 500 ppm) or abscisic acid(ABA) (5, 25, and 50 ppm) were applied to young,developing fruit on mature grapefruit (Citrusparadisi) trees during the period from April to June,beginning just after fruit set. The fruit were allowedto mature and were harvested early the following year.Harvested fruit were evaluated for weight, juicecharacteristics, and flavonoid concentrations.GA3 application resulted in larger mature fruit,which yielded juice with the same soluble solids valueas juice from control fruit, but with slightly loweracid percentages and lower concentrations of naringin.ABA treatment had little effect on juice solublesolids, acid content and naringin content except atthe highest concentration of 50 ppm, which lowerednaringin levels slightly in juice.  相似文献

In this study we investigated the enantioselective degradation of (2RS,3RS)‐paclobutrazol in peach and mandarin fruits under field conditions after foliar treatment at 500 mg active ingredient/L using a Lux Cellulose‐1 chiral column on a reverse‐phase liquid chromatography–tandem mass spectrometry system. Degradations of paclobutrazol in both fruits followed first‐order kinetics, with half‐lives of about 9 days. Initial deposits were 1.63 mg/kg on peach and 1.99 mg/kg on mandarin; terminal concentrations were lower than 0.05 mg/kg, which was acceptable in most cases. As anticipated, paclobutrazol levels in peels of mature mandarin were about 6.3 times higher than in pulp, indicating the potential risk of peel consumption. We also observed that paclobutrazol degradation in mature mandarin was relatively slow, indicating it might not be efficient enough to hold mandarin fruits on trees for lowering paclobutrazol concentrations. Significant enantioselectivity was observed: the (2R,3R)‐enantiomer was preferentially degraded in mandarin (whole fruit, peels, and pulp) but enriched in peach. Because of its more rapid degradation in mandarin and the lower levels observed in pulp compared with peels, potential endocrine‐related side effects due to the (2R,3R)‐enantiomer pose less of a risk in mandarin than in peach. Flower buds of peach (Prunus persica L.) trees, cv Novedad de Cordoba (Argentina), were collected near the end of the dormant period and immediately before anthesis. After removal of scale leaves, morphological observations of representative buds, made on transverse and longitudinal microtome sections, showed that all verticils making up the flower are present in an undifferentiated form during the dormant period (June). Flower buds collected at the end of dormant period (August) showed additional growth and differentiation, at which time formation of two ovules was beginning in the unicarpelar gynoecium. Dehiscence of anthers had not yet occurred 10 days before full bloom, and the ovules were still developing. Free endogenous gibberellin (GA)-like substances were quantified by bioassay (Tan-ginbozu dwarf rice microdrop) after SiO2 partition column chromatography, reversed phase C18-high performance liquid chromatography, and finally Nucleosil [N(CH3)2]high performance liquid chromatography. Bioactive fractions were then subjected to capillary gas chromatography-mass spectrometry-selected ion monitoring (GC-MS-SIM). Gibberellins A1, A3, and A8 were tentatively identified in peach flower buds using GC-SIM and Kovat's retention indices, and relative amounts approximated by GC-SIM (2:8:6 for GA1, GA3, and GA8, respectively). The highest concentration (330 nanograms per gram dry weight) of free GA1/GA3 was found in dormant buds (June) and diminished thereafter. The concentration free of GA1/GA3 did not increase immediately prior to bud break. However, high GA1/GA3 concentrations occurred during stages where rate of growth and cellular differentiation of (mainly fertile) verticils can be influenced.  相似文献

