In susceptible oat, toxaphene inhibits photosynthetic electron flow and concomitant ATP synthesis. Although the rate of 14CO2 assimilation is apparently not affected markedly there is an increase in dry weight of leaves contacting the pesticide. The labelling patterns in leaf sections exposed to 14CO2 are similar for both toxaphene-treated and untreated seedlings. However, if given a period in darkness before extraction it is evident that assimilation products in leaf sections from toxaphene-treated leaves remain as small M, materials, including substantial amounts of sugars, whereas in untreated controls these were converted to polymeric materials. In toxaphene-treated seedlings the translocation of assimilation products to the roots is decreased and sucrose accumulates in the leaves.  相似文献

Changes in leaf growth, photosynthetic efficiency, and incorporation pattern of photosynthetically fixed 14CO2 in leaves 1 and 2 from plant apex, in roots, and rhizome induced in Curcuma by growing in a solution culture at Fe concentration of 0 and 5.6 g m–3 were studied. 14C was incorporated into primary metabolites (sugars, amino acids, and organic acids) and secondary metabolites (essential oil and curcumin). Fe deficiency resulted in a decrease in leaf area, its fresh and dry mass, chlorophyll (Chl) content, and CO2 exchange rate at all leaf positions. The rate of 14CO2 fixation declined with leaf position, maximum being in the youngest leaf. Fe deficiency resulted in higher accumulation of sugars, amino acids, and organic acids in leaves at both positions. This is due to poor translocation of metabolites. Roots and rhizomes of Fe-deficient plants had lower concentrations of total photosynthate, sugars, and amino acids whereas organic acid concentration was higher in rhizomes. 14CO2 incorporation in essential oil was lower in the youngest leaf, as well as incorporation in curcumin content in rhizome. Fe deficiency influenced leaf area, its fresh and dry masses, CO2 exchange rate, and oil and curcumin accumulation by affecting translocation of assimilated photosynthates.  相似文献

Changes in 14C2H4 metabolism in the abscission zone were monitored during cotton (cv. Deltapine 16) leaf abscission. Rates of 14C2H4 oxidation to 14CO2 and tissue incorporation in abscission zone segments cut from the second true leaf of nonabscising leaves of intact plants were similar (about 200 disintegrations per minute per 0.1 gram dry weight per 5.5 hours) and relatively constant over a 5-day period. Deblading to induce abscission caused a dramatic rise in 14C2H4 oxidation, but tissue incorporation was not markedly affected. This rise occurred well before abscission, reaching a peak of 1,375 disintegrations per minute per 0.1 gram dry weight per 5.5 hours 2 days after deblading when abscission was 40%. The rate then gradually declined, but on day 5 when abscission reached completion, it was still nearly three times higher than in segments from nonabscising leaves. Application of 0.1 millimolar abscisic acid in lanolin to the debladed petiole ends increased the per cent abscission slightly and initially stimulated 14C2H4 oxidation. In contrast, naphthaleneacetic acid applied in a similar manner delayed and markedly inhibited both abscission and 14C2H4 oxidation.  相似文献

Changes in leaf growth, net photosynthetic rate (P N), incorporation pattern of photosynthetically fixed 14CO2 in leaves 1–4 from top, roots, and rhizome, and in essential oil and curcumin contents were studied in turmeric plants grown in nutrient solution at boron (B) concentrations of 0 and 0.5 g m-3. B deficiency resulted in decrease in leaf area, fresh and dry mass, chlorophyll (Chl) content, and P N and total 14CO2 incorporated at all leaf positions, the maximum effect being in young growing leaves. The incorporation of 14CO2 declined with leaf position being maximal in the youngest leaf. B deficiency resulted in reduced accumulation of sugars, amino acids, and organic acids at all leaf positions. Translocation of the metabolites towards rhizome and roots decreased. In rhizome, the amount of amino acids increased but content of organic acids did not show any change, whereas in roots there was decrease in contents of these metabolites as a result of B deficiency. Photoassimilate partitioning to essential oil in leaf and to curcumin in rhizome decreased. Although the curcumin content of rhizome increased due to B deficiency, the overall rhizome yield and curcumin yield decreased. The influence of B deficiency on leaf area, fresh and dry masses, CO2 exchange rate, oil content, and rhizome and curcumin yields can be ascribed to reduced photosynthate formation and translocation.  相似文献

