1. Extracts of the plasmodia of the myxomycete, Physarum polycephalum, exhibit reversible decreases in viscosity in response to the addition of ATP under appropriate conditions. The protoplasm material prepared by extraction with KCl solution can apparently exist in either a high or a low viscosity state. As prepared, it is in the low viscosity condition. Rapid and extensive increases in viscosity of the extract are brought about by addition of AMP, inorganic phosphate, or, under certain conditions, of ATP. Only after the high viscosity state has been attained does addition of appropriate quantities of ATP cause a reversible decrease in viscosity. 2. The active principle of crude plasmodial extracts may be concentrated by fractional precipitation with ammonium sulfate and is found in the fraction precipitated between 30 and 40 per cent saturation. This material possesses a higher viscosity than does the original crude extract and is apparently in the high viscosity state since the addition of ATP causes an immediate reversible decrease in viscosity. 3. The ATP-sensitive fraction of myxomycete plasmodia possesses a viscosity which is dependent upon its previous thermal treatment. Extracts incubated at 0° for a period of a few hours increase greatly in viscosity when they are returned to 24.5°. This increased viscosity is structural in nature, is destroyed by mechanical agitation of the solution, and may be reversibly destroyed by addition of ATP. 4. It is suggested that the ATP-responsive protein of myxomycete plasmodia may be related to sol-gel transformations which have been observed in intact plasmodia and may participate in the protoplasmic streaming of the intact organism. This suggestion is based upon the following facts: (a) the protoplasmic streaming of myxomycete plasmodia is increased by microinjection of ATP; (b) the gel portion of the cytoplasm at the site of the microinjection of ATP is extensively converted to the sol state. The changes in structure of the intact cytoplasm are thus similar in nature to the changes exhibited in response to ATP by the purified ATP-sensitive protein. 5. The ATP-sensitive protein of myxomycete plasmodia appears to undergo reversible aggregation to form a high viscosity state. The function of ATP is to break down the aggregates thus formed. Since a specific ATPase activity is associated with the purified material, added ATP is gradually destroyed and recovery of viscosity attends the spontaneous reconstitution of aggregates.  相似文献

Ultracentrifugal analysis of an extract of pea epicotyls, previously freed of debris and larger particles by centrifugation at 40,000 g for 10 minutes, has revealed the presence of a major component which possesses a sedimentation coefficient of 74 S. This component constitutes about 25 per cent of the TCA-precipitable material in the clarified epicotyl extract and is estimated to make up 1 to 2 per cent of the dry weight of the original tissue. In size, chemical composition, and morphology, the 74 S component resembles the nucleoproteins of the microsomes from animal tissues. The 74 S component of pea epicotyl extracts has been purified by repeated cycles of differential centrifugation to yield a preparation which is 80 per cent homogeneous in the analytical ultracentrifuge. It has been found to contain 30 to 37 per cent RNA as judged by a variety of analytical techniques. Approximately 55 per cent of the weight of the material is protein and a further 4.5 per cent phospholipide. Electron micrographs of air-dried specimens of the purified preparation show the 74 S constituent to be flattened spheres with an average height of 180 A and an average diameter of approximately 280 A. The molecular weight of the 74 S particles is computed from sedimentation, viscosity, and partial specific volume data to be 4.5 million ± 10 per cent in agreement with the value estimated from electron micrographs. The 74 S or microsomal component of pea epicotyls is rapidly aggregated in the presence of low concentrations of Mg ions or by somewhat higher concentrations of Ca or K salts. ATP on the contrary causes resolution of electrolyte-induced microsomal aggregates with simultaneous degradation of the particles to an ultracentrifugally inhomogeneous mixture of lower molecular weight materials.  相似文献

The phenomenon of contraction of a striated muscle down to below 50 per cent rest length has been examined for the scutal depressor of the barnacle Balanus nubilus by a combination of phase contrast and electron microscopy. It was found that neurally evoked contraction down to 60 per cent rest length results from the shortening of the I band. At the same time the Z disc changes in structure by an active process which results in spaces opening up within it. Thick filaments can now pass through these spaces from adjacent sarcomeres, interdigitating across the discs. Interdigitation permits repetitive contraction in the living muscle to below 30 per cent rest length. In non-neurally evoked contractions most thick filaments do not find spaces in the Z disc and bend back, giving rise to contraction band artifacts. Expansion of the Z disc can be produced in glycerinated material by the addition of solutions containing a high concentration of ATP.  相似文献

