Ten-year-old trees from four Italian populations of Pinus leucodermis (populations A, B, C and D), which were collected from different sites at different altitudes, were grown near Florence, Italy. Needle CO2 gas exchange and chlorophyll fluorescence response to increasing light intensities were evaluated; gas exchange and chlorophyll fluorescence variation between April and July were also monitored. Populations A, B and C showed a similar photosynthetic response to increasing photosynthetic photon flux density (PPFD) intensities, while at various light intensities population D, which originated from the highest altitude, showed the highest photosynthetic rates. In this population photosynthesis was saturated at PPFDs higher than 900 µmol m-2s-1 and a slow decrease of effective photosystem II quantum yield and F'V/F'M in response to increasing PPFDs were found. The same trees also showed a faster recovery in photosynthesis from limitations induced by winter temperatures than the other three populations. This work showed that photosynthetic response to light in population D was different from the other populations; trees from this population were probably naturally selected to prevent photoinhibition due to excess light.  相似文献

Summary: Stingless bee queens have for long been assumed to mate once on a nuptial flight, early in life. To evaluate critically monandry in one stingless bee, Scaptotrigona postica, worker offspring (adults or brood) were genetically analysed with microsatellite loci, five of which were developed specifically for the species. Marker loci were highly variable; unbiased estimates of heterozygosity were > 0.5. "Foreign" workers, either those having drifted from other colonies (circa 2%) or those of a replacement queen, were identified with the genetic markers and removed from further analysis. Worker genotypes were consistent with some queens having mated once and others having mated with up to six different males. Scaptotrigona postica queens are therefore facultatively polyandrous. Effective mating frequencies, me, were generally lower than the number of patrilines observed. Relatedness estimates of nestmates from individual colonies concurred with those derived from direct counts of the number of patrilines and their proportional representation. Putative genotypes of a colony's queen and her mates were deduced from those of her workers. Queens were generally not related to their mates. For one polyandrous queen, her six mates were related to each other, possibly because of numerically biased representation of males from different colonies at mating sites. However, males at an aggregation outside a colony came from numerous colonies.  相似文献

Estradiol (E2) was measured in spring and winter tissues in several populations of dioecious Populus tremuloides Michx. and in four trees with perfect flowers. A correlation between the concentration of E2 and sex expression was observed. In catkins, E2 concentrations increased before anthesis, peaked during flowering, and decreased as flowers matured. The increases were concurrent with sporogenesis and development of gametophytes. Statistically significant seasonal variation in E2 concentration was also evident. Dormant winter tissues showed significantly lower E2 concentrations than spring tissues. Branches exposed to relatively more light had significantly higher E2 content in both spring and winter. Seasonal variation in E2 content was also observed in trees of the same sex, among branches of the same tree, and among different organs.  相似文献

The correlation between 99 clone female and male fertilities in a first generation seed orchard of Pinus densiflora was studied over 6 years. The effective number of the parent (Np) and the variance effective population number [Ne(v)] were used to assess the impact of total (OT), female (Nf) and male (Nm) fertility variation. A theoretical framework was developed to account for female and male fertility correlations as well as the impact of possible pollen contamination. Total fertility variation was described by the sibling coefficient (OT: the probability that two genes randomly chosen from the gamete gene pool originate from the same parent), which was further subdivided into Nf and Nm. These parameters were compared under various conditions including the total seed harvest, imposing on equal seed harvest among the orchard's clones and two contamination scenarios (M = 0 and 20%). Fertility variations among females, males and clones were observed within and among years. Sibling coefficients (OT) were lower, but the effective number of parent (Np) and variance effective population number (Ne(v)) were higher in years with moderate female and good male strobilus production. Np for female and male reproductive outputs varied from 49 to 82 and from 57 to 93, respectively. Np was higher for males than females. When the crop of the 6 years was pooled, Np for female, male and the clone were 73, 87 and 85, respectively. The impact of female-male fertility correlation for conditions with no-, positive- and negative-correlations were assessed and their impact on OT, Np and Ne(v) was also evaluated. It was demonstrated that the practice of equal seed harvesting from every clone, or the mixing of seeds from several years, would substantially improve the genetic diversity and the genetic representation of the seed orchard population when a positive correlation between gender fertilities was observed. The relevance of these results to supplemental-mass-pollination was discussed under two cases where equal- and un-equal amounts of pollen from clones were included in the pollen mixes.  相似文献

