The ability of the Luteinizing Hormone-Releasing Hormone (LH-RH) analogs to displace LH-RH from its pituitary receptors was evaluated invitro. The two superactive analogs tested showed higher potency than the antagonists and LH-RH itself, D-Trp6-LH-RH being the most potent. The LH-RH specific binding activity in the pituitary fluctuated throughout the age of the rats. The highest number of LH-RH binding sites were seen on day 35 of age (276 fmol × 10?2/pit) and an increment was induced by 0.05 μg D-Trp6-LH-RH (400 fmol × 10?2/pit). However, 1 μg D-Trp6-LH-RH reduced the binding of LH-RH at all the times studied. In the control animals the number of estradiol binding sites increased on day 42 of age, and 0.05 μg D-Trp6-LH-RH augmented them on day 35 of age. On the contrary, 1 μg D-Trp6-LH-RH diminished the estradiol uterine receptors at all the times studied. Similar results were obtained in the ovariectomized-hypophysectomized rats on day 35 of age. Our studies demonstrated a biphasic action of D-Trp6-LH-RH on LH-RH pituitary receptors and a direct effect on uterus which could be mediated through the uterine estradiol receptors.  相似文献

We have recently demonstrated that gonadotropin releasing hormone (GnRH) acts directly on ovarian granulosa cells to inhibit the follicle stimulating hormone (FSH)-induced increase in granulosa cell steroidogenesis invitro. A GnRH antagonist, [D-pGlu1, D-Phe2, D-Trp3,6] GnRH (A), which is known to antagonize GnRH-stimulated gonadotropin release by cultured pituitary cells, was tested in the granulosa cell system. GnRH (10?8M) inhibited estrogen and progesterone production by FSH-treated granulosa cells invitro, whereas the antagonist A (10?6M) did not affect FSH stimulation of steroidogenesis. Antagonist A, when added together with GnRH and FSH, blocked the GnRH inhibition of FSH-induced steroidogenesis. Estrogen and progesterone production by granulosa cells was increased by 50% at a molar ratio (IDR50) of 201and121 ([antagonist]/[GnRH]), respectively. At 10?6M, antagonist A completely prevented the GnRH (10?8M) inhibition. A similar effect of antagonist A was seen in FSH-induced increase of luteinizing hormone (LH) receptor content. FSH treatment for 2 days invitro induced an 8-fold increase in LH receptor content in cultured granulosa cells; concomitant treatment with 10?8M GnRH completely inhibited the FSH effect. Antagonist A (10?6M), by itself, had no effect on the FSH action. However, when added together with FSH and GnRH, antagonist A completely abolished the inhibitory effect of GnRH. These results demonstrate that the direct inhibitory effect of GnRH on granulosa cell function can be prevented by a GnRH antagonist and that the GnRH action at the ovarian level may require stringent stereospecific interactions of these peptides with putative GnRH recognition sites.  相似文献

Rat serum is 2–5 fold more active than human serum in cleaving the three hypothalamic releasing hormones, luteinizing hormone releasing hormone (LH-RH), thyrotropin releasing hormone (TRH), and somatostatin. LH-RH was degraded by two distinct enzymatic mechanisms; 1) endopeptidase cleavage, 2) C-terminal cleavage. The C-terminal cleaving enzyme was active in rat serum but present only in trace levels in human. These mechanisms were substantiated by the use of suitably substituted analogs; D-Ala at position 6 of LH-RH prevented cleavage at the -Tyr5-D-Ala6-Leu7-site and the presence of ethylamide (C2H5NH2) at position 10 inhibited significantly the action of the second enzyme. These analogs have an enhanced biological activity invivo which correlates well with their decreased rate of degradation. Somatostatin was degraded by endopeptidase cleavage at one or more sites. D-Trip in position 8 blocked cleavage of the -Trp8-Lys9-bond, reducing significantly the rate of degradation. This also correlates well with the enhanced biological potency of the (D-Trp8)-somatostatin analog. TRH was degraded by cleavage of the pyroGlu-His and His-Pro.NH2 bonds with the release of free His and Pro. The analog (3-Me-His2)-TRH was degraded by a similar mechanism with the release of 3-Me-His.  相似文献

