The analysis of integumental pore pattern has identified two intraspecific groups of postlarval shrimp Litopenaeus vannamei (Boone, 1931), a species of which three to five natural populations or groups are expected to be found from Mexico to Peru. Natural distribution and man-made redistributions have not been typified on a population basis in any penaeid, but it is important to do so in this commercially significant group. Important factors to be defined on a geographic population or group basis include differences in domesticity (eg resistance to pathological agents); food conversion indexes and food preferences; growth indices; tolerance to stress of various origins and other attributes useful in aquaculture. This consideration is valid not only for L. vannamei but also for any other shrimp species that has been the object of artificial redistribution. 相似文献

We analized acute toxicity in white shrimp (Litopenaeus vannamei) postlarvae exposed to two chlorinated pesticides, DDT and endosulfan, under laboratory conditions during 168 hours, with controlled temperature (29 +/- 1 degrees C), salinity (3 +/- 1%o) and pH (8 +/- 1). Median lethal concentrations (LC50), "incipient" LC50, median lethal time (LT50) the "maximum acceptable concentration of the toxic compound" (MACT) and "the safety level" (SL) were determined. The concentration of the compounds at which organism growth was reduced by 5 and 50% (EC5 and EC50), as well as changes in oxygen consumption patterns were determined in the surviving postlarvae. They were very sensitive to both compounds and DDT was thrice as toxic as endosulfan. Growth rate decreased 50% and 80% with endosulfan and DDT, respectively, at the experimental pestice concentration. The low resistance of postlarvae to DDT and endosulfan suggests that additional inflow of these pesticides into the aquatic system could affect the rate of shrimp production in the area. 相似文献

We rated some reproductive characteristics of white shrimp Litopenaeus vannamei (Boone, 1931) males using 46 farmed individuals (weighing 21.42 +/- 0.56 g) and 40 wild individuals (weighing 36.10 +/- 0.72 g). In farmed shrimps, spermatophore mean weight was 8.94 +/- 0.51 mg; total mean sperm count was 3.90 +/- 0.27 x 10(6) in each spermatophore; and mean percentage of normal sperm was 86.9 +/- 0.37%. In wild individuals, the respective values were 30.68 +/- 2.32 mg; 6.22 +/- 1.09 x 10(6); and 62.1 +/- 3.56%. In both groups, the differences between right and left spermatophore were not significant (p < 0.01). There were significant differences in spermatophore weight and percentage of normal sperm between farmed and wild shrimps; sperm counts differences, however, were not significant (p < 0.01). The relationship between spermatophore weight (Ws) and individual weight (Wo) was Ws (mg)=1.23 (Wo)-17.34 (r2=0.89), in farmed shrimps; and Ws (mg) = 2.57 (Wo)-60.04 (r2 = 0.64), in wild ones. In cultivated organisms, the relationship between sperm counts (Cs) and individual weight (Wo) was Cs (x 10(6)) = 1.13 * 10(-4*) (Wo) 3.361 (r2 = 0.85); and versus spermatophores weight was Cs (x 10(6)) = 0.439* (Ws) 0.984 (r2 = 0.90). In wild organisms, there was no correlation. The proportion of normal sperm ranged from 79.8 to 95.2 % (86.9 +/- 0.37%) and from 14.0 to 91.5% (62.1 +/- 2.52%), in farmed and wild shrimps, respectively. The most frequent abnormalities in both farm and wild animals were sperm without spike (49.3% and 76.6%, respectively) and irregular shape (35.8 % and 17.7 %). Ryanodine receptor/calcium release channel is a large protein that plays an essential role in muscle contraction; mutations in the ryanodine receptor gene affect sensitivity to stress. As a first step towards investigating the relationship between the ryanodine receptor and shrimp cramped muscle syndrome, we cloned, partially sequenced, and examined single-nucleotide polymorphisms (SNPs) of the ryanodine receptor gene of the Pacific white shrimp (Litopenaeus vannamei). The nucleotide sequence of a 15.06-kb L. vannamei genomic DNA segment containing a partial ryanodine receptor gene sequence was determined (deposited in GenBank nucleotide database: HM367069). Direct sequencing of PCR-amplified ryanodine receptor exons with their intron-flanking regions in 10 cramped muscle syndrome shrimp and 10 healthy shrimp, revealed seven SNPs. Five of them (1713A/G, 1749T/C, 1755T/C, 3965G/A, and 8737C/T) are located in exons; however, they appear to be neutral (synonymous), since they do not alter the encoded amino acid. The other SNPs (1553C/T and 13337A/G) are in introns. The SNPs identified in the ryanodine receptor gene could be useful for association studies aimed at determining the physiological role of the ryanodine receptor in cramped muscle syndrome of shrimp. 相似文献

