Aims:  To evaluate quorum sensing (QS) inhibitory activity of plant essential oils using strains of Chromobacterium violaceum (CV12472 and CVO26) and Pseudomonas aeruginosa (PAO1).
Methods and Results:  Inhibition of QS-controlled violacein production in C. violaceum was assayed using disc diffusion and agar well diffusion method. Of the 21 essential oils, four oils showed varying levels of anti-QS activity. Syzygium aromaticum (Clove) oil showed promising anti-QS activity on both wild and mutant strains with zones of pigment inhibition 19 and 17 mm, respectively, followed by activity in cinnamon, lavender and peppermint oils. The effect of clove oil on the extent of violacein production was estimated photometrically and found to be concentration dependent. At sub-MICs of clove oil, 78·4% reduction in violacein production over control and up to 78% reduction in swarming motility in PAO1 over control were recorded. Gas chromatography–mass spectrometry analysis of clove oil indicated presence of many phytocompounds. Eugenol, the major constituent of clove oil could not exhibit anti-QS activity.
Conclusions:  Presence of anti-QS activity in clove oil and other essential oils has indicated new anti-infective activity. The identification of anti-QS phytoconstituents is needed to assess the mechanism of action against both C. violaceum and Ps. aeruginosa .
Significance and Impact of the study:  Essential oils having new antipathogenic drugs principle because of its anti-QS activity might be important in reducing virulence and pathogenicity of drug-resistant bacteria in vivo .  相似文献

Effect of growing seedling, seeds and seedlings extracts from seven leguminous plants (Pisum sativum, Vigna radiata, Vigna mungo, Cajanus cajan, Lentil culinaris, Cicer arietinum and Trigonella foenum graecum) were screened for their ability to influence quorum sensing controlled pigment production in Chromobacterium violaceum indicator strains (CV12472 and CVO26). Germinating seedling and seedling extracts of only P. sativum (pea) showed inhibition of violacein production. Interestingly, the T. foenum graecum (fenugreek) seed extracts enhances the pigment production. Quorum sensing regulated swarming motility in Pseudomonas aerugionsa PAO1 was reduced by pea seedling extract while enhanced by the fenugreek seed extracts. These findings suggest that plant metabolites of some legumes interact actively with bacterial quorum sensing and could modulate its associated functions.  相似文献

目的:从海洋真菌中筛选得到新型群体感应抑制剂,并对其进行活性评价。方法:首先利用紫色杆菌CV026指示菌株对真菌发酵粗提物进行活性筛选。其次通过18S r DNA序列比对进行菌种鉴定,同时采用硅胶柱色谱、凝胶柱色谱和高效液相色谱等技术并结合活性追踪检测分离纯化的活性化合物,再通过核磁质谱分析确定其结构。最后利用定量测定方法检测其在亚抑菌浓度下对紫色杆菌紫色菌素产量影响以及RT-PCR检测与QS调控相关基因的m RNA表达的影响。结果:从海藻共生菌中筛选到一株具有紫色杆菌群体感应抑制活性的海洋真菌Penicillium sp.QF046,其次级代谢产物中纯化到的活性化合物根据结构鉴定为一种星形曲霉毒素(asteltoxin)。该化合物对于紫色杆菌群体感应抑制浓度低于阳性对照化合物呋喃酮C30,同时抑制了群体感应相关基因m RNA水平的表达。结论:从海洋真菌Penicillium sp.QF046代谢产物中发现了一种抑制紫色杆菌群体感应的星形曲霉毒素,为进一步通过结构改造研发新型抗菌药物提供良好的前体化合物。  相似文献

许多致病菌的致病机制依赖于群体感应系统的调控,经实验证明群体感应系统突变或缺失的菌株致病能力显著下降,筛选高效的群体感应抑制剂有望成为解决细菌感染以及细菌耐药性问题的一个有效途径。从海洋软体动物体内分离海洋真菌69株,发酵液粗提物经QSIS2 (Quorum Sensing Inhibitor Selector 2) 筛选模型和紫色杆菌CV026指示菌株筛选后得到编号QY013的粗提物具有群体感应抑制活性,进一步实验表明该粗提物能够显著降低铜绿假单胞菌群体感应调控的毒力因子绿脓菌素的产量,以及紫色杆菌群体感应调控的紫色菌素的产量,且在有效浓度范围内对细菌生长不产生影响。形态学特征和18S rDNA序列分析表明菌株QY013为Penicillium属。文中筛选到一株具有细菌群体感应抑制活性的海洋来源真菌,其发酵液粗提物中的有效活性成分可用于新型抗菌药物的研究。  相似文献

