We quantified metabolic power consumption as a function of wind speed in the presence and absence of simulated solar radiation in rock squirrels, Spermophilus variegatus, a diurnal rodent inhabiting arid regions of Mexico and the western United States. In the absence of solar radiation, metabolic rate increased 2.2-fold as wind speed increased from 0.25 to 4.0 m·s-1. Whole-body thermal resistance declined 56% as wind speed increased over this range, indicating that body insulation in this species is much more sensitive to wind disruption than in other mammals. In the presence of 950 W·m-2 simulated solar radiation, metabolic rate increased 2.3-fold as wind speed was elevated from 0.25 to 4.0 m·s-1. Solar heat gain, calculated as the reduction in metabolic heat production associated with the addition of solar radiation, increased with wind speed from 1.26 mW·g-1 at 0.25 m·s-1 to 2.92 mW·g-1 at 4.0 m·s-1. This increase is opposite to theoretical expectations. A mathematical model for photoinhibition of leaf photosynthesis was developed by formalising the assumptions that (1) the rate of photoinhibition is proportional to irradiance; and (2) the rate of recovery, derived from the formulae for a pseudo first-order process, is proportional to the extent of inhibition. The photoinhibition model to calculate initial photo yield is integrated into a photosynthesis-stomatal conductance (g s) model that combines net photosynthetic rate (P N), transpiration rate (E), and g s, and also the leaf energy balance. The model was run to simulate the diurnal courses of P N, E, g s, photochemical efficiency, i.e., ratio of intercellular CO2 concentration and CO2 concentration over leaf surface (C i/C s), and leaf temperature (T 1) under different irradiances, air temperature, and humidity separately with fixed time courses of others. When midday depression occurred under high temperature, g s decreased the most and E the least. The duration of midday depression of g s was the longest and that in E the shortest. E increased with increasing vapour pressure deficit (VPD) initially, but when VPD exceeded a certain value, it decreased with increasing VPD; this was caused by a rapid decrease in g s. When air temperature exceeded a certain value, an increase in solar irradiance raised T 1 and the degree of midday depression. High solar radiation caused large decrease in initial photon efficiency (). P N, E, and g s showed reasonable decreases under conditions causing photoinhibition compared with non-photoinhibition condition under high irradiance. The T 1 under photoinhibition was higher than that under non-photoinhibition conditions, which was evident under high solar irradiance around noon. The decrease in C i/C s at midday implies that stomatal closure is a factor causing midday depression of photosynthesis.  相似文献

Summary Intercrossing and irradiation were successfully used in pearl millet (Pennisetum typhoides) to develop multiple interchanges involving up to the total complement of all the chromosomes in one complex. In interchange heterozygotes showing 12 + 1 II and 14, 90.9 and 87.8 per cent of the cells, respectively, had chromosome configurations other than that of 12 and 14 chromosomes. In general, the frequency of such cells resulting from breakdown of the expected complex configuration increased with the increase in the number of translocated chromosomes in the complex. The higher the number of chromosomes involved in the interchange ring, the higher were the pollen and ovule sterility. The results indicated that meiotic instability, deficiency-duplication gametes, and unequal distribution of chromosomes account for increased sterility of multiple interchange heterozygotes. Even though interchanges in pearl millet predominantly show the alternate type of segregation, sterility seems to be the major barrier for the exploitation of the multiple interchange method for gamete selection and the establishment of homozygous lines in this plant species.  相似文献

Batch transglutaminase (MTG) fermentations by Streptovertivillium mobaraense WSH-Z2 at various temperatures ranging from 25 to 35 °C were studied. Dry cell weight and MTG activity could reach their maximal values of 25.1 g/l and 2.94 U/ml, respectively at 30 °C. One typical equation was used to describe the relationship between specific growth rate and culture temperature by comparing several typical equations. Different lag time was observed under various culture temperature. The low lag time was observed under high culture temperature. X = –a 0(TT 0)2 + X 1 + a 1 (1 – exp(a 2 (TT 1))) and U = –a 0 (TT 0 )2 + U 0 + a 1 (1 – exp(a 2 (TT 1 ))) could be used to describe the relationship between temperature and the maximal dry cell weight as well as the maximal MTG activity at each temperature.  相似文献

