The involvement of ethylene in zygotic embryogenesis is a little known aspect of the growth and development in higher plants. In the present work, we study the alterations of the last step of the ethylene biosynthesis pathway during the formation period of turnip tops ( Brassica rapa cv. Rapa) seeds and its repercussions in the germination process and post-germinative growth. For this, we chose 11 different phases of silique development, the first being the recently fertilized pistil and the last being the silique just prior to its dehiscence (ca. 2 months post-anthesis). In the 11 phases, ethylene production was detected in both whole silique (with or without seeds) and in the seeds enclosed by the silique wall. The levels of ACC, ACO and ethylene production proved high in seeds belonging to: (1) the pod in the very early phases, when the seeds were growing but without photosynthetic competence; (2) the silique at maximum growth, in which the seeds will initiate desiccation and loss of photosynthetic activity. During the phases prior to dehiscence, there was a marked inhibition in the last step of the ethylene biosynthesis pathway. In viable dry seeds, no ACO activity was detected and the ACC levels were 4-fold lower than at the onset of the silique senescence. Germination brings about a net synthesis of ACC with respect of the stores dry seed. This fact, together with other results presented in this work, point towards, as in other seeds, a dependence of ethylene synthesis for radicle emergence. The possible role played by the silique wall in the control of ethylene biosynthesis during zygotic embryogenesis, as well as the participation of ethylene as a hormonal signal in the triggering of seed desiccation in Brassica rapa cv. Several hydrolytic enzyme activities were detected in the wall of developing cells of Rubus truticosus in suspension culture. The corresponding substrates of the enzymes are mostly polysaccharide wall constituents, except for chitinase activity. The activities measured when the enzymes were in the free state or wall-bound showed the positive influence of the cell wall micro-environment. Changes in the activities during a cell culture cycle demonstrated that those enzymes acting on xyloglucans behaved differently from the others, and suggest that xyloglucans undergo modifications in vivo over a longer period of time during the exponential growth phase. The same activities were identified in the culture medium. Endo-1,4-β-d-glucanase activities which depolymerized car☐ymethylcellulose (CMC) and xyloglucans (XG) were assayed viscosimetrically. It was found that XG oligosaccharides exhibited an inhibitory effect on the depolymerization of xyloglucans but not on that of CMC. This suggests that true xyloglucanases are present in the culture of Rubus cells.  相似文献

Abstract: The plant cell wall is a dynamic apparatus responsible for both morphogenesis and responsiveness to environmental conditions. In the cell wall of most seed plants, cellulose microfibrils are cross-linked by xyloglucans to form a cellulose/xyloglucan framework, which functions as the mechanical underpinning of the cell wall. Endoxyloglucan transferases are a class of enzymes that play a central role in construction and modification of the plant cell wall. These enzymes are encoded by a large multi-gene family termed xyloglucan-related proteins (XRPs). More than 24 members of the XRP family have so far been identified in Arabidopsis thaliana. Each member of this family functions as either a hydrolase or a transferase acting on xyloglucans. The primary structures of proteins and gene-expression profiles have strongly suggested their potentially divergent roles in plant morphogenesis: different members of this family are expressed in different types of tissues at distinct developmental stages and respond differentially to individual hormones as well as environmental stimuli. These facts imply that each member of this gene family is individually committed to a specific process that proceeds in a specific tissue at a specific stage of development. Probably the generation and maintenance of the cell walls in a whole organ, and thus in the whole plant, is achieved by the ensemble of individual members of the XRP family.  相似文献

Background and Aims

Although xyloglucans are ubiquitous in land plants, they are less abundant in Poales species than in eudicotyledons. Poales cell walls contain higher levels of β-1,3/1,4 mixed-linked glucans and arabinoxylans than xyloglucans. Despite the relatively low level of xyloglucans in Poales, the xyloglucan endotransglucosylase/hydrolase (XTH) gene family in rice (Oryza sativa) is comparable in size to that of the eudicotyledon Arabidopsis thaliana. This raises the question of whether xyloglucan is a substrate for rice XTH gene products, whose enzyme activity remains largely uncharacterized.

Methods

This study focused on OsXTH19 (which belongs to Group IIIA of the XTH family and is specifically expressed in growing tissues of rice shoots), and two other XTHs, OsXTH11 (Group I/II) and OsXTH20 (Group IIIA), for reference, and measurements were made of the enzymatic activities of three recombinant rice XTHs, i.e. OsXTH11, OsXTH20 and OsXTH19.