细菌性疾病是危害水产业健康发展的主要原因之一,从抑制调控致病菌毒力表达的群体感应(Quorum sensing,QS)入手是水产动物疾病防治研究新的热点方向。研究以紫色杆菌 (Chromobacterium violaceum)为报告菌株,采用T型划线和滤纸片法,对分离自异育银鲫肠道及实验室保存的芽孢杆菌F3-1、F3-2、F6-4、F6-5、X77、X93、X3914进行分析测测试,筛选具有群体感应抑制效果的菌株。T型划线结果显示菌株F3-1能够抑制紫色杆菌紫色菌素的产生;滤纸片法结果显示抑制紫色杆菌紫色菌素产生的主要物质存在于菌株F3-1的胞外产物中。试验提取了菌株F3-1的培养物的粗提物,测定粗提物浓度分别在0、50、100、200、300 mg/L时紫色杆菌菌素在585 nm处的光密度值,结果显示随着粗提物浓度的增加其色素的产量显著降低(P0.05),当粗提物浓度超过200 mg/L时,试管中观察不到紫色菌素存在;利用硫酸铵盐析的方法,提取了菌株F3-1的粗提物中蛋白产物,滤纸片扩散法结果显示表明其胞外蛋白是紫色杆菌QS系统的抑制物质。通过分子生物学16S rDNA鉴定,确定菌株F3-1为短小芽孢杆菌(Bacillus pumilus,GenBank accession No. JX984444)。电镜照片显示该菌具有短杆状特征,因此,分离自异育银鲫肠道的短小芽孢杆菌F3-1具有抑制细菌群体感应的作用。    相似文献

An ethyl acetate extracts isolated from a marine fungal strain, Penicillium chrysogenum DXY-1, obtained from marine sediments surrounding the East Sea, was found to exhibit anti-quorum sensing (anti-QS) activity. Interestingly, a novel active compound was identified as tyrosol by the purification and structural characterization. At a concentration of 0.5 mg/mL, tyrosol decreased QS-regulated violacein production in Chromobacterium violaceum CV026 by 53.5% and decreased QS-regulated pyocyanin production, elastase activity and proteolytic activity in Pseudomonas aeruginosa PA01 by 63.3%, 57.8% and 9.9%, respectively. SEM images showed that tyrosol inhibited biofilm formation in P. aeruginosa PA01 without having any effect on bacterial growth. Molecular docking results revealed that the natural signal molecule C6HSL and tyrosol bound to different receptor pockets of CviR, and tyrosol inhibited the QS activity of CviR in C. violaceum by binding to the DNA-binding domain and blocking pathogenic gene expression. All the data suggest that tyrosol may act as a potential inhibitor of the QS systems to solve the looming crisis of bacterial resistance. We believe that there are other active compounds with relatively high anti-QS activity or synergistic inhibitory effects on QS in the crude extract, which warrants further research.  相似文献

Inhibition of quorum sensing (QS)-regulated virulence factors including biofilm is a recognized anti-pathogenic drug target. The search for safe and effective anti-QS agents is expected to be useful to combat diseases caused by multidrug-resistant bacteria. In this study, effect of a commonly used antibiotic, doxycycline on QS was evaluated using sensor strains of Chromobacterium violaceum (ATCC 12472 and CVO26) and Pseudomonas aeruginosa PAO1. Sub-MICs of doxycycline reduced QS-controlled violacein production in C. violaceum to a significant degree (70 %) and showed a significant reduction of LasB elastase (67.2 %), pyocyanin (69.1 %), chitinase (69.8 %) and protease (65 %) production and swarming motility (74 %) in P. aeruginosa PAO1 over untreated controls. Similar results were also recorded against a clinical strain of P. aeruginosa (PAF-79). Interestingly, doxycycline at respective sub-MICs (4 and 32 μg ml?1) significantly reduced the biofilm-forming capability and exopolysaccharide production in both the strains of P. aeruginosa (PAO1 and PAF-79) over untreated controls. The results of this study highlight the multiple actions of doxycycline against QS-linked traits/virulence factors and its potential to attenuate virulence of P. aeruginosa.  相似文献