Studies of genetic variation can elucidate the structure of present and past populations as well as the genetic basis of the phenotypic variability of species. Taxodium distichum is a coniferous tree dominant in lowland river flood plains and swamps of the southeastern USA which exhibits morphological variability and adaption to stressful habitats. This study provides a survey of the Mississippi River Alluvial Valley (MAV) and Florida to elucidate their population structure and the extent of genetic differentiation between the two regions and sympatric varieties, including bald cypress (var. distichum) and pond cypress (var. imbricatum). We determined the genotypes of 12 simple sequence repeat loci totaling 444 adult individuals from 18 natural populations. Bayesian clustering analysis revealed high levels of differentiation between the MAV and the Florida regions. Within the MAV region, there was a significant correlation between genetic and geographical distances. In addition, we found that there was almost no genetic differentiation between the varieties. Most genetic variation was found within individuals (76.73?%), 1.67?% among individuals within population, 15.36?% among populations within the regions, and 9.23?% between regions within the variety. Our results suggest that (1) the populations of the MAV and the Florida regions are divided into two major genetic groups, which might originate from different glacial refugia, and (2) the patterns of genetic differentiation and phenotypic differentiation were not parallel in this species.  相似文献

Annual and short-lived perennial plant performance during wet years is important for long-term persistence in the Mojave Desert. Additionally, the effects of elevated CO2 on desert plants may be relatively greater during years of high resource availability compared to dry years. Therefore, during an El Niño year at the Nevada Desert FACE Facility (a whole-ecosystem CO2 manipulation), we characterized photosynthetic investment (by assimilation rate-internal CO2 concentration relationships) and evaluated the seasonal pattern of net photosynthesis (Anet) and stomatal conductance (gs) for an invasive annual grass, Bromus madritensis ssp. rubens and a native herbaceous perennial, Eriogonum inflatum. Prior to and following flowering, Bromus showed consistent increases in both the maximum rate of carboxylation by Rubisco (VCmax) and the light-saturated rate of electron flow (Jmax) at elevated CO2. This resulted in greater Anet at elevated CO2 throughout most of the life cycle and a decrease in the seasonal decline of maximum midday Anet upon flowering as compared to ambient CO2. Eriogonum showed significant photosynthetic down-regulation to elevated CO2 late in the season, but the overall pattern of maximum midday Anet was not altered with respect to phenology. For Eriogonum, this resulted in similar levels of Anet on a leaf area basis as the season progressed between CO2 treatments, but greater photosynthetic activity over a typical diurnal period. While gs did not consistently vary with CO2 in Bromus, it did decrease in Eriogonum at elevated CO2 throughout much of the season. Since the biomass of both plants increased significantly at elevated CO2, these patterns of gas exchange highlight the differential mechanisms for increased plant growth. The species-specific interaction between CO2 and phenology in different growth forms suggests that important plant strategies may be altered by elevated CO2 in natural settings. These results indicate the importance of evaluating the effects of elevated CO2 at all life cycle stages to better understand the effects of elevated CO2 on whole-plant performance in natural ecosystems.  相似文献

Four hybrids (4 F1s) were chosen out of crosses in the urdbean [Vigna mungo (L.) Hepper, 2n = 22] having contrasting morphological characters. Zymograms for isozyme peroxidase were drawn from the patterns obtained from parents and their respective F1 hybrids on the basis of relative similarities to parental bands. The selfed or crossed nature of hybrid pods was determined from the zymograms and their analysis. The number of bands and their intensities gave an idea about the extent of crossing in F1 populations. Genetic identity (I) values were indicative of their selfed nature. Dendrograms were constructed on the basis of genetic identity values to display the relative similarities between the populations. Analysis was based on individual pods to confirm their hybrid or selfed nature. Possible use of this technique for identification of F1 pods and elimination of selfed pods might be implemented to shorten the breeding operations during crossing.  相似文献