Key Results

All three OsXTH gene products have xyloglucan endohydrolase (XEH, EC 3·2·1·151) activity, and OsXTH11 has both XEH and xyloglucan endotransglycosylase (XET, EC 2·4·1207) activities. However, these proteins had neither hydrolase nor transglucosylase activity when glucuronoarabinoxylan or mixed-linkage glucan was used as the substrate. These results are consistent with histological observations demonstrating that pOsXTH19::GUS is expressed specifically in the vicinity of tissues where xyloglucan immunoreactivity is present. Transgenic rice lines over-expressing OsXTH19 (harbouring a Cauliflower Mosaic Virus 35S promoter::OsXTH19 cDNA construct) or with suppressed OsXTH19 expression (harbouring a pOsXTH19 RNAi construct) did not show dramatic phenotypic changes, suggesting functional redundancy and collaboration among XTH family members, as was observed in A. thaliana.

Conclusions

OsXTH20 and OsXTH19 act as hydrolases exclusively on xyloglucan, while OsXTH11 exhibits both hydrolase and XET activities exclusively on xyloglucans. Phenotypic analysis of transgenic lines with altered expression of OsXTH19 suggests that OsXTH19 and related XTH(s) play redundant roles in rice growth.  相似文献

Xyloglucan endotransglucosylase/hydrolase (XTHs: EC 2.4.1.207 and/or EC 3.2.1.151), a xyloglucan modifying enzyme, has been proposed to have a role during tomato and apple fruit ripening by loosening the cell wall. Since the ripening of climacteric fruits is controlled by endogenous ethylene biosynthesis, we wanted to study whether XET activity was ethylene-regulated, and if so, which specific genes encoding ripening-regulated XTH genes were indeed ethylene-regulated. XET specific activity in tomato and apple fruits was significantly increased by the ethylene treatment, as compared with the control fruits, suggesting an increase in the XTH gene expression induced by ethylene. The 25 SlXTH protein sequences of tomato and the 11 sequences MdXTH of apple were phylogenetically analyzed and grouped into three major clades. The SlXTHs genes with highest expression during ripening were SlXTH5 and SlXTH8 from Group III-B, and in apple MdXTH2, from Group II, and MdXTH10, and MdXTH11 from Group III-B. Ethylene was involved in the regulation of the expression of different SlXTH and MdXTH genes during ripening. In tomato fruit fifteen different SlXTH genes showed an increase in expression after ethylene treatment, and the SlXTHs that were ripening associated were also ethylene dependent, and belong to Group III-B (SlXTH5 and SlXTH8). In apple fruit, three MdXTH showed an increase in expression after the ethylene treatment and the only MdXTH that was ripening associated and ethylene dependent was MdXTH10 from Group III-B. The results indicate that XTH may play an important role in fruit ripening and a possible relationship between XTHs from Group III-B and fruit ripening, and ethylene regulation is suggested.  相似文献

Flowering plants usually require fertilization to form fruit and seed and to initiate floral organ abscission in structures that do not contribute to the fruit. An Arabidopsis mutant that initiates seedless fruit without fertilization (fwf) or parthenocarpy was isolated and characterized to understand the factors regulating the transition between the mature flower and the initiation of seed and fruit development. The fwf mutant is fertile and has normal plant growth and stature. It sets fertile seed following self-pollination and fertilization needs to be prevented to observe parthenocarpy. The initiation of parthenocarpic siliques (fruit) was found to be dependent upon carpel valve identity conferred by FRUITFULL but was independent of the perception of gibberellic acid, shown to stimulate parthenocarpy in Arabidopsis following exogenous application. The recessive nature of fwf is consistent with the involvement of FWF in processes that inhibit fruit growth and differentiation in the absence of fertilization. The enhanced cell division and expansion in the silique mesocarp layer, and increased lateral vascular bundle development imply FWF has roles also in modulating silique growth post-fertilization. Parthenocarpy was inhibited by the presence of other floral organs suggesting that both functional FWF activity and inter-organ communication act in concert to prevent fruit initiation in the absence of fertilization.  相似文献

Background

Mannans are hemicellulosic polysaccharides in the plant primary cell wall with two major physiological roles: as storage polysaccharides that provide energy for the growing seedling; and as structural components of the hemicellulose–cellulose network with a similar function to xyloglucans. Endo-β-mannanases are hydrolytic enzymes that cleave the mannan backbone. They are active during seed germination and during processes of growth or senescence. The recent discovery that endo-β-mannanase LeMAN4a from ripe tomato fruit also has mannan transglycosylase activity requires the role of endo-β-mannanases to be reinterpreted.

Aims

In this review, the role of endo-β-mannanases as mannan endotransglycosylase/hydrolases (MTHs) in remodelling the plant cell wall is considered by analogy to the role of xyloglucan endotransglucosylase/hydrolases (XTHs). The current understanding of the reaction mechanism of these enzymes, their three-dimensional protein structure, their substrates and their genes are reported.