Inhibition of parathyroid hormone (PTH)-sensitive adenylate cyclase by {Nle-8, Nle-18, Tyr-34} bPTH-(3–34) amide was studied in thyroparathyroid-ectomized dogs. The inhibitory effect was shown to be markedly enhanced by the addition of calcium ions into the in, vitro assay system. At 0.1 mM Ca2+, complete inhibition by the antagonist was obtained. Chelation of exogenous Ca2+ by EGTA eliminated the Ca2+-induced inhibition. Both the basal and hormone-stimulated activities were decreased in the presence of 0.1 mM Ca2+, whereas the addition of EGTA increased both activities. Our results suggest that Ca2+ modulates canine renal PTH-sensitive adenylate cyclase and its inhibition by substituted bPTH-(3–34).  相似文献

Met5-enkephalin amide, D-Ala2-Met5-enkephalin amide, D-Ala2-Leu5-enkephalin amide, morphine sulfate and naloxone hydrochloride were examined for their effects on growth hormone and prolactin release invivo and invitro. Subcutaneous injection of D-Ala2-Met5 enkephalin amidea, D-Ala2-Leu5 enkephalin amideb and morphine sulfate, but not Met5-enkephalin and amidec, resulted in significant elevations in the serum growth hormone and prolactin of immature female rats. Naloxone blocked the hormone-stimulatory effect of the opioid receptor agonists and when administered alone significantly reduced serum growth hormone and prolactin concentrations. None of the drugs demonstrated a direct action on anterior pituitary tissue growth hormone or prolactin release invitro.  相似文献

Synthesis and biological activity of 2′-acetyltaxol and 7-acetyltaxol are reported. Activity is measured invivo by cytotoxicity toward the macrophage-like cell line J774.2, and invitro by promotion of microtubule assembly in the absence of exogenous GTP. Addition of an acetyl moiety at C-2′ results in loss of invitro activity but not cytotoxicity. The properties of 7-acetyltaxol are similar to those of taxol in its effects on cell replication and on invitro microtubule polymerization. Therefore a free hydroxyl group at C-7 is not required for invitro activity and this position is available for structural modifications.  相似文献

The effect of morphine sulfate (MS) on adenylate cyclase (AC) and phosphodiesterase (PDE) activities in the rat striatum was investigated. MS produced a dose-dependent increase in basal AC activity and did not alter sodium fluoride-induced stimulation both invivo (7.5–30 mg/kg, 1 hr pretreatment, i.p.) and invitro (1–100μM). invitro, when submaximal effective concentrations of dopamine and MS were combined, there was an additive effect. However, administration of MS invivo did not alter dopamine-induced stimulation of AC activity. MS, invitro and invivo inhibited PDE activity in a dose-dependent manner only with the high substrate concentration (3.3 × 10−3M cyclic AMP). Preliminary results from this study indicate that morphine affects the cyclic AMP system.  相似文献

The synthetic decapeptide “luteinizing hormone-releasing hormone” (LH-RH) was rendered antigenic by reaction of its histidine or tyrosine residues (7 : 3 approx.) with p-diazonium phenylacetic acid and coupling of the azo-derivatives formed to bovine serum albumin (BSA). Immunization of rabbits yielded antisera that bound 125I-labeled LH-RH (approx. 50 pg) at dilutions up to 1:200, 000 and showed no cross-reaction with unrelated hypothalamic and pituitary hormones, extracts from rat cerebral cortex, and with small fragments of LH-RH. Cross-reaction was minimal (0.2%) with the free acid analogue of LH-RH, and moderate with des-pGlu LH-RH (20%), des-pGlu-His-LH-RH (2.4%) and with LH-RH analogues in which a single residue (No. 4–6 or No. 8) was exchanged by an amino-acid of similar character (1.2–12%). Biologically active hypothalamic extract and LH-RH produced parallel 125I-LH-RH-binding inhibition curves, providing immunochemical support for the identity of the native releasing hormone with synthetic LH-RH.  相似文献

A series of peptide analogs of luteinizing hormone releasing hormone (LH-RH), altered at position 6 and 10, was synthesized and evaluated in vivo for the ability to induce ovulation in the diestrous rat and in vitro for ability to release pituitary luteinizing hormone and follicle stimulating hormone. All the analogs with D-amino acid substitutions at position 6, even those with large bulky side chain, exhibited an amazingly high potency compared with the parent hormone, LH-RH. On the basis of the biological activities, structure-activity relationships in the central part of this molecule were discussed in detail.  相似文献

