During vegetative growth in controlled environments, the patternof distribution of 14C-labelled assimilates to shoot and root,and to the meristems of the shoot, was measured in red and whiteclover plants either wholly dependent on N2 fixation in rootnodules or receiving abundant nitrate nitrogen but lacking nodules. In experiments where single leaves on the primary shoot wereexposed to 14CO2, nodulated plants of both clovers generallyexported more of their labelled assimilates to root (+nodules),than equivalent plants utilizing nitrate nitrogen, and thiswas offset by reduced export to branches (red clover) or stolons(white clover). The intensity of these effects varied with experiment.The export of labelled assimilate to growing leaves at the terminalmeristem of the donor shoot was not influenced by source ofnitrogen. Internode elongation in the donor shoot utilized nolabelled assimilate. Whole plants of white clover exposed to 14CO2 on seven occasionsover 32 days exhibited the same effect on export to root (+nodules),which increased slightly in intensity with increasing plantage. Nodulated plants had larger root: shoot ratios than theirequivalents utilizing nitrate nitrogen. Removal of the blade from the leaf subtending the first flowerbud on Cyclamen persicum ‘Swan Lake’ plants causedthe petiole of that leaf to senesce, but had no effect on thegrowth of the flower peduncle in the debladed petiole's axil.A 10 mg NAA l–1 application generally had no effect onpetiole senescence, peduncle elongation or flowering date whenapplied to the cut end of the petiole after blade removal. A25 mg GA3 l–1 application or a combination of 25 mg GA3l–1 application or a combination of 25 mg GA3 l–1plus 10 mg NAA l–1 delayed petiole senescence and enhancedpeduncle elongation and subsequent flowering. A quantitative analysis of the 14C-labelled assimilate suppliedby the expanded leaves on the primary shoot to growing leaves,stem, lateral shoots (branches or stolons) and roots in redand white clover was conducted during vegetative growth. Stem growth of the primary shoot was inhibited in both cloversand utilized no energy resources. The growing leaves at theprimary shoot apex of white clover imported 4 per cent of theshoot's assimilate compared with 10 per cent in red clover.At the basal end of the primary shoot, the tap root of whiteclover imported 16 per cent of the shoot's assimilate comparedwith 22 per cent in red clover. Branches in red clover and stolonsin white clover were by far the largest sinks for primary shootassimilate, importing 39 per cent and 63 per cent of the labelledassimilate, respectively. Analyses of the translocation of assimilate from individualprimary shoot leaves demonstrated that in both clovers olderleaves exported more of their assimilate to branches or stolons,whereas younger leaves exported more of their assimilate toroots, and possibly in white clover, to growing leaves at thetip of the shoot. Of the labelled assimilate exported to branchesor stolons, each primary shoot leaf exported preferentiallyto the branch or stolon in its own axil, but in addition exportedsubstantial quantities of assimilate to all other axillary shoots,particularly those arising from basal axils where the subtendingleaf had died. Differences in assimilate partitioning in response to heat stresswere observed between the heat-sensitive tomato cultivar, RomaVF and the heat-tolerant cultivar, Saladette. Transport of carbonto the trusses and apex was inhibited in both cultivars, particularlyin Roma VF. Basipetal transport to the roots was inhibited inRoma VF only. Assimilate partitioning within the young trusswas also affected by heat stress in both cultivars, but wasmore pronounced in Roma VF: at higher temperatures more 14Cwas found in the peduncle and in the older flower buds thanin the younger flower buds. The youngest flower buds were moresensitive than other parts of the truss to heat stress. Theuptake of [14C]sucrose by detached flower buds from agar mediumwas lower at higher temperatures. Alteration of assimilate partitioningwas also observed following the application of GA3+Kinetin tothe first truss. The relative assimilatory activity of the inflorescence, itsindividual components, and the leaves of flowering tillers ofPoa annua L. and Lolium perenneL. was determined over the periodfrom inflorescence emergence to seed shedding. The pattern of14CO2 fixation was similar for both species and the inflorescencewas by far the most important assimilatory organ of the reproductivetiller, particularly over the latter period of seed developmentas leaf senescence progressed. With the exception of the seedsall parts of the inflorescence showed significant assimilatoryactivity and the lemmas and paleas accounted for 40–50per cent of the total 