Tissue-culture-propagated own-rooted cv. Spartan apple trees (Malus domestica Borkh.) planted in 1979 were treated in 1983 and 1985 via a soil-line trunk drench with the plant growth retardant paclobutrazol [(2RS, 3RS)-1-(4-chlorophenyl)-4.4-dimethyl-2-(1,2, 4-triazol-1-yl) pentan-3-ol]. Seeds of immature fruits from untreated and treated trees were sampled in 1989 ca 75 days after full bloom. After seeds were freeze-dried, gibberellins (GAs) were extracted, purified and fractionated via C18 reversed-phase high-performance liquid chromatography (HPLC). Gibberellins A1, A3, A4, A7, A8, A9, A15, A17, A19, A20, A24, A34, A35, A44, A51, A53, A54, A61, A62, A63 and A68 were identified by using C18 HPLC, gas chromatography-selected ion monitoring and Kovats retention indices. Eight of the GAs identified were also quantified by using deuterated internal standards. The paclobutrazol applications caused a 55% reduction of vegetative shoot elongation in 1989, but both treated and untreated trees had developed a biennial bearing pattern by that time (heavy bloom or “on year’in 1989). Levels of early 13-hydroxylation pathway GAs, viz. GA53, GA19, GA20, GA1 and also GA3, were not altered by treatment. However, GA4, GA7 and GA9 were increased 13.4, 6.5 and 3.8 times, respectively, in seeds of fruit from treated compared to untreated trees.  相似文献

The specific inhibitor of gibberellin biosynthesis, (2RS,3RS)-1-(4-chlorophenyl)-4,4-dimethyl-2-(1,2,4-triazol-1-yl)pentan-3-ol (paclobutrazol), inhibited germination ofAmaranthus caudatus L. seeds. Addition of gibberellic acid (GA3), 2-chloroethylphosphonic acid (ethephon), or 1-aminocyclopropane-1-carboxylic acid (ACC) effectively antagonized inhibition. Ethephon was found to be the most efficient antagonist. The transfer of seeds after 1 day's incubation in paclobutrazol to solutions of GA3 or ethephon reversed the inhibition, the effect increasing with increasing concentration of GA3 or ethephon. Seeds incubated in paclobutrazol for 5 days decreased sensitivity to GA3 and ethephon.  相似文献

Seeds from heavily fruiting (on-year), mature untreated, and paclobutrazol-treated apple trees (Malus domestica Borkh. cv. Spartan) were sampled in mid-June 1987, mid-July 1987, and mid-July 1990. After seeds were freeze-dried, gibberellins (GAs) were extracted, purified, and fractionated via C18 reversed-phase high-performance liquid chromatography (HPLC). Nine GAs (GA1, GA3, GA4, GA7, GA8, GA9, GA19, GA20, and GA53) were quantified by the use of deuterated GA internal standards. Paclobutrazol trunk drench treatments reduced vegetative shoot elongation in the seasons that seeds were sampled by 55% or more. Between June 17, 1987 and July 15, 1987, the dry weight of seeds from both untreated and treated trees increased about 2.5 times and there were reductions, on a per seed basis, of GA4 in seeds from both untreated and treated trees, of GA7 in seeds from treated trees, and of GA9 in seeds from untreated trees. However, GA9 increased in seeds from treated trees. Changes in levels of some of the early-13-hydroxylation pathway GAs (GA15 GA3, GA8, GA19, GA20, and GA53) also occurred during the month. For mid-July harvested seeds, the pattern, with some exceptions, was that 2 years after paclobutrazol treatment (1987), levels of early-13-hydroxylation pathway GAs in seeds from treated trees were lower compared to controls but after 5 years (1990) their levels tended to increase. For the non-13-hydroxylated GAs (GA4, GA7, and GA9), 2 years after paclobutrazol treatment, GA4 levels were equal in seeds from untreated and treated trees, GA7 decreased in seeds from treated trees compared with controls, but GA9 levels increased. Levels of these three GAs were higher in seeds from treated trees 5 years after treatment (1990) but levels of GA4, GA7, and GA9 dramatically increased in seeds from treated trees 4 years after treatment (1989), as we previously reported.  相似文献

