Benign and malignant connective tissue tumors consist of a fibrous component that contains varying amounts of one or more types of bone or other calcified tissue. Diagnosis of these connective tissue tumors often poses challenges for pathologists, because it is difficult to differentiate the organic matrix of osteoid from hyalinized stroma. To establish a definitive diagnosis, it sometimes is advantageous to demonstrate histologically by special staining either the type of calcification or the presence or absence of calcification. We compared the efficacy of methylene blue-acid fuchsin (MB-AF) to hematoxylin and eosin (H-E) for connective tissue tumors suspected to contain calcifications and to devise an optimal staining technique for calcification that would be specific, simple, and cost- and time-effective. We examined 50 benign and 45 malignant connective tissue tumors that were suspected to contain calcifications. Sections were stained with H-E and MB-AF and evaluated. MB-AF stained bone pink, which contrasted with blue soft tissue. After MB-AF staining, osteoid was faint pink in a blue stromal background. Osteoid was not visualized in H-E stained sections; it was stained the same shade of pink as stromal tissue. Dystrophic calcification and cementum could be identified equally well using either staining technique, but contrast was better after H-E staining. MB-AF staining of bone was comparable to H-E staining and could be used effectively to stain bone and osteoid. MB-AF is a simple, single step procedure. It also stains cementum blue with faint blue rimming and dystrophic calcification bluish-pink, but it cannot be used as a specific stain for types of calcification other than bone and osteoid.  相似文献

High concentrations of dimethyl sulfoxide (DMSO) have been shown to cause gross structural changes in collagen fibers in vitro. As endometrial fibrosis is a major cause of infertility in mares, the effect of serial DMSO infusions on periglandular fibrosis of Category II endometria was investigated. Initially, six mares with Category I endometria were serially infused with DMSO to determine whether such therapy would incite fibrosis. Four mares, serially infused with saline, served as controls in each experiment. One hundred milliliters of 75% DMSO was infused initially, followed by the same volume of 25% DMSO daily for 6 d. Endometrial biopsies obtained 21 d after the last infusion displayed no significant difference in the incidence of periglandular fibrosis when compared with the Day 0 biopsies. Examination of treated uteri at necropsy showed no gross evidence of adhesions or ulcerations.  相似文献

Fluid, particles, and cells are taken up from the peritoneal cavity by lymphatic drainage units, which, in the mouse and rat, are located along the peritoneal surface of the muscular portion of the diaphragm. The drainage units are composed of three specifically differentiated components: a lymphatic lacuna, a covering of lacunar mesothelium, and intervening submesothelial connective tissue. The units are drained by connecting lymphatic vessels that cross the diaphragm to empty into collecting lymphatic vessels running along the pleural surface of the diaphragm. The collecting lymphatics empty into parasternal lymphatic trunks. In this report, we briefly review critical features of the drainage apparatus and describe new observations, summarized below, about their structure. Around the rim of stomata, the mesothelial openings that lead into the lymphatic lacunae, plasma membranes of lacunar mesothelial cells and of lacunar endothelial cells abut but are not linked to one another by recognizable junctional specializations. Lacunar endothelial cells often extend valve-like processes that bridge the distal end of the channel beneath the stoma. The configuration of the endothelial processes may be complex. Occasionally, processes from fibroblasts in the submesothelial connective tissue adjacent to stomata make contact with the interstitial surface of lacunar endothelial cells. A discontinuous elastic layer in the submesothelial connective tissue spans the roof of each lacuna. Connecting and collecting lymphatics, which drain lymphatic lacunae, possess endothelial valves. Possible functions for each of these newly described structural features are discussed.  相似文献

An accurate identification of telocytes (TCs) was limited because of the heterogeneity of cell types expressing the markers attributed to TCs. Some endothelial lineage cells also could fit within the pattern of TCs. Such endothelial cells could line conjunctival lacunae previously assessed by laser confocal microscopy. We have been suggested that an accurate distinction of TCs from endothelial cells in the human eye conjunctiva could be achieved by use of CD31, CD34 and D2‐40 (podoplanin); and that the conjunctival lacunae are in fact lymphatic. We aimed as testing the hypothesis by an immunohistochemical study on human eye conjunctiva biopsy samples. Samples of human eye conjunctiva from 30 patients were evaluated immunohistochemically by use of the primary antibodies: CD34, D2‐40 and CD31. D2‐40 was equally expressed within epithelia and laminae propria. Basal epithelial cells were D2‐40 positive. Within the stromal compartment, the lymphatic marker D2‐40 labelled several lymphatic vessels. CD31 labelled both vascular and lymphatic endothelial cells within the lamina propria. When capillary lymphatics were tangentially cut, they gave the false appearance of telocytes. Blood endothelial cells expressed CD34, whereas lymphatic endothelial cells did not. Stromal CD34‐expressing cells/telocytes were found building a consistent pan‐stromal network which was equally CD31‐negative and D2‐40‐negative. The conjunctival lymphatic lacunae seem to represent a peculiar anatomic feature of eye conjunctiva. They are embedded within a CD34‐expressing stromal network of TCs. The negative expression of CD31 and D2‐40 should be tested when discriminating CD34‐expressing TCs.  相似文献

