According to recent researches, angle shear connectors are appropriate to transfer longitudinal shear forces across the steel-concrete interface. Angle steel profile has been used in different positions as L-shaped or C-shaped shear connectors. The application of angle shear connectors in tilted positions is of interest in this study. This study investigates the behaviour of tilted-shaped angle shear connectors under monotonic loading using experimental push out tests. Eight push-out specimens are tested to investigate the effects of different angle parameters on the ultimate load capacity of connectors. Two different tilted angles of 112.5 and 135 degrees between the angle leg and steel beam are considered. In addition, angle sizes and lengths are varied. Two different failure modes were observed consisting of concrete crushing-splitting and connector fracture. By increasing the size of connector, the maximum load increased for most cases. In general, the 135 degrees tilted angle shear connectors have a higher strength and stiffness than the 112.5 degrees type.  相似文献

The vascular connection between lateral roots and stem in the Ophioglossaceae and in two leptosporangiate fern species was examined. Two types of connections were found: “gradual” connections, which resemble leaf traces in ontogeny and morphology, and “abrupt” connections, which resemble the connections between lateral roots and their parent roots. Gradual root-stem connections occur in the genera Ophioglossum and Helminthostachys and in Woodwardia virginica. They are initiated in shoot apices distal to the level where cauline xylem elements mature. They resemble leaf traces in being provascular (procambial) strands that connect the cauline stele with the future vasculature of lateral appendages. As with leaf traces, gradual connections are part of the provascular and, later, protoxylem continuity between stems and lateral appendages. Gradual connections have many features in common with leaf traces, and the term root trace is applicable to them. The order of radial maturation of the primary xylem in gradual connections varies in different parts of the connections. It is endarch near the intersection with the cauline stele and exarch where the connections intersect root steles. Gradual connections resemble the transition regions of certain seed plants where protoxylem is also continuous from stem to root and the order of maturation is found to change continuously from stem to root. Abrupt connections occur in Botrychium and Osmunda cinnamomea. They develop in shoot apices at levels where cauline xylem is mature or maturing. The mature xylem does not dedifferentiate, so provascular and protoxylem continuity of the kind found in root traces does not occur. Also, reorientation of the order of maturation does not occur in abrupt connections. Xylem connectors are found in the region where radially oriented elements of the connections abut the longitudinally oriented cauline elements. Abrupt connections resemble the connection of secondary roots with their parent root systems since xylem connectors and the lack of continuity are also features found in these vascular systems. The resemblance of the vascular pattern of the fern root trace to the transition region of seed plants suggests that the radicle is more closely comparable to the cladogenous roots of pteridophytes than hitherto supposed.  相似文献

We have investigated the thermodynamic parameters and binding of a regulatory subunit of cAMP-dependent protein kinase (PKA) to its natural low-molecular-weight ligand, cAMP, and analogues thereof. For analysis of this model system, we compared side-by-side isothermal titration calorimetry (ITC) with surface plasmon resonance (SPR). Both ITC and SPR analyses revealed that binding of the protein to cAMP or its analogues was enthalpically driven and characterised by similar free energy values (DeltaG=-9.4 to -10.7 kcal mol-1) for all interactions. Despite the similar affinities, binding of the cyclic nucleotides used here was characterised by significant differences in the contribution of entropy (-TDeltaS) and enthalpy (DeltaH) to DeltaG. The comparison of ITC and SPR data for one cAMP analogue further revealed deviations caused by the method. These equilibrium parameters could be complemented by thermodynamic data of the transition state (DeltaHnot equal, DeltaGnot equal, DeltaSnot equal) for both association and dissociation measured by SPR. This direct comparison of ITC and SPR highlights method-specific advantages and drawbacks for thermodynamic analyses of protein/ligand interactions.  相似文献