We report three sets of DNA hybridization experiments conducted to determine relationships among species ofDidelphis (D. albiventris, D. marsupialis, D. virginiana). The 1989 and 1991 sets had fewer replicates per cell than the 1990 series (3.4 and 5.4 vs 9), but in 1991 we distinguished two populations ofD. marsupialis and utilized several individuals for each heterologous comparison. BothPhilander opossum andLutreolina crassicaudata were used as outgroups in 1989, but onlyLutreolina was included in subsequent sets. For each set, we calculated all four standard indices of thermal stability (T mode,T m,T 50 H, and NPH) and constructed trees by least-squares (FITCH) and neighbor-joining methods, both before and after correction for asymmetric reciprocal cell values. Subsets of the 1989 data lacking eitherPhilander orLutreolina were analyzed similarly. To explore measurement imprecision, the corrected and uncorrected matrices for each of the four indices were bootstrapped 100 times for the 1989 set and subsets and 1000 times for the 1990 and 1991 sets. Again, for the 1990 and 1991 data, an additional 100 bootstrapped distances were fitted to user trees representing the three possible pairings ofDidelphis spp. to determine the significance of the FITCH branch lengths. The successive experimental sets generated increasingly consistent evidence for pairingD. marsupialis withD. albiventris. The 1989 experiments involved just 85 comparisons, and only T mode's pairedD. marsupialis withD. albiventris (at bootstrap percentages of 70% or above), but did so whetherPhilander orLutreolina or both were included as the outgroup(s). FITCH and neighbor-joining trees had identical topologies for T mode's but sometimes differed for the other measures. In contrast, all but one of the corresponding FITCH and neighborjoining trees matched for the 1990 and 1991 data, and three of the four distance measures (T mode, T m, and T 50 H) unitedD. marsupialis withD. albiventris at bootstrap percentages averaging 81%; NPH gave a different result for 1990, associatingD. marsupialis withD. virginiana. Further, all but 2 of the 16 matrices for 1990 and 1991 gave mean bootstrapped branch lengths for a consensus pairing that were positive and at least one standard deviation from zero, despite the very short internodes recovered. These results illustrate that the potential of DNA hybridization for resolving very close relationships depends on both the index and the experimental design employed. We conclude that of the three species,D. albiventris andD. marsupialis shared a more recent common ancestor and estimate thatDidelphis spp. have diverged at about 0.39% in nucleotide sequence per myr.Deceased.  相似文献

The effect of H2O2 on photosynthetic O2 evolution and photosynthetic electron transfer in cells of cyanobacteria Anabaena variabilis and Anacystis nidulans was studied. The following experiments were performed: 1) directly testing the effect of exogenous H2O2; 2) testing the effect of intracellular H2O2 generated with the use of methyl viologen (MV); 3) testing the effect of inhibiting intracellular H2O2 decomposition by salicylic acid (SA) and 3-amino-1,2,4-triazole (AT). H2O2 inhibited photosynthetic O2 evolution and light-induced reduction of p-benzoquinone (BQ) + ferricyanide (FeCy) in the Hill reaction. The I50 value for H2O2 was 0.75 mM. Photosynthetic electron transfer in the cells treated with H2O2 was not maintained by H2O2, NH2OH, 1,5-diphenylcarbazide, tetraphenylboron, or butylated hydroxytoluene added as artificial electron donors for Photosystem (PS) II. The H2O CO2, H2O MV (involving PSII and PSI) and H2O BQ + FeCy (chiefly dependent on PSII) electron transfer reactions were inhibited upon incubation of the cells with MV, SA, or AT. The N,N,N",N"-tetramethyl-p-phenylenediamine MV (chiefly dependent on PSI) electron transfer was inhibited by SA and AT but was resistant to MV. The results show that H2O2 inhibits photosynthetic electron transfer. It is unlikely that H2O2 could be a physiological electron donor in oxygenic photosynthesis.  相似文献

