The objective of this study was to assess the impacts of land use changes and irrigation water resource on the nitrate contamination in shallow groundwater. 394 water samples were sampled from the same irrigation wells during a period of five years (from 2002 to 2007) in Huantai County in the North China Plain. NO3-N concentration in irrigation wells was measured. Geostatistical method combined with GIS technique was used to analyze the spatio-temporal distribution of groundwater NO3-N concentrations in Huantai County. Land use type and irrigation water resource were combined with the variation of NO3-N concentrations by statistical approach to investigate the relationship between them. The distribution map showed that the percentages of area increased by 13.06%, 14.37%, 12.23% and 3.85% for that had nitrate concentrations of 10–15, 15–20, 20–30 mg L?1 and greater than 30 mg L?1 for shallow groundwater, respectively, while decreased by 28.87% and 14.63% for 0–5 and 5–10 mg L?1. In the well-irrigated field, the NO3-N concentrations in shallow groundwater had increased for vegetables, wheat–vegetables and wheat–maize rotations. In contrast, fast-growing tree system could act as a buffer to retain shallow groundwater nitrate content which resulted in reduced NO3-N concentrations. Under the same land use condition, irrigation with sewage, or well and sewage by turns would both enormously add nitrate to groundwater.  相似文献

Due to the enviornmental problems associated with disposal of livestock sludge, many stabilization studies emphasizing on the sludge volume reduction were performed. However, little is known about the microbial risk present in sludge and its stabilized products. This study microbiologically explored the effects of anaerobic lagoon fermentation (ALF) and autothermal thermophilic aerobic digestion (ATAD) on pathogen-related risk of raw swine manure by using culture-independent 16S rDNA cloning and sequencing methods. In raw swine manure, clones closely related to pathogens such as Dialister pneumosintes, Erysipelothrix rhusiopathiae, Succinivibrioan dextrinosolvens, and Schineria sp. were detected. Meanwhile, in the mesophilic ALF-treated swine manure, bacterial community clones closely related to pathogens such as Schineria sp. and Succinivibrio dextrinosolvens were still detected. Interestingly, the ATAD treatment resulted in no detection of clones closely related to pathogens in the stabilized thermophilic bacterial community, with the predominance of novel Clostridia class populations. These findings support the superiority of ATAD in selectively reducing potential human and animal pathogens compared to ALF, which is a typical manure stabilization method used in livestock farms.  相似文献

The use of manure as a fertilizer in agriculture includes the risk of spreading pathogenic infectious agents to the environment, to animals and humans. The treatment of manure can help avoid or reduce these risks. Even if the treatment is dominated by economic considerations such as biogas production, ammonia stripping or phosphorous precipitation, the hygienic aspect should be kept in mind. Otherwise, new infection chains may be established by the use of insufficiently treated manure by-products such as fertilizers still containing infective pathogens. Treatment plants should use a concept according to HACCP principles that includes hazard analysis, risk assessment, the determination of process relevant CCPs and the validation of the process by determining the hygienizing efficiency using representative test organisms as well as microbial end product supervision. Treatment methods can be divided into physical, chemical and microbiological treatment, sometimes used in combination. For economical reasons, only composting or anaerobic treatment (biogas) or, to a minor extent, aerobic thermophilic stabilization (ATS) are used as routine preventive measures on a farm level. In cases of outbreaks of notifiable diseases both physical and chemical treatment of manure can lead to reliable disinfected/pasteurised end products which can be used in agriculture without long-lasting risks for soil fertility or the environment.  相似文献

At Department of Energy Site 300, beneficial hydrocarbon cocontaminants and favorable subsurface conditions facilitate sequential reductive dechlorination of trichloroethene (TCE) and rapid oxidation of the resultant cis-dichloroethene (cis-DCE) upon periodic oxygen influx. We assessed the geochemistry and microbial community of groundwater from across the site. Removal of cis-DCE was shown to coincide with oxygen influx in hydrocarbon-containing groundwater near the source area. Principal component analysis of contaminants and inorganic compounds showed that monitoring wells could be differentiated based upon concentrations of TCE, cis-DCE, and nitrate. Structurally similar communities were detected in groundwater from wells containing cis-DCE, high TCE, and low nitrate levels. Bacteria identified by sequencing 16S rRNA genes belonged to seven phylogenetic groups, including Alpha-, Beta-, Gamma- and Deltaproteobacteria, Nitrospira, Firmicutes and Cytophaga-Flexibacter-Bacteroidetes (CFB). Whereas members of the Burkholderiales and CFB group were abundant in all wells (10(4)-10(9) 16S rRNA gene copies L(-1)), quantitative PCR showed that Alphaproteobacteria were elevated (>10(6) L(-1)) only in wells containing hydrocarbon cocontaminants. The study shows that bacterial community structure is related to groundwater geochemistry and that Alphaproteobacteria are enriched in locales where cis-DCE removal occurs.  相似文献