To investigate the changes of cardiomyocyte inflammation and fibrosis factors in heart of carotid artery balloon injury inflammatory rat model. Using rat carotid artery balloon injury model to detect left ventricular characteristics at 2 h, 2 days and 14 days after surgery using hematoxylin-eosin (H&E) gross stain, Masson's trichome stain and Western blot analysis for inflammatory and fibrosis-induced factors, tumour necrosis factor α (TNFα), JNK1, P38α, connective tissue growth factor (CTGF), SP1 and transforming growth factor β (TGFβ) protein expressions. The rat carotid arteries were injured after 2 h, 2 days and 14 days. Balloon-angioplasty to H&E stain results showed the increasing trend of left ventricular wall at 2 h and 2 days; then, the left ventricular wall became thinner, and the left ventricular chamber became enlarged and dilated after 14 days of carotid artery balloon injury. In addition, the Masson's trichome stain results showed that the left ventricular section has fibrosis-related blue staining (collagen) at 2 and 14 days after rat carotid artery balloon injury, and became even more severe at 14 days. Furthermore, we observed the protein expression level changs, which include TNFα, JNK1, P38α, CTGF, SP1 and TGFβ using Western blotting assay. All proteins were induced at 2 h, 2 days and then reached the maximal level at 14 days. The vessel inflammation was associated with cardiac inflammatory and fibrosis effects during or after carotid artery balloon injury.  相似文献

The effects upon equine endometrial histological features produced by 10 to 30% concentrations of dimethyl sulfoxide (DMSO) in sterile saline were compared with the effects of sterile saline (0.9% NaCl) alone as an intrauterine infusion therapy in 16 barren mares. No harmful histological changes were noted (P > 0.05) as a result of the therapy. Thirty percent intrauterine DMSO therapy produced a significant (P < 0.01) improvement (i.e., reduction of chronic inflammatory cell infiltrates and reduction of periglandular fibrosis) in endometrial biopsy classification in 18 of the 27 barren mares evaluated; whereas only 2 of 18 barren mares improved following intrauterine saline treatment in the control group. In subsequent breeding trials, the pregnancy rates following intrauterine therapy were no different between DMSO-treated mares and saline-treated control mares (P > 0.05); however, there was a trend toward a higher pregnancy rate following DMSO therapy. The inability to control for sire variability in the breeding trials may have contributed toward the result that no significant difference could be demonstrated in the pregnancy rates between treated and control mares.  相似文献

We investigated the distribution of myofibroblasts and CD34-positive stromal cells in normal gallbladder and its pathological conditions (cholecystitis, n=25) using immunohistochemistry and in situ hybridization. In the wall of normal gallbladder, myofibroblasts were generally absent from all layers, but many CD34-positive stromal cells were observed in the connective tissue layer. In chronic cholecystitis with mild perimuscular fibrosis, a small to moderate number of myofibroblasts appeared in the mucosal layer. In chronic cholecystitis with marked perimuscular fibrosis, a small to large number of myofibroblasts appeared predominantly in the connective tissue layer, whereas the number of CD34-positive stromal cells decreased at the same location, although the number of myofibroblasts increased. In chronic cholecystitis with xanthogranulomatous foci, a small to large number of myofibroblasts were observed in the periphery of the xanthogranulomatous reaction and adjacent area. In contrast, CD34-positive stromal cells were completely absent or were limited to the area just around the xanthogranulomatous reaction. Induction of collagen type I and III mRNA was predominantly observed in the cytoplasm of myofibroblasts associated with the marked fibrosis, which consisted primarily of mature collagen fibers, and in the cytoplasm of myofibroblasts around the xanthogranulomatous reaction, respectively. Finally, myofibroblasts were observed in all subtypes. The increased number of myofibroblasts was most prominent in the connective tissue layer of chronic cholecystitis with marked perimuscular fibrosis or in the area adjacent to xanthogranulomatous foci of chronic cholecystitis. Under these conditions, CD34-positive stromal cells tended to disappear from the connective tissue layer, which exhibited an increase in myofibroblasts.  相似文献

