Levels of various protein fractions, (sarcoplasmic, myosin, actin, non-collagen and collagen) and the rate of their degradation by proteases were studied in phasic and tonic muscles of marine prawn, Penaeus indicus following acute (2 d) and chronic (15 d) exposure to sublethal concentration of phosphamidon. During exposure, greater decrease in sarcoplasmic protein fraction was observed in phasic muscle as compared to other myofibrillar proteins. But the sarcoplasmic protein content showed an elevation in tonic muscle. The changes in protein fractions were more pronounced during acute exposure than chronic exposure both in phasic and tonic muscles. These changes were correlated with the elevation of the acidic, neutral and basic protease activities during acute and chronic exposure. Free amino acids were increased during acute exposure, while they showed a significant decrease during chronic exposure in both the muscles. These results indicate that protein metabolism in both phasic and tonic muscles was significantly altered following phosphamidon exposure. These differential responses observed at acute and chronic exposure indicate the operation of compensatory mechanisms to mitigate the phosphamidon toxic stress.  相似文献

The muscle collagen of marine prawn,Penaeus indicus, was isolated by limited pepsin digestion. Based on selective salt precipitation, amino acid composition and gel electrophoretic pattern, the major collagen was found to be a homotrimer of á 1 chain, similar to type V collagen of vertebrates. Electron microscopy of reconstituted fibrils, made for the first time from a crustacean species, revealed a characteristic 64 nm periodicity. Biochemical studies indicate a less than normal amount of associated carbohydrates and an increased alanine content The major collagen had a denatu ration temperature of 37°C with an intrinsic viscosity of 11.3 dl/g. Spectral characteristics of the major collagen were studied. Results suggest the presence of genetically distinct collagen types and acid resistant cross links in crustacean muscle.  相似文献

Partial characterization of a natural agglutinin in the hemolymph of the lobster showed: (1) Calcium ion stabilized the agglutinin against inactivation at moderate (35–45°C) temperatures as well as against a reversible increase in activity at nonphysiological pH levels (pH 6 and 9). (2) Serum frozen at ?20°C in the absence of calcium showed an increase in agglutinin activity. (3) Heat inactivation curves indicated a single component for the agglutinin. (4) Heat inactivation destroyed the ability of the agglutinin to be adsorbed to the erythrocyte antigen. (5) The chemical nature of the antigenic reactive site differed with the species of erythrocyte tested; D-glucosamine blocked or inhibited adsorption of the agglutinin to erythrocytes of most species tested. (6) In vivo the agglutinin is located in the hemocytes.  相似文献

Juveniles of freshwater prawn M. malcolmsonii were exposed to 1/6th concentration of LC50 of Hg and Cu for 48 hr. Sampling was done at 1 8, 12, 16, 24, 30, 36, 42 and 48 hr of exposure. Gill and hepatopancreas were dissected and subjected to one-dimensional electrophoresis. Western blotting was employed to determine the relative concentration of heat shock protein, hsp 70 (stress-70) in each sample. In the gill tissue of the prawn that had been exposed to Hg (0.024 mgHg 1(-1)), stress-70 was detected from the 1st hr till the 16th hr of exposure. But in the gills of Cu exposed prawn, synthesis of stress-70 appeared from the 1st hr till the 24th hr. Synthesis of hsp70 was not recorded after the 24th hr in the gills of exposed prawns. Synthesis of stress-70 was also found to be tissue-specific for both metals in this prawn. When the antibody probe raised against stress-70 was used, synthesis of stress-70 was not observed in hepatopancreas of prawns exposed to Hg or Cu, during the entire period of exposure of 48 hr. The expression of stress-70 in M. malcolmsonii following exposure to Hg and Cu is apparently only transient, and also a differential expression of stress-70 between gill and hepatopancreas was observed for both the metals.  相似文献