The penaeid prawn, Litopenaeus vannamei, was employed to investigate intracellular isosmotic regulation in situations where invertebrates encounter hyposmosis. Hemolymph osmolality was first analyzed to confirm osmoregulatory conditions in the experimental animals, followed by analysis of amino acids in muscle and hemolymph using high-performance liquid chromatography. Total muscle amino acid levels decreased when hemolymph osmolality was extremely low, whereas glycine and l-serine levels increased in the hemolymph. These results suggest that tissue amino acids were released into the hemolymph to lower the osmolality of the tissues for purposes of low-salinity adaptation. Next, oxygen consumption and ammonia excretion rates were examined, and the O/N ratio was determined. Oxygen consumption levels and ammonia excretion rates increased, and the O/N ratio decreased when the animals were exposed to low salinity. These results suggest that amino acids were abundantly consumed as an energy source when animals were exposed to low salinity. To confirm the consumption of particular amino acids, the specific activity of l-serine ammonia lyase was also examined. Specific activity was highest when l-serine levels in the hemolymph were highest. Thus, it appears that l-serine levels increased under hyposmotic conditions due to the consumption of l-serine as an energy source. It was concluded that particular amino acids as osmolytes are likely metabolized as energy sources and consumed for purposes of hyposmotic adaptation. 相似文献

As in other countries, the presence of disease has affected and limited the development of shrimp culture in Venezuela. Vibriosis is one of the most prevalent, causing high mortality not only in larval cultures but also in shrimp production. The present work shows the histological changes in hepatopancreas of Litopenaeus vannamei experimentally induced by a strain of Vibrio alginolyticus. The shrimps were infected by the immersion technique, being exposed to a concentration of 5.2 x 10(7) bacteria/ml. As soon as their behavior indicated a moribund condition, the animals were sacrificed, and their hepatopancreata were fixed and processed for histological observation. Light microscopical observations revealed loss of the acinar structure of the digestive gland and sloughing off of cellular lining because of cytolisis. The acinar cellular content was dispersed within the lumina and among the acini. Clusters of hemocytes, in the majority loaded with bacteria, were observed under the connective tissue capsule surrounding the hepatopancreatic lobes and in the hemal spaces of the connective tissue sheath surrounding the tubules. 相似文献

Shrimp head waste is a major byproduct of crustacean processing in North-eastern Brazil and represents an interesting source of bioactive molecules. Additionally, its use increases the sustainability of processing fishery products. The present study reports a process developed for recovering bioactive molecules from shrimp heads through autolysis. A protein hydrolysate (120 ± 0.4 g) formed by a 9% (w/v) solution was recovered and lyophilized from 1 kg of shrimp heads. Approximately 195 ± 0.5 mg of carotenoids was recovered as an ethanolic extract. The recovery of chitin and chitosan were 25 ± 2 g kg?1 and 17 ± 4 g kg?1 wet processing waste, respectively. Chitosans were characterized by 13C NMR, and FT-IR analysis and exhibited a variable degree of deacetylation (60–80%). Sulfated glycosaminoglycans that exhibited electrophoretic migration similar to mammalian standards were also recovered (79 ± 2 mg kg?1 wet processing waste), and their degradation products suggested the presence of C6-sulfated heparan sulfate. These data point to the feasibility of an integrated process for isolating highly bioactive molecules, such as sulfated- and amino-polysaccharides, with a broad spectrum of applications from shrimp processing waste. 相似文献