目的建立混合菌液致SD大鼠盆腔结缔组织炎模型,研究大鼠盆腔黏连组织的病理学改变与层黏连蛋白表达。方法用注射器将细菌混悬液0.5mL,注人大鼠盆腔,并结合子宫穿孔手术,建立SD大鼠盆腔结缔组织炎模型。采用Philips分级评分法对大鼠盆腔黏连程度分级评分,切取黏连组织,用多聚甲醛固定,进行病理检测,并采用免疫组化法测定层黏连蛋白表达情况。结果模型组大鼠盆腔黏连Philips分级评分以Ⅲ级为主,与正常对照组比较差异极显著。HE染色镜检模型组可见大量炎细胞浸润,脓肿形成,伴有多少不等的纤维组织增生。免疫组化染色镜检模型组可见层黏连蛋白高表达,纤维结缔组织增生程度越高,层黏连蛋白表达越多。结论盆腔注射混合菌液并结合子宫穿孔手术,可作为建立大鼠盆腔结缔组织炎模型的方法。层黏连蛋白在盆腔结缔组织炎发病中起重要作用,参与整个炎症过程并维持盆腔黏连组织纤维化,可作为判断盆腔结缔组织炎的炎症和纤维化程度的参考指标。  相似文献

Summary Active fibrous septa are a common feature in liver fibrosis and cirrhosis. Their etiology and formation were studied using cultures of tissue fragments or cells included in collagen gels. Liver fragments obtained from patients with cirrhosis or severe schistosomal fibrosis were able to reorganize the gel and to form discrete, interconnecting fibrous septa composed of parallel arrays of collagen, subsequently colonized by migrating connective tissue cells. The same was obtained in cultures of fibrogranulomatous lesions isolated from schistosome-infected mice livers. However, fragments of normal human and murine liver tissue did not show the capacity to form fibrous septa. Septa formation was also obtained in cultures of cell spheroids formed by liver connective tissue cells isolated from human fibrotic or cirrhotic liver tissues, but not with spheroids of normal skin fibroblasts or smooth muscle cells. This experimental model may represent the fibrous septa formation in vivo, depending on the activity of liver connective tissue cells. The ability of tissue fragments or cell spheroids to form septa in collagen gels might reflect the degree of fibrosis present in the liver tissue in vivo. ABSTRACT

Mast cells are large cells with granular cytoplasm that participate in wound healing, angiogenesis and defense against pathogens. They also contribute to inflammation by initiating innate and acquired immunity. The granules of these cells exhibit characteristic staining properties. We investigated toluidine blue, astra blue, Alcian blue-pyronin Y and May-Grunwald Giemsa stains for mast cells in various oral lesions and assessed the efficacy of each for identifying mast cells. Sections were obtained from 10 each of diagnosed cases of inflammatory fibrous hyperplasia, periapical cyst, mild dysplasia, oral submucous fibrosis and oral squamous cell carcinoma and stained using the stains listed above. Mast cells were assessed for their presence, contrast of the mast cell in the connective tissue background and number. We found that May-Grunwald Giemsa stain was the best for identification of mast cells, although toluidine blue staining is less time-consuming. Overall we obtained better results using May-Grunwald Giemsa and toluidine blue for staining mast cells.  相似文献

Systemic sclerosis (SSc) is a complex connective tissue disease characterized by fibrosis of the skin and various internal organs. In SSc, telocytes, a peculiar type of stromal (interstitial) cells, display severe ultrastructural damages and are progressively lost from the clinically affected skin. The aim of the present work was to investigate the presence and distribution of telocytes in the internal organs of SSc patients. Archival paraffin‐embedded samples of gastric wall, myocardium and lung from SSc patients and controls were collected. Tissue sections were stained with Masson's trichrome to detect fibrosis. Telocytes were studied on tissue sections subjected to CD34 immunostaining. CD34/CD31 double immunofluorescence was performed to unequivocally differentiate telocytes (CD34‐positive/CD31‐negative) from vascular endothelial cells (CD34‐positive/CD31‐positive). Few telocytes entrapped in the fibrotic extracellular matrix were found in the muscularis mucosae and submucosa of SSc gastric wall. In the muscle layers and myenteric plexus, the network of telocytes was discontinuous or even completely absent around smooth muscle cells and ganglia. Telocytes were almost completely absent in fibrotic areas of SSc myocardium. In SSc fibrotic lung, few or no telocytes were observed in the thickened alveolar septa, around blood vessels and in the interstitial space surrounding terminal and respiratory bronchioles. In SSc, the loss of telocytes is not restricted to the skin, but it is a widespread process affecting multiple organs targeted by the fibrotic process. As telocytes are believed to be key players in the regulation of tissue/organ homoeostasis, our data suggest that telocyte loss might have important pathophysiological implications in SSc.  相似文献