Changes in growth parameters, carbon assimilation efficiency, and utilization of 14CO2 assimilate into alkaloids in plant parts were investigated at whole plant level by treatment of Catharanthus roseus with gibberellic acid (GA). Application of GA (1 000 g m−3) resulted in changes in leaf morphology, increase in stem elongation, leaf and internode length, plant height, and decrease in biomass content. Phenotypic changes were accompanied by decrease in contents of chlorophylls and in photosynthetic capacity. GA application resulted in higher % of total alkaloids accumulated in leaf, stem, and root. GA treatment produced negative phenotypic response in total biomass production but positive response in content of total alkaloids in leaf, stem, and roots. 14C assimilate partitioning revealed that 14C distribution in leaf, stem, and root of treated plants was higher than in untreated and variations were observed in contents of metabolites as sugars, amino acids, and organic acids. Capacity to utilize current fixed 14C derived assimilates for alkaloid production was high in leaves but low in roots of treated plants despite higher content of 14C metabolites such as sugars, amino acids, and organic acids. In spite of higher availability of metabolites, their utilization into alkaloid production is low in GA-treated roots.  相似文献

Fast growth and early development in barley are used in breeding programmes to improve the water use efficiency and transpiration efficiency of this crop in Mediterranean conditions. Here, we examine the use of several simple traits based on the structure and stable isotope composition of seedling leaves to assess differences in early vigour, phenology and grain yield, and also the interaction with low temperatures in barley. A set of 260 F8 lines of two-row barley (Hordeum vulgare L.) derived from the cross of Tadmor and WI 2291 were cultivated in two locations in northwest Syria. Total chlorophyll content on an area basis (SPAD) and specific leaf dry weight (SLDW) were measured in recently fully expanded intact leaves of seedlings. Total leaf area and total dry weight per seedling were evaluated in the same seedlings. The stable isotope compositions of carbon and nitrogen (δ13C and δ15N, respectively) were analyzed in the same leaves on a subset of 75 genotypes. Number of days from planting to heading and grain yield were recorded at both sites. The grain yield measured at both locations was positively correlated with the SPAD value of seedlings, but showed no relationship with SLDW. Days to heading was negatively correlated with SPAD values. Regarding early vigour, a negative relationship between the SLDW and the total leaf area of seedlings was observed. However, no relationship between the δ13C of seedlings and early vigour was observed, except when only the genotypes most resistant to low temperatures (i.e. showing the highest SPAD values) were considered. This subset of genotypes showed negative relationships between δ13C and either total leaf area or total dry weight. In addition, δ15N was negatively correlated with SPAD only within the high-SPAD genotypes. This suggests that within the genotypes resistant to low temperatures, those with higher chlorophyll content assimilate more nitrogen from nitrate.  相似文献

