Summary In a pot experiment the effects were estimated of varying nitrogen and sulphur applications on the total and nitrate nitrogen, free amino acid and amide and total amino acid concentrations of barley plants at five early stages of growth.Variations with time in contents of total and nitrate nitrogen differed at low and high nitrogen levels. Concentrations of free amino acids and amides were influenced by light intensity preceding sampling, age of plant and by levels of nitrogen and sulphur. At the fifth harvest (55 days old plants), high nitrogen addition without sulphur caused a fivefold increase in asparagine content and lesser increases in glycine and serine. Alterations in total amino acid contents with time were largely dependent on variations in total nitrogen. Sulphur deficiency, as judged by contents of S-amino acids and amides, became more pronounced with time. A positive interaction was found between effects of nitrogen and sulphur on the yield and cystine and methionine content of grain. Absence of sulphur reduced the cystine content of grain much more than that of methionine, particularly at the high nitrogen level.  相似文献

In order to increase the concentration of the nutritionally essential sulphur amino acids in seed protein, a transgene encoding a methionine- and cysteine-rich protein, sunflower seed albumin (SSA), was transferred to chickpeas (Cicer arietinum L). Transgenic seeds that accumulated SSA contained more methionine and less oxidized sulphur than the controls, suggesting that additional demand for sulphur amino acids from the expression of the transgene stimulated sulphur assimilation. In addition, the activity of trypsin inhibitors, a known family of endogenous, sulphur-rich chickpea seed proteins, was diminished in transgenic, SSA-containing seeds compared with the non-transgenic controls. Together, these results indicate that the reduced sulphur sequestered into SSA was supplied partly by additional sulphur assimilation in the developing transgenic seeds, and partly by some diversion of sulphur amino acids from endogenous seed proteins. Growth of chickpeas on nutrient with a high sulphur-to-nitrogen ratio increased the total seed sulphur content and the accumulation of sulphur amino acids in the seeds, and partly mitigated the effect of SSA accumulation on the trypsin inhibitor amount. The results suggest that free methionine and O-acetylserine (OAS) acted as signals that modulated chickpea seed protein composition in response to the variation in sulphur demand, as well as in response to variation in the nitrogen and sulphur status of the plant.  相似文献

Summary The effect of N and S fertilizers on yield, protein content and amino acid composition of the tropical cereal finger millet was studied in field and sand culture experiments. In the field the grain yield measured from 2400 to 4100 kg/ha, and the seed crude protein from 7 to 13% with rates of N up to 150 kg/ha. Seed crude protein was high in methionine and cystine, but contained less sulphur amino acids when the seed was high in protein.Supplies in excess failed to increase the lower methionine and cystine contents of the crude protein.Sulphur deficiency drastically reduced the synthesis of sulphur amino acids, concomitantly raising the aspartic acid/asparagine content of the leaves up to 45 per cent of their total N content. An S:N weight ratio of 1:10 in the seeds and 1:15 in the leaves indicates adequate supply of sulphur. A lysimeter study was conducted on Cajeme wheat (Triticum aestivum L.) to investigate the impact of salinity on protein and free amino acid content of the grain. Cross correlations were obtained between 16 different soil-plant-water based parameters and the concentration and total accumulation of amino acids. The results indicated that after 3 years of irrigation, the majority of protein bound and free amino acids increased in concentration in the grain. However, both free tryptophan and free proline revealed decreasing concentrations with increasing salinity. Free tryptophan showed a synergism between total accumulation, yield and concentration. Free proline concentrations decreased in association with increasing protein concentrations. Cross correlations of the 16 soil-plant-water based parameters with free and protein bound amino acids revealed significant correlations for free aspartic acid and glycine with total accumulation but not with concentrations. Only methionine plus cystine was lower than suggested FAO levels for essential amino acids and was lower in the third year than in the first year.  相似文献

Three prototrophic strains of Cephalosporium acremonium with different potentials for cephalosporin C production show pronounced differences in pools of intracellular sulphur amino acids. The superior strain exhibits the largest pool, particularly of cysteine (four times that of the non-producing strain). In fermentations with this strain methionine was found to be a much better sulphur source than sulphate for both cysteine and antibiotic synthesis. In the presence of methionine the utilization of sulphate is strongly depressed.  相似文献