In a study involving lactating does with normal-sized and enlarged pendulous udders, 85.5% of milk samples obtained from does with unilateral udder enlargement and 84.8% of samples from bilaterally enlarged udders indicated the presence of intramammary inflammation based on results of the modified Whiteside test. In contrast, only 47.6% of milk samples obtained from normal-sized udders showed evidence of intramammary inflammation. Bacterial cultures were performed on all milk samples which showed evidence of intramammary inflammation. 16 species of bacteria and other microorganisms were cultured from these samples. Bilaterally enlarged udders collectively yielded 13 different microbial isolates, while unilaterally enlarged udders yielded 11, and normal-sized udders yielded 9. The most frequent isolates in all groups were Staphylococcus spp. and Corynebacterium spp. On histopathologic examination, severe pathological changes in the teat and udder tissues of enlarged pendulous udders were observed. Lesions in parenchymatous tissues showed mild to severe inflammation with involution of the parenchyma of glands. This led to shrinkage of the alveoli and in severe cases, their complete disappearance and replacement by fibrous connective tissue which was observed to have a high degree of proliferation. Sections of non-pendulous udder tissue showed proper arrangement of alveoli with hypertrophic epithelial cells, indicating active secretory activity. The results provide some definitive evidence that enlarged pendulous udder development in some Nigerian breeds of goats is not spontaneous but occurs largely as a result of incursion of the mammary glands by pathogenic organisms which may cause mastitis resulting in progressive dysfunction of mammary tissue. In chronic cases, there is severe atrophy of glandular tissue leading to fibrosis.  相似文献

Ferrocyanide was perfused through the vascular system of the isolated legs of 10 dogs. Ferric ion was then injected intramuscularly (not intravascularly) into the leg. The injected materials spread along the trabeculae of the tissues, rupturing the sheets of fibrous connective tissue (frenulae) inpeded the movement of materials in the trabeculae. The barriers consist of sheets of connective tissue which insert on adjacent fibrous capsules. The capsules enclose each cluster of muscle, glandular or other cells. These barriers appear to function in conjunction with the lymphatic drainage system.  相似文献

A method for the combined staining of elastic, muscle and connective tissue for routine use in histopathology is described. The elastica, stained black by Verhoeffs technique, is contrasted with the muscle and connective tissue stained red and green or blue respectively by a modification of Masson's trichrome. Cell nuclei stain blue-black with Weigert's iron hematoxylin. The procedure takes approximately two hours and is most suitable for the study of vascular pathology in surgical and autopsy sections.  相似文献

Ulcerative colitis (UC) is characterized by chronic relapsing intestinal inflammation finally leading to extensive tissue fibrosis and resulting in a stiff colon unable to carry out peristalsis or to resorb fluids. Telocytes, a peculiar type of stromal cells, have been recently identified in the human gastrointestinal tract. Several roles have been proposed for telocytes, including mechanical support, intercellular signalling and modulation of intestinal motility. The aim of the present work was to investigate the presence and distribution of telocytes in colonic specimens from UC patients compared with controls. Archival paraffin‐embedded samples of the left colon from UC patients who underwent elective bowel resection and controls were collected. Tissue sections were stained with Masson's trichrome to detect fibrosis. Telocytes were identified by CD34 immunohistochemistry. In early fibrotic UC cases, fibrosis affected the muscularis mucosae and submucosa, while the muscularis propria was spared. In advanced fibrotic UC cases, fibrosis extended to affect the muscle layers and the myenteric plexus. Few telocytes were found in the muscularis mucosae and submucosa of both early and advanced fibrotic UC colonic wall. In the muscle layers and myenteric plexus of early fibrotic UC, telocytes were preserved in their distribution. In the muscularis propria of advanced fibrotic UC, the network of telocytes was reduced or even completely absent around smooth muscle bundles and myenteric plexus ganglia, paralleling the loss of the network of interstitial cells of Cajal. In UC, a loss of telocytes accompanies the fibrotic remodelling of the colonic wall and might contribute to colonic dysmotility.  相似文献

Locust neural lamella and Calpodes connective tissue fixed in glutaraldehyde have a fibrous component which stains after reaction with DAB and osmication and after staining sections with PTA.