Penaeoidean shrimp pleonal muscle is a valuable economic resource worldwide, but little is known of its development during larval stages. The development of pleonal muscle in Penaeus (Litopenaeus) vannamei was studied by rhodamine-phalloidin staining and laser-scanning confocal microscopy. Dorsal pleonal muscle was first evident at the protozoea I stage while ventral pleonal muscle was present by the protozoea II stage. Identifiable ventral pleonal muscles were evident by the protozoea III stage and all ventral muscle types were present in the mysis I. The tail flex response began at the mysis stage and growth of existing pleonal muscles continued. The pleopods formed during the mysis stages, with coxal and basis muscles developed by mysis III. The pleopods became functional beginning with the first post-larval stage. We conclude that the pleonal muscle pattern of P. vannamei larvae is similar to that of adult Penaeus setiferus, and that homologous muscles are present. The major formation of dorsal pleonal muscles occurs during the protozoea II stage, while significant development of ventral pleonal muscles occurs during the protozoea III stage. 相似文献

Five polymorphic microsatellite loci were characterized for Penaeus (Litopenaeus) vannamei. Loci were isolated using a partial Sau3A1 genomic library by the sequencing of randomly selected clones and by a biotinylated (CT)10 and (GT)10 probes screening procedure. The last strategy resulted in the most useful data. About 40% of the clones showed a previously reported satellite/microsatellite (PVS1), reducing the chance of finding new microsatellite regions. Whereas two of the microsatellite loci with more than 10 alleles will be useful for mating analysis in a breeding program, the others might prove useful for population genetic studies. 相似文献

Mating behavior has been described for very few species of penaeoid shrimps. We describe some of the environmental conditions under which mating of Litopenaeus vannamei took place in the ocean, as inferred by the presence of attached spermatophores on the thelycum of females, combined with the presence of mature males with empty ampulla terminalis, both evidence of recent copulation. Out of a total of twelve 24 hr samplings on board the research vessel B/O "EL PUMA", one was selected to examine mating. There were four periods of observation. The highest frequency of females with attached spermatophores were found during the daytime whilst the lowest was registered during the night. Females with attached spermatophore were mostly at stages IV and V of ovarian maturity and fell within the 30-50 mm of C.L. range. Copulating male size ranged between 30 and 40 mm of C.L. and there was a close relation between the percentage of mating females and males. Male to female ratio varied throughout the sampling period but it was never 1:1. The female mean size was, in all cases, bigger than the male mean size. Among the environmental factors, salinity and oxygen varied very slightly throughout the sampling period; temperature variation was more pronounced and those changes were attributed to the tidal oscillation. 相似文献

We report the development of 11 polymorphic microsatellite loci in pacific white shrimp (Litopenaeus vannamei) using an unenriched genomic library. The number of the alleles ranged from two to 18 and observed hererozygosity ranged from 0.0286 to 0.9429, indicating that these markers will be useful for population studies and mapping in pacific white shrimp. Seven loci were detected deviated from Hardy–Weinberg, caused by deficiency of heterozygote, suggesting population genetic structure across the sampled population. Ontogeny of the gut in Penaeus setiferus was investigated by reconstruction of serial sections examined by light microscopy. Development of the gut into the adult form is protracted over several weeks beyond metamorphosis in steps that may be directly related to the unique postlarval life history of Penaeus. The gastric mill is lacking in larval stages of P. setiferus. In protozoeal stages Z1-Z3, the pyloric ampullae are blind sacs that do not communicate with the midgut. The gland filter first appears in mysis stage M2. The gastric mill in early postlarval (PL) stages consists of poorly chitinized lobes with flexible setae. By PL21 the ossicles of the gastric mill are rigid and setae are replaced by spine-like denticles, but even by PL35 the gastric mill is neither as massive nor heavily chitinized as in adults. During the mysis stages and early PL stages, the hepatopancreas communicates freely with both the foregut and the midgut trunk. By PL35 the hepatopancreatic ducts are essentially isolated from the remainder of the midgut by foregut ossicles. The midgut in Z1 consists of two pairs of simple caeca and the midgut trunk. During larval growth, each of the lateral midgut caeca develops into a number of lobes. After metamorphosis these lobes begin to ramify into small-diameter tubules, and by PL35 have completely ramified into the hepatopancreas of adults. From M1 to PL4, the anterior midgut caeca decrease in absolute size and become a single anterior diverticulum. The posterior midgut diverticulum first appears in PL21 as a simple sac and thereafter increases in size and complexity. 相似文献

