SYNOPSIS. A reduction in the growth temperature of Tetrahymena pyriformis strain WH-14 from 35 C to 15 C resulted in distinct alterations in the fatty acid composition of the glycerophospholipids. The proportion of normal saturated acids declined from 26 to 19%; palmitoleic acid increased by 6%, and the composition of the polyunsaturated fatty acids increased in 18:2 Δ6,11(n) and decreased in 18:2 Δ9,12(n) and 18:3 Δ6,9,12(n). The unsaturation index (the average number of double bonds/100 molecules) did not change with a shift in temperature.
Two biosynthetic pathways exist in Tetrahymena for the formation of unsaturated fatty acids. The observed changes in fatty acid composition that accompany a lowering of the environmental temperature can be accounted for by a reduction in the accumulation of products of the fatty acid pathway leading to the formation of γ-linolenic acid [16:0(n) → 18:0(n) → 18:1 Δ9(n) → 18:2 Δ9,12(n) → 18:3 Δ6,9,12(n)] and an increase in the components of the pathway leading to the formation of 18:2 Δ6,11(n) [16:0(n) → 16:1 Δ9(n) → 18:1 Δ11(n) → 18:2 Δ6,11(n)]. The data suggest that the regulatory mechanism in Tetrahymena differs from that found in some bacteria where a simple substitution of unsaturated fatty acids for saturated fatty acids occurs at low culture temperatures.  相似文献

Plants of garden pea ( Pisum sativum L.) were exposed to charcoal-filtered air with or without addition of 65 ± 5 l−1 ozone. Plants were harvested daily for 9 days and lipids were extracted from the second-oldest leaf. Visible injury of this leaf was evident from day 5 on, while the differences in lipids between ozone and control treatments were observed earlier. Ozone caused large decreases in the contents of monogalactosyldiacylglycerol (MGDG), digalactosyldiacylglycerol (DGDG) and sulfoquinovosyldiacylglycerol (SQDG), a slower decrease in the content of phosphatidylcholine (PC), but an increase in the content of phosphatidylethanolamine (PE) per leaf area, compared with exposure to charcoal-filtered air. The content of phosphatidylglycerol (PG) was unaffected by ozone. Compared with charcoal-filtered air, fumigation with ozone resulted in a decrease in the proportion of linolenic acid (18:3) of the total lipid extract, with a concomitant increase in the proportion of linoleic acid (18:2). For individual lipids, ozone caused a similar pattern of decreased 18:3 and increased 18:2 in MGDG, SQDG, PC and PE, while the fatty acid composition of DGDG was unaffected. In PG, ozone decreased the proportions of 18:3 and trans -Δ3-decenoic acid (16:1trans), balanced by increased proportions of palmitic and oleic acids. The contents of chlorophylls and carotenoids were unaffected by ozone. Our results show that moderately elevated levels of ozone cause significant changes in the polar lipid composition of garden pea leaves and in the level of unsaturation of the lipid acyl groups and, furthermore, that ozone has different effects, which could be direct or indirect, on chloroplast lipids (MGDG, DGDG, SQDG, PG acylated with 16:1trans) and cytosolic membrane lipids.  相似文献

Abstract The biosynthesis of the positional isomers of the monounsaturated fatty acids of Methylococcus capsulatus (Bath) has been investigated by studying the incorporation of [2-14C]malonyl CoA into long-chain fatty acids in vitro. The major unsaturated products were Δ 9 16:1 and Δ 11 18:1; however, Δ 8, Δ 10 and Δ 11 16:1, as well as, Δ 10, Δ 12 and Δ 13 18:1 were also synthesized. The exclusion of O2 from the reaction vessel did not affect the synthesis of unsaturated fatty acids or the double bonds positions. Cerulenin inhibited the synthesis of unsaturated fatty acid more than saturated fatty acid. The use of both [1-14C] octanoate and [1-14C] decanoate as substrate resulted in the synthesis of long-chain fatty acids, however, unsaturates were only synthesized from octanoate. These results imply that the unique positional isomers of M. capsulatus are not synthesized by an aerobic mechanism.  相似文献

