研究了柠檬酸、草酸和乙酸对污泥中Cd、Pb、Cu和Zn等重金属的去除效果,并分析了污泥中重金属的形态变化和生物有效性.结果表明,随着反应时间和酸浓度的增加,污泥中重金属(Cu除外)的去除率也相应增加.污泥加入柠檬酸溶液反应7h即可去除污泥中52.0%的Pb和74.2%的Zn,24h后可去除76.0%的Pb和92.5%的Zn,草酸和乙酸对重金属的去除率较低.柠檬酸去除的Pb和Zn主要以稳定态存在,并导致污泥中不稳定态重金属的比例上升,其中可交换态重金属浓度有不同程度的增加.虽然有机酸对Cd和Cu的去除率较低,但反应后可交换态Cd和Cu的浓度仍有小幅增加.  相似文献

重金属铜、锌、镉复合胁迫对麻疯树幼苗生理生化的影响   总被引:2,自引:0,他引:2  
该研究以Cu~(2+)、Zn~(2+)、Cd~(2+)单一胁迫为对照,探讨不同浓度的Cu~(2+)、Zn~(2+)、Cd~(2+)复合胁迫对麻疯树幼苗生理生化指标的影响。结果表明:随着Cu~(2+)、Zn~(2+)、Cd~(2+)浓度的增加,麻疯树幼苗叶片中的蛋白质(Pro)、丙二醛(MDA)含量均逐渐增加,其叶片叶绿素含量随着Zn~(2+)胁迫浓度的增加呈现出先降后升的趋势,在中等浓度(100 mg·L-1)的Zn~(2+)胁迫时含量最低、随着Cu~(2+)胁迫浓度的增加叶绿素含量先升高后降低,在Cu~(2+)浓度为200 mg·L-1时含量最高,达到1 200 mg·g-1FW; Cd~(2+)胁迫对叶绿素含量和根系活力无明显影响。根系活力在Zn~(2+)浓度为100 mg·L~(-1)时最强,随着Cu~(2+)浓度的增加而减弱。低浓度的Cu~(2+)、Zn~(2+)、Cd~(2+)对过氧化物酶活性和可溶性糖含量都具有促进作用。Cu~(2+)、Zn~(2+)、Cd~(2+)复合胁迫时对可溶性蛋白、叶绿素和丙二醛含量均无明显影响,随着复合胁迫时浓度的增加,可溶性糖含量和根系活力先增后减。这表明麻疯树对三种重金属的胁迫具有一定的抗性,过高浓度的胁迫会影响麻疯树幼苗生理生化的一些指标,但是麻疯树可以通过自身的防御系统使伤害降到最小。此外,重金属复合胁迫可以在一定程度上减轻单一胁迫对麻疯树幼苗造成的毒害作用。  相似文献

【目的】了解水环境中重金属铜对异育银鲫肠道微生物组成及多样性的影响。【方法】采用试剂盒提取异育银鲫肠道总DNA,然后对总DNA进行16S r RNA进行扩增,构建异育银鲫肠道微生物16S r RNA基因克隆文库,最后进行数据分析。【结果】厚壁菌门、变形菌门和拟杆菌门为异育银鲫肠道中主要的细菌类群,在不同浓度的重金属铜胁迫处理后,厚壁菌门的含量明显降低。稀释性曲线、Venn图和多样性指数分析结果表明,重金属铜胁迫处理后异育银鲫肠道微生物多样性明显降低。【结论】重金属铜会使异育银鲫肠道微生物组成及多样性降低。此结果为研究重金属污染对异育银鲫健康状况的影响及异育银鲫养殖过程中病害的诊断奠定基础。  相似文献

通过室内模拟实验,研究了孔石莼和重金属(铜、镉、锌和铅)共同作用下,海水无机碳体系及碳源汇格局的变化.结果表明,t=7d时,无机碳体系各参数的变化幅度(Δ)与重金属种类和浓度有关.与对照相比,低浓度(<1μmol · L-1)的重金属添加组中,DIC、HCO-3 和PCO2的下降幅度都很明显(P<0.01* *).当铜、镉浓度大于"转折浓度"后, DIC、HCO-3和Pco2 均要大于初始值,其增幅随着重金属浓度的增加而增大.对于锌和铅,二者浓度高达50μmol · L-1时,水体中无机碳各参数与初始值相比仍呈现下降趋势.此外,当重金属浓度和种类不同时,水体中的碳源汇格局亦做不同的变化.当铜和镉浓度小于转折浓度时,水体表现为大气CO2的汇;而当铜和镉超出转折浓度时,水体会由大气CO2的汇过渡到源,并且当水体成为CO2的源后,其CO2的释放量是随着铜、镉浓度的增加而增大.在本实验设计的各浓度锌、铅添加组水体始终表现为碳汇,但当锌、铅浓度分别高于15μmol · L-1和20μmol · L-1时,其碳汇强度开始小于对照组(P<0.05*).  相似文献

