By the spectrophotometric method the age changes of total iodine content, its hormonal and non-hormonal compounds in rats and men tissues (muscles, liver, kidney, heart, lungs, brain, spleen and blood) have been studied. It was established that the total iodine content decreased at the expense of the protein-bound iodine concentration and also of the butyl-alcohol extractable iodine. The content of total iodine, its hormonal and nonhormonal fractions as well the level of protein-bound iodine in blood and basic tissue targets in representatives of 4 classes of animals: Esox lucius L., Rana esculenta, Streptopelia decaocto Priv., Lepus europaeus Pall. inhabiting the mountain regions with iodine deficiency in environment and in the lowlands of Transcarpathia with higher iodine provision have been investigated. A considerable decrease of general and hormone iodine level in the animal tissues of the mountain area accompanied by the suppression of the thyroid function has been stated. The utilization of thyroid hormones under the iodine deficiency condition is increased in the majority of cases and the level of protein-bound iodine is lowered that testifies to the transition of animal organism in the iodine-deficient areas to the lower level of thyroid homeostasis.  相似文献

Experiments on male rats maintained at the diet with a low amount of cobalt have revealed a decrease in values of iodine indices in organs and tissues except for the liver tissues where a sharp increase in the inorganic iodine concentration as well as disturbances of the protein and carbohydrate metabolism were observed. Mathematical analysis has shown a high correlation of changes in the carbohydrate and protein metabolism and concentration of protein-bound iodine in blood. It has permitted a conclusion on an indirect effect of the cobalt deficiency on the metabolism processes as a result of changes in the concentration of protein-bound iodine in blood and tissues.  相似文献

We have characterized the thyroid iodoprotein of a hagfish, Eptatretus burgeri, one of the lowest marine vertebrates. The iodoprotein was not very homogeneous in its apparent molecular mass which decreased with the increase in hormone/iodotyrosine ratio. Four subfractions with an apparent molecular mass of about 400 kDa were purified from one major fraction by size-exclusion and Mono Q ion-exchange HPLC. The subfractions appeared to have the same peptide backbone, since they showed a single band with the same mobility as a 160-kDa protein in SDS/PAGE and the same amino acid composition. However they differed from each other in having increasing iodine contents (1.9% to 5.9% by mass of total amino acids) associated with the increase in hormonal iodine proportion (8.4% to 16.7% of total iodine) and carbohydrate content (35.6% to 53.5% by mass). These values are strikingly different from those of thyroglobulin with an iodine content of less than 1%, hormonal iodine of 20-40% and carbohydrate content of less than 10%. The amino acid composition of the hagfish iodoprotein, especially the cysteine content of less than 1%, was also entirely different from that of thyroglobulin. These results suggest that most, if not all, tyrosine residues of the hagfish thyroid glycoprotein with a less rigid structure are susceptible to an iodinating system, but hormone residues are formed by a much less efficient mechanism than those in thyroglobulin, when poorly iodinated.  相似文献

Using 131I as tracer, iodine uptake by various thyroid and non-thyroid tissues has been reported in Channa punctatus in which vitamin C deficiency was experimentally induced by feeding vitamin C deficient diet to a level when morphological deficiency syndromes appeared. This was compared with a parallel control, fed a complete diet. The study showed a highly significant decrease in iodine uptake by thyroid tissues in vitamin C deficient fish as compared to the controls. However, the blood and other non-thyroid tissues recorded a significant increase in absorption and distribution of iodine. These findings suggested hypofunctioning of thyroid gland due to a prolonged deficiency of vitamin C.  相似文献

Sodium hypochlorite (NaOCl) and sodium dichloroisocyanurate (NaDCC) were more active against Bacillus subtilis 8236 spores in both viability and in germination and outgrowth studies than were polyvinylpyrrolidone-iodine (PVP-I) and Lugol's solution. Of the two chlorine compounds studied NaOCl proved to be the more active. The two iodine-containing compounds gave contrasting results with the Lugol's solution demonstrating increased antibacterial activity with increasing available iodine concentration. The antibacterial behaviour of PVP-I, however, reflected the more complex nature of aqueous iodine-surfactant mixtures. Initially, non-complexed iodine concentration (the active species) increased with increasing total available iodine concentration, resulting in increasing antibacterial activity. However, due to changes in the physical properties of the mixture, a maximum concentration of non-complexed iodine was reached so that a further increase in total available iodine resulted in a decrease in non-complexed iodine concentration and consequently a decrease in the antibacterial activity of the solution was observed. A greater inhibitory effect was observed in subsequent germination and outgrowth studies when spores were pre-treated with respective biocide than when untreated spores were added to germination media containing biocide at t = 0. This may reflect a combination of different contact times plus the neutralizing effect of the germination media on such halogen compounds.  相似文献

