The Tehuantepec jackrabbit (Lepus flavigularis) is an endangered species restricted to a small area in the Isthmus of Tehuantepec, Oaxaca, Mexico. To evaluate its phylogeographic
structure, population genetics, and demographic history we sequenced the mitochondrial Control Region hypervariable domain
(CR-1) for 42 individuals representing the entire species range. Phylogenetic patterns indicated that this species is subdivided
into two highly divergent clades, with an average nucleotide genetic distance of 3.7% (TrN) between them. Clades A and B are
geographically distributed in non-overlapping areas to the west and to the east of the Isthmus of Tehuantepec, respectively.
Genetic diversity indices showed reduced genetic variability in L. flavigularis when compared to other species of Lepus within main clades and within populations. This low genetic diversity coupled with the restricted distribution to very small
areas of occurrence and limited gene flow suggest that genetic drift has played an important role in the evolution of this
species. Historical demographic analysis also pointed out that these two clades underwent a recent population expansion that
started about 9,000 years ago for clade A and 3,200 years ago for clade B during the Holocene. Consequently, from the conservation
perspective our results suggest that populations included in clades A and B should be regarded as distinct evolutionary lineages. 相似文献

Random amplified polymorphic DNA (RAPD) and microsatellite markers were applied to evaluate the genetic variation in endemic
and endangered yellow catfish, Horabagrus
brachysoma sampled from three geographic locations of Western Ghat, South India river systems. In RAPD, of 32 10-mer RAPD primers screened
initially, 10 were chosen and used in a comparative analysis of H. brachysoma collected from Meenachil, Chalakkudy and Nethravathi River systems. Of the 124 total RAPD fragments amplified, 49 (39.51%)
were found to be shared by individuals of all 3 populations. The remaining 75 fragments were found to be polymorphic (60.48%).
In microsatellites, six polymorphic microsatellite loci were identified by using primers developed for Pangasius
hypophthalmus, Clarias
macrocephalus and Clarias
gariepinus. The identified loci were confirmed as microsatellite by sequencing after making a clone. The nucleotide sequences of 6 loci
were published in NCBI genbank. The number of alleles across the six loci ranged from 4 to 7 and heterozygosities ranged from
0.07 to 0.93. The mean number of alleles and effective number of alleles per locus were 5.00 and 3.314, respectively. The
average heterozygosity across all investigated samples was 0.72, indicating a significant deficiency of heterozygotes in this
species. RAPD and microsatellite methods reported a high degree of gene diversity and genetic distances depicted by UPGMA
dendrograms among the populations of H. brachysoma. 相似文献

Allothrombium
pulvinum Ewing is a common natural enemy of aphids and some other arthropods. So far, there are no studies that have addressed genetic
variation of this predatory mite. We investigated genetic variation of A. pulvinum across its whole known range in Iran. A 410 bp portion of the mitochondrial cytochrome c oxidase subunit I gene (coxI) and 797–802 bp portion of the internal transcribed spacer 2 of rDNA (ITS2) were sequenced for 55 individuals from 11 populations, resulting in 12 and 26 haplotypes, respectively. In the coxI region, haplotype and nucleotide diversities varied among populations from 0.00 to 0.90 and from 0.0000 to 0.0110, respectively.
In the ITS2 region they varied from 0.20 to 0.91 and from 0.0006 to 0.0023, respectively. For both gene regions the highest haplotype
and nucleotide diversities were detected in population Mahmoud Abad from northern Iran. Statistically significant population
differentiation (F
ST) was detected in most pair-wise population comparisons. The results of population differentiation for both gene regions were
generally congruent indicating that A. pulvinum from Iran consists of genetically different populations. This suggests that A. pulvinum comprises at least two geographically distinct populations or even more than one species. This study is an initial step towards
understanding genetic variation of A. pulvinum, a taxon for which little molecular information is available. More intensive sampling and analysis of additional DNA regions
are necessary for more detailed classification of this taxon. 相似文献