Changes in growth, CO2 exchange rate, and distribution of photosynthetically fixed 14CO2 into the primary photosynthetic metabolic pool (sugars, amino acids and organic acids) and essential oil accumulation were determined in leaves (leaf positions 1-6 from apex) of developing peppermint grown in a solution culture at Zn concentrations of 0 and 0.05 g m-3. There was a significant decrease in 14C incorporation in total, ethanol-soluble and ethanol-insoluble fractions in Zn deficient plants at all leaf positions. 14C incorporated in essential oil and in sugars were significantly higher in leaf pairs 1 to 3 than in leaf pairs 4 to 6. 14C incorporation into amino acids and organic acids was higher in all leaf pairs in Zn deficient plants. Statistical analysis showed a positive significant association between Zn content of leaf and 14C incorporation into ethanol-soluble fraction and sugars and a negative correlation with 14C incorporation into amino acids and organic acids. Hence the content of sugars in leaves significantly influences essential oil accumulation under Zn stress. Regenerants from tobacco(Nicotiana tabacum L. cv. White Burley) leaf segments cultivatedin vitro in vessels with solid agar medium under usual conditions (plantlets) grew under very low irradiance (I = 40 μxmol m?2 s?1), very high relative humidity (more than 90%) and decreased CO2 concentration (ca) during light period. In comparison with seedlings of a similar number of leaves and similar total leaf area grown in sand and nutrient solution, the plantlets had lower dry mass of shoots and roots per plant and thinner leaves almost without trichomes and epicuticular waxes. Due to a low transpiration rate under high relative humidity the water potential of plantlet leaves was higher than that of seedling leaves and the difference in water potential between leaves and medium was lowei. The rate of water loss from leaves detached from plantlets was considerably faster than that from seedlings under the same conditions (I = 110 μrnol m?2; s?1, temperature 30 °C, relative humidity 50 %). Net photosynthetie rates (Pn) of leaves of plantlets and seedlings measured under saturating I, natural ca and the leaf temperature 20 °C were similar, nevertheless the shape of curves relating Pn to c» indicated some differences in photosynthetie parameters(e.g. saturation of Pn under lower ca> higher CO2 compensation concentration in plantlets than in seedlings). Similarly compensation and saturating I were lower in plantlets than in seedlings. The shape of transpiration curves as well as the expressive linear phases of PN(ca) and PN(I) curves of plantlet leaves indicated ineffective stomatal control of gas exchance. A comparative study of tryptophan conversion in different regions of the sunflower seedling indicates that the regions most active in converting tryptophan on a pathway to auxin are the root apical segments and young leaves; next highest in activity is the cotyledonary tissue. The stem apex proper with leaf primordia is less active than the above regions in converting the auxin precursor. Hypocotyl tissue was observed to be least active. Pre-treatment of the apical bud region of the stem with gibberellic acid (GA) gives rise to tryptophan conversion rates which are 2.1 times those in untreated seedlings. The enhanced tryptophan conversion in the apical bud is followed by an increased elongation rate of the 1st internode which is 2.2 times that in the 1st internode of untreated seedlings. Treatment of the seedlings with Cycocel [(2-chloroethyl)trimethylamnionium chloride] does not reduce tryptophan conversion in the apical bud region of the seedling although elongation of the stem is greatly retarded. Indoleacetic acid (IAA) destruction in cell free preparations as well as in whole sections of the elongating region of the seedling stem was studied. IAA-1-14C destruction rates with the release of 14CO2 in whole sections of 1st internode tissue were approximately 3 times those in cell free preparations of the same region. No significant changes in IAA destruction rates in seedlings pre-treated with GA or Cycocel were observed.  相似文献

