Mosaicism in silkworms is easily inducible by applying a cold shock to the hybrid eggs between two strains with different markers at the early developmental stage. When BF1 eggs from the cross between a tetraploid female (which had been induced from the cross between a red egg and a white egg strain) and a male of the red egg strain were exposed to a cold shock, hexaploid eggs and two types of mosaic eggs were obtained: black-black (BB-) mosaic eggs, which possessed two kinds of black serosa cells with different-sized nuclei, and black-red (BR-) mosaic eggs. Cytological studies on the embryos from these mosaic eggs revealed that the BB-mosaic type was a 3n/6n mixoploid, while the BR-mosaic type was mixoploid with n/2n/3n, n/3n, or 2n/6n constitution. The hatchability of BB- and BR-mosaic eggs was considerably lower than that of either n/2n or 2n/4n mixoploid eggs obtained by a cold treatment of diploid eggs. Since abnormal development was observed at a high frequency in the embryos from BB- and BR-mosaic eggs at the blastokinesis stage, it is speculated that the coexistence of hexaploid or triploid cells may be a factor in the induction of such abnormality in early embryogenesis.  相似文献

Tetraploid and hexaploid silkworms, Bombyx mori L. (Lepidoptera Bombycidae) were induced by applying a cold shock to diploid and triploid eggs at the first cleavage stage. Male meiosis in the primary spermatocytes of these silkworms having a different ploidy was observed. In the polyploid cells, chromosome bridges, 2 for the tetraploid and 3 for the hexaploid, occurred between the newly formed daughter nuclei at telophase. Observation in the living spermatocytes showed that tetraploid cells needed a longer time than the diploid ones to complete the first meiotic division. The delay in the cell division may be responsible for the high sterility of the tetraploid males.  相似文献

Summary Wild-type nuclei from eggs ofDrosophila melanogaster at various developmental stages and from different regions of the egg—cleavage nuclei, pole nuclei from preblastoderm, and lateral nuclei from syncytial blastoderm—were singly implanted into unfertilizedy w sn 3 lz 50e eggs to determine their developmental potencies.All three types of transplanted nuclei were almost equally effective in initiating development of unfertilized eggs. Development was arrested in one of five critieal embryonic stages or in one of the three larval instars. The frequency of individuals reaching a distinct stage was approximately the same for all three types of donor nuclei. The stage-specific pattern of defects was independent of the type of nucleus transplanted.The deviations from normal development were broadly similar to those seen in controls developing from fertilized eggs which had only been punctured or into which cytoplasm had been injected. Many defective embryos also occurred in these control experiments. These and other observations indicate that a large proportion of irregularly developed individuals found after nuclear transfer can be ascribed to loss of egg material, disturbances in the internal organization of the egg during nuclear implantation, and the difficulty the implanted nucleus has in adjusting to the autonomous processes within the egg, such as the formation and migration of cytoplasmic islands.Some of the defective embryos and larvae originating from nuclear transfer were implanted into adult hosts. After culture for 14 days the early embryonic stages had formed several larval structures, and the late embryonic and larval stages had developed all larval organs. The proliferated imaginal primordia of thesein vivo cultured embryos and larvae, as well as the imaginal disks of the third instar larva, were then implanted into larval hosts with which they passed through metamorphosis and differentiated into imaginal structures. All three types of donor nuclei were capable of producing all adult structures derivedin situ from imaginal disks. The phenotype of these structures waswild-type, thus demonstrating their origin from the transplanted nuclei.The problem as to why not all transplanted nuclei initiated development, and why development after nuclear transplantation was arrested at the third larval instar, at the latest, is discussed.This article is dedicated to Professor Friedrich Seidel on the occasion of his 75th birthday.  相似文献

Nuclear transplants of loach were produced by transplantating blastula cell nuclei into nonenucleated unfertilized eggs, using recipient eggs and donor cells distinguished by different polymorphic microsatellites. Of the total of 2,847 operated eggs, 143 hatched and 119 developed to the feeding larval stage. For 15 nuclear transplants (i.e., 11.1-year-old fish and 4.2-year-old fish) that survived up to the adulthood, DNA analysis and karyotype analysis were performed. Results showed that, of the 15 fish, 14 had only a nucleus derived from the donor; moreover, 12 were diploids, 1 was a triploid, 1 was a tetraploid, and 1 was a diploid-tetraploid mosaic with both donor and recipient nuclei. For the 12 fish with only a 2n donor nucleus, morphometric analysis was performed, and two female fish and two male fish were mated with normal fish. The fish with only a 2n donor nucleus were determined to be morphometrically identical to normal fish: they had normal gametogenesis and were able to reproduce. Currently, nuclear transplantation technology is beginning to be adopted in fisheries. Biological information on nuclear transplants obtained in this study can be used in the development of nuclear transplantation technology.  相似文献

