Aggregation of mouse embryos produced 11 chimaeras Miwh/+C/C----+/+c/c and 8 chimaeras +/+C/C----+/+c/c (control). Chimaerism was detected by mosaicism of coat retinal pigment epithelium and by electrophoretic pattern of glucose phosphate isomerase. All chimaeras showed a common pattern of pigmented and unpigmented hair regions that alternated as stripes of different length and width and extended from spine in lateral-ventral direction. However, white coat color predominated in Miwh/+C/C----+/+c/c chimaeras due to a higher proportion of unpigmented zones as well as to weakening of hair color in pigmented areas. Besides, distal regions of limbs were always unpigmented in Miwh/+C/C----+/+c/c chimaeras and completely or partially pigmented in +/+C/C----+/+c/c chimaeras. Pigmented hair regions are often located on the ventral trunk surface where the Miwh/+ heterozygotes usually had an unpigmented spot. The examination of hairs, taken from the same regions of gray coloration, revealed the presence of pigmented, unpigmented and mosaic hairs. The proportion of unpigmented hairs was much higher in Miwh/+C/C----+/+c/c chimaeras than in +/+C/C----+/+c/c chimaeras. The data obtained indicate that a single Miwh gene dose reduced proliferative activity of melanoblasts which resulted in weakening of coat pigmentation.  相似文献

In vitro clonal analysis of quail cardiac neural crest development   总被引:1,自引:0,他引:1  
The developmental potentials of cardiac neural crest cells were investigated by in vitro clonal analysis. Five morphologically distinct types of clones were observed: (1) "pigmented" clones contained melanocytes only; (2) "mixed" clones consisted of pigmented and unpigmented cells; (3) "unpigmented dense" clones consisted of flattened, closely aligned unpigmented cells; (4) "unpigmented loose" clones consisted of a few loosely arranged, flattened cells; and (5) "unpigmented large" clones included a large number of small, stellate cells that were highly proliferative. The binding patterns of antibodies against lineage-specific markers showed that cells in the different clones expressed characteristic phenotypes. The following phenotypes were expressed in addition to pigment cells: smooth muscle cells, connective tissue cells, chondrocytes, and cells in the sensory neuron lineage. Mixed clones expressed all five phenotypes. Unpigmented dense clones contained smooth muscle cells, connective tissue cells, chondrocytes, and sensory neurons. Unpigmented loose clones exclusively consisted of smooth muscle cells, whereas unpigmented large clones contained chondrocytes and sensory neuron precursors. Based on these results, the following conclusions can be drawn: (1) Pigmented and unpigmented loose clones are most likely formed by precursors that are committed to the melanogenic and myogenic cell lineages, respectively. (2) Mixed and unpigmented dense clones are derived from pluripotent cells with the capacity to give rise to four or five phenotypes. (3) Unpigmented large clones originate from progenitor cells that appear to have a partially restricted developmental potential, that is, these cells are capable of generating two phenotypes in clonal cultures. Thus, the data indicate that the early migratory cardiac neural crest is a heterogeneous population of cells, consisting of pluripotent cells, cells with a partially restricted developmental potential, and cells committed to a particular cell lineage.  相似文献

The differentiation of Callitriche hermaphroditica in Sweden expressed in fruit morphology has been studied. Variation among 50 wild-scored populations is illustrated by canonical variates analysis and cluster analysis and shown to be geographically correlated. Two major geographical races differing in fruit size are distinguished and confirmed by the study of herbarium collections. The anatomy of the fruits is described, based on light microscopical studies. An anatomically aberrant fruit form was found in two different areas in Sweden. Comparisons are made with herbarium collections from Denmark, Norway, Finland and Soviet Karelia. The biogeography, in the light of postglacial events, and the taxonomy of C. hermaphroditica are discussed and a distribution map of C. hermaphroditica s. lat. in Sweden is presented.  相似文献