1. The cross-striation in the indirect flight myofibrils of Calliphora has been studied by phase contrast and polarised light microscopy. The band pattern at rest-length has been determined in flies killed in osmium tetroxide vapour while their wings remained in the resting position. All other observations have been made on unfixed fibrils. Although length changes in situ are probably very slight (about 2 per cent), isolated fibrils, by treatment with crude muscle extract or with ATP, can be induced to elongate to 104 per cent rest-length, or to shorten by 8 per cent but no more. Over the range 98 to 104 per cent rest-length, experimentally induced length changes are reversible. The fibrils can also be stretched beyond 104 per cent rest-length, but the process is irreversible. During the course of glycerol extraction the fibrils elongate to 104 per cent rest-length. 2. The changes in band pattern observed over the range 104 to 92 per cent rest-length are qualitatively the same as the changes observed over a wider range (about 130 to 40 per cent rest-length) in the skeletal myofibrils of rabbits. The earlier stages of shortening appear to be effected by retraction of the I bands into the A bands where they fill up the H zones. No evidence has been found that any changes in band pattern are due to a migration of the A substance. 3. Two components of the sarcomere can be extracted from it and a third component remains behind. These three components, which have also been demonstrated in skeletal myofibrils of the rabbit, where they behave in the same way, are: (a) the A substance which does not change its position as the fibril changes its length, and which can be extracted by the same procedures as remove myosin (shown elsewhere to be the A substance) from rabbit fibrils; (b) a material which extends from the Z lines to the borders of the H zone and which moves inwards during contraction and outwards during elongation; it can capture rabbit myosin from solution and form with it a contractile system, and it is thought to be actin; (c) a "backbone" or stroma bearing Z and M lines. 4. Since all these features of the cross-striation are the same in the insect fibrils as in rabbit fibrils, it is considered very probable that the sarcomere is similarly organised in both types of muscle and contracts by essentially the same mechanism.  相似文献

1. Mitochondria isolated from rat liver were disrupted with 0.3 per cent deoxycholate and a number of subfractions were isolated from this preparation by differential centrifugation. 2. The protein N, RNA and phospholipide content, as well as the succinoxidase, cytochrome c oxidase, adenylate kinase, and DPNH-cytochrome c reductase of these fractions were determined. 3. Two of these subfractions, found to consist of mitochondrial membranes (2), contained ~ 12 per cent of the protein N and ~ 35 per cent of the phospholipide of the whole mitochondria and accounted for ~ 70 per cent of the succinoxidase and cytochrome c oxidase activity of the original mitochondrial preparation. There was no discernible adenylate kinase, DPNH-cytochrome c reductase, or phosphorylating activities in these fractions, nor could they oxidize other substrates of the Krebs's cycle. 4. The most active fraction (60 minutes at 105,000 g pellet) had a higher phospholipide/protein value than the whole mitochondria and showed a seven-to elevenfold concentration of succinoxidase and cytochrome c oxidase activities. 5. Evidence has been given to indicate that the various components of the succinoxidase complex are present in this membrane fraction in the same relative proportions as in the whole mitochondria. 6. Abstract— The effect of stimulating the electric organ of Torpedo marmorata , anaesthetized with 0.01% Tricaine methane sulphonate, by means of electrical stimulation (5/s) administered via an electrode placed on the electric lobe has been studied electrophysiologically, biochemically and morphologically. The response of the organ declined to about 50 per cent of its initial value after about 500 stimuli, by a further 10 per cent after another 500 stimuli and then to about 12 per cent of the initial value after a further 1000 stimuli. Thereafter the response fell off progressively. However, even when the response was less than 1 per cent of its initial value, the organ had considerable powers of recuperation during a 30-s rest period, to 30–50 per cent of its initial value.
The fall in response was accompanied by a reduction in vesicle size and number, an increase in the area of the presynaptic membrane and a fall in the protein, total nucleotide, ATP and acetylcholine content of the vesicle fraction isolated from the stimulated tissue. However, whereas vesicle numbers and the protein and total nucleotide content of the vesicle fraction fell by only about 50 per cent, vesicular ATP and acetylcholine levels were reduced to about 10 per cent. An analysis of the covariance of vesicular ATP and acetylcholine showed an initial loss of an acetylcholine-rich (relative to ATP) population of vesicles. The early loss of vesicular protein and nucleotide and vesicle numbers as well as the morphological changes seen would be consistent with a loss of vesicles due to fusion with the external membrane. The preferential loss of acetylcholine and ATP from the vesicle fraction indicates that the vesicles surviving the stimulation procedure have been utilized in a number of cycles causing the progressive fall in vesicle volume, and acetylcholine and ATP content.  相似文献