Changes in growth parameters and 14CO2 and [U-14C]-sucrose incorporation into the primary metabolic pools and essential oil were investigated in leaves and stems of M. spicata treated with etherel and gibberellic acid (GA). Compared to the control, GA and etherel treatments induced significant phenotypic changes and a decrease in chlorophyll content, CO2 exchange rate, and stomatal conductance. Treatment with etherel led to increased total incorporation of 14CO2 into the leaves wheras total incorporation from 14C sucrose was decreased. When 14CO2 was fed, the incorporation into the ethanol soluble fraction, sugars, organic acids, and essential oil was significantly higher in etherel treated leaves than in the control. However, [U-14C]-sucrose feeding led to decreased label incorporation in the ethanol-soluble fraction, sugars, organic acids, and essential oils compared to the control. When 14CO2 was fed to GA treated leaves, label incorporation in ethanol-insoluble fraction, sugars, and oils was significantly higher than in the control. In contrast, when [U-14C]-sucrose was fed the incorporation in the ethanol soluble fraction, sugars, organic acids, and oil was significantly lower than in the control. Hence the hormone treatment induces a differential utilization of precursors for oil biosynthesis and accumulation and differences in partitioning of label between leaf and stem. Etherel and GA influence the partitioning of primary photosynthetic metabolites and thus modify plant growth and essential oil accumulation. To obtain basic information for evaluating critical loads of acid deposition for protecting Japanese beech forests, growth, net photosynthesis and leaf nutrient status of Fagus crenata seedlings grown for two growing seasons in brown forest soil acidified with H2SO4 or HNO3 solution were investigated. The whole-plant dry mass of the seedlings grown in the soil acidified by the addition of H2SO4 or HNO3 solution was significantly less than that of the seedlings grown in the control soil not supplemented with H+ as H2SO4 or HNO3 solution. However, the degrees of reduction in the whole-plant dry mass and net photosynthetic rate of the seedlings grown in the soil acidified by the addition of H+ as H2SO4 solution at 100 mg l–1 on the basis of air-dried soil volume (S-100 treatment) were greater than those of the seedlings grown in the soil acidified by the addition of H+ as HNO3 solution at 100 mg l–1 (N-100 treatment). The concentrations of Al and Mn in the leaves of the seedlings grown in the S-100 treatment were significantly higher than those in the N-100 treatment. A positive correlation was obtained between the molar ratio of (Ca+Mg+K)/(Al+Mn) in the soil solution and the relative whole-plant dry mass of the seedlings grown in the acidified soils to that of the seedlings grown in the control soil. Based on the results, we concluded that the negative effects of soil acidification due to sulfate deposition are greater than those of soil acidification due to nitrate deposition on growth, net photosynthesis and leaf nutrient status of F. crenata, and that the molar ratio of (Ca+Mg+K)/(Al+Mn) in soil solution is a suitable soil parameter for evaluating critical loads of acid deposition in efforts to protect F. crenata forests in Japan.  相似文献

Hamamelitol is an unusual branched-chain sugar alcohol previously suggested to function as a leaf compatible solute. In this study, we have examined the leaf metabolism and intracelluiar compartmentalization of hamamelitol and other soluble sugars during long-term water stress treatment of Hedera helix (English ivy). Total leaf hamamelitol content was relatively low in greenhouse control plants, but increased 2-fold during water stress treatment to levels approaching those observed in field-grown plants (6–7 μmol g?1 fresh weight). Using density gradient fractionation with non-aqueous solvents, we showed that hamamelitol occurs primarily in the cytoplasm and vacuoles of leaf mesophyll cells. During water stress treatment most of the increase in leaf hamamelitol occurred in the mesophyll cytoplasm, compensating osmotically for a decrease in cytoplasmic sucrose concentration. The maximum concentration of cytoplasmic hamamelitol was 155 mol m?3 and occurred in field-grown plants. Labelling experiments showed that hamamelitol is slowly synthesized from 14CO2 in leaves of H. helix, but is very long-lived (estimated t1/2 of 4 years). Together, these data indicate that hamamelitol probably functions during long-term stress conditions as an osmotically active, compatible solute in plant leaves. We suggest that the signal for enhanced accumulation of hamamelitol during the water stress treatment was initiated by decreased plant growth and increased leaf sucrose hydrolysis.  相似文献

Partitioning of current photosynthates towards primary metabolites and its simultaneous incorporation in leaf alkaloids was investigated in developing leaves of medicinally important Catharanthus roseus. Of the total 14CO2 assimilated, the leaves at positions 1–6 fixed 8, 22, 25, 19, 13, and 8 %, respectively, and stem 3 %. Leaf fresh mass, chlorophyll content, and CO2 exchange rate increased up to the third leaf. The total alkaloid content was highest in young actively growing leaves, which declined with age. Total 14C fixed and its content in ethanol soluble fraction increased up to the third leaf and then declined. The 14C content in primary metabolites such as sugars and organic acids was also highest in the 3rd leaf. The utilization of 14C assimilates into alkaloids was maximum in youngest leaf which declined with leaf age. Hence the capacity to synthesize alkaloids was highest in young growing leaves and metabolites from photosynthetic pathway were most efficiently utilized and incorporated into alkaloid biosynthetic pathway by young growing leaves.  相似文献