Understanding levels of connectivity among scleractinian coral populations over a range of temporal and spatial scales is vital for managing tropical coral reef ecosystems. Here, we use multilocus microsatellite genotypes to assess the spatial genetic structure of two molecular operational taxonomic units (MOTUs, types α and β) of the widespread coral Pocillopora damicornis on the Great Barrier Reef (GBR) and infer the extent of connectivity on spatial scales spanning from local habitat types to latitudinal sectors of the GBR. We found high genetic similarities over large spatial scales spanning > 1000 km from the northern to the southern GBR, but also strong genetic differentiation at local scales in both MOTUs. The presence of a considerable number of first‐generation migrants within the populations sampled (12% and 27% for types α and β, respectively) suggests that genetic differentiation over small spatial scales is probably a consequence of stochastic recruitment from different genetic pools into recently opened up spaces on the reef, for example, following major disturbance events. We explain high genetic similarity among populations over hundreds of kilometres by long competency periods of brooded zooxanthellate larvae and multiple larval release events each year, combined with strong longshore currents typical along the GBR. The lack of genetic evidence for predominantly clonal reproduction in adult populations of P. damicornis, which broods predominantly asexually produced larvae, further undermines the paradigm that brooded larvae settle close to parent colonies shortly after the release.  相似文献

The use of a thermosensitive genic male sterility (TGMS) system in two-line hybrid rice breeding is affected greatly by the sterility instability of TGMS lines caused by temperature fluctuation beyond their critical temperatures for fertility reversion. To prevent seed production from self contamination, we have developed a system to secure seed purity using a herbicide-sensitive TGMS mutant, M8077S, obtained by radiation. Genetic analysis, using the F1, F2 and F3 populations derived from this mutant and other normal varieties, revealed that bentazon lethality/sensitivity was controlled by a single recessive gene, which was named bel. The mutant can be killed at the seedling stage by bentazon at 300 mg/l or higher, a dosage that is safe for its F1 hybrids and all other normal varieties. This mutant is also sensitive to all the tested sulfonylurea herbicides. Response of segregating plants to these two types of herbicide indicated that sulfonylurea sensitivity was also controlled by bel. By crossing this mutant with Pei-Ai 64S, an F2 population was developed for genetic mapping. Surveying the two DNA pools from sensitive and non-sensitive F2 plants identified four markers that were polymorphic between the pools. The putative linked markers were then confirmed with the F2 population. The bel locus was located on chromosome 3, 7.1 cM from the closest microsatellite marker RM168. Phenotypic analysis indicated that the bel gene had no negative effect on agronomic traits in either a homozygous or heterozygous status. The mutant M8077S is valuable in the development of a TGMS breeding system for preventing impurity resulting from temperature fluctuation of the TGMS. Several two-line hybrid rice crosses using this system are under development.  相似文献

The responses of Quercus robur (oak) and Fagus sylvatica (beech) seedlings to four different light environments (full, 50%, 40% and 15% sunlight) and to a rapid increase in irradiance were explored during the summer, after 2 years of growth in a forest nursery at Nancy (France). Significant differences between the two species were found for most variables. Phenotypic plasticity for morphological variables (root-shoot ratio, leaf size, leaf weight ratio) was higher in beech than in oak, while the reverse was true for anatomical (stomatal density, epidermis thickness, exchange surface area of the palisade parenchyma) and physiological (maximum photosynthetic rate, stomatal conductance, Rubisco activity) variables. Predawn photochemical efficiency (Fv/Fm) was higher in oak than in beech in all light environments except in 15% sunlight. Fv/Fm was significantly lower in 100% sunlight than in the other light environments in beech but not in oak. Maximum photosynthetic rates (Amax) increased with increasing light availability in the two species but they were always higher in oak than in beech. Oak exhibited higher Rubisco activity than beech in full sunlight. The transfer of shade-adapted seedlings to the open caused a decrease of Fv/Fm, which was larger for beech than for oak. Transferred oak but not beech plants recovered gradually to the control Fv/Fm values. The decreased chlorophyll content and the increased non-photochemical quenching observed in high-light beech seedlings were not enough to avoid photoinhibition. The results suggest that a greater tolerance of strong irradiance is linked to an enhanced physiological plasticity (variables related to photosynthesis), while shade tolerance relies on an enhanced plasticity in light-harvesting variables (crown morphology and chlorophyll content).  相似文献