1. The rigor which takes place when completely frozen frog sartorius muscle is thawed ("thaw rigor"), is accompanied by a decrease in length of 70 per cent and a loss in weight of 35 per cent, whether the muscle is frozen in the resting or the exhausted condition, or during isometric tetanus. Muscle tetanized to maximal shortening shows a loss in weight of 25 per cent on thawing. 2. A load of 8 gm. is sufficient to prevent the decrease in length on thawing, but after its removal the muscle will shorten almost to the normal extent. 3. Inhibitors such as azide, cyanide, 2:4 dinitrophenol, p-chloromercuribenzoate, Cu, and hydrogen peroxide, when used for periods not exceeding 1 hour, have little effect on the shortening; although in some cases these poisons render the muscle inexcitable. 4. Muscles poisoned with iodoacetic acid and stimulated to exhaustion, or maintained at fixed length in nitrogen, show little or no shortening on thawing. ATP can produce shortening in the muscles in which it has been prevented. 5. The phenomenon is considered to be due to an in situ synaeresis of the actomyosin of the myofibrils. As a result of the disorganisation of the muscle protoplasm produced by the freezing and subsequent thawing, the ATP, which must be bound or localized in the resting muscle, can act on the myofibril in a similar manner to its in vitro effect on the actomyosin thread.  相似文献

On royal jelly, 1- to 2-day-old honeybee worker larvae have been reared in vitro to adults in a yield of 67±18 per cent. Up to 100 per cent and, on an average, 60 per cent of them were queens and intercaster. The preparation of a basic food from royal jelly by extensive alcohol extraction is described. With this control food, a survival rate of 47±18 per cent was achieved; 15 per cent of the adults were determined, 4·3 per cent of them were queens. Rearing of 1- to 2-day-old worker larvae on a basic food, to which unknown fractions may be added, was used as a biological test for the partial purification of queen bee determinator from royal jelly. By chromatography of the ethanol extract, previously treated with charcoal, on the cation exchanger Dowex 50 WX4 and rechromatography on silica gel, a 105-fold purification of determinator was achieved. Chemical properties of the highly hydrophilic, low molecular active fraction are described.  相似文献

Investigations dealing with the determination of the major chemical constituents of the axoplasm of the giant nerve fiber of the squid are described. Particular emphasis has been placed on determining the components involved in acid-base balance. It was found that 72 per cent of the total solids of axoplasm, representing 13.5 per cent of the wet material, are of relatively low molecular weight (dialyzable) and consist mainly of charged ionic or dipolar constituents. Of the 520 micromoles per gm. of total base, 72 per cent are balanced by organic acids: aspartic acid (65 micro equivalents per gm.), glutamic acid (10 micro equivalents), fumaric and succinic acids (15 micro equivalents), a new polycarboxylic acid (35 micro equivalents), and isethionic acid, a biologically novel sulfonic acid (220 micro equivalents). Besides potassium, sodium, small amounts of calcium, and magnesium there is a considerable fraction of organic (nitrogenous) base. Other features of the chemical composition of squid axoplasm include a relatively high concentration of taurine (100 micro equivalents) and an ultraviolet absorbing substance possibly identical with N-methylpicolinic acid. The distribution of the phosphates, especially the concentration of ATP, has been investigated. Specific techniques elaborated in connection with this study have been described and the biochemical implications of the analytical results are discussed.  相似文献

1. An investigation of the physicochemical properties of myosin has been carried out. Prepared under standard conditions, the ratio of flow-birefringence to protein concentration is uniform. The effect of electrolytes, pH, and urea on the flow-birefringence and viscosity (relative and anomalous) of myosin has been examined. 2. Decrease or abolition of flow-birefringence does not necessarily imply far reaching denaturation, since such effects can be reversed by a variety of means. 3. When a myosin solution is treated with adenosinetriphosphate, its flow-birefringence is decreased (average 48 per cent), its anomalous viscosity is retained, and its relative viscosity is decreased (average 14 per cent). The full effect of adenosinetriphosphate is obtained at 0.004 M; a molarity very much less than that of other substances which decrease the flow-birefringence of myosin. 4. The changes in the physicochemical properties of myosin brought about by adenosinetriphosphate are spontaneously reversible, and are connected with the enzymatic action of the protein as adenosinetriphosphatase. 5. Effects similar to those of adenosinetriphosphate on the physicochemical properties of purified myosin have been obtained so far only with inosinetriphosphate. 6. Inorganic phosphate is split off by myosin from inosinetriphosphate as well as from adenosinetriphosphate. Inorganic triphosphate is split by 1 to 2 per cent solution of three times precipitated myosin. 7. Adenosinediphosphate and inorganic triphosphate act as competitive inhibitors with adenosinetriphosphate, blocking the fall of flow-birefringence. 8. The implications of the results, and the conception of active enzymic groups attached to proteins participating in cell structure, whether contractile or non-contractile, are discussed in relation to present views on muscle physiology and other biological problems.  相似文献