AxenicTrentepohlia odorata was cultured at three different NH4Cl levels (3.5 × 10–2, 3.5 × 10–3, 3.5 × 10–4 M) and three different light intensities (48, 76, 122 µmol m–2 s–1). Chloride had no effect on growth over this range of concentration. High light intensity and high NH4Cl concentration enhanced the specific growth rate. The carotenoid content increased under a combination of high light intensity and low N concentration. WhenD. bardawil was exposed to the same combination of growth conditions, there was an increase in its carotenoid content. The light saturation and the light inhibition constants (K s andK i, respectively) for growth, and the saturation constant (K m) for NH4Cl were determined. TheK s andK i values were higher inT. odorata (66.7 and> 122 mol m–2 s–1, respectively) than inD. bardawil (5.1 and 14.7 µmol m–2 s–1, respectively). Summary The relationship between available methionine concentration and the levels of phaseolin — the major seed storage proteins of the common bean — was studied using three groups of genetic materials: First, the F2 progenies of interspecific crosses between P. vulgaris cultivars and aP. coccineus subsp. coccineus line (cv. Mexican Red Runner) having no detectable phaseolin; second, the F2 progenies and segregating F3 families of crosses between cultivated P. vulgaris lines and a Mexican wild bean accession (PI 325690-3) carrying a gene producing a reduction in phaseolin content; third, two inbred backcross populations: SanilacxBush Blue Lake 240 (population 2) and Sanilacx15R 148 (population 6). Total seed N levels were determined by micro-Kjeldahl, phaseolin levels by rocket immunoelectrophoresis and available methionine levels by the Streptococcus zymogenes bioassay. Our results indicate that in all the genetic materials studied, with the exception of population 6, higher phaseolin levels lead to increased available methionine concentration. Although phaseolin has a low methionine concentration, it is actually a major source of available methionine in common bean seeds, because it represents a large part of total seed nitrogen and because limited differences exist between the methionine concentrations of the different protein fractions. This contrasts with the situation in cereals such as maize, barley and sorghum, where increased levels of the major limiting amino acid (lysine) can be achieved through a decrease in the amounts of the main seed storage protein fraction (prolamines). In population 6, no relationship was observed between available methionine and phaseolin content. Other factors, such as additional methionine-rich polypeptides or the presence of tannins, might obscure the positive relationship between phaseolin and available methionine content in population 6.  相似文献

Summary The effects of ambient temperatures of 10°C and 30°C and of E. coli endotoxin on brain temperature and plasma iron level were investigated in unrestrained wild house mice, Mus musculus. In control animals (i.p. saline-injected) exposed to cold environmenta the brain temperature decreased and plasma iron levels were lower than those observed under thermoneutral conditions (30°C). Animals injected i.p. with endotoxin (0.5 g·kg-1) and placed at 30°C showed a drop in plasma iron level during the fever episode. The results provide strong evidence for a relationship between brain temperature and plasma iron level in control mice under thermoneutral conditions, and show that during cold exposure or after injection of endotoxin, there is no linear correlation between brain temperature and plasma iron. Homogeneity-time is defined and introduced as the criterion for mixing quality in bioreactors. The criterion could replace the mixing time, in the case, when more than one measuring point (sensors) is included in the measuring system. Summary European green lizards, Lacerta viridis, show a distinct annual cycle in their day and nighttime selected body temperature (T b) levels when monitored under natural photoperiod. The amplitude between daily photophase and scotophase temperatures varies throughout the year. Highest body temperatures with smallest day/night variation are selected from May through July. Throughout fall, the difference between day and nighttime selected T b levels increases. Lizards inevitably enter a state of winter dormancy which terminates daily rhythmicity patterns. Under natural photoperiodic conditions, cessation of dormancy occurs spontaneously by mid-March, regardless whether high temperatures are available or not. Lacerta viridis respond to an artificial long photoperiod (16 h light, 8 h dark) at all times of the year with modifications in both diel patterns and levels of selected T b to summer-like conditions. When, however, the natural photoperiod at different phases in the annual cycle is held constant for six to eight weeks, T b selection of Lacerta viridis also remains stable at the level corresponding to the prevailing photoperiod. These results implicate that the photoperiod is a more prominent Zeitgeber for seasonal cueing of temperature selection than has been surmised in the past. Further, we suggest that the large variations recorded in daily T b cycles do not imply that this lizard is an imprecise thermoregulator, but rather indicates an important integral process necessary for seasonal acclimatization.  相似文献