Eucalypt plantations have been trialled in recent years as a control measure for shallow groundwater associated with secondary salinity. Uncertainty still remains as to the potential growth and water use of these plantations; these relate mainly to the problems associated with drought stress and accumulated solutes in the root zone resulting from saline groundwater uptake. This study investigates the water balance and identifies water sources of a 21 year old unirrigated Eucalyptus grandis W. Hill ex Maiden (flooded or rose gum) and E. camaldulensis Dehnh. (river red gum) plantation over shallow saline groundwater in the Shepparton Irrigation Region of northern Victoria, Australia. Water sources used by the plantation were identified using a monthly water balance approach, together with investigations of stable isotopes of water (deuterium and oxygen-18), soil water and chloride. We found these trees to be heavily reliant on rainwater, and derive approximately 15% of their transpiration requirements from saline (10,000 mg l?1) groundwater at the capillary fringe. Rainfall at the site is relatively low (465 mm year?1 on average) and groundwater uptake provides a stable water source that leads to a slight extension of the growing period of these trees. There is little potential for recharge, with subsurface water moving into the groundwater depression created by tree water uptake.  相似文献

Aims: To isolate and characterize multiple antibiotic resistance plasmids found in swine manure and test for plasmid‐associated genetic markers in soil following manure application to an agricultural field. Methods and Results: Plasmids were isolated from an erythromycin enrichment culture that used liquid swine manure as an inoculant. Plasmids were transformed into Escherichia coli DH10β for subsequent characterization. We isolated and DNA sequenced a 22 102‐bp plasmid (pMC2) that confers macrolide, and tetracycline resistances, and carries genes predicted to code for mercury and chromium resistance. Conjugation experiments using an pRP4 derivative as a helper plasmid confirm that pMC2 has a functional mobilization unit. PCR was used to detect genetic elements found on pMC2 in DNA extracted from manure amended soil. Conclusions: The pMC2 plasmid has a tetracycline‐resistant core and has acquired additional resistance genes by insertion of an accessory region (12 762 bp) containing macrolide, mercury and chromium resistance genes, which was inserted between the truncated DDE motifs within the Tn903/IS102 mobile element. Significance and Impact of the Study: Liquid swine manure used for manure spreading contains multiple antibiotic resistance plasmids that can be detected in soil following manure application.  相似文献

Concern has greatly increased about the potential for contamination of water, food, and air by pathogens present in manure. We evaluated pathogen reduction in liquid swine manure in a multi-stage treatment system where first the solids and liquid are separated with polymer, followed by biological nitrogen (N) removal using nitrification and denitrification, and then phosphorus (P) extraction through lime precipitation. Each step of the treatment system was analyzed for Salmonella and microbial indicators of fecal contamination (total coliforms, fecal coliforms, and enterococci). Before treatment, mean concentrations of Salmonella, total coliforms, fecal coliforms, and enterococci were 3.89, 6.79, 6.23 and 5.73 log(10) colony forming units (cfu)/ml, respectively. The flushed manure contained 10,590 mg/l TSS, 8270 mg/l COD, 688 mg/l TKN and 480 mg/l TP, which were reduced >98% by the treatment system. Results showed a consistent trend in reduction of pathogens and microbial indicators as a result of each step in the treatment system. Solid-liquid separation decreased their concentrations by 0.5-1 log(10). Additional biological N removal treatment with alternating anoxic and oxic conditions achieved a higher reduction with average removals of 2.4 log(10) for Salmonella and 4.1-4.5 log(10) for indicator microbes. Subsequent P treatment decreased concentration of Salmonella and pathogen indicators to undetectable level (<0.3 log(10) cfu/ml) due to elevated process pH (10.3). Our results indicate that nitrification/denitrification treatment after solids separation is very effective in reducing pathogens in liquid swine manure and that the phosphorus removal step via alkaline calcium precipitation produces a sanitized effluent which may be important for biosecurity reasons.  相似文献