Phenol oxidase (PO) of Penicillium cyclopium belongs to the laccases (E.C. 1.14.18.1). By PAA-gel electrophoresis three forms of the enzyme were detected. Most of the PO in emerged sporulating cultures accumulates in the cell wall of the conidiospores. However, a high percentage is not bound to the wall constituents and can be easily removed with aqueous solutions. This soluble fraction was 570-fold enriched by (NH4)2SO4-precipitation, followed by chromatography on sephadex G 200 and on hydroxylapatite-cellulose, The necessity of PO for conidiospore pigmentation was shown by means of suspended spores whose rates of pigment accumulation resemble those of conidia maturing in situ. Pigmentation is stopped if PO is inhibited by diethyldithiocarbamate. This corresponds to the fact that in strains with heavily pigmented conidiospores PO activity is much higher than in strains with weakly pigmented or unpigmented spores. Also the time course of PO accumulation in the latter strains is different from that of the former. Furthermore, in mutant res-eth 1 which has unpigmented conidiospores, PO isoenzyme no. 3 could not be detected.  相似文献

Intricate coordinated mechanisms that govern the synchrony of hair growth and melanin synthesis remain largely unclear. These two events can be uncoupled in prematurely gray hair, probably due to oxidative insults that lead to the death of oxidative stress-sensitive melanocytes. In this study, we examined the gene expression profiles of middle (bulge) and lower (hair bulb) segments that had been micro-dissected from unpigmented and from normally pigmented hair follicles from the same donors using quantitative real-time RT-PCR (qPCR) arrays. We found a significant down-regulation of melanogenesis-related genes (TYR, TYRP1, MITF, PAX3, POMC) in unpigmented hair bulbs and of marker genes typical for melanocyte precursor cells (PAX3, SOX10, DCT) in unpigmented mid-segments compared with their pigmented analogues. qPCR, western blotting and spin trapping assays revealed that catalase protein expression and hydroxyl radical scavenging activities are strongly repressed in unpigmented hair follicles. These data provide the first clear evidence that compromised antioxidant activity in gray hair follicles simultaneously affects mature hair bulb melanocytes and their immature precursor cells in the bulge region.  相似文献

The early marsh orchid, Dactylorhiza incarnata (L.) Soó s. l., grows in medium-rich to rich fens and marshes over much of Europe and parts of Asia. The species is highly polymorphic and different forms may grow together at the same site. In the present study, I tested the hypothesis that these forms represent different migrant populations that have colonized Scandinavia independently of each other, possibly from different source areas. Accessions from Scandinavia and elsewhere were screened for variation at three size-variable plastid marker loci, one polyA repeat, one polyA-polyTA-polyT repeat and one 9 bp indel. Ten haplotypes were defined on basis on the combined variation pattern. The common occurrence of several haplotypes in southern Scandinavia and adjacent areas to the south and the east of the Baltic Sea suggests that D. incarnata has been dispersed on repeated occasions across the Baltic. Also, there was some correlation between haplotype composition and morphological form on the island of Gotland, in agreement with the independent colonization hypothesis. Material from northernmost Sweden, Finland and northwest Russia was fixed for a single widespread haplotype, indicating that populations in this area are located farther away from the Pleistocene refugia. Dactylorhiza incarnata ssp. lobelii from southwest Norway was characterized by a haplotype that was not encountered elsewhere in Scandinavia. Given its proximity to British populations dominated by the same haplotype, it is suggested that D. incarnata ssp. lobelii was established independently of the other Scandinavian populations, from coastal refugia located in western Europe.  相似文献