Cypripedium macranthos var. rebunense is an endangered plant endemic to Rebun Island, Japan. A proper understanding of genetic diversity is needed when conducting
conservation programs for rare and endangered species. We therefore examined the genetic diversity of C. macranthos var. rebunense using allozyme markers with a view to future conservation. Our study revealed that C. macranthos var. rebunense has relatively high genetic diversity (P was 0.62, n

e were 0.163 and 0.187, respectively) when compared with other plant taxa. The natural habitats of C. macranthos var. rebunense are geographically separated into northern and the southern populations. Disappearance of alleles and increase in homozygosity
expected as a result of the bottleneck effect were observed, particularly in the southern populations composed of a small
number of plants. As additional negative effects (inbreeding depression and further genetic drift) due to fragmentation are
predicted in these populations, the southern populations may show deterioration of genetic diversity in the near future. 相似文献

We have investigated the floral ontogeny of Arillastrum, Allosyncarpia, Stockwellia and Eucalyptopsis (of the eucalypt group, Myrtaceae) using scanning electron microscopy and light microscopy. Several critical characters for establishing relationships between these genera and to the eucalypts have been determined. The absence of compound petaline primordia in Arillastrum, Allosyncarpia, Stockwellia and Eucalyptopsis excludes these taxa from the eucalypt clade. Post-anthesis circumscissile abscission of the hypanthium above the ovary in Stockwellia, Eucalyptopsis and Allosyncarpia is evidence that these three taxa form a monophyletic group; undifferentiated perianth parts and elongated fusiform buds are characters that unite Stockwellia and Eucalyptopsis as sister taxa. No floral characters clearly associate Arillastrum with either the eucalypt clade or the clade of Stockwellia, Eucalyptopsis and Allosyncarpia.We gratefully acknowledge Clyde Dunlop and Bob Harwood (Northern Territory Herbarium) for collecting specimens of Allosyncarpia, and Bruce Gray (Atherton) for collecting specimens of Stockwellia. The maT clade of transposons is a group of transposable elements intermediate in sequence and predicted protein structure to mariner and Tc transposons, with a distribution thus far limited to a few invertebrate species. We present evidence, based on searches of publicly available databases, that the nematode Caenorhabditis briggsae has several maT-like transposons, which we have designated as CbmaT elements, dispersed throughout its genome. We also describe two additional transposon sequences that probably share their evolutionary history with the CbmaT transposons. One resembles a fold back variant of a CbmaT element, with long (380-bp) inverted terminal repeats (ITRs) that show a high degree (71%) of identity to CbmaT1. The other, which shares only the 26-bp ITR sequences with one of the CbmaT variants, is present in eight nearly identical copies, but does not have a transposase gene and may therefore be cross mobilised by a CbmaT transposase. Using PCR-based mobility assays, we show that CbmaT1 transposons are capable of excising from the C. briggsae genome. CbmaT1 excised approximately 500 times less frequently than Tcb1 in the reference strain AF16, but both CbmaT1 and Tcb1 excised at extremely high frequencies in the HK105 strain. The HK105 strain also exhibited a high frequency of spontaneous induction of unc-22 mutants, suggesting that it may be a mutator strain of C. briggsae. 相似文献

Summary A new species of Dioscorea from Morondava prefecture in Western Madagascar is described. Dioscorea bako Wilkin differs from D. alatipes Burkill & H. Perr. by its usually shallowly and irregularly lobed leaf margins, broader leaf blade (grey-green below), longer
petiole and the broadly cuneate area where the petiole is inserted onto the leaf blade in the basal sinus. It is pubescent,
especially densely on young shoots and inflorescences. It is endemic to Morondava Préfecture and is endangered under IUCN
Red List category criteria (IUCN 2001). The unusual morphological features of its inflorescences, some of which are shared
with D. alatipes are discussed. Dioscorea bako is a favoured food source for people of the Menabe region and is reported by them to be increasingly hard to find. Immediate
conservation measures are necessary to ensure that it remains extant and, in the medium to long term, research should be undertaken