14C fixed by the inflorescence in bothspecies. The importance of the grass inflorescence as a photosyntheticstructure is discussed in relation to similar studies on cereals. Embryogenic callus was induced from immature inflorescence segmentsof Java citronella (Cymbopogon winterianus) and maintained for2 years on Murashige and Skoog's medium supplemented with 2,4-D(l mg l–1). The callus cells retained the original chromosomenumber of 2n = 20. The somatic embryos germinated into plantletson MS basal medium or medium with IAA, NAA, BAP or KN individually(l mg l–1). The regenerated plantlets developed a goodroot system on full strength solid MS inorganics medium withIAA (1 mg l–1). The regenerated plants were similar tothe donor plant in morphology and had the same chromosome number,but showed some variation in the essential oil content. Uptake and partitioning through the xylem and phloem of K+,Na+, Mg2+ , Ca2+ and Cl were studied over a 9 d intervalduring late vegetative growth of castor bean (Ricinus communisL.) plants exposed to a mean salinity stress of 128 mol m–3NaCl. Empirically based models of flow and utilization of eachion within the whole plant were constructed using informationon ion increments of plant parts, molar ratios of ions to carbonin phloem sap sampled from petioles and stem internodes andpreviously derived information on carbon flow between plantsparts in xylem and phloem in identical plant material. Salientfeatures of the plant budget for K+ were prominent depositionin leaves, high mobility of K+ in phloem, high rates of cyclingthrough leaves and downward translocation of K+ providing theroot with a large excess of K+ . Corresponding data for Na+showed marked retention in the root, lateral uptake from xylemby hypocotyl, stem internodes and petioles leading to low intakeby young leaf laminae and substantial cycling from older leavesback to the root. The partitioning of the anionic componentof NaCl salinity, Cl, contrasted to that of Na+ in thatit was not substantially retained in the root, but depositedmore or less uniformly in stem, petiole and leaf lamina tissues.The flow pattern for Mg2+ showed relatively even depositionthrough the plant but some preferential uptake by young leaves,generally lesser export than import by leaf laminae, and a returnflow of Mg2+ from shoot to root considerably less than the recordedincrement of the root. Ca2+ partitioning contrasted with thatof the other ions in showing extremely poor phloem mobility,leading to progressive preferential accumulation in leaf laminaeand negligible cycling of the element through leaves or root.Features of the response of Ricinus to salinity shown in thepresent study were discussed with data from similar modellingstudies on white lupin (Lupinus albus L.) and barley (Hordeumvulgare L.) The effects of temperature and photoperiod on d. wt partitioningand 14C translocation were studied in three potato varieties.High temperatures and long days enhanced plant growth in termsof plant height and number of leaves, and also affected d. wtpartitioning between the plant organs. However, no temperatureeffect was noted on total plant d. wt, nor on the export of14C from the source leaf. Translocation of 14C to the vegetativeorgans (leaves and stems) was greater at higher temperatures,while translocation to the tubers was less under these conditions.We suggest that, under the temperature regimes studied, themain effect of high temperature is on assimilate partitioningand not on total plant productivity. The size of the apical dome of Chrysanthemum morifolium Ramat.at the transition to inflorescence initiation in continuouslight (long days) was not systematically influenced by eitherthe temperature or the irradiance under which the plants weregrown. It was generally 0.26 mm in diameter and c. 3.6 x 10–3mm3 in volume when the first bract was initiated. The dimensionsof the apical dome of plants in short days were only slightlysmaller at this stage. Similarly, each step in the further developmentof the chrysanthemum inflorescence was associated with a narrowrange of apex sizes, indicating that inflorescence initiationand development are closely related to apex size. Floret elongation and levels of precarthamin were investigatedin freshly collected flowers of Carthamus tinctorius. Accumulationof precarthamin was found to be induced at the early stagesof floret elongation. [U-14C]Acetate and [U-14C]phenylalaninewere incorporated into precarthamin in the detached floretsfrom the flower bud. Effects of temperature on floral initiation of ten white clovervarieties growing in controlled environments are described.Plants grown under long days (16 h) were subjected to constanttemperatures of 26, 18 and 10 °C. Relationships betweenmorphological and physiological