A cell surface glycoprotein induced by the mating pheromone alpha factor in Saccharomyces cerevisiae a cells has been purified to homogeneity. At 4 x 10(-9) M it strongly inhibits mating-type-specific agglutination between a and alpha cells. The protein is solely O-glycosylated. It consists of 29% carbohydrate and its apparent molecular mass is 22 kd on SDS gels. After HF treatment it behaves like a protein of 13 kd; therefore its true molecular mass probably is close to 18 kd. Mild periodate treatment destroys the biological activity of the purified protein. The protein contains one cysteine, no arginine, and 27% of the amino acids are serine and threonine residues, two thirds of which are glycosylated. With a polyclonal antibody the glycoprotein can already be detected at the cell surface 15 min after pheromone addition. The inducible antigen is not expressed in a specific phase of the cell cycle; it first appears exclusively on the growing bud. Mother cells express the antigen on their surface only after the daughter cells have separated; it is then localized at the tip of the pear-shaped 'shmoo'. Using the secretory ts-mutant sec 18 is shown that a mannosylated precursor of a agglutinin accumulates at the endoplasmic reticulum.  相似文献

A natural agglutinin from the serum of the hermit crab Diogenes affinis was purified to homogeneity by a single-step affinity chromatography using N-acetylglucosamine-coupled Sepharose 6B. The purified serum agglutinin (PSA) showed a strong affinity for rat RBC, and its hemagglutinating (HA) activity was specifically dependent on Ca2+ and reversibly sensitive to EDTA. PSA in active form has a molecular mass estimate of 185 kDa and is composed of four non-identical subunits (51, 49, 42 and 39 kDa) cross-linked by interchain disulfide bonds. The homogeneity of PSA was corroborated by immunodiffusion and immunoelectrophoretic analyses using rabbit antiserum raised against the agglutinin. The antibodies in this antiserum appear to be specific for RBC-binding sites of the agglutinin molecules as revealed by the ability of the antiserum to neutralize HA activities of both whole serum and PSA of D. affinis. In HA-inhibition assays performed with several carbohydrates and glycoproteins, PSA showed a distinct and unique specificity for acetyl group in carbohydrates independently of the presence of this group on C-2 or C-5 and its stereochemical arrangement in the axial or equatorial orientation. Besides, this agglutinin appears to recognize the terminal N- and O- acetyl groups in the oligosaccharide chain of glycoconjugates. The HA activity of D. affinis agglutinin was also susceptible to inhibition by lipopolysaccharides from diverse gram-negative bacteria, which might indicate a significant in vivo role of this humoral agglutinin in the host immune response against bacterial infections.  相似文献

Fucus vesiculosus agglutinin has been purified to homogeneity by conventional chromatographic procedures and characterized as a mucopolysaccharide with 90% carbohydrate content. Estimated molecular weight is about 2 X 10(6) daltons. It has no sub-unit structure and its isoelectric point is 3.2. It contains 1.23% S, 0.24% Ca and 0.06% P. Agglutinin mediated sheep red blood cell agglutination was inhibited only by glycoproteins with complex lateral oligosaccharide chains resembling some of the oligosaccharide chains found in the erythrocyte membrane glycoproteins. Metaperiodate treatment of the sheep red cells rendered them non-agglutinable. Sequential degradation of the oligosaccharide chains with glycosidases suggests that inner mannose residues are implicated in the receptor binding-sites for the agglutinin. Consequently we think that this agglutinin can be a lectin or a lectin-like molecule with complex saccharide specificity.  相似文献