Fischberg's method, thanks to which tetraploid individuals can be obtained, has been adapted to the newt Pleurodeles waltlii. — If at the beginning of the first furrow formation, the eggs are submitted for 10 minutes to a sudden increase in temperature (37.2–37.3°C), the embryos that subsequently develop are tetraploid in 100% of the cases. The percentage obtained of viable individuals is about 30% of the number of heated eggs. — A temperature treatment between 36.2 and 36.4°C applied at the same period and for the same length of time may lead to the formation of viable animals diplo-tetraploid mosaic. — Among adult tetraploids or diplo-tetraploid mosaic males and females can be found. — The cells of tetraploids can easily be distinguished from those of diploids by their larger size. Therefore, the former can be used as cellular markers.  相似文献

Summary Quantitative variation in seven morphological characteristics (leaf length and width, leaf length/ width ratio, flower, petal and stomata length, and number of chloroplasts in guard cells) were studied in Petunia hybrida plants regenerated from anther tissue culture and belonging to four different classes of ploidy (2n, 2n–3n, 3n–2n, 4n–8n). Results showed that leaf size is not a good characteristic for discriminating between plants of different ploidy — flower and stomata characteristics being more adequate for this purpose. After applying stepwise discriminant analysis the association chloroplast number — leaf length/width ratio — petal length was verified to be more appropriate for the discrimination of ploidy classes.  相似文献

Summary The ability of the highly condensed chromatin of small thymocyte nuclei and the more loosely organized chromatin of hepatocyte nuclei to interact with nine DNA-specific fluorochromes was assessed by microfluorometry. Although the results obtained with five of the fluorochromes—mithramycin, 7-aminoactinomycin d, Hoechst 33258, DAPI, and propidium iodide—were found to be virtually unaffected by differences in the degree of condensation of the chromatin, the values obtained with the remaining fluorochromes—proflavine, quinacrine mustard, berberine sulfate, and pyronin Y—appeared to be affected significantly by organizational differences of the chromatin. All of the latter structural probes, except quinacrine mustard, produced fluorescence values which were higher in the 2c nuclei of hepatocytes than in the nuclei of small thymocytes. Quinacrine mustard yielded higher values in thymocyte nuclei; and in the hepatocyte polyploid series (2, 4, and 8c), it did not produce the expected multiples of the 2c value. Pretreatment of the two types of nuclei with RNase affected their total fluorescence in unpredictable ways. While RNase extraction lessened the differences between thymocyte and 2c hepatocyte nuclei stained with propidium iodide, Hoechst 33258, proflavine, and berberine sulfate, it increased the differences between nuclei stained with mithramycin, quinacrine mustard, pyronin Y, and 7-aminoactinomycin d. The ability of RNA-depleted chromatin to interact with various types of fluorochromes might be a useful parameter in subsequent studies of chromatin organization.  相似文献

Summary Microfluorometric estimates of total RNA were made in selected test material stained with berberine sulfate, acridine orange, and Hoechst 33258. These measurements were compared with those obtained with propidium iodide, which is known to interact only with double-stranded nucleic acids. It was observed that all of the fluorochromes, including propidium iodide, yielded very similar patterns of fluorescence in the various types of material tested. In isolated thymocyte and hepatocyte nuclei stained with either propidium iodide or Hoechst 33258 at pH 2, it was evident that RNA could be estimated only indirectly by measuring the amount of fluorescence before and after extraction with RNase. It was apparent that the total fluorescence of small thymocyte nuclei was affected much less by RNase extraction than that of 2c hepatocyte nuclei. Attempts to obtain direct estimates of RNA by exposing the preparations to DNase were not successful: the fluorescence of thymocyte nuclei dropped almost to zero, and hepatocyte nuclei could no longer be assigned to distinct ploidy classes. In addition, since the highly condensed chromatin of thymocyte nuclei was stained much more prominently than the looser chromatin of hepatocyte nuclei with Hoechst 33258, it was apparent that this fluorochrome — when used at pH 2 — has potential usefulness as a probe of organizational differences in chromatin.  相似文献

