以酿酒酵母乙醇发酵高产工业菌株MF1002为始发菌株,对其营养细胞进紫外线诱变,筛选得到两株性状稳定的呼吸缺陷型突变体MF15c和MF11a。菌体细胞对2,3,5-氯化三苯四氮唑(TTC)显色测定呼吸强度的结果表明,两突变菌株的相对呼吸强度分别只有始发菌株的57.77%和47.25%。与现有报道的呼吸缺陷突变体不同,两株突变体的细胞生长速率只在培养初期略低于始发菌株,总体生长速率与始发菌株几乎没有差异,在YPD平板上培养也不形成小菌落。比较蔗糖发酵试验表明,两株突变体的乙醇产量较始发菌株只分别略提高6.48%-6.59%(MF15c)和1.66%-1.97%(MF11a),但发酵终止的残总糖含量却显著低于始发菌株,分别减少34.85%和19.70%,发酵效率较始发菌株分别显著提高6.69%和4.71%,表明这两株突变体为新型的呼吸缺陷型突变。鉴于提高乙醇发酵的发酵效率可显著降低生产成本,认为这两株突变菌株具有较高的潜在工业利用价值。  相似文献

高渗透压胁迫是降低生物法制备丁二酸生产效率的关键因素之一。为提高丁二酸生产菌株对高渗透压胁迫的耐受性能,本研究考察了外源引入全局调控蛋白IrrE提高大肠杆菌耐高渗透压胁迫性能的可行性。试验结果表明,在不同浓度Na+胁迫下,重组菌生长和发酵性能明显提升。在5 L罐发酵中,重组菌最大细胞干重、糖耗和丁二酸产量比对照菌分别提高了15.6%、22%和23%,表明引入IrrE蛋白可提高菌株对高渗透压胁迫的耐受能力。进一步比较重组菌和对照菌胞内相容性物质海藻糖和甘油的浓度后发现,重组菌胞内海藻糖和甘油浓度明显提高,其最大积累量分别是对照菌的1.3和3.8倍,推测IrrE可通过增加胞内相容性物质的积累提高菌株对高渗透压胁迫的耐受性。  相似文献

活性氧胁迫促进枯草芽孢杆菌WSHDZ-01过量合成过氧化氢酶   总被引:2,自引:0,他引:2  
研究了乙醇和H2O2胁迫对芽孢杆菌(Bacillussp.)WSHDZ-01过量合成过氧化氢酶(Catalase,简称CAT)的影响。在Bacillussp.WSHDZ-01培养体系中添加2.0%(V/V)乙醇,胞内过氧化氢酶酶活达到11151U/mL,是对照组的2.5倍。而在培养体系中添加0.3%(V/V)H2O2,则使胞内过氧化氢酶不断分泌到胞外,胞外酶占总酶活比率增加至27%。基于上述发现,分别以不添加胁迫物(a)、乙醇胁迫(b)、H2O2胁迫(c)为对照,在培养体系中维持持续的乙醇和H2O2胁迫,结果表明:1)胞外酶比例达到82.5%;2)发酵周期缩短为42h,比对照a延长了6h,比对照b和c分别缩短了8h和6h;3)生产强度达到470U/(mL·h),比对照a提高了18.6%,为过氧化氢酶工业化生产奠定了基础。  相似文献

【目的】研究酿酒酵母(Saccharomycesc erevisiae)中乙酰辅酶A合成酶基因ACS1和ACS2的生理作用。【方法】将来源于S.cerevisiae的ACS1和ACS2分别进行过量表达,研究过量表达ACS1和ACS2后S.cerevisiae胞内乙酰辅酶A含量、ATP水平、甲羟戊酸途径转录和乙醇耐受性等生理学特性变化。【结果】与出发菌株相比,过量表达ACS1和ACS2使得:(1)胞内乙酰辅酶A含量提高了2.19倍(ACS1)和5.02倍(ACS2);(2)胞内ATP含量提高了3.93倍(ACS1)和2.05倍(ACS2);(3)甲羟戊酸途径8个关键基因表达量显著上调;(4)S.cerevisiae对乙醇胁迫抵御能力显著增强。过量表达ACS1对乙醇胁迫的耐受能力强于过量表达ACS2。【结论】增加胞内乙酰辅酶A的含量可以显著增加甲羟戊酸途径碳代谢流量,并增强S.cerevisiae对发酵过程主要副产物乙醇的耐受能力。  相似文献