The effects of corticostriatal deafferentation (decortication) and destruction of intrinsic neurons (intrastriatal kainate injection) on the extracellular concentration, and veratrine-releasable pools, of endogenous amino acids in the rat striatum were examined using the in vivo brain dialysis technique. Intracellular amino acid content was also determined. Decortication reduced selectively intra- and extracellular levels of glutamate (Glu) and aspartate (Asp). Extracellular changes were more pronounced than those in tissue content. gamma-Aminobutyric acid (GABA), taurine (Tau), and phosphoethanolamine (PEA) levels were not affected, whereas nonneuroactive amino acids were increased at 1 week but not at 1 month post-lesion. The intracellular pool of Glu and Asp was also reduced in kainate-lesioned striata. However, extracellular levels of these compounds were not affected significantly by this treatment. The tissue content of all other amino acids was decreased, the most prominent change being in the concentration of GABA. Extracellular GABA concentration was also reduced dramatically, whereas the concentrations of noneuroactive amino acids were increased to varying degrees. These data suggest that transmitter pools of neuroactive amino acids are an important supply for their extracellular pools. Lesion-induced alterations in nonneuroactive amino acids are discussed with regard to the loss of metabolic pools, glial reactivity, and changes in blood-brain barrier transport. Veratrine induced a massive release of neuroactive amino acids such as Glu, Asp, GABA, and Tau into the extracellular fluid, and a delayed increase in PEA. The intensification of livestock have increased the emission of pollutants to the environment, leading to a growing interest in seeking strategies that minimise these emissions. Studies have shown that it is possible to manipulate diets by reducing CP levels and thus reducing nitrogen (N) excretion, without compromising performance. However, there is no knowledge of any study that has focused on reducing N excretion and relating this reduction to individual amino acids. This study investigated the effect of dietary methionine+cysteine (MC) and threonine (THR), the two most limiting amino acids for broiler production, on nitrogen excretion (NE) and nitrogen deposition (ND) and determined the efficiency of utilisation of both amino acids for protein deposition. Six trials were conducted to measure the NE and ND in broiler chickens during three rearing phases in response to dietary amino acid. The efficiency of utilisation of the amino acids was calculated by linear regression of body protein deposition and the amino acid intake. Despite the differences between sexes and phases, the efficiency of utilisation was the same, being 0.60 and 0.59 for MC and THR, respectively. The rate of NE behaved exponentially, increasing with amino acid intake, and can exceed 50% of N intake, being higher than ND. On average, for a reduction in intake of each unit of MC or THR (mg) there is a reduction of 0.5% of NE. Although this reduction seems low, considering that it corresponds to changes in one amino acid only, the impact on a large scale would be significant. Knowledge of how animals respond to NE and ND/protein deposition according to amino acid dietary content may represent new efforts towards reducing the impact on environment.  相似文献

Summary The behaviour of E. coli B culture grown on SO4 2--free minimal glucose-salt medium was examined in the presence of exogenous cysteine at various concentrations. This was done by means of using the following parameters: length of lag, growth rate and total population. Up to a concentration of cysteine at 0.2mm the growth sets in without a lag phase, the growth rate is optimal (identical with that of cultures grown on media containing Na2SO4 as source of sulphur), only the size of total population being decreased by cysteine. At concentrations of 0.2mm and upwards, after a concentration-dependent lag-period, the cultures were found to increase at various lower growth rates.The toxic effect of cysteine was reduced by leucine itself, as well as by a mixture of leucine, isoleucine, valine and threonine. The anti-cysteine action of these amino acids showed itself in the shortening of the lag period and in the recovery of the growth rate which, however, failed to reach the original level.Cysteamine failed to provide sulphur for cultures of E. coli B grown on the above medium. Neither was the utilization of cysteamine affected by the application of amino acids possessing an anti-cysteine action.We have postulated that beside the inhibition of the biosynthesis of amino acids having an anti-cysteine effect, toxic concentrations of cysteine posses additional sites of action.  相似文献