【目的】从污染土壤中分离筛选一株多环芳烃降解菌,并探究其与Pseudomonas aeruginosa B6-2构建的混菌体系对菲-镉复合污染的修复效能,以及微生物代谢特性对不同镉浓度赋存的响应特性,以期为复合污染的生物修复提供优良菌株资源及应用技术参考。【方法】采用富集驯化、筛选纯化方法得到一株多环芳烃降解菌,通过生理生化特征和16S rRNA基因序列分析进行鉴定。利用高效液相色谱法和电感耦合等离子体质谱法评估不同镉浓度赋存下各反应体系对菲和镉的去除效能;通过菌体细胞形态的扫描电镜观测及菌株代谢活性检测,探讨镉胁迫对菲生物降解过程的影响机制。【结果】筛选得到一株具有重金属耐受性和多环芳烃高效降解菌SZ-3,经鉴定为节杆菌属;降解菌协同体系(M)具有良好的菲降解效能和抗镉胁迫优势。镉胁迫浓度为0.5、10 mg/L时,M对菲和镉的去除率分别高于85%、80%;镉胁迫浓度为25、50 mg/L时,2种污染物的去除率均大于65%。扫描电镜分析表明,镉胁迫导致菌体表面粗糙且出现不同程度变形,菌体间黏附性和聚集性提高。反应周期内,邻苯二酚1,2-双加氧酶活性与电子传递体系活性随镉浓度增加而降低,两者变化与菲降解速率变化一致。【结论】Arthrobacter sp.SZ-3是一株PAHs高效降解菌,能与Pseudomonas aeruginosa B6-2协同高效修复菲-镉复合污染,随着初始镉胁迫浓度增加,混菌协同对目标污染物去除的优势显著。  相似文献

研究了钝顶螺旋藻和极大螺旋藻在含CdCl2水体中的生长状况与摄Cd能力.结果表明:两种螺旋藻皆对CdCl2有较强的耐受能力,但是有不同的摄Cd行为.当CdCl2浓度为6~24mg.L-1,培养96h时,两种螺旋藻对Cd的摄取作用主要表现为藻细胞外的表面吸附;培养10d时,钝顶螺旋藻的胞内Cd含量依然甚微,而极大螺旋藻对Cd的细胞内吸附量却明显增加,24mg.L-1CdCl2处理的极大螺旋藻胞内的Cd吸附量为12mg.L-1CdCl2处理的11.6倍,且略超过细胞表面吸附量.表明在高浓度Cd的长时间胁迫下,两种螺旋藻的摄Cd行为和对Cd的耐受机制具有明显差异,其中钝顶螺旋藻为胞外机制,而极大螺旋藻却为胞内、胞外混合机制,且以胞内机制为主.  相似文献

重金属复合污染是主要土壤污染类型之一。为了探明蜈蚣草修复土壤重金属复合污染的能力,土培试验研究了分别添加不同浓度铅、铜、锌条件下蜈蚣草生物量变化,结果表明,不同浓度铅和较低浓度锌处理下蜈蚣草生物量呈显著性地增加,土壤Pb、Zn添加浓度分别为1750mg/kg和1350mg/kg,蜈蚣草生物量最大。说明蜈蚣草有极强的耐Pb、Zn毒性能力,能在较高有效态Pb或Zn污染土壤上正常生长。蜈蚣草具有一定的耐Cu毒性能力,在Cu耐性方面,蜈蚣草可能存在生态型的差异。以上结果说明,利用蜈蚣草修复萃取Pb—As,Zn—As,Cu-As等复合污染土壤上As有重要的意义。  相似文献

一株抗重金属铜镉细菌的分离、鉴定及其16S rDNA的序列分析   总被引:12,自引:0,他引:12  
从湖北省大冶市矿区土壤中分离到一株高抗铜和镉的菌株,命名为NTG-01。该菌株可以单抗4.5mmol/L的铜和2mmol/L的镉,因此它是研究抗铜或镉机制的重要菌株。对分离到的NTG-01进行了形态学观察和生理生化鉴定以及16S rDNA序列分析。结果显示菌株NTG-01为细菌,革兰氏染色阴性,短杆状,鞭毛周生,菌体大小约为0.8μm×2.0μm,V-P实验阳性,甲基红实验阴性,利用葡萄糖产酸产气;通过对菌株NTG-01的16S rDNA序列进行测定和同源性比对,发现它与产气肠杆菌的16S rDNA有高达99%的同源性,结合形态和生理生化指标,将其鉴定为产气肠杆菌(Enterobacter aerogenes)。通过测定NTG-01对9种重金属的M IC值,可知它对多种重金属具有普遍较高的抗性。  相似文献