An earliness per se gene, designated Eps-Am1, was mapped in diploid wheat in F2 and single-seed descent mapping populations from the cross between cultivated (DV92) and wild (G3116) Triticum monococcum accessions. A QTL with a peak on RFLP loci Xcdo393 and Xwg241, the most distal markers on the long arm of chromosome 1Am, explained 47% of the variation in heading date (LOD score 8.3). Progeny tests for the two F2:3 families with critical recombination events between Xcdo393 and Xwg241 showed that the gene was distal to Xcdo393 and linked to Xwg241. Progeny tests and replicated experiments with line #3 suggested that Eps-Am1 was distal to Xwg241. This gene showed a large effect on heading date in the controlled environment experiments, and a smaller, but significant, effect under natural conditions. Eps-Am1 showed significant epistatic interactions with photoperiod and vernalization treatments, suggesting that the different classes of genes affecting heading date interact as part of a complex network that controls the timing of flowering induction. Besides its interactions with other genes affecting heading date, Eps-Am1 showed a significant interaction with temperature. The effect of temperature was larger in plants carrying the DV92 allele for late flowering than in those carrying the G3116 allele for early flowering. Average differences in heading date between the experiments performed at 16 °C and 23 °C were approximately 11 days (P < 0.001) for the lines carrying the Eps-Am1 allele for early flowering but approximately 50 days (P < 0.0001) for the lines carrying the allele for late flowering. The large differences in heading time (average 80 days) observed between plants carrying the G3116 and DV92 alleles when grown at 16 °C, suggest that it would be possible to produce very detailed maps for this gene to facilitate its future positional cloning.  相似文献

Habitat loss and fragmentation can influence the genetic structure of biological populations. We studied the genetic consequences of habitat fragmentation in Florida black bear (Ursus americanus floridanus) populations. Genetic samples were collected from 339 bears, representing nine populations. Bears were genotyped for 12 microsatellite loci to estimate genetic variation and to characterize genetic structure. None of the nine study populations deviated from Hardy–Weinberg equilibrium. Genetic variation, quantified by mean expected heterozygosity (H E), ranged from 0.27 to 0.71 and was substantially lower in smaller and less connected populations. High levels of genetic differentiation among populations (global F ST = 0.224; global R ST = 0.245) suggest that fragmentation of once contiguous habitat has resulted in genetically distinct populations. There was no isolation-by-distance relationship among Florida black bear populations, likely because of barriers to gene flow created by habitat fragmentation and other anthropogenic disturbances. These factors resulted in genetic differentiation among populations, even those that were geographically close. Population assignment tests indicated that most individuals were genetically assigned to the population where they were sampled. Habitat fragmentation and anthropogenic barriers to movement appear to have limited the dispersal capabilities of the Florida black bear, thereby reducing gene flow among populations. Regional corridors or translocation of bears may be needed to restore historical levels of genetic variation. Our results suggest that management actions to mitigate genetic consequences of habitat fragmentation are needed to ensure long-term persistence of the Florida black bear.  相似文献

Six south Florida populations of the endangered red-cockaded woodpecker (Picoides borealis) were sampled to examine genetic diversity and population structure in the southernmost portion of the species' range relative to 14 previously sampled populations from throughout the species range. Random amplified polymorphic DNA (RAPD) analyses were used to evaluate the populations (n= 161 individuals, 13 primers, one band/primer). Results suggested that south Florida populations have significant among-population genetic differentiation (FST= 0.17, P < 0.000), although gene flow may be adequate to offset drift (Nm= 1.26). Comparison of Florida populations with others sampled indicated differentiation was less in Florida (FST for all populations = 0.21). Cluster analyses of all 20 populations did not reflect complete geographical predictions, although clustering of distant populations resulted in a significant correlation between genetic distance and geographical distance. Overall, results suggest populations in south Florida, similar to the remainder of the species, have low genetic diversity and high population fragmentation. Exact clustering of distant populations supports the ability of RAPDs to differentiate populations accurately. Our results further support past management recommendations that translocations of birds among geographically proximate populations is preferable to movement of birds between distant populations.  相似文献