The fatty acid composition of the total lipids from two Conidiobolus species was studied by gas—liquid chromatography. The major fatty acids of C. lamprauges were palmitic acid (C16:0), oleic acid (C18:1), linolenic acid (C18:3), and arachidonic acid (C20:4). For C. eurymitus , myristic acid (C14:0), C16:0, and linoleic acid (C18:2) were the most abundant acids. The fatty acid composition of C. eurymitus was quite different from that of the Conidiobolus species as mentioned in other reports. The lipid composition of the total lipids of C. lamprauges and C. eurymitus was also studied by thinchrography on quartz rods. Triglycerides and phospholipids were the major components in the two Conidiobolus species.  相似文献

Female tadpoles of Rana catesbeiana were laparotomized at metamorphic stages XI-XIII and an empty capsule or one containing cyanoketone (CK), which is an inhibitor of Δ5-3β-hydroxysteroid dehydrogenase (Δ5-3β-HSD), was implanted intraperitoneally. Ovarian activity of Δ5-3β-HSD was examined histochemically 2 months later, estradiol-17β (E2) secretion by the ovaries was measured by RIA 4 months later and histological changes of the ovaries were examined 6 months later. The Δ5-3β-HSD activity of the CK-treated ovaries was much lower than that of controls. E2 secretion per froglet by CK-treated ovaries was about one third that of controls (p<0.001). Histological examination showed various degrees of masculinization of the ovaries, about 28% of which were totally transformed into testis-like structures.
As a result of suppressed Δ5-3β-HSD activity, dehydroepiandrosterone would have accumulated, resulting in deficient E2 secretion and, therefore, ovarian masculinization. In tadpoles, this effect does not depend on the pituitary, whereas interrenal hyperplasia and hyperactivity do, indicating that interrenal function is not essential for ovarian masculinization. From these findings and our previous results, we suggest that disturbance of steroidogenesis by CK in the ovaries results in their masculinization.  相似文献

This study investigated the impact of lipid extraction, CaCO3 removal and of both treatments combined on fish tissue δ13C, δ15N and C:N ratio. Furthermore, the suitability of empirical δ13C lipid normalization and correction models was examined. δ15N was affected by lipid extraction (increase of up to 1·65‰) and by the combination of both treatments, while acidification alone showed no effect. The observed shift in δ15N represents a significant bias in trophic level estimates, i.e. lipid-extracted samples are not suitable for δ15N analysis. C:N and δ13C were significantly affected by lipid extraction, proportional to initial tissue lipid content. For both variables, rates of change with lipid content (ΔC:N and Δδ13C) were species specific. All tested lipid normalization and correction models produced biased estimates of fish tissue δ13C, probably due to a non-representative database and incorrect assumptions and generalizations the models were based on. Improved models need a priori more extensive and detailed studies of the relationships between lipid content, C:N and δ13C, as well as of the underlying biochemical processes.  相似文献

The effects of NaCl and replacement of K+ by Na+ on the lipid composition of the two sugar beet inbred lines FIA and ADA were studied (a) with increasing additions of NaCl to the basal medium, and (b) with increasing replacement of K+ by Na+ at the same total concentration as in the basal medium. Direct relations were noted between NaCl concentration of the nutrient solution and the phospholipid concentration in the roots of FIA, the genotype characterized by a low K+/Na+ ratio, as well as between NaCl in the medium and the phospholipid concentration in the shoots of ADA, the genotype with a high K +/Na + ratio. The sulfolipid level in the roots of FIA was maintained at higher NaCl concentrations, while it was decreased in ADA. The glycolipid concentration in the shoots of ADA and the degree of unsaturation of the fatty acids of the total lipid fraction were decreased by salinity, indicating reduced biosynthesis of chloroplast glycolipids and/or accelerated oxidation of these lipids in the presence of NaCl.
In the Na+ for K+ replacement experiment a low content of K+ in the medium resulted in decreased levels of total lipids, phospholipids and sulfolipid in the roots of both genotypes, which did not relate to root growth. K+-leakage from the roots at low K+-level in the medium may be reduced by the increase in saturation of the lipids. In the shoots of ADA increased levels of total lipids, phospholipids and Sulfolipid were noted at a low K+-concentration of the nutrient solution.  相似文献