The distribution of metals within the hepatopancreas of Oniscus asellus (Crustacea, Isopoda) from two uncontaminated sites, and two sites contaminated with zinc, cadmium and lead, has been studied by atomic absorption spectrophotometry, light microscopy, transmission and scanning electron microscopy and X-ray microanalysis. The hepatopancreas contains two types of intracellular granule. The first type, in the S cells, are spherical granules which contain copper, sulphur and calcium. In woodlice from contaminated sites, these ‘copper’ granules, also contain zinc, cadmium and lead. The second type, in the B cells, are flocculent deposits which contain iron. In woodlice from contaminated sites, these ‘iron’ granules also contain zinc and lead. Moribund woodlice from contaminated sites have large numbers of ‘copper’ and ‘iron’ granules in the hepatopancreas and a fine deposit of zinc and lead on the membranes of the cells. There are numerous microorganisms in close association with the microvillous border of the hepatopancreas of woodlice from all four sites. Within the microorganisms of Oniscus asellus from contaminated sites, there are deposits of material which contain zinc, lead, calcium and phosphorus ‘Copper’ and ‘iron’ granules could have evolved as storage sites for essential metals to be utilized when demand from the body exceeds uptake from the food. Woodlice in contaminated sites may be able to ‘detoxify’ potentially harmful amounts of essential and non-essential metals by storing them in a relatively insoluble form within these granules.  相似文献

The effects of cadmium, copper, and zinc on the growth of ten strains ofaquatic hyphomycetes were investigated. On a solid medium, Cd and Cu reducedradial growth of most strains by 50% at concentrations between150–400 µM; in a liquid medium, the strains were more sensitive.The inhibitory effects of zinc were much less severe. Two isolates(Articulospora tetracladia and Tetracladium marchalianum) from a copper-minestream were more resistant against copper than conspecific strains from anon-polluted stream. Heliscus lugdunensis and Varicosporium elodeaeresponded to Cd exposure, but not to Cu or Zn exposure, by increasedsynthesis of SH-containing compounds. Glutathione levels showed a unimodalresponse to increasing Cd and Zn exposure. With copper, glutathionedecreased at intermediate levels of contamination. In the presence of Cd, H. lugdunensis synthesized several unknown sulfur-rich substances that wereabsent or produced at reduced rates in control cultures.  相似文献

Interactions of cadmium (Cd) ions with bovine serum albumin (BSA), bovine hepatic metallothionein (MT), calf thymus histone and deoxyribonucleic acid (DNA), and bovine hepatic chromatins were studied in the presence and absence of divalent zinc (Zn), copper (Cu), mercury (Hg), or lead (Pb) ions, using equilibrium dialysis at pH 7 and at 37°C. The BSA had 3.5 Cd-binding sites with an apparent affinity constant of 1×105. The other metal ions inhibited the binding by reducing the affinity constant and the number of Cd-binding sites in BSA. There were 6 high affinity and 13 low affinity Cd-binding sites in the MT. Zinc ions had poor efficacy in reducing the binding of Cd to the MT. However, the Cu2+ and Hg2+ ions inhibited the Cd binding to a considerable extent, the former ions being more potent in this respect. Histone did not bind Cd. There were two kinds of Cd-binding sites in DNA: One mole of Cd per four moles DNA-phosphorus at low affinity sites, and one mole of Cd per 6.7 moles DNA-phosphorus at high affinity sites. Their apparent association constants were 8.3×105 and 4.4×106 M, respectively. The other metal ions had inhibitory effects on the binding of Cd to DNA. Histone reduced the Cd-DNA interactions to only a minor extent. The other metal ions reduced the binding of Cd to DNA-histone complex to a small extent. Cadmium binds to the euchromatin (Euch), heterochromatin (Het), and Euch-Het mixture almost equally. The other metal ions reduced the binding maximally in Euch-Het followed next in order by Het and Euch. Cupric ions were the most potent inhibitors of the interactions of Cd with the nuclear materials.  相似文献