1. A method for the preparation of 8 to 10 liter quantities of T2 virus lysates, titering 2 to 5 x 1011 infectious units per ml. has been described. 2. Procedures have been developed for the concentration and purification of virus to a high specific infectivity. No fractionation procedure of the several used succeeded in further raising the specific infectivity of these purified preparations. 3. Some of the general properties of the better preparations have been determined. They exhibited titers of 2 x 1015 infective units per gm. of material or 1.2 x 1016 per gm. of nitrogen. 4. A study of the distribution of nitrogen among the various fractions of the virus showed that about 6 per cent of the total nitrogen is soluble in 4 per cent trichloracetic acid; that the protein nitrogen is about 40 per cent of the total and the nucleic acid nitrogen is 53 per cent. At least 96 per cent of the total phosphorus is in the nucleic acid fraction. Less than 0.5 per cent quantities of lipid and PNA were found.  相似文献

A system has been developed for the isolation of photosynthetically active chloroplasts from leaves of Populus deltoides. A high proportion of the chloroplasts appeared intact. The maximum rates of different photosynthetic processes were as follows: CO2 fixation 3.5 micromoles per milligram chlorophyll per hour, noncyclic ATP synthesis 10 micromoles per milligram chlorophyll per hour, and cyclic ATP synthesis 300 micromoles per milligram chlorophyll per hour.  相似文献

1. Viscosity and pH curves of casein dissolved in NaOH, KOH, LiOH, and NH4OH are shown and it is found that a maximum viscosity occurs at about the same pH point with each alkali; i.e., 9.1 to 9.25. The magnitude of the viscosity is largest in ammonia solutions. 2. The maximum viscosity occurs in 8 to 10 per cent solutions of casein in alkalies when about 98 x 10–5 gram equivalents of base are combined with 1 gram of casein. 3. A maximum viscosity occurs in the same region (pH 9.1 to 9.25) when casein is dissolved in Na2CO3, Na3AsO4, Na2SO3, NaF, and Na2PO3. 4. The maximum viscosity obtained with borax solutions of casein occurs at 8.15 to 8.2 pH. It is suggested that casein acts like mannitol, glycerol, etc., in increasing the dissociation of boric acid. 5. The flattening of the viscosity curves of casein solutions, following the decline from maximum, is shown to be due to alkaline hydrolysis whence casein no longer exists as such but is cleaved into a major protein containing no phosphorus or sulfur and less nitrogen. This cleavage commences at pH 10.0 to 10.5. 6. When casein is prepared from solutions that have been subjected to high temperatures (60°C. and above) or has otherwise been heated during its preparation, it yields solutions in alkalies of high viscosity.  相似文献

Possible preparation of 10 per cent sodium sulfapyridazine ophthalmic drops containing aubazidan, a microbial polysaccharide, providing prolongation of the drops action and stability on the storage was studied. The pharmacokinetic studies showed that aubazidan which is a natural polymer provided high prolongation of the sulfapyridazine effect in the tissues of the anterior part of the eye in rabbits. The prolonged effect was similar to the previously observed effect of the solutions containing synthetic polymers such as 1 per cent polyacrylamide and polyvinyl . Satisfactory tolerance by the rabbit eye tissues of 6-fold daily instillations of the 10 per cent sodium sulfapyridazine solution with aubazidan for the observation period of 1 month was stated. When stored in vials the 10 per cent sodium sulfapyridazine ophthalmic drops with 0.5 per cent of aubazidan preserved the stability for 24 months with respect to the colour, transparency, viscosity, pH and drug content. It was demonstrated with using the agar diffusion method and Staphylococcus aureus, Proteus vulgaris, Escherichia coli and Pseudomonas aeruginosa as the test cultures that sodium sulfapyridazine completely preserved its antimicrobial activity in the presence of aubazidan. The data suggested that clinical trials of the 10 per cent sodium sulfapyridazine solution with 0.2-0.5 per cent of aubazidan were promising in prevention and therapy of bacterial conjunctivitis.  相似文献

