Halotolerant yeast, Pichia farinosa, is a valuable yeast strain in fermentation industry because it produces high yield of glycerol and xylitol, and can tolerate both contamination and high-density growth during fermentation. However, the lack of genetic manipulation tools makes it less popular as a gene engineering strain. Expression systems commonly used in other yeast systems, such as Saccharomyces cerevisiae and Pichia pastoris cannot be used in P. farinosa because it translates universal Leu codon CUG as Ser. Here we reported a modified expression vector and a transformation system with enhanced efficiency in P. farinosa. The results showed that cells of OD600 0.8–1.0 with DTT treatment can obtain high transformation efficiency. The optimized electroporation condition was 900 V, 25 μF, and 200 Ω. The DNA concentration did not influence the transformation. Our system provides the potential not only for applying P. farinosa as an industrial strain of gene engineering, but also for studying gene function in its native host.  相似文献

Strophomenid brachiopods belonging to the generaLeptaena andLepidoleptaena are described from the uppermost Llandovery — Ludlow succession of Gotland, Sweden. In Gotland,Lepidoleptaena comprises the single speciesL. poulseni, andLeptaena includes four species:L. rhomboidalis, L. sperion, L. depressa andL. parvirugata n. sp.L. depressa shows a considerable amount of morphological variation, which is recognised in the two new subspeciesLeptaena depressa visbyensis n. ssp. from the Llandoverian — Wenlockian Visby Formation, andL. depressa lata n. ssp. from the mid-Wenlockian Slite Group. The distribution of the different species is largely substrate-dependent, withLeptaena rhomboidalis andLepidoleptaena poulseni adapted to high-energy environments with firm substrates, whereas the remaining species preferred fine-grained substrates in low-energy environments.   相似文献

Twenty isolates of four species of entomopathogenic hypocrealean fungi (Beauveria bassiana, Beauveria brongniartii, Isaria farinosa, and Metarhizium anisopliae) were found to be pathogenic to adults of the Asian longhorned beetle, Anoplophora glabripennis. Survival times for 50% of the beetles tested (ST50) ranged from 5.0 (M. anisopliae ARSEF 7234 and B. brongniartii ARSEF 6827) to 24.5 (I. farinosa ARSEF 8411) days. Screening studies initially included strains of B. brongniartii, which is registered as a microbial control agent in Europe, Asia and South America but not in North America. At that time, we could not confirm that this fungal species is native to North America which added uncertainty regarding future registration of this species for pest control in the USA. Therefore, subsequent bioassays documented median survival times for three M. anisopliae isolates (5–6 days to death) and two of these isolates are suggested for further development because they are already registered for pest control in the USA. Cenomanian-Turonian sediments from the Sarbay locality in north-western Kazakhstan have yielded a rich assemblage of plant fossils including well preserved flowers, fruits, and seeds of angiosperms. This work describes fossil seeds assigned to theMagnoliaceae and theIlliciaceae. Three new species of the extinct magnoliaceous genusLiriodendroidea, L. asiatica, L. costata, andL. tenuitesta, are established and new information on the previously described species,L. alata, is provided. TheLiriodendroidea seeds are closely similar to seeds of extantLiriodendron, but are distinguished in being much smaller and winged. A new genus and species,Illiciospermum pusillum, is established based on seeds with close similarity to those of the extant genusIllicium. The seeds are small, anatropous and exotestal with outer epidermis of testa forming a palisade layer. The facets of the palisade cells have deeply undulate anticlinal walls. The micropyle area is seen on the outer integument as a transverse slit placed on a raised strophiole-like structure close to the hilum. TheIlliciospermum seeds represent the first unequivocal record of theIlliciaceae in the Cretaceous. Another seed of possible illiciaceous affinity is described as aff.Illiciospermum sp.  相似文献

Six carabidicolous species of Laboulbeniales are reported as new for the Japanese mycoflora. They areLaboulbenia finitima, L. separata, L. borneensis, L. manubriolata, Dimeromyces caribaeus, andDixomyces ornatus. In Japan, the former three species are parasitic onMochtherus luctuosus and the latter three species onPerigona nigriceps. Laboulbenia finitima occurs on the left legs (tarsi and tibiae);L. separata on the left elytral margin:L. borneensis on the pronotum;L. manubriolata on the pronotum and elytra;Dimeromyces caribaeus on the right inferior posterior surface of the prothorax (pronotal hypomeron); andDixomyces ornatus on the outer margin of the right elytron. Comments and photographs are given for each fungus.  相似文献