Effects of four amino acids, L-asparagine, L-cysteine, L-citrulline, and L-glutamine in different concentrations (0, 0.5, 1, and 2 mg dm-3) combined with 2 mg dm-3 indole-3-butyric acid, on in vitro rooting and biochemical constituents of cherry rootstocks CAB-6P (Prunus cerasus L.) and Gisela 6 (P. canescens × P. cerasus) were investigated. In CAB-6P, root number and root fresh mass (FM) were maximum at 0.5 mg dm-3 cysteine. All amino acids reduced root length in CAB-6P and root number as well as root FM in Gisela 6. In Gisela 6, 0.5 mg dm-3 asparagine or 2 mg dm-3 glutamine reduced root length. In CAB-6P, 100 % rooting was achieved in the control and with 1 and 2 mg dm-3 cysteine or 1 mg dm?3 citrulline. In Gisela 6, the rooting percentage was maximum (76.92 %) with 0.5 mg dm?3 asparagine. Callus FM in CAB-6P was the greatest at 1 mg dm?3 and in Gisela 6 at 2 mg dm?3 citrulline. Callusing was 100 % in the majority of treatments for CAB-6P and 92.31 % for Gisela 6 with 0.5 or 2 mg dm?3 citrulline. Cysteine, citrulline, and glutamine diminished chlorophyll content in Gisela 6 whereas in CAB-6P all four amino acids hardly affected it. Carotenoid and porphyrin content in CAB-6P was decreased due to asparagine (0.5 or 1 mg dm?3). Porphyrin content in CAB-6P was also reduced by adding 0.5 or 1 mg dm?3 cysteine or 2 mg dm?3 citrulline. In Gisela 6, all amino acids decreased carotenoid and porphyrin content. In CAB-6P, all treatments except 0.5 mg dm?3 glutamine or 2 mg dm?3 asparagine increased leaf sucrose content. In roots, both sucrose and proline content were increased only at 1 mg dm?3 cysteine whereas in leaves only 0.5 mg dm?3 asparagine caused a 3-fold increase in proline content. A decrease in root proline in CAB-6P was observed due to asparagine, citrulline, or glutamine. In Gisela 6, decreased leaf sucrose and proline content was recorded at 2 mg dm?3 cysteine. All amino acids did not alter root sugar content remarkably whereas root proline content was raised by adding 0.5 mg dm?3 glutamine or 1 mg dm?3 cysteine.  相似文献

Nitrogen and sulphur, both vital structural elements, are especially needed for the synthesis of proteins and oils. Investigations revealed the required application of sulphur is one half to one third the amount of nitrogen, and the ratio becomes narrower in mustard (Brassica juncea L.), followed by wheat and rice. The efficiency of an increased level of nitrogen required a proportionately higher amount of sulphur. A critical investigation on the effective utilization of applied vis-à-vis absorbed nitrogen in wheat and mustard envisaged accumulation of NO3-N in vegetative parts when sulphur remained proportionately low. Application of sulphur hastened the chemical reduction of absorbed NO3- for its effective utilization. The effect was more pronounced in mustard than in wheat. Easily available forms of sulphur, like ammonium sulphate and gypsum, as compared to pyrite or elemental sulphur, maintained adequate N to S ratio in rice, resulting in a reduction in the percent of unfilled grain, a major consideration in rice yield. A narrow N to S ratio, with both at higher levels, increased the oil content but raised the saponification value of the oil, a measure of free fatty acids. Whereas, a proportionately narrow N to S ratio at moderate dose resulted in adequately higher seed and oil yield with relatively low saponification value, associated with increased iodine value of the oil, indicating respectively low free fatty acids and higher proportion of unsaturated fatty acids, an index for better quality of the oil.  相似文献

7S globulins were extracted from common bean (Phaseolus vulgaris L.) seeds and characterized. SDS-PAGE showed major bands corresponding to the phaseolin subunits (43-53 kDa). An amino acid analysis indicated that, in spite of the limited amounts of sulphur amino acids and tryptophan, the globulins contained very high levels of essential amino acids. The protein solubility profiles of native and denatured (120 degrees C for 20 min) 7S globulins in water and in 0.5 M NaCl showed that NaCl had a limited effect on increasing the solubility of either the native or denatured proteins. The in vivo small intestinal digestibility of the 7S globulins was 90%, this being decreased to 86% after a thermal treatment. Fourier transform infrared spectroscopy revealed a high content of beta-sheet and beta-turn structures, together with a contribution at 1687 cm(-1) that was assigned to intramolecular beta-sheets. These features are diagnostic of a high propensity to irreversible aggregation that may be related to an adverse effect on the protein quality.  相似文献