The bivoltine silkworm Bombyx mori (Lepidoptera: Bombycidae) exhibits a maternally controlled embryonic diapause. Maternal silkworms decide whether to lay diapause or nondiapause eggs depending on environmental factors such as the temperature and photoperiod during the egg and larval stages, and then induce diapause eggs during the pupal stage. However, little is known about the molecular mechanism that conveys the outcome of whether to produce diapause or nondiapause eggs from the egg or larval stages to the pupal stage. This study used microarray analysis to investigate differentially expressed genes in the larval brains of diapause‐ and nondiapause‐egg producers, to which bivoltine silkworms were destined by thermal or photic stimulation during the egg stage. The cytochrome P450 18a1 and Krüppel homolog 1 genes were upregulated in producers of diapause eggs compared with those of nondiapause eggs under both experimental conditions. Cytochrome P450 18a1 encodes a key enzyme for steroid hormone inactivation and Krüppel homolog 1 is an early juvenile hormone‐inducible gene that mediates the repression of metamorphosis. The upregulation of these genes during the larval stage might be involved in the signaling pathway that transmits information about the diapause program from the egg stage to the pupal stage in the silkworm.  相似文献

Honey bees (Apis mellifera) exhibit age polyethism, whereby female workers assume increasingly complex colony tasks as they age. While changes in DNA methylation accompany age polyethism, other DNA modifications accompanying age polyethism are less known. Changes in endopolyploidy (DNA amplification in the absence of cell division) with increased larval age are typical in many insect cells and are essential in adults for creating larger cells, more copies of essential loci, or greater storage capacity in secretory cells. However, changes in endopolyploidy with increased adult worker age and polyethism are unstudied. In this study, we examined endopolyploidy in honey bee workers ranging in age from newly emerged up to 55 days old. We found a nonsignificant increase in ploidy levels with age (P < 0.1) in the most highly endopolyploid secretory cells, the Malpighian tubules. All other cell types decreased ploidy levels with age. Endopolyploidy decreased the least amount (nonsignificant) in neural (brain) cells and the stinger (P < 0.1). There was a significant reduction of endopolyploidy with age in leg (P < 0.05) and thoracic (P < 0.001) muscles. Ploidy in thoracic muscle dropped from an average of 0.5 rounds of replication in newly emerged workers to essentially no rounds of replication (0.125) in the oldest workers. Ploidy reduction in flight muscle cells is likely due to the production of G1 (2C) nuclei by amitotic division in the multinucleate striated flight muscles that are essential to foragers, the oldest workers. We suggest that ploidy is constrained by the shape, size and makeup of the multinucleate striated muscle cells. Furthermore, the presence of multiple 2C nuclei might be optimal for cell function, while higher ploidy levels might be a dead-end strategy of some aging adult tissues, likely used to increase cell size and storage capacity in secretory cells.  相似文献

For the functional analysis of insect genes as well as for the production of recombinant proteins for biomedical use, clonal transgenic silkworms are very useful. We examined if they could be produced in the parthenogenetic strain that had been maintained for more than 40 years as a female line in which embryogenesis is induced with nearly 100% efficiency by a heat shock treatment of unfertilized eggs. All individuals have identical female genotype. Silkworm transgenesis requires injection of the DNA constructs into the non-diapausing eggs at the preblastodermal stage of embryogenesis. Since our parthenogenetic silkworms produce diapausing eggs, diapause programing was eliminated by incubating ovaries of the parthenogenetic strain in standard male larvae. Chorionated eggs were dissected from the implants, activated by the heat shock treatment and injected with the transgene construct. Several transgenic individuals occurred in the daughter generation. Southern blotting analysis of two randomly chosen transgenic lines VTG1 and VTG14 revealed multiple transgene insertions. Insertions found in the parental females were transferred to the next generation without any changes in their sites and copy numbers, suggesting that transgenic silkworms can be maintained as clonal strains with homozygous transgenes. Cryopreservation was developed for the storage of precious genotypes. As shown for the VTG1 and VTG14 lines, larval ovaries can be stored in DMSO at the temperature of liquid nitrogen, transferred to Grace’s medium during defrosting, and then implanted into larvae of either sex of the standard silkworm strains C146 and w1-pnd. Chorionated eggs, which developed in the implants, were dissected and activated by the heat shock to obtain females (nearly 100% efficiency) or by a cold shock to induce development to both sexes in 4% of the eggs. It was then possible to establish bisexual lines homozygous for the transgene.  相似文献