Two methods, centrifugation and flocculation, were evaluated to determine their efficiencies of recovery of Toxoplasma gondii oocysts from contaminated water samples. Demineralized and tap water replicates were inoculated with high numbers of sporulated or unsporulated T. gondii oocysts (1 x 10(5) and 1 x 10(4) oocysts). The strain, age, and concentration of the seeded oocysts were recorded. Oocysts were recovered either by centrifugation of the contaminated samples at various g values or by flocculation with two coagulants, Fe(2)(SO(4))(3) and Al(2)(SO(4))(3). The recovery rates were determined with the final pellets by phase-contrast microscopy. Sporulated oocysts were recovered more effectively by flocculation with Al(2)(SO(4))(3) (96.5% +/- 21.7%) than by flocculation with Fe(2)(SO(4))(3) (93.1% +/- 8.1%) or by centrifugation at 2,073 x g (82.5% +/- 6.8%). For the unsporulated oocysts, flocculation with Fe(2)(SO(4))(3) was more successful (100.3% +/- 26.9%) than flocculation with Al(2)(SO(4))(3) (90.4% +/- 19.1%) or centrifugation at 2,565 x g (97.2% +/- 12.5%). The infectivity of the sporulated oocysts recovered by centrifugation was confirmed by seroconversion of all inoculated mice 77 days postinfection. These data suggest that sporulated Toxoplasma oocysts purified by methods commonly used for waterborne pathogens retain their infectivity after mechanical treatment and are able to induce infections in mammals. This is the first step in developing a systematic approach for the detection of Toxoplasma oocysts in water.  相似文献

Biological nitrogen fixation (BNF) in sugarcane is considered one of the principal reasons for the success of the Brazilian Ethanol Program (PRO-ALCOOL) for motor car fuel. The BNF influences positively the energy balance of sugarcane crops for alcohol production. Gluconacetobacter diazotrophicus is a nitrogen-fixing bacterium associated with sugarcane, and is particularly abundant and active in the early stages after germination. The objective of this work was to evaluate the effect of the addition of increasing amounts of two sources of mineral nitrogen (ammonium sulphate and calcium nitrate) on the population of G. diazotrophicus and also to evaluate its effect on nitrogenase (acetylene reduction) activity and accumulation of N by two sugarcane hybrids, SP 701143 and SP 792312. The results showed that both varieties differed in the form of nitrogen they prefer to uptake from the soil. The variety SP 701143 preferred ammonium sulphate, whilst the variety SP 792312 preferred N from calcium nitrate. In both varieties, the addition of increased doses of ammonium and nitrate inhibited the population of G. diazotrophicus but in the variety SP 701143 the inhibition was more pronounced in the presence of calcium nitrate. The acetylene reduction activity was inhibited in both varieties, especially in variety SP 792312 in the presence of either of the two nitrogen sources.  相似文献

To manage public health and make better use of groundwater resources, the concentration characteristics and a health risk assessment of eight heavy metals in shallow groundwater were studied. Besides this, this paper introduced triangular fuzzy numbers into the USEPA health risk assessment model to assess the health risk posed to local children and adults through different exposure pathways. The results showed that Mn levels exceeded the WHO’s guideline values of 100?µg/L with the proportion of 27.98% and Sr were over the health reference level (HRL) of 1500?µg/L with the proportion of 56.25%, while other heavy metals were below the corresponding standard. The results of the HRA showed that the non-carcinogenic risks from Sr and Mn in the district were relatively higher, while those from the remaining six heavy metals were relatively lower. All hazard index (HI) values did not exceed the safety level of 1 for either age group. The average carcinogenic risk from Cr was slightly higher than the acceptable level of 1?×?10?6 for adults. Sensitivity analyses conducted using Monte Carlo simulation indicated that Sr and Cr concentrations were the most influential variables contributing to the non-carcinogenic and carcinogenic risk values, respectively, while body weight had a minor contribution.  相似文献

An alternative to land spreading of manure effluents is to mass-culture algae on the N and P present in the manure and convert manure N and P into algal biomass. The objective of this study was to determine how the fatty acid (FA) content and composition of algae respond to changes in the type of manure, manure loading rate, and to whether the algae was grown with supplemental carbon dioxide. Algal biomass was harvested weekly from indoor laboratory-scale algal turf scrubber (ATS) units using different loading rates of raw and anaerobically digested dairy manure effluents and raw swine manure effluent. Manure loading rates corresponded to N loading rates of 0.2 to 1.3 g TN m−2 day−1 for raw swine manure effluent and 0.3 to 2.3 g TN m−2 day−1 for dairy manure effluents. In addition, algal biomass was harvested from outdoor pilot-scale ATS units using different loading rates of raw and anaerobically digested dairy manure effluents. Both indoor and outdoor units were dominated by Rhizoclonium sp. FA content values of the algal biomass ranged from 0.6 to 1.5% of dry weight and showed no consistent relationship to loading rate, type of manure, or to whether supplemental carbon dioxide was added to the systems. FA composition was remarkably consistent among samples and >90% of the FA content consisted of 14:0, 16:0, 16:1ω7, 16:1ω9, 18:0, 18:1ω9, 18:2 ω6, and 18:3ω3.  相似文献