A mutant Escherichia coli leucyl-tRNA synthetase has been evolved for the selective incorporation of the methionine homolog 1 into proteins in yeast. This single aminoacyl-tRNA synthetase is capable of charging an amber suppressor EctRNA(CUA)(Leu) with at least eight different amino acids including methionine and cysteine homologs, as well as straight chain aliphatic amino acids. In addition we show that incorporation yields for these amino acids can be increased substantially by mutations in the editing CP1 domain of the E. coli leucyl-tRNA synthetase.  相似文献

Summary The effects of P nutrition under various salinity levels on the protein, amino acids, and nutrients in mature wheat grain were studied. Mexican dwarf wheat (Triticum aestivum L. var Inia) was grown to maturity in solution cultures with variable concentrations of P (0.5-, 5-, 25- and 50 mg P/l) in combination with NaCl at concentrations producing osmotic potentials (s) of –0.4-, –1.4-, –2.4- and –4.4-bars. All other essential nutrients were present in adequate concentrations for vigorous plant growth.Increasing levels of P in the nutrient solutions tended to decrease the grain yield, N, Cl, protein-glutamic acid,-proline,-leucine,-glycine, and-serine, while P, K, Mg, Zn, Mn and Cu in the grain were increased. The sum of all protein amino acids in the grain decreased as the concentration of P increased in the nutrient solution. The effect of P on the individual and sum of amino acids tended to show peak amounts at the 5.0 mg P/l treatment level.Increased levels of salinity significantly reduced grain yield, N, proteinglutamic acid,-proline,-leucine,-glycine,-serine,-aspartic acid,-alanine, and-phenylalanine in the grain. The sum of the protein amino acids (mol/g dry wt.) was decreased in the grain from plants grown at –4.4 bars salinity level, but not in the grain from plants receiving less saline treatments. The concentrations of free amino acids: serine, aspartic acid, glutamic acid, alanine, and arginine were lower in the grain produced at the –4.4 bars salinity than at –0.4 bars salinity level. The sum of free amino acids (mol/g dry wt.) in the grain were decreased at the highest salinity level as compared with concentrations found for grain produced at lower salinity levels.There were some interactions found between P and salinity on the protein amino acids and nutrients in the grains.The amounts of essential amino acids expressed in mg/g of protein were not significantly affected by the increasing levels of P and salinity in the nutrient solution and they were found in adequate or greater amounts than those recommended by the World Health Organization and FAO.  相似文献

Summary An application of sulphur (upto 90 ppm) alone or in combination with phosphorus and nitrogen, increased the content of total nitrogen, protein nitrogen and total soluble nitrogen but decreased amide, amino, ammoniacal and nitrate nitrogen and with further increase in dose, all the nitrogen fractions decreased. Higher doses of nitrogen in combination with sulphur increased the content of amide nitrogen and nitrate nitrogen in leaves only at 15 and 30 days stages. Higher doses of phosphorus in combination with sulphur and nitrogen decreased the content of all nitrogen fractions in leaves. All the nitrogen fractions decreased in age except the amide and ammoniacal nitrogen which increased upto 30 days and then started decreasing.  相似文献

The effects of melatonin and dopamine precursor L-3,4-dihydroxyphenylalanine (L-DOPA) intraperitoneal administration on the rhythms of free amino acids content in the retina of rats were studied. The authors found that the levels of those amino acids, which are protein constituents but not neurotransmitters in the rat retina, change diurnally with maximum at 3-6 h after light onset. Diurnal changes of Ala, Arg, Asn, Ile, Met, Ser, Trp, and Val content persisted in the retina of rats maintained at constant darkness. This fact confirms the true circadian nature of these rhythms. Constant lighting abolished diurnal changes of the content of all amino acids with the exception of Trp. Daytime but not nighttime administration of melatonin decreased the levels of Ala, Asn, Gln, Ile, Met, and Ser down to nocturnal values. Diurnal changes of amino acids content vanished in melatonin-injected rats. The effect of melatonin administration disappeared when the protein synthesis was inhibited by cycloheximide. The effect of intraperitoneal administration of L-DOPA on the levels of free amino acids was opposite the effect of melatonin administration. L-DOPA increased nocturnal levels of Gly, Thr, Trp, and Val but had no effect on the daytime amino acids content. As in the case of melatonin administration, significant diurnal changes of amino acid levels disappeared in L-DOPA-injected rats. The authors hypothesize that melatonin and dopamine can serve as zeitgebers-antagonists of amino acids content rhythms in the rat retina.  相似文献