Psidium guajava L., which has been used traditionally as a medicinal plant, was explored for anti‐quorum sensing (QS) activity. The anti‐QS activity of the flavonoid (FL) fraction of P. guajava leaves was determined using a biosensor bioassay with Chromobacterium violaceum CV026. Detailed investigation of the effects of the FL‐fraction on QS‐regulated violacein production in C. violaceum ATCC12472 and pyocyanin production, proteolytic, elastolytic activities, swarming motility and biofilm formation in Pseudomonas aeruginosa PAO1 was performed using standard methods. Possible mechanisms of QS‐inhibition were studied by assessing violacein production in response to N‐acyl homoserine lactone (AHL) synthesis in the presence of the FL‐fraction in C. violaceum ATCC31532 and by evaluating the induction of violacein in the mutant C. violaceum CV026 by AHL extracted from the culture supernatants of C. violaceum 31532. Active compounds in the FL‐fraction were identified by liquid chromatography–mass spectrometry (LC–MS). Inhibition of violacein production by the FL‐fraction in a C. violaceum CV026 biosensor bioassay indicated possible anti‐QS activity. The FL‐fraction showed concentration‐dependent decreases in violacein production in C. violaceum 12472 and inhibited pyocyanin production, proteolytic and elastolytic activities, swarming motility and biofilm formation in P. aeruginosa PAO1. Interestingly, the FL‐fraction did not inhibit AHL synthesis; AHL extracted from cultures of C. violaceum 31532 grown in the presence of the FL‐fraction induced violacein in the mutant C. violaceum CV026. LC–MS analysis revealed the presence of quercetin and quercetin‐3‐O‐arabinoside in the FL‐fraction. Both quercetin and quercetin‐3‐O‐arabinoside inhibited violacein production in C. violaceum 12472, at 50 and 100 μg/mL, respectively. Results of this study provide scope for further research to exploit these active molecules as anti‐QS agents.  相似文献

Quorum sensing (QS) is a widespread mechanism utilized by both Gram (+) and Gram (?) bacteria for communication and regulation of specific virulence traits and phenotypes due to population density. Plants are known to produce a number of compounds that can inhibit this communication and most of them have been discovered through well-known Chromobacterium violaceum biomonitor strains assays. In this study, we have analyzed 36 extracts, from 26 Turkish plant species, for their effects on bacterial growth and inhibition or induction of QS in the said biomonitoring assay. Four of the crude plant extracts from Tanacetum balsamita L. subsp. balsamitoides (Compositaceae), Epilobium angustifolium L. (Onograceae), Quercus frainetto Ten. (Fagaceae) and Quercus robur L. (Fagaceae) showed QS inhibitory activity and significantly reduced violacein production in C. violaceum. Particularly, ethyl acetate soluble compounds extracted from leaves of Quercus spp. were efficient in QS inhibition without any apparent increase in bacterial growth. On the other hand, extracts from Mentha longifolia subsp. longifolia and Hypericum orientale showed enhanced violacein production, thus increasing QS-dependent behaviour. Our results clearly demonstrate the QS-inhibitory effects of Q. frainetto Ten., E. angustifolium and T. balsamita L. extracts, while two other species showed QS-inducing effects, for the first time.  相似文献

Inhibition of marine biofouling by bacterial quorum sensing inhibitors   总被引:1,自引:0,他引:1  
Seventy eight natural products from chemical libraries containing compounds from marine organisms (sponges, algae, fungi, tunicates and cyanobacteria) and terrestrial plants, were screened for the inhibition of bacterial quorum sensing (QS) using a reporter strain Chromobacterium violaceum CV017. About half of the natural products did not show any QS inhibition. Twenty four percent of the tested compounds inhibited QS of the reporter without causing toxicity. The QS inhibitory activities of the most potent and abundant compounds were further investigated using the LuxR-based reporter E. coli pSB401 and the LasR-based reporter E. coli pSB1075. Midpacamide and tenuazonic acid were toxic to the tested reporters. QS-dependent luminescence of the LasR-based reporter, which is normally induced by N-3-oxo-dodecanoyl-L-homoserine lactone, was reduced by demethoxy encecalin and hymenialdisin at concentrations >6.6 μM and 15 μM, respectively. Hymenialdisin, demethoxy encecalin, microcolins A and B and kojic acid inhibited responses of the LuxR-based reporter induced by N-3-oxo-hexanoyl-L-homoserine lactone at concentrations >0.2 μM, 2.2 μM, 1.5 μM, 15 μM and 36 μM, respectively. The ability to prevent microfouling by one of the compounds screened in this study (kojic acid; final concentrations 330 μM and 1 mM) was tested in a controlled mesocosm experiment. Kojic acid inhibited formation of microbial communities on glass slides, decreasing the densities of bacteria and diatoms in comparison with the control lacking kojic acid. The study suggests that natural products with QS inhibitory properties can be used for controlling biofouling communities.  相似文献