traits and flowering were examined. Most plants flowered at the two higher temperatures but only10 per cent of plants flowered at 10 °C. Larger leaved typestended to produce more reproductive buds per stolon at the highertemperatures than did smaller leaved varieties. Of the floral characters studied, floret number was least affectedby temperature. Ovule number and peduncle length were greatestat 18 °C. The import of photosynthate labelled with 11C from a sourceleaf into the two halves of a split root system of an intactbarley plant was studied. When applied to one half of a splitroot system sugars that are absorbed and metabolized reducesubsequent import of 11C into that root half. The non-metabolizedsugar analogue 3-O-methyl glucose has no effect on import, whilstmannose and 2–deoxyglucose inhibit both root elongationand import of 11C. EDTA, PCMBS, and apoplastic pH in the range4–7, have little effect on partitioning. These resultsare interpreted in terms of a suggestion that phloem unloadsdirectly into expanding cells in the elongation zone of roottips. By direct somatic embryogenesis in vitro a clone of asepticplantlets can be raised from a single immature embryo of Trifoliumrepens (white clover) within about 6 weeks of pollination. Embryoidsare induced directly from intact zygotic embryonic tissue ona culture medium containing 0·025 or 0·05 mg 1–1BAP and 1·0 g 1–1 yeast extract. Similar directsomatic embryogenesis has also been achieved for Trifolium pratense(red clover) and Medicago sativa (lucerne). Simulated mixed swards of Perennial Ryegrass (Lolium perenneL.) cv. S23 and White clover (Trifolium repens L.) cv. S100were grown from seed under a constant 20 °C day/15 °Cnight temperature regime and harvested at intervals over and88 d growht period. The swards received a nutrient solutiondaily, which was either High (220 mg l1) or Low (10 mgl–1) in nitrate N. The nitrate was labelled with the 15Nisotope. An acetylene reduction assay was carried out on eachsward just prior to harvest. Rates of acetylene reduction agreed qualitatively with the l5Nanalyses but absolute values did not match (assuming a 4:1 C2H4:N2ratio) and errors in the acetylene assay are discussed. In theLow-N swards clover relied almost entirely on symbioticallyfixed N2, fixing more than ten times as much as the High-N cloverplants. In the Low-N treatment the grass was N-deficient despiteobtaining much more nitrate per unit root dry weight than clover.In the High-N swards, however, clover took up more nitrate perunit root weight than grass. The High-N clover plants also fixedsome N2 and maintained a higher total-N content than grass throughoutthe period. There was no evidence of transfer of symbioticallyfixed N from the clover to the grass in either treatment. The pattern of breakdown of seed coat impermeability of seedwithin the burrs of two cultivars of subterranean clover (Trifoliumsubterraneum L.) was closely associated with relative differencesin flowering sequence within the inflorescence. The patternof seed weight variation showed very little relationship withthe pattern of breakdown, while that for seed moisture contentshowed no relationship. Within a burr it was almost invariably the seed which developedfrom the latest flower within the inflorescence which was thefirst to become permeable. An ordering of the timing of breakdownwas evident only within the burr; there was no evidence fora pattern of breakdown of impermeability between the differentburrs along the runner. The adverse effect of lodging on grass seed yield may be attributed,in part, to assimilate limitation during the seed filling period.This investigation examined plant dry matter assimilate partitioningand seed yield as affected by lodging in three species thatare closely related but phenotypically different: tall fescue(Festuca arundinacea Schreber.), Italian ryegrass (Lolium multiflorumLam.), and perennial ryegrass (L. perenne L.). Studies wereperformed in field plots at Corvallis, Oregon, USA. Seed yieldcomponents (seed number per inflorescence, seed yield per inflorescence,and single seed mass) and leaf, stem (lower, middle, and peduncle)and seed inflorescence dry mass were measured just prior toanthesis to seed maturity. Dry mass and water soluble carbohydrates(WSC) were determined for shoot components. The reduction indry mass and WSC in leaves and stem components following anthesiswas often greater in lodged plants compared to upright plants.The relatively low seed yield depression in lodged tall fescuesuggested a higher compensation potential for partitioning reserveassimilate from leaves and stems to support seed growth anddevelopment. This potential does not appear to be present tothe same degree in Italian ryegrass and to an even lesser extentin perennial ryegrass. These