The present study aimed to evaluate the effect of soil-applied Zn and Cu on absorption and accumulation of Cd applied through irrigation water in legume (chickpea and mung bean) and cereal (wheat and maize) crops. The results revealed that Cd in irrigation water at higher levels (2 and 5 mg L?1) significantly (p < 0.05) reduced the plant biomass while the soil application of Zn and Cu, singly or combined, favored the biomass production. Plant tissue Cd concentration increased linearly with the increasing application of Cd via irrigation water. While Cd application caused a redistribution of metals in grains, straw, and roots with the highest concentration of Cd, Zn, and Cu occurred in roots followed by straw and grains. Zinc addition to soil alleviated Cd toxicity by decreasing Cd concentration in plant tissues due to a possible antagonistic effect. The addition of Cu to the soil had no consistent effects on Zn and Cd contents across all crops. Inhibitory effects of Cd on the uptake and accumulation of Zn and Cu have also been observed at higher Cd load. Thus, soil-applied Zn and Cu antagonized Cd helping the plant to cope with its toxicity and suppressed the toxic effects of Cd in plant tissues, thus favoring plant growth.  相似文献

The plant cytoskeleton orchestrates such fundamental processes in cells as division, growth and development, polymer cross-linking, membrane anchorage, etc. Here, we describe the influence of Cd2+, Ni2+, Zn2+, and Cu2+ on root development and vital organization of actin filaments into different cells of Arabidopsis thaliana line expressing GFP-FABD2. CdSO4, NiSO4, CuSO4, and ZnSO4 were used in concentrations of 5–20 µM in this study. It was found that Cd, Ni, and Cu cause dose-dependent primary root growth inhibition and alteration of the root morphology, whereas Zn slightly stimulates root growth and does not affect the morphology of Arabidopsis roots. This growth inhibition/stimulation correlated with the various sensitivities of microfilaments to Cd, Ni, Cu, and Zn action. It was established that Cd, Ni, and Cu affected predominantly the actin filaments of meristematic cells. Cells of transition and elongation zones demonstrated strong actin filament sensitivity to Cd and Cu. Microfilaments of elongating root cells were more sensitive to Ni and Cu. Although Cd, Ni, and Cu stimulated root hair growth after long-term treatment, actin filaments were destroyed after 1 h exposure with these metals. Zn did not disrupt native actin filament organization in root cells. Thus, our investigation shows that microfilaments act as sensitive cellular targets for Cd, Ni, and Cu. More data on effects on native actin filaments organization would contribute to a better understanding of plant tolerance mechanisms to the action of these metals.  相似文献

Summary The hydrolysis of phosphate from phytic acid by the acid soil phytase system was reduced in the presence of metal ions. Copper was most effective in this respect — zinc and cadmium were less inhibitory. Binding to metals did not completely inhibit the hydrolysis of phytic acid. At higher metal concentrations, where binding to other soil constituents, like humic acids, interfered less, the inhibition of the phytase activity was stronger than that of acid phosphatase.  相似文献

This study investigates the comparative strategies of accumulation under standardised laboratory conditions of the essential metals zinc and copper, and the non-essential metal cadmium by three crustaceans of different taxa; vizPalaemon elegans Rathke (Malacostraca: Eucarida: Decapoda),Echinogammarus pirloti (Sexton & Spooner) (Malacostraca: Peracarida: Amphipoda) and the barnacleElminius modestus Darwin (Cirripedia: Thoracica).The decapodP. elegans regulates body zinc concentrations to a constant level (ca. 79 µg Zn g–1) over a wide range of dissolved metal availabilities until regulation breaks down at high Zn availabilities and net accumulation begins. The amphipodE. pirloti accumulates zinc at all dissolved zinc concentrations but at a low net rate such that the accumulation strategy approaches that of regulation. The barnacleE. modestus accumulates zinc to high body concentrations with no significant excretion of accumulated zinc. In the case of copper,P. elegans similarly regulates body copper concentrations to a constant level (ca. 129 µg Cu g–1) over a range of dissolved copper availabilities until regulation breaks down at high copper concentrations. Both the amphipodE. pirloti and the barnacleE. modestus on the other hand accumulate copper at all dissolved copper exposures with no evidence of regulation. All three crustaceans accumulate the non-essential metal cadmium at all dissolved cadmium concentrations without regulation.Heavy metal accumulation strategies therefore vary between crustacean taxa and between metals. Uptake rates for zinc and cadmium have been estimated for the three crustaceans and can be interpreted in terms of cuticle permeability and way of life of each crustacean. Examination of these uptake rates provides an insight into possible reasons behind the adoption of particular metal accumulation strategies.  相似文献