A linear analogue of the luteinizing hormone-releasing hormone, longer than a decapeptide, is described for the first time, which is equivalent in potency to the best known inhibitors of ovulation, and which constitutes an important new lead to the design of inhibitors of even greater potency. At a dosage of 200 μg/rat, the undecapeptide [(1, D-Phe2, D-Trp3, D-Trp6]-LH-RH caused 100% inhibition of ovulation. The related analogues, [(1, D-Phe2, D-Trp3, D-Trp6]-LH-RH and [(Gly-Pro)1, D-Phe2, D-Trp3, D-Trp6]-LH-RH, were less active, invivo. All of these undecapeptides inhibited the action of 0.6 ng/ml of LH-RH by greater than 50% at the very low level of 10 ng/ml.  相似文献

When murine sarcoma virus-transformed cells are labeled with [3H]lysine invivo for various periods, 5 of 6 isoaccepting lysine tRNAs separable by RPC-5 chromatography are aminoacylated in 1 hr to the same extent that they are aminoacylated invitro. The sixth isoacceptor, tRNA6Lys, is not aminoacylated invivo to a measurable extent in 1 hr, although it is present in the tRNA prepared from the cells. All six isoacceptors are aminoacylated with [3H]lysine invivo when the labeling period is 2 or 3 hr. These results further show that invitro correlations of the amount of tRNA4Lys with cell division accurately reflect the situation invivo. Results of differential centrifugation indicate that tRNA6Lys occurs in mitochondria.  相似文献

Three-day old rats were injected subcutaneously either with natural purified pig ACTH (ACTH 1–39), ACTH 1–24, or ACTH analogues as long-acting zinc-phosphate preparations. ACTH 1–39, ACTH 1–24, ACTH 1–18, ACTH 1–16 accelerated the time of eye-opening, whereas an extract of corticosteroids produced invitro by excised adrenals of ACTH-treated three-day old rats was ineffective. Injections with ACTH on the twelfth day of life had no effect on eye-opening. It is concluded that a neonatal injection with ACTH or closely related analogues with markedly less corticotropic activity can accelerate the time of eye-opening. This effect is not mediated by the adrenal cortex. The sensitive period for it appears to be shortly after birth.  相似文献

The zymosan particles induced a time-dependent release of the chloride-dependent arginine aminopeptidase from rat peritoneal macrophages during invitro incubations. Intraperitoneal injections of zymosan, a streptococcal cell preparation and a Micrococcu-suspension caused the release of the chloride-activated arginine aminopeptidase into the peritoneal fluid. The arginine aminopeptidases obtained both from the cell cultivation media and the peritoneal washes were partly purified. The enzymes were similar with regard to the following properties: chloride activation with an optimum at physiological concentrations; strong inhibition by 10?6M p-chloromercuribenzoate; similar elution properties and preferential hydrolysis of mainly the N-L-aminoacyl-2-naphthylamines of arginine and lysine. The chloride-activated arginine aminopeptidase released into the media in invitro conditions was inactivated in contrast to the enzyme released into the peritoneal fluid as a result of the intraperitoneal injections. The timing of the release of the chloride-activated arginine aminopeptidase both in and invitro suggests that the enzyme plays a role in the initial phases of inflammation.  相似文献

A nonapeptide analog of luteinizing hormone-releasing hormone (LH-RH), [D-Ala6, des-Gly-NH210]-LH-RH ethylamide, was prepared by solid-phase methodology. The peptide was assayed against LH-RH in two in vivo systems and was found to be many times more potent than the naturally occurring hormone. In one of the tests, based on elevation of LH and FSH levels after infusion into immature male rats, the analog showed LH-releasing activity of 1600% and FSH-releasing activity of 1200% compared to LH-RH.  相似文献