Changes in hepatopancreas, muscle and gill tissue nitrogen metabolic profiles were studied in a penaeid prawn, Penaeus indicus, following its exposure to sublethal concentrations of methylparathion, carbaryl and aldrin. In all the insecticide exposed prawn tissues, Ammonia levels were significantly increased and a shift in the nitrogen metabolism towards the synthesis of urea and glutamine was observed. Inhibition of glutamate oxidation to ammonia and alpha-ketoglutarate by glutamate dehydrogenase suggests a mechanism whereby hyperammonemia is reduced by minimizing the addition of further ammonia to the already existing elevated ammonia pool. Increased alanine and aspartate aminotransferases demonstrates the onset of gluconeogenesis. Mechanisms to detoxify the ammonia by enhancing the synthesis of urea and glutamine at the cellular level was observed in the selected tissues pave way for the survivability of prawns in insecticide polluted environs.  相似文献

The subtropical grapsid crab Percnon gibbesi (H. Milne Edwards, 1853) is one of the most recent alien decapods found in the Mediterranean, where it was discovered at Linosa (Pelagie Islands, Sicily Strait) in summer 1999. At present, the invasion of this species has been recorded in several other insular localities. We studied the feeding habits of the crab in an attempt to understand its success in the Mediterranean. The morphological characteristics of the chela, the feeding adaptations of the gastric mill and the analysis of stomach contents indicate that P. gibbesi is a strictly herbivorous species, a characteristic not shared with any other large-sized infralittoral Mediterranean crab.  相似文献

A purified preparation of trypsin inhibitor was obtained from the hemolymph of a solitary ascidian, Halocynthia roretzi, by a procedure including trypsin-Sepharose chromatography, DEAE-cellulose chromatography, and Sephadex G-50 gel filtration. The product was a mixture of two isoinhibitors, inhibitors I and II. They were separated from each other by high-performance liquid chromatography on an anion exchanger column, and showed almost identical amino acid compositions. They were also indistinguishable in terms of apparent specific inhibitory activity against bovine trypsin when the activity was assayed with the inhibitors at rather high concentrations (greater than 50 nM). A large difference was observed between them, however, in the inhibition constants, which correspond to the dissociation constants of the inhibitor-trypsin complexes; the inhibition constant of inhibitor I was 90 pM, whereas that of inhibitor II was 4.7 nM. The molecular weights of inhibitors I and II were estimated to be 6,000 and 4,500, respectively, by SDS-polyacrylamide gel electrophoresis, while an almost identical value, 9,000, was obtained for both of them by gel filtration. The molecular weight calculated from the amino acid compositions was 5,929 for both. The isoelectric points were also identical, that is about 5.0. Both of the inhibitors were heat-stable. Ascidian inhibitor I also inhibited other trypsin-like enzymes of mammalian origin, as well as those of ascidian origin.  相似文献

A biliverdin binding protein, insecticyanin, has been isolated from the hemolymph of the fourth instar tobacco hornworm Manduca sexta. The protein has been purified to apparent homogeneity by conventional chromatography with a cumulative yield of 40-50%. The protein (Mw 71 600) is composed of three subunits (Mr 23 000). Each subunit binds one biliverdin molecule. Proton magnetic resonance spectroscopy and absorption spectroscopy demonstrate that the bilin is the biliverdin IX gamma isomer.  相似文献

A lectin from the hemolymph of the banana shrimp Fenneropenaeus merguiensis was purified by affinity chromatography on a fetuin-agarose column following by gel filtration on a Superose-12 column. The native molecular mass of purified F. merguiensis lectin (FmL) determined by gel filtration was 316.2 kDa and its carbohydrate content was estimated to be 4.4%. By SDS-PAGE analysis, purified FmL consisted of 32.3 kDa and 30.9 kDa subunits. These data suggest that this lectin is an oligomer. Two-dimensional electrophoresis showed that it had a pI value of 6.0 and was mainly composed of glycine, serine, histidine, glutamic acids and glutamine, with relatively lower amounts of methionine and tyrosine. Purified FmL expressed higher agglutination activity against rabbit and rat erythrocytes than with those from human, and its activity was Ca(2+)-dependent. The hemagglutinating activity of FmL was stable up to 55 degrees C and at pH 7.5-8. N-acetylated sugars, such as ManNAc, GlcNAc, GalNAc, and NeuNAc were strong inhibitors of the FmL induced hemagglutinating activity with NeuNAc being most effective. Porcine stomach mucin and fetuin were the most potent inhibitors of FmL. Purified FmL caused selective agglutination of Vibrio harveyi, and Vibrio parahemolyticus both pathogens of this Penaeus species and to a lesser extent Vibrio vulnificus but had no effect on the non-pathogenic strains; Vibrio cholerae, Salmonella typhi and Escherichia coli. Its bacterial agglutination was also completely inhibited by NeuNAc, mucin, fetuin and also anti-FmL antibody. This observation indicates that FmL may contribute to the defense response of this species of penaeid shrimps to potentially pathogenic bacteria.  相似文献

