Time-sequence analyses of carbohydrate breakdown in germinating rice seeds shows that a rapid breakdown of starch reserve in endosperm starts after about 4 days of germination. Although the major soluble carbohydrate in the dry seed is sucrose, a marked increase in the production of glucose and maltooligosaccharides accompanies the breakdown of starch. Maltotriose was found to constitute the greatest portion of the oligosaccharides throughout the germination stage. α-Amylase activities were found to parallel the pattern of starch breakdown. Assays for phosphorylase activity showed that this enzyme may account for much smaller amounts of starch breakdown per grain, as compared to the amounts hydrolyzed by α-amylase. There was a transient decline in the content of sucrose in the initial 4 days of seed germination, followed by the gradual increase in later germination stages. During the entire germination stage, sucrose synthetase activity was not detected in the endosperm, although appreciable enzyme activity was present in the growing shoot tissues as well as in the frozen rice seeds harvested at the mid-milky stage. We propose the predominant formation of glucose from starch reserves in the endosperm by the action of α-amylase and accompanying hydrolytic enzyme(s) and that this sugar is eventually mobilized to the growing tissues, shoots or roots.  相似文献

The ultrastructural changes occurring in the scutellar epithelium cells of rice seeds have been studied during germination and early seedling growth. During this time, several prominent structural changes occur, including (a) formation, development, and proliferation of organelles such as mitochondria, rough endoplasmic reticulum, free ribosomes, and Golgi apparatus; (b) folded structural modification of plasmamembranes in later stages; and (c) conspicuous decrease in lipid-storing spherosomes. Glyoxysome-like electron dense particles are detectable but their formation is much less prominent. It is conceivable that all these structural changes are related to the enhancement of the metabolic activities of the epithelial cells including the synthesis of hydrolytic enzymes such as α-amylase and their secretion into the endosperm tissues. Some enzyme activities characteristic of mitochondria and glyoxysomes have been determined using the crude scutellar extracts, and the results dealing with the low activities of the glyoxylate cycle enzymes and palmitoyl-coenzyme A oxidase appear to indicate that fatty acid breakdown is possibly via mitochondrial β-oxidation, although we reserve a definitive conclusion on the glyoxysomes being nonfunctional in fatty acid oxidation in rice seedlings.  相似文献

Some enzymic Properties of a partially purified preparationof sucrose phosphate synthetase (E.C.2.4.1.14) from germinatingrice seed scutella were studied. Examination of the reactionkinetics revealed that the rate of synthesis of sucrose phosphatefollows the Michaelis-Menten equation at an optimum PH of 7.5,having Km of 25 mM for UDP-glucose, and of 4.9 mM for fructose6-phosphate. UDP inhibited the enzyme reaction competitively;K1 of 3.3 mM. Fe++ and Fe+++ activated the enzyme reaction about2-fold; Ka, 0.3 mM and 2.0 mM, respectively. Co++, Co(NH3)6+++,Mg++ and Mn++ also activated the enzyme reaction. At high concentrationK+ activated the enzyme reaction with the maximum activationof 24% at 400 mM. The molecular weight and S20,w value of theenzyme were determined as 4.5 ? 105 and 10.4S, respectively. 1Part IV of this series is Ref. The formation of multiple forms of amylases in germinating rice (Oryza sativa L. cv Kimmaze) grains was examined by means of isoelectric focusing, cross-immunoelectrophoresis, and rocket-line immunoelectrophoresis followed by a reaction of enzymic characterization by using β-limit dextrin or starch as substrate. The constituents detected by isoelectric focusing were identified as three electrophoretically heterogeneous antigens. The major α-amylase bands A and B corresponded to a same antigen, the main portion of which was produced within 2 days' germination. The bulk of α-amylase D appeared between 2 and 4 days' germination. Component E, a debranching enzyme according to its action on the β-limit dextrin, already exists in the ungerminated seeds; its amount decreases within the first 2 days of germination and increases again thereafter.

Evidence showing that β-amylase (band C) is produced by the scutellum at an early stage of germination was provided. The enzyme appeared in a suspension of the scutellum after a prolonged incubation.

The time sequence analysis of the starch digestion pattern of the thin sectioned germinating rice (Oryza sativa L.) seed specimens using the starch film method showed that at the initial stage amylase activity was almost exclusively localized in the epithelium septum between the scutellum and endosperm. Starch breakdown in the endosperm tissues began afterward; amylase activity in the aleurone layers was detectable only after 2 days. Polyacrylamide gel electrofocusing (pH 4 to 6) revealed nearly the same zymogram patterns between endosperm and scutellum extracts, although additional amylase bands appeared in the endosperm extracts at later germination stages (4 to 6 days). These are presumably attributable to the newly synthesized enzyme molecules in the aleurone cells.  相似文献