The allometric equation, y=gxh, was applied monthly to the relationships between two different dimensions of tree seedlings of hinoki cypress (Chamaecyparis obtusa) during a course of self-thinning from April 1990 to March 1991 to detect differences in biomass allocation among individuals. As the h-value in the allometry of crown length and seedling height was greater than unity for all seasons, crown ratio became greater as seedling height increased. Leaf weight ratio increased with increasing seedling size because the h-value in the allometry of leaf dry weight and whole seedling dry weight was greater than unity in every month. Therefore, smaller seedlings are disadvantageous to photosynthetic production by leaves. In contrast, the leaf area ratio was constant irrespective of seedling size because the h-value in the allometry of leaf area and whole seedling dry weight was nearly equal to unity in most seasons. In addition, because the h-value in the allometry of leaf area and leaf dry weight was less than unity in all seasons, specific leaf area decreased with an increase in leaf dry weight, indicating that smaller seedlings adapt to low light environments by possessing shade leaves. Root weight ratio decreased with increasing seedling size because the h-value in the allometry of root dry weight and whole seedling dry weight was less than unity in most seasons.  相似文献

Sugar maple (Acer saccharum Marsh.) seedlings were grown in a nursery for three years in 13, 25, 45 and 100 per cent of full daylight. During the third year of growth, the rates of their apparent photosynthesis and respiration were measured periodically with an infra-red gas analyzer at various light intensities and normal CO2 concentration. In addition, the rates of apparent photosynthesis of a single attached leaf of the same seedlings were measured at saturating light intensity, hut varying CO2 concentrations. An increase in the light intensity in which seedlings were grown had no effect on their height or mean leaf area, hut resulted in thicker leaves, an increase in the total leaf area per seedling due to an increase in the number of leaves, an increase in the dry weight especially of roots and a decrease in the chlorophyll content of leaves. Throughout the growing season seedlings grown in full daylight, as compared with those grown in lower light intensities, had the lowest rates of apparent photosynthesis measured at standard conditions (21,600 lux light intensity and 300 ul/l of CO2), when this was expressed per unit leaf area, hut the highest rates on a per seedling basis. Thus dry matter production attained at the end of the growing season correlated positively with the photosynthetic rate per seedling, but not per unit leaf area. The rates of apparent photosynthesis of seedlings grown at lower light intensities were more responsive to changes in light intensity or CO2 concentration than those of seedlings grown in full daylight intensity.  相似文献

Sink-to-source transition was studied in developing sugarcane (Saccharum interspecific variety L62–96) leaves. Fully-expanded, mature sugarcane leaves were fed 14CO2 for 20 minutes, incorporating about 617 Bq. After five hours the leaves of each plant were cut into 1-cm-length segments that were weighed and then placed in scintillation cocktail for counting. All leaves younger than the leaf fed 14CO2 imported labeled photoassimilate. Three to four leaves had both importing and non-importing regions within the blade and a distinct transition region between them. A transition region was observed in leaves which had expanded to between 30 and 90 % of final blade length. Radioactivity per gram fresh weight was calculated as a measure of sink strength. Sink strength was greatest in the youngest leaf and declined with leaf age. The results of this study indicate that 1) import of photosynthate by developing sugarcane leaves occurs over a longer span of developmental ages than in dicotyledonous leaves and 2) the actual tissue region undergoing transition within such a leaf can be resolved as narrow zone between the importing and non-importing regions.  相似文献