The primary xylem connection between the diarch parent root and the diarch lateral root was derived from the pericycle and stelar parenchyma. Early in lateral root development stelar parenchyma that was positioned between the parent xylem and the primordium divided transversely. These transverse divisions produced a plate of cells, most of which subsequently differentiated into vessel element connectors. After emergence of the lateral root, xylem maturation began in the stelar vessel element connectors and maturation proceeded acropetally into the lateral root. Protoxylem of the lateral root was connected to the metaxylem of the parent root via stelar vessel element connectors. The circular phloem connection was pericyclic in origin. Axial phloem connections which vascularized the lateral root were established with sieve tube elements of both parent phloem poles. Maturation of the phloem connection occurred prior to lateral root emergence. Transaxial phloem, positioned in arches above and below the lateral root vascular cylinder, was derived from the pericycle; and each arch consisted of three to four sieve tube elements. No transfer cells were found in the transaxial phloem.  相似文献

Current surface plasmon resonance (SPR) modes based on the attenuated total reflection (ATR) method can broadly be categorized as: conventional SPR, long-range SPR (LRSPR), coupled plasmon-waveguide resonance (CPWR), and waveguide-coupled SPR (WCSPR). Although the features of optical biosensors are dependent upon their particular SPR mode, a common requirement for all biosensors utilized for biomolecular interaction analysis (BIA) is a high degree of sensitivity. The current paper presents a theoretical analysis and comparison of the sensitivity and resolution of these four types of SPR biosensors when employed in three of the most prevalent detection methods, namely angular interrogation, wavelength interrogation, and intensity measurement. This study develops a detailed understanding of the influences of various biosensor design parameters in order to enhance the sensitivity and detection limit capabilities of such devices.  相似文献

Flexibility, the ability to deform in response to loads, is a common property of biological beams. This paper investigates the mechanical behavior of multi-jointed beams, which are characterized by a linear series of morphologically similar joints. Flexural stiffness and torsional stiffness were measured in two structurally distinct beams, crinoid arms (Echinodermata, Comatulida) and crustacean antennae (Arthropoda, Decapoda). Morphological data from these beams were used to determine the relative contributions of beam diameter and joint density (number of joints per millimeter of beam length) to the flexural and torsional stiffness of these two structures. As predicted by beam theory, beam diameter influenced stiffness in both crinoid arms and crustacean antennae. In crinoid arms, increases in joint density were associated with decreases in stiffness, but joint density had no significant influence on stiffness in crustacean antennae. In both crinoid arms and crustacean antennae, the magnitudes of flexural and torsional stiffness, as well as the ratio of these two variables, were similar to previously reported values for non-jointed biological beams. These results suggest that the structural design of a biological beam is not a limiting factor determining its mechanical properties.  相似文献

A semiparametric estimate of an average regression effect with right-censored failure time data has recently been proposed under the Cox-type model where the regression effect beta(t) is allowed to vary with time. In this article, we derive a simple algebraic relationship between this average regression effect and a measurement of group differences in k-sample transformation models when the random error belongs to the G(rho) family of Harrington and Fleming (1982, Biometrika 69, 553-566), the latter being equivalent to the conditional regression effect in a gamma frailty model. The models considered here are suitable for the attenuating hazard ratios that often arise in practice. The results reveal an interesting connection among the above three classes of models as alternatives to the proportional hazards assumption and add to our understanding of the behavior of the partial likelihood estimate under nonproportional hazards. The algebraic relationship provides a simple estimator under the transformation model. We develop a variance estimator based on the empirical influence function that is much easier to compute than the previously suggested resampling methods. When there is truncation in the right tail of the failure times, we propose a method of bias correction to improve the coverage properties of the confidence intervals. The estimate, its estimated variance, and the bias correction term can all be calculated with minor modifications to standard software for proportional hazards regression.  相似文献