The triazole plant growth regulators, paclobutrazol and uniconazole, reduced in vitro growth of moth bean callus by 20–25% when added to the culture medium at 1 mg/L (3.4 μM). The addition of 10 mg/L (29 μM) gibberellic acid (GA3) to the culture medium in combination with the triazoles restored callus growth to a level equivalent to that of the untreated control. GA3 alone had little effect on callus growth. When added to a regeneration medium at 1 mg/L both paclobutrazol and uniconazole reduced the percentage of cultures that formed roots, as well as the mean number of roots per culture. In contrast, GA3 increased root formation and counteracted the inhibitory effects of the triazoles on rooting. The addition of triazoles or GA3 to the regeneration medium reduced the formation of green meristematic nodules, which are precursors of shoots in moth bean callus. When callus was grown in the presence of either paclobutrazol or uniconazole, subsequent root and green meristematic nodule formation were reduced upon transfer to a growth regulator-free regeneration medium. The results of this study indicate that exposure of moth bean callus tissue to micromolar concentrations of triazoles or GA3 can significantly alter in vitro growth and differentiation.  相似文献

The involvement of gibberellins in the control of flowering of sunflower was studied by direct application of GA3 to the apex of the plants, analysis of the endogenous levels of gibberellin-like substances at different plant ages, and indirectly by the application of paclobutrazol, an inhibitor of gibberellin synthesis. GA3 speeded-up flower initiation and floral apex development. The time of GA3 application was more critical than the amount of GA3 applied. The endogenous levels of gibberellin-like compounds increased significantly by day 15 after sowing. The application of paclobutrazol markedly delayed floral initiation and this effect was also depedent on plant age. Both GA3 and paclobutrazol had their greatest effects between 10 and 20 days after sowing suggesting that an increase in gibberellins in that time period plays a role in floral initiation.  相似文献

A reduction in concentration of gibberellins has been implicated in the phase change from juvenile to mature forms of ivy (Hedera helix L.). Attempts were made to increase the effective internal concentration of gibberellins by exogenous application of GA3, and to decrease them by various applications of abscisic acid (ABA) and paclobutrazol (PP333), alone or in combination with GA3. ABA and GA3 were fed directly into the xylem of ivy plants by a wick system (a less drastic procedure than the defoliation or decapitation used by earlier workers) whereas PP333 was applied as a soil drench.Mature ivy responded to the application of GA3 by reversion to the juvenile form, although this reversion was incomplete with respect to leaf lobing and red (anthocyanin) pigmentation and could occur spontaneously without the application of GA3. Contrary to expectation, applications of ABA and PP333 caused the stimulation of growth in juvenile ivy. No adult characteristics were induced. As similar concentrations of ABA and PP333 produced severe retardation of growth (which could be alleviated by the application of GA3) in other species, it is suggested that ivy may be an unsuitable model system for the investigation of phase change in woody plants.  相似文献

Endogenous gibberellins (GAs) were extracted from safflower (Carthamus tinctorius L.) stems and detected by capillary gas chromatography-mass spectrometry from which GA1, GA3, GA19,, GA20, GA29, and probably, GA44 were detected. The detection of these GAs suggests that the early 13-OH biosynthetic pathway is prevalent in safflower shoots. Deuterated GAs were used as internal standards and GA concentrations were determined in stems harvested at weekly intervals. GA1 and GA19 levels per stem increased but concentrations per gram dry weight decreased over time. GA20 was only detected in young stem tissue.Gibberellic acid (GA3) was also applied in field trials and both GA3 and the GA biosynthetic inhibitor, paclobutrazol, were applied in growth chamber tests. GA3 increased epidermal cell size, internode length, and increased internode cell number causing stem elongation. Conversely, paclobutrazol reduced stem height, internode and cell size, cell number and overall shoot weight. In field tests, GA3 increased total stem weight, but decreased leaf weight, flower bud number and seed yield. Thus, GA3 promoted vegetative growth at the expense of reproductive commitment. These studies collectively indicate a promotory role of GAs in the control of shoot growth in safflower, and are generally consistent with gibberellin studies of related crop plants. The effects of NAA (naphthaleneacetic acid), GA3 (gibberellic acid), CPPU (N-(2-chloro-4-pyridyl)-N'-phenylurea) and pollination on fruit set, cell division and enlargement were studied in Lagenaria leucantha, an important vegetable. NAA and GA3 were ineffective in inducing parthenocarpy, whereas CPPU induced parthenocarpic fruit significantly larger than fruit that resulted from pollination. Cell division, which occurred during the first 4 days after pollination was not reactivated by NAA or GA3, but was effectively reactivated by CPPU. The cell number of the total cross-section of CPPU-treated fruit was 117.4% of that of pollinated fruit and 154.4% of that of unpollinated at 12 DAA (days after anthesis) respectively. The CPPU-induced parthenocarpic fruit had the largest cell cross-sectional area followed, successively, by pollinated fruit, NAA-treated fruit, GA3-treated fruit and unpollinated fruit. These results indicate that CPPU induced parthenocarpic fruit growth by directly reactivating cell division and expansion.  相似文献