findings suggest that the potentialto compensate for reduced assimilate supply during the periodof high assimilate demand by seeds may be attributed, in part,to the total assimilate reserve accumulated prior to photoassimilatereduction caused by the lodged condition. The fourth fully expanded leaf on the main stolon of white cloverplants was exposed to 14CO2. Thereafter, quantitative and fractionalanalysis of the partitioning, storage and remobilization afterdefoliation of the 14C labelled assimilate was sequentiallyconducted over a 2- to 3-week period. In undefoliated plants, most 14C reached its final destinationwithin 24 h of feeding. Forty percent of assimilated 14C waslost through respiration, while the rest was exported, predominantlyto meristems, but also to roots, stolons and leaves. The 14Cinitially translocated to meristems was subsequently recoveredin stolon and leaf tissue as the plants matured. Approximately 10% of assimilated 14C was invested into long-termstorage in roots and stolons. These reserves were remobilizedafter both partial and total defoliation, and a portion of theremobilized 14C was incorporated into new growth, Partly defoliatedplants regrew more rapidly than totally defoliated plants, butmore 14C reserve depletion took place in the totally defoliatedtreatment. Microswards of white clover (Trifolium repens L.) were grownin controlled environments at 10/7, 18/13 and 26/21 °C day/nighttemperatures. The vertical distribution of leaves of differentages and their rates of 14CO2-uptake in situ were studied. Extending petioles carried the laminae of young leaves throughthe existing foliage. A final position was reached within 1/4to 1/3 of the time between unfolding and death. Newly unfoldedleaves had higher rates of 14CO2-uptake per leaf area than olderones at the same height in the canopy. At higher temperatures,the decrease with age was faster. However, the light-photosynthesisresponse of leaves which were removed from different heightsin the canopy varied much less with leaf age than did the ratesof 14CO2-uptake in situ. The comparison of the rates of 14CO2-uptake in situ with thelight-photosynthesis response curves suggests that young leavesreceive more light than older ones at the same height in thecanopy. This would imply that young white clover leaves havethe ability to reach canopy positions having a favourable lightenvironment. This ability may improve the chances of survivalof white clover in competition with other species. A model of nitrogen uptake and distribution is presented whichdescribes these processes in relation to the amount of availablesoil nitrate and the rate of plant growth. Nitrogen uptake iseither sink or source limited. Sink limitation is based on maximumN-concentrations of plant compartments. The N-uptake model iscombined with a photosynthesis model based on the productivity-nitrogenrelationship at the single-leaf level. The model is parameterizedusing cauliflower as an example crop. Applied to an independentdata set, the combined model was able to predict leaf, stemand inflorescence nitrogen concentrations with correlation coefficientsbetween predicted and simulated values of 0.89, 0.66 and 0.86,respectively. The influence of nitrogen supply and light intensityon leaf nitrate-N could also be predicted with good accuracy(r2 = 0.87). Dry matter production based on the productivity-Nrelationship and the partitioning into leaf, stem and inflorescencewas also reproduced satisfactorily (r2 = 0.91, 0.93 and 0.92,respectively). The net efflux of H+ from lucerne (Medicago saliva L.), redclover (Trifolium pratense L.) and white clover (Trifolium repensL.) growing in flowing solution culture and dependent upon symbioticfixation of atmospheric N, was measured over a 75 d experimentalperiod. Considerable and rapid increases in acidity of the nutrientsolution of up to 1.45 pH units were recorded when the pH wasriot held constant over a 30 h period. There was little differencein H+ efflux when solution pH was held constant at 4.75, 5.75or 6.75, but there was an immediate cessation when it was adjustedto 3.75. Differences in the daily net efflux of H+ closely followedthe pattern of daily differences in incoming radiation, andthere was also evidence of a diurnal pattern of H+ efflux. Althoughthere were initially distinct differences between the speciesin the calculated rate of net H+ efflux (µg H+ g–1dry shoot d), by day 75 these had diminished. In allspecies, however, the maximum rate of efflux per unit of shootsoccurred during the earlier rapid phases of growth. The measuredefflux of H+ was well equated with the plant content of excesscations (as measured by ash alkalinity) and, on average, theratio of acidity produced to N assimilated (expressed as anequivalent) was 0-24.