Future outlook

There are likely to be more endohydrolases within the plant cell wall that can carry out hydrolysis and transglycosylation reactions. The challenge will be to demonstrate that the transglycosylation activities shown in vitro also exist in vivo and to validate a role for transglycosylation reactions during the growth and development of the plant cell wall.Key words: Cell wall, endo-β-mannanase, endohydrolase, mannan, endotransglycosylase  相似文献

Auxin-induced elongation of epicotyl segments of azuki bean ( Vigna angularis Ohwi and Ohashi cv. Takara) was suppressed by fucose-binding lectins from Tetragonolobus purpureus Moench and Ulex europaeus L. These lectins also inhibited auxin-induced cell wall loosening (decrease in the minimum stress-relaxation time of the cell walls) of segments. Auxin caused a decrease in molecular mass of xyloglucans extracted with 24% KOH from the cell walls. The lectins inhibited auxin-induced changes in molecular mass of the xyloglucans. The autolytic release of xylose-containing products from the pectinase-treated cell walls was also suppressed by the lectins. Fucose-binding lectins pretreated with fucose exhibited little or no inhibitory effect on auxin-induced elongation, cell wall loosning, or breakdown of xyloglucans. These results support the view that the breakdown of xyloglucans is involved in the cell wall loosening responsible for auxin-induced elongation in dicotyledons.  相似文献

The involvement of anti-gravitational polysaccharides in gravity resistance, one of two major gravity responses in plants, was discussed. In dicotyledons, xyloglucans are the only cell wall polysaccharides, whose level, molecular size, and metabolic turnover were modified under both hypergravity and microgravity conditions, suggesting that xyloglucans act as anti-gravitational polysaccharides. In monocotyledonous Poaceae, (1-->3),(1-->4)-beta glucans, instead of xyloglucans, were shown to play a role as anti-gravitational polysaccharides. These polysaccharides are also involved in plant responses to other environmental factors, such as light and temperature, and to some phytohormones, such as auxin and ethylene. Thus, the type of anti-gravitational polysaccharides is different between dicotyledons and Poaceae, but such polysaccharides are universally involved in plant responses to environmental and hormonal signals. In gravity resistance, the gravity signal may be received by the plasma membrane mechanoreceptors, transformed and transduced within each cell, and then may modify the processes of synthesis and secretion of the anti-gravitational polysaccharides and the cell wall enzymes responsible for their degradation, as well as the apoplastic pH, leading to the cell wall reinforcement. A series of events inducing gravity resistance are quite independent of those leading to gravitropism.  相似文献

Green and red tomato pericarp tissues were subjected to stress-relaxation analyses to evaluate their physical properties. Significant decreases in the initial stress, minimum stress-relaxation and maximum stress-relaxation times in the red tissues predict the losses of both viscosity and elasticity in the tissue. Cell walls of red fruit yielded more water-soluble polysaccharides and less pectin, hemicelluloses and cellulose. Average molecular mass of pectin determined by gel filtration chromatography was similar in the green and red, but molecular mass of hemicellulose of red fruit walls was reduced to 50% of that of the green fruit. The decreases in the amount of hemicellulose B and in the average molecular mass were associated primarily with the degradation of xylo-glucans. These data demonstrate that pectin solubilization, depolymerization of xyloglucans and over-all changes in the quantity of cell wall polysaccharide fractions contribute to tomato fruit softening.  相似文献

Hemicellulosic polysaccharides from persimmon fruit ( Diospyros kaki L.) pericarp were extracted from depectinated cell walls with 0.5, 1 and 4 M KOH at different stages of development: (I) maximal growth corresponding to the first sigmoidal growth phase; (II) cessation of growth corresponding to the lag between the first and the second sigmoidal phases; (III) maximal growth corresponding to the second sigmoidal phase; and (IV) cessation of growth when the fruit had reached its maximum size and the change in colour (green to red) had taken place. During fruit development the amount of total hemicelluloses per unit dry mass cell wall decreased twofold. Xyloglucan was present in the three hemicellulosic fractions, and also decreased with fruit age, although its amount relative to other hemicelluloses increased. The amount of xyloglucan was especially high in the hemicelluloses extracted with 4 M KOH, representing more than 50% at stages III and IV. The average molecular mass of xyloglucan increased from stage I through stage II (0.5 M hemicellulosic fraction) or through stage III (I and 4 M hemicellulosic fractions) and decreased after that. The xyloglucan endotransglycosylase (XET: EC 2.4.1.-) activity was measured as the incorporation of [3H]XXXGol (reduced xyloglucan heptasaccharide labelled at position 1 of the glucitol moiety) into partially purified persimmon fruit xyloglucan. XET specific activity increased greatly between stages I and II. The importance of this enzyme during fruit ripening is discussed.  相似文献