We studied the effect of the quorum-sensing (QS) blockers 5-hydroxy-3[(1R)-1-hydroxypropyl]-4-methylfuran-2(5H)-one (FUR1), (5R)-3,4-dihydroxy-5-[(1S)-1,2-dihydroxyethyl]furan-2(5H)-one (FUR2) and triclosan (TRI) on the formation of bacterial biofilms, and the effect of these biofilms on the larval attachment of the polychaete Hydroides elegans and the bryozoan Bugula neritina. 14-day-old subtidal biofilms were harvested from artificial substrata and were allowed to develop in the laboratory with and without QS blockers. QS blockers inhibited the production of violacein by the QS reporter strain Chromobacterium violaceum CV026 and did not affect the metabolic activity of bacteria in multispecies biofilms. At a concentration of 10(-3) M all three tested compounds inhibited the establishment of microbial communities, but at one of 10(-4) M only FUR2 inhibited establishment. The tested QS blockers caused changes in bacterial density and bacterial community structure, as revealed by terminal restriction fragment length polymorphism and FISH. The groups most affected by QS blockers were Alphaproteobacteria, Gammaproteobacteria and the Cytophagales. Larvae of H. elegans and B. neritina avoided settling on biofilms that had developed in the presence of QS blockers. Our results suggest that QS blockers directly control the formation of multi-species biofilms, and indirectly - by means of biofilm properties - affect larval attachment on these modified biofilms.  相似文献

Inhibition of bacterial quorum sensing by vanilla extract   总被引:2,自引:0,他引:2  
AIMS: The purpose of this study was to search for a novel quorum sensing inhibitor and analyse its inhibitory activity. METHODS AND RESULTS: Quorum sensing inhibition was monitored using the Tn-5 mutant, Chromobacterium violaceum CV026. Vanilla beans (Vanilla planifolia Andrews) were extracted using 75% (v/v) aqueous methanol and added to C. violaceum CV026 cultures. Inhibitory activity was measured by quantifying violacein production using a spectrophotometer. The results have revealed that vanilla extract significantly reduced violacein production in a concentration-dependent manner, indicating inhibition of quorum sensing. CONCLUSIONS: Vanilla, a widely used spice and flavour, can inhibit bacterial quorum sensing. SIGNIFICANCE AND IMPACT OF THE STUDY: The results suggest that the intake of vanilla-containing food materials might promote human health by inhibiting quorum sensing and preventing bacterial pathogenesis. Further studies are required to isolate specific substances from vanilla extract acting as quorum sensing inhibitors.  相似文献

Quorum sensing inhibition activity of garlic extract and p-coumaric acid   总被引:1,自引:0,他引:1  
Aims:  The goal of this work was to investigate the influence of DMSO, garlic extract and p -coumaric acid on bacterial quorum sensing (QS).
Methods and Results:  The decreases in the QS responses of QS reporter strains Escherichia coli pSB401 and pSB536, Agrobacterium tumefaciens NTL4, Chromobacterium violaceum 5999 and wt 494, Pseudomonas putida IsoF/gfp and environmental Pseudomonas chlororaphis were quantified in relation to growth inhibitory effects. DMSO showed no significant QS-specific effects on the strains tested even at close-to-lethal concentrations. Garlic extracts antagonized the activity of QS receptors LuxR, AhyR and TraR, but were toxic at higher concentrations. P -coumaric acid fully inhibited QS responses of 5999, NTL4 and P. chlororaphis , with no influence on cell viability.
Conclusions:  The quorum sensing inhibition activity of garlic was extended to novel receptors, and p -coumaric acid was found to possess previously undescribed QS antagonist properties.
Significance and Impact of the Study:  The results suggest that p -coumaric acid might act as QS inhibitor. Further studies are required to understand its role in the regulation of QS and investigate structurally related compounds.  相似文献