Six carabidicolous species of Laboulbeniales are reported as new for the Japanese mycoflora. They areDixomyces stomonaxi, Laboulbenia picardii, L. tenera, L. slackensis, L. aristata andL. kwangjuensis. Two forms ofD. stomonaxi are distinguished, one form of which resemblesD. nigromarginatus. A remarkable ornamentation consisting of a coillike pattern occurs on the receptacle ofL. picardii. InL. tenera andL. slackensis, the outer appendage has somewhat constricted, blackened septa near the base.Laboulbenia aristata andL. kwangjuensis have spirally arranged outer wall cells. Antheridia were observed inL. tenera, L. slackensis, L. aristata andL. kwangjuensis.  相似文献

C-band patterns are described for 20Lilium spp. distributed across six sections. All species have a similar basic karyotype (n = 12) but C-bands differ markedly between them. The patterns are characterized by a dispersed scattering of thin intercalary bands as well as centric and NOR bands. Only one species,L. canadense, shows a clear equilocal pattern with intercalary C-bands occurring proximally in all of the longer chromosome arms. Comparing species, similar patterns are revealed forL. regale andL. sulphureum, forL. formosanum andL. longiflorum (all in sect.Leucolirion) and to a lesser extent forL. hansonii, L. martagon, andL. tsingtauense (sect.Martagon). The pattern forL. henryi (previously classed in sect.Sinomartagon) matches those ofL. regale andL. sulphureum quite well and its transfer to sect.Leucolirion is proposed. This is consistent with results from interspecies hybrids betweenL. henryi andL. regale (and related species) which are reportedly fertile. No other clear similarities in C-band patterns were seen across species. It seems that C-band patterns change rapidly inLilium and hence their usefulness in classification will be restricted to identifying closely related species.Dedicated to Prof.D. G. Catcheside on the 80th anniversary of his birth.  相似文献

The taxonomic positions ofRetzia, Desfontainia, andNicodemia have been much discussed, and all three genera have been included inLoganiaceae (Gentianales). We have made a cladistic analysis ofrbcL gene sequences to determine the relationships of these taxa toGentianales. Four newrbcL sequences are presented; i.e., ofRetzia, Desfontainia, Diervilla (Caprifoliaceae), andEuthystachys (Stilbaceae). Our results show thatRetzia, Desfontainia, andNicodemia are not closely related toLoganiaceae or theGentianales. Retzia is most closely related toEuthystachys and is better included inStilbaceae. The positions ofDesfontainia andNicodemia are not settled, butDesfontainia shows affinity for theDipsacales s.l. andNicodemia for theLamiales s.l.  相似文献

In 83 species of the familiesMonotropaceae, Apocynaceae, Oleaceae, Scrophulariaceae, Lentibulariaceae, Bignoniaceae, Martyniaceae, Myoporaceae, Verbenaceae, Lamiaceae, Campanulaceae, andLiliaceae, protein bodies in the cell nuclei have been found, in 68 of these species for the first time. On the basis of their structure in accordance with morphological characters the generaBurgsdorfia, Hesiodia, Olisia, andPhlomoides of theLamiaceae are accepted;Lamium is divided intoLamium, Lamiastrum andOrvala; two new combinations are established:Kickxia campyloceras (Rech. fil. &Esfandiari)Speta andEtornotus papilionaceus (Burm. in L.)Speta. Deviating shape or lack of protein bodies corroborate former taxonomic decisions, e.g. the transfer ofMonotropa toMonotropaceae or the separation ofGaleopsis andLadanum; Veronica schmidtiana should not be included inPseudolysimachion. Systematic affinities are discussed primarily withinScrophulariaceae because nuclear protein bodies have been studied already in many species of this family. ForCampanula patula two 2 x populations are reported.