The stability of brain networks with randomly connected excitatory and inhibitory neural populations is investigated using a simplified physiological model of brain electrical activity. Neural populations are randomly assigned to be excitatory or inhibitory and the stability of a brain network is determined by the spectrum of the network’s matrix of connection strengths. The probability that a network is stable is determined from its spectral density which is numerically determined and is approximated by a spectral distribution recently derived by Rajan and Abbott. The probability that a brain network is stable is maximum when the total connection strength into a population is approximately zero and is shown to depend on the arrangement of the excitatory and inhibitory connections and the parameters of the network. The maximum excitatory and inhibitory input into a structure allowed by stability occurs when the net input equals zero and, in contrast to networks with randomly distributed excitatory and inhibitory connections, substantially increases as the number of connections increases. Networks with the largest excitatory and inhibitory input allowed by stability have multiple marginally stable modes, are highly responsive and adaptable to external stimuli, have the same total input into each structure with minimal variance in the excitatory and inhibitory connection strengths, and have a wide range of flexible, adaptable, and complex behavior.  相似文献

When primary endpoints of randomized trials are continuous variables, the analysis of covariance (ANCOVA) with pre-treatment measurements as a covariate is often used to compare two treatment groups. In the ANCOVA, equal slopes (coefficients of pre-treatment measurements) and equal residual variances are commonly assumed. However, random allocation guarantees only equal variances of pre-treatment measurements. Unequal covariances and variances of post-treatment measurements indicate unequal slopes and, usually, unequal residual variances. For non-normal data with unequal covariances and variances of post-treatment measurements, it is known that the ANCOVA with equal slopes and equal variances using an ordinary least-squares method provides an asymptotically normal estimator for the treatment effect. However, the asymptotic variance of the estimator differs from the variance estimated from a standard formula, and its property is unclear. Furthermore, the asymptotic properties of the ANCOVA with equal slopes and unequal variances using a generalized least-squares method are unclear. In this paper, we consider non-normal data with unequal covariances and variances of post-treatment measurements, and examine the asymptotic properties of the ANCOVA with equal slopes using the variance estimated from a standard formula. Analytically, we show that the actual type I error rate, thus the coverage, of the ANCOVA with equal variances is asymptotically at a nominal level under equal sample sizes. That of the ANCOVA with unequal variances using a generalized least-squares method is asymptotically at a nominal level, even under unequal sample sizes. In conclusion, the ANCOVA with equal slopes can be asymptotically justified under random allocation.  相似文献

针对混合家系遗传参数估计,本文在假定公畜方差组分和母畜方差组分相等这一理论基础上,通过对方差分析的期望均方组成分析,提出了新的遗传力估计方法,以及某些特殊情况下的近似估计方法。通过一个估测实例比较了几种遗传力估计方法,结果表明,本文方法与全同胞组分估计最为接近,而且遗传力标准误最小,本文近似估计方法的效果也较好。对各种方法而言,资料越不平衡其差异越大。本文方法可以在一定程度上弥补全同胞分析时,因实际资料的公母畜方差组分差异过大的缺陷,具有实际可行性。此外,由于本文方法是用单因方差分析解决二因方差分析问题,计算更为简便,并可免于计算混合家系平均亲缘相关系数。  相似文献

The variance in intensities of MRI scans is a fundamental impediment for quantitative MRI analysis. Intensity values are not only highly dependent on acquisition parameters, but also on the subject and body region being scanned. This warrants the need for image normalization techniques to ensure that intensity values are consistent within tissues across different subjects and visits. Many intensity normalization methods have been developed and proven successful for the analysis of brain pathologies, but evaluation of these methods for images of the prostate region is lagging.In this paper, we compare four different normalization methods on 49 T2-w scans of prostate cancer patients: 1) the well-established histogram normalization, 2) the generalized scale normalization, 3) an extension of generalized scale normalization called generalized ball-scale normalization, and 4) a custom normalization based on healthy prostate tissue intensities. The methods are compared qualitatively and quantitatively in terms of behaviors of intensity distributions as well as impact on radiomic features.Our findings suggest that normalization based on prior knowledge of the healthy prostate tissue intensities may be the most effective way of acquiring the desired properties of normalized images. In addition, the histogram normalization method outperform the generalized scale and generalized ball-scale methods which have proven superior for other body regions.  相似文献