Phytoplankton photosynthesis was measured during spring-summer 1991-1992 in the inner and outer part of the shallow Potter Cove, King George Island. Strong winds characterise this area. Wind-induced turbulent mixing was quantified by means of the root-mean square expected vertical displacement depth of cells in the water column, Zt. The light attenuation coefficient was used as a measure of the influence of the large amount of terrigenous particles usually present in the water column; 1% light penetration ranged between 30 and 9 m, and between 30 and 15 m for the inner and outer cove, respectively. Obvious differences between photosynthetic capacity [P*max; averages 2.6 and 0.6 µg C (µg chlorophyll-a)-1 h-1] and photosynthetic efficiency {!*; 0.073 and 0.0018 µg C (µg chlorophyll-a)-1 h-1 [(µmol m-2 s-1)-1]} values were obtained for both sites during low mixing conditions (Zt from 10 to 20 m), while no differences were found for high mixing situations (Zt>20 m). This suggests different photoacclimation of phytoplankton responses, induced by modifications of the light field, which in turn are controlled by physical forcing. Our results suggest that although in experimental work P*max can be high, wind-induced mixing and low irradiance will prevent profuse phytoplankton development in the area.  相似文献

利用SRAP (Sequence-related amplified polymorphism)分子标记, 对放牧干扰下的高山嵩草(Kobresia pygmaea)种群进行了遗传多样性研究, 获得了下述结果: 1) 20对SRAP引物组合共检测出448条清晰条带, 其中376条条带具有多态性, 多态位点百分率为83.93%, 随着放牧强度的增加, 高山嵩草种群多态位点百分数、Nei’s遗传多样性指数、Shannon信息指数均下降。2)高山嵩草种群具有较高的遗传多样性和较低的遗传分化(总的遗传多样性Ht为0.276 6, 种群内遗传多样性Hs为0.243 6, 遗传分化系数Gst为0.119 4, 基于Gst估计的基因流Nm*为1.843 4), 但随着放牧强度的增加, Gst增加, Nm*降低, 说明放牧限制了种群间的基因交流, 使种群发生遗传分化。3)不同放牧梯度的高山嵩草种群间的遗传距离很小, 但是随着放牧强度的增加, 种群间的遗传距离逐渐增加, 遗传一致度降低。根据遗传距离所构建的UPGMA聚类图中高山嵩草4个种群随着牧压的增加, 逐级聚在一起。  相似文献

The genus Amaranthus includes several important monoecious and dioecious weed species, and several populations of these species have developed resistance to herbicides. These species are closely related and two or more species often coexist in agricultural settings. Collectively, these attributes raise the concern that herbicide resistance might transfer from one weedy Amaranthus species to another. We performed research to determine if a dominant allele encoding a herbicide-insensitive form of acetolactate synthase (ALS) could be transferred from a monoecious species, A. hybridus, to a dioecious species, A. rudis. Numerous F1 hybrids were obtained from controlled crosses in a greenhouse between A. rudis and herbicide-resistant A. hybridus, and most (85%) of these hybrids were herbicide-resistant. Molecular analysis of the ALS gene was used to verify that herbicide-resistant hybrids contained both an A. rudis and an A. hybridus ALS allele. Although hybrids had greatly reduced fertility, 42 BC1 plants were obtained by backcrossing 33 hybrids with male A. rudis. Fertility was greatly restored in BC1 progeny, and numerous BC2 progeny were obtained from a second backcross to A. rudis. The herbicide-resistance allele from A. hybridus was transmitted to 50% of the BC1 progeny. The resistance allele was subsequently transmitted to and conferred herbicide resistance in 39 of 110 plants analyzed from four BC2 families. Parental species, hybrids, and BC2 progeny were compared for 2C nuclear DNA contents. The mean hybrid 2C nuclear DNA content, 1.27 pg, was equal to the average between A. rudis and A. hybridus, which had 2C DNA contents of 1.42 and 1.12 pg, respectively. The mean 2C DNA content of BC2 plants, 1.40 pg, was significantly (! < 0.01) less than that of the recurring A. rudis parent and indicated that BC2 plants were not polyploid. This report demonstrates that herbicide resistance can be acquired by A. rudis through a hybridization event with A. hybridus.  相似文献