Two characteristic temperatures were identified from measurements of the temperature dependence of O2 evolution by Chlorella vulgaris and Anacystis nidulans: T1, the threshold temperature for inhibition of O2 evolution under saturating light conditions, and T2, the upper temperature limit for O2 evolution. Measurement of delayed light emission from photosystem II (PSII) showed that it passed through a maximum at T1 and was virtually eliminated on heating the samples to T2. Related changes were observed in low-temperature (77K) fluoresence emission spectra. Heat-stress had little effect on the absorption properties of the cells at temperatures below T1 but incubation at higher temperatures, particularly under high-light conditions, resulted in extensive absorption losses. An analysis of these measurements suggests that this increased susceptibility to photobleaching is triggered by an inhibition of the flow of reducing equivalents from PSII that normally serves to protect the light-harvesting apparatus of the cells from photo-oxidation. Discovery of permanent hybridity in the very large chromosomes of Gibasis pulchella (Commelinaceae) has allowed specific identification of segmental interchanges in complex heterozygotes. The interchanges are confined to terminal regions, are sometimes very small, and may be unequal in size. Breakpoints have occurred close to major C-bands, probably at euchromatin/heterochromatin boundaries. Complete and disjunctional ring formation at meiosis results in the segregation of two Renner complexes, each of which can be specifically identified with C-banding. The complex carrying the interchanges is usually transmitted through the pollen. Certain chromosomes that have undergone more extensive change than the rest of the complement may have some special significance. There is evidence of small duplications within heterozygous genomes. Lectins from Canavalia brasiliensis (CnBr), Cratylia floribunda (CFL), Vatairea macrocarpa (VML) and Phaseolus vulgaris (PHA) aggregate Rhizobiumbacteria. The relationship between specific sedimentation rate, (based on bacterial dry biomass) of bacterial aggregates and lectin concentrations was hyperbolic and showed bacterial surface affinity by lectins. R. tropici (Rt), R. leguminosarum bv. phaseoli (Rlp) and R. etli (Re) surfaces showed predominantly receptors of galactosidic nature. The Rt surfaces showed very high affinities (k s = ±8.6 × 10–8 ag lectin protein ml–1) by Gal-specific lectins (PHA and VML), and very low affinities (ks=± 4.9 × 10–6) by Glc-specific lectins (CnBr and CFL). The Rlp surface had intermediate affinities by lectins. The Re surface showed high affinities by PHA (ks= ±1.26 × 10–8) and intermediate affinities by VML, CnBr and CFL. The relationship between sedimentation specific (based on lectin weight) and bacterial density was a sigmoid and showed lectin affinity by Rt surfaces. The bacterial sedimentation showed positive cooperative binding of lectins. The Vmax induced by Glc-specific lectins was ±20 of that produced by Gal-specific lectins. The PHA affinity (ks= 1.19 mg dry biomass ml–1) was larger than VML (ks = 1.23). The Glc-specific lectin affinities were smaller than those of Gal-specific. The apparent binding site number of lectins (napp) was: 2.7-PHA; 2.2-VML; 3.2-CFL and 3.2-CnBr. The dissociation constant, ks, of lectin-binding kinetics decreased with sugar-hapten treatment (10 M). The napp decreased in PHA and CFL, increasing in VML + sugar-hapten treatment. This study showed that there is a difference in Rhizobium surfaces for lectin binding.  相似文献