The DEAE-cellulose column chromatography has shown two differentforms of starch synthetase, which are referred to as fractionsI and II in extracts of rice seeds (non-waxy and waxy varieties)harvested at the milky stage. Similarly treated leaf extractsof rice (non-waxy) and maize (non-waxy) also demonstrate dieexistence of two major isozyme fractions. In all enzyme preparationstested, ADP-glucose was the sole glucosyl donor and UDP-glucosewas totally inactive. Rechromatography, on a DEAE-cellulosecolumn, of two enzyme fractions (I and II) separated from non-waxyrice seed extracts did not alter their elution patterns. Someof their enzymic properties were compared, in particular, theirglucosyl-acceptor (primer) specificities. Regardless of potentamylase activities in the two fractions, notable differenceswere observed in that fraction I utilized the long-chain oligosaccharides[maltododecaose] and various types of high molecular -glucansmore readily than fraction II. However, short-chain oligosaccharides[maltose, maltotriose and maltotetraose] were utilized morereadily by fraction II than by fraction I. Microsomes prepared from the rice seed scutellum were incubated in wheat germ extracts (S-100 fraction) to direct the synthesis of alpha- amylase, a secretory protein subject to proteolytic processing (cleavage of the N-terminal signal sequence) as well as glycosylation during its biosynthesis. The characterization and identification of the immunoprecipitable products synthesized were performed by SDS gel electrophoresis and subsequent fluorography. The molecular weight of the alpha-amylase synthesized by the microsomes was found to be identical with that of the mature secretory form of the enzyme on the basis of electrophoretic mobilities. A significant portion of the enzyme molecules synthesized was shown to be segregated into the microsomal vesicles and protected against digestion by endo-beta-N- acetylglucosaminidase, indicating that both proteolytic processing and glycosylation of the precursor polypeptide chains take place in the microsomes. The modification of the polypeptide chains was further examined by disrupting the microsomal membranes with Triton X-100. Detergent treatment of the microsomes prior to protein synthesis caused an inhibition of both proteolytic processing and glycosylation of the polypeptide chains, leading to the synthesis of the unprocessed nascent (precursor I), processed but nonglycosylated nascent (precursor II) forms, in addition to the mature form of alpha-amylase. Furthermore, the results of time-sequence analysis of the inhibitory effect of Triton X-100 on the modification of the polypeptide chains have led us to conclude that both proteolytic processing and subsequent glycosylation occur in the microsomes during the biosynthesis of alpha- amylase.  相似文献

Determination of seed vigor was attempted by comparing ATP-levels of deteriorating seed to germination percentage and production of dry matter. Immediately after imbibition of any seed lot investigated, a production of ATP took place. This ATP-accumulation invariably reached a plateau after 6 h of imbibition. Two well germinating seed lots of rape, one of cauliflower and one of sugar beet, were artificially aged by means of elevated storage temperature and humidity. Every second week through 16 weeks of deterioration the levels of ATP, ADP and AMP after 7 h of imbibition were compared with the germination percentage. While ADP- and AMP-contents of germinating seed displayed no change (when imbibed 7 h) during the period of artificial aging, seed deterioration was reflected in the ATP-levels long before loss of viability could be detected by the conventional germination test.
When ATP-levels per seed were related to germination percentage throughout the aging, all four seed lots displayed similar patterns although the absolute figures differed. In contrast to the conventional "per seed' basis, however, ATP per gram seed not only displayed similar deterioration patterns, but the absolute values were also of the same magnitude.  相似文献

Biosynthesis and excretion of hydrolases in germinating cereal seeds   总被引:1,自引:0,他引:1  
The initial formation site of hydrolases in germinating cerealseeds and their subsequent release were examined using the substrate-filmtechnique. Early in the germination of cereal seeds, e.g., barley,wheat, rye, oat and maize, -amylase invariably appeared in theregion of the epithelial cells of the scutellum, whereas itlater gradually diffused into the entire region of the endospermtissues. The initial formation site of proteinase and RNA-asein germinating barley seeds was also confirmed to be in theepithelium. We conclude that the epithelium has a more importantrole in the enzymic breakdown of reserve substances stored inthe endosperm tissues than the aleurone layer. (Received October 19, 1979; )  相似文献