应用ISSR 分子标记方法对采自云南的8 个居群的小桐子( Jatropha curcas) 共158 个个体进行遗传多样性分析。8 个ISSR 引物共扩增到了67 个位点, 其中61 个是多态性位点。分析结果表明: (1) 云南小桐子的遗传多样性水平很高。在物种水平上, 平均每个位点的多态位点百分率PPB = 91.04% , 有效等位基因数Ne = 1.5244, Nei′s 基因多样性指数He= 0.3070, Shannon 多样性信息指数Ho = 0.4618; 在居群水平上, PPB = 55.04%, Ne = 1.3826, He = 0.2171, Shannon 多样性信息指数Ho = 0.3178。(2) 居群间的遗传分化低于居群内的遗传分化。基于Nei''s 遗传多样性分析得出的居群间遗传多样性分化系数Gst = 0.2944。AMOVA分析显示: 云南小桐子的遗传变异主要存在于居群内, 占总变异的63.50%, 居群间的遗传变异占36.50%。(3) 居群间的地理距离及遗传一致度并不存在相关性。鉴于以上指标, 我们推测云南小桐子可能来自不同的地区。  相似文献

The rice heterochronic gene plastochron1, pla1, shows shorter plastochron and ectopic expression of the vegetative program during the rice reproductive phase resulting in aberrant panicle formation. A genetic and physical map was constructed to isolate the causal gene for the pla1 syndrome. Small-scale mapping was carried out to determine the approximate map position of the pla1 locus, and then a high-resolution genetic map was made for pla1-1, one of the pla1 alleles, using an F2 population comprising 578 pla1-1 homozygous plants. In a high-resolution genetic map, the pla1-1 locus was found to map between RFLP markers C961 and R1738A on chromosome 10, within a 3.6-cM genetic distance. A physical map encompassing the pla1-1 locus was constructed by overlapping Bacterial Artificial Chromosome (BAC) clones through chromosome walking. PCR-based RFLP markers from BAC-end clones were developed and mapped relative to the pla1 locus. Physical map construction using BAC clones indicated that a BAC clone, B44A10 (167-kb), contained the pla1 locus within 74-kb corresponding to a 0.52-cM genetic distance. Gene prediction of 74-kb region carrying the pla1 locus suggested several candidate genes for the pla1 gene. Identification of a candidate gene for pla1 will be made by sequence analysis of allele variation and cDNA screening.  相似文献

The photosynthetic induction response under constant and fluctuating light was examined in naturally occurring saplings (about 0.5-2 m in height) of three shade-tolerant tree species, Pourouma bicolor spp digitata, Dicorynia guianensis, and Vouacapoua americana, growing in bright gaps and in the shaded understorey in a Neotropical rain forest. Light availability to saplings was estimated by hemispherical photography. Photosynthetic induction was measured in the morning on leaves that had not yet experienced direct sunlight. In Dicorynia, the maximum net photosynthesis rate (Amax) was similar between forest environments (ca 4 µmol m-2 s-1), whereas for the two other species, it was twice as high in gaps (ca 7.5) as in the understorey (ca 4.5). However, the time required to reach 90% of Amax did not differ among species, and was short, 7-11 min. Biochemical induction was fast in leaves of Pourouma, as about 3 min were needed to reach 75% of maximum carboxylation capacity (Vcmax); the two other species needed 4-5 min. When induction continued after reaching 75% of Vcmax, stomatal conductance increased in Pourouma only (ca 80%), causing a further increase in its net photosynthesis rate. When fully induced leaves were shaded for 20 min, loss of induction was moderate in all species. However, gap saplings of Dicorynia had a rapid induction loss (ca 80%), which was mainly due to biochemical limitation as stomatal conductance decreased only slowly. When leaves were exposed to a series of lightflecks separated by short periods of low light, photosynthetic induction increased substantially and to a similar extent in all species. Although Amax was much lower in old than in young leaves as measured in Dicorynia and Vouacapoua, variables of the dynamic response of photosynthesis to a change in light tended to be similar between young and old leaves. Old leaves, therefore, might remain important for whole-plant carbon gain, especially in understorey environments. The three shade-tolerant species show that, particularly in low light, they are capable of efficient sunfleck utilization.  相似文献