This study was conducted to determine if exposure of shrimp, Litopenaeus vannamei, to a commercial anti-vibrio vaccine caused changes in antibacterial and cellular (phagocytosis) defences. Shrimp post-larvae were administered either Vibromax™ vaccine or a blank preparation. Whole body homogenates were prepared before (day 0), during (day 10) and after (day 20) vaccination and incubated with a selection of pathogenic vibrios. Homogenate from day 0 animals showed natural antibacterial activity towards Vibrioanguillarum which was significantly enhanced for bacteria-exposed shrimp at 10 days post-challenge. This effect of the vaccine was short-term in its duration. No antibacterial activity was observed in day 0 shrimp homogenate against Vibrio alginolyticus but it was significantly enhanced for both vaccinated and blank-vaccinated shrimp by day 10. No natural or inducible antibacterial activity was observed against Vibrio harveyi at 0, 10 or 20 days post-challenge. To determine if prior exposure of shrimp to inactivated vibrios results in elevated hemocyte phagocytic activity, juveniles were injected with either a mixture of formalin-inactivated vibrios or saline. Hemocyte monolayers made from these shrimp were overlaid with a 1:1 mix of Bacillus subtilis and these vibrios. Hemocytes from vibrio-exposed animals showed elevated levels of internalised vibrios compared with those from the saline injected group. These studies show selectively enhanced cellular defences of shrimp following ‘vaccination’. 相似文献

Bioinformatics and re-sequencing approaches were used for the discovery of sequence polymorphisms in Litopenaeus vannamei . A total of 1221 putative single nucleotide polymorphisms (SNPs) were identified in a pool of individuals from various commercial populations. A set of 211 SNPs were selected for further molecular validation and 88% showed variation in 637 samples representing three commercial breeding lines. An association analysis was performed between these markers and several traits of economic importance for shrimp producers including resistance to three major viral diseases. A small number of SNPs showed associations with test weekly gain, grow-out survival and resistance to Taura Syndrome Virus. Very low levels of linkage disequilibrium were revealed between most SNP pairs, with only 11% of SNPs showing an r 2 -value above 0.10 with at least one other SNP. Comparison of allele frequencies showed small changes over three generations of the breeding programme in one of the commercial breeding populations. This unique SNP resource has the potential to catalyse future studies of genetic dissection of complex traits, tracing relationships in breeding programmes, and monitoring genetic diversity in commercial and wild populations of L. vannamei . 相似文献

Catalase is an antioxidant enzyme that plays a very important role in the protection against oxidative damage by breaking down hydrogen peroxide. It is a very highly conserved enzyme that has been identified from numerous species including bacteria, fungi, plants and animals, but the information about catalase in crustaceans is very limited. A cDNA containing the complete coding sequence for catalase from the shrimp Penaeus (Litopenaeus) vannamei was sequenced and the mRNA was detected by RT-PCR in selected tissues. Catalase was detected in hepatopancreas crude extracts by Western blot analysis with anti-human catalase polyclonal antibodies. The nucleotide sequence is 1692 bp long, including a 72-bp 5′-UTR, a coding sequence of 1515 bp and a 104-bp 3′-UTR. The deduced amino acid sequence corresponds to 505 amino acids with high identity to invertebrate, vertebrate and even bacterial catalases and contains the catalytic residues His71, Asn144, and Tyr354. The predicted protein has a calculated molecular mass of 57 kDa; which coincides with the size of the subunit (55 kDa) and the tetrameric protein (230 kDa) detected in hepatopancreas extracts under native conditions. - 1.1. Rostrum, carapace and abdominal regions of the exoskeletons of juvenile Penaeus californiensis (3–4 g) were analyzed for calcium, magnesium and phosphorus levels at 0. 12, 24, 48, 72 and 96 hr postmolt.
- 2.2. Calcium and magnesium reached relatively constant levels within 24 hr postmolt. Rostrum and carapace calcium values (maxium) were higher, 12–14% than those of the abdomen, 8%. Likewise, rostrum and carapace magnesium values (maxium) were higher, 0.9–1.2%. than those of the abdomen, 0.6%. There was no consistent pattern in changes in phosphorus with regard to body parts and time. Phosphorus values (maxium) varied from 0.5 to 1.2%.
- 3.3. Dry weight of all body parts increased rapidly between 0 and 24 hr postmolt. Gradual increases from 24 to 96 hr suggested that there was continuous, but reduced, synthesis of matrix during that period.