The influence of different feeding levels below and slightly above maintenance on whole body δ13C and δ15N values of Nile tilapia Oreochromis niloticus was examined. The energy budget of each fish was determined by indirect calorimetry. The δ13C values of the lipid-free material of Nile tilapia fed below and slightly above maintenance level did not differ between the feeding groups, but the δ13C values in the lipids and the δ15N values of the lipid-free material showed small but significant differences. Those fish with a negative lipid retention had significantly higher δ13C values in the lipid fraction compared to fish that synthesized fatty acids. There was a significant negative correlation between the amount of energy metabolized by the fish and both the δ13C values in the lipids and the δ15N values of the lipid-free material. Fasting and feeding below the maintenance level may influence the isotopic composition of animals and should therefore be considered in ecological and nutritional studies.  相似文献

The question of whether membrane expansion, which is caused by anesthetics in animal systems, alters the lipid composition of plant cell membranes was investigated. We have measured the effects of several anesthetics on the relative amounts of the principal fatty acids from the polar lipids of barley (Hordeum vulgare L.) root membranes. Procaine, dibucaine, tetracaine, chloroform and, to a lesser degree, methanol increased the proportions of palmitic, stearic and oleic acids and decreased the proportions of linoleic and linolenic acids. Ethanol had no significant effect. Total amounts of the fatty acids from the polar lipids of roots in procaine solution decreased markedly so that all of the acids decreased in amount. The anesthetic was effective as soon as the roots were introduced to the solution and the changes progressed at constant rates for 6 h. Only the polar membrane lipids were altered; other lipids were not affected. Increased hydrostatic pressure of about 1.0 MPa largely prevented the anesthetic effects, including the decrease in the total amounts of the fatty acids. Hydrostatic pressure as high as 2 MPa had no effect per se on the membrane lipid composition. These results indicate that anesthetics cause expansion of the root membranes which results in the lipid changes. That a compositional change in the membrane lipids involves a conformational change such as expansion is an indication of the nature of the link between changes in the membrane lipids and changes in function of areas where hydrophilic ions permeate.Abbreviations 16:0 palmitic acid - 18:0 stearic acid - 18:1 oleic acid - 18:2 linoleic acid - 18:3 linolenic acid  相似文献

Boraginaceae seeds are particularly rich in Γ -linolenic acid (6,9,12-octadecatrienoic acid, Γ -18:3). In microsomes, the analysis of phosphatidylcholine (PC) molecular species by HPLC led to identification of 15 different molecular species; among them 4 contained Γ -18:3, mostly at position 2 of sn -glycerol. Time courses of acylation and desaturation in PC molecular species were examined when [14C]oleoyl-CoA or [14C]linoleoyl-CoA was provided as substrates to isolated microsomes. With [14C]oleoyl-CoA or [14C]linoleoyl-CoA and in the absence of NADH, 3 main labelled PC molecular species were found: 18:2/[14C]18:1, 16:0/[14C]18:1 and 18:1/[14C]18:1. When NADH was present in the incubation medium, the fatty acids were progressively desaturated by the Δ12- and Δ6-desaturases successively (with [14C]oleoyl-CoA as precursor) or by the Δ6-desaturase alone (with [14C]linoleoyl-CoA as precursor). In both types of experiments, 7 final desaturation products in microsomes were evidenced; among them, 3 contained radioactive Γ -18:3, i.e . 18:2/[14C] Γ -18:3, 18:1/[14C] Γ -18:3 and 16:0/[14C] Γ -18:3. While the Δ12-desaturase had no specificity for position on the glycerol backbone, labelled Γ -linolenic acid was recovered exclusively in the sn -2 position.  相似文献