Variability is an important but neglected aspect of connectional neuroanatomy. The quantitative density of the 'same' corticocortical or thalamocortical connection may vary by over two orders of magnitude between different injections of the same tracer. At present, however, the frequency distribution of connection densities is unknown. Therefore, it is unclear what kind of sampling strategies or statistical methods are appropriate for quantitative studies of connectivity. Nor is it clear if the measured variability represents differences between subjects, or if it is simply a consequence of intra-individual differences resulting from experimental technique and the exact placement of tracers relative to local spatial and laminar variation in connectivity. We used quantitative measurements of the density of a large number of corticocortical and thalamocortical connections from our own laboratories and from the literature. Variability in the density of given corticocortical and thalamocortical connections is high, with the standard deviation of density proportional to the mean. The frequency distribution is close to exponential. Therefore, analysis methods relying on the normal distribution are not appropriate. We provide an appendix that gives simple statistical guidance for samples drawn from exponentially distributed data. For a given corticocortical or thalamocortical connection density, between-individual standard deviation is 0.85 to 1.25 times the within-individual standard deviation. Therefore, much of the variability reported in conventional neuroanatomical studies (with one tracer deposited per animal) is due to within-individual factors. We also find that strong, but not weak, corticocortical connections are substantially more variable than thalamocortical connections. We propose that the near exponential distribution of connection densities is a simple consequence of 'patchy' connectivity. We anticipate that connection data will be well described by the negative binomial, a class of distribution that applies to events occurring in clumped or patchy substrates. Local patchiness may be a feature of all corticocortical connections and could explain why strong corticocortical connections are more variable than strong thalamocortical connections. This idea is supported by the columnar patterns of many corticocortical but few thalamocortical connections in the literature.  相似文献

An upright, muscle-relaxed human spine, suffering from a mild functional scoliosis, caused by a small difference in leg length, is modeled as an anisotropic, elastic beam. The lower end of the beam is built-in in a fixed body, i.e., the laterally tilted pelvis. The upper end is rigidly attached to a rigid body, i.e., the supported upper part of the trunk, which is supposed to move freely in the frontal plane. It is shown that the characteristic scoliotic curvature of the spine, observed on an X-ray picture, can be reproduced by means of buckling analysis of the beam model, using realistic values of geometric and loading parameters and a properly chosen bending stiffness, which is found to be in reasonable agreement with earlier experimental findings. The analysis also shows that the muscle-relaxed upright equilibrium position of the spine is mechanically unstable.  相似文献

The binding interaction between amphotericin B and human serum albumin (HSA) has been studied using surface plasmon resonance (SPR) spectroscopy combined with a fluorescence quenching method to confirm the binding kinetic results. In this paper, the SPR method used to study the drug-protein interaction has been described in detail. The association rate constant, dissociation rate constant and the equilibrium association constant of amphotericin B binding to HSA were obtained using this method. To confirm the feasibility of the SPR method, a fluorescence quenching method was performed to obtain the equilibrium constant. In order to obtain more accurate results, experiment design was used to optimize the fluorescence quenching process. The two equilibrium association constants obtained using the two methods were 4.017 x 10(4) M(-1) (SPR) and 3.656 x 10(4) M(-1) (fluorescence quenching method) respectively.  相似文献