The phylogenetic position of Orbiniidae within Annelida is unresolved. Conflicting hypotheses place them either in a basal taxon Scolecida, close to Spionida, or in a basal position in Aciculata. Because Aciculata have a specific type of eye, the photoreceptive organs in the orbiniid Scoloplos armiger were investigated to test these phylogenetic hypotheses. Two different types of prostomial photoreceptor‐like sense organs were found in juveniles and one additional in subadults. In juveniles there are four ciliary photoreceptor‐like phaosomes with unbranched cilia and two pigmented eyes. The paired pigmented eyes lie beside the brain above the circumoesophageal connectives. Each consists of one pigmented cell, one unpigmented supportive cell and three everse rhabdomeric sensory cells with vestigial cilia. During development the number of phaosomes increases considerably and numerous unpigmented sense organs appear consisting of one rhabdomeric photoreceptor cell and one supportive cell. The development and morphology of the pigmented eyes of S. armiger suggest that they represent miniaturized eyes of the phyllodocidan type of adult eye rather than persisting larval eyes resulting in small inverse eyes typical of Scolecida. Moreover, the structure of the brain indicates a loss of the palps. Hence, a closer relationship of Orbiniidae to Phyllodocida is indicated. Due to a still extensive lack of ultrastructural data among polychaetes this conclusion cannot be corroborated by considering the structure of the unpigmented ciliary and rhabdomeric photoreceptor‐like sense organs. Data from the national dairy cow recording systems during 1997 were used to calculate lactation-specific cumulative risk of mastitis treatments and cumulative risk of removal from the herds in Denmark, Finland Norway and Sweden. Sweden had the lowest risk of recorded mastitis treatments during 305 days of lactation and Norway had the highest risk. The incidence risk of recorded mastitis treatments during 305 days of lactation in Denmark, Finland, Norway and Sweden was 0.177, 0.139, 0.215 and 0.127 for first parity cows and 0.228, 0.215, 0.358 and 0.204 for parities higher than three, respectively. The risk of a first parity cow being treated for mastitis was almost 3 times higher at calving in Norway than in Sweden. The period with the highest risk for mastitis treatments was from 2 days before calving until 14 days after calving and the highest risk for removal was from calving to 10 days after calving in all countries.The study clearly demonstrated differences in bovine mastitis treatment patterns among the Nordic countries. The most important findings were the differences in treatment risks during different lactations within each country, as well as differences in strategies with respect to the time during lactation mastitis was treated.  相似文献

Mice homozygous for the recessive patchwork (pwk) mutation are characterized by a variegated pigment pattern with a mixture of unpigmented and normally pigmented hairs. The pigmented hair bulbs contain functional melanocytes. By contrast, the unpigmented hair bulbs contain no melanocytes. This lack results from the death of melanoblasts in the hair follicle at the end of embryogenesis. Here, we report that melanoblasts and melanocytes are found in the epidermis of pwk/pwk mice. Furthermore, these epidermal pigment cells are able to colonize new hair follicles after skin wounding. Despite the presence of epidermal pigment cells with a colonization potential, a follicle that had produced an unpigmented hair produces a new unpigmented hair during the successive hair growth cycles. This hair color continuity is also true for the pigmented hair follicles. Endosperm maize cultures derived from a strain homozygous for all genes required for anthocyanin synthesis develop an intense pigmentation. Pigmenting ability is generally maintained in successive subcultures, altough colourless areas are frequently observed in pigmented cultures. The isolated colourless cell clusters show a growth rate higher than the coloured ones. These calli nevertheless do not lose the ability to synthesize anthocyanins, and in successive subcultures turn red again.The different growth rates associated with the ability of cells to accumulate pigments suggest the existence of different physiological states of the culture. To investigate this possibility we analyzed the polypeptide patterns of coloured and colourless cultures. SDS gel electrophoresis has demonstrated differences in soluble protein fractions, among which a 26 kD peptide, characteristic of pigmented tissues, has been evidenced. Retinal pigmented epithelia (RPE) isolated from chicken embryos of various developmental stages were dissociated into single cells, and their ability to re-express defferentiated characteristics in clonal culture was investigated. The lighty pigmented, columnar cells isolated from stage 25 to 29 embryos dissociated more easily than the heavily pigmeted, cuboidal cells from embryos of stages 30 to 34. The yield of RPE cells per embryo increased with donor age, paralleling the growth of the epithelium in vivo . However, the potential these cells to attach, to proliferate, and to form typical, welldifferentiated RPE colonies declined with donor age. Cells from stage 25 embryos developed exclusively into large, typical epithelial colonies which expressed all stages of differentiation from flat, unpigmented cells at the margin to cuboidal, pigmented cells in the centre. At the other end of the spectrum, cells from stage 34 embryos frequently formed small, atypical colonies of unpigmented cells, in addition to typical but relatively small colonies. The plating efficiency (calculated on the basis of pigmented colonies formed within 3 weeks) dropped from more than 2% at stage 25 to 0.01% at stage 34.  相似文献