Manure slurries stored in pits under slatted floors of both finishing and nursery barns were sampled at four different depths to study stratifications of total solids (TS) and nutrients (nitrogen and phosphorus), and to determine the relationship between the stratification of TS and nutrients. The results obtained can be used to improve the management and handling of swine manure in the under-slat storage pits. A management scheme that can be adopted for both the finishing and the nursery barns' pits is the layer-by-layer harvesting of the manure. The thinner manure, which is lower in nutrients, can be spread on land near the production units in larger volumes or it may pumped to land remote from the production units without causing many clogging problems. The thicker manure, higher in nutrients, can be transported to land further away and spread in smaller volumes. The TS content of each stratum can be used to accurately estimate the nitrogen and phosphorus levels in the respective strata so that application rates can easily be adjusted accordingly during the time of land application.  相似文献

Supplementary nitrogen (N) side-dressing via the irrigation system is needed in organic cropping. The aim here was to improve N-extraction efficiency, by testing five extraction protocols with guano, layer and broiler manures. The manure-N released by the different methods and manures was mainly in the form of ammonia and ranged from 50% to 85% with no differences among extraction methods. Volatilised ammonia from the extract solution was trapped. At the end of the extraction period, the pH of the extract solution was raised and the rest of the volatilised ammonia was trapped. In the case of guano, about 89% of the manure-N that was mineralised to the extract solution volatilised (after a pH increase), whereas in the layer and broiler manures, 59% and 54% were volatilised, respectively. Extraction of ammonia, its volatilisation and entrapment could provide a significantly more efficient N source than using the extract solution as currently recommended.  相似文献

南方丘陵红壤茶园长期受到酸沉降的胁迫,但茶树根际氮(N)、磷(P)转化过程对酸雨的响应及其机制尚不清楚.以江西典型丘陵红壤25年茶园为对象,开展pH 4.5、pH 3.5、pH 2.5及对照4种不同强度酸雨处理的原位模拟试验,于试验第3年测定根际和非根际土壤矿质N、速效P和相关酶的活性,并估测土壤N、P矿化速率,计算各变量的根际效应.结果表明: 与对照相比,pH 4.5、pH 3.5和pH 2.5处理根际土壤NO3--N含量分别降低了7.1%、42.1%和49.9%,矿质N分别降低了6.4%、35.9%和40.3%,速效P分别降低了10.5%、41.1%和46.9%;根际氨化速率分别降低了18.7%、30.1%和44.7%,N净矿化速率分别降低了3.6%、12.7%和38.8%,P矿化速率分别降低了31.5%、41.8%和63.0%,但不同处理之间根际硝化速率差异不显著;根际土壤脲酶和酸性磷酸酶活性均表现为随酸雨加重呈增强的趋势(P<0.05).非根际土壤除NH4+-N外,其他有效N和P含量未随酸雨加重而改变;不同酸雨处理对非根际土壤氨化、硝化、N净矿化速率和P矿化速率的影响差异均不显著.根际NH4+-N、NO3--N、矿质N、氨化和净矿化速率均随着酸雨强度加重由正效应转变为负效应,而脲酶和酸性磷酸酶活性由负效应转变为正效应,但速效P和P矿化速率始终表现为负效应,硝化速率始终为正效应.综上所述,连续酸雨加重总体上抑制了根际N、P转化,降低其有效性,且不同程度改变其根际效应,从而影响茶园养分循环.  相似文献

An in situ co-metabolic air sparging (CAS) study was carried out at McClellan Air Force Base (MAFB), Sacramento, CA, USA, in a trichloroethene- (TCE) and cis-dichloroethene (cis-DCE)-contaminated aquifer where one test zone received 2% propane in air and the other served as a control and received only air. As part of that study, bacterial population shifts were evaluated by length heterogeneity polymerase chain reaction (LH-PCR). The results showed that an organism(s) that had a fragment size of 385 bp was positively correlated with propane removal rates. The 385 bp fragment consisted of up to 83% of the total fragments in the analysis when propane removal rates peaked. A 16S rRNA clone library made from the bacteria sampled from the propane-sparged groundwater included clones of a TM7 division bacterium that had a 385 bp LH-PCR fragment; no other bacterial species with this fragment size were detected. Both propane removal rates and the 385 bp LH-PCR fragment decreased as nitrate levels in the groundwater decreased. Extinction culturing in natural unamended groundwater medium was used to assess the bacterial diversity of the culturable fraction of microorganisms in both CAS and air-sparged groundwater and to bring novel species into culture for further study. The dominant cultures acquired from the CAS groundwater were from the Herbaspirillum/Oxalobacter clade. The dominant cultures from the air-sparged groundwater were from a novel beta-Proteobacterial clade, which we named after isolate HTCC333.  相似文献