Rapid mobilisation of storage products, including xyloglucan, in cotyledons of germinating nasturtium (Tropaeolum majus L.) normally starts about 7–8 d after imbibition and growth of the seedling at 20–25° C. Levels of activity of endo-1,4--glucanase (EC 3.2.1.4) in cotyledons, as assayed viscometrically with xyloglucan as substrate, varied in parallel with the rate of breakdown of xyloglucan. When cotyledons were excised from the seedling axis and incubated on moist filter paper at any point before 7 d, the catabolic reactions which normally occurred in the intact seedling were suspended. If, however, cotyledons excised at 8 d were incubated in 10–6 M 2,4-dichlorophenoxyacetic acid, a rise in endo-1,4--glucanase (xyloglucanase) activity was observed and a sharp decrease in fresh and dry weight as well as xyloglucan levels ensued at rates comparable to those observed in cotyledons attached to the seedling. Neither gibberellin nor kinetin treatments promoted xyloglucan breakdown or enhanced xyloglucanase activity. Addition of auxin to excised cotyledons before 7 d did not evoke premature breakdown, indicating that the tissue became receptive to auxin only at this time. The triggering process took place in darkness and was unaffected by various light-dark cycles. We isolated and identified 10 alpha-amylase isoforms by using beta-cyclodextrin Sepharose affinity column chromatography and two-dimensional polyacrylamide gel electrophoresis from germinating rice (Oryza sativa L.) seeds. Immunoblots with anti-alpha-amylase I-1 and II-4 antibodies indicated that 8 isoforms in 10 are distinguishable from alpha-amylase I-1 and II-4. Peptide mass fingerprinting analysis showed that there exist novel isoforms encoded by RAmy3B and RAmy3C genes. The optimum temperature for enzyme reaction of the RAmy3B and RAmy3C coding isoforms resembled that of alpha-amylase isoform II-4 (RAmy3D). Furthermore, complex protein polymorphism resulted from a single alpha-amylase gene was found to occur not only in RAmy3D, but also in RAmy3B.  相似文献

Abstract Cultivars of barley (Hordeum vulgare L.) were tested for germination sensitivity to progressively higher concentrations of salt, mannitol, and betaine. The three solutes were equally inhibitory at equal osmotic potential, but there was a consistent difference in osmotic sensitivity between two cultivars, CM-67 and Briggs (Briggs was the most sensitive). There was no difference between the two cultivars in salt or water uptake from salt solutions during imbibition. Brief presoaking in water did not improve salt resistance, indicating that a hydration-dependent decrease in membrane permeability is not involved in salt tolerance. The calcium content of Briggs was higher than CM-67. These results suggest that salt inhibits barley germination primarily by osmotic effects, and that salt influx during imbibition does not play a role in this inhibition. A hypothesis regarding salt effects on germination is discussed.  相似文献

Sucrose synthetase (UDP-glucose:d-fructose-2-glucosyltransferase, EC 2.4.1.13) from ripening rice seeds was purified by ammonium sulfate fractionation and column chromatography of microgranular DEAE-cellulose (DE-32) and Neusilin (MgO· Al2O3·2SiO2). An enzyme preparation obtained was homogeneous as examined by polyacrylamide gel electrophoresis. The enzyme, having a molecular weight, 4.0 × 105, consists of 4 identical subunits, each having a molecular weight, 1.0 × 105.Examination of reaction kinetics of both sucrose synthesis and cleavage catalyzed by sucrose synthetase revealed that the rate of synthesis follows a Michaelis-Menten equation having the following parameters: Km(fructose)UDP-glucose, 6.9 mm; Km(fructose)ADP-glucose, 40 mm; Km(UDP-glucose), 5.3 mm; and Km(ADP-glucose), 3.8 mm. The cleavage reaction yielded the following values: Km(UDP), 0.8 mm; Km(ADP), 3.3 mm; and Km(sucrose)UDP, 290 mm. In the latter reaction the rate deviated from the Michaelis equation when ADP was used as the glucose acceptor, the n value being 1.6 by the Hill plot analysis and S0.5(sucrose)ADP, 400 mm. At high concentration of ADP the cleavage reaction was inhibited, while the synthesis reaction was inhibited with high concentrations of fructose.  相似文献

Summary Ultrastructural aspects of the breakdown of starch grains during the mobilisation of reserves in Phaseolus aureus Roxb. seed germination are described. The starch grains show erosion from within leading to the formation of a hollow shell. The erosion is accompanied by intrusion of cytoplasm into the shell. The biosynthesis of nonspecific lipid transfer proteins (ns-LTPs) in germinating castor bean (Ricinus communis L.) seeds were investigated. Lipid transfer activities of ns-LTPs in the cotyledons, axis, and endosperm increased with growth after germination. The activity increases were accompanied by increased amounts of ns-LTPs in each tissue, as measured by immunoblot using anti-ns-LTP serum. These results suggest that the ns-LTPs are synthesized de novo in each tissue after germination and not activated from inactive proteins synthesized before germination. Comparison of the immunoblot products in each tissue from 4-day-old seedlings indicate the occurrence of tissue-specific isoforms of ns-LTPs; 9 kilodaltons (major) and 7 kilodaltons (minor) in the cotyledons, and 7 kilodaltons (major) and 9 kilodaltons (minor) in the axis, whereas only the 8-kilodalton ns-LTP is present in the endosperm. In vitro translation from poly(A)+ RNAs from three tissues of castor bean seedlings and the detection of immunoprecipitated products indicate that translatable mRNAs for ns-LTPs exist in the three tissues a day before the synthesis of ns-LTPs; the translation products, which are 3.5 to 4.0 kilodaltons larger than ns-LTPs, were processed to the mature ns-LTPs. The production of mature ns-LTPs from translatable mRNAs without any delay suggests that gene expression of ns-LTPs in castor bean seedlings is controlled at a step before the formation of translatable mRNAs.  相似文献