SUMMARY

The incubation period and percentage hatching of eggs of pigmented and unpigmented Biomphalaria glabrata at constant temperatures were investigated in the range 14 °C to 34 °C. In order to determine the influence of extreme temperatures on adult snails, specimens of the same species were exposed to 0 °C and 40 °C for selected time periods. The results indicate that sustained temperatures below 16 °C and above 32 °C are detrimental to the development and hatching of B. glabrata embryos. The optimum temperatures for incubation period and hatching differ from each other. As far as temperature is concerned, this foreign snail species should be capable of successfully colonizing the warmer parts of southern Africa.  相似文献

Lythrum salicaria, now a widespread invasive species, exhibits tristyly, a form of heteromorphic selfincompatibility. In tristyly, each plant exhibits one (and only one) of three morphologically different floral forms. Moreover, each flower produces two types of stamens, and these two exhibit different incompatibility reactions. Differences between stamens of a single flower must be the result of epigenetic phenomena and for that reason, we performed two-dimensional gel electrophoresis (2-DE) to analyze fractions of soluble proteins derived from the pollen coat and protoplast including three hydrolytic enzymes from the six different stamen types (two from each of three floral forms). There were significant differences in the 2-D protein profiles both between pollen from the same flower and between the same type of pollen from two different flowers, in the pollen coat as well as in the protoplast extracts. In five of the six samples of pollen fractions, characteristic peptides were found. Quantitative differences between pollen from the same flower were observed in case of esterases. Furthermore, analysis of proteases and acid phosphatases revealed also qualitative differences between these enzymes in pollen from the same flower.  相似文献

Superoxide dismutase, catalase, glutathione peroxidase, and glutathione reductase activities in pigmented and unpigmented liver tissues of frog and albino rat, respectively, were studied. Our results show that pigmented tissue is lacking in manganese superoxide dismutase activity and that the main enzymatic activity utilized in the cytosol by pigmented cells to reduce the hydrogen peroxide to water is represented by catalase; on the contrary, for the same reaction, the cells of albino rat liver primarily utilize the glutathione peroxidase activity. Both a low glutathione peroxidase activity and a low glutathione reductase activity were found in pigmented tissue of frog liver when compared with unpigmented tissue of rat liver. In light of our results, we also report a hypothetical interrelationship between melanin and reduced glutathione: We believe that in pigmented cells the melanin could act as a reducing physiological agent replacing the glutathione in the reduction of hydrogen peroxide. This reducing action of melanin could cause a diminished need for GSH and therefore could provoke the low glutathione peroxidase and reductase activities in pigmented tissue.  相似文献

Seven cases of human cutaneous malignant melanomas, some of them associated with distant metastases, were analyzed by electron microscopy. The obtained results indicate that the polymorphism of melanosomes can not be used to distinguish between melanomas developed on Dubreuilh's precancerous melanosis and those formed on nevi. The features of tumoral cells in pigmented tumors were different from those of cells within unpigmented tumors, and there were no cytologic differences between the primary tumor and metastases.  相似文献