Epidermis-origin hemolymph protein (EOHP) was identified and purified from the last instar larval hemolymph of Galleria mellonella by anion exchange chromatography, chromatofocusing chromatography, and Sephadex G-100. The EOHP has a native molecular mass of 47 kDa and is composed of one subunit. The isoelectric point of the EOHP was determined to be 5.3. The amino acid composition of the EOHP was rich in aspartic acid, glutamic acid and lysine, but poor in tyrosine, methionine, and tryptophan. EOHP is present in hemolymph over the period from the 4th instar larvae to the adult stage examined. Concentration of EOHP is high during the larval stage but gradually decreased during the developmental stage from pupal to adult stage. EOHP is present in the cuticle, fat bodies and trachea but not in hemocytes, fore gut, mid gut and hind gut.  相似文献

Midgut gland carbohydrate metabolism of penaeid prawn, Penaeus indicus was studied on acute and acclimation to sublethal concentration of phosphamidon. The midgut gland tissue of acclimated prawn showed inhibited glycolysis with an onset of gluconeogenesis. In general acclimation to sublethal concentration resulted in the elevation of the synthetic phase of midgut gland carbohydrate metabolism.  相似文献

Cyclograpsus lavauxi H. Milne Edwards, 1853 occurs under boulders near the littoral fringe, where it is wetted only briefly, if at all, in each tide. Mean water content of sea-water equilibrated crabs was 62.3 % of body weight. On warm, windy days crabs on the shore lose > 17.5% of body water. Water loss for crabs of 1.6 g mean body weight was 0.29 % water content · h −1 · mm Hg saturation deficit −1, which is similar to that of species occupying positions lower on the shore. C. lavauxi is, however, able to tolerate comparatively greater losses of body water (up to 36%) compared with species from lower shore levels.

As the gills dry, the lamellae separate into regular clumps which may help to maintain gas exchange. Resting oxygen consumption (Vo2) after 3–5 h settlement was similar in water and air. During enforced activity, Vo2 increased by factors of 5.3 in water and 2.6 in air at a standardized body weight of 1.5 g, indicating an appreciable aerobic scope in both media. Loss of up to 16% of body water did not depress resting aerial Vo2 or aerobic scope.

In resting crabs there was no change in haemolymph pH after 24 h of either immersion or emersion, but haemolymph [Ca2+], [HCO3] + [CO2−3], and calculated Pco2 all rose in air relative to aquatic values. These results suggest that cuticular or other endogenous CaCO3 is mobilized to compensate the respiratory acidosis in air. The implications of such physiological properties for aquatic and aerial activity on the shore are discussed.  相似文献

Freshwater prawn, Macrobrachium malcolmsonii, has potential for aquaculture. Larvae of M. malcolmsonii were reared in synthetic brackishwater using commercial salts. Live and artificial feeds were provided to larvae either individually or in combination. Survival was significantly higher when larvae were fed with live feed only. However, larvae fed with artificial feed showed significantly lower survival. Survival was significantly higher when Artemia combined together with other feeds. Despite its limitations, synthetic brackishwater can be used for freshwater prawn seed production.  相似文献