In mainland China, the most popular pineapple cultivar is ‘Comte de Paris’. Gibberellic acids have been widely applied to enhance fruit growth in various species. To evaluate the effect of gibberellic acid (GA3) on ‘Comte de Paris’ pineapple production and quality, pineapple fruits were sprayed with GA3 at concentrations of 5, 20, 50, or 100 mg l−1 at both 0 and 15 days after flowering (DAF). Fruits were sampled every 15 days from 0 to 60 DAF (maturation) for flow cytometric analysis and histological observation. The results showed that the treatments with the three highest concentrations of GA3 significantly increased fruit weight, and the most effective concentration was 50 mg l−1 GA3, which increased the flesh weight by 20.3% compared to the control. Although treatment with GA3 had little effect on the total soluble solids and fruit juice pH, it increased the vitamin C content. Although flow cytometric analysis showed that the 50 mg l−1 GA3 treatment had only a slight impact on the number of S phase cells, histological observations indicated that the increase of fruit volume and flesh weight under this GA3 treatment was not due to the increase of cell number but a result of the increase of cell area in the fruit flesh.  相似文献

A significant depression of callus growth resulted from low concentrations of abscisic acid (ABA) added to the medium recommended by Linsmaier and Skoog. Low concentrations also decreased the chlorogenic acid and lignin content of the callus, and generally decreased amounts of scopolin and scopoletin in the tissue. Gibberellic acid (GA3) stimulated callus growth in a low concentration (0.1 mg/1) and inhibited growth at a high concentration (10.0 mg/1). Both levels of GA3 increased scopoletin accumulation in tobacco callus. A high concentration of GA3 increased the accumulation of scopolin and chlorogenic acids, whereas a low concentration decreased the amounts of these two phenolic compounds. In comparison with the control, lignin synthesis was stimulated by a low GA3 concentration, but a high GA3 concentration did not have a significant effect. Both low and high concentrations of GA3 overcame ABA inhibition of growth and lignin synthesis, and partially reversed ABA inhibition of scopoletin production. However, GA3 did not reverse the inhibitory effect of ABA on scopolin production. The low concentration of GA3 overcame the inhibition of chlorogenic acid production resulting from a 0.01 mg/1 concentration of ABA, but this was the only reversal of chlorogenic acid inhibition resulting from addition of GA3 to the medium.  相似文献

Background  

Acidity is an essential component of the organoleptic quality of fleshy fruits. However, in these fruits, the physiological and molecular mechanisms that control fruit acidity remain unclear. In peach the D locus controls fruit acidity; low-acidity is determined by the dominant allele. Using a peach progeny of 208 F2 trees, the D locus was mapped to the proximal end of linkage group 5 and co-localized with major QTLs involved in the control of fruit pH, titratable acidity and organic acid concentration and small QTLs for sugar concentration. To investigate the molecular basis of fruit acidity in peach we initiated the map-based cloning of the D locus.  相似文献