旨在构建优良的高温耐受酿酒酵母菌株,并探究其高温耐受机制。通过CRISPR/Cas9技术在絮凝性工业酿酒酵母KF-7中敲除ASP3(编码L-天冬酰胺酶II)并进一步高表达CRZ1(编码具有锌指结构的转录因子Crz1p),通过比较转录组解析重组菌株的高温耐受机制。结果显示,在44℃高温条件下,ASP3敲除菌株KAS11利用98.36 g/L葡萄糖产生43.68 g/L乙醇。在KAS11基础上高表达CRZ1后,菌株KASCR7发酵105.37 g/L葡萄糖产48.02 g/L乙醇。与KF-7相比,两个重组菌株的乙醇产量分别提升了4.77%和15.18%。比较转录组分析结果表明,在高温胁迫下,重组菌株的核糖体生物合成及翻译相关基因受到抑制,而热休克蛋白基因以及NAD+、NADH、嘌呤、甘油、脯氨酸等合成相关基因受到诱导,这些响应可能共同导致重组菌株的高温耐受性提升。研究结果可为构建高温耐受酿酒酵母菌株提供优良菌株资源和理论基础。  相似文献

为了研究Mbp1基因对工业酿酒酵母(Saccharomyces cerevisiae)耐受性的影响,在敲除野生型工业酿酒酵母MF1015菌株Mbp1基因的基础上,分析Mbp1基因缺失菌株(突变菌株)与野生型在乙醇耐受性、耐热性、细胞壁完整性、呼吸强度、海藻糖含量、过氧化氢酶(CAT)、过氧化物酶(POD)、超氧化物歧化酶(SOD)、乙醇脱氢酶(ADH)活性等方面的差异。结果表明:在含刚果红或SDS的平板上突变菌株的菌落比野生型小、菌体存活率比野生型低;突变菌株的乙醇耐受性和耐热性均下降,在乙醇浓度为9%的液体培养基中,突变菌株的最大OD600约为3.15,而野生型最大OD600值约为3.48;在30℃和37℃时突变菌株和野生型的生长趋势基本一致,而在40℃时,突变菌株最大OD600约为3.0,而野生型约为4.5;用9%乙醇处理后,突变菌株胞内海藻糖含量、过氧化氢酶、过氧化物酶、超氧化物歧化酶、乙醇脱氢酶酶活分别比野生型低49%、80%、24%、37%、73%,而未经乙醇处理的突变菌株和野生型酶活无明显差别。推测Mbp1基因有助于工业酿酒酵母适应外部不良环境。  相似文献

以亚硫酸盐甘蔗渣浆酶解液作为原料,利用C. shehatae发酵制取燃料乙醇。结果表明:还原糖最适初始质量浓度为葡萄糖140 g/L、木糖60 g/L、酶解液总糖80 g/L。利用初始葡萄糖55.06 g/L、木糖11.18 g/L、纤维二糖4.51 g/L的亚硫酸盐甘蔗渣浆酶解液发酵,经18 h获得乙醇22.98 g/L。乙醇得率为67.23%,葡萄糖利用率为99.27%,木糖利用率为32.96%,C. shehatae适合作为蔗渣为原料的乙醇发酵菌株。  相似文献