Photodynamic therapy (PDT) as a regime for melanoma is of limited success due to factors such as the efficacy of the photosensitizer used, penetration depth and the presence of pigment. We characterised a pigmented and an unpigmented melanoma cell line with respect to their phenotypes. Cell viability was assessed after exposure to hypericin, a UVA-activated photosensitizer. Exposure to 3 μM activated hypericin induced a cytoprotective (autophagic) response from both cell lines. However, the pigmented cells accumulated a large amount of glycogen in their cytoplasm. We hypothesise that the treatment induces an initial cytoprotective response through autophagy, but with increased stress results in a different mode of cell death in pigmented melanoma cells from unpigmented cells. These results indicate that hypericin-PDT could be an adjuvant therapy for melanoma.  相似文献

Summary The retina of Cataglyphis bicolor was investigated by electron microscopy. Three types of structurally distinct retinulae were found and mapped throughout the compound eye: Type I is composed of four unpigmented thin cells, four larger pigmented cells as well as a basal ninth cell. Its rhabdom possesses a round cross section and four microvilli directions. This type occupies most of the dorsal two-thirds of the retina. Type II consists of two thin cells, two intermediate cells and four large cells. A basal ninth cell is also present; the rhabdom is as in type I. Type II retinulae are located in the ventral third of the retina. Type III ommatidia are unique within the Hymenoptera: there are four large pigmented cells, four thinner unpigmented cells and a basal ninth cell. The rhabdom, however, has a dumb-bell shaped cross section; two small cells lie at its opposed extremities and the remaining six cells have mutually perpendicular microvilli orientations. This type of retinula is found at the dorso-medial eye margin. Serial sectioning in this region revealed a conical shaped rhabdom without any torsion along the longitudinal axis. The rhabdomere cross section was calculated from distal and proximal thin sections. Angular statistics were applied to the microvilli directions of all three ommatidial types to determine the degree of order. A simple method to measure early euphausiid larvae is tested on 35 specimens of Euphausia superba obtained in the Weddell-Scotia Confluence region (ranging from Calyptopes I to Furcilia III) against the measures obtained on the same specimens with a graduated eyepiece. The arrangement of the test includes two observers, four magnifications (from 7.5× to 62.5×) whenever feasible and two replications of each measurement. A total of 953 measurements were analyzed in an incomplete random blocks ANOVA design not finding significant differences between magnifications, methods, observers and their interactions. It was found that the relative differences between methods were of the same magnitude as the differences between replications (approximately 5%). The proposed method is less demanding on laboratory work, thus allowing the measurement of the large number of specimens needed to estimate size frequency distributions.  相似文献

【目的】北二星蝽Eysarcoris aeneus是一种广泛分布于古北区的重要农业害虫,可危害多种经济作物。其刺肩型和钝肩型前胸背板后侧角长度存在差异,暗示其种内变异的存在。【方法】基于几何形态学的多元回归分析(multivariate regression)、主成分分析(principal component analysis, PCA)、典型变量分析(canonical variate analysis, CVA)和判别函数分析(discriminant function analysis, DFA),对采集于中国19个地区的142头标本(98头北二星蝽标本和44头作为外群的广二星蝽E.ventralis标本)的前翅、后翅、头和小盾片4个性状进行分析比较,研究北二星蝽钝肩型和刺肩型之间的形态变异。【结果】对于所研究的4个性状来说,北二星蝽钝肩型和刺肩型标本间均未检测到异速生长的存在。主成分分析中,钝肩型和刺肩型的标本均有重叠,典型变量分析则显示它们之间存在显著差异(马氏距离和普氏距离的P值均小于0.01)。判别函数分析显示,依据这4个性状两型样本间的正确判别率在67%~89%之间。【结论】结果表明,前翅、后翅、头和小盾片都可以作为北二星蝽钝肩型和刺肩型之间形态变异的评价指标,小盾片特征具有最高水平的鉴别价值;对于所研究的这4个性状来说,北二星蝽钝肩型和刺肩型之间都表现出显著的形状差异,而它们的质心距离差异不显著,说明相对于体型大小差异分析,形状差异分析能更灵敏地揭示谱系间的变异。本研究表明几何形态学可以有效地描述北二星蝽的种内形态变异,为蝽类昆虫种内变异的研究奠定了基础。  相似文献