We studied the role of bacterial secondary metabolites in the context of grazing protection against protozoans. A model system was used to examine the impact of violacein-producing bacteria on feeding rates, growth, and survival of three common bacterivorous nanoflagellates. Freshwater isolates of Janthinobacterium lividum and Chromobacterium violaceum produced the purple pigment violacein and exhibited acute toxicity to the nanoflagellates tested. High-resolution video microscopy revealed that these bacteria were ingested by the flagellates at high rates. The uptake of less than three bacteria resulted in rapid flagellate cell death after about 20 min and cell lysis within 1 to 2 h. In selectivity experiments with nontoxic Pseudomonas putida MM1, flagellates did not discriminate against pigmented strains. Purified violacein from cell extracts of C. violaceum showed high toxicity to nanoflagellates. In addition, antiprotozoal activity was found to positively correlate with the violacein content of the bacterial strains. Pigment synthesis in C. violaceum is regulated by an N-acylhomoserine lactone (AHL)-dependent quorum-sensing system. An AHL-deficient, nonpigmented mutant provided high flagellate growth rates, while the addition of the natural C. violaceum AHL could restore toxicity. Moreover, it was shown that the presence of violacein-producing bacteria in an otherwise nontoxic bacterial diet considerably inhibited flagellate population growth. Our results suggest that violacein-producing bacteria possess a highly effective survival mechanism which may exemplify the potential of some bacterial secondary metabolites to undermine protozoan grazing pressure and population dynamics.  相似文献

Quorum sensing inhibitors (QSIs) that specifically interfere with bacterial cell-to-cell communication are considered as an alternative approach to conventional antibacterial therapy. In our study, a set of twenty-six fumardiamides with a quinoline head-group were evaluated as potential QSIs. Two strains of Gram-negative Chromobacterium violaceum (violacein-producing strain ATCC31532 and violacein-negative, mini-Tn5 mutant derivative CV026) were used as QS reporters for testing anti-QS and bactericidal activity of various quinoline fumardiamides. The initial screening of eighteen fumardiamides with primaquine, mefloquine and chloroquine scaffolds identified chloroquine derivatives as the most promising QSIs. Tail-group optimization of chloroquine fumardiamides led to the most active compounds 27, 29 and 30 bearing aminoethyl or piperidine moieties. At 400 µM concentration, these compounds inhibited the QS of C. violaceum strains in a manner similar to quercetin (the model QSI), while at the 40 µM concentration their inhibitory effect was twice less than that of quercetin. As none of the compounds displayed a bactericidal effect and that the QS inhibition was specific to the CV026 strain, our findings indicate that the structurally optimized chloroquine derivatives could function as quorum quenching (QQ) agents with a potential to block the signaling without entering the cell. In conclusion, our finding provides an important step toward the further design of agents targeting cell-to-cell communication.  相似文献

Many Gram-negative bacteria use N-acyl homoserine lactone signal molecules to monitor their own population density and coordinate gene regulation in a process called quorum sensing (QS). Increasing evidence implies that certain eukaryotes produce QS-inhibitory compounds. In this work, we tested 46 terrestrial plants materials for their ability to inhibit QS-regulated behaviors in different bacterial species. Plant materials were dried and extracted using different solvents. The chloroform-soluble compounds extracted from Scorzonera sandrasica were found to inhibit violacein production, a QS-regulated behavior in Chromobacterium violaceum. In addition, the chloroform extract was also able to inhibit QS-regulated carbapenem antibiotic production in Erwinia carotovora. Because the regulation of many bacterial processes is controlled by QS systems, the finding of natural compounds acting as QS inhibitors suggests an attractive tool to control and handle detrimental infections caused by human, animal, and plant pathogens.  相似文献

Macroalgae are an important source of antimicrobial compounds. However, it is unclear if these compounds are produced by the algae themselves, by their associated bacteria, or by both. The main aim of this study was to investigate the potential of macroalgae and their associated microorganisms to inhibit bacterial quorum sensing (QS) and growth. Before extraction, half of the algal specimens were treated with 30% ethanol to remove surface associated bacteria. Canistrocarpus cervicornis extracts were able to inhibit QS of the reporter Chromobacterium violaceum CV017, where extracts with associated bacteria were more efficient than those without bacteria. However, not all algal extracts that inhibited QS of CV017 were able to inhibit bacterial attachment of Pseudomonas aeruginosas PA01, showing specific activity of algal metabolites. Only 58% of the extracts showed antibacterial activity against eight marine fouling and pathogenic bacterial strains tested. Our data suggests that algae and their associated microbiota are important sources of antimicrobial compounds which potentially can be used in future biotechnological applications.  相似文献