Some solubility, oxidation, reduction, and compound-forming characteristics of extracts of Cypridina luciferin have been presented. A method of purification has been described which increased the amount of luciferin per unit of dry weight, as measured by the total light emitted, to about two thousand times that in the dry starting material. 23Sodium binding to a partially purified beef brain and purified dogfish rectal gland (sodium + potassium)-activated adenosinetriphosphatase (NaK ATPase) has been studied by pulsed nmr. In both preparations addition of ATP (in the absence of Mg) increased the amount of Na bound to the enzyme protein. In the less-pure brain preparation there was some binding of Na to the protein in the absence of ATP but in the purer preparation from the rectal gland there was little or no binding without ATP. With the dogfish enzyme, potassium readily displaced bound sodium. The KD for sodium determined by nmr agreed closely with that determined kinetically. This, coupled with the fact that the dogfish enzyme required ATP for sodium binding suggests that the sodium detected by nmr in this preparation is due to binding at its specific site(s).  相似文献

From microdensitometer measurements on electron micrographs of sectioned sperm heads it has been found that the electron stains, triiodobenzoyl chloride, and triiodophenylisocyanate, increase the image contrast of the cell membrane above its immediate background by about 40 per cent and 70 per cent respectively, while the nucleus remains unstained. Assumptions based on current electron scattering theory have been used to deduce the uptake by weight of the stains in terms of the density of the nucleus, which was estimated from complementary measurements made with the interference microscope and electron microscope. The uptake of the stains was found to be about 7 per cent and 12 per cent by weight respectively. It is suggested that the method used in this work could be applied generally for the density measurement of cell structures unresolved by the light microscope.  相似文献

1. Whole bovine albumin, homogeneous in diffusion and sedimentation, and essentially homogeneous in electrophoresis, has been prepared by a method involving ammonium sulfate precipitation of the globulins in the cold and of the albumin at room temperature, isoelectric precipitation of the euglobulins, and reprecipitation of the albumin. 2. The product has been characterized by chemical analysis and by viscosity, diffusion, sedimentation, and electrophoresis measurements. The carbohydrate content is 0.38 per cent, the nitrogen content, 15.2 per cent. The molecular shape approximates that of a prolate ellipsoid with an axial ratio of 3.1, assuming 33 per cent hydration; the average molecular weight is 65,000. 3. Bovine albumin is readily denatured by concentrated solutions of urea or guanidine hydrochloride, gross changes in molecular shape resulting. 4. Regeneration of bovine albumin denatured in solutions of 8 M urea or guanidine hydrochloride yields a material closely resembling the native in carbohydrate content, in molecular size and shape, and in electrophoretic properties. However, the regenerated protein differs from the native in susceptibility to tryptic digestion, and, in this respect, appears to be in a denatured state. 5. In 8 M solutions of guanidine hydrochloride a limiting yield of regenerated albumin equivalent to 95 per cent of the original protein is approached. 6. Bovine crystalbumin, a crystalline carbohydrate-free fraction of the whole albumin, appears to be more susceptible to denaturation than whole bovine albumin.  相似文献

Ribonucleoprotein (RNP)1 particles isolated by DOC treatment from pancreatic microsomes have a RNA content of 35 to 45 per cent of their dry weight. In the analytical ultracentrifuge about 85 per cent of the material has a sedimentation coefficient of ∼85 S. These particles contain amylase, RNase, and trypsin-activatable proteolytic activities which cannot be washed off or detached by incubation in 0.44 M sucrose. The enzymes are released, however, by incubation in the presence of low concentrations of ATP, PP, or EDTA, and high concentrations of IP and AMP. At the same time, and at the same concentrations, ∼80 per cent of the RNA and ∼25 per cent of the protein of the particles becomes also non-sedimentable. The simultaneous addition of Mg++ to the incubation medium prevents these losses. This finding, together with the observation that all the Mg++ of the particles is released by the same agents, makes it likely that Mg++ holds the particles together, and that its removal by the chelators used causes the particles to disintegrate. These findings are discussed in relation to the molecular structure of the RNP particles.  相似文献

1. Pepsin solutions were iodinated at pH 5.0–6.0 until 10–20 per cent of the activity was lost and 1/20 (0.7 per cent) of the saturating amount of iodine had been introduced into the protein molecule. After alkaline hydrolysis 65 per cent of the original iodine was accounted for as mono-iodotyrosine although only 42 per cent was isolated as a crystalline product. No evidence was obtained to support the possibility that any group other than tyrosine in pepsin was iodinated. 2. Some of the properties of the crystalline l-mono-iodotyrosine were determined and compared to those of di-iodotyrosine. 3. One iodinated pepsin preparation was crystallized. The crystal form was the same as that of the original pepsin. A solubility curve of the crystals demonstrated that it was very different from pepsin and had nearly constant solubility.  相似文献