试验以黄河鲤(Cyprinus carpio)为研究对象,以饲料中杜仲(Eucommia ulmoides)叶粉0添加组为对照组,2%、4%、6%和8%添加组为实验组,每组3个平行,对黄河鲤[体重:(505.13±1.37) g]进行55d的投喂试验,旨在探讨杜仲叶粉在黄河鲤饲料中的应用效果。结果表明:(1)黄河鲤背肌脂肪含量随饲料中杜仲叶粉添加量升高显著下降,粗蛋白含量显著上升(P<0.05)。除6%添加组外,鱼体粗灰分、粗蛋白和粗脂肪鲜重含量随杜仲叶粉添加量升高呈上升趋势(P<0.05),鱼体鲜重水分含量呈显著下降趋势(P<0.05);(2)在黄河鲤肌肉中检出16种氨基酸,杜仲叶粉添加显著影响肌肉中苏氨酸、丝氨酸和组氨酸含量(P<0.05)。酸味氨基酸总量与总氨基酸比值在2%添加组显著高于其他处理组,药效氨基酸总量与总氨基酸比值则在2%添加组显著低于其他处理组(P<0.05);(3)黄河鲤血浆和肝脏中GSH-PX、MDA、AKP和ACP活性随饲料中杜仲叶粉添加量增加整体呈先升高后降低趋势,血浆和肝脏中SOD活性呈显著上升趋势(P<0.05)。血浆...  相似文献

Two mutants of Chlorella vulgaris characterized by higher and lower content of sulphur amino acids compared with the wild strain were assayed for the efficiency of the sulphate uptake mechanism. In both mutants uptake exhibited positive cooperation kinetics and was strongly stimulated by sulphate starvation. Stimulation was depressed by cysteine and to a higher extent by methionine. Mutations affected the uptake efficiency concordantly with the level of sulphur amino acids. Addition to the starved strains of sulphate or chromate reduced the induced transport to the lower values of the non-starved strains. Addition of cycloheximide during the induction period prevented a further enhancement of transport without depressing the attained rate in the low sulphur mutant; it was followed by a rapid fall to the non-induced rate in the high sulphur mutant.  相似文献

Summary The effect of selenium and phosphorus application on Berseem (Trifolium alexandrium) was studied in the green house. Each dose of selenium from O to 16 ppm decreased the dry matter yield of Berseem significantly at all the levels of applied phosphorus. Phosphorus stimulated the dry matter yield at all levels of selenium. Yields in the second cutting were of a higher order than in the first cut. Selenium application increased selenium and phosphorus content in Berseem, but at 8 and 16 ppm of selenium the phosphorus content decreased significantly. Sulphur concentration decreased with selenium application but application of phosphorus increased sulphur concentration showing synergetic effect.Studies on residual effect of selenium revealed that the application of 50 ppm of phosphorus increased the yield while that of 100 ppm decreased it. Selenium concentration which increased with increasing phosphorus was many fold higher in the first cutting than in the second cutting and similar was the trend with phosphorus and sulphur.  相似文献

The urinary excretion of thiocyanate by hens after dosage with cyanide (30 mumol) has been studied in a series of acute experiments involving 6 hr urine collection periods. More than half of the dose could be recovered as thiocyanate when cyanide was given by intravenous infusion and the rate of excretion closely paralleled plasma thiocyanate concentration. Little cyanide was excreted directly. The excretion of thiosulphate fell by an amount that suggested that availability of sulphane sulphur might limit the extent of conversion. However, neither thiosulphate nor sulphur amino acids enhanced thiocyanate excretion when they were infused together with cyanide; indeed, thiocyanate excretion decreased as the level of sulphur compound given was increased. Both nitrite and sulphite depressed thiocyanate excretion also but they differed in their effects on plasma thiocyanate levels and the pattern of urinary excretion. Comparison of excretion from both sides of the kidneys separately emphasised the importance of the first pass of cyanide in its conversion to thiocyanate. The results suggest that although sulphur availability may be limited the in vivo production of sulphite also restricts cyanide detoxication.  相似文献