The effects of narcotic analgesics, narcotic-antagonist analgesics and narcotic antagonists on ganglionic transmission in the superior cervical ganglia of the rat were studied invivo and invitro. Invivo administration of morphine, meperidine, methadone, pentazocine or naltrexone blocked ganglionic transmission. Levorphanol, cyclazocine, nalorphine and naloxone had no effect on ganglionic transmission in this procedure. Invitro studies confirmed the invivo results with the exception of levorphanol, cyclazocine and nalorphine, which were also found to block ganglionic transmission invitro. In both preparations, naloxone did not antagonize the effect of morphine, suggesting that the effects of morphine and the other opiates were nonspecific. Similar potency of d- and l-isomers of pentazocine and cyclazocine support this conclusion. The observation that naltrexone blocked ganglionic transmission, but the other pure narcotic antagonist, naloxone, was inactive is somewhat unique to this test procedure and possibly significant.  相似文献

The biological sensitivity of cultured human skin-derived fibroblasts was examined in terms of the ability of insulin and insulin analogues to stimulate the uptake of alpha-aminoisobutyrate in these cells. The relative biological activity measured: insulin=desalanine insulin > proinsulin > desoctapeptide insulin parallels exactly the relative activity of these compounds on fat and muscle preparations both invitro and invivo. Inactive insulin analogues do not stimulate alpha-aminoisobutyrate uptake. It is concluded that the chemical specificity of human fibroblast insulin receptors is retained in culture and that the biological responsiveness (alpha-aminoisobutyrate uptake) of such cells to insulin should prove useful for comparative studies of receptors obtained from different individuals.  相似文献

An agonist of chicken hypothalamic luteinizing hormone-releasing hormone (cLH-RH). [D-Trp6] cLH-RH, was synthesized and tested for luteinizing hormone (LH)-releasing activity using dispersed chicken anterior pituitary cells, as well as for binding to rat anterior pituitary membrane receptors. cLH-RH and mammalian LH-RH (mLH-RH) gave identical dose-response curves in stimulating chicken LH release (ED50=1.6 and 1.8×10?9M respectively) and similar estimates of potency. The [D-Trp6] analogs of cLH-RH and mLH-RH stimulated LH release at lower doses (ED50=7.0 and ~7.0×10?11M respectively) and were approximately 20-fold more potent. In contrast to the activity in the chicken bioassay, cLH-RH bound to rat anterior pituitary membrane receptors with a much lower affinity than did mLH-RH and had a relative potency of 2%. [D-Trp6] cLH-RH was approximately 100-fold more potent than cLH-RH in the rat receptor assay while [D-Trp6] mLH-RH was 28-fold more active than mLH-RH. These data demonstrate that substitution of Gly6 of LH-RH with D-Trp enhances the LH release from chicken pituitary cells to a similar extent to that observed in mammals, and indicate that the approaches used to produce active LH-RH analogs in mammals are likely to be applicable to birds.  相似文献

N-Palmitoyl-cysteine methyl ester and N-palmitoyl-glutamic acid α-methyl ester, which are analogues to the lipophilic N-terminal part of the lipoprotein from the outer membrane of Escherichia coli, were synthesized and tested for biological activity in an invitro lymphocyte culture system: in spleen cells from several inbred mouse strains the fatty acid derivatives exhibited mitogenic activity towards B-lymphocytes comparable to the effect of lipoprotein, as measured by 3H-thymidine incorporation and by hemolytic plaque assays. These results confirm former investigations, which have shown that the mitogenic principle of the lipoprotein molecule resides in its N-terminal fatty acid-containing part. The proper dispersion of the water-insoluble substances was critical for their mitogenic activity. Optimal mitogenicity was obtained by sonicating the substances at concentrations of 0.1 mg/ml in culture medium.  相似文献

Substitution of cyclopentylcarbonyl-(Cpc) for 1 in the effective and potent antiovulatory inhibitor, [D-Phe2, Pro3, D-Phe6]-LHRH (I) retained the in vitro potency. We know of no other inhibitor of the luteinizing hormone releasing hormone (LHRH) with a modification at position 1, which is as potent in vitro. This result agrees with the concept of the role of 1, D-Phe2, Pro3, D-Phe6]-LHRH did not inhibit ovulation in rats at the same dosage as did I; this result is under study to circumvent. Des-Gly10-[D-Phe2, Pro3, D-Phe6]-LHRH ethylamide and [Glu1, D-Phe2, Pro3, D-Phe6]-LHRH were significantly less active in vitro than I.  相似文献