To develop a noninvasive, early-detection method for skin cancers, a feasibility study of multispectral image analysis was investigated. The three most frequently occurring skin cancer types, ten basal-cell carcinomas (BCCs), ten squamous-cell carcinomas (SCCs) and five malignant melanomas (MMs) were studied, along with ten normal moles. Images were acquired by a charge-coupled device camera using eight narrow-band filters ranging from 450 nm to 800 nm, at 50-nm intervals. To extract main features of these tumors, principal components analysis (PCA) was performed, because it projects the multidimensional (here, eight-dimensional) data in the direction of maximum data variance. Then, the primary PCA components for red, green, and blue subset images were analyzed in terms of hue-saturation-intensity (HSI). By hue distributions, the BCCs and SCCs were differentiated from the MMs and normal moles. Texture information was used to further classify tumor types after the HSI analysis. The texture analysis, performed using a spatial gray-level co-occurrence matrix (SGCM), could separate MMs from normal moles. The BCCs and SCCs were further studied by Fisher's linear discriminant analysis. Distribution was described as a Gaussian mixture model. By this classification procedure, seven BCCs, eight SCCs, five MMs, and ten NMs were correctly classified. Three BCCs and two SCCs were unseparable. Thus, multispectral skin cancer image analysis has potential to diagnose skin cancers.  相似文献

Recombinant measles virus nucleoprotein (N) was produced in insect cells where it bound to cellular RNA to form helical N-RNA structures. These structures were observed by electron microscopy but were too flexible for high-resolution image analysis. Removal of the C-terminal tail of N by trypsin treatment resulted in structures that were much more rigid and seemed more regular. Several methods of image analysis were employed in order to make a helical reconstruction of the digested N-RNA. During this analysis, it became clear that the apparently regular coils of digested N-RNA consisted of a series of closely related helical states. The iterative helical real space reconstruction method allowed the identification of two helical states for which a reconstruction could be calculated. The model with the highest resolution shows N monomers that consist of three domains and that are connected to their neighbours by two narrow connections, one close to the helical axis and another toward the middle of the monomers. There are no connections between N molecules in subsequent helical turns. After labelling the RNA in the structure with cis-platinum, the connection closest to the helical axis increased in density, suggesting the position of the RNA. The shapes of the monomers of the nucleoproteins of influenza virus, rabies virus (both determined before) and that of measles virus (determined here) are all similar, whereas the overall shapes of their respective N-RNAs (nucleocapsids) is very different. This is probably due to the position and number of the connections between the N subunits in the N-RNA, one for influenza virus allowing much flexibility, two for rabies virus at either end of the N molecules leading to ribbons and two for measles virus leading to the typical paramyxovirus helical nucleocapsid.  相似文献

Dynamic stabilization devices have been introduced to clinics as an alternative to rigid fixation. The stiffness of these devices varies widely, whereas the optimal stiffness, achieving a predefined stabilization of the spine, is unknown. This study was focused on the determination of stiffness values for posterior stabilization devices achieving a flexible, semi-flexible or rigid connection between two vertebrae.An extensively validated finite element model of a lumbar spinal segment L4-5 with an implanted posterior fixation device was used in this study. The model was exposed to pure moments of 7.5 and 20 Nm around the three principal anatomical directions, simulating flexion, extension, lateral bending and axial rotation. In parametrical studies, the influence of the axial and bending fixator stiffness on the spinal range of motion was investigated. In order to examine the validity of the computed results, an in-vitro study was carried out. In this, the influence of two posterior stabilization devices (DSS? and rigidly internal fixator) on the segmental stabilization was investigated.The finite element (FE)-model predicted that each load direction caused a pairing of stiffness relations between axial and bending stiffness. In flexion and extension, however, the bending stiffness had a neglectable effect on the segmental stabilization, compared to the axial stiffness. In contrast, lateral bending and axial rotation were influenced by both stiffness parameters. Except in axial rotation, the model predictions were in a good agreement with the determined in-vitro data. In axial rotation, the FE-model predicted a stiffer segmental behavior than it was determined in the in-vitro study.It is usually expected that high stiffness values are required for a posterior stabilization device to stiffen a spinal segment. We found that already small stiffness values were sufficient to cause a stiffening. Using these data, it may possible to develop implants for certain clinical indications.  相似文献