A parsimony analysis of DNA sequences of the chloroplast-encoded generbcL from twelve members of theAcanthaceae s.l., including members of the sometimes segregateThunbergioideae andNelsonioideae, and other families in theBignoniales sensuThorne (1992) is presented. The results largely agree with the classification of theAcanthaceae presented byBremekamp (1965) andThorne (1992) and supportNelsonioideae as a sister group to the rest of theAcanthaceae. Thunbergioideae are placed as a sister toAcanthaceae s.str.Acanthus andAphelandra, both representatives ofAcanthoideae, form a sister group toRuellioideae. An analysis of branch support found that many branches throughout theBignoniales are weakly upheld. This points to the need for further studies in the group using more sequences ofrbcL as well as other data. None of the families ofBignoniales as presently circumscribed (includingAcanthaceae s.l.) were strongly supported, although the larger clade containing the families of theBignoniales was robust.  相似文献

A new taxonomic treatment is proposed for thePottia starckeana species complex. The peristome development is not considered to be a useful feature to separate the taxa. On the basis of spore morphology only two species are accepted:P. starckeana, with spores wavy in outline, andP. davalliana, with variously-shaped and developed processes on the spores.Pottia starckeana var.brachyoda is reduced to synonymy withP. starckeana; P. conica andP. commutata are treated as synonyms ofP. davalliana. The speciesP. mutica, P. affinis, P. salina, P. microphylla, P. texana, andP. arizonica (included var.mucronulata) are considered taxa of doubtful affinity, as they have spore features intermediate between the two spore types established for the group. The identity ofP. appertii andP. recurvifolia has not been elucidated because the type material has been destroyed.  相似文献

The tribal name Bocageeae Endlicher is reestablished and the tribe is circumscribed on the basis of solitary internodal ebracteate pedicels that are articulated at the base, and pollen shed in polyads of eight or more grains. Septate anther locules, large pollen size, and seed appendages are prevalent in the tribe. Intectate pollen with free-standing columellae, rare in Annonaceae, occurs not only in the genusTrigynaea but also in some species ofBocagea andHornschuchia. As defined here, the Bocageeae include seven neotropical genera:Cymbopetalum, Porcelia, Bocagea, Cardiopetalum, Froesiodendron, Hornschuchia, andTrigynaea. The latter five genera are revised and the treatments include ten new species:Cardiopetalum plicatum, Froesiodendron urceocalyx, Hornschuchia lianarum, H. santosii, H. leptandra, Trigynaea cinnamomea, T. lanceipetala, T. triplinervis, T. lagaropoda, andT. axilliflora, all from tropical South America. A new combination,Froesiodendron longicuspe, changes the rank of that taxon from subspecies to species.Cardiopetalum surinamense is removed fromFroesiodendron and reassigned toCardiopetalum on the basis of its connate petals, dehiscent monocarps, and seeds with bilobed arils. A cladogram provides an explicit hypothesis of intergeneric relationships in the tribe. The new combinationOnychopetalum periquino, based onTrigynaea periquino, is made.  相似文献

The degree of relationships withinLactuca sativa and three wild relativesL. serriola, L. saligna, andL. virosa was studied by observing the performance, vigour and fertility of the F 1 hybrids obtained from crosses made in and between the four species. The crosses ofL. saligna ×L. virosa and the reciprocal crosses produced no hybrids.L. saligna andL. virosa are the least related of the four species.L. sativa ×L. serriola and the reciprocal crosses were successful and produced fertile hybrids These two species are genetically very closely related.L. saligna is known to produce, as a female parent, hybrids withL. sativa andL. serriola. Now the reciprocal cross was successful for the first time, so the unability to obtain hybrids in the past was based on the choice of accessions and not caused by unilateral incompatibility.L. virosa ×L. sativa and the reciprocal combination produced hybrids. The combinationL. serriola ×L. virosa produced hybrids with very limited fertility. In contrast to earlier reports (sterile hybrids) one combination of the reciprocal cross too produced hybrids with very limited fertility.—Some of theL. saligna ×L. sativa (and reciprocal) hybrids were found to look strikingly likeL. serriola. This adds evidence for the descent ofL. serriola andL. sativa:L. saligna also made part of the ancestral complex of the cultivated lettuce.  相似文献

Soils from various areas in Japan were screened for isolation of cleistothecial ascomycetes. From the recent collections, two new species of onygenalean fungi,Gymnascella japonica andGymnostellatospora frigida, are described and illustrated. Three interesting fungi,Aphanoascus saturnoideus, Arachnotheca glomerata andLeucothecium emdenii, are also described as the first records from Japan.  相似文献