Kalopanax pictus (Araliaceae) is a long-lived woody species primarily distributed throughout East Asia. This species is regarded as medically
and ecologically important in Korea. We used starch gel electrophoresis to investigate its allozyme variation and genetic
structure in samples from Korean populations (both natural and cultivated). Our analysis of 10 enzymes revealed 18 loci, of
which 10 were polymorphic (35.6%). Genetic diversity at the species and population levels were 0.200 and 0.149, respectively,
with the mean for cultivated populations (0.124) being lower than for natural (wild) populations (0.181). Asexual and sexual
reproduction modes, perennial habitat, and longevity all were possible factors contributing to such high diversity. An indirect
estimate of the number of migrants per generation (N
m
= 1.08) indicated that gene flow was not extensive among these Korean populations. Therefore, we suggest that geographical
distance as well as reproductive isolation between wild and cultivated plants may play roles in shaping the population structure

ANGUSTIFOLIA (AN), a plant homolog of C-terminal binding protein, controls the polar elongation of leaf cells and the trichome-branching pattern in Arabidopsis thaliana. In the present study, degenerate PCR was used to isolate an ortholog of AN, referred to as LgAN, from larch (Larix gmelinii). The LgAN cDNA is predicted to encode a protein of 646 amino acids that shows striking sequence similarity to AN proteins from other plants. The predicted amino acid sequence has a conserved NAD-dependent 2-hydroxy acid dehydrogenase (D2-HDH) motif and a plant AN-specific LxCxE/D motif at its N-terminus, as well as a plant-specific long C-terminal region. The LgAN gene is a single-copy gene that is expressed in all larch tissues. Expression of the LgAN cDNA rescued the leaf width and trichome-branching pattern defects in the angustifolia-1 (an-1) mutant of Arabidopsis, showing that the LgAN gene has effects complementary to those of AN. P transposons belong to the eukaryotic DNA transposons, which are transposed by a cut and paste mechanism using a P-element-coded transposase. They have been detected in Drosophila, and reside as single copies and stable homologous sequences in many vertebrate species. We present the P elements Pcin1, Pcin2 and Pcin3 from Ciona intestinalis, a species of the most primitive chordates, and compare them with those from Ciona savignyi. They showed typical DNA transposon structures, namely terminal inverted repeats and target site duplications. The coding
region of Pcin1 consisted of 13 small exons that could be translated into a P-transposon-homologous protein. C. intestinalis and C. savignyi displayed nearly the same phenotype. However, their P elements were highly divergent and the assumed P transposase from C. intestinalis was more closely related to the transposase from Drosophila melanogaster than to the transposase of C. savignyi. The present study showed that P elements with typical features of transposable DNA elements may be found already at the base of the chordate lineage.

A new source of resistance to the pathotype 4 isolate of Turnip mosaic virus (TuMV) CDN 1 has been identified in Brassica napus (oilseed rape). Analysis of segregation of resistance to TuMV isolate CDN 1 in a backcross generation following a cross between a resistant and a susceptible B. napus line showed that the resistance was dominant and monogenic. Molecular markers linked to this dominant resistance were identified using amplified fragment length polymorphism (AFLP) and microsatellite bulk segregant analysis. Bulks consisted of individuals from a BC1 population with the resistant or the susceptible phenotype following challenge with CDN 1. One AFLP and six microsatellite markers were associated with the resistance locus, named TuRB03, and these mapped to the same region on chromosome N6 as a previously mapped TuMV resistance gene TuRB01. Further testing of TuRB03 with other TuMV isolates showed that it was not effective against all pathotype 4 isolates. It was effective against some, but not all pathotype 3 isolates tested. It provided further resolution of TuMV pathotypes by sub-dividing pathotypes 3 and 4. TuRB03 also provides a new source of resistance for combining with other resistances in our attempts to generate durable resistance to this virus. 相似文献

Notocactus scopa cv. Soonjung was subjected to in planta Agrobacterium tumefaciens-mediated transformation with vacuum infiltration, pin-pricking, and a combination of the two methods. The pin-pricking combined with vacuum infiltration (20-30 cmHg for 15 min) resulted in a transformation efficiency of 67-100%, and the expression of the uidA and nptII genes was detected in transformed cactus. The established in planta transformation technique generated a transgenic cactus with higher transformation efficiency, shortened selection process, and stable gene expression via asexual reproduction. All of the results showed that the in planta transformation method utilized in the current study provided an efficient and time-saving procedure for the delivery of genes into the cactus genome, and that this technique can be applied to other asexually reproducing succulent plant species. 相似文献