Summary Conditions for the production of microbial uricase byCandida utilis were studied. For the selected strain, hypoxanthine proved to be the most effective inducer of uricase formation. Effects of water stress duration and intensity on gas exchange and leaf water potential were investigated in 7-month-old seedlings of a humid coastal provenance (Gympie) and a dry inland (Hungry Hills) provenance of E. cloeziana F. Muell. and in a dry inland (Chinchilla) provenance of E. argophloia Blakely supplied with 100% (T100), 70% (T70), 50% (T50) of their water requirements, or were watered only after they were wilted at dawn (T0). Seedlings of E. argophloia had the highest midday net photosynthetic rate (A), stomatal conductance (g s), stomatal density and predawn leaf water potential (pd) in all treatments. The E. cloeziana provenances did not differ in these attributes. The T70 and T50 treatments caused reductions in A of 30% in E. argophloia, and 55% in the E. cloeziana provenances. Under the T0 treatment, E. argophloia maintained higher rates of gas exchange at all levels of water stress than E. cloeziana provenances. The estimates of pd and midday water potential (md) at which plants remained wilted overnight were respectively: –2.7 and –4.1 MPa for E. cloeziana (humid), –2.8 and –4.0 MPa for E. cloeziana (dry) and, –3.7 and –4.9 MPa for E. argophloia. Following stress relief, both A and g s recovered more quickly in E. argophloia and in the dry provenance of E. cloeziana than in the humid provenance. We conclude that E. argophloia is more drought tolerant and has a potential for cultivation in the humid and semi humid climates, whilst E. cloeziana has greater potential in the humid subtropical climates.  相似文献

Summary Body temperatures (T b) and daily activity patterns of free-living arctic ground squirrells (Spermophilus undulatus) were determined via telemetry at a field site in northern Alaska. Simultaneous measurements were made of ambient temperature (T a), wind speed (V), and incident solar radiation. The operative environmental temperature (T e) for ground squirrels was obtained from fur-covered, thin metal taxidermic models of the animals. Standard operative temperature (T es), a comparative index of heat flow, was calculated from T e, V, and laboratory measurements of thermal conductivity.During the period of the study (August), S. undulatus were active for about 14 h per day (06.00 to 20.00 h). T b was high throughout the daily cycle, averaging 38–39°C. Circadian variations in T b were slight; average T b values dropped <1°C at night. Daytime T b fluctuations were not closely correlated to activity or to changes in environmental conditions. Air temperatures during the study were low, usually between 10 and 15°C during the day. However, T es in exposed areas was normally higher, even though skies were generally overcast. During periods of sunshine, T es may be as high as 34°C. The absence of nocturnal activity may result from increased costs of thermoregulation at night, which sharply reduces foraging efficiency. The high and stable body temperatures of S. undulatus probably result from thermoneutral daytime T es, low activity levels, and the use of well-insulated nests.  相似文献