代谢工程与全基因组重组构建酿酒酵母抗逆高产乙醇菌株   总被引:1,自引:0,他引:1  
将酿酒酵母海藻糖代谢工程与全基因组重组技术相结合,改良工业酿酒酵母菌株的抗逆性和乙醇发酵性能。对来源于二倍体出发菌株Zd4的两株优良单倍体Z1和Z2菌株进行杂交获得基因组重组菌株Z12,并对Z1和Z2先进行(1)过表达海藻糖-6-磷酸合成酶基因 (TPS1) ,(2)敲除海藻糖水解酶基因 (ATH1), (3)同时过表达 TPS1和敲除ATH1, 经此三种基因工程操作后再进行杂交获得代谢工程菌株的全基因组重组菌株Z12ptps1、Z12 Δath1和Z12pTΔA。与亲株Zd4相比,Z12及结合代谢工程获得的菌株在高糖、高乙醇浓度与高温条件下生长与乙醇发酵性能都有不同程度的改进。对比研究结果表明:在高糖发酵条件下,同时过表达 TPS1和敲除ATH1 的双基因操作工程菌株胞内海藻糖积累、乙醇主发酵速率和乙醇产量相对于亲株的提高幅度要大于只过表达 TPS1,或敲除ATH1 的工程菌。结合了全基因组重组后获得的二倍体工程菌株Z12pTΔA,与原始出发菌株Zd4及重组子Z12相比,主发酵速率分别提高11.4%和6.3%,乙醇产量提高7.0%和4.1%,与其胞内海藻糖含量高于其它菌株、在胁迫条件下具有更强耐逆境能力相一致。结果证明,海藻糖代谢工程与杂交介导的全基因组重组相结合,是提高酿酒酵母抗逆生长与乙醇发酵性能的有效策略与技术途径。  相似文献

【目的】探究外源添加不同氨基酸和相容性溶质对谷氨酸棒杆菌(Corynebacterium glutamicum)在高糖胁迫环境下生长的影响及可能的作用机理。【方法】通过在培养基中外源添加各种氨基酸和相容性溶质,研究其对谷氨酸棒杆菌在高葡萄糖和高蔗糖胁迫下生长的影响,并分析添加精氨酸对高葡萄糖胁迫下菌株糖转运和代谢途径中关键酶转录水平的影响,以及对菌株发酵产氨基酸的影响。进一步探究了碱性氨基酸在其它棒状杆菌属中抵御高葡萄糖胁迫的潜在作用。【结果】在高葡萄糖胁迫条件下,外源添加赖氨酸、精氨酸和组氨酸后谷氨酸棒杆菌的生物量分别提高54.7%、50.0%和37.6%;而在高蔗糖胁迫条件下,添加脯氨酸和四氢嘧啶后菌株生物量增加20%以上。进一步研究表明,在高葡萄糖胁迫下,外源添加精氨酸后谷氨酸棒杆菌的葡萄糖利用速率提高约2.5倍,谷氨酸的发酵产量也增加了127.5%。此外,碱性氨基酸对其它4种棒状杆菌也具有一定的渗透保护效应。【结论】精氨酸对谷氨酸棒杆菌在高葡萄糖胁迫下具有良好的渗透保护作用,可能归因于其能促进葡萄糖的转运和代谢能力,同时发现碱性氨基酸的渗透保护效应对棒状杆菌属具有一定的普遍性。  相似文献

燃料乙醇发酵过程中酿酒酵母细胞活性被高浓度乙醇严重抑制而导致发酵提前终止,生产强度严重降低,因此构建同时具有高耐受性、高发酵性能的菌株一直是发酵工业追求的目标。选取酿酒酵母细胞形态调节关键基因小GTP酶家族成员Rho1,构建易错PCR产物文库,以酿酒酵母S288c为出发菌株采取“富集-自然生长-复筛”的筛选策略,成功筛选得到两株乙醇胁迫耐受性与发酵性能均提高的突变株M2和M5。测序发现突变株过表达的Rho1序列出现了3~5个氨基酸的突变和大片段的缺失突变。以300 g/L起始葡萄糖进行乙醇发酵,72 h时,M2和M5的乙醇滴度比对照菌株分别提高了19.4%和22.3%,超高浓度乙醇发酵能力显著提高。本研究为利用蛋白定向进化方法改良酵母菌复杂表型提供了新的作用靶点。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. We also discuss the development and practicality of various cell culture and animal model systems to study KSHV replication and pathogenesis.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