Summary The occurrence in triticale of four different genes causing hybrid necrosis is described: Ne1 and Ne2 in the B genome of wheat and Ner1 and Ner2 in the rye genome. Hybrid necrosis develops due to dominant complementary interaction of two genes. This interaction in triticale, however, may take place not only between genes belonging to the same genome but also between genes of different genomes. In triticale, these genes can cause hybrid necrosis in four different combinations. The inheritance of the phenomenon in triticale is, therefore, more complicated than it is in wheat or rye. To avoid hybrid necrosis in triticale, attention should be paid that no necrosis genes are introduced into the primary triticale stocks from the wheat and rye parents. The expression of necrosis genes is influenced by the level of ploidy. Any additional genome — A, B, D, or R — may exert a suppressing effect on the expression of necrosis genes. Therefore, when identifying genotypes of triticale with regard to their necrosis genes, the level of ploidy has to be accounted for. Moreover, the present results illustrate that gene expression in polyploids is not only determined by interactions with other single genes but that it may also be modified by the total genotype of the respective individual.  相似文献

Polyspermy occurs frequently in the fertilization of mammalian eggs, but little is known about whether polyspermic eggs have developmental ability in vitro or in vivo. We previously reported that poly-pronuclear (PPN; 3 or more pronuclei) pig eggs developed normally to the blastocyst stage despite having fewer inner cell mass cell numbers as compared to blastocysts derived from two-pronuclear (2PN) eggs. Here it is shown that most PPN pig eggs have abnormal cleavage patterns (having 3 or more cells) in the first cell division and retarded development of pronuclei prior to syngamy as compared to 2PN eggs. Most blastocysts (14 of 18) that developed from PPN eggs showed abnormal ploidy (were haploid, triploid, and tetraploid) whereas 20 of 22 blastocysts derived from 2PN embryos were diploid. The size and morphology of most Day 40 fetuses that developed from PPN eggs appeared to be normal. Of 8 Day 40 fetuses analyzed, 1 was triploid (XXY) and another was a mosaic with both diploid (XX) and tetraploid cells (frequency of less than 10%, XXXX), and the others were diploid. Anomalies of chromosomal composition were not detected in these fetuses. Five live piglets and one dead piglet were born from two recipients of PPN eggs. It is proposed that not all pronuclei of PPN pig eggs participate in syngamy, resulting in diploid cells in the conceptus. Our data suggest that there are two types of pronuclei location in polyspermic pig eggs and that the resulting ploidy is determined at the zygote stage before the first cell division according to pronuclear location.  相似文献

The karyotype of Rana temporaria is analysed from preparations of squashed tadpole epidermis, the karyotype of Rana dalmatina from preparations of squashed larval epidermis and adult tissues. For each of these two species, the 26 chromosomes of the karyotype have been divided in two groups according to their length. Inside these two groups, three morphological criteria — relative length, arm-ratio and presence of secondary constriction — permit the identification of each of the component parts. For Rana dalmatina, the analysis of the two meiotic divisions shows that the classification remains possible in two groups. There is no evidence of an heteromorphic pair of chromosomes in these two species. The studies on Rana temporaria and Rana dalmatina are compared to recent ones on other Ranidae.  相似文献

Mosaic cycles in agricultural landscapes of Northwest Europe   总被引:2,自引:0,他引:2  
Mosaic cycles were originally understood as cyclical regeneration phases in forests. In this review, we shall examine how far the concept can be extended towards cyclical mosaics of habitat quality in patterned landscapes as a special case of ‘dynamic landscapes’. We will concentrate on habitats and plants in European temperate agricultural landscapes and grasslands in particular. Mosaic cycles of habitat quality are characterised by spatiotemporal shifts between disturbance and secondary succession. We found evidence for mosaic cycles in traditional agricultural systems, modern crop farming, and in recent conservation management. The relevant disturbance parameters to describe land-use drivers of mosaic cycles are spatial extent, frequency, and magnitude (biomass loss). Land-use-related drivers are usually regular and deterministic in space and time, with the exception of year-round grazing by free-ranging large herbivores. Fluctuating soil resources such as water and nutrients in interaction with climate variability add a stochastic component to these (land-use-related) drivers. The proportion of deterministic and stochastic components and their autocorrelation in time and space divides purely deterministic mosaic cycles from purely stochastic dynamic landscapes. In a second part, we briefly review plant life-history traits that may facilitate survival of plants in mosaic cycles of habitat quality. Theoretical studies emphasise (i) dispersal functions for extinction and recolonisation processes of metapopulations, (ii) storage effects as a component of buffered population growth in response to temporal fluctuations of habitat quality, and (iii) competitive ability in metacommunities. We propose a simple scheme relating these functions to the temporal and spatial correlation of patterned landscapes. There are only a very limited number of field studies available that give some support for the proposed scheme. We provide perspectives for further research in this field.  相似文献