Effects of indole-3-acetic acid (IAA) on the mechanical properties of cell walls and structures of cell wall polysaccharides in outer and inner tissues of segments of dark grown squash (Cucurbita maxima Duch.) hypocotyls were investigated. IAA induced the elongation of unpeeled, intact segments, but had no effect on the elongation of peeled segments. IAA induced the cell wall loosening in outer tissues as studied by the stress-relaxation analysis but not in inner tissues. IAA-induced changes in the net sugar content of cell wall fractions in outer and inner tissues were very small. Extracted hemicellulosic xyloglucans derived from outer tissues had a molecular weight about two times as large as in inner tissues, and the molecular weight of xyloglucans in both outer and inner tissues decreased during incubation. IAA substantially accelerated the depolymerization of xyloglucans in outer tissues, while it prevented that in inner tissues. These results suggest that IAA-induced growth in intact segments is due to the cell wall loosening in outer tissues, and that IAA-accelerated depolymerization of hemicellulosic xyloglucans in outer tissues is involved in the cell wall loosening processes.  相似文献

The internodes of Merremia emarginata plant showed exceptionally high stretchability throughout the development period. Therefore, it provides excellent material to study the changes undergoing during cell elongation. In this study, the influence of the hormone treatments (GA3, PAA and NAA) on the wall component synthesis was analyzed in relation to elongation growth during internode development. A clear increasing trend of wall components was observed with increase in internode length. The non-esterified pectic substances were markedly correlated with internode length while esterified pectic substances showed correlation only in hormone treated internodes. Low molecular weight xyloglucans content showed correlation only in GA3 and NAA treated internodes, while high molecular weight xyloglucans were significantly correlated with length of internodes treated with PAA and NAA.  相似文献

During fruit development in tomato (Solanum lycopersicum), cell proliferation and rapid cell expansion occur after pollination. Cell wall synthesis, alteration, and degradation play important roles during early fruit formation, but cell wall composition and the extent of cell wall synthesis/degradation are poorly understood. In this study, we used immunolocalization with a range of specific monoclonal antibodies to examine the changes in cell wall composition during early fruit development in tomato. In exploring early fruit development, the ?1 day post-anthesis (DPA) ovary and fruits at 1, 3, and 5 DPA were sampled. Paraffin sections were prepared for staining and immunolabeling. The 5 DPA fruit showed rapid growth in size and an increase in both methyl-esterified pectin and de-methyl-esterified pectin content in the pericarp, suggesting rapid synthesis and de-methyl esterification of pectin during this growth period. Labeling of pectic arabinan with LM6 antibody and galactan with LM5 antibody revealed abundant amounts of both, with unique distribution patterns in the ovule and premature pericarp. These results suggest the presence of rapid pectin metabolism during the early stages of fruit development and indicate a unique distribution of pectic galactan and arabinan within the ovule, where they may be involved in embryogenesis.  相似文献

In order to gain insight into the mechanism of cell extension growth, enzymic processes involved in structural modification of cell wall xyloglucans were investigated, using an apoplastic enzyme preparation from epicotyls of dark grown Vigna angularis Ohwi et Ohashi cv. Takara and purified xyloglucans derived from cell walls of Vigna. The reaction of Vigna xyloglucan (mass average molecular weight=420 kDa) with the apoplastic enzyme preparation gave three fractions: (1) a waterinsoluble high molecular weight (820 kDa) xyloglucan fraction (WI), (2) a watersoluble low molecular weight (149 kDa) xyloglucan fraction (WS), and (3) an 80% ethanol-soluble monosaccharide fraction (ES). WI and WS were chiefly composed of t -galactosyl-, t -xylosyl-, 2-xylosyl-, 4-glucosyl- and 4,6-glucosyl residues, whereas ES was composed of fucose, galactose, glucose and xylose monomers. The data indicate that WI is generated by the linking of xyloglucan molecules by some alkali stable linkages, probably of glycosidic nature. The optimal pH for the WI-producing activity of the apoplastic enzyme preparation was 5.4. Higher WI-producing activity was detected in the upper juvenile than in the lower non-elongating regions of the epicotyl. Our data suggest the possible involvement of a transglycosylation reaction in the structural changes of the xyloglucans that are responsible for cell extension growth of the Vigna angularis epicotyl. The data are also consistent with the idea that the enzymic processes are regulated by hydrogen ions in the apoplastic space.  相似文献