Lipids comprising the stenols, stanols, polar lipid fatty acids, alkanes and alkenes of blue-green algal-(diatomaceous)-microbial mats and cores (modern cold water stromatolites) collected from three Antarctic lakes were identified and compared with those of other algae. The major stenols were: (cholesta-5, 22-dien-3β-ol, cholest-5-en-3β-ol, 24-methylcholesta-5, 22-dien-3β-ol, 24-methyl-cholest-5-en-3β-ol, 24-ethylcholesta-5, 22-dien-3β-ol, and 24-ethylcholest-5-en-3β-ol). The presence of C28 Δ3, 22 stenols, as well as other C28 stenols, was suggestive of diatom input. C29 stenols may have originated from blue-grern algae. However, the high concentrations of stenols present and the lack of Δ7 stenols was atypical for known stenol components of several blue-green algal species previously reported. The occurrence of these stenols and other lipid markers strongls implicate diatoms as well as blue-green algae as important biogenetic sources of lipids and has established the potential for studies of lipid diagenesis in these unique cold, freshwater stromatolites .  相似文献

Abstract: [1-3H, 1-14C]Palmitaldehyde(3H:14C= 15) was injected intracerebrally to 18-day-old rats and incorporation of radioactivity into brain lipids was followed over a 24-h period. The substrate was metabolized primarily by oxidation to palmitic acid with loss of tritium and, to a lesser extent, by reduction to hexadecanol. The alkyl moieties of the ethanolamine phospholipids showed considerably lower 3H:14C ratios than the substrate, indicating a substantial participation in ether lipid synthesis by tritium-free alcohols derived from 14C-labeled fatty acids. Virtually no 3H radioactivity was found in alkenyl moieties, indicating stereospecificity of both reduction of aldehyde and dehydrogenation of alkyl to alkenyl glycerolipid. The data are consistent with the general concept that plasmalogen biosynthesis proceeds exclusively through fatty alcohols and alkyl glycerolipids and that fatty aldehydes cannot be utilized directly.  相似文献

The biosynthesis of lipids in the mycelium and sporophore of Pleurotus sajor caju was studied. Whereas in the mycelium the biosynthesis of lipids was directly primarily towards storage (e.g. tri-acylglycerols), in the sporophore it was directed towards structural components (e.g. sterols). The incorporation of 14C precursors into non-polar and polar lipid fractions was generally similar for 14C acetate, 14C palmitate, 14C oleate and 14C linoleate in the case of mycelium and sporophore. It appears that linoleic acid was utilised as a source of acetate for lipid biosynthesis in the sporophore. A significantly higher incorporation of label was seen in sporophore sterol than in mycelial sterol. Malate dehydrogenase activity increased in the mycelium grown in the presence of lipids. Lipase of P. sajor caju was inducive. The growth of P. sajor caju was enhanced by increased lipid utilisation. The implications of these results on commercial cultivation of this fungus are discussed.  相似文献

Ladderane lipids are unusual membrane lipids of bacteria that anaerobically oxidize ammonium to dinitrogen gas (anammox). Ladderane lipids contain linearly concatenated cyclobutane rings for which the pathway of biosynthesis is currently unknown. To investigate the possible biosynthetic routes of these lipids, 2-13C-labelled acetate was added to a culture of the anammox bacterium Candidatus Brocadia fulgida. Labelling patterns obtained by high-field 13C nuclear magnetic resonance spectroscopy of isolated lipids indicated that C . Brocadia fulgida synthesizes C16:0 and iso C16:0 fatty acids according to the known pathway of type II fatty acid biosynthesis. The 13C-labelling pattern of the C8 alkyl chain of the C20 [3] ladderane monoether also indicated the use of this route. However, carbon atoms in the cyclobutane rings and the cyclohexane ring were nonspecifically labelled and did not correspond to known patterns of fatty acid synthesis. Taken together, our results indicate that it is unlikely that ladderane lipids are formed from the cyclization of polyunsaturated fatty acids as hypothesized previously and suggest an alternative, although as yet unknown, pathway of biosynthesis.  相似文献