Thyroid lysosomes have been studied by means of EPMA combined with an electron microscope. In 4 months old mice, lysosomes are a few number and contain very low iodine concentrations and sulfur load is not significant. In 10 months old mice, iodine only or associated with sulfur is observed in homogeneous lysosomes varying in size and density. Those two types of localizations reflect two eventual steps in hormonal metabolism pathway of iodine.  相似文献

The distribution and chemical species of iodine in various subcellular fractions of human liver were studied by using epithermal neutron activation analysis combined with chemical and biochemical separation techniques, such as gradient centrifugation and gel chromatography. It was found that the total iodine content orders in various subcellular fractions is as follows: nuclei > cytosol > mitochondria > lysosome > microsome. In the lysosomal fraction, iodine is mainly bound to macromolecules, whereas in the nuclei and mitochondrial fractions, mainly with lower-molecular-weight organic compounds. In the cytosol fraction, iodine is combined with three proteins, in which iodine is chiefly bound with mid- and high-molecular-weight proteins.  相似文献

Changes are described in the content of butanol-extracted iodine in the blood serum and total iodine in thyroid tissue of white rat males under ultrasonic effect of various intensity (0.2, 0.6, 1.0 Watt/cm2) and analysed at different time intervals after ultrasound application. A decrease in total iodine content in thyroid glands (at initial level 0.315 mg%) and an increase of butanol-extracted iodine content in the blood serum were observed in all the experimental groups. The data obtained compared with the earlier found increase of the thyroid hormone content in the serum and thyreotropic hormone at a decreased content of thyroxins in the thyroid gland seem to point to the activation of the thyroid function after the application of ultrasound.  相似文献

In the presented study the effect of different iodine (I) levels and sources in hen feed on the iodine concentration of different tissues, blood serum, and eggs of laying hens was studied. For this purpose, two experiments were conducted with 30 laying hens each. In these experiments feed was enriched with KI and Ca(IO(3))(2), respectively, at 0 (Control), 0.25, 0.5, 2.5 and 5 mg I/kg feed, resulting a analysed iodine level from 0.44 to 4.20 mg/kg feed. After four weeks experimental feeding the iodine concentrations of thyroid glands, blood, meat, liver, abdominal fat and eggs were measured with inductively coupled plasma mass spectrometry. The experimental treatment did not affect hen performance. The iodine supplementation significantly increased the iodine concentration of eggs (144-1304?μg/kg), thyroid glands (3367-5975?μg/g), blood serum (16-67?μg/kg) and liver (13-43?μg/kg). Meat (about 14?μg I/kg) and abdominal fat (about 12?μg?I/kg) were not significantly affected by iodine treatment. Comparative regression analyses showed that at a similar iodine intake, the supply via KI resulted in significantly higher iodine deposition into eggs than Ca(IO(3))(2). Due to the high carry-over of iodine into eggs, eggs may considerably contribute to the iodine supply of the consumers.  相似文献

Environmental iodine deficiency continues to be a significant public health problem worldwide. On the other hand, iodide excess results principally from the use of iodine-containing medicinal preparations or radiographic contrast media. For this reason we intended to explore iodide excess impairment on prooxidant/antioxidant balance of the thyroid gland, hepatic tissue and in blood and the effect of selenium administration on oxidative stress markers under the same circumstances. Experiments were performed for 10 days with white, male, Wistar rats, as follows: group 1: control-normal iodine supply group; 2: high iodine diet, group; 3: high iodine diet and selenium; group 4: high iodine diet and Carbimasole. Oxidative stress markers such as lipid peroxides were determined in thyroid gland, hepatic tissue and in blood. Measuring H+ donor ability of the sera and catalase activity in thyroid gland and in hepatic tissue assessed antioxidant defense. Iodide excess had prooxidant effects, leading to an increased lipid peroxides level and catalase activity in target tissues and in blood and to a decreased H+ donor ability of the sera. Selenium supplementation had opposite effects. Present data allow us to conclude that the alterations due to iodide excess in thyroid gland, hepatic tissue and in blood are mediated through oxidative stress.  相似文献

Iodine is a key ingredient in the synthesis of thyroid hormones and also a major factor in the regulation of thyroid function. A local reduction of iodine content in follicular lumen leads to overexpression of local thyroid-stimulating hormone receptor (TSHr), which in turn excessively stimulates the regional thyroid tissue, and result in the formation of nodular goiter. In this study, we investigated the relationship between iodine content and sodium iodide symporter (NIS) expression by using the clinical specimens from patients with nodular goiter and explored the pathogenesis triggered by iodine deficiency in nodular goiter. In total, 28 patients were clinically histopathologically confirmed to have nodular goiter and the corresponding adjacent normal thyroid specimens were harvested simultaneously. Western blot and immunohistochemistry were performed to assay NIS expression and localization in thyrocytes of both nodular goiter and adjacent normal thyroid tissues. NIS expression mediated by iodine in follicular lumen was confirmed by follicular model in vitro. Meanwhile, radioscan with iodine-131were conducted on both nodular goiter and adjacent normal thyroid. Our data showed that NIS expression in nodular goiter was significantly higher than that in adjacent normal tissues, which was associated with low iodine in the follicular lumen. Abnormal localization of NIS and lower amount of radioactive iodine-131 were also found in nodular goiter. Our data implied that low iodine in the follicular lumen caused by cytoplasm mis-localization of NIS may induce nodular goiter.  相似文献