A failure in the aeration system of Litopenaeus vannamei rearing with biofloc technology can decrease the oxygen concentrations rapidly and also increase the carbon dioxide (CO2) concentrations at the same rate. We report here an evaluation of the effect of CO2 on the oxygen consumption of L. vannamei. We used a continuous-flow respirometer with water recirculation equipped with a digital fiber-optic oximeter. Eight juveniles of L. vannamei (12.1 ± 1. 4 g) were used in each treatment with one per respiratory chamber (0.6 L). The shrimp were exposed to six concentrations of CO2 (5, 30, 60, 95, 150, and 300 mgCO2/L) with an acute exposure time of six hours. Upon treatment with 5–30 mgCO2/L, we observed a consumption of oxygen of 0.233 ± 0.129 and 0.33 ± 0.072 mgO2/g/h, respectively. Upon treatment with 60 mgCO2/L, an increase was observed in the oxygen consumption (0.521 ± 0.098 mgO2/g/h). Upon treatment with 95, 150, and 300 mgCO2/L however, the shrimp decreased their oxygen consumption and lost their equilibrium. The CO2 should therefore be maintained at a maximum of 5 mgCO2/L during shrimp rearing. 相似文献

Heat shock protein 70s (HSP70s) are fundamental chaperone proteins that are indispensable to most living organisms. In order to investigate the function of HSP70 and heat shock response in shrimp, a heat shock cognate (HSC70) gene of the white shrimp (Litopenaeus vannamei), containing a 1959-bp open reading frame, was cloned and characterized. The amino acid sequence, 71.5 kDa of molecular weight, shares 80-99.6% homology with 12 diverse species' HSP70s and HSC70s. In fact, some segments of the eukaryotic HSC70 sequence, such as ATP/GTP-binding site, cytoplasmic HSP70 C-terminal sequence, and GGMP/GAP repeats, are also found in the putative shrimp HSC70. Moreover, multi-tissue RT-PCR was performed to assay the basal expressions of HSC70 in the heart, gill, hepatopancreas, stomach, gut, and muscle. The results demonstrate that the basal expressions of HSC70 in theses organs are similar to that of beta-actin. Furthermore, quantitative real-time experiments showed that HSC70 was up-regulated in hepatopancreas (4.6-fold), stomach (5.9-fold), gut (2.6-fold), and muscle (3.5-fold) but not in the heart (1.7-fold) and gill (1.6-fold) after 2 h of heat shock. Nevertheless, the HSC70 was found to be highly expressed in the heart and gill following 6 h of heat shock. This suggests that HSC70 in white shrimp possess both short-term and long-term responses to heat shock stress, indicating this HSC70 may be a heat-dependent HSC70 member. Finally, we constructed an expression vector to generate HSC70 in Escherichia coli BL21, which displayed immune cross-reactivity with mouse HSP70 antibody. In conclusion, the identification and expression of white shrimp HSC70 gene present useful data for studying the molecular mechanism of heat shock response and the effect of heat shock proteins in shrimps' cytoprotection. 相似文献

Pyloric pattern-generating neurons that control the pyloric region of the foregut were identified in the stomatogastric ganglion
of the most primitive decapod genus Penaeus. Five types of motor neurons and one interneuron are involved in generation of pyloric motor pattern. One cell type of motor
neurons innervates muscles of both the gastric mill and the pylorus like the gastric motor neurons in Cancer, but unlike those in Panulirus. These identified neurons are connected to each other either by electrical or inhibitory chemical synapses to construct the
neural circuit. This pyloric circuit is similar to the homologous circuit of other crustacean species though some differences
are seen in synaptic connections, supporting the hypothesis that the basic design of the neural circuit has been conserved
during evolution of the Malacostraca, and that differences have occurred in the synaptic connectivity as the foregut structure
has become complex. The motor neurons use either acetylcholine or glutamate as a neurotransmitter like in reptantians. The
foregut structure, the number of the pyloric cells, muscle innervation, neurotransmitters, and circuitry are compared among
malacostracan crustaceans to provide insight into how the neural circuits change and evolve to produce the motor patterns
mediating behaviour.