When heterotrophic cell cultures of red goosefoot (Chenopodium rubrum) turned photoautotrophic, their contents of various glycolipids and phospholipids increased. The total lipids and the individual lipid classes, especially monogalactosyldiacylglycerols, became richer in linolenic and poorer in linoleic acids. When photoautotrophic cell cultures were rendered heterotrophic again a reversal of changes occurred; both the composition of lipids and the patterns of their constituent fatty acids became similar to those of the starting heterotrophic cultures.The results indicate that the biosynthesis of linolenic acid in photoautotrophic cell cultures involves mainly desaturation of linoleic acid and that chain extension of hexadecatrienoic acid is possibly another, though minor pathway. Monogalactosyldiacylglycerols are apparently the substrates preferred for linolenic acid biosynthesis, whereas various phospholipids are the substrates preferred for linoleic acid biosynthesis.During a growth period of 6 weeks, the levels of polyunsaturated fatty acids in the lipids from both heterotrophic and photoautotrophic cell cultures decrease with time, whereas the proportions of palmitic acid increase.  相似文献

Primary hepatocytes from wild northern pike Esox lucius were incubated with radiolabelled linolenic acid ([l-14C]-18:3(n-3)) to assess their ability to synthesize docosahexaenoic acid [22:6(n-3)]. The distribution of radioactivity in lipid classes and hepatocyte polyunsaturated fatty acids (PUFA) was measured over the time-course of 24h. The majority of radioactivity from [l-14C]-18:3(n-3) was recovered in hepatocyte triacylglycerols (TAG) and phosphatidylcholine (PC). The levels of radioactivity in TAG and in most of phospholipids, including PC, increased significantly over the incubation period. Radioactivity from [1-14C]-18:3(n-3) was recovered in several hepatocyte PUFA, including 22:6(n-3), and the Δ6 and Δ5-desaturation products 18:4(n-3) and 20:5(n-3). The presence of radioactivity in C24 (n-3) PUFA may be evidence that the biosynthesis of 22:6(n-3) in pike proceeds via a pathway independent of Δ4-desaturation. Analysis by radio gas chromatography revealed that radiolabelled 24:6(n-3) was present among the desaturation and elongation products of [l-14C]-18:3(n-3). The results establish that, under the in vitro conditions employed, pike hepatocytes are able to convert linolenic acid to 20:5(n-3) and 22:6(n-3).  相似文献

Abstract: Effects of altered composition of membrane lipids on high-affinity uptake of l -glutamate and taurine were studied in an established neuroblastoma cell line M1. Increase in participation of certain fatty acids (20: 5ω3 and 22: 5ω3) as constituents of membrane lipids resulted in a fourfold increase in the maximum initial rate of l -glutamate uptake (Vmax) while increase in Vmax of taurine uptake was much smaller. Neither structural requirements of l -glutamate uptake nor passive permeability of the membrane to l -glutamate or taurine seemed to be influenced by the alterations in the lipid composition. Since increased proportions of 20: 5ω3 and 22: 5ω3 in the membrane phosphatides caused no dramatic changes in kinetic parameters of taurine uptake and incorporation of either linoleic or linolenic acid alone into the phosphatides had only a relatively small effect on some of the measured parameters, the possibility of a specific relationship between 22: 5ω3 and/or 20: 5ω3 and l -glutamate uptake is discussed. Unlike l -glutamate uptake systems in other preparations, the high-affinity uptake system of l -glutamate in neuroblastoma cells did not readily accept l -aspartate as a substrate.  相似文献

The neutral fraction of nonstarch lipids in developing brown rice (Oryza sativa L., cv IR42) was accumulated up to 16 days after flowering (DAF), but phospholipids and glycolipids increased only up to 8 DAF. Fatty acids accumulated in nonstarch lipids until 12 DAF. However, the proportion of linolenic acid in the lipid fraction decreased and that of oleic acid increased during this period. Accumulation of fat-by-hydrolysis in the brown rice occurred until 20 DAF and followed closely that of starch. The proportion of linolenic acid decreased and that of linoleic acid increased until 16 DAF. The fatty acid composition of fat-by-hydrolysis and starch lipids were identical and fat-by-hydrolysis accounted for 48% by weight of starch lipids. Nonstarch lipids were mainly composed of triglycerides and were located in the bran and embryo of mature brown rice. Starch lipids were mainly composed of lysophosphatidyl choline, free fatty acids and lysophosphatidyl ethanolamine, and were located in the endosperm.  相似文献