Active and total phosphorylase activity, using labelled C14-glucose-1-phosphate as the substrate, is demonstrated by histoautoradiographic method. This method can demonstrate the polysaccharide synthesizedin vitro by phosphorylase without intervention from the unlabelled pre-existing glycogen. C14-glucose can not replace C14-glucose-1-phosphate as substrate. The distribution of phosphorylase in tissue sections, except in cases of very low activity, is similar to that obtained by customary dilute Lugol's iodine staining method. The relative difference of intensity between active and total phosphorylase, as revealed by iodine staining, is also reflected by histoautoradiographic method. Histoautoradiographic method has several advantages over the iodine staining method. This method is more sensitive for demonstration of very low phosphorylase activity which may escape detection by iodine staining. Branching enzyme activity, especially when it favors synthesis of glycogen type of polysaccharide instead of amylopectin type, can be better detected by this method. Active phosphorylase substrate medium can be used to demonstrate this activity in plant tissues, where the presence of pre-existing starch often prohibits the use of iodine staining method. Stripping film method for autoradiography is recommended for the study of this enzyme activity.  相似文献

A greenhouse pot experiment was carried out to investigate the availability of iodide and iodate to soil-grown spinach (Spinacia oleracea L.) in relation to total iodine concentration in soil solution. Four iodine concentrations (0, 0.5, 1, 2 mg kg−1) for iodide (I) and iodate (IO3) were used. Results showed that the biomass productions of spinach were not significantly affected by the addition of iodate and iodide to the soil, and that iodine concentrations in spinach plants on the basis of fresh weights increased with increasing addition of iodine. Iodine concentrations in tissues were much greater for plants grown with iodate than with iodide. In contrast to the iodide treatments, in iodate treatment leaves accounted for a larger fraction of the total plant iodine. The soil-to-leaf transfer factors (TFleaf) for plants grown with iodate were about tenfold higher than those grown with iodide. Iodine concentrations in soil solution increased with increasing iodine additions to the soil irrespective of iodine species. However, total iodine in soil solution was generally higher for iodate treatments than iodide both in pots with and without spinach. According to these results, iodate can be considered as potential iodine fertilizer to increase iodine content in vegetables.  相似文献

It known an oppressing action of radiation, including radioactive iodine isotopes on the reproductive system and fetus development. There are clinical data on a negative influence of iodine deficiency on the course of pregnancy and fetus development resulting from hormonal disfunction of thyroid gland and a mother-fetus system. There are no data about a character and mechanisms of interaction of radiation and iodine endemia at the combined action on the gonads and fetus. The urgency of this problem is caused by the fact that many regions of the country are characterized to some extent by iodine deficiency in local food and water (in Russian Federation such regions make approximately 50% of territories), and the opportunity of radiation accidents at nuclear plants with contamination of the environment with products of nuclear division (significant part of which is radioactive iodine isotopes is an objective reality. The analysis of a few published and own experimental data allows us to conclude that the combined influence of an external gamma-irradiation and iodine deficiency on reproductive function has a synergic character.  相似文献

An adequate intake of iodine in the diet of pregnant women is important to ensure normal growth and development of the fetus. It is difficult, however, to accurately determine iodine intakes using traditional methods of dietary assessment, primarily because the contribution of iodised salt use, at the table and in cooking, to total iodine intake is difficult to quantify. Given the limitations of dietary assessment, biochemical indices in blood and urine are typically used to assess iodine status in children and adults. Although reference ranges exist for thyroid hormones, there has been no consensus on the cut-offs needed in pregnancy to diagnose iodine deficiency. In contrast, a median urinary iodine concentration (UIC) of 150-249 μg/L has been established to determine the adequate iodine status of a group of pregnant women. However, the large intra-individual variation in UIC from either spot or 24h urine samples means that UIC cannot be used to assess iodine status in an individual pregnant woman. The difficulty in determining if an individual pregnant woman is iodine deficient is problematic for cross-sectional studies examining associations between iodine status in pregnancy and developmental outcomes in the child.  相似文献

This report describes normal histology of tissues from immature female dogs and sequential microscopic changes that occur during different stages of the estrous cycle in ovary, uterus, vagina and mammary glands. These observations are associated with a literature review of concurrent estrous cycle plasma hormone levels and expression of sex steroid receptors in these tissues. The combined review of different aspects of the estrous cycle in the dog provides readers with a comprehensive presentation and may guide investigators that are involved in testing compounds with hormonal effects in female dogs.  相似文献