Summary Long-term increased or decreased circulating levels of thyroid hormones significantly modify porphyrin concentrations and morphology in the Harderian glands of male and female hamsters. Administration of T3 reduced porphyrin concentrations in females; this treatment or decreasing thyroid hormone levels with KClO4 suppressed the post-castration rise of porphyrins in males. Hypophysectomy led to increased porphyrins in the Harderian glands of males; this rise was suppressed in hypophysectomized males by T3 or T4. In females, hypophysectomy reduced porphyrins which were further reduced by daily administration of T3 or T4. These modifications in the normal females were identical in castrated males. Mitotic activity in the Harderian glands of females was stimulated by KClO4 and by hypophysectomy with or without exogenous T3. In males, castration increased mitotic activity which was suppressed by T3 and exacerbated by KClO4. Increased mitotic activity seemingly follows loss of tissue mass. The data show that thyroid hormones act directly on the Harderian glands rather than indirectly through modification of TSH synthesis/release. Female type glands in males are a consequence of loss of gonadal androgens by castration, or by suppression or loss of thyroid hormones by hypophysectomy or by treatment with KClO4. Effects of environmental conditions on isoprene emission from live oak   总被引:12,自引:0,他引:12  
Live-oak plants (Quercus virginiana Mill.) were subjected to various levels of CO2, water stress or photosynthetic photon flux density to test the hypothesis that isoprene biosynthesis occurred only under conditions of restricted CO2 availability. Isoprene emission increases as the ambient CO2 concentration decreased, independent of the amount of time that plants had photosynthesized at ambient CO2 levels. When plants were water-stressed over a 4-d period photosynthesis and leaf conductance decreased 98 and 94%, respectively, while isoprene emissions remained constant. Significant isoprene emissions occurred when plants were saturated with CO2, i.e., below the light compensation level for net photosynthesis (100 mol m-2 s-1). Isoprene emission rates increased with photosynthetic photon flux density and at 25 and 50 mol m-2 s-1 were 7 and 18 times greater than emissions in the dark. These data indicate that isoprene is a normal plant metabolite and not — as has been suggested — formed exclusively in response to restricted CO2 or various stresses.Abbreviation PPFD photosynthetic photon flux density  相似文献

To gain information on extended flight energetics, quasi-natural flight conditions imitating steady horizontal flight were set by combining the tetheredflight wind-tunnel method with the exhaustion-flight method. The bees were suspended from a two-component aerodynamic balance at different, near optimum body angle of attack and were allowed to choose their own speed: their body mass and body weight was determined before and after a flight; their speed, lift, wingbeat frequency and total flight time were measured throughout a flight. These values were used to determine thrust, resultant aerodynamic force (magnitude and tilting angle), Reynolds number, total flight distance and total flight impulse. Flights in which lift was body weight were mostly obtained. Bees, flown to complete exhausion, were refed with 5, 10, 15 or 20 l of a 1.28-mol·l-1 glucose solution (energy content w=18.5, 37.0, 55.5 or 74.0 J) and again flown to complete exhaustion at an ambient temperature of 25±1.5°C by a flight of known duration such that the calculation of absolute and relative metabolic power was possible. Mean body mass after exhaustion was 76.49±3.52 mg. During long term flights of 7.47–31.30 min similar changes in flight velocity, lift, thrust, aerodynamic force, wingbeat frequency and tilting angle took place, independent of the volume of feeding solution. After increasing rapidly within 15 s a more or less steady phase of 60–80% of total flight time, showing only a slight decrease, was followed by a steeper, more irregular decrease, finally reaching 0 within 20–30 s. In steady phases lift was nearly equal to resultant aerodynamic force; tilting angle was 79.8±4.0°, thrust to lift radio did not vary, thrust was 18.0±7.4% of lift, lift was somewhat higher/equal/lower than body mass in 61.3%, 16.1%, 22.6% of all totally analysable flights (n=31). The following parameters were varied as functions of volume of feeding solution (5–20 l in steps of 5 l) and energy content. (18.5–74.0 J in steps of 18.5 J): total flight time, velocity, total flight distance, mean lift, thrust, mean resultant aerodynamic force, tilting angle, total flight impulse, wingbeat frequency, metabolic power and metabolic power related to body mass, the latter related to empty, full and mean (=100 mg) body mass. The following positive correlations were found: L=1.069·10-9 f 2.538; R=1.629·10-9 f 2.464; P m=7.079·10-8 f 2.456; P m=0.008v+0.008; P m=18.996L+0.022; P m=19.782R+0.021; P m=82.143T+0.028; P m=1.245·bm f 1.424 ; P mrel e=6.471·bm f 1.040 ; =83.248+0.385. The following negative correlations were found: V=3.939–0.032; T=1.324·10-4–0.038·10-4.