The reproductive biology and nonparasitic development of Boophilus microplus (Canestrini) and hybridized Boophilus ticks (B. annulatus (Say) male x B. microplus female) held under natural field conditions in south Texas throughout the year were compared. Comparisons between the two types of ticks indicated that the ovipositional biology (percentage of ovipositing females and number of eggs laid) of the females favored hybrid ticks during some months and B. microplus ticks during other months. However, on a yearlong basis, there was virtually no difference in the percentage of ovipositing females or in the number of eggs deposited by hybrid females as compared to B. microplus. The duration of each nonparasitic development period (preoviposition period, incubation period of eggs, and larval longevity) showed that both types of ticks had very similar developmental and survival rates during the year. Generally the difference in duration of each of the nonparasitic parameters was <8 days, leading to a high degree of synchrony of the nonparasitic developmental rates between the two types of ticks. On the other hand, egg hatchability of hybrid ticks was consistently lower than pure-strain B. microplus throughout the year with significantly lower hatch rates occuring in April, July, August, October, and November. Thus, results obtained on percentage of ovipositing females, number of eggs laid, preoviposition period, incubation period of eggs, and larval longevity provide positive encouragement for the possible use of sterile hybrid males as a means of eliminating a native B. microplus population. In contrast, results of the egg hatchability of the two types of ticks indicate that the selective advantage afforded toB. microplus could have an adverse affect on the success of a sterile hybrid male program by making the number of hybrid ticks necessary to eliminate a native population prohibitively high.This paper reports the results of research only. The genome of Prodiamesa olivacea (Diptera, Chironomidae) has a 2 C DNA content of 0.25 pg. Mitotic metaphases reveal 3 pairs of chromosomes: 2 metacentric ones and one submetacentric. The latter comprises 20.8% of total Feulgen DNA. During larval polytenization the complemental portion of the 3rd falls to 6.5%. Concomitantly the polytene 3rd chromosome is much shorter than expected. It has no constriction and is shaped like a ball sector. — Underreplication is understood as suppression of DNA syntheses mainly in the long arm of the 3rd chromosome at the first to third endoreplicative cycle. Most of the dense heterochromatin seen in the apex of the 3rd polytene element is not itself underreplicated; it conceals the underreplicated long arm of this chromosome. — In ovarian nurse cells which are closely connected with the germ line the longer heterochromatic arm of the 3rd polyneme chromosome is fully replicated. — Underreplication is discussed in the context of DNA silencing.  相似文献

The oostegites of amphipods attached to peraeopods 2–5 are of two main types — broad with relatively short marginal setae and narrow with long marginal setae. Broad oostegites are found in other peracarids and are considered the primitive type. Amphipods with broad oostegites tend to have smaller eggs than those with narrow oostegites. It is concluded that following the evolution of the major amphipod groups, the oostegites were modified as egg sizes have changed as part of the reproductive strategies of the species within these groups.  相似文献

Replication times for all important chromosome bands, of both types R and Q (277 structures) are analysed. — The R-bands form a group of structures whose DNA replicates during the early S-phase, while the DNA situated in the Q-bands replicates during the late S-phase. — There may not exist overlapping between replication times of these two types of structures. — The widest R-bands are those which are the earliest to replicate; in general, the most intense Q-bands are those which are the latest to replicate. Especially among these last ones, a certain asynchronism exists between the replication times. Finally the heterochromatin of chromosomes 1, 16 and Y and of the short arms of the acrocentrics could contain two types of DNA which replicate at different times.  相似文献

We present a new method to produce tetraploid Crassostrea gigas by cytochalasin B inhibition of polar body 2 expulsion in diploid females crossed with tetraploid males. This offers a means of direct introgression of genetic characters from selected diploid to tetraploid lines, avoiding a triploid step. Offspring larval ploidy shifted over time and depended on size, with tetraploids more frequent among the smaller larvae and triploids among the large. Viable tetraploids were found at 4 and 6 months, indicating the technique was successful. The possibility that gynogenesis occurred was tested by microsatellite analysis to confirm the presence of paternally inherited alleles. These were present in all animals of the 2n × 4n + CB (female first) cross. However, a 4n × 2n + CB cross produced triploids, including some gynogens. Our method illustrates for the first time that diploid C. gigas eggs, if selected for large size, can give viable tetraploid offspring.  相似文献