Our objective was to examine alterations in carbohydrate status of leaf meristems that are associated with nitrogen-induced changes in leaf elongation rates of tall fescue (Festuca arundinacea Schreb.). Dark respiration rates, concentrations of nonstructural carbohydrates, and soluble proteins were measured in leaf intercalary meristems and adjacent segments of elongating leaves. The two genotypes used differed by 43% in leaf elongation rate. Application of high nitrogen (336 kilograms per hectare) resulted in 140% higher leaf elongation rate when compared to plants receiving low nitrogen (22 kilograms per hectare). Leaf meristems of plants receiving high and low nitrogen had dark respiration rates of 5.4 and 2.9 microliters O2 consumed per milligram structural dry weight per hour, respectively. Concentrations of soluble proteins were lower while concentrations of fructan tended to be slightly higher in leaf meristems of low-nitrogen plants when compared to high-nitrogen plants. Concentrations of reducing sugars, nonreducing sugars, and takadiastase-soluble carbohydrate of leaf meristems were not affected by nitrogen treatment. Total nonstructural carbohydrates of leaf meristems averaged 44 and 39% of dry weight for low- and high-nitrogen plants, respectively. Within the leaf meristem, approximately 74 and 34% of the pool of total nonstructural carbohydrate could be consumed per day in high- and low-nitrogen plants, respectively, assuming no carbohydrate import to the meristem occurred. Plants were able to maintain high concentrations of nonstructural carbohydrates in leaf meristems despite a 3-fold range in leaf elongation rates, suggesting that carbohydrate synthesis and transport to leaf intercalary meristems may not limit leaf growth of these genotypes.  相似文献

A two-component model of growth and maintenance respiration is used to study the response of northern red oak (Quercus rubra L.) seedlings and 32-year-old trees to sub-ambient (10 μmol h; cumulative dose based on 7 h daily mean), ambient (43 μmol h), and twice-ambient (85 μmolh) ozone. The relative growth rates (RGR) of leaves sampled from seedlings and trees were similar across treatments, as were specific leaf respiration rates (SRR). Growth coefficients estimated from the SRR versus RGR relationship averaged 25-3 mol CO2 kg?1 leaf dry mass produced for seedlings and 21-5 mol kg?1 for trees. Maintenance coefficients ranged from 0-89 to 1-07 mol CO2 kg?1 leaf dry mass d?1 for seedlings and from 0-64 to 0-84 mol kg-1 d?1 for trees. Neither coefficient was affected by ozone. Leaves sampled throughout the growing season also showed little response of respiration to ozone. This occurred despite a 30% reduction in net photosynthesis for trees grown at twice-ambient ozone. These results suggest that growth and maintenance respiration in young northern red oak leaves are not affected by ozone and that in older leaves injury can occur without a parallel increase in so-called ‘maintenance’ respiration.  相似文献

The application of 6N-benzyladenine (BA) to primary bean-leaves attached to the intact plant resulted in increased leaf area. This was due to an extension of the duration of the period of leaf expansion. The dry-weight percentage of untreated leaves decreased continuously from emergence to abscission. In BA treated leaves this decrease stopped with the cessation of leaf expansion. Net chlorophyll synthesis occurs in untreated leaves as long as they continue to expand, shortly afterwards net chlorophyll decomposition starts. Benzyladenine treatments diminished the rate of chlorophyll synthesis immediately after its first application, but prevented the loss of chlorophyll afterwards. The abscission of primary leaves was also delayed by BA. During the period of rapid leaf expansion in slices cut from BA treated leaves, sodium absorption rates were lower than in those from untreated leaves. From the age of 14 days after sowing, till abscission time of untreated primary leaves, the BA treatment did not significantly affect sodium absorption rates by leaf slices.  相似文献

To understand the physiology of rice under seawater salinity, potted rice plants were irrigated with different concentrations of Japan seawater (electrical conductivity 0.9, 5.7, 11.5, or 21.5 mS cm-1) from 10 d after transplanting (DAT) to 35 DAT, and from 75 to 100 DAT. Seawater salinity decreased the net photosynthetic rate, stomatal conductance, intercellular CO2 concentration, transpiration rate, leaf water and osmotic potentials, and relative water content, and increased leaf temperature. The contents of chlorophylls, carotenoids, and total sugars significantly decreased in the leaves but content of non-reducing sugars decreased only slightly. With increasing salinity the Na+ concentration increased, while Ca2+, Mn2+, and K+ concentrations decreased. Salinity decreased the contents of sugars and proteins, dry mass, and rate of dry mater accumulation in developing grains.  相似文献
